Process for the preparation of a pyridazone The subject of the present patent is a process for the preparation of 6-dimethylamino-2- (p-dimethylaminophenyl) -pyridazon- (3) of formula
EMI0001.0006
The new compound is obtained when 2- (p-nitrophenyl) -pyridazon- (3), which has an exchangeable substituent in the 6-position, is reacted with dimethylamine in the 6-dimetlrylamino-2- (p-nitrophe) obtained - rryl) -pyridazon- (3)
the nitro group in a manner known per se, e.g. B. with hydrogen in the presence of a nickel catalyst, reduced to the amino group and the resulting 6 - Dimethy lamino - 2 - (p - aminophenyl) - pyrid- azon- (3) with a methyl group introduced the agent treated, in particular with a reactive ester of - Methanol, such as with an alkyl halide, e.g.
B. 14iethyl iodide. An exchangeable substituent is, for example, a halogen, in particular a chlorine atom, or a substituted oxy, such as phenoxy, or hlereapto, such as methylmereapto group.
The reaction can be carried out in the presence or absence of thinning agents and / or condensing agents and / or catalysts in an open or closed vessel under pressure.
The 6-Dimethyla.mino-2- (p-dimethylaminophenyl) -pyridazon- (3) obtained in this way melts at 15.0-152. The new compound shows an antipyretic and analgesic effect and is said to be used as a remedy.
The invention is described in more detail in the following example. The relationship between part by weight and part by volume is the same as that between grams and cubic centimeters. Example 6 parts by weight of 2- (p-nitrophenyl) -6-chloropyridazon- (3) are mixed in a closed tube with 10 parts by volume of a 7n-ethyl alcoholic dimethylamine solution,
40 parts by volume abs. Ethanol and 2 parts by volume of triethylamine heated to 165-175 hours for 10 hours. After cooling down, the pipe contents are checked. The residue is crystallized from glacial acetic acid / water. The 6-dimethyl = 1- amino-2- (p-nitrophenyl) pyridazon- (3) of the formula obtained in this way
EMI0002.0004
melts at 210-212.
5.8 parts by weight of 6-dimethylamino-2- (p-nitrophenyl) - pyridazon - (3) are dissolved in 300 parts by volume of absolute ethanol and in the presence of 2.5 parts by weight of Raney nickel for 16-18 hours under normal pressure and hydrogenated at 40. The solution is filtered off from the catalyst and evaporated in vacuo.
The crystallized residue is dissolved in hot water, the hot solution is filtered through animal charcoal and the filtrate is cooled and suction filtered. The pale yellow 6-dimethylamino-2- (p-aminophenyl) -pyridazon- (3) obtained in this way melts at 170 to 172.
5.0 parts by weight of this compound are heated to 110-120 for 6 hours in a closed vessel with 40 parts by volume of absolute alcohol, 7.7 parts by weight of diethyl iodide and 1.3 parts by weight of sodium hydroxide. The reaction mixture is then evaporated in vacuo, taken up in water, made alkaline and extracted several times with chloroform. The chloroform solution is dried over sodium sulfate, filtered and evaporated. The remaining dark brown oil is suspended in hot water. and brought into solution by adding alcohol.
The solution is filtered with the addition of animal charcoal, cooled, the yellow crystals are suction filtered, washed with water and dried.
The 6-dimethylamino-2- (p-dimethylaminophenyl) -pyridazon- (3) obtained in this way melts at 150-152 0 . The 2- (p-nitrophenyl) -6-chloro-pyridazon- (3) used as starting material can be obtained as follows: 40 parts by weight of 2-phenyl-6-oxy-pyridazon- (3) are stirred with 80 parts by volume of concentrated nitric acid and 80 parts by volume of concentrated sulfuric acid at 0-5 slowly added within 60 minutes. The.
The mixture is stirred for a further 2 hours at 0-10 and then slowly mixed with 320 parts by volume of water. The temperature then rises to 35. After cooling, the light yellow nitro derivative is sucked on. The crude product is taken up in soda solution, the solution is filtered with animal charcoal and the 2- (p-nitrophenyl) - 6-oxy-pyridazon- (3) is precipitated with 2N hydrochloric acid, suction filtered and dried. It melts at 289-291.
10 parts by weight of this compound who heated with 25 parts by volume of phosphorus oxychloride 21/2 hours on a boiling water bath. The mixture is then poured into water at 40 to 50, with the addition of ice being kept at this temperature. The 2- (p-nitrophenyl) -6-chloro-pyridazon- (3) crystallizes out. It can be recrystallized from ethyl acetate: and then melts at 195-196.
