Das so erhaltene Produkt, das eine neue Verbindung darstellt, ist ein blassgelbes, kri stallines festes Produkt vom Smp. 300 C (Zer setzung).
Es kann durch Umsetzung mit Na triuunchlorid oder Natriiunbromid in wässriger Lösung in 4-Amino-6- (2'-amino-6'-methyl- pyrimidyl-4'-amino) -chinolin-1,1'- dimethochlo- rid vom Smp. 313 C (Zersetzung) bzw. 4 Amino-6- (2'-amino-6'-methyl-pyrimidyl-4'- amino)-chinolin-1,1'-dimethobromid vom Smp. 300 C (Zersetzung) übergeführt werden.
Die Umsetzung kann durch Erhitzen der vereinigten Ausgangsstoffe, zweckmässiger weise, wenn auch nicht notwendigerweise, in einem flüssigen Medium und in Gegenwart. einer Säure, durchgeführt werden. Als ge eignetes flüssiges Medium ist beispielsweise Wasser und als geeignete Säure beispielsweise Salzsäure zu nennen.
Als Jodanionen abgebende Verbindung kann man beispielsweise Hydrogenjodid oder Alkalimetalljödide, wie z. B. Kaliumjodid, verwenden.
Das nach dem erfindungsgemässen Verfah ren erhaltene Produkt besitzt eine starke trypanozide Wirksamkeit, wie sich bei dessen Prüfung in Mäusen gegen Trypanosomaarten gezeigt hat.
Im folgenden Beispiel bedeuten Teile Ge wichtsteile.
<I>Beispiel:</I> 2,4 Teile 4,6-Diaminochinolin-methjodid, 2,3 Teile 4-Chlor-2-amino-6 methylpyrimidin-1 methjodid iuid 20 Teile 1,5 % iger Salzsäure werden zusammen während 1 Stunde unter Rückfluss zum Sieden erhitzt. Nach Abk_ üh- lüng wird das Produkt abfiltriert und mit wenig kaltem Wasser gewaschen.
Man löst das Produkt anschliessend in 200 Teilen heissem Wasser, worauf die Lösung mit Hohle behan delt und filtriert wird. Durch Zugabe eines Überschusses an- Kaliumjodid zum Filtrat wird 4-Amino-6- (2'-amino-6'-methyl-pyrimidyl- 4'-amino)-chinolin-1,1'-dimethjodid ausgefällt, das aus 50 % igem Alkohol in Form blassgelber Nadeln vom Smp. 300 C (Zersetzung) aus kristallisiert.
Das Ausgangsmaterial, 4,6-Diaminochino- lin-methjodid, kann wie folgt hergestellt wer den: Man löst 7,8 Teile Natriiunhydroxyd in 95 Teilen Wasser, versetzt die Lösung mit 46,7 Teilen 6-Acetylamino-4-oxychinolin-carbon- säure-äthylester (Hermack und Weatherhead, J. Chem. Soc. 1940, 1167) und erhitzt das Ge misch unter Rückfluss während 2 Stunden zum Sieden.
Es werden 50 Teile Wasser zu gesetzt, worauf die Lösung mit Kohle behan delt und filtriert wird. Das sich beim Ab kühlen abscheidende Natriumsalz wird mit wenig kaltem Wasser gewaschen und aus 200 Teilen kaltem Wasser kristallisiert.
Man löst das auf diese Weise gereinigte Natriumsalz in 800 Teilen Wasser und behandelt die Lö sung bei Siedetemperatur mit 100 Teilen 2n- Essigsäure. Die auf diese Weise ausgefällte 6 - Acetyl - amino - 4 - oxy-.chinolin-2-carbonsäure wird abfiltriert, mit Wasser gewaschen und getrocknet. Sie weist einen Smp. von 322 bis 323 C (Zersetzung) auf.
12 Teile 6-Acetyl-amino-4-oxy-chinolin-2- carbonsäure, 120 Teile Chinolin und 1,2 Teile Kupferbronze werden gerührt und während 2 Stunden unter Rückfluss zum Sieden erhitzt. Nach Abkühlung wird das abgeschiedene Pro- dukt abfiltriert, mit Aceton gewaschen und getrocknet.
Das auf diese Weise erhaltene 6-Acetyl-amino-4-oxy-chinolin wird durch Kristallisation aus einem grossen Volumen Wasser gereinigt und anschliessend während 6 Stunden bei 140 C im Vakuum getrocknet. Es weist einen Smp. von 280 bis 282 C auf.
Man vermischt 28 Teile 4-Oxy-6-acetyl- amino-chinolin mit 75 Teilen Phosphoroxy- chlorid und erhitzt das Gemisch unter Rühren und unter Rüekfluss auf 120 bis 130 C. Diese Temperatur wird aufrechterhalten, bis alles in Lösung gegangen ist. Das Reaktionsgemisch wird abgekCihlt und einem Gemisch von 1200 Teilen Eis und 527 Teilen 37 % iger wässriger Natrimnhydroxydlösung zugesetzt.
Das aus gefällte Produkt wird abfiltriert, mit Wasser alkalifrei gewaschen, getrocknet und aus Essig säure-Äthylester kristallisiert, wobei man 4- Chlor-6-acetylamino-chinolin in Form farb loser Kristalle vom Smp. 223 bis 224 C er hält.
14,8 Teile 4-Chlor-6-acetylamino-chinolin und 20 Teile Phenol werden zusammen auf 170 bis 175 C erhitzt, worauf unter Rühren während 3,25 Stunden Ammoniak eingeleitet wird. Hierauf wird das Reaktionsgemisch ab gekühlt und mit 100 Teilen Wasser unter Rühren versetzt. Dann wird Natrium hydroxydlösung zugesetzt, um die Lösung gegen Claytongelb alkalisch zu stellen. Das ausgefällte 4-Amino-6-acetylamino-chinolin wird isoliert, mit wenig kaltem Wasser ge waschen und aus Wasser kristallisiert, wobei es in Form praktisch farbloser Kristalle vom Smp. 244 bis 245 C erhalten wird.
Man suspendiert 4,2 Teile 4-Amino-6- acetylamino-chinolin in 30 Teilen Nitro- benzol und erhitzt das Gemisch bei 100 C. Man versetzt allmählich unter Rühren mit 2,73 Teilen Dimethylsulfat und erhitzt das Gemisch während 2 Stunden bei 100 C. Das abgeschiedene feste Produkt wird abfiltriert und durch Waschen mit Aceton vom Nitro- benzol befreit.
Dieses rohe 4-Amino-6-acetyl- amino-chinolin-methosulfat wird mit 14 Teilen konz. Salzsäure und 7 Teilen Wasser wäh rend 10 Minuten aufgekocht. Die heisse Lö sung wird mit Natriiuncarbonat neutralisiert. Durch Zugabe von Natriumjodid zur erhal tenen Lösung wird 4,6 - Diaminochinolin- methjodid ausgefällt, das abfiltriert und mit wenig Wasser gewaschen wird.
Es kristalli siert aus Wasser in Form von gelborange ge färbten Kristallen vom Smp. 254 bis 255 C.
