Verfahren zur Extraktion und Vakuumverdampfung und Vorrichtung zu seiner Durchführung.
Gegenstand der Erfindung ist ein Verfah- ren zur Extraktion und Vakuumverdampfung, welches dadurch gekennzeichnet ist, dass die Extraktion unter Vermeidung von ErwÏrmung durchgef hrt wird, wÏhrend die Regeneration der Extraktionsfl ssigkeit im Destilliergefϯ unter Vakuum und unterhalb einer die Wirkstoffe des jeweiligen Extraktes schädigenden Temperatur durchgeführt und kondensierte Extraktionsfl ssigkeit bei Raum temperatur periodisch wiederholt demEx- traktionsgefäss zugeführt wird.
Weiter betrifft die Erfindung eine Vor richtung zur Durchführung des erfindungs- gemassen Verfahrens, die dadureh gekenn zeiclmet ist, dass ein durch periodiseh ange legtes Vakuum in ein unter Vakuum peri odisch arbeitendes Destillationsgefäss entlee- rendes Extraktionsgefϯ vorgesehen ist, das mit einem mit dem Flüssigkeitsspiegel der in das Destillationsgefϯ abgesaugten, angereicherten Extraktionsfl ssigkeit sich senkenden.
nach Entleerung des Extraktionsgefässes gegen die Absaugleitung sperrenden Schwim nerventil versehen ist, und dass ein von dem Destillationsgefϯ mit regenerierter Extrak tionsflüssigkeit beschicktes, das Extraktions- gefäss mittelbar speisendes Vorratsgefäss vorgesehen ist.
Bei bisher übliehen Extraktionsverfahren unter Verwendung der bekannten Einrich- tungen wird die sich anreichernde Extrak tionsflüssigkeit entweder lange Zeit auf ver hältnismässig hoher Temperatur gehalten oder laufend so weit erhitzt, dass das Extraktions- mittel verdampft und nach Kondensation wieder mit dem Extraktionsgut in Berührung gebracht wird. Eine Schädigung des Extraktes lässt sich dabei insbesondere dann nicht vermeiden, wenn es sich um das Extrahieren von Stoffen handelt, die gegen Wärme und insbesondere langandauernde Einwirkung von Wärme empfindlich sind.
Die Erfindung ermöglicht ein schonendes Arbeiten, so dass eine Schädigung der Wirkstoffe des Extraktes vermieden wird.
In der Zeichnung ist eine beispielsweise Ausführungsform der erfindungsgemässen Vorrichtung schematisch dargestellt, an Hand welcher auch nachfolgend das Verfahren naeh der Erfindung beispielsweise erlÏutert wird.
In einem Extraktionsgefäss 1 wird Extraktionsgut ausserhalb des koaxialen Siebein- satzes 5 angeordnet. Das Extraktionsgefäss besitzt einen Ansatz 2, der durch einen Gummi- stopfen 26 verschlossen ist, durch den ein Kapillarrohr 28 mit der ausgezogenen Spitze nach au¯en durchgefilhrt ist. Oben ist das Extraktionsgefäss durch einen Ansatz mit Gummistopfen 3 verschlossen, durch den das Kapillarrohr 6 der Schwanenhalsrohrverbin- dung 7, die das Extraktionsgefϯ 1 mit dem Zwischengefäss 25 verbindet, durchgeht.
Der Schwimmerventilkorper 4 mit seiner F h rungsstange 27 dient zum Verschlu¯ der Leitung 8, durch die bei Anlegung von Vakuum die Extraktionslosung über die Zuführungs ieitung 10 in das Destillationsgefäss 9 geführt wird, das in dem Wärmebad 1 4 angeordnet ist und mit einem Kapillarrohr 12 (wie es üblicherweise bei Vakuumverdampfern angewendet wird) versehen ist und aus dem der
Extrakt durch einen verschliessbaren Ablauf
11 am Boden des Gefässes 9 zum Beispiel naeh Fortnahme des Warmebades abgezogen wird,
wÏhrend die DÏmpfe des regenerierten Ex traktionsmittels dureh den Kolbenhals 15 und das mit Thermometer 13 versehene Rohr 16 in einen Kühler 17 abgezogen werden, aus dem das Kondensat in das Vorratsgefäss IS gelant, an das Vakuum unter Zwischenschaltung eines K hlers 21 durch eine Leitung 24 angelegt ist, die-und damit die gesamte Vor- richtung gesteuert wird durez den Hahn 22. der die Vorrichtung entweder an die Va kuumleitung 24 oder die Leitung zur Aussen- atmosphäre 23 anschliesst.
Das Vorratsgefäss 18 ist durch ein Tellerventil 19. 20 gegenüber dem Zwisehengefäss 25 abgeschlossen, das stÏndig mit der Aul3enatmosphäre in Verbin- dung steht, also als einziger Teil der Vorrichtung nicht unter Vakuum gesetzt zu werden braucht.
Aus diesem Zwischengefϯ 25 flie¯t unter dem Druck der Aussenatmosphäre bzw. dem Vakuum im Extraktionsgefäss 1, das an dieses Extraktionsgefäss über die Leitung 8 angelegt wird und in ihm auch dann noeh herrscht, wenn der Schwimmerventilkörper 4 die Verbindung zwischen Extraktionsgefäss l und Saugleitung 8 geschlossen hat, regeneriertes Extraktionsmittel über die Schwanenhalsroh r- ve. rbindung 7 und das Kapillarrohr 6 ein, wobei dieses Vakuum im Extraktionsgefϯ I bzw. die Druckdifferenz zwischen dem Zwisehengefäss 25 und dem Extraktionsgefϯ 1 durch zusätzliche VIittel 2, 26, 28 stetig in geringem Grade vermindert wird.
Durch das Kapillarrohr 28 strömt nämlich Luft. in das Extraktionsgefäss 1 ein, wobei die Kapillaren 6 und 28 derart aufeinander abgestimmt sind. dass die Überführung der in dem Zwisehen- gefϯ 25 vorhandenen Extraktionsmittelmenge vor Abdampfen des Extraktionsmittels in dem
Destillationsgefϯ 9, das hei¯t vor Absehalte des Vaknums, beendet ist.
An Einzelheiten ist zu bemerken, dass das
Extraktionsgefäss zweckmässig'nur zu zwei
Drittel gefüllt wird. Das Destillationsgefäss 9 besitzt einen eingezogenen Boden mit sich dreimal verjüngender Form : dies soll dem
Umstand Rechnung tragen, da¯ im Destilla tionsgefass mit versehiedenen Flüssigkeits- mengen eingedampft wird. Je kleiner die Venge ist, um so tiefer wird der Flüssigkeitsspiegel liegen. Damit wird aber auch der Durchmesser der Spiegeloberfläehe kleiner, so da¯ praktisch keine vollkommene Austroek- nung stattfinden kann. I) ies ist deshalb von Bedeutung, weil viele gegen Hitze empfindliche Substanzen ihre Wirkstoffe schon bei einmaliger Austrocknung verlieren k¯nnen.
Die Arbeitsweise der Vorrichtung zur Durchführung des Extraktions-und Vakuumverdampfungsverfahrens ist im Prinzip folgende :
Das Extraktionsgefäss wird, wie oben beschrieben, mit Substanz gefüllt und so viel Extraktionsflüssigkeit hinzugegeben, dass der Flüssigkeitsspiegel über der Substanz steht.
In das Zwischengefäss 25 wird ebenfalls Ex- traktionsfl ssigkeit bis etwa zur Hälfte ein- gefüllt. Auch in das Destillationsgefäss 9 wird etwas Extraktionsflüssigkeit eingefüllt. Es sei angenommen, dass die Dauer der jeweiligen Extraktion etwa 10 Minutez beträgt. Dann wird vor dieser Zeit die Vakuumpumpe ein- geschaltet, jedoch der Dreiweghahn 22 so eingestellt, da¯ die Vorrichtung nicht evakniert wird.
Zwischen der Vakuumpumpe und dem Dreiweghahn mu¯ sich, wie blich, ein entsprechend grosses Vakuumvorratsgefäss be- finden, damit die Evakuierung des Destilla tionsgefässes plötzlieh erfolgt.
Wenn nun der Dreiweghahn 22 so gestellt wird, dass das Vakuumvorratsgefäss mit der Vorrichtung direkt verbunden wird unter Ab sperrung des Lufteintrittsstutzens 23, so wird las GummiplÏttchen 20 an den Ventilsitz 19 gesaugtunddamitdasVorratsgefäss18ab- geschlossen.
Der Abschlul des Destillationsgefϯes 9 gegen das Extraktionsgefäss l erfolgt derge stalt, dass die Flüssigkeit und damit der Schwimmerventilkörper 4, welcher im Flüssig keitsspiegel im Extraktionsgefäss schwimmt, langsam nach unten gesaugt wird, so dass die gesamte Extraktionslösung aus dem Extraktionsgefäss in die Leitung 8 gesaugt wird. Sobald dies geschehen ist, schliesst der Schwimmerventilkörper den Ventilsitz dicht ab. Für ein gerades Auftreffen sorgt die aus einem feinen Glasstiel bestehende Führungsstange 27.
Durch das durch das Absaugen entstehende Vakuum im Extraktionsgefäss wird uber die Schwanenhalsrolirverbindung 7 aus dem Zwischengefäss 25 regenerierte Extrak tionsflüssigkeit in das Extraktionsgefäss 1 gesaugt. I) ies vollzieht sich langsam durch die kapillare Uffnung 6. Auf diese Weise kann keine regenerierte Extraktionsfliissigkeit mit dem Absaugen in das Destillationsgefäss 9 gelangen. Das Extraktionsgefäss 1 füllt sich mit der Menge Flüssigkeit wieder. die vorher abgesaugt wurde.
Nun kommt die Verdampfung im Destil lationsgefäss 9 in Gang und das Vorratsgefäss 18 füllt sich langsam. Die extrahierten Stoffe werden im Geläss 9 eingeengt. Während dieser Zeit wird die Substanz mit der regenerierten Extraktionsflüssigkeit im Extraktionsgefäss extrahiert.
Die Durchlassöffnungen der Schwanenhals- rohrverbindung 7 und der an diese angeschlossenen Kapillare 6 einerseits und der Kapillare 28, die das Extraktionsgefäss 1 in ständiger Verbindung mit der Aussenatmosphäre hilt, anderseits sind in ihren Abmessungen derart aufeinander abgestimmt, dass bei angelegtem Vakuum die Füllung des Extraktionsgefässes mit einer in dem Zwischengefϯ 25 vorhandenen Charge regenerierten Extraktionsmittels vor der Abdampfung des Extraktions- mittels in dem Destillationsgeiass 9, das heisst vor Abschaltung des Vakuums, beendet wird.
Nach je etwa 10 Minuten wird der Drei- weghahn 22 so gestellt, da¯ durch den Stut- zen 3 in die Vorrichtung ausschliesslich Liift einströmen kann. Dadurch fällt der Teller 20 des Ventils 19, 20 herunter und die im Vor ratsgefäss befindliche Flüssigkeit läuft in das Zwischengefϯ 25, der Schwimmerventilkörper 4 steigt in dem Siebring 5 wieder bis zum Flüssigkeitsspiegel. Sobald dies geschehen ist, verstellt man den Dreiweghahn 22 derart, dass unter Luftabschluss die Vorrichtung wieder evakuiert wird. Nun beginnt der gleiehe Prozess von neuem. Statt der Verstellung von Hand kann mit grossem Vorteil eine Verste lung durch eine periodisch funktionierende Betätigungsvorrichtung, die an sich bekannt ist, erfolgen.
Mit der Zeit sammeln sich im Destillationsgefäss 9 die extrahierten Stoffe an. Durch die sich immer wieder regenerierende Extrak tionslösung kann die Extraktion sehr weit getrieben werden.
Die Temperatur im Destillationsgefäss wird durch die Regelung der Gasflamme stets unterhalb der Temperatur gehalten, bei welcher die Wirkstoffe des jeweiligen Extraktes geschädigt würden. Die Temperatur im Extraktionsgefäss ist mit der Zimmertemperatur identisch.
Für den Fall, dass Extraktionsflüssigkeiten in Anwendung kommen, die bei gutem Vakuum durch den Kühler 21 hindurchgesaugt werden, empfiehlt sich die Anwendung einer K hlfalle mit entspreehendem Rüekfluss in das Vorratsgefäss an Stelle des Kühlers 21.
Die Kühlung dieser Kühlfalle muss mit einem den Verhältnissen angepassten Kühlmittel erfolgen.
Für alle Verbindungen zwischen den ein zelnen Glasteilen werden zweckmässig Gummi- schläuche verwendet. Soll die Vorrichtung maschinell bedient werden, so darf der Drei weghahn 22 nicht fest mit dem Kühler 21 ver bunden sein, sondern beide Teile müssen mit einem Stück Gummischlauch aneinander angeschlossen werden. Der Griff des Hahn kükens wird mit Hilfe eines lletallarmes ver- längert, an dessen Ende die Kraft'zum Verdrehen des Hahnes wirkt. Nach der gew nschten Dauer der Extraktion muB der Hahn um 90¯ gedreht werden, um nach einigen Sekunden wieder in die alte Stellung zuriiekgezogen zu werden.
Die praktische Ausführung einer solehen selbsttÏtigen Steuerung bietet keine Schwicrigkeiten, so dass eine nähere Erläu terung nicht erforderlich ist. Dadurch, dass das Abtreiben der Extraktionsflüssigkeit gesondert und in Abwesenheit des Extraktions- gutes vorgenommen wird, und zwar r unter sehonendsten Bedingungen, und dem Extrak tionsgut periodiseh regenerierte Extraktionsflüssigkeit wieder zugeführt wird und diese Perioden beliebig oft wiederholt werden k¯nnen, was z.
B. durch Zeitschaltung eines den Hahn bedienenden rotors möglieh ist, kann auch in solchen Fällen ohne Erwärmung extrahiert werden, in denen bei Benutzung der üblichen Extraktionsvorrichtungen die Anwendung von Wärme nicht zu umgehen ist.
PATENTANSPR¯CHE: I. Verfahren zur Extraktion und Vakuumverdampfung, dadurch gekennzeichnet, dass die Extraktion unter Vermeidung von ErwÏrmung durchgeführt wird, während die Regeneration der Extraktionsfl ssigkeit im De stilliergefäss unter Vakuum und unterhalb einer die Wirkstoffe des jeweiligen Extraktes schÏdigenden Temperatur durchgeführt und kondensierte Extraktionsflüssigkeit bei Raumtemperatur periodisch wiederholt dem Extraktionsgefäss zugeführt wird.

By regulating the gas flame, the temperature in the distillation vessel is always kept below the temperature at which the active ingredients of the respective extract would be damaged. The temperature in the extraction vessel is identical to the room temperature.
In the event that extraction liquids are used which are sucked through the cooler 21 with a good vacuum, the use of a cold trap with a corresponding return flow into the storage vessel is recommended instead of the cooler 21.
The cooling of this cold trap must take place with a coolant adapted to the conditions.
It is advisable to use rubber hoses for all connections between the individual glass parts. If the device is to be operated by machine, the three way valve 22 must not be firmly connected to the cooler 21, but both parts must be connected to one another with a piece of rubber hose. The handle of the stopcock is lengthened with the help of an all-out arm, at the end of which the force to turn the stopcock acts. After the desired duration of the extraction, the stopcock must be turned 90º and then pulled back into the old position after a few seconds.
The practical implementation of such an automatic control does not present any difficulties, so that a more detailed explanation is not necessary.
