PROCEDE POUR LA PREPARATION DE NOUVEAUX AGENTS TENSIOACTIFS
CATIONIQUES.
La présente invention a pour objet de nouveaux agents tensioâctifs, analogues aux substances dites "détergents cationiques" et ayant des proprié- tés bactéricides particulières, qui frappent aussi des types de germes'contre lesquels les détergents cationiques connus ont peu d'activité, et qui onbun pouvoir irritatif plus faible que ceux-ci.
En particulier, les nouveaux composés suivant la présente invention sont des sels des éthers du 2,4-diguanidine-phénol, de la formule (I)

Due caractéristique donnant un intérêt particulier aux substances du type (I) est la présence du groupe diguanidins-phénol-éthérique, avec les deux fonctions guanidiniques en position appropriée sur le noyau beznènbique.
Déjà les inventeurs, voir? entre autres, la communication faite au XXIIIe Congrès de 6himie Appliquée, Septembre 1950, et publiée ultérieurement (A. Di Marco, G.Boretti, C. Pasini, "11 Farmaco"6, 300 (1951)), avaient

trouvé que ledit groupe donnait auxdites substances'ayant R = méthyle, por- pyle, des propriétés antibactériques définies quoique faibles.
Subséquèmment, il a été trouvé, d'une façon surprenante, que dans la série des composés de formule (I) avec R ayant de 8 à 16 atomes de carbone (chine normale), ces propriétés sont extraordinairement développées, au point que les substances ainsi constituées peuvent égaler dans l'action et, par rapport à certains micro-organismes, même surpasser les plus actifs parmii les détergents catio- niques.
Si l'on compare, par exemple, le chlorure de cétyl-pyridine, qui est un des plus actifs des détergents cationiques,connus (et -qui, comme les nouveaux agents,suivant l'invention, mais contrairement aux autres, présente une action constante, même en cas de variation du pH du milieu), avec un re- présentant de la nouvelle série de composés, par exemple celui avec,R avant 12 atomes de carbone (di-chlorhydrate de l'éther laurylique du 2,4-diguani- dinephénol) on trouve que, tandis que l'activité bactéricide sur les germes Gram-positifs et sur les germes résistants- aux acides, est à peu près égale chez les deux, elle est considérablement plus grande chez le deuxième sur les germes Gram-négatifs.
Même pour ce qui concerne le pouvoir irritatif - soit comme degré, soit comme persistance du phénomène d'irritation locale - la comparaison en- tre le chlorure de cétyl-pyridine et le di-chlorhydrate de l'éther lauryli- que du 2,4-diguanidine-phénol, est nettement favorable au deuxième.
Le fait que les solutions des substances suivant l'invention ont moins de pouvoir irritant est probablement dû aussi à leur pH proche de la neutralité.
Les substances formant l'objet de la présente invention n'avaient jamais été préparées auparavant.
Forme aussi l'objet de la présente invention le procédé pour ob- tenir les substances correspondant à la formule (I) avec R-;-- alcoyle; il est essentiellement caractérisé par le fait que l'on fait réagir avec la cyanamide les éthers du 2,4-diaminophénol, soit en partant des bases libres, suivant le schéma de réaction :

II a été trouvé qu'avec les sels des 2,4-diaminophényl-éthers, la cyanamide réagit en deux stades, selon le schéma suivante donné à titre d'exemple, pour les di-chlorhydrates :

I1 a été trouvé que lorsqu'à la solution chaude des 2,4-diamino- phényl-éthers on ajoute la cyanamide, même-en excès, il se produit entre 10-15 minutes un dégagement considérable de chaleur (à ce point s'est formé le premier groupe guanidimique,qui est celui en position ortho par rapport à l'OR) et que pourrobtenir le produit désiré, il faut prolonger le temps d'ébullition.
Pour les premiers termes de la série (R = méthyle, éthyle, propyle, butyle), l'influence de la chaîne latérale est très faible ; -il- reste essen- tiellement des sels ayant en particulier la propriété de bien se dissoudre dans l'alcool éthylique. Pour eux, la méthode de préparation est -pratique- ment la suivante : réaction dans l'alcool éthylique, concentration (éventuel- le) et précipitation par addition d'un solvant apolaire (éther).. La -présence d'eau en petites quantités est nécessaire (hormis pour le méthyl-dérivé) puisqu'ils en ont tous 'besoin pour construire leur édifice cristallin.
Lorsque, au contraire, la chaîne latérale est allongée de 8 à 16 atomes de C, les substances en question acquièrent, pour ainsi dire, des pro- priétés intermédiaires entre les sels et les paraffines.. On a troqué, en effet, que pour les produits à chaîne latérale avec 8 à 16 atomes:de 0, ;si l'on effectue la réaction dans un solvant seulement alcoolique, la précipi- tation subséquente avec un solvant apolaire ne donne pas de résultat 'satis- faisant.
D'autre part, la séparation par -précipitation est importante parce qu'elle permet d'éliminer aisément les produits .secondaires de 'la réaction.
On a trouvé que l'on obtient, au contraire, de bons rendements en 'employait comme milieu de réaction un mélange de solvants constitué -en ordre principal par un solvant apolaire avec addition de petites quantités de solvants polai- res, en particulier par l'acétate d'éthyle additionné de, petites quantités d'eau et d'alcool métbylique, et ayant un point d'ébullition compris entre environ 50 c et-environ 150 C pour pouvoir appliquer un temps de réaction compris entre des limites utiles. De cette façon, on a, en outre, l'avantage d'obtenir; directement du liquide de réaction, la séparation d'un produit avec des caractéristiques satisfaisantes et avec de bons rendements.
'Quelques exemples de réalisation seront décrits ci-après à titre illustratif et nbn limitatif.
EXEMPLE i.
10 g de di-chlorhydrate de l'éther n-propylique du 2,4-diaminophénol sont dissous à chaud dans 100 cc d'alcool. éthylique à 96%. On ajoute 9 g de cyanamide anhydre et on maintient sous reflux pendant environ 5 heures; on traite à chaud avec du charbon décolorant et l'on filtre. On concentre

à moitié @ volume, on ajoute 2 ce d'eau et on dilue avec précaution? en agitant, avec de l'éther éthylique jusqu'à un commencement dé bouchement.
Par frottement des parois du matras commence la séparation du dichlorhydrate de l'éther n-propylique du 2,4-diguanidine-phénol, qui est blanc et qui est filtré et lavé avec un peu d'acétone. L'analyse montre qu'il possède 1 mol. d'eau de cristallisation. Il n'a pas de point de fusion net, mais commence à se concrétionner à environ 190 C et se décompose au-dessus de 220 C.
On peut l'obtenir dans une forme différente - qui abandonne l'eau de cristallisation à 130-135 C, simulant une fusion avec décomposition - en chauffant à reflux pendant 30 minutes en solution aqueuse à 1%,'en concentrant rapidement à 13-15 mm. Hg de pression et à 30-35 C presque jusqu'à se, en reprenant avec un peu d'alcool et en précipitant avec de l'éther.
EXEMPLE 2. la g de dichlorhydrate de l'éther n-butylique du 2,4-diaminophé- nol sont dissous à chaud dans 100 ce d'alcool éthylique à 96%. On ajoute 8 g de cyanamide anhydre et on maintient à reflux pendant environ 5 heures; on traite avec du charbon décolorant et on filtre. On ajoute 2 ce d'eau et on dilue avec précaution, en agitant, avec de l'éther éthylique jusqu'au commencement du bouchement. Par frottement des parois du, matras commence la séparation du dichlorhydrate de l'éther n-butylique du 2,4-diguanidine- phénol, blanc, qui est filtré et lavé avec un peu d'acétone.
L'analyse montre qu'il possède 1 mol. d'eau de cristallisation.
11 n'a pas de point du fusion net, il commence à se concrétionner à plus de 170 C et il se décompose au-dessus de 230 C.
EXEMPLE 3.
10 g de dichlorhydrate de l'éther diamino-phéhyl-laurylique sont chauffés sur bain-marié à reflux avec 100 ce d'acétate d'éthyle, 2,5 ce d'eau et 5 ce d'alcool méthylique; au commencement de l'ébullition on ajoute 5 g de cyanamide anhydre et on continue à reflux pendant 2 heures. Au liquide encore chaud on ajoute encore 5 ce d'alcool méthylique et on laisse refroidir ; après quelque temps, commence la séparation du produit qui est achevée en laissant ensuite reposer pendant une nuit,à O C. On filtre sur Büchner et on lave avec 5 cc d'acétone. On recristallise depuis de l'eau avec un peu d'alcool, après traitement préalable avec du charbon décolorant.
Le dichlo- rhydrate de l'éther laurylique du 2,4-diguanidine-phénol ainsi obtenu possède une molécule d'eau de cristallisation, n'a pas de point de fusion net, mais commence à se concrétionner à environ 135 C; la concrétion se complétant à environ 210 C, puis il brunit et se décompose à environ 250 C.
EXEMPLE 4.
10 g de dichlorhydrate de l'éther 2,4-di-aminophényl-cétylique sont chauffés à reflux avec 80 ce d'acétate d'éthyle, 2,5 ce d'eau et 4 cc d'alcool méthylique ; au commencement de l'ébullition on ajoute 4 g de cyana- mide anhydre et on continue à reflux pendant deux heures. Au liquide encore chaud on ajoute encore 12 ce d'alcool méthylique et on laisse refroidir, avec quoi commence la séparation du produit qui est achevée en laissant à 0 C pen- dant une nuit. On filtre sur B#chner, on lave avec 5 ce d'acétone et on cris- tallise à partir d'alcool méthylique-eau, après un traitement préalable avec du charbon décolorant.
Le dichlorhydrate de l'éther cétylique du 2,4-digua- nidine-phénol ainsi obtenu possède une molécule d'eau de cristallisation, il n'a pas de point de fusion net, mais se concrétionne entre 80 et 100 et se décompose à environ 275 C; après un lent brunissement préalable.

It has been found that when cyanamide, even in excess, is added to the hot solution of 2,4-diaminophenyl-ethers, a considerable evolution of heat takes place between 10-15 minutes (at this point s' the first guanidimic group is formed, which is the one in position ortho to the OR) and that to obtain the desired product, the boiling time must be prolonged.
For the first terms of the series (R = methyl, ethyl, propyl, butyl), the influence of the side chain is very weak; There remain essentially salts having in particular the property of dissolving well in ethyl alcohol. For them, the preparation method is -practically the following: reaction in ethyl alcohol, concentration (if any) and precipitation by addition of an apolar solvent (ether). The -presence of water in small quantities is necessary (except for the methyl derivative) since they all need it to build their crystalline edifice.
When, on the contrary, the side chain is lengthened by 8 to 16 C atoms, the substances in question acquire, so to speak, properties intermediate between salts and paraffins. It has been exchanged, in fact, for side chain products with 8 to 16 atoms: 0. If the reaction is carried out in an alcoholic solvent only, the subsequent precipitation with an apolar solvent does not give a satisfactory result.
On the other hand, the precipitation separation is important because it allows easy removal of the side products of the reaction.
On the contrary, it has been found that good yields are obtained by employing as reaction medium a mixture of solvents consisting mainly of an apolar solvent with the addition of small quantities of polar solvents, in particular of the solvent. Ethyl acetate with the addition of small amounts of water and metbyl alcohol, and having a boiling point of between about 50 ° C and -about 150 ° C to be able to apply a reaction time between useful limits. Add 2 cc of water and dilute carefully, with stirring, with ethyl ether until the start of plugging. By rubbing the walls of the matras begins the separation of the dihydrochloride of n-butyl ether from 2,4-diguanidine-phenol, white, which is filtered and washed with a little acetone.
Analysis shows that it has 1 mol. water of crystallization.

EXAMPLE 3.
10 g of diamino-phéhyl-lauryl ether dihydrochloride are heated on a reflux water bath with 100 cc of ethyl acetate, 2.5 cc of water and 5 cc of methyl alcohol; at the start of the boiling, 5 g of anhydrous cyanamide are added and the mixture is continued at reflux for 2 hours. To the still hot liquid is added another 5 cc of methyl alcohol and allowed to cool; after some time, the separation of the product begins which is completed by then allowing to stand overnight at 0 C. It is filtered through Buchner and washed with 5 cc of acetone. It is recrystallized from water with a little alcohol, after pretreatment with decolorizing charcoal.
The 2,4-diguanidine-phenol lauryl ether dichlorhydrate thus obtained has one molecule of water of crystallization, has no sharp melting point, but begins to solidify at about 135 C; the concretion completing at around 210 C, then it turns brown and decomposes at around 250 C.
EXAMPLE 4.
10 g of 2,4-di-aminophenyl-cetyl ether dihydrochloride are heated to reflux with 80 cc of ethyl acetate, 2.5 cc of water and 4 cc of methyl alcohol; at the start of boiling, 4 g of anhydrous cyanamide are added and the mixture is continued at reflux for two hours. To the still hot liquid, another 12 cc of methyl alcohol is added and the mixture is allowed to cool, with which the separation of the product begins, which is completed by leaving at 0 ° C. overnight. It is filtered through B # chner, washed with 5 cc of acetone and crystallized from methyl alcohol-water, after pretreatment with decolorizing charcoal.
The 2,4-diguanidine-phenol cetyl ether dihydrochloride thus obtained has one molecule of water of crystallization, it has no clear melting point, but solidifies between 80 and 100 and decomposes at about 275 C; after a slow preliminary browning.
