Abstract

The invention discloses an edible fungus culture medium based on disposable bamboo chopsticks and preparation technology and application thereof, it relates to the field of culture medium preparation. The culture medium comprises the following components: 20-30 parts of peat soil, 35-45 parts of sawdust, 7-9 parts of bran, 1-2 parts of gypsum powder, 1-2 parts of lime powder, 0.1-0.6 part of vitamin B, 1-2 parts of calcium superphosphate and 40-55 parts of water; The ratio of auxiliary materials to bamboo chopsticks is 1 part of auxiliary materials contains 120-140 bamboo chopsticks. The preparation technology of that culture medium is simple, and when apply it to the cultivation of edible fungi, it can improve quality and production efficiency of the strains, shortening the production cycle of the strains remarkably and improving the economic benefit greatly. Thus, the culture medium of the present invention has a good potential of popularization and application.

TECHNICAL FIELD
The invention relates to the technical field of strain cultivation, in particular to an edible fungus culture medium based on disposable bamboo chopsticks and a preparation method and application thereof.
BACKGROUND
Edible fungi production is pollution-free, pure natural, recyclable, renewable and environmentally friendly. Edible fungi has become the sixth largest agricultural product after grain, cotton, oil, fruit and vegetables in the traditional agricultural production field; In the process of agricultural modernization, edible fungi industry has become the vanguard and main force of modern agricultural development; Hou Zhengxiang of the Development Research Center of the State Council said that edible fungi as a sunrise industry will soon surpass China's grain, cotton and oil industry as well as China's automobile industry.
In the new century, the production technology of edible fungi in China is developing with each passing days, and having breakthroughs in many technical fields, which let China become a well-developed market to edible fungi with powerful demand, however, due to the long production cycle, such demand is difficult to meet.
At the same time, disposable bamboo chopsticks are widely used in China, and most of them become waste, causing pollution and waste. The main component of bamboo chopsticks is cellulose, which is a suitable nutrient medium for the growth of edible fungi and can be used as a carrier for cultivating edible fungi.
Therefore, it brings a great practical significance to fungi cultivation by providing an edible fungus culture medium based on disposable bamboo chopsticks that can shorten its production cycle.
SUMMARY
The purpose of the invention is to provide an edible fungus culture medium based on disposable bamboo chopsticks and a preparation method and application thereof, so as to solve the problems existing in the prior art.
The invention provides an edible fungus culture medium based on disposable bamboo chopsticks, which comprises auxiliary materials and bamboo chopsticks, wherein the auxiliary materials comprise the following raw materials in parts by mass: 20-30 parts of peat soil, 35-45 parts of sawdust, 7-9 parts of bran, 1-2 parts of gypsum powder, 1-2 parts of lime powder, 0.1-0.6 part of vitamin B and 1-2 parts of calcium superphosphate. Among them, the ratio of auxiliary materials to bamboo chopsticks is that one part of auxiliary materials contains 120-140 bamboo chopsticks.
The invention also provides a preparation method of the edible fungus culture medium based on disposable bamboo chopsticks, which comprises the following steps:
(1) The bamboo chopsticks are cleaned to absorb water, and then the bamboo chopsticks are roughened to make their surface rough;
(2) Stirring peat soil, sawdust, bran, gypsum powder, lime powder, vitamin B, calcium superphosphate and water uniformly to prepare culture medium auxiliary materials;
(3) Arranging the bamboo chopsticks processed in the step (1) in a culture medium auxiliary material, and covering gaps and surfaces of the bamboo chopsticks with the culture medium auxiliary material to obtain a preliminarily processed culture medium;
(4) Putting the culture medium preliminarily processed in step (3) into a steam sterilization pot for sterilization, and cooling it to obtain the edible fungus culture medium based on disposable bamboo chopsticks.
Further, in Step(4), the condition of the sterilization is 0.103MPa and 121°C for 40-50 minutes.
The invention also provides the application of the edible fungus culture medium based on disposable bamboo chopsticks in edible fungus strain culture.
Further, breaking the fermented edible fungus liquid hyphae balls, diluting it by 300-500 times sterile water to obtain liquid strains, and inoculating the liquid strains on the edible fungus culture medium based on disposable bamboo chopsticks according to the inoculation amount of 20-25ml, and culturing the culture medium at 26°C for 3-10 days.
The invention discloses the following technical effects:
In the technical research and production application of edible fungi, the general sterilization conditions of solid substrates (bamboo chopsticks, etc.) are 0.138MPa, 126°C, and sterilization is executed for about 120 minutes, which is time-consuming and energy-consuming, and causes the loss of nutrient components of bamboo chopsticks substrate. However, the sterilization parameters of the present invention not only achieves a good sterilization effect, but also lowers the energy waste of sterilization and reduces the loss of nutrient components of bamboo chopsticks substrate. The liquid hyphae can quickly spread on the surface of bamboo chopsticks and grow inward, making the growth of edible fungi hyphae to be in a dominant state, hence inhibiting the growth of miscellaneous bacteria. Therefore, by reducing sterilization parameters properly will not affect the normal growth of edible fungi hyphae on the surface of bamboo chopsticks, On the other hand, due to the sterilization parameters of the invention, the treatment temperature is reduced and the treatment time is shortened, thereby reducing the nutrient components loss and lower the energy use.
The inoculation amount directly affects the germination and growth effect of hyphae, if the amount is small, the hyphae grow slowly, and the long growth cycle can easily cause the pollution of miscellaneous bacteria. If the amount of inoculation is large, it causes waste and compels a high requirement for sanitary environment, refrigeration and ventilation equipment, which does not suit the factory with weak technical force and insufficient technical personnel, and it especially does not suit individual farmers. More importantly, when the amount of inoculation is large, it will form a layer of bacterial mucosa on the surface of bamboo chopsticks that blocks the entry of fresh air, causing growth failure of internal hyphae, ultimately leading to the pollution of miscellaneous bacteria and increasing the use risk of liquid strains. In this invention, the inoculation amount is adjusted. The fermented edible fungus liquid hypha balls is broken and diluted by 300-500 times with sterile water to obtain liquid strains, then inoculated the strains on the prepared culture medium according to the inoculation amount of 20-25m, by which it can reduce the waste of resources and ensure the germination effect, meanwhile inhibit a layer of bacterial mucosa on the surface of bamboo chopsticks that blocks the entry of fresh air, guaranteeing the oxygen supply for hyphae to grow properly.
Peat soil and disposable bamboo chopsticks are also used in the invention, and peat soil stems from many years of naturally fermentation, it has good maturity and strong water holding capacity and suitable fertility, besides it is abundant in China with low purchasing price. Peat soil can improve the fertility and lower the cost of culture medium. The main component of bamboo chopsticks is cellulose, which is a suitable nutrient substrate for the growth of edible fungi. The invention also reduces the environmental pollution caused by disposable bamboo chopsticks.
The culture medium of the invention is also added with vitamin B, which can promote the good development of edible fungi hypha; The culture medium provided by the invention can improve the quality and production efficiency of strains, shorten the period of strain production, and greatly improve the economic benefit, overall, it has a great potential of popularization and application.

(2) Stirring peat soil, sawdust, bran, gypsum powder, lime powder, vitamin B, calcium superphosphate and water uniformly to prepare culture medium auxiliary materials;
(3) Arranging the bamboo chopsticks processed in the step (1) in a culture medium auxiliary material, covering gaps and surfaces of the bamboo chopsticks with the culture medium auxiliary material to obtain a preliminarily processed culture medium;
(4) Putting the culture medium preliminarily treated in the step (3) into a steam sterilization pot for sterilization at the temperature of 0.103MPa and 121°C for 40 minutes, and cooling it to obtain the edible fungus culture medium based on disposable bamboo chopsticks.
(5) Breaking fermented edible fungus liquid hypha ball, diluting them by 300 times sterile water to obtain liquid strains, inoculating liquid hypha ofpleurotus eryngii on the edible fungus culture medium based on disposable bamboo chopsticks according to the inoculation amount of 20ml, and cultured under condition of 26°C for 3 days, making the surface of the bamboo chopsticks covered with hyphae, and keep culturing it for 5 days, so that the interior of the bamboo chopsticks is covered with hyphae.
The hyphae cultivated is thick and dense, and the production cycle of strain was 23 days.
Embodiment 2
The edible fungus culture medium in this embodiment comprises auxiliary materials and bamboo chopsticks, wherein the auxiliary materials comprise the following components in parts by mass:
parts of peat soil, 45 parts of sawdust, 9 parts of bran, 2 parts of gypsum powder, 2 parts of lime powder, 0.1 part of vitamin b, 1 part of calcium superphosphate and 40 parts of water; the ratio of auxiliary material to bamboo chopsticks is 1:120
Preparation and inoculation of edible fungi culture medium;

(5) Breaking fermented edible fungus liquid hypha ball, diluting them by 400 times sterile water to obtain liquid strains, inoculating liquid hypha ofpleurotus eryngii on the edible fungus culture medium based on disposable bamboo chopsticks according to the inoculation amount of 22ml, and cultured under condition of 26°C for 4 days, making the surface of the bamboo chopsticks covered with hyphae, and keep culturing it for 4 days, so that the interior of the bamboo chopsticks is covered with hyphae.
The hyphae cultivated is thick and dense, and the production cycle of strain was 20 days.
Embodiment 3
The edible fungus culture medium in this embodiment comprises auxiliary materials and bamboo chopsticks, wherein the auxiliary materials comprise the following components in parts by mass:
parts of peat soil, 35 parts of sawdust, 7 parts of bran, 1 part of gypsum powder, 1 part of lime powder, 0.6 part of vitamin B, 2 parts of calcium superphosphate and 55 parts of water; the ratio of auxiliary material to bamboo chopsticks is 1:140
Preparation and inoculation of edible fungi culture medium;

(5) Breaking fermented edible fungus liquid hypha ball, diluting them by 500 times sterile water to obtain liquid strains, inoculating liquid hypha ofpleurotus eryngii on the edible fungus culture medium based on disposable bamboo chopsticks according to the inoculation amount of 25ml, and cultured under condition of 26°C for 5 days, making the surface of the bamboo chopsticks covered with hyphae, and keep culturing it for 3 days, so that the interior of the bamboo chopsticks is covered with hyphae.
The hyphae cultivated is thick and dense, and the production cycle of strain was 25 days.
Comparative Embodiment 1
Different from Example 1, if the sterilization conditions were 0.138MPa, 126°C and 120 minutes, the prepared hyphae are sparse and in poor state.
Comparative Embodiment 2
Different from Example 1, if the inoculation amount was 30ml, the prepared hyphae are sparse and in poor state.

An edible fungus culture medium based on disposable bamboo chopsticks, which is characterized in that the edible fungus culture medium comprises auxiliary materials and bamboo chopsticks, wherein the auxiliary materials comprise the following raw materials in parts by mass: 20-30 parts of peat soil, 35-45 parts of sawdust, 7-9 parts of bran, 1-2 parts of gypsum powder, 1-2 parts of lime powder, 0.1-0.6 part of vitamin b, and 1-2 parts of vitamin b.Among them, the ratio of auxiliary materials to bamboo chopsticks is that one part of auxiliary materials contains 120-140 bamboo chopsticks.

5. According to either one of claims 1-2, the edible fungus culture medium based on disposable bamboo chopsticks is applied to the cultivation of edible fungus strains.

According to claim 5, the application of culture is characterized in breaking the fermented edible fungus liquid hyphae balls, diluting them by 300-500 times sterile water to obtain liquid strains, and inoculating the liquid strains on the edible fungus culture medium based on disposable bamboo chopsticks according to the inoculation amount of -25ml, and culturing the culture medium at 26°C for 3-10 days.
