Abstract

HRASV12와 같은 종양유전자(Oncogene)에 의해 유도된 노화(Oncogene Induced Senescence; OIS)는 악성 형질전환의 위험이 있는 세포의 성장 및 증식을 억제하여 악성 종양으로의 발전을 억제한다. 이러한 노화 관련 종양유전자(Oncogene)의 변이로 인한 노화 기능의 상실은 종양을 유발할 수 있다(Serrena, M, et al., Cell 88, 593-602 (1997).
종양 세포의 노화는 종양의 치료를 위한 다양한 항암요법, 예를 들어 화학요법 및 방사선요법의 주요한 기전으로 사용된다 (Wang, B., Kohli, J. & Demaria, M. Trends Cancer 6, 838-857 (2020)). 학계 및 임상에서 보고된 다양한 화학요법 및 방사선 요법은 치료 요법의 1차 반응으로 세포의 노화를 유도한다. 예를 들어 과활성 발암 신호, 방사선 요법, 전통적인 화학요법, CDC7 억제제, 및 텔로머라제 억제제와 같은 치료는 DNA의 손상 및 p53-p21 신호전달을 통한 노화를 유도한다. 이러한 p53-p21 신호 전달 매개 노화는 VO-OHpi와 같은 PTEN 억제제, nutlin 3, RG7112 또는 UBX0101와 같은 MDM2-p53 상호작용 교란제, 및 vorinostat와 같은 히스톤 데아세틸라아제 억제제와 같은 약물에 의해서 유발될 수 있다. 또한 Desitabine과 같은 DNA 메틸트렌스퍼라제 억제제, CDK 억제제, 오로라 키나아제 억제제(AURK) 및 PLK1 억제제는 세포 주기의 G2/M 진행을 차단하여 노화를 유도한다.
이러한 치료에 의해 유도된 노화(Therapy-Induced Senescence; TIS) 암세포는 SASP를 통해 인접 암세포를 어레스트하고, 약물전달을 위한 혈관 구조를 개선하며, 면역세포를 모집하는 등 다양한 기전을 통해 종양 미세 환경에 영향을 미침으로써, 종양세포의 성장 및 증식을 억제하는 것으로 알려져 있다(Faget, D. V. et al., Nat. Rev. Cancer 19, 439-453 (2019); Krizhanovsky, V. et al. Cell 134, 657-667 (2008); Ritschka, B. et al., Genes Dev. 31, 172-183 (2017); Chang, B. D. et al. Oncogene 18, 4808-4818 (1999)).
한편, 치료 유도된 노화(Therapy-Induced Senescence; TIS) 및 TIS 유래 SASP에 의한 초기 항종양 효과에도 불구하고, 장기적으로는 오히려 염증의 유발, 면역 억제 미세 환경의 조성, 혈관 신생 촉진, 인접 암세포의 전이와 같은 친종양성 특징을 나타낼 수 있는 것으로 보고된다(Coppe, J. P. et al. PLoS Biol. 6, 2853-2868 (2008); Yang, L., Discov. 3, 17049 (2017); Watanabe, S., et al., Cancer Sci. 108, 563-569 (2017)). 구체적으로, SASP 인자로 알려진 MMP는 혈관 신생, 암세포의 성장 및 침습을 촉진할 수 있으며(Open Biol. 11, 200360 (2021)), IL-6 및 IL-8은 만성 염증 미세환경을 형성하여 종양 형성 및 다양한 부작용을 유발할 수 있다.
이에, 치료 유도된 노화 세포를 선택적으로 사멸시킴으로써, 노화세포의 장기적인 부작용을 방지하는 치료 요법이 보고되었다. 원-투 펀치 요법(One-Two punch theraphy)이라고도 명명되는 본 방법은 최근 화학 요법 및 방사선 치료를 통해 종양 세포의 노화를 유도하여 초기 항 종양효과를 통한 1차 치료효과를 유도하고, 후속적으로 노화 세포의 선택적 사멸 요법(senolytic therapy)을 통해 노화 세포에 의한 종양의 재발 및 장기적인 부작용을 방지하는 새로운 치료요법이다(Nature Reviews Cancer volume 22, pages340-355 (2022)). 원-투 펀치 요법(One-Two punch theraphy)은 약물의 병용 투여를 기반으로 하지 않고 각 약물을 사용한 순차적 치료를 기반으로 하기 때문에, 약물의 조합으로 발생하는 조합 독성을 회피할 수 있으며, 따라서 기존에 보고된 다양한 노화 세포 사멸 약물(senolytic drug)을 사용할 수 있다는 장점이 있다.
현재, 종양의 치료를 위한 원-투 펀치 요법(One-Two punch theraphy)에 기존에 보고된 노화 세포 사멸 약물(senolytic drug)을 사용하려는 시도가 다양하게 연구되고 있다. 구체적으로, Navitoclax(ABT-263) 및 BCL-2, BCL-XL 및 BCL-W의 선택적 억제제가 노화 암세포를 포함한 광범위한 유형의 노화 세포를 효과적으로 제거할 수 있는 것으로 보고되었다(Aging Cell 15, 428-435 (2016); Nat. Med. 22, 78-83 (2016); US11331328B). 종양 노화 유도제인 리툭시맙과 Navitoclax의 병용 요법은 2상 연구에서, 높은 효과를 나타내었으며, 내약성이 우수한 것으로 나타났다(Leuk. Lymphoma 56, 2826-2833 (2015).).
원-투 펀치 요법의 뛰어난 효과에도 불구하고, 지금까지 보고된 종양의 치료에 사용될 수 있는 노화 세포 사멸 약물(senolytic drug)의 종류는 현저히 부족하다(Nature Reviews Cancer volume 22, pages340-355 (2022)). 특히, 현재 보고된 Navitoclax, ABT737, NAav-Gal등은 모두 BCL-2/XL/W 기반의 약리기전을 통한 노화 세포 사멸 약물이기 때문에, 치료 유도된 노화 세포 외의 다양한 노화세포에 무분별하게 작용하여 예상치 못한 부작용을 유발할 수 있다. 이에, 다양한 기전을 통한 노화 세포 사멸 약물의 개발, 치료 유도된 노화 종양 세포에 특이적인 바이오마커의 발굴 및 이에 기반한 치료 유도된 노화 세포 특이적 사멸 약물의 개발의 필요성이 대두되고 있다.
이러한 배경기술 아래에서, 본 발명자들은 치료 유도된 노화 종양 세포의 신규 바이오마커를 발굴하고 원-투 펀치 요법에 사용될 수 있는 신규 노화 세포 사멸 약물(senolytic drug)을 개발하기 위해 예의 노력한 결과 다양한 항 종양 요법에 의해 유도된 치료 유도된 노화 종양 세포에서 Pim-1 Kinase의 발현이 공통적으로 향상되어 치료 유도된 노화 종양 세포의 신규 바이오마커로 사용할 수 있음을 확인하였으며, 나아가 Pim-1 kinase 억제제를 처리하는 경우, 치료 유도된 노화 종양세포에 특이적인 사멸 효과를 나타내는 것을 확인하고, 본 발명을 완성하게 되었다.
발명의 요약
본 발명의 목적은 Pim-1 키나아제 억제제를 포함하는 노화 세포의 사멸용 조성물을 제공하는 데 있다.

도 1은 유방암 세포주(MCF7, ZR75-1, Hs578T, MDA-MB-231)에 아드리아마이신 100nM를 처리하여 치료 유도 노화(TIS)를 진행시킨 뒤, 각 암 세포주의 세포 크기 증가 및 SA-β-gal 염색 결과(1a), SA-β-gal 양성 세포의 비(1b, 왼쪽) 및 p21 mRNA 발현 양(1b, 오른쪽)을 나타낸 것이다.
도 2a는 치료 유도 노화(TIS) 후, MCF7 및 MDA-MB-231에서의 차세대 염기서열 분석 (NGS)를 통해 확인 한 mRNA 발현량 변화를 나타낸 벤다이어그램을 나타낸 것이다.
도 2b는 치료 유도 노화 후, 노화 전과 비교하여 MCF7과 MDA-MB-231 세포 주에서도 공통으로 통계적으로 의미있게 발현이 증가된 유전자 중, FDA 승인된 약물의 표적이 되는 유전자 28종을 선정하여 각각의 세포주에서 TIS 시 발현 변화를 나타낸 volcano plot이다.
도 2c는 RNAseq를 진행하지 않은 다른 유방암 세포주(BT474, Hs578T, T47D, HCC1937)의 치료 유도 노화 후, 앞서 선정된 28종 유전자의 발현 변화를 확인한 결과이다.
도 3a는 최종 선정된 유전자를 표적으로 하는 억제제(항체를 제외한 저분자 화합물)를 MCF7 대조군 세포와 TIS 세포에 농도 별로 처리 후, 대조군(Ctrl) 및 치료 유도 노화된 세포(TIS)에 대한 세포 분열 억제 및 사멸능을 CCK8 assay를 통하여 확인한 결과이다.
도 3b는 MCF7 세포주에서 PIM1 키나아제 억제제(PIM1/2 Kinase Inhibitor VI, Calbiochem, CAS 587852-28-6) 및 노화 세포 선택적 사멸 약물(Senolytic drug)으로 보고된 ABT-263를 처리하여 대조군에 비해 치료 유도 노화된 세포군에서 세포 생존이 유의하게 억제됨을 CCK8 assay를 통해 확인한 결과이다.
도 3c는 MCF7 세포주의 대조군과 치료 유도 노화된 세포군에 다양한 농도의 PIM1/2 Kinase Inhibitor VI (Calbiochem, CAS 587852-28-6)를 처리한 뒤 세포의 사멸 효과를 현미경으로 확인한 결과이다.
도 3d 및 3e는 MCF7 세포주(Ctrl 및 TIS)에 대해 다른 PIM1 키나아제 억제제인 SGI-1776(Selleckhem, Catalog No.S2198, CAS 1025065-69-3)을 처리한 뒤 세포의 사멸 효과를 CCK8 assay와 현미경으로 확인한 결과이다.
도 4는 유방암 세포를 이외에도 다양한 암 세포 주에 (MKN1, MKN74, SNU601: 위암, SNU478, SNU1196, SNU869: 담도암, Hct116, SW480: 대장암, BxPC3: 췌장암, HeLa S3: 자궁경부암, PC3: 전립선암, HSC4: 두경부암) 치료 유도 노화(TIS)를 진행시키고, PIM1 키나아제 단백질의 발현을 확인한 결과이다.
도 4a는 표기된 암세포주에서 아드리아마이신으로 치료 유도 노화를 진행하고 PIM1를 포함한 PIM 키나아제 패밀리 유전자의 mRNA 발현 변화를 확인한 결과이다.
도 4b는 동일한 조건에서 PIM1의 단백질의 양적 변화를 표기한 다양한 세포주에서 확인한 결과이다.
도 4c는 다양한 암 세포에 시스플라틴을 처리하여 치료 유도 노화(TIS)를 진행시키고, PIM1 키나아제 mRNA 발현 변화를 확인한 결과이다.
도 5a 및 5b는 각 암세포(HSC4: 두경부암, SNU601: 위암, SW480: 대장암)에서, PIM 1/2 inhibitor VI를 처리한 뒤 대조군(Ctrl) 및 치료 유도 노화된 세포(TIS)에 대한 사멸능을 현미경과 CCK8 assay로 확인한 결과이다.
도 6a는 MCF7 세포주에 Pim1 키나아제 유전자를 표적으로 하는 siRNA를 처리후 48시간 경과 뒤, PIM1 키나아제 단백질의 발현 변화를 나타낸 것이다.
도 6b 및 6c는 Pim1 키나아제 유전자를 표적으로 하는 siRNA를 해당 농도로 48시간 처리하여, PIM1 키나아제 단백질의 발현을 억제하는 경우에 대조군(Ctrl) 및 치료 유도 노화된 세포(TIS)에 대한 사멸능을 현미경과 CCK8 assay 를 통해 확인한 결과이다.
다른 식으로 정의되지 않는 한, 본 명세서에서 사용된 모든 기술적 및 과학적 용어들은 본 발명이 속하는 기술분야에서 숙련된 전문가에 의해서 통상적으로 이해되는 것과 동일한 의미를 갖는다. 일반적으로 본 명세서에서 사용된 명명법은 본 기술분야에서 잘 알려져 있고 통상적으로 사용되는 것이다.
종양 세포의 노화는 종양의 치료를 위한 다양한 항암요법, 예를 들어 화학요법 및 방사선요법의 주요한 기전이다. 노화된 세포의 SASP는 1차적으로 항종양 효과를 나타내는 것으로 보고되었으나, 장기적인 관점에서 노화 세포는 전종양성 효과를 나타내어, 종양의 형성, 재발 및 전이를 촉진하고 다양한 부작용을 나타낼 수 있다. 이러한 관점에서, 최근 종양의 노화 유도에 의한 1차 치료 이후, 노화 세포 사멸 약물(senolytic drug)을 투여하여, 치료 유도된 노화 세포(TIS cell)를 선택적으로 사멸시켜 장기적인 부작용을 예방하는 원-투 펀치 요법(One-Two punch theraphy)이 주목받고 있으나, 보고된 노화 세포 사멸 약물(senolytic drug)의 종류가 현저히 부족하다.
본 발명의 일 실시예에서는 치료 유도된 노화 세포(TIS cell)의 유전자 발현 프로파일을 분석하여 이를 기반으로 노화 세포 사멸 효과(senolytic effects)를 분석한 결과, 치료 유도된 노화 세포(TIS cell)에서 Pim-1 키나아제의 발현이 현저히 향상되고 Pim-1 키나아제의 활성 억제제 또는 유전자 발현을 억제한 결과 치료 유도된 노화 세포에 선택적으로 높은 독성을 나타내는 것을 확인하였다.
따라서, 본 발명은 일 관점에서, Pim-1 키나아제 억제제를 포함하는 노화 세포의 사멸용 조성물에 관한 것이다.
본 발명의 용어, “Pim-1 키나아제(Pim-1 kinase)”는 PIM-1 키나아제 유전자에 의해 암호화된 세린/트레오닌 단백질 키나아제이다. Pim-1 키나아제는 MYC 전사활성 조절 세포 주기 진행, 세포 자멸사 및 전사활성화 등의 경로에 관여하는 것으로 알려져 있으다. Pim-1 키나아제는 종양유전자(oncogene)로도 잘 알려져 있다(The International Journal of Biochemistry & Cell Biology. 37 (4): 726-30).
본 발명의 용어, “Pim-1 키나아제 억제제(Pim-1 kinase inhibitor)”는 Pim-1 키나아제 유전자 발현의 억제 또는 Pim-1 키나아제 단백질의 활성을 억제할 수 있는 모든 화학적 또는 생물학적 제제를 포함하는 의미로 사용된다.
본 발명에 있어서, 상기 Pim-1 키나아제 억제제는 Pim-1 키나아제 유전자 발현 억제제, 또는 PIM1 키나아제 단백질 활성 억제제인 것을 특징으로 할 수 있다.
본 발명의 용어 "유전자 발현 억제제"는 표적 유전자가 전사 및 번역을 통해 발현되는 것을 억제 또는 저해하는 모든 제제를 의미한다. 표적 유전자 특이적으로 발현을 억제하는 모든 유형의 제제가 포함될 수 있으며, 전사단계의 조절 및 RNAi와 같은 전사 후 번역 조절에 의한 억제제를 모두 포함한다. 상기 Pim-1 키나아제 유전자 발현 억제제는 예를 들어, PIM-1 키나아제 유전자를 표적으로 하는 안티센스 뉴클레오타이드(antisense nucleotide), siRNA(small interfering RNA), shRNA(small hairpin RNA), 리보자임(ribozyme), 압타머(aptamer), anti-microRNA, MicroRNA mimic, Zinc Finger Nucleases(ZFNs) 및 Transcription activator-like effector nucleases (TALENs)와 같은 뉴클라아제, CRISPR-Cas9 시스템 제제(예, gRNA, sgRNA), polycistronic tRNA-gRNA 시스템 제제, 이용한 self-ribozyme-flanked RNAs 등이 있으나(Gao & Zhao 2014, Xie et al. 2015, Zetsche et al. 2017), 이에 제한 되는 것은 아니다.
본 발명에 있어서, 상기 Pim-1 키나아제 유전자 발현 억제제는 바람직하게는 Pim-1 키나아제 유전자를 표적으로 하는 siRNA인 것을 특징으로 할 수 있으며, 보다 바람직하게는 상기 siRNA는 서열번호 1로 표시되는 서열을 포함할 수 있으나, 이에 제한되는 것은 아니다.
상기 안티센스 뉴클레오타이드는 DNA, 미성숙-mRNA 또는 성숙된 mRNA의 상보적 염기서열에 결합(혼성화)하여 DNA에서 단백질로서 유전정보의 흐름을 방해하는 것이다. 상기 siRNA는 안티센스와 작동 원리는 유사하나 21~25개의 뉴클레오타이드가 연결된 두 가닥(double strand)으로, RNase H가 아닌 RISCs(RNA-induced silencing complexes)라는 효소복합체를 통해 mRNA를 파괴한다. 상기 CRISPR-Cas9(크리스퍼 유전자 가위)은 특정 염기서열을 인지해 해당 부위의 DNA를 절단하는 제한효소로 작용한다.
본 발명의 용어 "단백질 활성 억제제"는 표적 단백질의 활성을 억제 또는 저해하는 모든 제제를 의미한다. 상기 단백질 활성 억제제는 표적단백질의 활성을 직접적으로 억제하거나, 다른 단백질과의 상호작용을 방해하여, 이의 기능을 억제하고, 하위 신호 전달 경로를 차단하는 것을 특징으로 할 수 있다. 상기 Pim-1 키나아제 단백질 활성 억제제는 예를 들어, Pim-1 키나아제 단백질에 특이적으로 결합하는 화합물, 펩티드, 펩티드 모방체, 기질 유사체, 압타머 및 항체 등일 수 있으나, 이에 제한되는 것은 아니다.
본 발명에 있어서, 상기 Pim-1 키나아제 억제제는 당업계에 공지된 다양한 Pim-1 키나아제 억제제로부터 제한 없이 선택되어 사용될 수 있다.
본 발명에 있어서, Pim-1 키나아제 억제제로서 상기 화합물은 표적 단백질인 Pim-1 키나아제의 활성을 억제할 수 있는 임의의 화합물을 모두 포함한다. 예를 들어, Pim-1 키나아제 억제제로 플라보노이드 억제제, isoxazolo[3,4-b]quinoline-3,4(1H,9H)-dione, ETP-45299, 이미다조피리다진(imidazopyridazine) 기반의 화합물(SGI-1776), 티아졸리다인다이온(Thiazolidinedione) 기반의 화합물(AZD1208), 플루오로피콜린아마이드(Fluoropicolinamidine) 기반의 화합물(PIM447), PIM1/2 Kinase Inhibitor VI 등이 있으나, 이에 제한되는 것은 아니다.
본 발명에 있어서, 상기 앱타머(aptamer)는 단일 사슬 DNA 또는 RNA 분자로서, SELEX(systematic evolution of ligands by exponential enrichment)라 불리는 올리고 뉴클레오티드 (oligonucleotide) 라이브러리를 이용한 진화적인 방법에 의해 특정 화학 분자나 생물학적 분자에 높은 친화력과 선별력을 갖고 결합하는 올리고머를 분리하여 수득할 수 있다. 앱타머는 표적 단백질에 특이적으로 결합하고 표적 단백질의 활성을 조정할 수 있는데, 예컨대, 결합을 통하여 표적 단백질의 활성을 차단할 수 있다. 상기 항체는 하나 이상의 상보성 결정 영역을 포함하는 항체 단백질 분자의 집합, 하나의 항체 단백질 분자, 결합 단편 또는 이의 유도체를 의미한다.
본 발명에 있어서, 상기 "억제제"는 "길항제(antagonist)" 또는 "저해제"와 상호호환적으로 사용될 수 있다.
본 발명의 용어 "표적 단백질", "표적 유전자", "표적으로 하는" 등에서 사용되는 "표적"은 활성을 조절하고자 하는 대상을 의미하며, 특히 본 발명에서, 억제제는 표적으로 하는 대상 유전자의 발현 또는 활성을 조절할 수 있는 것을 특징으로 한다. 본 발명에 있어서, 표적 단백질은 Pim-1 키나아제이다.
본 발명의 용어, “노화(Senescence)”는 활발하게 분열하는 세포에서, 대사적으로 활성화된 비분열 세포로의 진행을 의미한다. 본 발명의 용어 “노화 세포”는 노화 과정에 있는 중간 노화 세포 및 완전히 분열이 비활성화된 완전 노화 세포를 모두 포함하는 개념으로 사용된다.
본 발명에 있어서, 상기 노화 세포는 다음과 같은 특징을 나타내는 것을 특징으로 할 수 있으나, 이에 제한되는 것은 아니다:
i) 성장의 억제;
ii) 생리학적 유사분열제(physiological mitogens)의 처리에도 세포 주기로의 재진입 억제;
iii) 세포 사멸에 대한 내성; 및/또는
iv) 차별화된 기능의 획득.
본 발명에 있어서, 노화는 다음과 같이 비제한적으로 분류될 수 있다.
“복제 세포 노화(replicative cellular senescence)”는 텔로미어의 열화로 인한 분화 세포의 세포 분열 횟수의 제한을 의미한다.
“조기 세포 노화 (premature cellular senescence)"는 텔로미어의 열화 없이 자외선, 활성산소종, 화학요법, 환경 독소, 이온화 방사선, 흡연, 종양유전자 돌연변이(Oncogenic mutation) 등의 다양한 스트레스에 의한 세포 주기의 정지를 의미한다. “종양유전자 유도된 노화(Oncogene induced senescence)는 조기 세포 노화의 한 예로서, 종양유전자의 발현 향상 또는 변이에 의한 세포 노화를 의미한다.
“치료 유도된 노화(Therapy induced senescence)는 특정 질병의 치료를 위해 투여된 치료제에 의해 세포가 노화 상태로 강제되는 것을 의미한다. 일반적으로 치료 유도된 노화는 종양의 치료를 위한 항암제의 투여 또는 항암 요법으로 인해 발생된다. 종양학적 관점에서, 상기 치료 유도된 노화는 일반적으로 다양한 화학 요법, 생물학적 요법, 및 방사선 요법 등과 같은 항암 치료에 의해 발생하는 것으로 보고된다(Nature Reviews Cancer volume 22, pages340-355 (2022)).
본 발명에 있어서, 상기 노화 세포는 성장 및/또는 분열이 중단된 상태의 세포인 것을 특징으로 할 수 있다. 본 발명에 있어서, 상기 노화 세포는 대사 활성이 유지되는 것을 특징으로 할 수 있다. 본 발명에 있어서 상기 노화 세포는 영구적으로 성장 및/또는 분열이 중단된 세포인 것을 특징으로 할 수 있으나, 이에 제한되는 것은 아니며, 외부적인 요인에 의해 다시 분열 주기에 재진입할 수도 있다. 본 발명에 있어서, 상기 노화 세포는 복제 세포 노화, 조기 세포 노화, 또는 치료 유도 노화에 의해 노화된 것을 특징으로 할 수 있다.
본 발명에 있어서, 상기 노화세포는 혈관계, 조혈 시스템, 상피기관 및 기질을 포함하는 재생 가능한 조직에 존재하는 것 또는 이로부터 분리된 것을 특징으로 할 수 있다.
본 발명에 있어서, 상기 노화세포는 노화 관련 병변 부위에 존재하는 것 또는 이로부터 분리된 것을 특징으로 할 수 있다. 예를 들어 노화 관련 병변 부위는, 만성 노화 관련 질환, 염증성 질환, 골관절염, 동맥경화증, 면역 질환, 신생물성 질환, 및 전종양 또는 종양 조직 등이 있으나 이에 한정되는 것은 아니다.
본 발명의 일 실시예에서는 암 세포에 아드리아마이신을 처리하여 치료 유도된 노화를 유도하였으며, SA-b-gal 염색 및 p21 mRNA 발현 수준을 통해 노화세포를 분리하였다. 따라서, 본 발명에 있어서, 상기 노화 세포는 노화 암 세포 인 것을 특징으로 할 수 있다.
본 발명에 있어서, 상기 암은 예를 들어, 유방암, 위암, 대장암, 두경부암, 자궁경부암, 담도암, 폐암 또는 췌장암인 것을 특징으로 할 수 있으나, 이에 제한되는 것은 아니다.
본 발명에 있어서, 상기 노화 세포는 바람직하게는 종양유전자 유도된 노화 세포(Oncogene-induced senescence cell; OIS cell) 또는 치료 유도된 노화 세포(Therapy induced senescence cell; TIS cell)인 것을 특징으로 할 수 있으며, 보다 바람직하게는 치료 유도된 노화 세포(Therapy induced senescence cell; TIS cell)인 것을 특징으로 할 수 있다.
본 발명에 있어서, 상기 치료 유도된 노화 세포는 화학요법, 생물학적 요법, 및/또는 방사선 요법에 의해 노화가 유도된 것을 특징으로 할 수 있다.

본 발명의 노화 세포 사멸용 조성물은 하나 이상의 담체 또는 부형제를 이용하여 제제화함으로써 단위 용량 형태로 제조되거나 다용량 용기 내에 내입시켜 제조될 수 있다.
본 발명에서 사용되는 용어, "담체"란 생물체를 자극하지 않으면서, 주입되는 화합물의 생물학적 활성 및 특성을 저해하지 않는 담체 또는 희석제를 의미할 수 있다. 본 발명에 사용 가능한 상기 담체의 종류는 특별히 제한되지 아니하며 당해 기술 분야에서 통상적으로 사용되는 담체라면 어느 것이든 사용할 수 있다. 상기 담체의 비제한적인 예로는, 식염수, 멸균수, 링거액, 완충 식염수, 알부민 주사 용액, 덱스트로즈 용액, 말토 덱스트린 용액, 글리세롤, 에탄올 등을 들 수 있다. 이들은 단독으로 사용되거나 2 종 이상을 혼합하여 사용될 수 있다. 상기 담체는 비자연적 담체(non-naturally occuring carrier)를 포함할 수 있다. 또한, 필요한 경우 항산화제, 완충액 및/또는 정균제 등 다른 통상의 첨가제를 첨가하여 사용할 수 있으며, 희석제, 분산제, 계면활성제, 결합제, 윤활제 등을 부가적으로 첨가하여 수용액, 현탁액, 유탁액 등과 같은 주사용 제형, 환약, 캡슐, 과립 또는 정제 등으로 제제화하여 사용할 수 있다.
상기 조성물을 제제화할 경우에는 보통 사용하는 충진제, 증량제, 결합제, 습윤제, 붕해제, 계면활성제 등의 희석제 또는 부형제를 사용하여 조제된다.
본 발명에 따른 노화 세포 사멸용 조성물은 통상의 방법에 따라 다양한 형태로 제형화하여 사용될 수 있다. 적합한 제형으로는 정제, 환제, 산제, 과립제, 당의정, 경질 또는 연질의 캡슐제, 용액제, 현탁제 또는 유화액제, 주사제, 에어로졸 등의 경구형 제형, 외용제, 좌제 및 멸균 주사용액 등이 있으나, 이에 한정되는 것은 아니다.
본 발명에 따른 노화 세포 사멸용 조성물은 약학적으로 불활성인 유기 또는 무기 담체를 이용하여 적합한 제형으로 제조할 수 있다. 즉, 제형이 정제, 코팅된 정제, 당의정 및 경질 캡슐제인 경우 락토스, 수크로스, 전분 또는 그 유도체, 탈크, 칼슘 카보네이트, 젤라틴, 스테아르산 또는 그 염을 포함할 수 있다. 또한, 제형이 연질 캡슐제인 경우에는 식물성 오일, 왁스, 지방, 반고체 및 액체의 폴리올을 포함할 수 있다. 또한, 제형이 용액 또는 시럽 형태인 경우, 물, 폴리올, 글리세롤, 및 식물성 오일 등을 포함할 수 있다.
본 발명에 따른 노화세포 사멸용 조성물은 상기의 담체 외에도 보존제, 안정화제, 습윤제, 유화제, 용해제, 감미제, 착색제, 삼투압 조절제, 산화방지제 등을 더 포함할 수 있다.
본 발명의 용어, "투여"는 어떠한 적절한 방법으로 대상에게 본 발명의 조성물을 도입하는 것을 의미하며, 본 발명의 조성물의 투여 경로는 목적 조직에 도달할 수 있는 한 경구 또는 비경구의 다양한 경로를 통하여 투여될 수 있다. 본 발명에 따른 약학 조성물의 투여 방식은 특별히 제한되지 아니하며, 당해 기술 분야에서 통상적으로 사용하는 방식에 따를 수 있다. 투여의 방식은, 예를 들어, 정맥내, 동맥내, 복강내, 근육내, 동맥내, 복강내, 흉골내, 경피, 비측내, 흡입, 국소, 직장, 경구, 안구내 또는 피내 경로를 통해 통상적인 방식으로 투여될 수 있으며, 상기 예에 제한되는 것은 아니다.
본 발명의 노화 세포 사멸용 조성물은 투여 목적, 투여 경로, 대상의 연령, 성별 및 체중 및 질환의 중등도 등의 여러 관련 인자와 함께, 활성성분인 약물의 종류에 따라 결정된다.
본 발명에 있어서, 상기 노화 세포는 SA-b-gal 염색에 양성을 타나내는 것을 특징으로 할 수 있다. 본 발명에 있어서, 상기 노화 세포는 p21의 발현이 향상된 것을 특징으로 할 수 있다.
본 발명의 일 실시예에서는 치료 유도 노화된 종양 세포의 유전자 발현을 분석하여 발현이 향상된 유전자를 동정하였다.
본 발명에 있어서, 상기 노화 세포는 ATP2B4, CPE, CXCR4, DGAT2, F2R, FYN, ICAM1, IL6, INSR, KDM7A, MAOA, MAP2, MAST4, PDE4B, PIM1, PINK1, PLD1, PSMB9, QPRT, S1PR3, SLC6A8, UGCG 및 VEGFA로 구성된 군에서 선택되는 어느 하나 이상의 유전자의 발현이 정상 대조군 세포에 비해 증가한 것을 특징으로 할 수 있다.
본 발명에 있어서, 상기 노화 세포는 바람직하게는 PIM1 유전자의 발현이 정상대조군 세포에 비해 증가한 것을 특징으로 할 수 있다.

본 발명에 있어서, 상기 노화 관련 질환은 암 또는 종양인 것을 특징으로 할 수 있으며, 보다 바람직하게는 노화 암 또는 노화 종양인 것을 특징으로 할 수 있다.
본 발명에 있어서, 상기 노화 관련 질환은 노화 세포를 검출하여 진단될 수 있으며, 상기 노화 관련 질환의 진단은 BrdU 또는 3H-티리비딘의 혼읩에 의한 DNA 복제 부족 검출, 증식 세포 핵 항원(PCNA) 및 Ki-67과 같은 단백질에 대한 면역 염색, SA-β-gal 조직화학 염색, p16, p19, Pai1, Igfbp2, IL-6, Mmp13, Nrg1, DEC1(differentiated embryo-chondrocyte expressed-1), p15, CDK1 및 DCR2(decoy death receptor-2)의 발현 검출, 노화 관련 이질염색질 병소(senescence-associated heterochromatin foci, SAHFs) 및/또는 노화 관련 DNA 손상 병소(senescence-associated DNA-damage foci, SDFs)의 검출을 통해 수행될 수 있다. 상기 SAHFs의 검출은 H3 Lys9 메틸화와 같은 특정 이질염색질 관련 히스톤 변형 및/또는 HP-1(heterochro-matin protein-1)의 존재 및 DAPI와 같은 DNA 염료의 우선적 결합 등을 통해 검출될 수 있다. 또 다른 예로서, 노화세포는 미국특허 제5,491,069호 및 미국 특허출원제2010/0086941호에 기재된 것과 같은 방법으로 검출될 수 있으나, 이에 제한되는 것은 아니다.본 발명에 있어서, 상기 노화 세포 사멸용 조성물은 약학적 조성물, 식품 조성물, 식품 보충제 조성물, 화장료 조성물 또는 화장품의 형태로 제조될 수 있다.

일 실시예에서, 화학요법제로 노화가 유도된 종양 세포에 Pim-1 키나아제 활성 억제제(PIM1/2 Kinase Inhibitor VI)를 처리하거나, PIM-1 키나아제 유전자를 표적으로하는 siRNA를 처리하는 경우 정상 종양세포에 비해 치료 유도된 노화 종양 세포에 대해 현저한 사멸효과를 나타내어, 선택적으로 치료 유도된 노화세포를 사멸시킬 수 있음을 확인하였다.
본 발명의 Pim-1 키나아제 억제제 및 이를 포함하는 노화 세포의 사멸용 조성물은 1차적인 항암 요법에 의한 종양 세포의 노화 후, 노화된 종양 세포를 사멸시켜 장기적인 종양의 재발 및 부작용을 방지하는 원-투 펀치 요법(One-Two punch theraphy)의 노화 세포 사멸 약물(senolytic drug)로서 사용될 수 있다.
따라서, 본 발명은 또 다른 관점에서, Pim-1 키나아제 억제제를 포함하고, 종양 세포의 노화 유도 요법 후 투여되는, 종양의 예방 또는 치료용 약학적 조성물에 관한 것이다.
pim-1이 종양유전자로 밝혀지면서 종양을 치료하기 위한 다양한 Pim-1 키나아제 억제제가 개발되었으며, 독소루비신과 같은 항암제와의 병용요법도 보고된 바 있다(Jeffrey D. Altenburg et al. Blood(2014) 124 (21) 4505). 그러나, Pim-1 키나아제가 종양유전자 유도된 노화(Oncogene-induced senescence)에 관여하는 것으로 알려지면서, Pim-1 키나아제의 발현 억제가 정상세포에서 종양 형성을 유도할 수 있으며, 장기적인 관점에서 Pim-1 키나아제에 의해 유도된 종양 세포의 노화가 종양의 치료에 도움이 될 수 있다고 보고되었으며, Pim-1 키나아제의 억제가 종양의 생성 및 증식을 유발할 수 있다는 점이 보고되었다(Bo Jin et al. Aging cell 2014, 13, pp879-889; Wu Wu et al. Med Sci Monit, 2019, 25: 8651-8659). Pim-1 키나아제의 위와 같은 특성은 Pim-1 키나아제 억제제가 종양의 치료를 위한 단독요법 또는 세포의 노화를 유도하는 다른 항암요법, 방사선 요법 등과의 병용 요법에서, 효과가 반감되거나 예기치 못한 부작용을 나타낼 수 있음을 의미한다.
따라서, 본 발명의 약학적 조성물은 종양 세포의 노화 유도 요법 뒤에 투여되는 것을 특징으로 할 수 있다.
본 발명의 약학적 조성물은 종양의 치료를 위한 원-투 펀치 요법(One-Two punch theraphy)에 있어서, 노화를 유도하는 항암 요법에 의해 형성되는 치료 유도된 노화 종양 세포를 선택적으로 사멸시키는 노화세포 사멸 약물(Senolytic drug)의 용도로 사용되는 것을 특징으로 할 수 있다.
본 발명의 용어, “원-투 펀치 요법”은 종양의 노화를 유도하는 기존의 항암요법, 예를 들어 화학요법, 생물학적 제제, 방사선 요법 등을 이용한 1차 치료 후, 노화 세포를 선택적으로 사멸시키는 세놀리틱 약물을 투여하여 상기 치료에 의해 형성되는 치료 유도된 노화 종양 세포를 선택적으로 사멸시키는 요법을 의미한다. 원-투 펀치 요법은 약물의 병용 투여를 기반으로 하지 않고 각 약물을 사용한 순차적 치료를 기반으로 하기 때문에, 약물의 조합으로 발생하는 조합 독성을 회피할 수 있으며, 치료 유도된 노화 종양 세포에 의한 장기적 부작용을 예방할 수 있다.
본 발명에 있어서, 상기 Pim-1 키나아제 억제제는 Pim-1 키나아제 유전자 발현 억제제, 또는 PIM1 키나아제 단백질 활성 억제제인 것을 특징으로 할 수 있다.

본 발명에 있어서, 상기 Pim-1 키나아제 유전자 발현 억제제는 바람직하게는 Pim-1 키나아제 유전자를 표적으로 하는 siRNA인 것을 특징으로 할 수 있으며, 보다 바람직하게는 상기 siRNA는 서열번호 1로 표시되는 서열을 포함할 수 있으나, 이에 제한되는 것은 아니다.
본 발명에 있어서, 상기 Pim-1 키나아제 단백질 활성 억제제는 표적단백질의 활성을 직접적으로 억제하거나, 다른 단백질과의 상호작용을 방해하여, 이의 기능을 억제하고, 하위 신호 전달 경로를 차단하는 것을 특징으로 할 수 있다. 상기 Pim-1 키나아제 단백질 활성 억제제는 예를 들어, Pim-1 키나아제 단백질에 특이적으로 결합하는 화합물, 펩티드, 펩티드 모방체, 기질 유사체, 압타머 및 항체 등일 수 있으나, 이에 제한되는 것은 아니다.

본 발명의 용어, “종양 세포의 노화 유도 요법”은 종양 세포의 노화를 유도하여 종양 세포의 성장과 증식을 억제하고, 노화 세포에서 분비되는 SASP에 의해 인접 암세포의 어레스트, 면역세포 모집과 같은 영향을 통한 항암 요법을 의미한다. 본 기술분야에서 통상적인 항암요법, 예를 들어 통상의 화학요법 및 방사선 요법 등은 종양 세포의 노화를 유도하는 것으로 알려져 있으며(Faget, D. 본 발명에 있어서, 상기 항암제는 바람직하게는 암을 예방 또는 치료하는 것으로 알려진 화학적 또는 생물학적 물질인 것을 특징으로 할 수 있다.
본 발명에 있어서, 상기 항암제는 액티노마이신 D, 아드리아마이신, 암사크린, 아라-C, 9-(3-D-아라비노실2-플루오로아데닌, BCNU, 블레오마이신, 캄토테신, 카보플라틴, 2-클로로-2-데옥시아데노신, CPT-11, 사이클로 포스파미드, 도세탁셀, 독소루비신, 에도테카린, 에토포시드, 플루다라빈, 5-플루오로유라실(5-FU), 젬시타빈, HU-젬자, 이리노테칸, 메토트렉세이트, 6-엠퓨린, 마이토미신-C, 패클리탁셀, 시스-플라틴, SN-38, 탁솔, 티오테파, 6-티오구아닌, 트라이메트렉세이트, 빈블라스틴, 빈크리스틴, 및 VP-16으로 이루어진 군으로부터 선택되는 것을 특징으로 할 수 있으나, 이에 제한되는 것은 아니다.
본 발명에 있어서, 상기 항암제는 DNA 손상제인 것을 특징으로 할 수 있다. 상기 DNA 손상제는 예를 들어 알킬화제, 대사길항제, 항종양 항생제, 백금 동족체, 국소이성질화효소 I 억제제 및 국소이성질화효소 II 억제제로 구성된 군에서 선택될 수 있으나, 이에 제한되는 것은 아니다.
본 발명에 있어서, 상기 항암제는 유사 분열 억제제인 것을 특징으로 할수 있다.
본 발명에 있어서, 상기 항암제는 표적 항암제인 것을 특징으로 할 수 있다. 상기 표적 항암제는 예를 들어, CDC7 억제제(예, XL413, TAK-931), 텔로머라아제 억제제(예, GRN163L, BIBR15), 후성유전학적 조절제(예, Decitabine, Vorinostat), CDK 억제제(예, Palbociclib, Abemaciclib, Ribociclib, PF-06873600), AURK 억제제(예, Alisertib, barasertib), PLK1 억제제(예, BI-2535, BI-6727), MDM-p53 신호전달 붕괴제(MDM2-p53 disruptor)(예, Nitlin3, RG7112, UBX0101), PTEN 억제제(예, VO-OHpic) 등이 있으나, 이에 제한되는 것은 아니다.
본 발명에 있어서, 상기 항암제는 항호르몬제, 아로마타제 억제제, 항안드로겐제, 단백질키나아제 억제제, 안티센스 올리고뉴클레오타이드, 리보자임, 종양백신 등을 포함하는 것을 특징으로 할 수 있다.
본 발명에 있어서, 상기 항암제는 치료적 항체 및 항체-약물 접합체(Antibody-Drug conjugates)인 것을 특징으로 할 수 있다. 상기 치료적 항체는 예를 들어, 알렘투주마브 (alemtuzumab, Campath), 베바시주마브 (AVASTIN, Genentech); 세툭시마브 (cetuximab, Imclone); 파니투무마브 (panitumumab, Amgen), 리툭시마브 (rituximab, Genentech/Biogen Idec), 페르투주마브 (OMNITARG™, 2C4, Genentech), 트라스투주마브(HERCEPTIN, Genentech), 및 토시투모마브 (tositumomab, Corixia) 등이 있으나, 이에 제한되는 것은 아니다.
본 발명에서 예시적으로 나열된 "알킬화제", "대사길항제", "항종양 항생제", "백금 동족체", "국소이성화효소 I 억제제", "국소이성화효소 II 억제제", "유사분열 억제제", “표적 항암제”, “치료용 항체”와 같은 용어는 세포의 노화를 유도할 수 있는 것이라면 임상적으로 사용되는 화학적 또는 생물학적 항암제를 제한없이 포함하는 의미로 사용된다.
본 발명의 약학적 조성물은 종양 세포의 노화 유도 요법에 의해 유도된 치료 유도된 노화 종양 세포를 선택적으로 사멸시키는 것을 특징으로 할 수 있다.
본 발명에 있어서, 상기 치료 유도된 노화 종양 세포는 SA-b-gal 염색에 양성을 타나내는 것을 특징으로 할 수 있다. 본 발명에 있어서, 상기 노화 세포는 p21의 발현이 향상된 것을 특징으로 할 수 있다.
본 발명의 일 실시예에서는 치료 유도 노화된 종양 세포의 유전자 발현을 분석하여 발현이 향상된 유전자를 동정하였다.
본 발명에 있어서, 상기 치료 유도된 노화 종양 세포는 ATP2B4, CPE, CXCR4, DGAT2, F2R, FYN, ICAM1, IL6, INSR, KDM7A, MAOA, MAP2, MAST4, PDE4B, PIM1, PINK1, PLD1, PSMB9, QPRT, S1PR3, SLC6A8, UGCG 및 VEGFA로 구성된 군에서 선택되는 어느 하나 이상의 유전자의 발현이 정상 대조군 세포에 비해 증가한 것을 특징으로 할 수 있다.
본 발명에 있어서, 상기 치료 유도된 노화 종양 세포는 바람직하게는 PIM1 유전자의 발현이 정상대조군 세포에 비해 증가한 것을 특징으로 할 수 있다.
본 발명의 약학적 조성물은 종양 세포의 노화 유도 요법, 바람직하게는 항암제의 투여 및/또는 방사선 요법 이후에 순차적으로 투여되는 것을 특징으로 할 수 있다.
본 발명의 약학적 조성물은 종양 세포의 노화 유도 요법이 대상에게 적용되고 난 이후 일정한 간격을 두고 투여되는 것을 특징으로 할 수 있다. 본 발명에 있어서, 상기 약학적 조성물은 바람직하게는 종양 세포의 노화 유도 요법에 의해 치료 유도된 노화 세포가 충분히 형성된 후 투여되는 것을 특징으로 할 수 있다.
본 발명에 있어서, 상기 약학적 조성물은 바람직하게는 노화 유도 요법 이후, 수시간, 수 일, 수 주, 수 개월 또는 수년의 간격을 두고 투여될 수 있으며, 보다 바람직하게는 약 1시간 내지 약 24시간, 약 1일 내지 약 6일, 약 1주일 내지 약 4주일, 약 1개월 내지 11개월, 또는 약 1년 이상의 간격을 두고 투여되는 것을 특징으로 할 수 있으나, 이에 제한되는 것은 아니다. 보다 바람직한 원-투 펀치 요법의 적용을 위해 상기 약학적 조성물은 대상으로부터 분리된 샘플에서, 노화 세포의 존재를 확인한 뒤 투여되는 것을 특징으로 할 수 있다.
본 발명에 있어서, 상기 약학적 조성물은 유효성분인 Pim-1 키나아제 억제제외에도, 다른 노화 세포 사멸 약물(senolytic drug)을 추가로 포함하는 것을 특징으로 할 수 있다. 암 또는 종양의 치료에 사용되는 노화 세포 사멸 약물(senolytic drug)은 당업계에 잘 알려져 있다(Wang, L., Lankhorst, L. & Bernards, R. Exploiting senescence for the treatment of cancer. Nat Rev Cancer 22, 340-355 (2022)).
본 발명은 또 다른 관점에서, 종양 세포의 노화 유도 요법 이후, Pim-1 키나아제 억제제를 투여하는 단계를 포함하는 상기 약학적 조성물의 투여방법에 관한 것이다.
본 발명은 또 다른 관점에서, Pim-1 키나아제 억제제를 포함하는 종양의 예방 또는 치료용 약학적 조성물의 제조를 위한 용도에 관한 것으로, 상기 약학적 조성물은 종양 세포의 노화 유도 요법 뒤에 투여되는 것을 특징으로 할 수 있다.
본 발명은 또 다른 관점에서, Pim-1 키나아제 억제제의 종양의 예방 또는 치료를 위한 원-투 펀치 요법에서, 세놀리틱 약물로서의 용도에 관한 것이다.
본 발명은 또 다른 관점에서, 종양 세포의 노화를 유도하는 단계; 및 Pim-1 키나아제 억제제를 투여하여 노화된 종양 세포를 사멸시키는 단계를 포함하는 종양의 예방 또는 치료 방법에 관한 것이다.

본 발명에 있어서, 별도로 언급되지 않는 한 상기 종양 세포의 노화 유도 요법은 상기 약학적 조성물의 관점에서 서술된 것과 동일한 특징을 공유할 수 있음은 통상의 기술자에게 쉽게 이해될 수 있다.
본 발명에 있어서, 상기 Pim-1 키나아제 억제제의 투여는 직접적인 Pim-1 키나아제 희석액으로 투여될 수 있으며, 본 발명의 노화 세포 사멸용 조성물 또는 약학적 조성물로 제조되어 투여될 수 있다.
본 발명에 있어서, 종양 세포의 노화 유도 요법과 Pim-1 키나아제 억제제의 순차적 투여 이외에도, 대상에 존재하는 노화 세포의 사멸을 통해 종양 세포를 치료할 수 있다.
따라서, 본 발명은 또 다른 관점에서, 대상 또는 대상에게서 분리한 샘플에서 노화 세포를 검출하는 단계; 노화 세포가 검출된 경우 대상에게 Pim-1 키나아제 억제제를 투여하는 단계를 포함하는 종양의 예방 또는 치료 방법에 관한 것이다.
본 발명에 있어서, 상기 노화 세포를 검출하는 단계는 생체 내 또는 시험관 내에서 수행될 수 있다. 본 발명에 있어서, 상기 노화 세포를 검출하는 단계는 세포의 크기 증가, 세포 수 증가와 같이 육안 또는 현미경을 통해 검출되는 것을 특징으로 할 수 있다.
본 발명에 있어서, 상기 노화 세포를 검출하는 단계는 검출은 당업계에 공지된 다양한 노화 세포 마커를 검출하여 수행될 수 있다. 예를 들어, 상기 노화 세포를 검출하는 단계는 BrdU 또는 3H-티리비딘의 혼읩에 의한 DNA 복제 부족 검출, 증식 세포 핵 항원(PCNA) 및 Ki-67과 같은 단백질에 대한 면역 염색, SA-β-gal 조직화학 염색, p16, p19, Pai1, Igfbp2, IL-6, Mmp13, Nrg1, DEC1(differentiated embryo-chondrocyte expressed-1), p15, CDK1 및 DCR2(decoy death receptor-2)의 발현 검출, 노화 관련 이질염색질 병소(senescence-associated heterochromatin foci, SAHFs) 및/또는 노화 관련 DNA 손상 병소(senescence-associated DNA-damage foci, SDFs)의 검출을 통해 수행될 수 있다. 상기 SAHFs의 검출은 H3 Lys9 메틸화와 같은 특정 이질염색질 관련 히스톤 변형 및/또는 HP-1(heterochro-matin protein-1)의 존재 및 DAPI와 같은 DNA 염료의 우선적 결합 등을 통해 검출될 수 있다. 또 다른 예로서, 노화세포는 미국특허 제5,491,069호 및 미국 특허출원제2010/0086941호에 기재된 것과 같은 방법으로 검출될 수 있으나, 이에 제한되는 것은 아니다.
본 발명에 있어서, 상기 노화 세포를 검출하는 단계는 ATP2B4, CPE, CXCR4, DGAT2, F2R, FYN, ICAM1, IL6, INSR, KDM7A, MAOA, MAP2, MAST4, PDE4B, PIM1, PINK1, PLD1, PSMB9, QPRT, S1PR3, SLC6A8, UGCG 및 VEGFA로 구성된 군에서 선택되는 어느 하나 이상의 유전자의 발현 수준을 확인하여 수행될 수 있다. 본 발명에 있어서, 상기 군에서 선택되는 어느 하나 이상의 유전자의 발현 수준이 정상 대조군 세포에 비해 증가한 경우 노화 세포로 검출하는 것을 특징으로 할 수 있다.
본 발명에 있어서, 상기 노화 세포를 검출하는 단계는 PIM1 유전자의 발현 수준을 확인하는 단계를 포함하는 것을 특징으로 할 수 있다.
본 발명에 있어서, 상기 PIM-1 키나아제 유전자의 발현 수준이 정상대조군 세포에 비해 증가한 경우, 노화 세포로 검출하는 것을 특징으로 할 수 있다.

본 발명의 용어, "치료"란, 본 발명의 노화 세포 사멸용 조성물 또는 약학적 조성물을 질환 발병 개체에 투여하여 상기 질환의 증세가 호전되도록 하거나 이롭게 되도록 하는 모든 행위를 의미할 수 있다.
본 발명에서 사용되는 용어, "개체" 또는 “대상”은 질환이 발병되었거나 발병할 가능성이 있는 인간을 포함한 모든 동물을 의미할 수 있다. 상기 동물은 인간뿐만 아니라 이와 유사한 증상의 치료를 필요로 하는 소, 말, 양, 돼지, 염소, 낙타, 영양, 개, 고양이 등의 포유동물일 수 있으나, 이에 제한되지는 않는다.
본 발명에 있어서, 상기 “대상”은 노화 세포를 갖는 것을 특징으로 할 수 있다. 본 발명에 있어서, 상기 “대상”은 치료 유도된 노화 세포를 갖는 것을 특징으로 할 수 있다.
본 발명에 있어서, 상기 “대상”은 종양 세포의 노화 유도 요법에 의해 노화된 세포를 가질 수 있다. 본 발명에 있어서, 상기 대상은 항암 요법을 적용받은 것을 특징으로 할 수 있으며, 상기 항암 요법에 의해 치료 유도된 노화 세포를 갖는 것을 특징으로 할 수 있다.
본 발명의 용어, "투여"는 어떠한 적절한 방법으로 대상에게 본 발명의 약학 조성물을 도입하는 것을 의미하며, 본 발명의 조성물의 투여 경로는 목적 조직에 도달할 수 있는 한 경구 또는 비경구의 다양한 경로를 통하여 투여될 수 있다. 본 발명에 따른 약학 조성물의 투여 방식은 특별히 제한되지 아니하며, 당해 기술 분야에서 통상적으로 사용하는 방식에 따를 수 있다. 투여의 방식은, 예를 들어, 정맥내, 동맥내, 복강내, 근육내, 동맥내, 복강내, 흉골내, 경피, 비측내, 흡입, 국소, 직장, 경구, 안구내 또는 피내 경로를 통해 통상적인 방식으로 투여될 수 있으며, 상기 예에 제한되는 것은 아니다.
본 발명에 있어서, 대상으로부터 분리된 샘플은 상기 대상으로부터 분리된 조직, 세포, 전혈, 혈청, 혈장 또는 타액 등을 포함할 수 있고, 바람직하게는 종양 조직 또는 종양 스페로이드를 포함할 수 있으나, 이에 제한되는 것은 아니다.
[실시예]
이하, 실시예를 통하여 본 발명을 더욱 상세히 설명하고자 한다. 이들 실시예는 오로지 본 발명을 예시하기 위한 것으로, 본 발명의 범위가 이들 실시예에 의해 제한되는 것으로 해석되지 않는 것은 당업계에서 통상의 지식을 가진 자에게 있어서 자명할 것이다.
실시예 1: 노화 종양 세포의 제조
치료 유도된 노화 종양 세포를 제조하기 위해 아드리아마이신(Selleckhem, Catalog No.S1208, CAS 25316-40-9)을 대조적인 성격을 나타내는 유방암 세포주 (여성 호르몬 수용체 발현 양성이면서 상대적으로 덜 악성이 성격을 보이는 MCF7, ZR75-1와 호르몬수용체 및 HER2 수용체 음성의 삼중응성 유방암 세포주로 상대적으로 좀더 공격적인 성격을 보이는 Hs578T, MDA-MB-231)에 100nM를 처리 후 48시간 후에 세포배양액을 교환한 후 5-7일 배양하여 치료 유도된 노화 종양 세포를 제조하였다.
도 1에 도시된 것과 같이, 아드리아마이신에 의한 치료 유도 노화 후, 각 유방암 세포주의 세포 크기 증가하는 것을 확인하였으며, 노화 마커인 SA-b-gal 양성 세포의 수가 증가한 것을 확인하였다. 또한 각 세포주에서 세포주기 억제 인자인 p21의 mRNA 발현도 현저히 증가함을 확인하여 치료 유도 노화된 유방암 세포를 수득하였다.
실시예 2: 치료 유도된 노화 유방암 세포의 유전자 발현 분석 및 약물의 표적 선정
다음으로, 실시예 1에서 제조된 치료 유도된 노화 유방암세포의 유전자 발현을 RNASeq를 통해 분석한 뒤, 두 세포주 모두에서 발현이 향상된 322개의 유전자를 동정하였다(도 2A).
발현이 향상된 322개의 유전자 중, FDA 승인된 약물에 의해 표적이 가능한 유전자 28개를 선정하였으며 MCF7 및 MDA-MB 231 세포주에서 치료에 의한 노화를 유도하는 경우 28개 유전자의 발현 수준 변화를 확인하였다(도 2B).
MCF7 및 MDA-MB 231 세포주 이외에도, BT474, Hs578t, T47D, HCC1937과 같은 다른 유방암 세포주에 대해 실시예 1과 동일한 방법으로 치료에 의한 노화를 유도하고, 상기 28개의 유전자 발현 변화를 확인하여 적어도 2가지 이상의 세포주에서 동일한 발현 향상 패턴을 나타내는 다음의 유전자 23종을 표적으로 최종 선정하였다:
ATP2B4, CPE, CXCR4, DGAT2, F2R, FYN, ICAM1, IL6, INSR, KDM7A, MAOA, MAP2, MAST4, PDE4B, PIM1, PINK1, PLD1, PSMB9, QPRT, S1PR3, SLC6A8, UGCG 및 VEGFA.
실시예 3: 각 표적에 대한 약물의 노화 세포 선택적 사멸효과(senolytic effect) 확인
실시예 2에서 최종 선정된 23종의 유전자에 대한 FDA 승인 억제제(표 1)를 치료 유도된 노화 유방암 세포에 처리하는 경우에 노화 세포 선택적 세포 분열 억제 및 사멸효과(senolyric effct)를 CCK8 assay (D-Plus CCK, cell viability assay kit, Cat CCK-3000, 동인LS)를 통하여 확인하였다(도 3A). 이 때 대조군(치료 유도되지 않은 암세포)의 경우는 해당 억제제를 48시간 표기된 농도로 처리한 후에 측정을 하였으며, 치료 유도 노화 세포군의 경우 실시예 1과 같이 암세포에 아드리아마신을 48시간 처리하여 치료 유도 노화를 진행시킨 후에 배양액을 교환하면서 해당 억제제를 표기된 농도로 48시간 동안 처리하고 측정하였다.
도 3A에 도시된 것과 같이, 표 1의 표적을 억제하는 것으로 알려진 FDA 승인된 약물을 대조군 및 치료 유도된 노화 종양 세포에 처리하여 세포 사멸능을 확인한 결과, PIM-1 키나아제 억제제를 처리하는 경우 만이 치료 유도된 노화 종양 세포에 선택적인 사멸효과를 나타내는 것을 확인하였다.
PIM-1 키나아제를 최종 표적으로 선정하고, 기존의 원-투 펀치 요법에 사용되는 노화 세포 선택적 사멸 약물(Senolytic drug)으로 보고된 ABT-263과의 노화 세포 선택적 사멸 효과를 비교 하였다. 그 결과, 도 3B 및 3C에 도시된 것과 같이, ABT 263은 대조군과 TIS 세포의 사멸 효과에 유의미한 차이는 있었으나, 대조군과 TIS 세포 모두에 거의 완전한 사멸효과를 나타낸 반면, PIM-1 키나아제 억제제는 대조군은 고농도에서 세포 성장을 저하하지만, 거의 사멸시키지 않는데 반해 TIS 세포는 효과적으로 사멸시켜, 노화 세포에 대한 선택적 사멸능이 ABT-263에 비해 현저히 뛰어난 것을 확인하였다.
또한, PIM1/2 Kinase Inhibitor VI외의 PIM-1 키나아제 억제제를 사용하는 경우에도, 위에서 확인한 것과 동일한 노화 세포 선택적 사멸능을 나타내는지 확인하기 위해, SGI-1776(Selleckhem, Catalog No.S2198, CAS 1025065-69-3)을 사용하여, 동일한 방법으로 노화 세포 선택적 사멸 효과를 비교 하였다. 도 3D에 도시된 것과 같이, SGI-1776 (CAS 1025065-69-3)를 처리한 경우에도, TIS 세포에서 대조군에 비애 높은 세포 독성을 나타내어 PIM1 키나아제 억제제의 종류에 관계없이 뛰어난 노화 세포에 대한 선택적 사멸효과를 나타낼 수 있음을 확인하였다.
실시예 4: 다양한 암 세포주에 대한 치료에 의한 노화 유도 및 PIM 1 키나아제의 발현 분석 및 노화 세포 선택적 사멸능 분석
PIM 키나아제는 다르게 인산화와 같이 활성을 조절하는 2차적인 변화가 잘 알려져 있지 않으며, PIM 키나아제의 발현량에 전체적인 활성이 비례하는 것으로 보고되어 있다.
유방암 세포주(MCF7 및 MDA-MB231) 이외에도, 위암(SNU601, MKN1, MKN74), 대장암(SW480, Hct116), 두경부암(HSC4), 자궁경부암(HeLa S3), 담관암 (SNU308, SNU478, SNU1196), 췌장 선암종(BxPC3), 전립선암(PC3) 세포주에 대해 실시예 1과 동일한 방법을 통해 치료에 의한 노화를 유도하여 치료 유도된 노화 세포를 수득하였다.
그 결과 도 4a 내지 4b에 도시된 것과 같이, 모든 암세포주에 대해 치료 유도된 노화가 진행되는 경우, PIM-1 키나아제 mRNA와 단백질의 발현이 증가하는 것을 확인하였다.
또한, 아드리아마이신 외의 항암제를 사용하는 경우에도, 치료 유도 노화(TIS)가 발생하는지 확인하기 위해, 시스플라틴(cisplatin Selleckhem, Catalog No.S1166 CAS 15663-27-1) 10uM을 다양한 암세포주(MCF7, MDA-MB 231, MKN1, HSC4, SNU869)에 처리하는 경우에도 치료 유도 노화(TIS)가 발생하였으며, PIM-1 키나아제 유전자의 mRNA 발현이 현저히 증가되는 것을 확인하였다(도 4c).
치료에 의해 노화가 유도된 위암(SNU601), 두경부암(HSC4), 대장암(SW480) 세포주에 대해 Pim-1 키나아제 억제제를 처리하는 경우의 노화세포 선택적 사멸효과를 확인하기 위해 실시예 3에 도시된 방법과 동일한 방법으로 농도에 따른 사멸능을 확인하였다.
도 5에 도시된 것과 같이, 모든 암세포 주에서, 대조군은 유의미한 사멸효과가 나타나지 않은데 반해, 치료 유도된 노화 암세포의 경우 현저한 사멸효과를 나타내어, Pim-1 키나아제 억제제가 노화 세포를 선택적으로 사멸시킬 수 있음을 확인하였다.
실시예 4: Pim-1 유전자 발현 억제를 통한 노화 세포 선택적 사멸능 분석
Pim-1 키나아제 단백질 억제제의 처리 외에도, Pim-1 유전자의 발현 억제시 노화 세포를 선택적으로 사멸할 수 있는지 확인하기 위해, Pim-1 유전자를 표적으로 하는 siRNA(GUAUGAUAUGGUGUGUGGAGAUAUUC, 서열번호 1)를 처리하여 Pim-1 유전자의 발현을 억제하고 세포 독성을 확인하였다.
그 결과 도 6에 도시된 것과 같이, Pim-1 유전자를 표적으로 하는 siRNA를 처리하여 Pim-1 키나아제의 발현을 유전자 단계에서 억제(knock down)할 수 있음을 확인하였으며(도 6a), 다양한 암세포(MCF7, HSC4, SNU601)에 Pim-1 유전자를 표적으로 하는 siRNA를 처리하여 Pim-1 키나아제의 발현을 유전자 단계에서 억제(knock down)하는 경우에도 노화 세포에 대해 선택적인 사멸 효과를 나타내는 것을 확인하였다(도 6b 및 6c).
본 발명의 실시예에서와 같이, Pim-1 키나아제 단백질의 활성을 억제하는 다양한 화합물 또는 Pim-1 키나아제 유전자를 선택적으로 억제하는 siRNA의 처리시 일관적으로 노화 세포에 대해 뛰어난 노화세포의 성장 억제 및 사멸능을 고려하면, Pim-1 키나아제의 발현 억제 또는 활성 억제가 노화세포의 성장 억제 및 사멸을 유도할 수 있는 신규한 작동 메커니즘임이 입증되었다 할 수 있다.
따라서, 상기 실시예의 결과로부터 Pim-1 키나아제 유전자 발현 억제제, 또는 Pim-1 키나아제 단백질 활성 억제제는 그 종류 및 형태를 불문하고, Pim-1 키 (아주대) PP-B2966.docx나아제의 억제를 통해 다양한 노화 세포의 사멸 용도(senolytic use)에 이용될 수 있음을 자명하게 이해할 수 있다.
이상으로 본 발명 내용의 특정한 부분을 상세히 기술하였는 바, 당업계의 통상의 지식을 가진 자에게 있어서 이러한 구체적 기술은 단지 바람직한 실시 양태일 뿐이며, 이에 의해 본 발명의 범위가 제한되는 것이 아닌 점은 명백할 것이다. 따라서 본 발명의 실질적인 범위는 첨부된 청구항들과 그것들의 등가물에 의하여 정의된다고 할 것이다.
본 발명에 따른 Pim-1 키나아제 억제제를 포함하는 노화 세포의 사멸용 조성물은 노화 세포, 특히 Pim-1 키나아제 발현이 향상된 노화 세포에 선택적인 사멸효과를 나타낸다. 본 발명의 노화 세포의 사멸용 조성물은 노화 세포에 대한 선택적 사멸효과를 통해 노화 관련 질환의 예방 또는 치료에 유용하게 사용될 수 있으며, 특히, 종양 세포의 노화 유도를 기반으로 하는 항암요법에 후속적으로 Pim-1 키나아제 억제제를 투여하여 치료 유도된 노화 세포(Therapy induced senescence cell)를 선택적 사멸시킴으로써 뛰어난 종양의 예방 또는 치료 효과를 나타낼 수 있다.

Claims (20)
- Pim-1 키나아제 억제제를 포함하는 노화 세포의 사멸용 조성물.
- 제1항에 있어서, 상기 Pim-1 키나아제 억제제는 Pim-1 키나아제 유전자 발현 억제제, 또는 Pim-1 키나아제 단백질 활성 억제제인 것을 특징으로 하는 조성물.
- 제2항에 있어서, 상기 Pim-1 키나아제 유전자 발현 억제제는 Pim-1 키나아제 유전자를 표적으로 하는 안티센스 뉴클레오타이드(antisense nucleotide), siRNA(small interfering RNA), shRNA(small hairpin RNA), 리보자임(ribozyme), 압타머(aptamer), anti-microRNA, MicroRNA mimic, nuclease, Zinc Finger Nucleases(ZFNs), Transcription activator-like effector nucleases (TALENs), gRNA, sgRNA 및 self-ribozyme-flanked RNA로 구성된 군에서 선택되는 것을 특징으로 하는 조성물.
- 제3항에 있어서, 상기 Pim-1 키나아제 유전자 발현 억제제는 서열번호 1로 표시되는 서열을 포함하는 siRNA인 것을 특징으로 하는 조성물.
- 제2항에 있어서, 상기 Pim-1 kinase 단백질 활성 억제제는 Pim-1 kinase 단백질의 활성을 특이적으로 억제하는 화합물, 펩티드, 펩티드 모방체, 기질 유사체, 압타머, 항체 및 이의 단편으로 구성된 군에서 선택되는 것을 특징으로 하는 조성물.
- 제1항에 있어서, 상기 노화 세포는 노화 암 세포인 것을 특징으로 하는 조성물.
- 제6항에 있어서, 상기 노화 암 세포는 종양유전자 유도된 노화 세포(Oncogene-induced senescence cell; OIS cell) 또는 치료 유도된 노화 세포(Therapy induced senescence cell; TIS cell)인 것을 특징으로 하는 조성물.
- 제1항에 있어서, 상기 노화 세포는 Pim-1 키나아제의 발현이 향상된 것을 특징으로 하는 조성물.
- 제7항에 있어서, 상기 치료 유도된 노화 세포(TIS cell)는 화학 요법, 생물학적 요법 또는 방사선 요법에 의해 노화가 유도된 세포인 것을 특징으로 하는 조성물.
- 제7항에 있어서, 상기 치료 유도된 노화 세포(TIS cell)는 항암제에 의해 노화가 유도된 암세포인 것을 특징으로 하는 조성물.
- Pim-1 키나아제 억제제를 포함하는 노화 관련 질환의 예방 또는 치료용 약학적 조성물.
- 제11항에 있어서, 상기 노화 관련 질환은 알츠하이머, 파킨슨, 백내장, 황반변성, 녹내장, 동맥경화증, 급성 관상동맥 증후군, 심근경색, 뇌졸중, 고혈압, 특발성 폐섬유증(IPF), 만성 폐쇄성 폐질환(COPD), 골관절염, 제1형 또는 제2형 당뇨병, 비만, 지방기능장애, 관상동맥질환, 뇌혈관 질환, 치주질환, 각종 조직의 위측, 섬유증과 같은 암 치료 관련 장애, 뇌 및 심장의 손상, 및 골수이형성증후군으로 구성된 군에서 선택되는 것을 특징으로 하는 약학적 조성물.
- Pim-1 키나아제 억제제를 포함하고, 종양 세포의 노화 유도 요법 뒤에 투여되는, 종양의 예방 또는 치료용 약학적 조성물.
- 제14항에 있어서, 상기 Pim-1 키나아제 억제제는 Pim-1 키나아제 유전자 발현 억제제, 또는 Pim-1 키나아제 단백질 활성 억제제인 것을 특징으로 하는 약학적 조성물.

- 제13항에 있어서, 상기 종양 세포의 노화 유도 요법은 화학요법, 생물학적 요법 또는 방사선 요법인 것을 특징으로 하는 약학적 조성물.
- 제13항에 있어서, 상기 종양 세포의 노화 유도 요법에 의해 생성된 치료 유도된 노화 종양 세포(TIS tumor cell)를 선택적으로 사멸시키는 것을 특징으로 하는 약학적 조성물.
- 제13항에 있어서, 상기 종양 세포의 노화 유도 요법으로 유도된 노화 종양 세포는 Pim-1 키나아제의 발현이 향상된 것을 특징으로 하는 약학적 조성물.
