Abstract
嗅覚受容体の活性化を介した角層形成促進剤、肌荒れ抑制剤、保湿剤、皮膚バリア機能改善剤、メラニン産生抑制剤、及びそれらのスクリーニング法の提供。 OR10A6を指標とすることで、角層形成促進剤、肌荒れ抑制剤、保湿剤、皮膚バリア機能改善剤、及びメラニン産生抑制剤、のスクリーニングを行うことができる。
Description
本発明は、角層形成促進剤、肌荒れ抑制剤、保湿剤、及び皮膚バリア機能改善剤、及びメラニン産生抑制剤、並びにかかる剤のスクリーニング方法に関する。特に、本発明は、OR10A6、OR5P2、又はOR5P3の活性化による角層形成促進剤、肌荒れ抑制剤、保湿剤、皮膚バリア機能改善剤、及びメラニン産生抑制剤、かかる剤のスクリーニング方法、角層形成の促進、肌荒れの抑制、保湿機能の向上、皮膚バリア機能、メラニン産生の抑制のためのOR10A6活性化剤、OR5P2活性化剤、及びOR5P3活性化剤、並びにそれらを含む組成物に関する。
嗅覚受容体は、主に鼻に存在し匂いを感知するセンサーとして機能するが、近年ある種の嗅覚受容体は鼻のみならず、皮膚などの器官にも存在し皮膚に好ましい効果をもたらすことが報告されている(非特許文献1)。例えば、嗅覚受容体OR2AT4は角化細胞にも存在し、OR2AT4のアゴニストであるサンダロールの適用により角化細胞におけるOR2AT4が活性化されると創傷治癒や角化細胞の増殖が促進されることや、OR2AT4が毛包の鞘細胞にも存在しておりサンダロールの適用により毛髪伸長を引き起こすことも報告されている(非特許文献2、3、特許文献1)。
しかしながら、嗅覚受容体は哺乳類の種類によっては1,000種類以上存在し、ヒトでは機能する嗅覚受容体だけでも約400種類の存在が確認されている。その上、非常に多様な種類の匂いをかぎ分けることを可能とするため、これらの嗅覚受容体はそれぞれ1つではなく複数の物質に反応し多岐にわたる機能に関与する(非特許文献4)。例えば、汗腺に発現している嗅覚受容体OR6C74、OR51A7、OR4A15は発汗調節に関与することが報告されている(特許文献2)。このように嗅覚受容体の種類及び機能は非常に多岐にわたるため、どのような嗅覚受容体皮膚に存在しその活性化により皮膚に好ましい影響を与える物質については未知の部分が多く、更なる探索が求められる。

本発明者らは、嗅覚受容体に着目したスキンケアについて鋭意研究を行っており、角層形成促進剤、肌荒れ抑制剤、保湿剤、皮膚バリア機能改善剤、メラニン産生抑制剤等の取得を目的とする。
本発明者らは、嗅覚受容体であるOR10A6、OR5P2、及びOR5P3が皮膚において発現すること、そしてこれら嗅覚受容体の1つであるOR10A6のアゴニストによりOR10A6の活性を向上することで角層形成促進等の皮膚に好ましい作用を奏することを見出し、本発明に至った。
そこで本発明は下記に関する:

プロパン酸3-フェニルプロピル、アンドロスタジエノン、アンドロステロン、酪酸、又はOR10A6活性化剤を適用することで、OR10A6の活性化を介して角層形成の促進、肌荒れの抑制、保湿機能の向上、皮膚バリア機能の向上、メラニン産生の抑制が可能になる。更に、OR10A6の活性を評価することにより角層形成促進、肌荒れ抑制、保湿、皮膚バリア機能改善、又はメラニン産生抑制作用の評価が可能になる。かかる評価方法を利用し、角層形成促進剤、肌荒れ抑制剤、保湿剤、皮膚バリア機能改善剤、又はメラニン産生抑制剤をスクリーニングすることが可能になる。
本発明は、プロパン酸3-フェニルプロピル、シトロネロール、フェネチルアルコール、αシンナミルアルコール、シクラメンアルデヒド、αイオノン、ネロール、ノナデカン、リナロール、リラール、フェニルプロピルアルコール、アンドロスタジエノン、アンドロステロン、及び酪酸から選ばれる1種又は複数を有効成分として含有する、角層形成促進剤、肌荒れ抑制剤、保湿剤、皮膚バリア機能改善剤、メラニン産生抑制剤を提供する。
プロパン酸3-フェニルプロピル(CAS番号:122-74-7)は、フローラル系の香気を有し、主に香料として使用される物質である。本明細書では、3PPPと称する場合がある。
シトロネロール(CAS番号:106-22-9)は、ゼラニウム油、シトロネラ油などの精油に含まれ、バラ様の香気を有する香気成分である。
フェネチルアルコール(CAS番号:60-12-8)は、バラや酒などに含まれ、バラ様の香気を有し香料として利用されるほか、その抗菌作用により保存料などに利用される。
αシンナミルアルコール(CAS番号:4407-36-7)は、桂皮アルコール等とも呼ばれ、シナモンの葉等に含まれ、ヒアシンス様の香気を有する。
シクラメンアルデヒド(CAS番号:103-95-7)は、シクラメン、スズラン様の香気を有するが天然には存在しない人工香料物質である。
ゲラニオール(CAS番号:106-24-1)は、ゼラニウム油、パルマローザ油、シトロネラ油等に含まれ、バラ様香気成分として知られている。
αイオノン(CAS番号:127-41-3)は、テルペノイドの一種であり、α-イオノン、β-イオノン、γ-イオノンの異性体のうちの1つである。異性体ごとに異なる香気を有し、α-イオノンは、スミレの花様の香気を有する。
ネロール(CAS番号:106-25-2)は、ネロリ油、バラ油等に含まれバラ様の香気を有する香気成分である。
ノナデカン(CAS番号:629-92-5)は、バラ油等に含まれワックス様のにおいを有する成分である。
リナロール(CAS番号:78-70-6)は、(R)体と(S)体が存在し、(S)体はオレンジ様、(R)体はラベンダー様の香気を有し香料として使用されるが、ゲラニオールやシトラールなどの合成原料としても使用される。ラセミ体が好ましく使用できる。
リラール(CAS番号:31906-04-4)は、リリアールとも呼ばれスズラン様の香気を有する香料である。
フェニルプロピルアルコール(CAS番号:122-97-4)は、スパイス様の香気を有し、香料、保存料等として使用される。
アンドロスタジエノン(CAS番号:4075-07-4)は、フェロモン作用を示すことが知られており、特有の香気を有し香水に調合されることもあるステロイドである。
アンドロステロン(CAS番号:453-41-8)は、体臭の原因物質にもなる男性ホルモンであるが、その特有の匂いのため香水に調合されることもあるステロイドである。
酪酸(CAS番号:107-92-6)は、特有の不快臭を有し、天然に広く分布する物質であるが、その特有の匂いのため香料に含まれることもある。
プロパン酸3-フェニルプロピル、シトロネロール、フェネチルアルコール、αシンナミルアルコール、シクラメンアルデヒド、ゲラニオール、αイオノン、ネロール、ノナデカン、リナロール、リラール、フェニルプロピルアルコール、アンドロスタジエノン、アンドロステロン、及び酪酸は、嗅覚受容体の一つであるOR10A6のアゴニストとして作用することが知られている。これらの物質は、主に匂い物質やフェロモン物質として知られているが、本発明者らにより角層形成促進、肌荒れ抑制、保湿、皮膚バリア機能改善、及びメラニン産生抑制作用といった皮膚に好ましい作用を発揮することが見出された。とりわけ、かかる作用は、OR10A6の活性化を介して発揮される。これらの成分は、市販のものを用いても合成してもよい。
また、上記以外の成分であっても、OR10A6を活性化することができれば角層形成促進、肌荒れ抑制、保湿、皮膚バリア機能改善、及びメラニン産生抑制作用等の皮膚に好ましい作用を発揮することができると考えられる。更に、OR5P2およびOR5P3もOR10A6と同様に角化細胞において発現することが発見されたため、これらの嗅覚受容体も活性化させることにより角層形成促進、肌荒れ抑制、保湿、皮膚バリア機能改善、及びメラニン産生抑制作用といった皮膚に好ましい作用を発揮することが期待される。従って、本発明は、角層形成促進、肌荒れ抑制、保湿、皮膚バリア機能改善、及びメラニン産生抑制のためのOR10A6活性化剤、OR5P2活性化剤、又はOR5P3活性化剤も提供する。OR10A6活性化剤の例としては、プロパン酸3-フェニルプロピル、シトロネロール、フェネチルアルコール、αシンナミルアルコール、シクラメンアルデヒド、ゲラニオール、αイオノン、ネロール、ノナデカン、リナロール、リラール、フェニルプロピルアルコール、アンドロスタジエノン、アンドロステロン、及び酪酸等のOR10A6アゴニスト作用を有する物質を有効成分として含む剤が挙げられる。中でも、プロパン酸3-フェニルプロピル、シトロネロール、フェネチルアルコールが好ましく、特にプロパン酸3-フェニルプロピルが好ましく使用できる。OR5P3活性化剤の例としては、1-オクタノール、アセトフォノン、及びクマリン等のOR5P3アゴニスト作用を有する物質を有効成分として含む剤が挙げられる。
OR10A6、OR5P2、又はOR5P3は、嗅覚受容体の一種であり、それぞれOR10A6、OR5P2、又はOR5P3遺伝子によってコードされるタンパク質である。嗅覚受容体は、鼻腔内の嗅細胞の繊毛等に存在し、匂い分子が結合することにより、細胞のイオンチァネルが開かれ、ナトリウムイオンが細胞内へ流入すると活動電位が発生し、これにより匂い情報が脳へと伝達され匂いの知覚を引き起こす。上述のようにいくつかの嗅覚受容体では、鼻以外の組織にも存在し、匂い情報の伝達以外の作用にも関与することが報告されているが、本発明者らにより、OR10A6、OR5P2、又はOR5P3が皮膚にも存在すること、更にはOR10A6の活性化により角層形成促進、肌荒れ抑制、保湿、皮膚バリア機能改善、及びメラニン産生抑制作用等の皮膚に好ましい作用を発揮することが発見された。
一態様では、OR10A6、OR5P2、又はOR5P3の活性化とは、例えば、生体試料におけるOR10A6、OR5P2、又はOR5P3のmRNA量又はタンパク質量が、本発明のOR10A6、OR5P2、又はOR5P3活性化剤を添加すると、添加しない場合と比べて増加することを指す。例えば、増加は、有意水準を5%とした統計学的有意差(例えばスチューデントのt検定)を有する増加、及び/又は、例えば10%以上、20%以上、30%以上、40%以上、50%以上、60%以上、70%以上、80%以上、90%以上、100%以上、またはそれ以上の増加を意味し得る。
OR10A6、OR5P2、又はOR5P3発現の測定は、生体試料におけるOR10A6、OR5P2、又はOR5P3のmRNA量又はタンパク質量を測定することにより決定することができる。mRNA量の測定としては、定量的PCRやノーザンブロティングなど本技術分野に既知の手法を用いて行うことができる。例えば、OR10A6、OR5P2、又はOR5P3のmRNAに対するプローブを用いてもよい。タンパク質量については、ウエスタンブロッティング、免疫染色、ICMなどの本技術分野に既知の手法を用いて行うことができる。
一態様では、OR10A6、OR5P2、又はOR5P3の活性化は、それぞれ各嗅覚受容体にプロパン酸3-フェニルプロピル、シトロネロール、フェネチルアルコール、αシンナミルアルコール、シクラメンアルデヒド、ゲラニオール、αイオノン、ネロール、ノナデカン、リナロール、リラール、フェニルプロピルアルコール、アンドロスタジエノン、アンドロステロン、酪酸、1-オクタノール、アセトフォノン、クマリン、その他匂い分子といったOR10A6、OR5P2、又はOR5P3に対する活性化作用を有する物質(例えば、上記各嗅覚受容体に対するアゴニスト等)が結合することにより、惹起される。OR10A6、OR5P2、又はOR5P3の活性化は、例えば、生体試料におけるこれらの受容体が存在する細胞内へのNa+、K+、又はCa2+等の陽イオンの流入により測定することができる。例えば、候補物質を添加すると添加前に比べて細胞内Ca2+といった陽イオンが増加する場合、OR10A6、OR5P2、又はOR5P3活性化作用があると判断できる。例えば、陽イオン濃度の増加は、候補物質を添加した場合に、例えば有意水準を5%とした統計学的有意差(例えば、ANOVA分散分析、Scheffe法等)をもって増加していること、あるいは、例えば10%以上、20%以上、30%以上、40%以上、50%以上、60%以上、70%以上、80%以上、90%以上、100%以上、またはそれ以上増加することを意味し得る。陽イオンの流入は、限定されないものの、蛍光顕微鏡などを用いる細胞内イメージング等を行うことによって測定してもよい。
対照のOR10A6、OR5P2、又はOR5P3活性化作用に比べて、候補物質を添加した場合の活性化作用が増加した場合、候補物質がOR10A6、OR5P2、又はOR5P3活性化作用を有するものと決定することもできる。対照のOR10A6、OR5P2、又はOR5P3活性化作用は、対照薬剤を添加した実験系や候補物質のみを含まない以外は同じ条件の実験系で決定されてもよいし、shRNAやsiRNAによるRNAi等を介したOR10A6、OR5P2、又はOR5P3ノックダウン細胞や、TALEN、CRISPRによるゲノム編集等を介するノックアウト細胞を含む実験系で決定されてもよい。
生体試料とは、OR10A6、OR5P2、又はOR5P3活性が測定可能な細胞を含む試料であればよく、角化細胞以外の細胞が含まれていてもよい。角化細胞以外の細胞としては、嗅細胞等のOR10A6、OR5P2、又はOR5P3の存在が確認される細胞、あるいは、OR10A6、OR5P2、又はOR5P3を有するように作成される細胞等が挙げられる。生体試料は、任意の動物由来であってよいが、化粧料や医薬品の開発の観点からはヒト由来が好ましい。生体試料としては、上記嗅覚受容体を測定可能な細胞、例えば、角化細胞又はその他のOR10A6、OR5P2、又はOR5P3活性が測定可能な細胞の培養物であってもよいし、皮膚試料であってもよいし、3次元に構築された培養皮膚モデルを用いてもよい。
角層形成促進、肌荒れ抑制、保湿、及び皮膚バリア機能改善作用は、例えば、角化外膜(CEと称することがある)を測定することにより確認できる。例えば、角化外膜の産生量は、角化細胞をβ-メルカプトエタノール等のアルコールやSDS等の界面活性化剤等を含む細胞溶解液等により溶解させ、溶液中で溶解せずに残った角化外膜の数をカウントすることにより求められる。例えば、このようにして測定された角化外膜の産生量が、候補物質を添加した場合の神経活性化作用が対照に比べて増加した場合、候補物質が角層形成促進、肌荒れ抑制、保湿、及び皮膚バリア機能改善作用を有するものと決定することができる。例えば、角化外膜の産生量の増加は、候補物質を添加した場合に、対照に比べて例えば有意水準を5%とした統計学的有意差(例えば、t検定、ダネットの検定等)をもって増加していること、あるいは、例えば10%以上、20%以上、30%以上、40%以上、50%以上、60%以上、70%以上、80%以上、90%以上、100%以上、又はそれ以上増加することを意味し得る。対照の角化外膜の産生量は、対照薬剤を添加した実験系や候補物質のみを含まない以外は同じ条件の実験系で決定されていてもよいし、OR10A6、OR5P2、又はOR5P3ノックダウン細胞やノックアウト細胞を含む実験系で決定してもよい。角層形成促進、肌荒れ抑制、保湿、及び皮膚バリア機能改善作用は、角化細胞の増殖マーカー、目視検査、TEWL、特定のタンパクや脂質の測定等のCE測定以外の方法によって確認することもできる。例えば、TGM1(Transglutaminase-1)、FLG(Filaggrin)、HAS3(Hyaluronan Synthase3)、GBA(β-Glucocerebrosidase)、IL-1α(Interleukin 1 Alpha)等の遺伝子発現量やタンパク質産生量を測定することにより確認することができる。メラニン産生抑制作用は、IL-1α、COX2等のメラニン産生に関与する遺伝子のmRNA量又はタンパク質量を測定することにより決定することができる。mRNA量の測定としては、定量的PCRやノーザンブロティングなど本技術分野に既知の手法を用いて行うことができる。例えば、TGM1、FLG、HAS3、GBA、IL-1α等のmRNAに対するプローブを用いてもよい。タンパク質量については、ウエスタンブロッティング、免疫染色、ICMなどの本技術分野に既知の手法を用いて行うことができる。例えば、角層形成促進、肌荒れ抑制、保湿、又は皮膚バリア機能改善は、例えば、生体試料におけるTGM1、FLG、HAS3、GBAのmRNA量又はタンパク質量が増加すること、及び/又はIL-1αのmRNA量又はタンパク質量が減少することにより確認できる。例えば、増加は、有意水準を5%とした統計学的有意差(例えばスチューデントのt検定、ダネットの検定)を有する増加、及び/又は、例えば10%以上、20%以上、30%以上、40%以上、50%以上、60%以上、70%以上、80%以上、90%以上、100%以上、またはそれ以上の増加を意味し得る。例えば、減少は、有意水準を5%とした統計学的有意差(例えばスチューデントのt検定、ダネットの検定)を有する減少、及び/又は、例えば10%以上、20%以上、30%以上、40%以上、50%以上、60%以上、70%以上、80%以上、90%以上、又は100%以上の減少を意味し得る。
OR10A6、OR5P2、又はOR5P3ノックダウン細胞とは、例えば、OR10A6、OR5P2、又はOR5P3遺伝子にノックダウン処置が施され、OR10A6、OR5P2、又はOR5P3の発現が損なわれた又は低減した細胞を指す。ノックダウン処置は、例えば、shRNAやsiRNAによるRNAi等の任意の公知技術を用いることができる。ノックダウン処置によりOR10A6、OR5P2、又はOR5P3発現が損なわれた又は低減したとは、例えば、ノックダウン処置を施した細胞のOR10A6、OR5P2、又はOR5P3遺伝子の発現量及び/又はOR10A6、OR5P2、又はOR5P3タンパク質量が、非ノックダウン細胞に比べて、例えば有意水準を5%とした統計学的有意差(例えば、t検定等)をもって低減していること、あるいは、例えば5%以上、10%以上、20%以上、30%以上、40%以上、50%以上、60%以上、70%以上、80%以上、90%以上、又は100%低減することを意味し得る。
OR10A6、OR5P2、又はOR5P3ノックアウト細胞とは、例えば、OR10A6、OR5P2、又はOR5P3遺伝子にノックアウト処置が施され、OR10A6、OR5P2、又はOR5P3遺伝子の機能が完全に除去された細胞を指す。ノックアウト処置は、例えば、TALEN、CRISPRによるゲノム編集等の任意の公知技術を用いることができる。
本発明のある実施形態は、プロパン酸3-フェニルプロピル、シトロネロール、フェネチルアルコール、αシンナミルアルコール、シクラメンアルデヒド、ゲラニオール、αイオノン、ネロール、ノナデカン、リナロール、リラール、フェニルプロピルアルコール、アンドロスタジエノン、アンドロステロン、及び酪酸から選ばれる1種又は複数、OR10A6活性化剤、OR5P2活性化剤、又はOR5P3活性化剤(以下、包括的に本発明の剤と称する場合がある)、あるいはそれを含む組成物を適用することを含む、対象における角層形成促進、肌荒れ抑制、保湿、皮膚バリア機能改善、又はメラニン産生抑制のための方法も提供する。本発明は、対象のOR10A6、OR5P2、又はOR5P3を活性化させることを含む、対象における角層形成促進、肌荒れ抑制、保湿、皮膚バリア機能改善、又はメラニン産生抑制のための方法も提供する。当該実施形態の一態様では、対象のOR10A6、OR5P2、又はOR5P3の活性化は、プロパン酸3-フェニルプロピル、シトロネロール、フェネチルアルコール、αシンナミルアルコール、シクラメンアルデヒド、ゲラニオール、αイオノン、ネロール、ノナデカン、リナロール、リラール、フェニルプロピルアルコール、アンドロスタジエノン、アンドロステロン、酪酸、1-オクタノール、アセトフォノン、及びクマリン、又は本発明の剤、あるいはそれを含む組成物を適用することにより達成される。
本発明の方法を適用する対象は、例えば、角層形成能が低下している対象、肌荒れを有するか又は予防又は軽減を望む対象、肌の乾燥等を有し保湿が必要であるか又は保湿を望む対象、皮膚バリア機能が低下しているか又は皮膚バリア機能改善を希望する対象、例えば、日焼け、しみ、くすみが気になる対象、美白を希望する対象等のメラニン産生抑制が必要又はメラニン産生抑制を希望する対象、OR10A6、OR5P2、又はOR5P3の活性が低下している対象、あるいは、嗅覚受容体の活性が低下している対象、例えば加齢などの原因により嗅覚が低下している対象等が挙げられる。
本願にかかる方法は、美容目的であり、医師や医療従事者が行う医療行為は除かれることがある。また、本願にかかる方法は、対象の美容行為を支援する方法であってもよい。
好ましい一実施形態は、プロパン酸3-フェニルプロピルを適用することを含む、対象における角層形成促進、肌荒れ抑制、保湿、皮膚バリア機能改善、又はメラニン産生抑制のための美容方法である。かかる実施形態の一態様では、角層形成促進、肌荒れ抑制、保湿、皮膚バリア機能改善、又はメラニン産生抑制は、対象のOR10A6を活性化させることにより達成される。
さらに本発明は、プロパン酸3-フェニルプロピル、シトロネロール、フェネチルアルコール、αシンナミルアルコール、シクラメンアルデヒド、ゲラニオール、αイオノン、ネロール、ノナデカン、リナロール、リラール、フェニルプロピルアルコール、アンドロスタジエノン、アンドロステロン、酪酸1-オクタノール、アセトフォノン、及びクマリンから選ばれる1種又は複数、又は本発明の剤を含む、角層形成促進、肌荒れ抑制、保湿、皮膚バリア機能改善、及び/又はメラニン産生抑制のための組成物も提供する。
本発明の剤又は組成物の適用は、経皮、経口、経粘膜、経鼻、静脈内、動脈内、皮下など任意の経路で適用することができるが、皮膚に作用させる観点から経皮投与が好ましい。経皮的に投与する場合、任意の皮膚、例えば顔面、頭部、頸部、四肢、体幹の皮膚に適用することができる。
本発明の剤は、それぞれ化粧料、医薬品又は医薬部外品に配合されてもよい。これらの薬剤は、経口、又は非経口、例えば経皮的に適用されてもよい。経皮的な適用の場合、皮膚外用剤に剤形することができる。皮膚外用剤は、皮膚に適用可能であれば特に限定されず、例えば、溶液状、乳化状、固形状、半固形状、粉末状、粉末分散状、水-油二層分離状、水-油-粉末三層分離状、軟膏状、ゲル状、エアゾール状、ムース状、スティック状等、任意の剤型が適用できる。皮膚外用剤に剤形される場合には、皮膚外用剤に通常用いられる基剤、及び賦形剤、例えば保存剤、乳化剤、pH調整剤などが用いられてもよい。化粧料に配合する場合、化粧水、乳液、美容液、クリーム、ローション、パック、エッセンス、ジェル等の顔用又は体用の化粧料や、ファンデーション、化粧下地、コンシーラー等のメーキャップ化粧料、さらには浴用剤などに配合することができる。本発明の剤を含む化粧品を用いることにより、例えばOR10A6、OR5P2、又はOR5P3の活性化を介して、角層形成促進、肌荒れ抑制、保湿、皮膚バリア機能改善、又はメラニン産生抑制をもたらす。したがって、本発明の剤は、角層形成促進、肌荒れ抑制、保湿、皮膚バリア機能改善、又はメラニン産生抑制を目的とした化粧料に使用されてもよい。
本発明の剤又は組成物における有効成分の量は、角層形成促進、肌荒れ抑制、保湿、皮膚バリア機能改善、又はメラニン産生抑制作用を発揮させる観点で任意に選択することができる。例えば、皮膚外用剤として配合する観点からは、プロパン酸3-フェニルプロピル、シトロネロール、フェネチルアルコール、αシンナミルアルコール、シクラメンアルデヒド、ゲラニオール、αイオノン、ネロール、ノナデカン、リナロール、リラール、フェニルプロピルアルコール、アンドロスタジエノン、アンドロステロン、酪酸、1-オクタノール、アセトフォノン、クマリン、又はその他OR10A6、OR5P2、又はOR5P3を活性化することができる有効成分を0.0005~100.0mMで配合することができる。効果を十分に発揮させる観点から、好ましくは0.05mM以上で配合でき、さらに好ましくは0.5mM以上、例えば1.0mMで配合することができる。一方、強いにおいを避ける観点から、好ましくは10.0mM以下で配合することができ、さらに好ましくは5.0mM以下で配合してもよい。上記成分を任意の比率で組み合わせてもよく、その場合、それらの成分の合計量が上記範囲内にあることが好ましい。
また、本発明は、OR10A6、OR5P2、又はOR5P3活性を指標とした角層形成促進剤、肌荒れ抑制剤、保湿剤、又は皮膚バリア機能改善剤、又はメラニン産生抑制剤のスクリーニング方法を提供する。
一態様では、本発明のスクリーニング方法は、以下の工程:
生体試料におけるOR10A6、OR5P2、又はOR5P3活性を測定すること、および
対照に比較してOR10A6、OR5P2、又はOR5P3活性が増加した場合に、候補物質を角層形成促進剤、肌荒れ抑制剤、保湿剤、皮膚バリア機能改善剤、又はメラニン産生抑制剤として決定することを含む。
生体試料におけるOR10A6、OR5P2、又はOR5P3活性を測定すること、および
対照に比較してOR10A6、OR5P2、又はOR5P3活性が増加した場合に、候補物質を角層形成促進剤、肌荒れ抑制剤、保湿剤、皮膚バリア機能改善剤、又はメラニン産生抑制剤として決定することを含む。
候補物質としては、化粧品素材ライブラリー、エキスライブラリー、医薬品ライブラリー、化合物ライブラリーなど任意のライブラリーに属する物質を使用することができる。例えば、嗅覚受容体に反応する匂い物質や香料原料等が挙げられるがこれらに限定されない。
さらに本発明は、角層形成促進、肌荒れ抑制、保湿、皮膚バリア機能改善、又はメラニン産生抑制のためのプロパン酸3-フェニルプロピル、シトロネロール、フェネチルアルコール、αシンナミルアルコール、シクラメンアルデヒド、ゲラニオール、αイオノン、ネロール、ノナデカン、リナロール、リラール、フェニルプロピルアルコール、アンドロスタジエノン、アンドロステロン、酪酸、1-オクタノール、アセトフォノン、又はクマリン等のOR10A6、OR5P2、又はOR5P3に対するアゴニストも提供し、ここで、好ましくは角層形成促進、肌荒れ抑制、保湿、皮膚バリア機能改善、又はメラニン産生抑制はOR10A6、OR5P2、又はOR5P3を活性化することにより達成される。好ましい実施形態は、角層形成促進、肌荒れ抑制、保湿、皮膚バリア機能改善、又はメラニン産生抑制のためのプロパン酸3-フェニルプロピルであり、ここで角層形成促進、肌荒れ抑制、保湿、皮膚バリア機能改善、又はメラニン産生抑制はOR10A6を活性化することにより達成される。
さらに、本発明は、角層形成促進、肌荒れ抑制、保湿、皮膚バリア機能改善、又はメラニン産生抑制のための薬剤の製造におけるプロパン酸3-フェニルプロピル、シトロネロール、フェネチルアルコール、αシンナミルアルコール、シクラメンアルデヒド、ゲラニオール、αイオノン、ネロール、ノナデカン、リナロール、リラール、フェニルプロピルアルコール、アンドロスタジエノン、アンドロステロン、酪酸等のOR10A6に対するアゴニスト、OR5P2に対するアゴニスト、又は1-オクタノール、アセトフォノン、クマリン等のOR5P3に対するアゴニストの使用も提供する。

以下に説明する本発明の実施例は例示のみを目的とし、本発明の技術的範囲を限定するものではない。本発明の技術的範囲は特許請求の範囲の記載によってのみ限定される。本発明の趣旨を逸脱しないことを条件として、本発明の変更、例えば、本発明の構成要件の追加、削除及び置換を行うことができる。
実施例1:角化細胞におけるOR10A6、OR5P2、及びOR5P3の発現
培養ケラチノサイト(販売元:クラボウ)を、EPILIFE-KG2培地(販売元:クラボウ)で、37℃加湿雰囲気下で培養し、コンフルエントになった培養物を回収し、Trizol(販売元:NIPPON GENE)を用いて、cDNAをVILO Master Mix(販売元:Invitrogen)の製品説明書にしたがって調製した。以下に示す配列のOR10A6増幅用プライマー(配列番号1,2)、OR5P2増幅用プライマー(配列番号3,4)、OR5P3増幅用プライマー(配列番号5,6)及び内部標準のGAPDH増幅用プライマー(配列番号7,8)用いて、リアルタイムPCRを行い増幅産物を電気泳動により確認した。

結果を図1に示す。OR10A6、OR5P2、及びOR5P3はいずれも角化細胞において発現することが確認された。そこで、以下の実験によりこれらの嗅覚受容体の一つであるOR10A6およびそのアゴニストである3PPPを使用して角化細胞における反応を調べた。
実施例2:3PPPによる細胞のカルシウム応答観察
培養角化細胞(販売元:クラボウ)を、EPILIFW-KG2培地(販売元:クラボウ)またはOR10A6 siRNA(販売元:Horizon)を含むEPILIFW-KG2培地で、37℃加湿雰囲気下で1日間培養した。コンフルエントになった細胞に対し、Fura2 AM(販売元:life technology)を5μMで含むEPILIFW-KG2に置換し1時間培養した。その後、NaCl 150mM、KCl 5 mM、CaCl2 1.8mM、MgCl2 1.2mM、HEPES 25mM、D-glucose 10mMを含むバッファー(pH7.4)に置換し、3-Phenylpropyl propionate(3PPP)(販売元:SigmaAldrich)を0.05~1mMの濃度で細胞に添加した。3PPPを添加した際の細胞内Ca2+濃度変化を、蛍光顕微鏡(販売元:ニコン社)により340nmと380nmの波長を観察することで、各時点における蛍光強度比(F340/380)として測定した。
培養角化細胞(販売元:クラボウ)を、EPILIFW-KG2培地(販売元:クラボウ)またはOR10A6 siRNA(販売元:Horizon)を含むEPILIFW-KG2培地で、37℃加湿雰囲気下で1日間培養した。コンフルエントになった細胞に対し、Fura2 AM(販売元:life technology)を5μMで含むEPILIFW-KG2に置換し1時間培養した。その後、NaCl 150mM、KCl 5 mM、CaCl2 1.8mM、MgCl2 1.2mM、HEPES 25mM、D-glucose 10mMを含むバッファー(pH7.4)に置換し、3-Phenylpropyl propionate(3PPP)(販売元:SigmaAldrich)を0.05~1mMの濃度で細胞に添加した。3PPPを添加した際の細胞内Ca2+濃度変化を、蛍光顕微鏡(販売元:ニコン社)により340nmと380nmの波長を観察することで、各時点における蛍光強度比(F340/380)として測定した。
結果を図2に示す。上図に示されるように、3PPPを添加するとCa2+濃度が3PPPを添加する前の値に対し大きく上昇した。また、そのCa2+濃度の上昇は3PPPの濃度が0.05mM、0.5mM、1.0mMと上昇するにつれ、有意に高くなり濃度に依存することもわかる。更に、下図により3PPPによる反応はOR10A6ノックダウン角化細胞では有意に低くなることが確認された。以上の実験により角化細胞に存在するOR10A6がそのアゴニストである3PPPにより活性化されたことがわかる。そこで、以下の実験によりOR10A6を3PPPにより活性化することによる皮膚への影響を調べた。
実施例3:OR10A6活性化による角層形成促進、肌荒れ抑制、保湿、及び皮膚バリア機能改善作用1
培養角化細胞(販売元:クラボウ)を、EPILIFW-KG2培地(販売元:クラボウ)またはOR10A6 siRNA(販売元:Horizon)を含むEPILIFW-KG2培地で、37℃加湿雰囲気下で1日間培養した。コンフルエントになった細胞に対し、3-Phenylpropyl propionate(3PPP)(販売元:SigmaAldrich)を1mMの濃度で細胞に添加しさらに3日間培養した。3日後培養細胞をトリプシン(販売元:ナカライテスク株式会社)処理により回収し、光学顕微鏡で全細胞数をカウントし、更にトリパンブルー(販売元:ナカライテスク株式会社)により染色された死細胞数をカウントすることにより、生細胞数を全細胞数-死細胞数として求めた。その後細胞を遠心後β-メルカプトエタノール1%、SDS(ラウリル硫酸ナトリウム)1%、20mMトリスHCl(pH7.5)を含むバッファーに置換し95℃、15分で細胞を溶解させ、溶液中に溶解せずに残った角化外膜を有する角化細胞の数を光学顕微鏡でカウントした。角層形成促進、肌荒れ抑制、保湿、皮膚バリア機能改善作用の指標として角化外膜の産生量を角化外膜を有する角化細胞の数/生細胞数×100として算出し、その値をCE産生率(%)として示した。
培養角化細胞(販売元:クラボウ)を、EPILIFW-KG2培地(販売元:クラボウ)またはOR10A6 siRNA(販売元:Horizon)を含むEPILIFW-KG2培地で、37℃加湿雰囲気下で1日間培養した。コンフルエントになった細胞に対し、3-Phenylpropyl propionate(3PPP)(販売元:SigmaAldrich)を1mMの濃度で細胞に添加しさらに3日間培養した。3日後培養細胞をトリプシン(販売元:ナカライテスク株式会社)処理により回収し、光学顕微鏡で全細胞数をカウントし、更にトリパンブルー(販売元:ナカライテスク株式会社)により染色された死細胞数をカウントすることにより、生細胞数を全細胞数-死細胞数として求めた。その後細胞を遠心後β-メルカプトエタノール1%、SDS(ラウリル硫酸ナトリウム)1%、20mMトリスHCl(pH7.5)を含むバッファーに置換し95℃、15分で細胞を溶解させ、溶液中に溶解せずに残った角化外膜を有する角化細胞の数を光学顕微鏡でカウントした。角層形成促進、肌荒れ抑制、保湿、皮膚バリア機能改善作用の指標として角化外膜の産生量を角化外膜を有する角化細胞の数/生細胞数×100として算出し、その値をCE産生率(%)として示した。
結果を図3に示す。角化細胞におけるCE産生率は3PPPにより有意に増加した(左図)。しかしながら、OR10A6ノックアウト角化細胞では3PPPを添加しても角化外膜の産生量に有意な増加は見られなかった。更に、3PPP無添加の場合、CE産生率はOR10A6ノックアウト角化細胞ではOR10A6非ノックアウト角化細胞に比べて有意に低下した(右図)。したがって、OR10A6の活性化によりCE産生が促進されることが示唆される。
以上の結果により、OR10A6、OR5P2、及びOR5P3は角化細胞に存在し、3PPPによるOR10A6の活性化によりCE産生が促進されることがわかった。
実施例4:OR10A6活性化による角層形成促進、肌荒れ抑制、保湿、及び皮膚バリア機能改善作用2
実施例3により3PPPがOR10A6などの嗅覚受容体の活性化を介してCEの形成を促進する可能性が示されたが、本実施例では、OR10A6活性化によるCEの形成および成熟を促進する酵素であるトランスグルタミナーゼ1をコードするTGM1遺伝子の発現を測定した。
4-1:細胞の培養
HaCaT細胞を、24ウェルプレート(Cat No.3526,Corning,USA)に10.0×104cells/wellの密度で10.0%(v/v)Fetal Bovine Serum(FBS,Cat No.SH30071.03,Hyclone,UK)および1.0%(v/v)抗真菌剤(Antibiotic-Antimycotic 100X,Cat No.15240-062,Invitrogen,USA)を含むDulbecco’s Modified Eagle Medium(DMEM,Cat No.043-30085,Wako,Japan)に播種し、CO2インキュベータ内(CO2濃度 5%、37℃)で24時間培養した。培地を除去後、終濃度が0.05mMとなるように3PPPを添加した培地に交換しCO2インキュベータ内で更に48時間培養した。対照には3PPPを含まない培地を用いた。
実施例3により3PPPがOR10A6などの嗅覚受容体の活性化を介してCEの形成を促進する可能性が示されたが、本実施例では、OR10A6活性化によるCEの形成および成熟を促進する酵素であるトランスグルタミナーゼ1をコードするTGM1遺伝子の発現を測定した。
4-1:細胞の培養

4-2:RNA抽出・精製、定量および純度測定
PureLinkTM RNA Mini Kit(Cat No.12183018A,Invitrogen,USA)を用いてRNA抽出・精製を行い、精製したRNAの一部をUV透過性96ウェルプレートに分取してTris-EDTA Bufferにより10倍希釈し、マイクロプレートリーダー(SPARK(登録商標)10M TECAN,Switzerland)を用いて230nm、260nm、280nmの吸光度(OD230、OD260、OD280)を測定した。OD260を用いてRNA濃度を算出し、TE Bufferで希釈してRNA濃度を10μg/mLに調製した。

4-3:リアルタイムPCR法による遺伝子発現解析
SuperScript(商標)IV VILOTM Master Mix with ezDNase(Cat No.11766050,Invitrogen,USA)を用いてRNAの逆転写を行った。8連チューブに1ウェルあたり4μLのSuperScript(商標)IV VILOTM Master Mixおよび6μLのNuclease-free Waterを添加し、リアルタイムPCR(QuantStudio(登録商標)3,Applied Biosystems,USA)を用いて25℃で10分間、50℃で10分間、85℃で5分間加熱して、cDNAを合成した。PCRプレートに1ウェルあたり10μLのTaqMan(登録商標)Fast AdvancedMaster Mix(Cat No.4444557,Applied Biosystems,USA)、1μLのTaqMan Gene Expressior、7μLのUltraPureTM Distilled Water(Invitrogen,Cat No.10977-015,USA)および2μLのcDNAを加え、密封した。TGM1のプライマー(Hs00165929_m1)及び内部標準遺伝子としてGAPDHのプライマー(Hs02786624_g1)を用いReal-Time qPCRを行い、3PPP添加培地におけるTGM1のThreshold Cycle(Ct)値を算出後、GAPDHによりCt値を補正してΔCt値とし、かつ、1サイクル当たり遺伝子量が2倍になると仮定し、対照の遺伝子発現量を1とした場合の3PPP添加培地における遺伝子発現量を求めた。遺伝子発現量の解析には1つの処理群につき24ウェルプレート×3ウェルの平均値を用いた。

表1に示すように、3PPPを添加するとTGM1の遺伝子発現が有意に上昇し、3PPPがOR10A6を活性化しTGM1の産生を促進することによりCEの形成および成熟を促進され、角層形成促進、肌荒れ抑制、保湿、及び皮膚バリア機能改善に寄与する可能性が示唆された。
実施例5:OR10A6活性化による角層形成促進、肌荒れ抑制、保湿、及び皮膚バリア機能改善作用3
OR10A6活性化による角層形成、肌荒れ抑制、保湿、皮膚バリア改善効果を確認するために、これらの機能に関与する他の遺伝子を用いて以下の実験を行った。より具体的には、NMFとして皮膚の水分保持機能に関与するフィラグリンをコードするFLG(Filaggrin)、表皮細胞で産生され皮膚の水分保持に関与するヒアルロン酸の合成酵素をコードするHAS3(Hyaluronan Synthase 3)、及び皮膚のバリア機能に関わるセラミドEOP(セラミド1)の産生酵素をコードするGBA(β-Glucocerebrosidase)の発現量を測定した。
OR10A6活性化による角層形成、肌荒れ抑制、保湿、皮膚バリア改善効果を確認するために、これらの機能に関与する他の遺伝子を用いて以下の実験を行った。より具体的には、NMFとして皮膚の水分保持機能に関与するフィラグリンをコードするFLG(Filaggrin)、表皮細胞で産生され皮膚の水分保持に関与するヒアルロン酸の合成酵素をコードするHAS3(Hyaluronan Synthase 3)、及び皮膚のバリア機能に関わるセラミドEOP(セラミド1)の産生酵素をコードするGBA(β-Glucocerebrosidase)の発現量を測定した。
TGM1のプライマー(Hs00165929_m1)の代わりに、FLG(Hs00856927_g1)、Human Hyaluronan Synthase 3(HAS3,Assay ID.Hs00193436_m1)、Human β-Glucocerebrosidase(GBA,Assay ID.Hs00986836_g1)のプライマーを用いる以外は実施例4と同じ材料及び方法にて、0.05mMの3PPPを含む又は含まない培地におけるこれらの遺伝子発現量を測定した。
表2~4に示すように、3PPPを添加するとFLG、HAS3、GBAの遺伝子発現が有意に上昇した。以上の結果から、3PPP等によるOR10A6活性化により、TGM1の促進以外にもこれらの遺伝子の発現を促進することによっても、角層形成促進、肌荒れ抑制、保湿、及び皮膚バリア機能改善に寄与している可能性が示唆された。
実施例6:OR10A6活性化による角層形成促進、肌荒れ抑制、保湿、及び皮膚バリア機能改善作用4
3PPP以外のOR10A6活性化剤についても同様の効果がみられるかについて調べるために、OR10A6に反応する物質として知られているシトロネロール及びフェネチルアルコール(非特許文献5)を用い、これらの物質が、3PPPと同様に角層形成作用を有するかを検討した。より具体的には、3PPPの代わりにシトロネロール(CAS番号:106-22-9、和光037-05993)又はフェネチルアルコール(CAS番号:60-12-8、TCI P0084)を下記濃度で用いる以外は実施例4と同じ材料及び方法にてTGM1遺伝子発現量を測定した。
3PPP以外のOR10A6活性化剤についても同様の効果がみられるかについて調べるために、OR10A6に反応する物質として知られているシトロネロール及びフェネチルアルコール(非特許文献5)を用い、これらの物質が、3PPPと同様に角層形成作用を有するかを検討した。より具体的には、3PPPの代わりにシトロネロール(CAS番号:106-22-9、和光037-05993)又はフェネチルアルコール(CAS番号:60-12-8、TCI P0084)を下記濃度で用いる以外は実施例4と同じ材料及び方法にてTGM1遺伝子発現量を測定した。
表6の結果より、3PPPのみならず他のOR10A6活性化剤であるシトロネロールやフェネチルアルコールを用いてもTGM1の発現が有意に上昇した。
実施例7:OR10A6活性化による角層形成促進、肌荒れ抑制、保湿、及び皮膚バリア機能改善作用5
実施例6について効果がみられたシトロネロール及びフェネチルアルコールについて、実施例5と同様にGBAを用いて以下の実験を行った。より具体的には、3PPPの代わりにシトロネロール(CAS番号:106-22-9、和光037-05993)又はフェネチルアルコール(CAS番号:60-12-8、TCI P0084)を下記濃度で用いる以外は実施例5と同じ材料及び方法にてGBA遺伝子発現量を測定した。
実施例6について効果がみられたシトロネロール及びフェネチルアルコールについて、実施例5と同様にGBAを用いて以下の実験を行った。より具体的には、3PPPの代わりにシトロネロール(CAS番号:106-22-9、和光037-05993)又はフェネチルアルコール(CAS番号:60-12-8、TCI P0084)を下記濃度で用いる以外は実施例5と同じ材料及び方法にてGBA遺伝子発現量を測定した。
表7の結果より、実施例6と同様、3PPP以外のOR10A6活性化剤であるシトロネロールやフェネチルアルコールでもGBAの発現が有意又は有意傾向で上昇した。
実施例8:OR10A6活性化による角層形成促進、肌荒れ抑制、保湿、及び皮膚バリア機能改善作用5
実施例6、7について効果がみられたシトロネロールについて、実施例5と同様にHAS3を用いて以下の実験を行った。より具体的には、3PPPの代わりにシトロネロール(CAS番号:106-22-9、和光037-05993)を下記濃度で用いる以外は実施例5と同じ材料及び方法にてHAS3遺伝子発現量を測定した。
実施例6、7について効果がみられたシトロネロールについて、実施例5と同様にHAS3を用いて以下の実験を行った。より具体的には、3PPPの代わりにシトロネロール(CAS番号:106-22-9、和光037-05993)を下記濃度で用いる以外は実施例5と同じ材料及び方法にてHAS3遺伝子発現量を測定した。
実施例9:
以上により、OR10A6を活性化することにより角層形成促進、肌荒れ抑制、保湿、及び皮膚バリア機能改善作用を奏することが示唆されたが、他の効果について確認するために炎症性サイトカインでありメラニン産生を促進することが知られているIL-αをコードするIL-1α(Interleukin 1 Alpha)の発現量を測定した。より具体的には、TGM1のプライマー(Hs00165929_m1)の代わりに、IL-1αのプライマー(Hs00174092_m1)を用いる以外は実施例4と同じ材料及び方法にて、下記濃度の3PPPを含む又は含まない培地におけるIL-1αの遺伝子発現量を測定した。IL-1α(Interleukin 1 Alpha)は、炎症性サイトカインであるIL-1αタンパクをコードする遺伝子であり、IL-1αはチロシナーゼ活性を高めることによりメラニンの過剰生成を引き起こすことが知られている。
以上により、OR10A6を活性化することにより角層形成促進、肌荒れ抑制、保湿、及び皮膚バリア機能改善作用を奏することが示唆されたが、他の効果について確認するために炎症性サイトカインでありメラニン産生を促進することが知られているIL-αをコードするIL-1α(Interleukin 1 Alpha)の発現量を測定した。より具体的には、TGM1のプライマー(Hs00165929_m1)の代わりに、IL-1αのプライマー(Hs00174092_m1)を用いる以外は実施例4と同じ材料及び方法にて、下記濃度の3PPPを含む又は含まない培地におけるIL-1αの遺伝子発現量を測定した。IL-1α(Interleukin 1 Alpha)は、炎症性サイトカインであるIL-1αタンパクをコードする遺伝子であり、IL-1αはチロシナーゼ活性を高めることによりメラニンの過剰生成を引き起こすことが知られている。
表10に示すように、3PPPを添加するとIL-αの遺伝子発現が濃度依存的に減少した。この結果から、OR10A6の活性化によりTGM1、FLG、HAS3、GBA発現促進による角層形成促進、肌荒れ抑制、保湿、及び皮膚バリア機能改善のみならず、IL-αの発現抑制によって肌荒れ抑制やメラニン産生抑制にも寄与している可能性が示唆された。
以上の結果により、OR10A6、OR5P2、及びOR5P3の活性化により角層形成促進、肌荒れ抑制、保湿、皮膚バリア機能改善、メラニン産生抑制作用等の皮膚に好ましい影響を及ぼし、これらの効果はOR10A6、OR5P2、OR5P3を活性化させる成分等により達成されることが期待される。例えば、OR10A6を活性化させる成分としては、プロパン酸3-フェニルプロピル、シトロネロール、フェネチルアルコール、αシンナミルアルコール、シクラメンアルデヒド、ゲラニオール、αイオノン、ネロール、ノナデカン、リナロール、リラール、フェニルプロピルアルコール、アンドロスタジエノン、アンドロステロン、酪酸、OR5P3を活性化させる成分としては、1-オクタノール、アセトフォノン、及びクマリン等が知られているため、これらの成分による効果が期待できる。
Claims (9)
- OR10A6活性化剤を有効成分として含有する、角層形成促進剤。
- プロパン酸3-フェニルプロピル、シトロネロール、フェネチルアルコール、αシンナミルアルコール、シクラメンアルデヒド、ゲラニオール、αイオノン、ネロール、ノナデカン、リナロール、リラール、フェニルプロピルアルコール、アンドロスタジエノン、アンドロステロン、及び酪酸から選ばれる1種又は複数を有効成分として含有する、角層形成促進剤。
- 請求項1又は2の角層形成促進剤を含有する、角層形成を促進するための組成物。
- プロパン酸3-フェニルプロピル、シトロネロール、フェネチルアルコール、αシンナミルアルコール、シクラメンアルデヒド、ゲラニオール、αイオノン、ネロール、ノナデカン、リナロール、リラール、フェニルプロピルアルコール、アンドロスタジエノン、アンドロステロン、及び酪酸から選ばれる1種又は複数を有効成分として含有する、肌荒れ抑制剤。
- プロパン酸3-フェニルプロピル、シトロネロール、フェネチルアルコール、αシンナミルアルコール、シクラメンアルデヒド、ゲラニオール、αイオノン、ネロール、ノナデカン、リナロール、リラール、フェニルプロピルアルコール、アンドロスタジエノン、アンドロステロン、及び酪酸から選ばれる1種又は複数を有効成分として含有する、保湿剤。
- プロパン酸3-フェニルプロピル、シトロネロール、フェネチルアルコール、αシンナミルアルコール、シクラメンアルデヒド、ゲラニオール、αイオノン、ネロール、ノナデカン、リナロール、リラール、フェニルプロピルアルコール、アンドロスタジエノン、アンドロステロン、及び酪酸から選ばれる1種又は複数を有効成分として含有する、皮膚バリア機能改善剤。
- プロパン酸3-フェニルプロピル、αシンナミルアルコール、シクラメンアルデヒド、ゲラニオール、αイオノン、ノナデカン、リナロール、リラール、フェニルプロピルアルコール、アンドロスタジエノン、アンドロステロン、及び酪酸から選ばれる1種又は複数を有効成分として含有する、メラニン産生抑制剤。
- 角層形成促進、肌荒れ抑制、保湿、皮膚バリア機能改善、又はメラニン産生抑制のためのOR10A6活性化剤。
- OR10A6活性を指標とした角層形成促進剤、肌荒れ抑制剤、保湿剤、皮膚バリア機能改善剤、又はメラニン産生抑制剤のスクリーニング方法。
