La présente invention concerne une nouvelle souche bactérienne appartenant au genre Christensenella, une composition la comprenant et ses utilisations, notamment comme médicament dans le traitement et/ou la prévention de maladies telles que les maladies métaboliques et/ou les maladies inflammatoires.
Description
Souche bactérienne dans la prévention et/ou le traitement de pathologies inflammatoires

[0002] L'invention concerne une nouvelle souche bactérienne appartenant au genre Christensenella, une composition la comprenant et ses utilisations, notamment comme médicament dans le traitement et/ou la prévention de maladies telles que les maladies métaboliques et/ou les maladies inflammatoires.
[0003] Etat de l'art
[0004] Notre microbiote intestinal ou « flore intestinale » est constitué d'un ensemble de bactéries, virus, parasites et champignons non pathogènes, soit 1012 à 1014 micro-organismes présents dans le tube digestif de chaque individu, ce qui représente 2 à 10 fois plus que le nombre de cellules présent dans notre corps. Dès lors, son rôle est de plus en plus étudié ces dernières années. En 2012, un microbiologiste a identifié et cultivé à partir de fèces humaines une nouvelle espèce : Christensenella minuta, appartenant aux Clostridiales gram-négatifs. (Morotomi et al., Description of Christensenella minuta gen. nov., sp. nov, isolated from human faeces, which forms a distinct branch in the order Clostridiales, and proposal of Christensenellaceae fam. nov., International Journal of Systematic and Evolutionary Microbiology (2012), 62, 144-149).
[0005] Il est dorénavant admis par la communauté scientifique que le microbiote intestinal humain est lié à l'apparition de certaines pathologies dites « non transmissibles » et associées à une dysbiose du microbiote, telles que l'obésité, le diabète, les maladies métaboliques, les maladies inflammatoires intestinales, ou encore la dépression. Or, la prévalence de ces maladies ne cesse d'augmenter, sans aucun traitement satisfaisant.
[0006] Les maladies inflammatoires chroniques de l'intestin (MICI) ou Inflammatory Bowel Disease (IBD) en Anglais regroupent les maladies liées à l'inflammation de l'intestin à caractère chronique. La maladie de Crohn et la rectocolite hémorragique en sont les principales. Il s'agit de maladies récentes, multifactorielles dont l'origine est inconnue. Dans un certain nombre de cas, l'apparition de ces maladies pourrait être liée à une prédisposition génétique, à l'environnement, mais encore au microbiote. Ces maladies ont toutes la particularité de présenter une inflammation de l'intestin ce qui induit une modification du microbiote et
entretient alors l'inflammation.
[0007] Dans le cadre de la maladie de Crohn, l'inflammation est le plus souvent regroupée au niveau de l'iléon et du côlon. La rectocolite hémorragique (RCH) ou colite ulcéreuse affecte quant à elle plus particulièrement l'extrémité distale du tube digestif, c'est-à-dire le côlon et le rectum. Son incidence est restée stable, à 4,4 cas pour 100 000 habitants en France alors que celle de la maladie de Crohn a augmenté de 5,3 à 7,6 cas pour 100 000 habitants en France entre 1988 et 2014.
[0008] Ce sont des pathologies handicapantes pour lesquels aucun traitement curatif n'est disponible et dont la prévalence augmente fortement dans les pays occidentaux. Ces maladies ont la particularité d'agir par poussée dont la période de rémission est plus ou moins longue. L'objectif des traitements actuels est de prolonger la durée des rémissions le plus possible et donc d'améliorer la qualité de vie des patients malgré des traitements à vie.
[0009] Malgré des progrès dans la compréhension, et le diagnostic de ces pathologies, la médecine moderne n'a pas de solution curative satisfaisante, ce qui cause des problèmes de santé publique majeurs, notamment chez les adolescents dont la prévalence a fortement augmenté ces dernières années.
[0010] Il existe donc un besoin médical fort pour un produit capable d'agir sur le microbiote intestinal, en particulier dans la prévention et le traitement des maladies inflammatoires chroniques de l'intestin.
[0011] Un objectif de la présente invention est donc d'apporter une solution qui soit simple, efficace et économique pour restaurer un microbiote intestinal sain et donc traiter la dysbiose du microbiote intestinal, en particulier traiter les maladies inflammatoires chroniques, préférentiellement en évitant l'administration d'un traitement à vie et ainsi faire reculer la prévalence de ces maladies.
[0012] Résumé de l'invention
[0013] Pour répondre à cet objectif, les inventeurs ont identifié, parmi la multitude de bactéries présentes dans le microbiote intestinal et parmi les souches déjà connues appartenant à l'espèce Christensenella minuta, une nouvelle souche bactérienne de Christensenella minuta spécifique déposée auprès de l'institut Leibniz DSMZ (Deutsche Sammlung von Mikroorganismen und Zellkulturen GmbH, en français Collection allemande de micro-organismes et de cultures cellulaires GmbH) sous le numéro DSM 33715, particulièrement adaptée dans la prévention et/ou le traitement d'une dysbiose intestinale,
préférentiellement les maladies inflammatoires chroniques de l'intestin.
[0014] Ainsi, l'invention concerne une souche bactérienne de Christensenella minuta déposée sous le numéro DSM 33715 et ayant pour séquence d'ADNr 16S, la séquence SEQ ID NO :1, ainsi que toute souche comprenant une séquence nucléotidique ayant au moins 99,90% d'identité d'ANI (Average Nucleotide Identity - Identité Nucléotidique Moyenne) avec la séquence nucléotidique du génome de la souche bactérienne DSM 33715.
[0015] L'invention vise aussi le surnageant de la culture de la souche bactérienne selon l'invention mais également à une composition comprenant (i) au moins la souche bactérienne selon l'invention et/ou (ii) le surnageant de la culture selon l'invention et (iii) au moins un excipient acceptable. Ledit excipient acceptable étant préférentiellement un excipient pharmaceutiquement acceptable lorsqu'il s'agit d'un produit destiné à être utilisé comme médicament pour un usage humain ou un usage vétérinaire.
[0016] Enfin, l'invention est particulièrement adaptée pour utiliser ladite souche bactérienne, ledit surnageant ou ladite composition selon l'invention comme médicament. En effet, la présente invention est particulièrement adaptée dans la prévention et/ou le traitement de pathologies dites « non transmissibles » et associées à une dysbiose du microbiote intestinal ou maladies associées à une dysbiose du microbiote intestinal, en particulier les maladies telles que les maladies métaboliques, les maladies inflammatoires et d'autres qui seront détaillées ci-après.
[0017] Préférentiellement, l'invention vise à prévenir et/ou traiter les maladies inflammatoires chroniques de l'intestin, plus préférentiellement la maladie de Crohn, ou la colite ulcéreuse.
[0018] Enfin, l'invention concerne également l'utilisation non thérapeutique de la souche bactérienne selon l'invention, et/ou du surnageant selon l'invention ou de la composition selon l'invention pour maintenir et/ou renforcer le microbiote intestinal et/ou soutenir la diversité du microbiote intestinal et/ou promouvoir l'augmentation de bactéries bénéfiques dans le microbiote intestinal chez un sujet sain.

Chaque espèce bactérienne comprend une diversité de souches bactériennes. La souche bactérienne selon l'invention appartient à la famille Christensenellaceae, le genre Christensenella et l'espèce minuta. Des résultats sur un modèle pré-clinique de colite induite par du TNBS chez le rat permettent de confirmer les effets anti-inflammatoires de la souche, en
particulier sur les maladies inflammatoires chroniques de l'intestin.
[0051] Les inventeurs ont également démontré que ladite souche bactérienne DSM 33715 permettait de restaurer ou renforcer et donc préserver l'intégrité de la barrière intestinale. Enfin, une synergie d'action a été établie lorsque la souche est utilisée en combinaison avec des agents anti-inflammatoires utilisés en clinique tel que le Pentasa (5-ASA), le Tofacitinib ou le Budésonide, permettant ainsi d'envisager l'administration de doses moins importantes de tels agents anti-inflammatoires connus pour leur toxicité et leurs effets indésirables auprès des patients.

Enfin, les génomes d'intérêt sont comparés deux à deux pour calculer l'ANI.

La densité optique à 600 nm a été prise à Oh, 24h, 48h, 72h et 96h.
[0121] Les tests d'antibiorésistance ont été réalisé par l'utilisation de concentration minimale
inhibitrice (CMI) selon la méthode de référence de dilution en milieu gélosé du CLSI (Clinical and laboratory Standards Institute). Les solutions stock de chaque antibiotique ont été préparées et stérilisées par filtration à 0,2 p : ampicilline (Sigma-Aldrich), tétracycline (Sigma- Aldrich, Chloramphénicol (Sigma-Aldrich), clindamycine (Sigma-Aldrich), méropénème (USP), pipéracilline (USP), tazobactam (USP), et ceftriaxone (USP). Le milieu Brucella agar (BD BBL) supplémenté avec 5 pg/ml d'hémine (Sigma-Aldrich), 1 pg/ml de vitamine Kl (Sigma-Aldrich) et 5% (v/v) de sang de mouton hémolysé (laked) (Hemostat) a été utilisé comme milieu de test. Les boîtes agar avec différentes dilutions d'antibiotique ont été préparées et dégazées dans la chambre anaérobique avant inoculation. L'inoculum de DSM 33715 a été resuspendu au standard 0,5 de McFarland en eau peptonée. 2 pl de l'inoculum ont été déposés à la surface de l'agar (chaque spot correspondant approximativement à 105 CFU/ml). Un triplicat est réalisé par concentration d'antibiotique. Les boîtes ont ensuite été incubées pendant 6 jours à 37°C en condition anaérobique. Les valeurs de CMI ont été déterminées selon les directives du CLSI et ont été interprétées comme sensible, intermédiaire ou résistante, basées sur les points de rupture des anaérobes du CLSI.
[0122] Résultats de caractérisation
[0123] La souche DSM 33715 est strictement anaérobique avec une phase exponentielle de croissance entre 20 et 40h et une phase stationnaire commençant à 40h, non motile, non sporulante, négative pour l'activité oxydase et catalase et est gram négative. DSM 33715 est négative pour la majorité des enzymes testées, à l'exception de l'acidification du glucose, du xylose, du rhamnose et de l'arabinose, ce qui est cohérent avec la description des Christensenella minuta faite par Morotomi et al. Elle présente les activités enzymatiques suivantes : 0-Glucosidase, 0-Arabinosidase, Glutamic Acid Décarboxylase, ainsi qu'Estérase, Acid Phosphatase et Napthol-AS-BI-Phosphatase. Une résistance aux acides biliaires a été observée jusqu'à 80g/l d'Oxgall, correspondant à 80% de bile. DSM 33715 présente également une croissance en milieu avec un pH allant de 6 à 9. Enfin, la souche DSM 33715 montre une résistance à l'ampicilline et la tétracycline.
[0124] La Figure IA représente l'image d'une bactérie Christensenella minuta selon l'invention réalisée en microscopie électronique à transmission en coloration négative (grossissement : 36kX). Les dimensions moyennes d'une bactérie de la souche DSM 33715 sont : longueur 1,27 +- 0,28 mm ; largeur= 0,507 +- 0,04 mm ; avec une épaisseur de membrane= 29,4 nm (moyenne réalisée à partir de 41 bactéries). La souche a une forme courte avec les extrémités
effilées, se présentant en singulet, paires ou rosaces.

Pour 2 des 3 donneurs DSM 33715 induit une production d'IL-10 supérieur à une souche de référence de C. minuta DSM 22607 (Figure 2A). [0143] L'induction de production d'IL-10, une cytokine anti-inflammatoire, par DSM 33715 a également été mise en évidence par le modèle cellulaire THP-1 différencié en macrophage type MO par traitement à la PMA (Figure 2B).

Les cellules ont été lavées au PBS-1X (Gibco) et cultivées pendant 24h supplémentaires en milieu McCoy's 5A à 2% de SVF. DSM 22607 est la souche de référence. Le 5-ASA (20 mM) est un agent anti-inflammatoire utilisé en contrôle. Après 6h, les surnageants de culture cellulaire ont été collectés et stockés à - 80°C jusqu'à la quantification de l'IL-8 par ELISA. Le dosage de l'IL-8 a été effectué par ELISA spécifique de BioLegend, selon les instructions du fabricant. L'absorbance à 460 nm a été lue en utilisant le lecteur de microplaques FluoStar Omega, BMG Labtech. 6 expériences indépendantes ont été réalisées en duplicat. [0149] Pour évaluer l'effet immunomodulateur de la souche DSM 33715, les cellules HT-29 ont été stimulées avec la cytokine pro-inflammatoire TNF-a et la bactérie DSM 33715 à une MOI de 50 en début de phase stationnaire de croissance ou de son surnageant, pendant 6H. La chimiokine IL-8 est sécrétée par les HT-29 dans un état inflammatoire induit par le TNF-a (Ctrl). Les valeurs représentent la moyenne normalisée en pourcentage par rapport au contrôle + TNF-a (PBS-Glycérol pour la bactérie et le milieu de culture bactérien pour le surnageant), représentant la sécrétion d'IL-8 induite par stimulation au TNF-a, soit 100%. En présence de DSM 33715 ou de son surnageant, la sécrétion de l'IL-8 est diminuée de 50% par rapport au contrôle (p<0.0001, Dunnett's multiple comparisons test). Ainsi, la souche DSM 33715 et son surnageant présente un effet immunomodulateur anti-inflammatoire.
[0150] Exemple 4 : Effet anti-inflammatoire de la souche selon l'invention.
[0151] Dans la présente étude, les inventeurs ont étudié l'effet anti-inflammatoire de la souche DSM 33715 sur les voies de signalisation de l'inflammation NF-0B et JAK/STAT.
[0152] Les cellules HT-29 à une densité de 3xl05 cellules/puits, en plaque 24 puits, ont été transfectées selon un protocole de reverse-transfection, avec 200 ng de vecteur pRelA-luc (voie de signalisation NF-0B) ou pGL4 luc2P GAS-RE (voie de signalisation JAK/STAT
dépendante de l'IFN-y) et 10 ng de vecteur pRL-TK (Promega), utilisant le réactif de transfection X-tremeGENE HP DNA avec un ration 3:1 (Roche). Brièvement, les complexes de transfection réactif/ADN ont été préparés dans un volume final de 50 pl de milieu McCoy's 5A sans sérum et sans antibiotique, la quantité appropriée de chaque plasmide a été ajoutée et homogénéisée, puis l'agent de transfection a été ajouté (3:1). Les complexes de transfection ont été mélangés délicatement et incubés à température ambiante pendant 15 minutes. Après incubation, les complexes de transfection ont été ajoutés aux cellules HT-29 fraîchement resuspendue à la densité appropriée en milieu McCoy's 5A supplémenté de 10 % de SVF, puis les mélanges cellules/complexe de transfection ont été délicatement homogénéisés et mis en plaque. Les plaques ont été incubées pendant 24h en atmosphère humide à 5% de CO2 et le milieu a ensuite été remplacé par du milieu à 2% de SVF pendant 12h, après lavage en PBS IX. Après 12h de privation de sérum, le milieu a été retiré et remplacé par du milieu 2% SVF +/- 5 ng/ml de TNF-a (système rapporteur NF-0B) ou 100 pg/ml d'IFN-y (système rapporteur JAK/STAT) et 10% de surnageant de DSM 33715 ou de milieu de culture bactérien comme contrôle ou l'inhibiteur spécifique de la voie de signalisation (5 pM BAY11-7082 pour la voie NF-0B et 1 pM Tofacitinib pour la voie JAK/STAT). [0154] Le surnageant de DSM 33715 conduit à une diminution de l'activation de la voie NF-0B induite par le TNF-a de 25% et une diminution de l'activation de la voie JAK/STAT induite par l'IFN-y de 40%. Or, ces voies sont particulièrement pertinentes car hyperactives chez les patients atteints d'inflammation chroniques comme dans la maladie de Crohn, mais également certains cancers. La voie N F-KB est impliquée dans les cancers d'origine épithéliale, tels que le cancer du sein ; dans les cancers gastro-intestinaux, tels que le cancer colorectal. La
voie Jak/STAT est suractivée dans les cancers solides principalement, tels que le cancer du sein, du poumon, du foie, de la tête, du cou et de l'estomac.
[0155] Exemple 5 : Effet anti-inflammatoire de la souche selon l'invention en combinaison avec un agent anti-inflammatoire.
[0156] Dans la présente étude, les inventeurs ont étudié l'effet anti-inflammatoire de la souche DSM 33715 en combinaison avec le 5-ASA ou le Budésonide.
[0157] Les cellules HT-29 (ECACC) ont été ensemencées en plaque 24 puits à 3xl05 cellules/ puits en milieu McCoy's 5A (Gibco) supplémenté de 10% de sérum de veau foetal (SVF, Gibco) et incubées à 37°C/5% CO2 pendant 24h. Les cellules ont été lavées au PBS-1X (Gibco) et cultivées pendant 24h supplémentaires en milieu McCoy's 5A à 2% de SVF. Les cellules ont ensuite été stimulées pendant 6 heures avec 5 ng/ml de TNF-a en présence de 10% de surnageant en phase stationnaire de DSM 33715 ou de milieu de culture bactérien comme contrôle (ctrl), en présence ou non d'agents anti-inflammatoires. Le 5-ASA (10 mM) et le budésonide (20 pM) sont des molécules utilisées dans le traitement de maladies inflammatoires, comme la rectocolite et la maladie de Crohn. Après 6h, les surnageants de culture cellulaire ont été collectés et stockés à - 80°C jusqu'à la quantification de l'IL-8 par ELISA. Le dosage de l'IL-8 a été effectué par ELISA spécifique de BioLegend, selon les instructions du fabricant. L'absorbance à 460 nm a été lue utilisant le lecteur de microplaques FluoStar Omega, BMG Labtech. [0159] Pour évaluer l'effet immunomodulateur de la souche DSM 33715 en combinaison avec un agent anti-inflammatoire utilisé en thérapeutique, les cellules HT-29 ont été stimulées avec la cytokine pro-inflammatoire TNF-a et 10% du surnageant de DSM 33715 en présence ou non de 5-ASA ou de Budésonide, pendant 6H. La chimiokine IL-8 est sécrétée par les HT-29 dans un état inflammatoire induit par le TNF-a (Ctrl). Les valeurs représentent la moyenne normalisée en pourcentage par rapport au contrôle + TNF-a, représentant la sécrétion d'IL-8 induite par stimulation au TNF-a, soit 100%.
[0160] En présence de DSM 33715, la sécrétion de ITL-8 est diminuée de 28% par rapport au contrôle, et de 38% en présence de 5-ASA. La combinaison du surnageant et du 5-ASA conduit à une diminution de 64% (Panel A). Il y a donc un effet amélioré du surnageant et du 5-ASA dans la modulation de l'inflammation.

[0163] Dans la présente étude, les inventeurs ont étudié l'effet anti-inflammatoire de DSM 33715 associé ou non au Tofacitinib sur la modulation de la voie de signalisation de l'inflammation JAK/STAT induite par l'IFN-y.
[0164] Les cellules HT-29 à une densité de 3xl05 cellules/puits, en plaque 24 puits, ont été transfectées selon un protocole de reverse-transfection, avec 200 ng de vecteur pGL4 luc2P GAS-RE (voie de signalisation JAK/STAT dépendante de l'IFN-y) et 10 ng de vecteur pRL-TK (Promega), utilisant le réactif de transfection X-tremeGENE HP DNA avec un ration 3:1 (Roche). Brièvement, les complexes de transfection réactif/ADN ont été préparés dans un volume final de 50 pl de milieu McCoy's 5A sans sérum et sans antibiotique, la quantité appropriée de chaque plasmide a été ajoutée et homogénéisée, puis l'agent de transfection a été ajouté (3:1). Les complexes de transfection ont été mélangés délicatement et incubés à température ambiante pendant 15 minutes. Après incubation, les complexes de transfection ont été ajoutés aux cellules HT-29 fraîchement resuspendue à la densité appropriée en milieu McCoy's 5A supplémenté de 10 % de SVF, puis les mélanges cellules/complexe de transfection ont été délicatement homogénéisés et mis en plaque. Les plaques ont été incubées pendant 24h en atmosphère humide à 5% de CO2 et le milieu a ensuite été remplacé par du milieu à 2% de SVF pendant 12h, après lavage en PBS IX. Après 12h de privation de sérum, le milieu a été retiré et remplacé par du milieu 2% SVF +/- 100 pg/ml d'IFN-y (système rapporteur JAK/STAT) et 10% de surnageant de DSM 33715 ou de milieu de culture bactérien comme contrôle ou l'inhibiteur spécifique de la voie de signalisation (5 pM BAY11-7082 pour la voie NF-0B et 1 pM Tofacitinib pour la voie JAK/STAT). [0167] Exemple 7 : Effet de la souche selon l'invention dans la protection de l'épithélium intestinal.
[0168] Dans la présente étude, les inventeurs ont étudié l'effet de la souche DSM 33715 sur l'intégrité de la barrière intestinale par mesure de la résistance transépithéliale.
[0169] Les cellules Caco-2 (ECACC) ont été ensemencées à 2xl04 cellules par puits en plaques 24 puits contenant des inserts avec une membrane perméable en polyesterTranswell (Corning Life Science). Les cellules ont été cultivées en milieu DMEM supplémenté de 20% de sérum de veau foetal (SVF, Gibco) et incubées à 37°C / 5% CO2. La résistance électrique transépithéliale a été vérifiée tous les jours à partir du septième jour de culture à l'aide d'un ohmmètre EVOM3 (WPI). Le milieu a été remplacé tous les 2 jours durant 8 à 10 jours, jusqu'à ce que la TEER optimale ait été atteinte (2000 Q.cm2). A l'atteinte de la TEER de référence, les cellules ont été traitées dans le compartiment apical avec DSM 33715 ou DSM 22607 à une MOI de 50 ou du PBS IX / Glycérol 10% (contrôle) pendant 3h, puis 100 ng/ml de TNF-a a été ajouté ou non dans le compartiment basal. La TEER a été mesurée juste avant l'addition de TNF-a (T0) et 6h (T6H) après l'ajout de TNF-a.

[0171] Pour évaluer la capacité de la souche DSM 33715 à restaurer ou renforcer la barrière intestinale, les cellules Caco-2 en monocouche polarisées ont été prétraitées avec DSM 33715, puis sensibilisées au TNF-a. La TEER a été comparée à T0 (avant ajout du TNF) et T6H. Les valeurs représentent la moyenne normalisée par rapport au contrôle - TNF-a, représentant la TEER basale. Comme attendu, le traitement des Caco-2 avec le TNF-a induit une augmentation de la perméabilité (Ctrl + TNF-a). La souche DSM33715 est capable de restaurer la barrière
épithéliale, avec un retour au niveau basal.
[0172] Exemple 8 : Effet de la souche selon l'invention dans un modèle in vivo de colite.
[0173] Dans la présente étude, les inventeurs ont étudié l'effet in vivo de colite induite par le TNBS chez le rat. L'essai a été réalisé par un prestataire accrédité (Intestinal Biotech Development, Lille) selon les directives gouvernementales.
[0174] Pour l'induction de la colite, des rats Sprague Dawley ont été anesthésiés pendant 2h et ont reçu une injection intra rectale de TNBS (Trinitrobenzene sulfonic acid, 80 mg/kg dissous en éthanol 40%).
[0175] Les animaux ont été sacrifiés 4 jours après l'injection de TNBS. DSM 33715 a été administrée par voie orale à une dose de 109 CFU/ml, gavage débutant 14 jours avant l'induction de la colite et jusqu'au jour de l'euthanasie. Les animaux du groupe "contrôle positif" ont reçu le Pentasa mixé dans leur alimentation, à une dose de 150 mg/kg. L'effet antiinflammatoire des produits testés a été évalué au sacrifice, suite à une période totale de traitement de 18 jours.
[0176] Les variations de poids ont été suivies durant la totalité de la période de l'expérience. La longueur du côlon a été évaluée le jour de l'autopsie. Les scores macroscopique et microscopique ont été réalisés par deux opérateurs différents pour valider les scores. Le côlon de chaque rat a été examiné pour évaluer les lésions macroscopiques selon le score de Wallace. Le score de Wallace est une évaluation des lésions macroscopiques sur une échelle de 0 à 10, et ce score est basé sur des caractéristiques reflétant l'inflammation, telles que l'hyperémie, l'épaisseur de l'intestin et l'étendue des ulcérations.

Les coupes obtenues ont été colorées selon la technique de coloration MGG (May-Grünwald Giemsa). Ce classement sur une échelle de 0 à 6 prend en compte le degré de l'infiltrat inflammatoire, la présence d'érosion, d'ulcération ou de nécrose, ainsi que la profondeur et l'étendue des lésions. La méthode est décrite par Ameho et al., 1997, Gut.

(cytokine pro-inflammatoire) (kit eBioscience), ainsi que le niveau de la Lipocaline-2 (Lcn-2), marqueur de l'infiltration par les neutrophiles (Clinisciences) par quantification ELISA. Brièvement, un centimètre de côlon distal a été collecté et homogénéisé dans une solution de Tris-HCI contenant un cocktail d'inhibiteurs de protéases (Sigma-Aldrich) par l'utilisation de l'homogénéisateur Precellys utilisant des billes de céramique (1,4 et 2,8 mm). Les échantillons obtenus ont été centrifugés pendant 20 min et les surnageants collectés et conservés à -80°C, jusqu'à leur utilisation pour les ELISAs.

[0181] Figure 8A : Les inventeurs ont observé une diminution significative de la longueur du côlon dans le groupe de rats traité au TNBS et ayant reçu le véhicule en traitement par rapport au groupe contrôle ayant reçu le véhicule, comme attendue. Une augmentation significative de la longueur du côlon a été constatée dans le groupe de rats traités au TNBS ayant reçu la souche DSM 33715 par rapport au groupe TNBS-Véhicule. La souche DSM 33715 a un effet protecteur sur le côlon dans le modèle de colite induite par le TNBS.
[0182] Figure 8B : L'intensité de l'inflammation et des lésions du côlon a été évaluée au niveau macroscopique selon le score de Wallace. Une augmentation significative du score de Wallace a été observée dans les rats du groupe TNBS-Véhicule par rapport au groupe de rats "sains", avec respectivement un score de 7,00+/-0,41 vs 0,0+/-0,0, p<0,0001. Ce résultat indique que les rats présentaient une colite sévère 4 jours après l'induction avec un site ulcératif ou inflammatoire supérieur à 3 cm de colon (correspondant au score de 7, sur une échelle allant de 0 à 10). Une diminution significative du score a été observée dans le groupe TNBS-Pentasa et le groupe TNBS-DSM 33715. DSM 33715 améliore de 38% les lésions inflammatoires observées au niveau macroscopique par rapport au groupe TNBS-véhicule, avec un score de Wallace respectivement de 4,33+/-0,33 vs 7,00+/-0,41, p=0,049. Il en résulte que le traitement induit un effet anti-inflammatoire important.
[0183] Figure 8C : Une amélioration significative du score histologique - intensité des lésions inflammatoires- est observée dans le groupe de rats TNBS ayant reçu le Pentasa, contrôle positif, et la souche DSM 33715, par rapport aux rats TNBS ayant reçu le véhicule. observée pour le Pentasa et une diminution significative de la LCN-2 est observée dans le groupe de rats TNBS traités par la souche DSM 33715.

L'administration de DSM 33715 chez les rats colitiques a montré une diminution significative des niveaux d'IL-1 , indiquant un effet anti-inflammatoire de DSM 33715 dans le modèle de colite aigue induite par le TNBS chez le rat.
[0186] L'ensemble de ces résultats est en corrélation pour mettre en évidence des propriétés anti-inflammatoire de la souche DSM 33715, en particulier par rapport au traitement de référence sur ce type de pathologies mais également en comparaison avec la souche de référence de C. minuta. Ainsi, la souche selon l'invention possède un potentiel thérapeutique certain dans la dysbiose intestinales et en particulier dans la prévention et/ou le traitement des maladies inflammatoires.
[0187] Exemple 9 : Essai comparatif dans un modèle in vivo de colite
[0188] Dans la présente étude, les inventeurs ont étudié l'effet in vivo de la souche selon l'invention et de la souche de référence C. minuta (DSM22607) dans un modèle de colite induite par le TNBS chez le rat. L'essai a été réalisé selon le même protocole que celui décrit dans l'exemple 8.
[0189] Une induction de TNBS ayant eu lieu après 2 semaines de prétraitement avec C. minuta ou le contrôle chez le rat, les scores macroscopiques et microscopiques ont été évalués.
[0190] Les scores macroscopiques (évaluations physiologiques reflétant l'inflammation sur la base de l'hyperémie, de l'épaississement de l'intestin, de la diarrhée et de l'étendue de l'ulcération), l'évolution du poids corporel et le poids du côlon ont été analysés 4 jours après l'induction au TNBS.

Les résultats présentés dans le panel A démontrent l'intensité de l'inflammation et des lésions du côlon qui ont été évaluées au niveau macroscopique selon le score de Wallace dans un modèle de colite aiguë induite par TNBS chez le rat. Ce résultat indique que les rats non traités présentaient une colite sévère 4 jours après l'induction. Un effet anti-inflammatoire a également été observé dans le groupe TNBS- DSM 22607 et plus encore dans le groupe TNBS-DSM 33715 (Invention). Ainsi, la souche DSM 22607 diminue de 27% l'apparition des lésions alors que la souche selon l'invention (DSM
33715) les décroît de 38% par rapport au groupe TNBS-véhicule. (Statistiques : p=0,02 et p= 0,0001 respectivement).
[0193] Les résultats présentés dans le panel B démontrent, dans ce même modèle de colite aiguë chez le rat, une diminution significative de 36% du score histologique des coupes de côlon dans le groupe de rats TNBS ayant reçu la souche selon l'invention (DSM 33715) alors que ce n'est pas le cas chez les animaux ayant reçu le traitement avec DSM 22607. Le score histologique étant le reflet de l'intensité des lésions inflammatoires au niveau tissulaire, ce résultat met en évidence une supériorité de la souche DSM 33715 par rapport à DSM 22607 dans le traitement de l'inflammation gastro-intestinale. [0194] Ainsi, l'ensemble de ces résultats indique des propriétés anti-inflammatoires importantes de la souche DSM 33715 et un effet supérieur par rapport à la souche de référence DSM 22607 dans le modèle de colite aiguë induite par le TNBS chez le rat.
