<5> 酵素剤を60℃以上の温度において油脂に作用させる、<4>に記載の方法。
<6> 配列番号2又は配列番号4のアミノ酸配列と99%以上の配列同一性を有するアミノ酸配列からなるリパーゼであって、50mM PIPES緩衝液(pH7.0)中で70℃で30分処理後も酵素活性が残存する、リパーゼ。
<7> 以下のステップ(1)及び(2)を含んでなる、リパーゼの製造法:
(1)<6>に記載のリパーゼの産生能を有する微生物を培養するステップ;

本発明のエステル交換用酵素剤は、配列番号2又は配列番号4のアミノ酸配列と90%以上の配列同一性を有するアミノ酸配列からなるリパーゼを有効成分として含有する。
本酵素は、エステル交換リパーゼを産生する微生物(エステル交換リパーゼ産生株)を培養することにより得ることができる。
(1)配列番号2又は配列番号4のアミノ酸配列と99%以上の配列同一性を有するアミノ酸配列からなるリパーゼであって、50mM PIPES緩衝液(pH7.0)中で70℃で30分処理後も酵素活性が残存するリパーゼの産生能を有する微生物を培養するステップ;
(2)培養後の培養液及び/又は菌体より、リパーゼを回収するステップ。
本発明によれば、本酵素剤を油脂に作用させることを含む、エステル交換された油脂の製造方法が提供される。即ち、本発明によれば、本酵素剤を用いた油脂のエステル交換法が提供される。本発明のエステル交換法では本酵素剤又は本酵素を油脂に作用させるステップを行い、当該ステップによって油脂のエステル交換、即ち、油脂中のトリアシルグリセロール(TG又はTAGと略称される)の構成脂肪酸を再編成(再配列)させる。
<サザンブロッティング用TDL2遺伝子プローブの調製>
鋳型として、Talaromyces thermophilus NBRC31798TのゲノムDNAを用いて、表1のプライマーを設計してPCR DIG Synthesis kit(Roche社製)を用いたPCRラベリングにてTDL2遺伝子プローブを調製した。
TDL2遺伝子を含むDNA断片の増幅を行うため、自己環化させたDNA(約3.0kbp)をテンプレートとして用いて、表2.のプライマーF3およびプライマーR3にてPCRを行った。
増幅したDNA断片を精製し、シーケンス解析を行った。

<精製酵素の調製>
Talaromyces thermophilus NBRC31798Tをオートクレーブした小麦ふすま培地で固体培養(40℃、4日間培養)して得られた培養液をろ過後、20mMリン酸緩衝液(pH8.0)と1.0M塩化ナトリウム含有20mMリン酸緩衝液(pH8.0)のリニアグラジエント条件にて陰イオン交換クロマトグラフィー(GE Healthcare製HiPrep DEAE FF)を行った後、1.0M硫酸アンモニウム含有20mMリン酸緩衝液(pH7.0)と20mMリン酸緩衝液(pH7.0)のリニアグラジエント条件にて疎水性クロマトグラフィー(GE Healthcare製Hitrap Butyl FF)によって精製酵素を得た。
TDL1およびTDL2の精製酵素をcOmplete ULTRA Tablets(Roche社製プロテインインヒビター)を含む、50mM PIPES緩衝液(pH7.0)を使用し2倍希釈後、水浴にて熱処理(70℃ あるいは 100℃ 30分間)を行い、熱処理サンプルとした。
<方法>
シリカゲルカラムを用いて、ヒマワリ油からトリオレインを分取後、ポリビニルアルコール溶液と混合し、乳化した。乳化トリオレイン溶液に各サンプルを添加し、加水分解反応(40℃、1時間)を行った。反応液に酢酸エチルを添加し、トリオレインおよび反応生成物(ジオレイン、モノオレイン、オレイン酸)を抽出した。抽出液をクロロホルム:アセトン=96:4の展開溶媒条件にて薄層クロマトグラフィーによる分離・検出を行った。
耐熱性評価の結果(図1)、TDL1は70℃処理サンプルにおいても、反応生成物(ジオレイン、オレイン酸)が確認できることから、70℃で30分間処理後も活性が残存していることが判明した。
<組換え酵素発現株の取得>
タラロマイセス・サーモフィラス NBRC31798T(サーモマイセス・デュポンティ NCBI:txid28565)のゲノム DNAを鋳型としたPCRによって、TDL1遺伝子(配列番号5)およびTDL2遺伝子(配列番号6)を増幅した。増幅したDNAを糸状菌発現ベクター(Aspergillus oryzae由来α-アミラーゼ改変プロモーター、Aspergillus oryzae由来FAD-GDHのターミネーター及びpyrG遺伝子を挿入したpUC19)に連結し、プロトプラスト-PEG法によってA. oryzaeに導入した。
得られた形質転換体を澱粉、酵母エキス、無機塩類を含む液体培地(pH6.0)に接種し、30℃で90時間、通気撹拌培養した。培養上清を珪藻土ろ過によって生産菌体除去後、限外ろ過膜で濃縮し、凍結乾燥にて酵素の乾燥粉末を得た。
固定化担体として珪藻土を使用した。酵素の乾燥粉末中のタンパク質量をBradford法キット(BIO-RAD)にて測定し、測定値をもとにタンパク質量0.1gになるように酵素の乾燥粉末を精製水1mLに溶解し、酵素溶解液とした。常法に従い、固定化担体(セライトNo.535(Celite社製))に固定化し、固定化酵素(TDL1-IM、TDL2-IM)を調整した。
50mLチューブに3.4gのトリオレイン(オレインリッチ、昭和産業製)と5gのパルミチン酸メチル(Wako製)を加え、反応温度(40℃、80℃)で30分保温した。固定化酵素としてLipozyme TL-IM(ノボザイムス社)、TDL1-IM、TDL2-IMを用いた。各固定化酵素を0.05g加えた後、振とうして反応させた。20時間後の上清を回収し反応サンプルとした。

全てのリパーゼにおいてOAMEの生成が確認できた。TDL1―IMとLipozyme TL-IMは同等の生成量であり、TDL2-IMは他の2つに比べて60%程度の生成量であった。各リパーゼを用いた場合のPOP比率(%)を表6に示す。
OAME生成量が同等のときの1,3位特異性を評価するため、OAME生成量が約20%となるように各リパーゼで反応後、POP比率を評価した。上記評価方法に準じた方法で反応温度は80℃で実施した。酵素添加量や反応時間を適宜調整してOAME生成量が約20%となった時点で上清を回収した。
各リパーゼを用いた場合のPOP比率(%)を表2に示す。
