본 발명은 화합물 N-[(4'-브로모[1,1'-바이페닐]-4-일)설포닐]-L-발린 또는 이의 약제학적으로 허용 가능한 염을 유효성분으로 포함하는 아밀로이드 베타의 올리고머화 및 피브릴화 억제용 조성물에 관한 것이다.
본 발명은 대한민국 과학기술정보통신부의 지원 하에서 과제고유번호 1711115820 및 세부과제번호 2020R1A2B5B01002463에 의해 이루어진 것으로서, 상기 과제의 연구관리전문기관은 한국연구재단, 연구사업명은 "개인기초연구(과기정통부)(R&D)", 연구과제명은 "한국형 알츠하이머병 관련 돌연변이의 병인적 메커니즘 및 바이오마커의 multi variate 분석과 신약 개발 위한 한국형 세포와 동물 모델 개발 연구”, 주관기관은 가천대학교, 연구기간은 2020.03.01.-2023.02.28.이다.
본 특허출원은 2021년 05월 21일에 대한민국 특허청에 제출된 대한민국 특허출원 제10-2021-0065823호에 대하여 우선권을 주장하며, 상기 특허출원의 개시사항은 본원에 참조로서 삽입된다.
글로벌 대형 제약회사들의 알츠하이머병(Alzheimer's disease) 치료제 개발이 연이어 실패하고 있다. 지금까지의 치료제는 대부분 아밀로이드 베타(Amyloid beta, Aβ)나 타우(tau)와 같은 단백질에 결합하는 항체를 중심으로 개발되어왔지만, 현재까지도 많은 한계점을 지니고 있다.
Aβ의 경우 그 자체가 서로 올리고머화(oligomerization)하여 올리고머(oligomer)가 생성되고 계속해서 응집(aggregation)되면 피브릴화(fibrillization)가 진행되어 피브릴 형태(fibril form)를 형성하게 된다. 아직까지도 올리고머와 피브릴 중에서 어떤 형태가 신경세포에 독성을 일으키는지에 대한 논의가 진행 중에 있다. 현재까지의 결과들을 종합하면 올리고머가 세포에 독성을 일으킬 수 있다는 주장이 우세하다.
화합물 기반의 약물 후보를 스크리닝(screening)하는 대표적인 방법으로서 Aβ의 피브릴화(fibrillization)를 저해하는 효과를 확인하는 방법이 있다. 하지만 Aβ의 피브릴화는 세포 독성을 보호하는 기작으로도 여겨지고 있기 때문에 적합한 스크리닝 방법이 아닐 수 있다. 또한 Aβ의 피브릴화 저해 효과가 독성 성질을 가진 올리고머의 안정성을 높임으로써 일어날 수 있다는 문제점을 가지고 있다.
기존의 Aβ 연구는 항체를 기반으로 하기 때문에 막대한 연구비가 소요된다. 항체와는 달리 화합물은 항체에 비해 비용이 저렴하여 안정성이 매우 높다는 장점이 있다. 따라서 Aβ에 특이적으로 결합하는 화합물을 발견한다면 기존의 항체를 기반으로 하는 알츠하이머병의 예방, 개선 또는 치료 기법의 한계점을 극복할 수 있다.
본 발명자들은 종래의 알츠하이머병 치료제 대비 안정성이 높고 비용이 저렴하며 아밀로이드 베타(Amyloid beta, Aβ)에 특이적으로 결합하는 알츠하이머병을 예방, 개선 또는 치료할 수 있는 물질을 개발하고자 예의 연구 노력하였다. 그 결과, N-[(4'-브로모[1,1'-바이페닐]-4-일)설포닐]-L-발린이 Aβ에 특이적으로 결합하여 Aβ의 올리고머화(oligomerization) 및 피브릴화(fibrillization)를 저해하는 화합물임을 규명함으로써, 본 발명을 완성하게 되었다.
따라서, 본 발명의 목적은 화합물 N-[(4'-브로모[1,1'-바이페닐]-4-일)설포닐]-L-발린 또는 이의 약제학적으로 허용 가능한 염을 유효성분으로 포함하는 아밀로이드 베타의 올리고머화 및 피브릴화 억제용 조성물을 제공하는 것이다.
본 발명의 다른 목적은 본 발명에 따른 아밀로이드 베타의 올리고머화 및 피브릴화 억제용 조성물을 포함하는 신경보호에 의한 퇴행성 신경질환의 예방 또는 치료용 약제학적 조성물을 제공하는 것이다.
본 발명의 또 다른 목적은 본 발명에 따른 아밀로이드 베타의 올리고머화 및 피브릴화 억제용 조성물을 포함하는 기억력 개선, 인지 능력 개선 또는 학습 능력 개선용 약제학적 조성물을 제공하는 것이다.
본 발명의 다른 목적은 본 발명에 따른 조성물을 포함하는 아밀로이드 베타의 올리고머화 및 피브릴화 억제용 기능성 식품 조성물을 제공하는 것이다.
본 발명의 또 다른 목적은 본 발명에 따른 조성물을 포함하는 기억력 개선, 인지 능력 개선 또는 학습 능력 개선용 기능성 식품 조성물을 제공하는 것이다.
본 발명의 일 양태에 따르면, 본 발명은 하기의 화학식 1로 표시되는 화합물 또는 이의 약제학적으로 허용 가능한 염을 유효성분으로 포함하는 아밀로이드 베타(Amyloid beta, Aβ)의 올리고머화(oligomerization) 및 피브릴화(fibrillization) 억제용 조성물을 제공한다:

본 명세서의 화합물 "N-[(4'-브로모[1,1'-바이페닐]-4-일)설포닐]-L-발린(N-[(4'-Bromo[1,1'-biphenyl]-4-yl)sulfonyl]-L-valine)"은 PD166793이라고도 불리며 MMP 억제제(matrix metalloproteinase inhibitor)의 일종으로서 하기의 화학식 1로 표시되는 화합물을 의미한다.

본 명세서의 용어 "유효성분으로 포함하는"이란, 본 발명의 N-[(4'-브로모[1,1'-바이페닐]-4-일)설포닐]-L-발린 또는 이의 약제학적으로 허용 가능한 염의 약리학적 효능 또는 활성을 달성하는 데 충분한 양을 포함하는 것을 의미하며, 약물의 전달, 안정화 및 제제화를 위하여 다양한 성분이 부가적으로 첨가될 수 있는 것을 포함하는 의미이다.
본 발명의 용어 "아밀로이드 베타(Amyloid beta, Aβ)"는 APP(amyloid precursor protein)로부터 유래된 폴리펩타이드 또는 이의 단편을 의미하고, 이를 코딩하는 핵산 서열 또는 이의 단편을 포함할 수 있다. 상술한 APP가 α-secretase에 의해 절단되기 시작하는 비-아밀로이드 형성 경로(non-amyloidogenic pathway)와 β-secretase에 의해 절단된 다음 γ-secretase에 의해 절단되는 아밀로이드 형성 경로(amyloidogenic pathway)가 있다. 아밀로이드 형성 경로의 γ-secretase는 정확성이 떨어지기 때문에 다양한 C-말단을 갖는 아밀로이드 베타 종(amyloid beta species)을 형성한다. 따라서 아밀로이드 베타 종에는 다양한 Aβ가 존재하나, 이 중 40개의 아미노산으로 이루어진 아밀로이드 베타인 Aβ40이 약 80% 내지 90%로 가장 다수이고, 그 다음으로 42개의 아미노산으로 이루어진 Aβ42가 약 5% 내지 10% 정도를 차지한다. 이 가운데, 다른 아밀로이드 베타보다 소수성인 Aβ42가 아밀로이드 플라크(Amyloid plaque)의 주요한 구성으로 알려져 있다. 게다가, 또 다른 아밀로이드 베타 종인 43개의 아미노산으로 이루어진 Aβ43은 알츠하이머병의 조기 발병에 기여하는 것으로 알려져 있다.

프리세닐린-1(Presenilin-1, PS-1)은 인간 PSEN1 유전자에 의해 인코딩되는 프리세닐린(presenilin) 단백질로 γ-secretase 복합체의 4개의 코어 단백질 중 하나이며 APP로부터 Aβ를 생성하는 데 중요한 역할을 한다고 알려져 있다.

본 발명의 구체적인 일 구현예에 있어서, 본 발명의 아밀로이드 베타는 Aβ1-33, Aβ1-34, Aβ1-36, Aβ1-37, Aβ1-38, Aβ1-39, Aβ1-40, Aβ1-42, Aβ1-43, Aβ2-40, Aβ2-42, Aβ2-43, Aβ11-40, Aβ11-42 및 Aβ11-43으로 이루어진 군으로부터 선택되는 적어도 하나 이상의 아밀로이드 베타 종(amyloid beta species)을 포함하나 이에 제한되는 것은 아니다. 달리 명시되지 않는 한, 본 발명에 기재된 Aβ40은 Aβ1-40을 의미하고, Aβ42는 Aβ1-42를 의미하며, Aβ43은 Aβ1-43을 의미한다.

본 발명의 일 구현예에 있어서, 상기 아밀로이드 베타는 조직(tissue), 뇌척수액(cerebrospinal fluid, CSF), 혈청(serum), 혈장(plasma), 전혈(whole blood) 및 간질액(interstitial fluid)으로 이루어진 군으로부터 선택되는 적어도 하나 이상에 포함되는 것이다.

본 발명의 일 구현예에 있어서, 상기 퇴행성 신경질환은 인지 장애(cognitive disorder), 치매(dementia), 루이소체치매(Lewy body dementia) 알츠하이머병(Alzheimer's disease), 파킨슨병(Parkinson's diseases), 헌팅턴병(Huntington's disease), 근위축성 측삭경화증(Amyotrophic lateral sclerosis), 다발성 경화증(Multiple sclerosis), 허혈성 뇌손상(ischemic brain injury), 뇌염(encephalitis), 뇌수막염(meningitis), 길랑-바레 증후군(Guillain-Barre syndrome), 또는 외상성 뇌손상(traumatic brain injury)이다.
본 발명의 일 구현예에 있어서, 본 발명의 대상체(subject)는 포유동물이다. 본 발명의 일 구체예에 있어서, 상기 포유동물은 개, 고양이, 소, 말, 돼지, 마우스, 랫트, 기니피그, 토끼, 비-인간 영장류(non-human primate) 및 인간을 포함하나, 반드시 이에 제한되는 것은 아니다. 본 발명의 일 구체예에 있어서, 상기 비-인간 영장류는 원원류(prosimians), 구세계 원숭이(old world monkeys), 신세계 원숭이(new world monkeys), 또는 유인원(anthropoid)이다.
본 발명의 퇴행성 신경질환의 치료 방법은 상술한 본 발명의 다른 일 양태인 아밀로이드 베타의 올리고머화 및 피브릴화 억제용 조성물을 포함하므로, 본 명세서 기재의 과도한 복잡성을 피하기 위해 중복되는 내용을 원용하며, 그 기재를 생략한다.
본 발명의 특징 및 이점을 요약하면 다음과 같다:
(a) 본 발명은 화합물 N-[(4'-브로모[1,1'-바이페닐]-4-일)설포닐]-L-발린 또는 이의 약제학적으로 허용 가능한 염을 유효성분으로 포함하는 아밀로이드 베타의 올리고머화 및 피브릴화 억제용 조성물을 제공한다.
(b) 본 발명은 본 발명에 따른 아밀로이드 베타의 올리고머화 및 피브릴화 억제용 조성물을 포함하는 신경보호에 의한 퇴행성 신경질환의 예방 또는 치료용 약제학적 조성물을 제공한다.
(c) 본 발명은 본 발명에 따른 아밀로이드 베타의 올리고머화 및 피브릴화 억제용 조성물을 포함하는 기억력 개선, 인지 능력 개선 또는 학습 능력 개선용 약제학적 조성물을 제공한다.
(d) 본 발명은 본 발명에 따른 조성물을 포함하는 아밀로이드 베타의 올리고머화 및 피브릴화 억제용 기능성 식품 조성물을 제공한다.
(e) 본 발명은 본 발명에 따른 조성물을 포함하는 기억력, 인지 능력 또는 학습 능력을 개선하는 것인 것을 특징으로 하는 기능성 식품 조성물을 제공한다.
(f) 본 발명의 조성물을 이용하는 경우, Aβ와 특이적으로 결합하여 아밀로이드 베타의 올리고머화 및 피브릴화가 저해되고, 아밀로이드 베타에 의해 유발되는 세포 사멸은 억제되며, 활성산소종 생성은 저해되고, 미토콘드리아 기능 장애는 회복됨으로써 신경보호 효과가 있어 퇴행성 신경질환, 특히 알츠하이머병의 예방, 개선 또는 치료에 사용될 수 있고, 인지 능력, 기억력 및 학습 능력을 개선하는 데에 사용될 수 있다.
도 1은 티오플라빈 T 분석의 결과로 총 11개의 약물 후보와 Aβ의 반응 후 시간에 따른 형광 시그널의 변화를 나타낸다.
도 2는 Aβ의 모노머 형태로부터 멀티머 형태를 구별할 수 있는 멀티머 검출 시스템(MDS) 실험의 ELISA 분석 후, Aβ를 음성 대조군(negative control; PBS만을 포함하며, 그래프 X-축에서 'X'로 표기됨) 및 총 11개의 약물 후보와 각각 반응시킨 다음 시간에 따른 발광 세기를 측정하고, 음성 대조군의 그래프 아래 면적(area under the curve, AUC)을 0% 기준으로 하여, 약물 후보의 아밀로이드 베타(Aβ) 올리고머화 저해율(oligomerization inhibition rate)을 계산한 결과를 나타낸다.

이하, 실시예를 통하여 본 발명을 더욱 상세히 설명하고자 한다. 이들 실시예는 오로지 본 발명을 보다 구체적으로 설명하기 위한 것으로, 본 발명의 요지에 따라 본 발명의 범위가 이들 실시예에 의해 제한되지 않는다는 것은 당업계에서 통상의 지식을 가진 자에 있어서 자명할 것이다.
실시예
실시예 1:
약제학적 조성물의 제조
TOCRIS 사로부터 구매한 N-[(4'-브로모[1,1'-바이페닐]-4-일)설포닐]-L-발린을 최종 농도가 1 μM, 10 μM, 20 μM, 50 μM 및 100 μM이 되도록 DMSO(dimethyl sulfoxide)에 용해하여 약제학적 조성물을 제조하였다. 후술할 알츠하이머병 마우스 모델에 투여하는 경우에는 생리 식염수(saline)를 비히클(vehicle)로 하여 약제학적 조성물을 제조하였다. 이하에서는 본 발명의 유효성분인 N-[(4'-브로모[1,1'-바이페닐]-4-일)설포닐]-L-발린을 PD166793으로 표기하였고, 실시예 1의 약물이라고 지칭하였다. 용매만을 포함하는 조성물을 in vitro 실험에서 대조군(control)으로 사용하였고, 생리 식염수만을 투여한 비히클 그룹을 in vivo 실험에서 대조군으로 사용하였다.
실시예 2: Aβ 피브릴화 저해 효과
(1) 티오플라빈 T 분석 방법
아밀로이드-베타(Aβ)의 피브릴화에 대한 실시예 1의 약물의 저해 효과를 in vitro에서 측정하기 위해 티오플라빈 T 분석(Thioflavin T assay, ThT assay)을 수행하였다.
인간 Aβ42(AggreSureTM, AnaSpec)를 PBS(phosphate buffered saline)에 pH 7.4 및 10 μM 농도가 되도록 용해하여 30 μl를 준비하였다. 실시예 1의 약물, 다른 후보 약물 49개 및 페놀 레드(phenol red, 50 μM, 대조군) 각 20 μl에 상술한 30 μl의 Aβ42를 첨가한 후, 37℃에서 24시간 동안 인큐베이션(incubation)하여 반응물을 준비였다. 그 다음, pH 9의 글라이신-수산화나트륨 완충용액으로 제조한 20 μl의 ThT 용액(50 μM, Sigma Aldrich)을 각 반응물에 첨가하여 혼합물을 제조하였다.
그 후, 각 혼합물과 ThT 용액의 형광 신호는 마이크로플레이트 판독기(microplate reader, PerkinElmer Victor-3®)를 사용하여 450 nm의 여기 파장(excitation wavelength) 및 510 nm의 방사 파장(emission wavelength)에서 측정하였다. 측정한 각 형광 흡광도(fluorescence absorbance)에서 ThT 용액의 배경 형광(background fluorescence)을 제하였다.
(2) 티오플라빈 T 분석 결과
첫 실험에서 실험군으로 실시예 1의 약물이 포함된 총 50개의 약물과 대조군으로 페놀 레드를 Aβ 및 ThT 용액과 혼합하여 반응시킨 후 형광 시그널의 변화를 관찰하였다. 실험 결과, 상기 50개의 약물 중 Aβ 피브릴화 억제 효과를 보인 약물 20개를 선별할 수 있었다.
선별된 약물 20개의 Aβ 피브릴화 억제 효과에 대한 검증 실험을 추가로 진행하였고, 그 결과 총 11개의 약물 후보를 선별할 수 있었다. 결과는 도 1에 나타내었다. 도 1의 그래프 범례에서 'No drug'는 대조군인 페놀 레드를 나타내고, '4-73'은 실시예 1의 약물(PD166793)을 나타낸다. 도 1의 그래프에서 X축은 시간(분)을 나타내고 Y축은 티오플라빈 T의 형광 세기를 나타낸다.
도 1에 나타낸 바와 같이, 대조군(도 1의 No drug)과 비교하여 실시예 1의 약물(도 1의 4-73)을 처리한 경우 피브릴 형광 시그널이 전혀 나타나지 않아, Aβ의 피브릴화가 100% 억제되었음을 확인하였다.
상기 결과로부터 실시예 1의 약물이 아밀로이드 베타의 피브릴화를 현저하게 억제함을 확인할 수 있었다.
실시예 3: Aβ 올리고머화 저해 효과
(1) 멀티머 검출 시스템 분석 방법
Aβ의 올리고머화에 대한 실시예 1의 약물의 저해 효과를 in vitro에서 측정하기 위해 멀티머 검출 시스템(Multimer detection system, MDS) 실험 기법을 다음과 같이 수행하였다. MDS 실험 기법에서 Aβ의 N-말단에 대해 특이적인 에피토프-중첩된 항체(epitope-overlapping antibody)를 사용함으로써 올리고머 형태의 Aβ 항원만을 포획하고 검출할 수 있었다.
인간 Aβ42(제조사 rPeptide)를 50 mM의 Tris 및 150 mM의 NaCl이 첨가된 pH 7.2의 용액에 용해하였고, TBST를 이용하여 희석하였다. 준비한 Aβ42를 실시예 1의 약물, 다른 후보 약물 10개 및 음성 대조군(PBS만을 첨가한 용액)에 각각 첨가해 37℃에서 24시간 동안 인큐베이션하여 Aβ의 올리고머 형성을 유도한 반응물을 준비하였다.
ELISA(enzyme-linked immunosorbent assay)를 이용하여 올리고머 형태의 Aβ만을 측정하기 위해서, 96-웰 플레이트(96-well plate)에 2 μg/ml의 항-β-아밀로이드1-42 항체(BioLegend®)를 코팅하였고, 항체가 코팅된 플레이트에 상기 반응물을 각각 웰(well) 당 100 μL씩 분주한 후 37℃에서 1시간 동안 인큐베이션하였다. 상기 플레이트를 TBST로 3회 세척한 후, HRP(horseradish peroxidase)가 표지된 항-β-아밀로이드1-42 항체(BioLegend®, 200 ng/ml)를 웰 당 100 μl씩 플레이트에 첨가하였고, 다시 37℃에서 1시간 동안 인큐베이션하였다. 그 다음, 플레이트를 TBST로 3회 세척한 후 웰 당 100 μl씩 ECL(enhanced chemiluminescence) 용액을 플레이트에 분주하고 발광(luminescence) 세기를 마이크로플레이트 판독기(PerkinElmer®)를 사용하여 측정하였다.
상술한 실시예 2에서 ThT 실험 결과 최종으로 선별된 11개의 약물(실시예 1의 약물이 포함됨)의 Aβ 올리고머화 저해 효과를 상기 MDS 실험 기법으로 확인하였다. 결과는 도 2에 나타내었다. 도 2의 그래프에서 범례 중 'X'는 약물을 처리하지 않고 완충용액(PBS)만을 처리한 음성 대조군을 나타내고, '4-73'은 실시예 1의 약물을 나타내며, 그래프의 Y축은 음성 대조군을 0% 기준으로 하여 다른 약물 후보의 올리고머화 저해율(%)을 나타낸다.
(2) 멀티머 검출 시스템 분석 결과
도 2에 나타낸 바와 같이, MDS 실험 기법을 통해 올리고머 형태의 Aβ만을 측정한 결과, 실시예 1의 약물(도 2의 4-73)이 PBS만을 처리한 음성 대조군(도 2의 X) 대비 약 23.4%로 Aβ의 올리고머화 저해율을 나타내는 것을 확인하였다.
상기 결과로부터 실시예 1의 약물이 다른 약물 후보에 비해 아밀로이드 베타의 올리고머화를 현저하게 억제함을 확인할 수 있었다.
상기 실시예 2 및 실시예 3의 결과로부터, 본 발명자들은 실시예 1의 약물이 대조군 대비 아밀로이드 베타의 피브릴화 및 올리고머화를 유의하게 저해하는 효과를 가지고 있음을 확인하였다.
이하에서는 실시예 1의 약물이 신경세포에서 세포 독성을 나타내는지 여부와 Aβ에 의해 유발된 세포 독성에 대해 신경보호 효과를 갖는지 여부를 in vitro에서 조사하였다.
실시예 4: SH-SY5Y 세포 배양
신경모세포종 SH-SY5Y 세포(neuroblastoma, ATCC CRL-2266)를 10% FBS, 1% 카나마이신(kanamycin) 및 1% 항생제-항균제(antibiotic-antimycotic)가 보충된 DMEM에서 37℃ 및 5% CO2 조건하에서 1주에 2회 계대하여 배양하였다. 96-웰 플레이트에서 세포를 배양하는 경우, 2 Х 104 세포/웰의 SH-SY5Y 세포를 96-웰 플레이트에서 계대 배양한 후, 24시간 동안 배양하였다.
이하의 실시예에서는 실시예 4의 방법으로 배양된 SH-SY5Y 세포가 80% 내지 90%의 세포밀집도에 도달하였을 때 실험을 수행하였다.
실시예 5: 실시예 1의 약물 자체의 세포 독성 확인
신경세포 SH-SY5Y에 실시예 1의 약물을 농도별로 처리한 후에 세포 생존율(cell viability)에 영향을 주는지, 즉 세포 독성(cytotoxicity)이 있는지 확인하였다.
(1) 세포 독성 확인 방법 - ATP 발광 분석
실시예 1의 약물 자체의 세포 독성을 확인하기 위해서, ATP 발광 분석(ATP luminescence assay)을 이용하여 세포 생존율을 측정하였다. 실시예 4에 나타낸 바와 같이, 2 Х 104 세포/well의 SH-SY5Y 세포를 96-웰 플레이트에서 계대 배양한 후, 24시간 동안 배양하였다. 이후, 세포에 24시간 동안 실시예 1의 약물을 1 μM, 10 μM, 20 μM 및 50 μM 농도로 처리하였다. 대조군으로서 용매만 첨가하였다. 배지(media)를 제거하고, 세포를 PBS로 세척(washing)한 뒤, 새로운 배지를 첨가하고, 추가로 30분 동안 배양하였다. 그런 다음, CellTiter-Glo® luminescent 시약을 첨가하고 발광을 다중-플레이트 판독기(multi-plate reader)에서 확인하였다. 세포 생존율 퍼센트(%)는 3번의 실험을 반복한 값의 평균 ± SD(standard deviation)로 표기하였고, 대조군의 값을 사용하여 정규화(normalization)하였다. 대조군 대비 통계학적으로 유의하게 세포 생존율(%)이 감소한 경우 세포 독성이 있다고 판단하였다. 결과는 도 3에 나타내었다.
(2) 세포 독성 확인 결과
도 3에 나타낸 바와 같이, 실시예 1의 약물 농도가 50 μM인 경우 SH-SY5Y 세포에서 세포 생존율이 감소하는 경향이 있었지만, 대조군과 비교하여 유의한 통계적인 차이는 보이지 않았다.
따라서, 실시예 1의 약물은 50 μM 농도까지 세포 독성이 없는 것으로 확인할 수 있었다. 그러나 이하의 실시예에서는 50 μM 농도는 제외하고 1 μM, 10 μM 및 20 μM 농도를 적용하여 신경보호 실험을 수행하였다.
실시예 6: Aβ 유발 독성에 대한 신경보호 효과
실시예 1의 약물이 신경세포에서 Aβ에 의해 유발되는 세포 독성에 대한 신경보호 효과(neuroprotective effect)가 있는지 확인하였다.
(1) Aβ 유발 세포 독성 실험 방법
실시예 4의 방법으로 SH-SY5Y 세포를 배양하여 안정화(stabilization)시킨 후, 세포에 5 μM의 아밀로이드 베타 1-42 (Aβ1-42)를 처리하기 6시간 전에 실시예 1의 약물을 1 μM, 10 μM 및 20 μM의 농도로 각각 전처리(pre-treatment)하였다. 대조군으로는 세포에 실시예 1의 약물과 Aβ1-42 모두 처리하지 않고 용매만을 처리한 대조군(control)을 사용하였다. 아밀로이드 베타 5 μM만을 단독으로 처리한 실험군(도 4의 Abeta 5 μM) 또한 실험하였다. 세포 생존율(%)은 상술한 실시예 5에 기재된 ATP 발광 분석 방법으로 측정하였다. 통계적으로 유의한 차이는 Aβ 5 μM 단독 처리군(Abeta μM)과 비교하여 p<0.05인 경우 * 표시로 나타내었다. 결과는 도 4에 나타내었다.
(2) Aβ 유발 세포 독성 실험 결과
도 4에 나타낸 바와 같이, 대조군(도 4의 control) 대비 아밀로이드 베타 5 μM 단독 처리군(도 4의 Abeta 5 μM)에서 세포 생존율이 50%까지 감소하였으나, 실시예 1의 약물을 함께 처리하였을 때 세포 사멸이 현저히 줄어드는 것을 관찰하였다. 아밀로이드 베타 5 μM 단독 처리군 대비 아밀로이드 베타 5 μM에 실시예 1의 약물을 함께 처리하였을 때 10 μM부터 세포 생존율이 증가, 즉 세포 사멸이 유의하게 감소한 것을 확인하였다(*p<0.05). 특히, 실시예 1의 약물을 20 μM 농도로 함께 처리하였을 때 아밀로이드 베타 5 μM 단독 처리군 대비 세포 생존율이 80%로 회복, 즉 세포 사멸이 유의하게 감소하였다(*p<0.05).
상기 결과로부터 신경세포에서 실시예 1의 약물이 아밀로이드 베타에 의해 유발되는 세포 독성에 대한 유의한 신경보호 효과가 있음을 확인하였다.
이하에서는 실시예 1의 약물이 어떤 메커니즘을 통해 신경보호 효과를 발휘하는지 조사하기 위해, 실시예 1의 약물이 아밀로이드 베타에 의해 유발되는 증가된 활성산소종 수준과 감소된 미토콘드리아 막 전위에 대해 미치는 영향을 조사하였다.
실시예 7: Aβ 유발 활성산소종의 생산 저해 효과
아밀로이드 베타는 활성산소종(Reactive Oxygen Species, ROS)을 증가시켜 세포 사멸을 일으키는 것으로 알려져 있다. 실시예 1의 약물이 Aβ에 의해 증가된 ROS를 저해하는 효과가 있는지 조사하였다.
(1) Aβ 유발 활성산소종 측정 방법
H2DCFDA(2',7'-dichlorodihydrofluorescein diacetate) 염색 방법을 이용하여 ROS를 측정하였다. 실시예 4의 방법으로 배양한 SH-SY5Y 세포를 24시간 안정화한 후, 2시간 동안 실시예 1의 약물을 1 μM, 10 μM 및 20 μM의 농도로 전-처리하였다. 이어서, Aβ 5 μM를 처리하고 추가로 24시간 동안 배양하였다(도 5의 1 μM + Abeta, 10 μM + Abeta 및 20 μM + Abeta 참조). 세포에 용매만을 처리한 대조군(control)을 사용하였고, Aβ 5 μM 단독 처리군(Abeta 5 μM, Aβ 처리 후 24시간 배양)과 실시예 1의 약물 단독 처리군(1 μM, 10 μM 및 20 μM, 약물 처리 후 26시간 배양) 또한 실험하였다.
인큐베이션 후, 생성된 ROS 수준을 평가하기 위해, 세포에 25 μM의 H2DCFDA를 처리하고 37℃에서 빛을 차단시킨 채 2시간 동안 추가 배양하였다. 495 nm의 여기 파장(excitation wavelength) 및 520 nm의 방사 파장(emission wavelength)에서 마이크로플레이트 판독기로 형광 강도를 측정하였다. ROS 수준은 3회 반복 측정되었으며, 대조군(control) 백분율(100%)을 기준으로 계산되었다. 통계적으로 유의한 차이는 Aβ 5 μM 단독 처리군(Abeta)과 비교하여 p < 0.05인 경우 * 표시로 나타내었다. 결과는 도 5에 나타내었다.
(2) Aβ 유발 활성산소종 측정 결과
도 5에 나타낸 바와 같이, 대조군(control) 대비 Aβ 5 μM을 단독 처리한 경우(도 5의 Abeta 5 μM) ROS가 유의하게 증가하였고(*p<0.05), 이에 반해 Aβ 5 μM 단독 처리군 대비 실시예 1의 약물을 함께 처리한 경우 10 μM부터 ROS가 유의하게 감소하는 것을 확인하였다(*p<0.05). 실시예 1의 약물 단독 처리군 또한 Aβ 5 μM 단독 처리군 대비 유의하게 낮은 ROS 수준을 보였다(*p<0.05).
상기 결과로부터 실시예 1의 약물이 아밀로이드 베타에 의해 유발된 ROS 생성을 저해함으로써 세포 생존율을 증가시키는 유의한 신경보호 효과를 가지고 있음을 확인하였다.
실시예 8: Aβ 유발 미토콘드리아 막 전위 손실에 대한 회복 효과
실시예 1의 약물이 나타내는 신경보호 효과의 가능한 메커니즘을 확인하기 위해 미토콘드리아 막 전위(mitochondrial membrane potential, MMP, ΔΨm)를 측정하였다.
(1) 미토콘드리아 막 전위 측정 방법
ΔΨm은 TMRE(tetramethylrhodamine, methyl ester) 염색 방법을 이용하여 측정하였다. 실시예 4의 방법으로 배양한 SH-SY5Y 세포를 24시간 순응화(acclimatization)한 후, 2시간 동안 세포에 실시예 1의 약물을 전-처리하였다. 이어서, 24시간 동안 5 μM의 아밀로이드 베타와 함께 배양하였다. 세포에 용매만을 처리한 대조군(control)을 사용하였고, Aβ 5 μM 단독 처리군(Abeta 5 μM, Aβ 처리 후 24시간 배양)과 실시예 1의 약물 단독 처리군(1 μM, 10 μM 및 20 μM, 약물 처리 후 26시간 배양) 또한 실험하였다.
그 후, ΔΨm을 측정하기 위해 1 μM의 TMRE 염색 용액을 첨가하고 37℃에서 30분 동안 인큐베이션하였다. 549 nm의 여기 파장(excitation wavelength) 및 575 nm의 방사 파장(emission wavelength)에서 마이크로플레이트 판독기로 형광 강도를 측정하였다. ΔΨm은 3회 반복 측정되었고 평균 ± SD로 나타내었으며 대조군(control)에 대한 백분율(100%)로 계산되었다. 통계적으로 유의한 차이는 Aβ 5 μM 단독 처리군(Abeta)과 비교하여 p<0.05인 경우 * 표시로 나타내었다. 결과는 도 6에 나타내었다.
(2) 미토콘드리아 막 전위 측정 결과
도 6에 나타낸 바와 같이, 대조군(control) 대비 Aβ 5 μM을 단독 처리한 경우(도 6의 Abeta 5 μM) 미토콘드리아의 기능이 60%까지 유의하게 감소하였지만(*p<0.05), Aβ 5 μM 단독 처리군 대비 실시예 1의 약물을 함께 처리한 경우 10 μM부터 미토콘드리아 막 전위가 회복되는 것을 확인하였다(*p<0.05). 실시예 1의 약물 단독 처리군 또한 아밀로이드 베타 5 μM 단독 처리군 대비 유의하게 높은 미토콘드리아 막 전위 수준을 나타내었다(*p<0.05). 이러한 결과는 실시예 1의 약물이 아밀로이드 베타에 의해 유발된 ROS 생성을 유의하게 저해하는 효과를 나타낸 상술한 실시예 7의 결과와 일치한다.
상기 결과로부터 실시예 1의 약물이 아밀로이드 베타에 의해 유발된 미토콘드리아 막 전위 감소(즉, 미토콘드리아 기능 저하)를 회복시킴으로써 미토콘드리아의 기능장애를 예방하는 신경보호 효과가 있음을 확인할 수 있었다.
상기 실시예 4 내지 실시예 8의 결과로부터 실시예 1의 약물 자체 신경세포 독성이 없고, 실시예 1의 약물은 신경세포에서 아밀로이드 베타에 의해 유발되는 세포 사멸을 저해하며, 아밀로이드 베타에 의해 유발되는 ROS 생성을 저해하고, 아밀로이드 베타에 의해 유발되는 미토콘드리아 막 전위 손실을 회복하는 신경보호 효과를 가지고 있음을 in vitro에서 확인하였다.
이하에서는 실시예 1의 약물이 알츠하이머병 동물 모델에서 인지 능력 개선 효과, 학습과 기억 개선 효과, 대뇌 피질 및 해마에서 플라크 감소 효과를 나타내는지 여부를 in vivo에서 조사하였다.
실시예 9: 알츠하이머병 동물 모델
인간 APP 및 PS1을 과발현(overexpression)함으로써 알츠하이머병(Alzheimer's disease, AD)이 유발되는 5xFAD 형절전환 마우스(transgenic mouse)를 이용하여 실험을 진행하였다. 5xFAD 마우스는 3개의 FAD(Familial Alzheimer's Disease) 돌연변이 Swedish (K670N, M671L), Florida (I716V) 및 London (V717I)을 가지는 인간 APP(695)(Amyloid beta Precursor Protein 695)와 2개의 FAD 돌연변이 M146L 및 L286V를 가지는 인간 PS1(Presenilin 1)를 과발현하는, 5개의 AD-관련 돌연변이를 가지는 알츠하이머병 동물 모델이다.
(1) 5xFAD 마우스 번식 방법
5xFAD 마우스(5XFAD, 5x-FAD 및 Tg6799로도 알려짐)는 The Jackson Laboratory 사로부터 구입한 후, 공급처에서 제공된 정보를 바탕으로 C57BL6/SJL F1 암컷(C57BL6/J Х SJL/J F1)과의 교미를 통해 동종 번식을 하였다.
(2) 5xFAD 마우스 유전자형분석
유전자형분석(genotyping, 지노타이핑)을 통해, 정상 대조군(WT, wild type)과 유전자변형군(5xFAD)으로 나누었다. 유전자형분석 결과는 도 7에 나타내었다.
도 7에 나타낸 바와 같이, PCR 산물이 검출되지 않은 마우스는 WT으로 결정하고, APP와 PS1(Presenilin 1) PCR 산물이 동시에 검출된 마우스는 5xFAD로 결정하였다.
(3) 5xFAD 마우스의 공지된 표현형 특성
당업계에 공지된 5xFAD 마우스의 플라크(plaque), 다발(Tangle), 신경세포 손실(neuronal loss), 신경아교종(gliosis), 시냅스 손실(synaptic loss), LTP/LTD 변화(changes in LTP/LTD) 및 인지기능장애(cognitive impairment)와 같은 표현형 특성을 요약하여 아래에 기재하였다.
플라크를 조사한 결과, 5xFAD 마우스는 약 2개월령에 처음에는 해마이행부(subiculum) 및 피질의 Layer V에서 세포 외 아밀로이드 침착을 보였다. 1.5개월령에서부터 Aβ42 또한 세포체(soma) 및 신경돌기(neurite) 내에 응집된 형태(aggregated form)로 신경세포 내에 축적되었다. 다발을 조사한 결과, 18개월령 이상까지 관찰되지 않았다. 신경세포 손실을 조사한 결과, 6개월령에서 피질 layer V 및 해마이행부에서 신경세포 손실이 관찰되었다. 신경아교종을 조사한 결과, 2개월령에서부터 관찰되었다. 시냅스 손실을 조사한 결과, 시냅스전 마커(presynaptic marker)인 시넵토피신(synaptophysin)의 수준이 4개월령부터 감소하기 시작하였고, 또 다른 시냅스전 마커인 신탁신(syntaxin) 및 시냅스후 마커(postsynaptic marker)인 PSD-95의 수준이 9개월령부터 감소하기 시작하였다. LTP/LTD 변화를 조사한 결과, 기저 시냅스 전달 및 해마 CA1 영역에서의 LTP가 4개월령 내지 6개월령 사이에 악화되기 시작하였다. 인지기능장애를 조사한 결과, 4개월령 내지 5개월령에 Y-미로 시험에서 공간 작업 기억(spatial working memory)의 손상을 나타내었고, 맥락적-공포-조건화 시험(contextual-fear-conditioning test)에서 장기 기억 안정화의 손상을 나타내었다.
이러한 신경학적 특성과 인지기능 장애를 나타내는, 알츠하이머병 동물 모델로서 잘 구축된 5xFAD 마우스를 이용하여, 이하의 실시예에서 실시예 1의 약물이 인지기능 장애와 아밀로이드 플라크 침착에 대한 개선 효과가 있는지 평가하기 위해, 다음과 같이 그룹화(grouping)하였다.
(4) 그룹화
2.5개월령의 5xFAD 마우스(Tg, transgenic)와 연령 대비 대조군(WT)을 포함하여 그룹당 11마리 내지 13마리가 되도록 그룹으로 나뉘었다. 실시예 9의 방법으로부터 얻은 WT 또는 Tg 그룹에, 비히클(vehicle) 또는 실시예 1의 약물(PD166793)을 10 mg/kg 용량으로 2개월간 주 5회 경구 투여하였다. 비히클(vehicle)은 생리 식염수(saline)를 사용하였다. 이하의 실시예에서 실시예 1의 약물이 알츠하이머병에 대한 치료 효과가 in vivo에서 있는지 여부를 테스트하기 위해, 실시예 9의 방법에 따른 알츠하이머병 마우스 모델을 이용하여, 4개 그룹(WT-vehicle; WT-PD166793; 5xFAD-vehicle; 및 5xFAD-PD166793)으로 나누었다.
실시예 10: 인지 능력 개선 효과 - 사물 인지 시험
실시예 1의 약물이 실시예 9에 따른 알츠하이머병 마우스 모델의 인지 기억(recognition memory)에 주는 영향을 평가하기 위해서 사물 인지 시험(Novel object recognition test, NOR)을 수행하였다.
(1) 사물 인지 시험 방법
NOR을 수행하여 비디오(video)로 추적한 동물의 행동 및 움직임을 분석할 수 있는 소프트웨어인 Ethovision XT 9 system을 이용하여 동물이 새로운 물체(novel object)에 대해 관심이 있는지 측정하였다.
상세하게는, 불투명한 박스에 동물이 사육되는 환경과 유사한 환경을 만들어 준 후, 동물을 박스에 1일간 적응(habituation)을 시켜주었다. 박스에 두 개의 동일한 물체를 넣어주고 탐색할 수 있게 한 다음, 두 개의 익숙한 물체(old object) 중 하나를 꺼내고 그 대신 실험동물이 새롭게 인지할 수 있는 새로운 물체(new object)를 넣어준 후, 익숙한 물체와 새로운 물체 사이에서 새로운 물체에 대해 얼마나 관심을 가지는지에 대해 조사하기 위해 각 물체 근처에 머문 시간, 빈도 등을 측정하였다. 실시예 1의 약물에 대한 효과로 새로운 물체에 대한 인지 능력이 그룹 간 차이가 있는지 확인하였다. 결과는 도 8에 나타내었다.
(2) 사물 인지 시험 결과
도 8에 나타낸 바와 같이, WT에 식염수를 투여한 그룹(WT-vehicle)과 WT에 실시예 1의 약물인 PD166793을 투여한 그룹(WT-PD166793) 모두 익숙한 물체를 탐색한 비율 대비 새로운 물체를 탐색한 비율이 현저하게 높았다. 반면에 5xFAD 마우스에 식염수를 투여한 그룹(5xFAD-vehicle)은 오히려 익숙한 물체(old object)에 대한 탐색 비율이 높았는데, 이는 5xFAD 마우스의 손상된 인지 능력을 나타낸다. 실시예 1의 약물인 PD166793을 투여한 5xFAD 마우스 그룹(5xFAD-PD166793)은 사물 인지 시험(NOR) 결과 새로운 물체를 탐색한 비율이 유의하게 높아, 생리 식염수를 투여한 5xFAD 마우스 그룹(5xFAD-vehicle)에 비해 회복된 인지 능력을 나타내어, Memory index 결과에서 유의한 차이를 나타내었다. 총 움직인 거리(total distance)와 속도(velocity)에서는 그룹 간 유의한 차이가 없었다(데이터 기재 안함).
상기 결과로부터 5xFAD 마우스는 실시예 1의 약물인 PD166793 투여에 의해 인지 능력(recognition ability)과 공간 관련 기억력이 개선되었음을 확인하였다. 즉, 실시예 1의 약물은 알츠하이머병 동물 모델의 인지 능력과 공간 관련 기억력을 유의하게 개선시키는 효과를 나타내었다.
실시예 11: 공간 기억력 개선 효과 - Y자 미로 시험
실시예 1의 약물이 실시예 9에 따른 알츠하이머병 마우스 모델의 기억 손상(memory impairment)에 주는 영향을 평가하기 위해서 Y자 미로 실험(Y-maze test)을 수행하였다.
(1) Y자 미로 시험 방법
Y자 미로 시험은 Y자 모양의 암(arm)을 수조 안에 설치하여 수행된다. 동물에게 단서(cue)가 보이도록 가지 모양 끝에 설치하고, EthoVision XT 9 system (Noldus)을 통해 동물의 움직임을 8분 동안 관찰하였다. 각 arm에 임의의 번호를 정한 후 동물이 들어가는 번호를 기록하였다. 동물이 갔었던 길을 기억하고, 새로운 길로 가려는 행동, 즉 변경 행동(spontaneous alteration)을 통하여 공간 기억력을 살펴보았다. 결과는 도 9a 및 도 9b에 나타내었다.
(2) Y자 미로 시험 결과
도 9a에 나타낸 바와 같이, 대조군(WT-Vehicle) 대비 식염수를 투여한 5xFAD 마우스 그룹(5xFAD-vehicle)은 유의하게 감소한 변경 행동을 보였다. 반면에, 실시예 1의 약물인 PD166793을 투여한 5xFAD 마우스 그룹(5xFAD-PD166793)은 식염수를 투여한 5xFAD 마우스 그룹(5xFAD-vehicle)에 비해 변경 행동(spontaneous alteration)이 유의하게 증가하였다($$ p<0.01). 도 9b에 나타낸 바와 같이, 총 암 출입 횟수(total arm entries, N)에서는 그룹 간 유의한 차이가 없었다.
이러한 결과로부터 5xFAD 마우스에서 실시예 1의 약물 투여에 의해 변경 행동 능력 즉, 공간 기억력(spatial memory)이 개선되었음을 확인할 수 있었다. 즉, 실시예 1의 약물은 알츠하이머병 동물 모델의 공간 기억력을 유의하게 개선시키는 효과를 나타내었다.
실시예 12: 학습과 기억 개선 효과 - 수동회피실험
실시예 1의 약물이 실시예 9에 따른 알츠하이머병 마우스 모델의 기억 손상(memory impairment)에 주는 영향을 평가하기 위해서 수동회피실험(passive avoidance test)을 수행하였다.
(1) 수동회피실험 방법
실험 쥐에게 외부에서 오는 불쾌한 자극(electric shock)을 학습시켜 해당 자극에 대한 기억력(learning and memory)을 평가하기 위해 수동회피실험을 진행하였다. 훈련(Training) 단계로, 어두운 환경을 좋아하는 쥐의 습성을 이용해 실험 쥐가 GEMINITM Avoidance System에 입장하면 20초 안에 어두운 방으로 이동하도록 학습시켰다(다음날 어두운 방에서 불쾌한 자극을 학습시키기 위함). 다음날 쥐에게 어두운 방에서는 불쾌한 자극이 주어진다는 것을 학습시킨 뒤, 해당 내용에 대한 기억력을 Step-through latency로 측정하여 평가하였다. 어두운 방에서 불쾌한 자극에 대해 학습하고 기억한 실험 쥐는 어두운 방으로 들어가기까지 걸리는 시간(초)인 Step-through latency가 길고, 이러한 학습이 이루어지지 않고/않거나 기억이 형성되지 않은 경우에는 Step-through latency가 짧다. 결과는 도 10에 나타내었다.
(2) 수동회피실험 결과
도 10에 나타낸 바와 같이, WT-vehicle 및 WT-PD166793 그룹 모두 Latency 시간이 상당히 길어 어두운 방에서의 불쾌한 자극에 대한 학습과 기억이 잘 이루어진 것을 확인할 수 있었다. 반면에, 생리 식염수를 투여한 WT-vehicle 그룹을 기준으로 다른 그룹과 비교하였을 때, 5xFAD-vehicle 그룹의 step-through latency 시간이 확연히 짧은 것을 확인할 수 있었다. 5xFAD-vehicle 그룹 대비 5xFAD 마우스에 PD166793을 투약한 그룹(5xFAD-PD166793)에서는 step-through latency 시간이 상당히 많이 회복된 것을 확인할 수 있었다($$$$ p<0.0001).
이러한 결과는, 실시예 1의 약물인 PD166793 투약이 알츠하이머병 동물 모델에서 학습과 기억 능력 개선에 유의한 효과가 있다는 것을 의미한다.
실시예 13: 아밀로이드 플라크 감소 효과 - 면역조직화학검사
실시예 1의 약물이 실시예 10 내지 실시예 12의 행동 실험을 마친 알츠하이머병 마우스 모델의 해마 및 피질 뇌 조직에서 아밀로이드 플라크 형성에 영향을 주었는지 평가하기 위해서 면역조직화학검사(immunohistochemistry)를 수행하였다.
(1) 면역조직화학염색 방법
면역조직화학염색은 조직 내에 있는 특정 단백질을 염색하여 확인하는 기법이다. 전술한 실시예 10 내지 실시예 12에서 수행한 동물 행동실험 진행 후, 면역염색화학법을 위해 실험동물들의 뇌 조직을 회수해 절편화 작업을 거쳐 MHCII(Major histocompatibility complex II) 항체와 티오플라빈 S(Thioflavin S, Thio-S)를 이용하여 뇌 조직 절편을 염색하였다. Thio-S는 아밀로이드 베타 피브릴에 결합하여 초록색 형광을 나타내므로, 아밀로이드 플라크를 염색하기 위해 사용하였다. 미세아교세포는 평소에는 MHCII를 표면에 거의 발현하지 않다가, 병리적 맥락에서 활성화된 미세아교세포는 MHCII를 발현하므로, 활성화된 미세아교세포(activated microglia)를 염색하기 위해 MHCII 항체를 사용하였다. 뇌(brain)의 아밀로이드 플라크 근처에 집중되어 있는 미세아교세포의 활성화(activation)와 증식은 알츠하이머병의 두드러진 특징 중 하나이다. DAPI (4',6-diamidino-2-phenylindole)는 아데닌-티아민이 많은 영역의 DNA에 결합하여 파란색 형광을 나타내므로, 핵을 관찰할 수 있다.
행동실험에 사용한 동물인 WT-vehicle, WT-PD166793, 5xFAD-vehicle 및 5xFAD-PD166793의 뇌 조직을 회수하여 한 반구(hemisphere)를 고정하고 얼려서 절편화 작업을 진행하였다. 각 절편(slice)은 22 μm 두께로 제작하였다. 절편화된 뇌 조직을 MHCII 항체와 티오플라빈 S(Thioflavin S)로 염색하고 슬라이드 글라스(slide glass)에 DAPI가 포함된 Vectashield®를 활용해 마운팅(mounting)하였다. 염색된 조직들은 형광 현미경을 통해 확인하고 이미지를 얻었다. 현미경 이미지는 도 11a 및 도 11b에 제시하였다. 현미경 이미지에서 Thio-S가 양성인 경우, 아밀로이드 플라크가 형성되었다고 판단하였으며, MHCII가 양성인 경우, 활성화된 미세아교세포(activated microglia)가 존재한다고 판단하였다.
(2) 면역조직화학염색 결과
도 11a 및 도 11b에 제시된 이미지를 분석하고 정량화하여 그 결과를 도 12a 내지 도 12d에 제시하였다. 도 11a는 피질 부분의 현미경 사진을 나타내고, 도 11b는 해마 치상회(hippocampal dentate gyrus, hippocampus DG) 부분의 현미경 사진을 나타낸다. 도 11a 및 도 11b에서 붉은색 점선 박스 부분을 확대하여 우측에 Thio-S와 MHCII 염색 이미지를 나타내었다. 도 12a 및 도 12c는 각각 피질과 해마 치상회의 슬라이드 당 플라크 수(N)를 나타낸 그래프이다. 도 12b 및 도 12d는 ROI(region of interest)의 형광 세기를 정량화하여 비율을 나타낸 그래프이다. 각 그룹에서 실험동물 5마리를 선정하여 분석하였다.
관찰 결과, WT-vehicle 그룹 동물들의 피질(cortex) 및 해마(hippocampus)에서는 아밀로이드 플라크(amyloid plaque)를 찾아볼 수 없었다. 이러한 결과는, 동물실험에 사용된 해당 동물이 조직학적으로 야생형(wild type)에 해당한다는 것을 보여준다.
5xFAD-vehicle 및 5xFAD-PD166793 그룹 동물들의 피질 및 해마에서 모두 아밀로이드 플라크가 관찰되었다. 이러한 결과는, 동물실험에 사용된 해당 동물이 조직학적으로 형질전환 마우스(Tg mouse)에 해당한다는 것을 보여준다. 그러나, 5xFAD 마우스에 실시예 1의 약물인 PD166793을 투약한 그룹에서는 아밀로이드 플라크가 유의하게 감소한 것으로 확인되었다. 10 mg/kg 용량의 PD166793 투약이 피질과 해마에서 아밀로이드 플라크를 감소시키는 효과를 나타내었음을 보여준다.
또한 WT-saline 그룹 동물들의 피질 및 해마에서는 MHCII 양성반응을 나타내는 활성화된 미세아교세포를 찾아볼 수 없었고, 5xFAD-vehicle 및 5xFAD-PD166793 그룹 동물들의 피질 및 해마에서 모두 MHCII 양성반응을 나타내는 활성화된 미세아교세포가 관찰되었다. 이러한 결과는, 동물실험에 사용된 형질전환 마우스(Tg mouse)에서 신경면역반응이 증가되어 있음을 보여준다. 그러나, 5xFAD 마우스에 PD166793을 투약한 그룹에서는 MHCII 양성반응을 나타내는 활성화된 미세아교세포의 수가 유의하게 감소되어 있는 것으로 확인되었다. 이러한 결과는 10 mg/kg 용량의 PD166793 투약이 피질과 해마에서 신경면역반응 및/또는 신경염증반응을 현저히 감소시키는 효과를 나타냈음을 보여준다.
상기 결과로부터 본 발명자들은 실시예 1의 약물이 알츠하이머병 마우스 모델의 아밀로이드 플라크 형성과 신경면역반응 및/또는 신경염증반응을 유의하게 감소시키는 효과가 있음을 in vivo에서 확인하였다.
결론
본 발명자들은 상기 실시예에서 살펴본 바와 같이 본 발명의 약물 N-[(4'-브로모[1,1'-바이페닐]-4-일)설포닐]-L-발린, 즉 PD166793이 i) 아밀로이드 베타의 피브릴화 및 올리고머화 모두를 저해하고(실시예 2 및 3), ii) 약물 자체의 신경세포 독성을 나타내지 않으며(실시예 5), iii) 신경세포에서 아밀로이드 베타에 의해 유도되는, 세포 사멸 억제 효과(실시예 6), ROS 생산 억제 효과(실시예 7) 및 미토콘드리아 기능 장애 회복 효과(실시예 8)가 in vitro에서 있고, vi) 알츠하이머병 마우스 모델의 인지 능력, 공간 기억력 및 학습과 기억 능력을 개선시키는 효과(실시예 10 내지 실시예 12)와 뇌 피질 및 해마에서 아밀로이드 플라크 형성 및 신경면역반응 또는 신경염증반응의 감소 효과(실시예 13)가 in vivo에서 있음을 확인하였다.
따라서, 상기 결과들로부터 본 발명의 N-[(4'-브로모[1,1'-바이페닐]-4-일)설포닐]-L-발린, 즉 PD166793을 유효성분으로 포함하는 조성물은 아밀로이드 베타(Amyloid beta, Aβ)의 올리고머화(oligomerization) 및 피브릴화(fibrillization) 억제하는 데 유용하게 사용될 수 있고, 알츠하이머병을 예방, 개선 또는 치료를 위해 유용하게 사용될 수 있으며, 기억력, 인지 능력 및 학습 능력을 개선하기 위해 사용될 수 있다.
Claims (16)
- 제1항에 있어서, 상기 아밀로이드 베타는 조직, 뇌척수액, 혈청, 혈장, 전혈 및 간질액으로 이루어진 군으로부터 선택되는 적어도 하나 이상에 포함되는 것인, 조성물.
- 제1항에 있어서, 상기 조성물은 신경세포 사멸을 억제하는 것인, 조성물.
- 제1항에 있어서, 상기 조성물은 아밀로이드 플라크(amyloid plaque) 형성을 억제하는 것인, 조성물.
- 제1항에 있어서, 상기 조성물은 활성산소종(reactive oxygen species, ROS)을 감소시키는 것인, 조성물.
- 제5항에 있어서, 상기 활성산소종은 과산화수소(hydrogen peroxide, H2O2), 과산화물(peroxide), 초과산화물(superoxide, O2 -), 하이드록실 라디칼(hydroxyl radical, ·OH), 알콕시 라디칼(alkoxy radical, RO·), 퍼옥시 라디칼(peroxy radical, ROO·), 유기 하이드로퍼옥사이드(organic hydroperoxide, ROOH), 일중항산소(singlet oxygen, 1O2) 및 알파-산소(alpha-oxygen, α-O)로 이루어진 군으로부터 선택되는 1종 이상인 것인, 조성물.
- 제1항에 있어서, 상기 조성물은 미토콘드리아 막 전위(mitochondrial membrane potential, ΔΨm) 손실을 회복시키는 것인, 조성물.
- 제1항에 있어서, 상기 조성물은 미세아교세포(microglia)의 활성화를 억제하는 것인, 조성물.
- 제1항 내지 제8항 중 어느 한 항에 따른 아밀로이드 베타(Amyloid beta, Aβ)의 올리고머화(oligomerization) 및 피브릴화(fibrillization) 억제용 조성물을 포함하는 신경보호에 의한 퇴행성 신경질환의 예방 또는 치료용 약제학적 조성물.
- 제9항에 있어서, 상기 퇴행성 신경질환은 인지 장애(cognitive disorder), 치매(dementia), 루이소체치매(Lewy body dementia), 알츠하이머병(Alzheimer's disease), 파킨슨병(Parkinson's diseases), 헌팅턴병(Huntington's disease), 근위축성 측삭경화증(Amyotrophic lateral sclerosis), 다발성 경화증(Multiple sclerosis), 허혈성 뇌손상(ischemic brain injury), 뇌염(encephalitis), 뇌수막염(meningitis), 길랑-바레 증후군(Guillain-Barre syndrome) 및 외상성 뇌손상(traumatic brain injury)으로 이루어진 군으로부터 선택된 것인, 약제학적 조성물.
- 제1항 내지 제8항 중 어느 한 항에 따른 아밀로이드 베타(Amyloid beta, Aβ)의 올리고머화(oligomerization) 및 피브릴화(fibrillization) 억제용 조성물을 포함하는 인지 능력 개선, 기억력 개선 또는 학습 능력 개선용 약제학적 조성물.
- 제1항 내지 제8항 중 어느 한 항에 따른 조성물을 포함하는 아밀로이드 베타(Amyloid beta, Aβ)의 올리고머화(oligomerization) 및 피브릴화(fibrillization) 억제용 기능성 식품 조성물.
- 제12항에 있어서, 상기 기능성 식품 조성물은 신경보호에 의해 퇴행성 신경질환을 예방 또는 개선하는 것인 것을 특징으로 하는, 기능성 식품 조성물.
- 제13항에 있어서, 상기 퇴행성 신경질환은 인지 장애(cognitive disorder), 치매(dementia), 루이소체치매(Lewy body dementia), 알츠하이머병(Alzheimer's disease), 파킨슨병(Parkinson's diseases), 헌팅턴병(Huntington's disease), 근위축성 측삭경화증(Amyotrophic lateral sclerosis), 다발성 경화증(Multiple sclerosis), 허혈성 뇌손상(ischemic brain injury), 뇌염(encephalitis), 뇌수막염(meningitis), 길랑-바레 증후군(Guillain-Barre syndrome) 및 외상성 뇌손상(traumatic brain injury)으로 이루어진 군으로부터 선택된 것인, 기능성 식품 조성물.
- 제1항 내지 제8항 중 어느 한 항에 따른 조성물을 포함하는 인지 능력 개선, 기억력 개선 또는 학습 능력 개선용 기능성 식품 조성물.
- 제1항 내지 제8항 중 어느 한 항에 따른 조성물을 이를 필요로 하는 대상체(subject)에게 투여하는 단계를 포함하는 퇴행성 신경질환의 치료 방법.
