本発明のキットは、AAVの製造に用いるものであって、第一のベクターと第二のベクターとを含むベクター配列の組み合わせを含んでいればよく、緩衝液組成物や精製水などに溶解した状態や分散した状態であってもよい。ベクターを導入する試薬などの本発明を実施するのに有用な組成物と組み合わせてもよいし、適切な容器、例えば、バイアル、チューブ、試験管またはほかの容器を含んでいてもよい。
本明細書において、ある遺伝子が天然に保持するプロモーターとは、人工的ではなく、天然状態においてその遺伝子を発現させることが知られているプロモーターを意味する。
第一のベクターは、1以上のウイルスヘルパー遺伝子を含み、上記ウイルスヘルパー遺伝子のうちの少なくとも一つが発現調節可能なプロモーターの制御下にある。
特に好ましくは、第一のベクターにおける発現調節可能なプロモーターは、テトラサイクリンにより発現調節可能なプロモーターであるTetオン/オフシステムであって、最も好ましくは、Tetオンシステムである。プロモーターは特に限定されないが、サイトメガロウイルス由来のプロモーター(所望によりエンハンサーを含む)、SV40初期プロモーター、ヒト伸長因子-1α(EF-1α)プロモーター、ヒトユビキチンCプロモーター、レトロウイルスのラウス肉腫ウイルスLTRプロモーター、ジヒドロ葉酸還元酵素プロモーター、β-アクチンプロモーター、及びホスホグリセリン酸キナーゼ(PGK)プロモーター、H1プロモーター(RNA PolymeraseIIIプロモーター)、U6プロモーター(RNA PolymeraseIIIプロモーター)などを挙げることができる。
VA-RNA遺伝子発現調節用のTetオンシステムに用いるプロモーターとしてはH1プロモーター(RNA PolymeraseIIIプロモーター)であってもよい。
Tetオンシステムにおいて、Tet operator配列は、発現を調節したいプロモーターの上流にあっても下流にあってもよい。発現をオフにしたときの発現漏れを減らすという点において、Tet operator配列は、発現調節可能なプロモーターの下流にあることが好ましい。
好ましくは、第一のベクターに含まれるウイルスヘルパー遺伝子が、E2遺伝子、E4遺伝子、およびVA-RNA遺伝子である。
より好ましくは、第一のベクターに含まれるウイルスヘルパー遺伝子は、E4遺伝子、およびVA-RNA遺伝子である。
E4遺伝子は、好ましくは、E4ORF6遺伝子である。
VA-RNA遺伝子は、好ましくは、VA-RNAI遺伝子である。
第一のベクターに含まれるウイルスヘルパー遺伝子のうち、E2遺伝子が発現調節可能なプロモーターの制御下にないことが好ましい。
第一のベクターに含まれるウイルスヘルパー遺伝子のうち、E4遺伝子のみが発現調節可能なプロモーターの制御下にあってもよい。第一のベクターに含まれるウイルスヘルパー遺伝子のうち、VA-RNA遺伝子のみが発現調節可能なプロモーターの制御下にあってもよい。好ましくは、第一のベクターに含まれるウイルスヘルパー遺伝子のうち、E4遺伝子およびVA-RNA遺伝子が発現調節可能なプロモーターの制御下にある。
第二のベクターは、1以上のアデノ随伴ウイルス遺伝子を含み、上記のアデノ随伴ウイルス遺伝子の少なくとも一つが、薬剤による発現調節プロモーターの制御下にはない。
好ましくは、第二のベクターにおいて、薬剤による発現調節プロモーターの制御下にないアデノ随伴ウイルス遺伝子のいずれか一つが、プロモーターの制御下にあり、より好ましくはアデノ随伴ウイルス遺伝子が天然に保持するプロモーターの制御下にある。
好ましくは、第二のベクターにおいて、全てのアデノ随伴ウイルス遺伝子が発現調節プロモーターの制御下になく、より好ましくは、全てのアデノ随伴ウイルス遺伝子が天然に保持するプロモーターの制御下にある。
rep遺伝子が天然に保持するプロモーターとしてはp5及びp19プロモーターを挙げることができる。cap遺伝子が天然に保持するプロモーターとしてはp40プロモーターを挙げることができる。
本発明はさらに、1以上のウイルスヘルパー遺伝子を含む第一のベクターであって、上記ウイルスヘルパー遺伝子のうちの少なくとも一つが発現調節可能なプロモーターの制御下にある、第一のベクターと、1以上のアデノ随伴ウイルス遺伝子を含む第二のベクターであって、上記アデノ随伴ウイルス遺伝子の少なくとも一つが、薬剤による発現調節プロモーターの制御下にはない、第二のベクターとを連結した状態で有する、統合ベクターに関する。連結した状態とは、直接もしくは別の配列で間接的につながっている状態を指す。第一のベクターまたは第二のベクターが環状であるときには、ベクターの配列の一部を切断することで、直鎖状にし、連結したものを統合ベクターとすることができる。
本発明の統合ベクターとしては、実施例で使用した図11に構造を示すSeq43のように全塩基配列を合成したものでもよい。図11に示すベクターにおいてはE4遺伝子とその上流のプロモータ及びVA-RNA遺伝子とその上流のプロモーターが第一のベクターに対応し、rep遺伝子及びcap遺伝子とその上流のプロモーター第二ベクターに対応し、図11に示す統合ベクターにおいては、第一のベクターと第二ベクターとが連結した状態で存在している。
統合ベクターにおける第一のベクター及び第二のベクターの構成は、本明細書中において上記の<キット>において説明した通りである。
第三のベクターは、所望の治療又は予防用遺伝子を含むベクターである。
第三のベクターとしては核酸を含むものであれば限定されるものではないが、例えば、プラスミド、人工染色体、ウイルスなどを使用することができ、好ましくは、プラスミドである。
本発明は、上記した第一のベクターと第二のベクターとを有する細胞に関する。本発明はさらに、上記した統合ベクターを有する細胞に関する。本発明はさらに、第一のベクターと第二のベクターと第三のベクターを有する細胞に関する。
本発明の細胞は、第一のベクターと第二のベクター、統合ベクター、または第一のベクターと第二のベクターと第三のベクターとを細胞内に取り込んだ後、1週間以上、前記導入されたベクターのうちのアデノ随伴ウイルスの産生に必要な遺伝子を細胞内に保持することができる。そのため、前記ベクターを導入した細胞を1週間以上培養した後、アデノ随伴ベクターを産生させることが可能となる。好ましくは、2週間以上培養した後に、アデノ随伴ベクターを産生させることが可能であり、更に好ましくは1か月以上培養した後に、アデノ随伴ベクターを産生させることが可能である。
本発明によれば、1以上のウイルスヘルパー遺伝子を含む第一のベクターであって、上記ウイルスヘルパー遺伝子のうちの少なくとも一つが発現調節可能なプロモーターの制御下にある、第一のベクターと、1以上のアデノ随伴ウイルス遺伝子を含む第二のベクターであって、上記アデノ随伴ウイルス遺伝子の少なくとも一つが、薬剤による発現調節プロモーターの制御下にはない、第二のベクターとを細胞に導入することを含む、アデノ随伴ウイルスを産生するための細胞の製造方法が提供される。更に第三のベクターを含む、アデノ随伴ウイルスを産生するための細胞の製造方法が提供される。第一のベクター、第二のベクター及び第三のベクターの構成は、本明細書中において上記の<キット>において説明した通りである。第一のベクター、第二のベクター及び第三のベクターが、発現されるように導入されていればよいが、永続的な発現となることがより好ましい。永続的な発現とは、細胞が***した場合に、第一のベクター、第二のベクター及び第三のベクターも複製され、***後の細胞も第一のベクター、第二のベクター及び第三のベクターを有することになることをいう。永続的な発現は、ベクターが細胞の染色体の中に組み込まれることや細胞質内で複製する能力をもつベクターを用いることによって行うことができる。
トランスフェクションとは、化学的手段(例えば、リン酸カルシウム仲介沈殿)、機械的手段(例えば、エレクトロポレーション)または物理的手段(例えば、生体弾(bioballistic)による送達)によって真核細胞の膜を横断して核酸を細胞に導入することをいう。
トランスダクションとは、ウイルス由来のベクターを介して、真核細胞の膜を横断して、核酸を細胞に導入することをいう。
リポフェクションとは、電気的な相互作用によりベクターと陽性荷電脂質などとの複合体を形成させ、エンドサイトーシスや膜融合により、核酸を細胞に導入することをいう。
本発明によるアデノ随伴ウイルスの製造方法の一例としては、
1以上のウイルスヘルパー遺伝子を含む第一のベクターであって、上記ウイルスヘルパー遺伝子のうちの少なくとも一つが発現調節可能なプロモーターの制御下にある、第一のベクターと、1以上のアデノ随伴ウイルス遺伝子を含む第二のベクターであって、上記アデノ随伴ウイルス遺伝子の少なくとも一つが、薬剤による発現調節プロモーターの制御下にはない、第二のベクターと、必要な場合には、所望の治療又は予防用遺伝子を含む第三のベクターとを含む細胞を培養する工程と、
上記発現調節可能なプロモーターを作動させることによって、発現調節可能なプロモーターの制御下にあるウイルスヘルパー遺伝子を発現させる工程と、
を含む方法が挙げられる。
ウイルスヘルパー遺伝子の一部と、アデノ随伴ウイルス遺伝子の一部を細胞に導入する第一の工程;
第一の工程で得られた細胞を培養して増殖させる第二の工程;

ウイルスヘルパー遺伝子の一部と、アデノ随伴ウイルス遺伝子と、必要な場合には、所望の治療又は予防用遺伝子とを細胞に導入する第一の工程;
第一の工程で得られた細胞を培養して増殖させる第二の工程;
上記第一の工程で得られた細胞が含んでいないウイルスヘルパー遺伝子を、上記第二の工程で得られた細胞に導入する第三の工程;及び

第二の工程では細胞の倍化時間は、200時間以内であればよく、100時間以内であれば好ましく、50時間以内であればより好ましく、30時間以内であれば特に好ましい。
第二の工程では細胞生存率は65%以上であればよく、80%以上が好ましく、95%以上であることがより好ましい。
アデノ随伴ウイルス遺伝子を細胞に導入する第一の工程;
第一の工程で得られた細胞を培養して増殖させる第二の工程;
ウイルスヘルパー遺伝子と、必要な場合には、所望の治療又は予防用遺伝子とを、上記第二の工程で得られた細胞に導入する第三の工程;及び

好ましくは、第一の工程で細胞に導入されるアデノ随伴ウイルス遺伝子がrep遺伝子およびcap遺伝子であり、第三の工程で細胞に導入されるウイルスヘルパー遺伝子がE2遺伝子、E4遺伝子及びVA-RNA遺伝子である。
ウイルスヘルパー遺伝子の一部と、必要な場合には、所望の治療又は予防用遺伝子とを細胞に導入する第一の工程;
第一の工程で得られた細胞を培養して増殖させる第二の工程;
上記第一の工程で得られた細胞が含んでいないウイルスヘルパー遺伝子と、アデノ随伴ウイルス遺伝子とを、上記第二の工程で得られた細胞に導入する第三の工程;及び
第三の工程で得られた細胞を培養する第四の工程
を含む方法が挙げられる。
好ましくは、第一の工程で細胞に導入されるウイルスヘルパー遺伝子がE2遺伝子であり、第三の工程で細胞に導入されるウイルスヘルパー遺伝子がE4遺伝子及びVA-RNA遺伝子であり、第三の工程で細胞に導入されるアデノ随伴ウイルス遺伝子がrep遺伝子およびcap遺伝子である。
CO2濃度は、一般的には3~10%CO2であり、好ましくは5~10%CO2であり、一例としては8%CO2である。
培養は振盪攪拌しながら行ってもよい。振盪攪拌する場合の攪拌速度は、一般的には50rpm~200rpmであり、好ましくは80~150rpmである。
攪拌培養は、リアクター内のプロペラ等による回転攪拌培養であっても良い。回転攪拌する場合の攪拌速度は、一般的には50rpm~200rpmであり、好ましくは80~150rpmである。また、波型振盪撹拌や、撹拌翼の上下動によって撹拌してもよいが、特に限定されない。
各工程における培養時間は、特に限定されないが、一般的には6時間~14日間であり、好ましくは12時間~7日間であり、より好ましくは24時間~144時間であり、さらに好ましくは24時間~96時間である。
<細胞培養>
浮遊化HEK293細胞(Thermo Fisher Scientific, R79007)を12mLのExpi29 Expression Medium(Thermo Fisher Scientific,A1435101)で1×107cells/mLとなるように懸濁し、125mL振とうフラスコで24時間培養した。培養液は120rpmで一定に攪拌しながら、37℃、8%CO2 条件下でインキュベートを行った。
AAVpro(登録商標)Helper Free System (AAV5)(タカラバイオ,6650)を使用した(Seq1、Seq2、Seq16)。
使用したプラスミド(Seq1~Seq16、Seq20~Seq23)の全体配列を、配列表の配列番号1~16、20~23に示す。

seq13(E4ORF6):PCRによりE4ORF6遺伝子の領域を増幅し、pEBMulti-Neoベクター(FUJIFILM Wako Pure Chemical Corporation)のApaI/XbaIサイトに挿入したプラスミド。

seq22(Switch2―VAI):U6プロモーター(RNA polymerase IIIプロモーター)とVA-RNAI(VAI)遺伝子をつないだ配列を合成し、pUC57ベクターのEcoRVサイトに挿入したプラスミド(GENEWIZ)。Switch1―VAIは、RNA polymerase IIIプロモーター上流にTetオペレーター配列を組み込んだ構造になっている。
seq23(tTS):合成したテトラサイクリン調節性の転写サイレンサー(tTS)遺伝子をpLVSIN-CMV HygベクターのClaI-XbaIサイトに挿入したプラスミド(GENEWIZ)。
Seq38(Switch―E4ORF6―Switch1―VAI―tTS):合成したE4ORF6遺伝子配列をコードしたプラスミド(GENEWIZ)に対して、PCRで増幅したSwitch1―VAI、tTSを挿入した。
Seq39(nRCp5):pRC5 Vector(seq2)をBsp14071/BglIIで制限酵素処理し、rep-cap断片を作製した。rep-cap断片をAAVS1 Safe Harbor cDNA/miRNA Donor Vector (pAAVS1D-PGK.MSC-Ef1α.copGFPpuro)(SBI社、GE603A-1)のBsp14071/BglIIサイト間に交換し、ライゲーションにより再環状化し、rep-capノックイン用プラスミド。
Seq40(p5RCp5):合成したp5プロモーター配列をSeq39に挿入したプラスミド。rep-cap遺伝子の前後にp5プロモーター配列を有する。
Seq41(E2-GOI):E2遺伝子およびAAV ITR配列に挟まれたGFP遺伝子およびpuromycin耐性遺伝子、およびpiggyBacトランスポゾン配列を持つ遺伝子配列を合成しpUC57ベクターのEcoRVサイトに挿入したプラスミド(GENEWIZ)。
Seq42:piggyBac トランスポザーゼ遺伝子配列を合成し、pUC57ベクターのEcoRVサイトに挿入したプラスミド(GENEWIZ)。
Seq43(all-in-one):合成によって作製したREP遺伝子、CAP遺伝子、E2遺伝子、Switch―E4ORF6、Switch1―VAI、tTSを組み込んだプラスミド。seq43のプラスミドマップを図11に示す。
鋳型10pg DNAに対して、終濃度0.2μmol/L Primer1およびPrimer2、PrimeSTAR Max Premix(1×)を混合し、ヌクレアーゼフリーwater(Thermo Fisher Scientific,10977015)で総量50μLに調製した。前述のプラスミド作製に使用したprimer1およびprimer2を以下に示す。seq24~seq37の塩基配列を、配列表の配列番号24~37に示す。
seq3とseq4をインバースPCRで作成するのには、鋳型としてseq2のプラスミドを用い、seq5~7、seq14、seq15、seq20をインバースPCRで作成するのには鋳型として、seq1を用いた。以下に(作成したプラスミド):(インバースPCRで用いたプライマーセット)を示す。

1.8mLのExpi29 Expression MediumにPolyethylenimine(PEI)(PolyPlus-transfection SAS, 115-0015)36μLと、後記する組み合わせのプラスミドを等量混合し総量18μgを懸濁し、15分間静置した。その後、前日に細胞濃度1x107cells/mLで播種した培養細胞(培養液量は12mL)に、上記試薬を添加し、37℃、8%CO2条件下でインキュベートを行った。
導入細胞の障害性は、遺伝子導入後day4、day7、day11においてtrypan blue染色による細胞の生存率によって評価を実施した。障害細胞率(%)=100-(細胞の生存率)で算出を行った。
細胞を遠心分離により回収し、RIPA Bufferで溶解した。細胞溶液にSample Buffer(ナカライ,09499-14)を添加し、100℃で5分間加熱してサンプルを調製した。ドデシル硫酸ナトリウム(SDS)ゲル電気泳動装置(アトー,2321670)を用いてサンプル中のタンパク質をサイズ分離後、セミドライブロッティング装置(アトー,2322490)を用いてメンブレンに転写を行った。転写後のメンブレンはBlocking One(ナカライテスク,03953-95)を用いて30分間ブロッキング後、ブロッキング溶液で70倍に希釈した抗REP抗体(OriGene Technologies)、カプシドの構造タンパク質に反応する抗VP抗体(PROGEN Biotechnik GmbH)、抗β-Actin抗体(Abcam)で常温、3時間反応させた。次に、二次抗体としてブロッキング溶液で10,000倍に希釈したanti-mouse IgG-HRP(Cytiva)を常温、2時間反応後、SuperSignal West Femto Maximum Sensitivity Substrate(Thermo Fisher Scientific,34095)によりREPタンパク質およびカプシドの構造タンパク質(VP1、VP2、VP3)を検出した。撮影にはWSE-6100LuminoGraphI(アトー,2006100)を用いた。取得画像の解析にはImageJを用いて定量評価を実施した。
遺伝子導入後72時間、37℃、8%CO2条件下で培養を行ったHEK細胞培養液を回収し、-80℃での凍結融解にて細胞を破砕した。その後、13,800×gの遠心分離によって上清を回収し、終濃度2mmol/L MgCl2、25U/mL Benzonaseを添加し、37℃で2時間反応を行い、gDNA(ゲノムDNA)や残存プラスミドを消化した。反応後のサンプルは、95℃で15分、70℃で3分、40℃で3分、20℃で3分の順にインキュベーションし、Benzonaseを失活させ、これをddPCR(Droplet Digital PCR)サンプルとした。ddPCRサンプルはTE buffer(pH8.0,0.05%Pluronic F-68 および10μg/mLウシ胸腺DNA含有)で50倍に希釈した。氷上でチューブにddPCRtm Supermix for Probes(no dUTP)(BIORAD社) 10μL、希釈済みのddPCRサンプル2μL、プライマー(seq17、seq18、最終濃度900nmol/L)およびプローブ(seq19、最終濃度250nmol/L)、ヌクレアーゼフリーwater(Thermo Fisher Scientific,10977015)を混合して、総液量22μLに調整し、ddPCRを行った。seq17、seq18及びseq19の配列を、配列表の配列番号17~19に示す。
1.8mLのExpi29 Expression MediumにPolyethylenimine(PEI)(PolyPlus-transfection SAS,115-0015)36μLと、後記する組み合わせのプラスミドを等量混合し総量18μgを懸濁し、15分間静置した。その後、前日に細胞濃度1x107cells/mLで播種した培養細胞(培養液量は12mL)に、上記試薬を添加し、37℃、8%CO2条件下でインキュベートを行った。遺伝子導入した細胞を3週間維持培養し、フローサイトメトリーを用いてRFPもしくはGFP蛍光陽性細胞をシングルセルで単離した。単離した細胞は、10%ウシ胎児血清を含有したDMEMに懸濁し、6ウェルプレート上で37℃、8%CO2 条件下で1ヵ月培養し、細胞株を樹立した。
HEK293細胞に、以下の組合せで遺伝子を導入し、細胞障害性を評価した結果を図1に示す。

図1の結果から、Day11においては、E2遺伝子、E4遺伝子、VA-RNA遺伝子を除くことで細胞障害が低減されていた。
さらに図1の結果から、E4遺伝子,VA-RNA遺伝子が無いことで細胞の障害を回避できることが確認された。

p5RCp5: p5前後付加rep-cap遺伝子挿入細胞、seq1、seq20、seq16
上記で樹立したrep-cap株について、1か月培養を行った。その後、ベクター配列存在時のみ増幅されるように設計したプライマーおよびプローブを用いたドロップレットデジタルPCR法によって、細胞から抽出した染色体をテンプレートとした場合にPCR増幅されることを確認することで、導入した配列が細胞の染色体中に組込まれていることを確認した。

図8の結果から、発現制御をしていないrep-cap遺伝子を挿入した細胞株が樹立でき、さらに、E2遺伝子、E4遺伝子、VA-RNA遺伝子、GOI遺伝子を後から導入することでAAVを産生できることを確認した。
p5RCp5について、NGS解析を実施し、細胞のシークエンスを読み込むことで、rep遺伝子およびcap遺伝子が染色体中に組み込まれていることを確認した。
上記で樹立した2株のE2-GOI株について、1か月培養を行った。その後、ベクター配列存在時のみ増幅されるように設計したプライマーおよびプローブを用いたドロップレットデジタルPCR法によって、細胞から抽出した染色体をテンプレートとした場合にPCR増幅されることを確認することで、導入した配列が細胞の染色体中に組込まれていることを確認した。

図9の結果から、E2遺伝子-GOI遺伝子を挿入した細胞株が樹立でき、さらに、REP遺伝子、CAP遺伝子、E4遺伝子、VA-RNA遺伝子を後から導入することでAAVを産生できることを確認した。

- 1以上のウイルスヘルパー遺伝子を含む第一のベクターであって、前記ウイルスヘルパー遺伝子のうちの少なくとも一つが発現調節可能なプロモーターの制御下にある、第一のベクターと、
1以上のアデノ随伴ウイルス遺伝子を含む第二のベクターであって、前記アデノ随伴ウイルス遺伝子の少なくとも一つが、薬剤による発現調節プロモーターの制御下にはない、第二のベクター、
とを含むキット。 - 第一のベクターにおける発現調節可能なプロモーターが、薬剤による発現調節可能なプロモーターである、請求項1に記載のキット。
- 第一のベクターに含まれるウイルスヘルパー遺伝子が、アデノウイルス由来である請求項1または2に記載のキット。
- 第一のベクターに含まれるウイルスヘルパー遺伝子のうち、E4遺伝子およびVA-RNA遺伝子が発現調節可能なプロモーターの制御下にある、請求項1から3のいずれか一項に記載のキット。
- 第一のベクターに含まれるウイルスヘルパー遺伝子の全てが、発現調節可能なプロモーターの制御下にある、請求項1から4のいずれか一項にに記載のキット。
- 第一のベクターに含まれるウイルスヘルパー遺伝子が、E2遺伝子、E4遺伝子、およびVA-RNA遺伝子である、請求項1から5のいずれか一項に記載のキット。
- E4遺伝子が、E4ORF6遺伝子である、請求項6に記載のキット。
- VA-RNA遺伝子が、VA-RNAI遺伝子である、請求項6または7に記載のキット。
- 第二のベクターにおいて、全てのアデノ随伴ウイルス遺伝子が、薬剤による発現調節プロモーターの制御下にはない、請求項1から8の何れか一項に記載のキット。
- 第二のベクターにおいて、薬剤による発現調節プロモーターの制御下にないアデノ随伴ウイルス遺伝子のいずれか一つが、アデノ随伴ウイルス遺伝子が天然に保持するプロモーターの制御下にある、請求項1から9の何れか一項に記載のキット。
- 第二のベクターに含まれるアデノ随伴ウイルス遺伝子が、rep遺伝子およびcap遺伝子である、請求項1から10のいずれか一項に記載のキット。
- 第二のベクターに含まれるアデノ随伴ウイルス遺伝子のうち、薬剤による発現調節プロモーターの制御下にないアデノ随伴ウイルス遺伝子がrep遺伝子である、請求項1から11のいずれか一項に記載のキット。
- 請求項1から12の何れか一項に記載のキットに含まれる第一のベクターと第二のベクターとを有する細胞。
- 細胞がヒト細胞である、請求項13に記載の細胞。
- 前記第一のベクターと前記第二のベクターとを染色体中に有する、請求項13又は14に記載の細胞。
- 1以上のウイルスヘルパー遺伝子を含む第一のベクターであって、前記ウイルスヘルパー遺伝子のうちの少なくとも一つが発現調節可能なプロモーターの制御下にある、第一のベクターと、
1以上のアデノ随伴ウイルス遺伝子を含む第二のベクターであって、前記アデノ随伴ウイルス遺伝子の少なくとも一つが、薬剤による発現調節プロモーターの制御下にはない、第二のベクター
とを連結した状態で有する、統合ベクター。 - 所望の治療又は予防用遺伝子を含む第三のベクターを更に有する、請求項16に記載の統合ベクター。
- 請求項16又は17に記載の統合ベクターを有する細胞。
- 細胞がヒト細胞である、請求項18に記載の細胞。
- 前記統合ベクターを染色体中に有する、請求項18又は19に記載の細胞。
- 1以上のウイルスヘルパー遺伝子を含む第一のベクターであって、前記ウイルスヘルパー遺伝子のうちの少なくとも一つが発現調節可能なプロモーターの制御下にある、第一のベクターと、1以上のアデノ随伴ウイルス遺伝子を含む第二のベクターであって、前記アデノ随伴ウイルス遺伝子の少なくとも一つが、薬剤による発現調節プロモーターの制御下にはない、第二のベクターとを細胞に導入することを含む、アデノ随伴ウイルスを産生するための細胞の製造方法。
- 1以上のウイルスヘルパー遺伝子を含む第一のベクターであって、前記ウイルスヘルパー遺伝子のうちの少なくとも一つが発現調節可能なプロモーターの制御下にある、第一のベクターと、1以上のアデノ随伴ウイルス遺伝子を含む第二のベクターであって、前記アデノ随伴ウイルス遺伝子の少なくとも一つが、薬剤による発現調節プロモーターの制御下にはない、第二のベクターと、必要な場合には、所望の治療又は予防用遺伝子を含む第三のベクターとを含む細胞を培養する工程と、
前記発現調節可能なプロモーターを作動させることによって、発現調節可能なプロモーターの制御下にあるウイルスヘルパー遺伝子を発現させる工程と、
を含む、アデノ随伴ウイルスの製造方法。 - ウイルスヘルパー遺伝子の一部と、アデノ随伴ウイルス遺伝子の一部を細胞に導入する第一の工程;
第一の工程で得られた細胞を培養して増殖させる第二の工程;
前記第一の工程で得られた細胞が含んでいないウイルスヘルパー遺伝子、前記第一の工程で得られた細胞が含んでいないアデノ随伴ウイルス遺伝子、及び必要な場合には、所望の治療又は予防用遺伝子を、前記第二の工程で得られた細胞に導入する第三の工程;及び
第三の工程で得られた細胞を培養する第四の工程
を含む、アデノ随伴ウイルスの製造方法。 - 第一の工程で細胞に導入されるウイルスヘルパー遺伝子がE2遺伝子であり、第一の工程で細胞に導入されるアデノ随伴ウイルス遺伝子がrep遺伝子であり、第三の工程で細胞に導入されるウイルスヘルパー遺伝子がE4遺伝子及びVA-RNA遺伝子であり、第三の工程で細胞に導入されるアデノ随伴ウイルス遺伝子がcap遺伝子である、請求項23に記載のアデノ随伴ウイルスの製造方法。
- ウイルスヘルパー遺伝子の一部と、アデノ随伴ウイルス遺伝子と、必要な場合には、所望の治療又は予防用遺伝子とを細胞に導入する第一の工程;
第一の工程で得られた細胞を培養して増殖させる第二の工程;
前記第一の工程で得られた細胞が含んでいないウイルスヘルパー遺伝子を、前記第二の工程で得られた細胞に導入する第三の工程;及び
第三の工程で得られた細胞を培養する第四の工程
を含む、アデノ随伴ウイルスの製造方法。 - 第一の工程で細胞に導入されるウイルスヘルパー遺伝子がE2遺伝子であり、第一の工程で細胞に導入されるアデノ随伴ウイルス遺伝子がrep遺伝子およびcap遺伝子であり、第三の工程で細胞に導入されるウイルスヘルパー遺伝子がE4遺伝子及びVA-RNA遺伝子である、請求項25に記載のアデノ随伴ウイルスの製造方法。
- アデノ随伴ウイルス遺伝子を細胞に導入する第一の工程;
第一の工程で得られた細胞を培養して増殖させる第二の工程;
ウイルスヘルパー遺伝子と、必要な場合には、所望の治療又は予防用遺伝子とを、前記第二の工程で得られた細胞に導入する第三の工程;及び
第三の工程で得られた細胞を培養する第四の工程
を含む、アデノ随伴ウイルスの製造方法。 - 第一の工程で細胞に導入されるアデノ随伴ウイルス遺伝子がrep遺伝子およびcap遺伝子であり、第三の工程で細胞に導入されるウイルスヘルパー遺伝子がE2遺伝子、E4遺伝子及びVA-RNA遺伝子である、請求項27に記載のアデノ随伴ウイルスの製造方法。
- ウイルスヘルパー遺伝子の一部と、必要な場合には、所望の治療又は予防用遺伝子とを細胞に導入する第一の工程;
第一の工程で得られた細胞を培養して増殖させる第二の工程;
前記第一の工程で得られた細胞が含んでいないウイルスヘルパー遺伝子と、アデノ随伴ウイルス遺伝子とを、前記第二の工程で得られた細胞に導入する第三の工程;及び
第三の工程で得られた細胞を培養する第四の工程
を含む、アデノ随伴ウイルスの製造方法。 - 第一の工程で細胞に導入されるウイルスヘルパー遺伝子がE2遺伝子であり、第三の工程で細胞に導入されるウイルスヘルパー遺伝子がE4遺伝子及びVA-RNA遺伝子であり、第三の工程で細胞に導入されるアデノ随伴ウイルス遺伝子がrep遺伝子およびcap遺伝子である、請求項29に記載のアデノ随伴ウイルスの製造方法。
