바이오센서는 그 변환방법에 따라, 전기화학센서, 열감지센서, 광학센서 등으로 구분되며, 최근에는, 분석하고자 하는 대상 물질의 종류에 따라, 글루코오스 센서, 세포 센서, 면역 바이오센서, DNA 칩 등으로 다양하게 명명된다.
이 중, 전기화학센서는 생물학적인 시료의 양을 정보처리가 쉬운 전기신호로 전환이 가능하다는 측면에서, 현재까지 바이오센서의 변환방식으로 널리 사용되고 있다.

상기 공개특허 제10-2013-0033330호의 바이오센서뿐만 아니라 대부분의 종래 바이오 센서, 특히 젖산 혹은 당뇨 측정기는, 비교적 일정한 산성도를 나타내는 혈액을 시료로 사용하거나, 낮은 농도의 타액이 일부 측정이 가능하여, 산성도에 따른 측정 편차가 크게 발생하지 않는다.
그러나, 인체로부터 발생하는 땀, 타액 등과 같이 pH가 약 4 내지 8 정도인 시료를 측정 시료로 사용하는 바이오센서의 경우 산성도에 따른 측정 편차가 크게 발생하게 되어 현재 개발된 바이오센서에 의해서는 정확한 측정이 어려운 문제가 있다.
따라서, 넓은 범위의 대상 물질(analyte)의 농도를 측정할 수 있으면서도, pH에 따른 영향성이 저감된 바이오센서의 개발이 필요하다.
본 발명은, 간접 및 비접촉 방법으로 Buffer strength를 향상시켜 측정 시료의 pH에 따른 측정 편차를 최소화 함으로써, 정확성과 정밀성이 향상된 바이오센서를 제공하는 것을 발명의 목적으로 한다.
또한, 본 발명은, 외부 불순물질의 여과력을 향상시킴으로써, 정확성과 정밀성이 향상된 바이오센서를 제공하는 것을 발명의 목적으로 한다.
본 발명은, 기판; 상기 기판 상에 형성되는 작업전극과 기준전극을 포함하며, 상기 작업전극은, 상기 기판 상에 형성되는 작업전극층; 상기 작업전극층 상에 형성되는 효소반응층; 상기 효소반응층 상에 형성되는 고분자막층; 및 상기 고분자막층 상에 형성되는 버퍼층을 포함하는, 바이오센서에 관한 것이다.
본 발명은, 그 제1 관점에 있어서, 상기 버퍼층은 pH 완충제를 포함하는 것일 수 있다.

본 발명의 일 실시 예에 따른 바이오센서는, 작업전극의 상부에 pH 완충제를 포함하는 버퍼층을 형성하여 측정 시료의 pH에 따른 측정 편차를 최소화 함으로써, 센서의 검출 정확성과 정밀성을 향상시키는 것이 가능하다.
또한, 본 발명의 일 실시 예에 따른 바이오센서는, 버퍼층에 포함되는 pH 완충제의 농도를 조절함으로써, 센서의 분해능을 향상시키는 것이 가능하다.
또한, 본 발명의 일 실시 예에 따른 바이오센서는, pH 완충제를 함유하는 필터를 구비하여, 외부 불순물질의 여과력을 향상시킴으로써, 센서의 검출 정확성과 정밀성을 향상시키는 것이 가능하다.

본 발명은, 작업전극의 상부에 버퍼층을 구비함으로써, 측정 시료의 pH에 따른 영향성을 저감시켜, 센서의 검출 정확성 및 정밀성을 향상시키기 위한, 바이오센서에 관한 것이다.
구체적으로, 본 발명은 기판; 상기 기판 상에 형성되는 작업전극과 기준전극을 포함하며, 상기 작업전극은, 상기 기판 상에 형성되는 작업전극층; 상기 작업전극층 상에 형성되는 효소반응층; 상기 효소반응층 상에 형성되는 고분자막층; 및 상기 고분자막층 상에 형성되는 버퍼층을 포함하는, 바이오센서에 관한 것이다.
본 발명의 바이오센서에 있어서, 검출 대상 시료는, 혈액, 체액(침, 땀, 눈물 등), 뇨 등의 생체 시료일 수 있으며, 그 외의 액체 시료일 수 있으나, 시료의 다양한 pH에 대한 영향성을 저감시키기 위한 측면에서, 체액(침, 땀, 눈물 등)이 바람직하다.
상기 검출 대상 시료의 pH는 특별히 한정되는 것은 아니나, 본 발명의 일 실시 예에 따른 바이오센서의 경우 바람직하게는 5 내지 8일 수 있다.

<바이오센서>
본 발명의 바이오센서는, 작업전극의 상부에 pH에 따른 영향성을 저감시킬 수 있는 버퍼층을 구비함으로써, 시료에 따른 측정 편차를 최소화하기 위한 것일 수 있다.
구체적으로, 기판; 상기 기판 상에 형성되는 작업전극과 기준전극을 포함하며, 상기 작업전극은, 상기 기판 상에 형성되는 작업전극층; 상기 작업전극층 상에 형성되는 효소반응층; 상기 효소반응층 상에 형성되는 고분자막층; 및 상기 고분자막층 상에 형성되는 버퍼층을 포함하는 것일 수 있다.
도 1은 본 발명의 일 실시 예에 따른 바이오센서의 개략적인 단면도이다.
도 1을 참조하면, 본 발명의 일 실시 예에 따른 바이오센서는, 기판(100), 작업전극(200), 기준전극(300) 및 절연막(400)을 포함하며, 상기 작업전극(200)은, 작업전극층(210), 효소반응층(220), 고분자막층(230) 및 버퍼층(240)을 포함한다.
기판(100)은, 바이오센서를 구성하는 구성요소들의 구조적인 기지(base)를 제공하는 역할을 수행한다.
기판(100)은, 유리 등과 같은 경성 재질을 갖거나, 플렉서블 특성을 갖는 필름 형태로 구현되는 것일 수 있고, 종래 또는 이후에 개발되는 것을 사용할 수 있으며, 일 또는 복수의 실시 형태에 있어서, 실리콘, 유리, 유리에폭시, 세라믹, 폴리에틸렌나프탈레이트(PET), 폴리부틸렌테레프탈레이트 등의 폴리에스테르계 수지; 디아세틸셀룰로오스, 트리아세틸셀룰로오스 등의 셀룰로오스계 수지; 폴리카보네이트계 수지; 폴리메틸(메타)아크릴레이트, 폴리에틸(메타)아크릴레이트 등의 아크릴계 수지; 폴리스티렌, 아크릴로니트릴-스티렌 공중합체 등의 스티렌계 수지; 폴리에틸렌, 폴리프로필렌, 시클로계 또는 노보넨 구조를 갖는 폴리올레핀, 에틸렌-프로필렌 공중합체 등의 폴리올레핀계 수지; 염화비닐계 수지; 나일론, 방향족 폴리아미드 등의 아미드계 수지; 이미드계 수지; 폴리에테르술폰계 수지; 술폰계 수지; 폴리에테르에테르케톤계 수지; 황화 폴리페닐렌계 수지; 비닐알코올계 수지; 염화비닐리덴계 수지; 비닐부티랄계 수지; 알릴레이트계 수지; 폴리옥시메틸렌계 수지; 에폭시계 수지 등과 같은 열가소성 수지로 구성된 필름을 들 수 있으며, 상기 열가소성 수지의 블렌드물로 구성된 필름도 사용할 수 있다. 또한, (메타)아크릴계, 우레탄계, 아크릴우레탄계, 에폭시계, 실리콘계 등의 열경화성 수지 또는 자외선 경화형 수지로 된 필름을 이용할 수 있으나, 이에 한정되는 것은 아니다.
기판(100)의 두께는 특별히 한정되는 것은 아니나, 일반적으로는 강도나 취급성 등의 작업성, 박층성 등을 고려하여, 1 내지 500㎛일 수 있으며, 1 내지 300㎛가 바람직하며, 5 내지 200㎛가 보다 바람직하다
일 실시 예에 있어서, 기판(100)은, 적절한 1종 이상의 첨가제가 함유된 것일 수 있으며, 첨가제는, 예를 들어, 자외선흡수제, 산화방지제, 윤활제, 가소제, 이형제, 착색방지제, 난연제, 핵제, 대전방지제, 안료, 착색제 등을 들 수 있다.
일 실시 예에 있어서, 기판(100)은, 기판의 일면 또는 양면에 하드코팅층, 반사방지층, 가스배리어층과 같은 다양한 기능성층을 포함하는 구조일 수 있으며, 기능성층은 전술한 것으로 한정되는 것은 아니며, 용도에 따라 다양한 기능성층을 포함할 수 있다.
일 실시 예에 있어서, 기판(100)은, 필요에 따라 표면 처리된 것일 수 있으며, 이러한 표면 처리로는, 예를 들어, 플라즈마(plasma) 처리, 코로나(corona) 처리, 프라이머(primer) 처리 등의 건식 처리, 검화 처리를 포함하는 알칼리 처리 등의 화학 처리 등을 들 수 있다.
작업전극(200) 및 기준전극(300)은, 기판(100) 상에 형성될 수 있다.
상기 작업전극(200)은, 시료에 포함되는 분석물의 반응에 의해 발생되는 전기적 신호를 감지하는 역할을 수행하기 위하여 구비되는 것일 수 있다.
작업전극층(210)은 기판(100) 상에 배치되는 것일 수 있다. 일 실시 예에 있어서, 작업전극층(210)은 기판(100)의 상면에 접촉하여 배치되는 것일 수 있다. 작업전극층(210)은 감지 대상 물질의 산화-환원 반응 등에서 발생된 전자 또는 정공이 전달되는 통로로 제공될 수 있다.
일 또는 복수의 실시 형태에 있어서, 상기 작업전극층(210)은 탄소전극층 및 금속전극층으로 이루어진 군에서 선택되는 1종 이상을 포함하는 것일 수 있다.
일 또는 복수의 실시 형태에 있어서, 상기 탄소전극층은 카본 페이스트(carbon paste) 및/또는 카본나노튜브(Carbon Nano Tube; CNT) 등이 혼합된 페이스트(paste)로 형성되는 것일 수 있다. 상기 탄소전극층은 효소반응층(220)에서 발생한 전자 및/또는 정공을 안정적으로 수송할 수 있다.
일 실시 예에 있어서, 상기 작업전극층(210)은 카본 페이스트(carbon paste) 단일층으로 형성될 수 있다. 상기 카본 페이스트(carbon paste) 단일층이 전극으로 제공됨으로써, 금속 전극이 생략될 수 있다. 따라서, 바이오센서의 박막화가 가능하다.
일 실시 예에 있어서, 상기 금속전극층은 금속층 및 상기 금속층 상면에 배치된 금속보호층을 포함하는 것일 수 있다.
일 또는 복수의 실시 형태에 있어서, 상기 금속층은 금(Au), 은(Ag), 구리(Cu), 백금(Pt), 티타늄(Ti), 니켈(Ni), 주석(Sn), 몰리브덴(Mo), 팔라듐(Pd), 코발트(Co) 및 이들의 합금으로 이루어진 군에서 선택된 1종 이상을 포함하는 것일 수 있다. 예를 들면, APC 합금(Ag-Pd-Cu alloy)이 사용될 수 있다.
상기 금속보호층은, 전기 전도성을 가지면서 금속층의 상면을 전체적으로 덮는 것일 수 있다. 일 실시 예에 있어서, 금속보호층은 상기 금속층의 상면에 접촉하여 배치되는 것일 수 있다. 상기 금속보호층은 작업전극(200)의 산화-환원 반응으로 인해 금속층이 산화-환원 되는 것을 방지하기 위한 것일 수 있다.

효소반응층(220)은 작업전극층(210) 상에 배치되는 것일 수 있다. 일 실시 예에 있어서, 효소반응층(220)은 작업전극층(210)의 상면에 접촉하여 배치되는 것일 수 있다. 효소 반응층(220)은 시료에 포함된 대상 물질(analyte)의 화학반응이 일어나는 층으로 제공될 수 있다.
일 또는 복수의 실시 형태에 있어서, 상기 효소반응층(220)은 산화 효소 및 탈수소 효소로 이루어진 군에서 선택되는 1종 이상을 포함하는 것일 수 있다. 산화 효소 및 탈수소 효소는 검출 대상 물질(analyte)의 종류에 따라 적절히 선택될 수 있는 것으로, 시료에 포함된 대상 물질(analyte)과 결합하여 효소-기질 복합체를 형성하여 화학반응의 활성화 에너지를 조절함으로써 물질 대사의 속도를 증가 혹은 감소시킬 수 있는 생체 촉매이면 특별히 한정되지 않는다.

상기 예시적인 산화 효소 또는 탈수소 효소에 따라 측정할 수 있는 검출 대상 물질(analyte)은, 글루코오스(glucose), 콜레스테롤(cholesterol), 락테이트(lactate), 아스코빅산(ascorbic acid), 알코올(alcohol) 및 글루탐산(glutamate) 중 하나 이상일 수 있으며, 이들의 농도를 측정할 수 있다.
일 실시 예에 있어서, 상기 산화 효소 또는 탈수소 효소는 바인더를 통해 고정될 수 있다. 상기 바인더는 본 발명이 속한 기술분야에서 통상적으로 사용되는 바인더를 포함하며, 일 실시 예에 있어서, 나피온(nafion) 이의 유도체 또는 키토산 등이 있을 수 있다.

상기 루테늄 화합물로서는 종래 또는 이후 사용되는 루테늄 화합물을 사용할 수 있으며, 루테늄 화합물은 산화형의 루테늄 착체로서 상기 반응계에 존재할 수 있는 것인 것이 바람직하다. 상기 루테늄 착체로서는 미디에이터(전자전달체)로서 기능하면 그 배위자의 종류는 특별히 한정되지 않는다.
상기 페나진메토설페이트 및 그 유도체는 종래 또는 이후 사용되는 화합물을 사용할 수 있으며, 예를 들어, 페나진메토설페이트 및 1-메톡시-5-메틸페나지늄메틸설페이트(1-메톡시PMS) 등이 있을 수 있다.
상기 완충액은 검출 대상 물질(analyte)의 종류와 농도에 따라 적절히 선택될 수 있는 것으로, 다양한 pH 범위를 나타내는 시료에 대해서도 일정한 검출 결과를 나타낼 수 있는 것이면 특별히 한정되지 않는다.

상기 효소반응층(220)에 의한 검출 원리를 예를 들어 설명하면, 검출 대상 물질(analyte)을 포함하는 시료를 바이오센서에 주입하면, 시료에 포함되어 있는 검출 대상 물질(analyte), 예컨대 기질(substrate)이 산화 효소 또는 탈수소 효소에 의해 산화되고, 산화 효소 또는 탈수소 효소는 환원된다. 이때, 전자전달매개체는 촉매 반응을 일으킴으로써 상기 효소의 반응을 빠르게 유도하게 되며, 산화 효소 또는 탈수소 효소를 산화시키고, 자신은 환원된다. 환원된 전자전달매개체는 일정 전압이 가해진 전극 표면에서 전자를 잃고 전기화학적으로 다시 산화된다. 시료 내 대상 물질(analyte)의 농도는 전자전달매개체가 산화되는 과정에서 발생되는 전류량 내지 전류밀도 등에 비례하므로, 이 전류량 내지 전류밀도 등을 측정함으로써 대상 물질(analyte)의 농도를 측정할 수 있다.
고분자막층(230)은 효소반응층(220) 상에 배치되는 것일 수 있다. 일 실시 예에 있어서, 고분자막층(230)은 효소반응층(220)과 버퍼층(240)의 사이에 구비되며, 효소반응층(220)의 상면에 접촉하여 배치되는 것일 수 있다. 고분자막층(230)은 효소의 산화를 방지하고, 외부 물질로부터 효소를 보호함으로써 효소의 안정성을 높여, 바이오센서의 검출 성능을 향상시키기 위한 층으로 제공될 수 있다.

pH 완충제의 pH는 다양한 pH 범위를 나타내는 시료에 대해서도 일정한 검출 결과를 나타낼 수 있는 것이면 특별히 한정되지 않는다. 일 실시 예에 있어서, 상기 pH 완충제의 pH는 5 내지 8일 수 있다.
일 또는 복수의 실시 형태에 있어서, pH 완충제의 농도는 10 내지 500mM 일 수 있으며, 바람직하게는 50 내지 200mM일 수 있다. pH 완충제의 농도가 상기 범위를 만족하는 경우, 다양한 pH 범위를 나타내는 시료에 대한 측정 편차를 최소화 할 수 있을 뿐만 아니라 측정 결과의 산포를 최소화하여 분해능이 증가할 수 있다.
일 실시 예에 있어서, 상기 버퍼층은 필터를 포함하는 것일 수 있으며, 바람직하게는 상기 pH 완충제를 함유하는 필터를 포함하는 것일 수 있다. 필터는 외부의 불순 물질을 제거함으로써 바이오센서의 정확성과 정밀성을 향상시킬 수 있는 것이면 특별히 제한되지 않는다.

상기 pH 완충제를 함유하는 필터는, pH 완충제에 침지 후 건조되어 작업전극(200)의 상부에 구비되는 것이 바람직하다. 이 경우, 보관 중에는 효소층에 영향을 미치지 않으나, 대상 물질(analyte)이 포함된 시료가 유입될 경우 pH를 일정수준으로 유지시켜 줄 뿐만 아니라 외부 불순 물질의 유입을 효과적으로 차단할 수 있다.
일 또는 복수의 실시 형태에 있어서, 상기 버퍼층(240)의 두께는 1 내지 500㎛ 일 수 있으며, 바람직하게는 50 내지 300㎛ 일 수 있다. 버퍼층(240)의 두께가 상기 범위를 만족하는 경우, 바이오센서의 pH 영향성을 효과적으로 저감시킬 수 있을 뿐만 아니라, 외부 불순 물질의 유입을 효과적으로 차단할 수 있다.
일 또는 복수의 실시 형태에 있어서, 상기 버퍼층은 단층 또는 복층으로 형성될 수 있으며, 바람직하게는 1 내지 3층의 구조로 형성되는 것일 수 있다. 일 실시 예에 있어서, 상기 복층 구조는 버퍼층 복수개가 서로 접하여 적층된 것일 수 있다. 버퍼층의 적층구조가 상기 범위를 만족하는 경우, 과다한 대상 물질(analyte)의 필요로 인한 반응 지연을 방지할 수 있다.
상기 기준전극(300)은, 전위가 일정하며 작업전극(200)의 발생 전위를 얻기 위한 전위의 기준이 되는 전극으로써의 역할을 수행하기 위하여 구비되는 것일 수 있다.
일 또는 복수의 실시 형태에 있어서, 상기 기준전극(300)은 은-염화은(Ag/AgCl) 전극, 칼로멜(calomel) 전극, 수은-황산수은(mercury sulfate) 전극, 및 수은-산화수은(mercury-oxide mercury) 전극 등으로 이루어진 군에서 선택되는 1종 이상을 포함하는 것일 수 있고, 온도 사이클에 대한 전위의 히스테리시스가 덜하고, 고온까지 전위가 안정하다는 점을 고려할 때, 은-염화은(Ag/AgCl) 전극인 것이 바람직하다. 은-염화은(Ag/AgCl) 전극은, Ag/AgCl 페이스트(paste)로부터 형성될 수 있다.
<바이오센서 제조방법>
본 발명은, 상기 바이오센서의 제조를 위한 바이오센서 제조방법을 포함한다.
일 실시 예에 있어서, 기판 상에 작업전극층을 형성하고, 작업전극층 상에 효소반응층을 형성하고, 효소반응층 상에 고분자막층을 형성하고, 고분자막층 상에 버퍼층을 형성함으로써, 작업전극을 제조할 수 있다.
일 또는 복수의 실시 형태에 있어서, 작업전극층을 형성하는 단계는 스크린 인쇄, 활판 인쇄, 음각 인쇄, 평판 인쇄 및 포토리소그래피(photolithography)로 이루어진 군에서 선택되는 하나 이상의 공정을 포함하여 수행되는 것일 수 있다.
예를 들어, 기판 상에 카본 페이스트(carbon paste)를 스크린 인쇄법으로 인쇄하거나, 금(Au), 은(Ag), 구리(Cu), 백금(Pt), 티타늄(Ti), 니켈(Ni), 주석(Sn), 몰리브덴(Mo), 팔라듐(Pd), 코발트(Co) 및 이들의 합금으로 이루어진 군에서 선택된 1종 이상을 포함하는 금속층을 형성하고 이를 포토리소그래피(photolithography) 공법 등에 의해 패터닝(patterning)하여 형성될 수 있다.
일 실시 예에 있어서, 작업전극층이 금속보호층을 더 포함할 경우, 금속층을 먼저 패터닝(patterning) 한 후 상기 금속보호층을 형성하거나, 상기 금속막 상에 ITO(Indium Tin Oxide) 또는 IZO(Indium Zinc Oxide) 도전성 산화물막을 형성한 후, 상기 금속막과 도전성 산화물 막을 함께 패터닝하여 금속층 및 금속보호층을 함께 형성할 수 있다.
효소반응층을 형성하는 단계 및 고분자막층을 형성하는 단계는 본 발명이 속한 기술분야에서 통상적으로 사용되는 도포법이 사용될 수 있다. 일 또는 복수의 실시 형태에 있어서, 흐름코팅(flow coating), 잉크젯(ink jet), 및 드롭 캐스팅(drop casting)으로 이루어진 군에서 선택되는 어느 하나로 수행될 수 있으며, 드롭 캐스팅(drop casting)인 것이 더욱 바람직하다.
일 실시 예에 있어서, 버퍼층을 형성하는 단계는 pH 완충제를 필터에 침전시킨 뒤 건조과정을 거쳐, 작업전극 상에 부착, 흡착, 드롭 등의 방법을 수행하여 형성하는 것일 수 있다.
일 실시 예에 있어서, 기준전극은 Ag/AgCl 페이스트(paste) 등을 사용하여 형성될 수 있으며, 실질적으로 상기 작업전극의 제조 방법과 동일한 방법으로 제조될 수 있다.
상기 제조방법에 의해 제조되는 바이오센서는, 상기 항목 <바이오센서>에서 서술한 모든 특성을 나타내는 것일 수 있다.
<바이오센서 신호 측정방법>
본 발명은, 상기 바이오센서 제조방법으로 제조된 바이오센서를 이용한 분석물의 전기화학적 신호 측정방법을 포함한다.
본 명세서에 있어서 「전기 화학적으로 측정한다」란, 전기 화학적인 측정 수법을 적용하여 측정하는 것을 말하며, 일 또는 복수의 실시 형태에 있어서, 전류 측정법, 전위차 측정법, 전량 분석법 등을 들 수 있고, 바람직하게는 전류 측정법일 수 있다.
일 실시 예에 있어서, 본 발명의 바이오센서 신호 측정방법은 시료와의 접촉 후에 상기 작업전극과 기준전극을 포함하는 전극부에 전압을 인가하는 것, 인가 시에 방출되는 응답 전류치를 측정하는 것, 및, 상기 응답 전류치에 기초하여 상기 시료 중의 검출 대상 물질(analyte)의 전기화학적 신호를 산출하는 것을 포함할 수 있다. 인가 전압으로서는 특별히 제한되는 것은 아니나, 일 또는 복수의 실시 형태에 있어서, 은-염화은 전극(Ag/AgCl 전극)을 기준으로, -500 내지 +500mV일수 있으며 바람직하게는 -200 내지 +200 mV일수 있다.
일 실시 예에 있어서, 본 발명의 바이오센서 신호 측정방법은 상기 시료와 접촉 후 소정 시간 비인가의 상태로 유지한 후, 상기 전극부에 전압을 인가해도 되고, 상기 시약과의 접촉과 동시에 전극부에 전압을 인가해도 된다.
본 발명의 바이오센서 신호 측정방법에 의하면, 다양한 pH 범위를 나타내는 시료에 대한 측정 편차를 최소화 함으로써, 바이오센서의 정확성과 정밀성을 향상시킬 수 있다.
<바이오센서 신호 측정시스템>
본 발명은 상기 바이오센서와, 상기 바이오센서의 전극부에 전압을 인가하는 수단과, 전극부에 있어서의 전류를 측정하기 위한 수단을 포함하는, 시료 중의 검출 대상 물질(analyte)의 전기화학적 신호를 측정하기 위한 바이오센서의 전기화학적 신호 측정시스템에 관한 것이다.
본 발명의 바이오센서 신호 측정시스템에 의하면, 다양한 pH 범위를 나타내는 시료에 대해서도, 높은 정확성과 정밀도로 측정이 가능하다.
인가 수단으로서는, 바이오센서의 전극부와 도통하고, 전압을 인가 가능하면 특별히 제한되는 것은 아니며, 공지의 인가 수단을 사용할 수 있다. 인가 수단으로서는, 일 또는 복수의 실시 형태에 있어서, 바이오센서의 전극부와 접촉 가능한 접촉자, 및 직류 전원 등의 전원 등을 포함할 수 있다.
측정 수단은, 전압 인가 시에 발생한 전극부에 있어서의 복수의 전류를 측정하기 위한 것으로서, 일 또는 복수의 실시 형태에 있어서, 바이오센서의 전극부로부터 방출되는 전자의 양에 상관하는 응답 전류치를 측정 가능한 것이면 되고, 종래 또는 이후 개발되는 바이오센서에 사용되고 있는 것을 사용할 수 있다.
이하, 구체적으로 본 발명의 실시예를 기재한다. 그러나, 본 발명은 이하에서 개시되는 실시 예들에 한정되는 것이 아니라 서로 다른 다양한 형태로 구현될 수 있으며, 단지 본 실시예들은 본 발명의 개시가 완전하도록 하고, 본 발명이 속하는 기술분야에서 통상의 지식을 가진 자에게 발명의 범주를 완전하게 알려주기 위해 제공되는 것이며, 본 발명은 청구항의 범주에 의해 정의될 뿐이다.
<실시 예 및 비교 예>
실시예 1
도 1에 도시된 바이오센서와 같은 구조의 실시예 1의 바이오센서를 제작하였다.
우선, 락트산 센서의 기판(100)으로서, PET제 기판(길이 30~40㎜, 폭 3~10㎜, 두께 188㎛)을 준비하고, 그 한쪽의 표면에, 작업전극층(210)으로는 Carbon paste(대주전자재료사제)를, 기준전극(300)으로는 Silver paste(대주전자재료사제)를 각각 10~30㎛ 두께로 250 메쉬 스크린을 사용하여 스크린 인쇄하였다. 그리고 Carbon paste는 100℃에서 20분간, Silver paste는 80℃에서 10분간 가열 처리를 하였다.
이후, 상기 작업전극층(210)과 기준전극(300)을 제외한 전극부의 상면 상에 절연성 페이스트(대주전자재료사제)를 10~50㎛ 두께로 250 메쉬 스크린을 사용하여 스크린 인쇄하였다. 그리고 120℃에서 15분간 가열 처리를 하였다.
이후, 작업전극층(210) 상에 용액 2.0㎕당 락트산 옥시다아제(상품명 「LCO301」, 토요보사제) 8U, Hexaammineruthenium(III) chloride, 98%(363340010, 아크로스사제) 1.5㎍, 1-m-PMS(M8640, 알드리치사제) 20nmol, PBS 완충액 0.4㎕를 포함하는 용액을 조제하여, 해당 용액 2.0㎕를 검출부에 드롭캐스팅(drop casting)하여 효소반응층(220)을 형성하였다.
이후, 상기 효소반응층(220) 상에 PVA(1% PVA in PBS, 알드리치사제) 1.2㎕를 도포하고 1시간동안 건조하여, 고분자막층(230)을 형성하였다.
이후, 필터페이퍼(상품명 「Grade 4 Qualitative Filter Paper」 Whatman사제)를 상기 효소반응층(220)에 사용한 PBS 완충액을 포함한 농도가 10X가 되도록 침지시킨 뒤 1 시간 이상 건조하고, 이를 상기 고분자막층(230) 상에 구비시켜 버퍼층(240)을 형성함으로써, 실시예 1의 바이오센서를 제작하였다.
실시예 2
필터페이퍼(상품명 「Grade 4 Qualitative Filter Paper」 Whatman사제)를 효소반응층(220)에 사용한 PBS 완충액을 포함한 농도가 40X가 되도록 침지시킨 것을 제외하고는, 실시예 1과 동일하게 실시예 2의 바이오센서를 제작하였다.
비교예 1
버퍼층(240)을 형성하는 단계를 생략한 것을 제외하고는, 실시예 1과 동일하게 비교예 1의 바이오센서를 제작하였다.
<실험예 1>: 버퍼층 유무에 따른 바이오센서 평가
상기 실시예 1 및 비교예 1에 따른 바이오센서를 하기 측정방법을 이용하여 시료 중의 락트산 농도에 따른 전류 값을 측정함으로써, 다양한 pH 범위의 시료에 대한 pH 안정성을 평가하였다.
시료로서는 5mM, 10mM, 15mM, 20mM, 25mM, 30mM, 및 40mM의 락트산 조합액 10㎕를 사용하고, 측정 장치는 CHI630를 이용했다. 측정은, 25℃ 60~70RH%로 설정한 항온항습의 환경 실험실 내에서, 바이오센서에 시료를 공급한 후, 검체 검지 후 15초 동안 200mV의 전압을 인가함으로써 행했다.
상기 실시예 1 및 비교예 1에 따른 바이오센서의 평가 결과를 하기 표 1에 나타내었다.

상기 표 1을 참조하면, 실시예 1의 바이오센서의 경우 pH 5 내지 8의 시료에 대한 검량 기울기 산포가 최대 0.04로, 비교적 일정한 검량 기울기를 나타내고 있으며, 시료 농도 5 내지 40mM에 대한 P-value 값이 0.05 이상을 나타내고 있어, 감응 범위 전체에 대하여 pH 영향성이 저감되었음을 확인할 수 있다.
반면, 비교예 1의 바이오센서의 경우 pH 5 내지 8의 시료에 대한 검량 기울기 산포가 최대 0.13으로, 실시예 1의 바이오센서 대비 검량 기울기 폭이 증가하였으며, 15mM 이상의 시료 농도에 대한 P-value가 0.000 내지 0.001을 나타내고 있어, 시료의 pH에 따른 측정 편차가 증가하였음을 확인할 수 있다.
<실험예 2>: pH 완충제의 농도 따른 바이오센서 평가
상기 실시예 1 및 2에 따른 바이오센서를 하기 측정방법을 이용하여 시료 중의 락트산 농도에 따른 전류 값을 측정함으로써, pH 완충제의 농도에 따른 바이오센서의 분해능을 평가하였다.
시료로서는 10mM 및 30mM의 락트산 조합액을 각각 10㎕를 사용하고, 측정 장치는 CHI630를 이용했다. 측정은, 25℃, 60~70RH%로 설정한 항온항습의 환경 실험실 내에서, 바이오센서에 시료를 공급한 후, 검체 검지 후 15초 동안 200mV의 전압을 인가함으로써 행했다.
상기 실시예 1 및 2에 따른 바이오센서의 평가 결과를 각각 도 2 및 도 3에 나타내었다.
도 2 및 도 3을 참조하면, PBS 농도가 40X가 되도록 침지된 실시예 2의 바이오센서의 pH에 대한 영향성 저감의 효과가 PBS 농도가 10X가 되도록 침지된 실시예 1의 바이오센서의 pH에 대한 영향성 저감의 효과와 유사한 수준으로, 버퍼층에 포함되는 pH 완충제의 농도가 증가하더라도 시료의 pH에 대한 영향성 저감의 효과가 이에 비례하여 증가하지 않는 것을 확인할 수 있다.
그러나, PBS 농도가 40X가 되도록 침지된 실시예 2의 바이오센서는 그 측정 산포가 PBS 농도가 10X가 되도록 침지된 실시예 1의 바이오센서 대비 증가하여, 버퍼층에 포함되는 pH 완충제의 농도가 과도하게 증가할 경우 측정 산포의 증가로 인한 분해능이 다소 감소함을 확인할 수 있다.
따라서, 버퍼층에 포함되는 pH 완충제의 농도를 적절히 조절함으로써, pH 영향성이 저감되면서도 분해능이 우수한 바이오센서를 제작할 수 있다.
본 발명의 일 실시 예에 따른 바이오센서는, 작업전극의 상부에 pH 완충제를 포함하는 버퍼층을 형성하여 측정 시료의 pH에 따른 측정 편차를 최소화 함으로써, 센서의 검출 정확성과 정밀성을 향상시키는 것이 가능하다.
또한, 본 발명의 일 실시 예에 따른 바이오센서는, 버퍼층에 포함되는 pH 완충제의 농도를 조절함으로써, 센서의 분해능을 향상시키는 것이 가능하다.
또한, 본 발명의 일 실시 예에 따른 바이오센서는, pH 완충제를 함유하는 필터를 구비하여, 외부 불순물질의 여과력을 향상시킴으로써, 센서의 검출 정확성과 정밀성을 향상시키는 것이 가능하다
Claims (19)
- 기판;상기 기판 상에 형성되는 작업전극과 기준전극을 포함하며,상기 작업전극은,상기 기판 상에 형성되는 작업전극층;상기 작업전극층 상에 형성되는 효소반응층;상기 효소반응층 상에 형성되는 고분자막층; 및상기 고분자막층 상에 형성되는 버퍼층을 포함하는, 바이오센서.
- 청구항 1에 있어서, 상기 버퍼층은 pH 완충제를 포함하는, 바이오센서.
- 청구항 2에 있어서, 상기 pH 완충제는 포스페이트 완충된 염수(PBS), 트리스(하이드록시메틸)아미노메탄(Tris), 트리스 염산(Tris-HCl), 암모늄 바이카보네이트, 4-(2-하이드록시에틸)-1-피페라진에탄설폰산(HEPES), 3-(N-모르폴리노)프로판설폰산(MOPS), 2-(N-모르폴리노)에탄설폰산(MES), 2,2-비스(하이드록시메틸)-2,2',2"-니트릴로트리에탄올(Bis-tris), N-(2-아세트아미도)이미노디아세트산(ADA), 피페라진-N,N'-비스(2-에탄설폰산)(PIPES), N-(2-아세트아미도)-2-아미노에탄설폰산(ACES), 3-(N-모르폴리닐)-2-하이드록시프로판설폰산 소듐 염(MOPSO), 1,3-비스(트리스(하이드록시메틸)메틸아미노)프로판(Bis-tris Propane), N,N-비스(2-하이드록시에틸)-2-아미노에탄설폰산(BES), 2-[[1,3-디하이드록시-2-(하이드록시메틸)프로판-2-일]아미노]에탄설폰산(TES), 3-(비스(2-하이드록시에틸)아미노)-2-하이드록시프로판-1-설폰산(DIPSO), 3-[[1,3-디하이드록시-2-(하이드록시메틸)프로판-2-일]아미노]-2-하이드록시프로판-1-설폰산(TAPSO), 트리즈마(Trizma), 피페라진-1,4-비스(2-하이드록시프로판설폰산) 이수화물(POPSO), 3-[4-(2-하이드록시에틸)-1-피페라지닐]프로판설폰산(HEPPS), N-(2-하이드록시-1,1-비스(하이드록시메틸)에틸)글리신(TRICINE), 글리실글리신(GLY-GLY), 2-(비스(2-하이드록시에틸)아미노)아세트산(BICINE), N-(2-하이드록시에틸)피페라진-N'-(4-부탄설폰산)(HEPBS), 3-[[1,3-디하이드록시-2-(하이드록시메틸)프로판-2-일]아미노]프로판-1-설폰산(TAPS), 2-아미노-2-메틸-1,3-프로판디올(AMPD), N-(1,1-디메틸-2-하이드록시에틸)-3-아미노-2-하이드록시프로판설폰산(AMPSO), N-사이클로헥실-2-아미노에탄설폰산(CHES), N-사이클로헥실-2-하이드록실-3-아미노프로판설폰산(CAPSO), 1-아미노-2-메틸-1-프로판올(AMP), N-사이클로헥실-3-아미노프로판설폰산(CAPS), 4-(사이클로헥실아미노)-1-부탄설폰산(CABS) 및 라이소제니 브로쓰(Lysogeny broth)로 이루어진 군에서 선택되는 1종 이상을 포함하는, 바이오센서.
- 청구항 2에 있어서, 상기 pH 완충제의 pH가 5 내지 8인, 바이오센서.
- 청구항 2에 있어서, 상기 pH 완충제의 농도는, 10 내지 500mM인, 바이오센서.
- 청구항 2에 있어서, 상기 버퍼층은 상기 pH 완충제를 함유하는 필터를 포함하는, 바이오센서.
- 청구항 6에 있어서, 상기 필터는 폴리프로필렌(Polypropylene; PP), 폴리테트라플루오로에틸렌(Polytetrafluoroethylene; PTFE), 폴리비닐리덴 플루오라이드(Polyvinylidene fluoride; PVDF), 혼합 셀룰로오스 에스테르(Mixed cellulose esters; MCE), 셀룰로오스 아세테이트(Cellulose acetate; CA), 니트레이트 셀룰로오스(Nitrate cellulose; NC), 폴리에테르 설폰(Polyether sulfone; PES), 및 나일론(Nylon; NYL)으로 이루어진 군에서 선택되는 1종 이상을 포함하는, 바이오센서.
- 청구항 6에 있어서, 상기 pH 완충제를 함유하는 필터는 건조된 것인, 바이오센서.
- 청구항 1에 있어서, 상기 버퍼층의 두께는, 1 내지 500㎛인, 바이오센서.
- 청구항 1에 있어서, 상기 버퍼층은 단층 또는 복층 구조인, 바이오센서.
- 청구항 1에 있어서, 상기 작업전극층은 탄소전극층 및 금속전극층으로 이루어진 군에서 선택되는 1종 이상을 포함하는, 바이오센서.
- 청구항 11에 있어서, 상기 금속전극층은 금속층 및 금속보호층으로 이루어진 군에서 선택되는 1종 이상을 포함하는, 바이오센서.
- 청구항 1에 있어서, 상기 효소반응층은 산화 효소 및 탈수소 효소로 이루어진 군에서 선택되는 1종 이상을 포함하는, 바이오센서.
- 청구항 13에 있어서, 상기 산화 효소는 글루코오스 산화 효소(glucose oxidase), 콜레스테롤 산화 효소(cholesterol oxidase), 락테이트 산화 효소(lactate oxidase), 아스코빅산 산화 효소(ascorbic acid oxidase) 및 알코올 산화 효소(alcohol oxidase)로 이루어진 군에서 선택되는 1종 이상을 포함하며,상기 탈수소 효소는 글루코오스 탈수소 효소(glucose dehydrogenase), 글루탐산 탈수소 효소(glutamate dehydrogenase), 락테이트 탈수소 효소(lactate dehydrogenase) 및 알코올 탈수소 효소(alcohol dehydrogenase)로 이루어진 군에서 선택되는 1종 이상을 포함하는, 바이오센서.
- 청구항 13에 있어서, 상기 효소반응층은 미디에이터, 완충액 및 용매로 이루어진 군에서 선택되는 1종 이상을 더 포함하는, 바이오센서.
- 청구항 15에 있어서, 상기 미디에이터는 페리시안화칼륨, 시토크롬C, 피로로퀴놀린퀴논(PQQ), NAD+, NADP+, 동착체, 루테늄 화합물, 페나진메토설페이트 및 그 유도체, 포타슘 페리시아나이드(Potassium ferricyanide, K3[Fe(CN)6]), 포타슘 페로시아나이드(Potassium ferrocyanide, K4[Fe(CN)6]), 염화헥사아민루테늄(Ⅲ)(hexaamineruthenium(Ⅲ) chloride), 페로센(ferrocene), 페로센 유도체, 퀴논(quinones), 퀴논 유도체, 하이드로퀴논(hydroquinone), 페로센(ferrocene) 및 페로센 유도체로 이루어진 군에서 선택되는 1종 이상을 포함하는, 바이오센서.

- 청구항 18에 있어서, 상기 수용성 고분자는 폴리비닐알코올(polyvinyl alcohol; PVA), 하이드록시에틸 셀룰로오즈(hydroxyethyl cellulose; HEC), 하이드록시프로필 셀룰로오즈(hydroxypropyl cellulose; HPC), 카르복시메틸 셀룰로오즈(carboxy methyl cellulose; CMC), 셀룰로오즈 아세테이트(cellulose acetate; CA), 및 폴리비닐피롤리돈(polyvinyl pyrrolidone; PVP)으로 이루어진 군에서 선택되는 1종 이상을 포함하며,상기 비수용성 고분자는 폴리우레탄(polyurethane, PU), 폴리카보네이트(polycarbonate; PC), 및 폴리비닐클로라이드(polyvinyl chloride; PVC)으로 이루어진 군에서 선택되는 1종 이상을 포함하는, 바이오센서.
