Abstract
본 발명은 표피층에서 비타민D의 기능을 활성화시키고 각질형성세포의 분화인자의 발현을 촉진하여 피부재생을 촉진할 수 있는 조성물을 제공하는 것이다. 본 발명에 따른 피부재생용 화장료 조성물은 아몬드, 타히보, 흰목이버섯을 포함하여, 칼슘, 비타민D 및 비타민E를 충분히 공급하며, 노화된 피부의 재생을 촉진시켜 피부각질을 제거하고 피부결을 개선시키는 효과가 있다.
Description
본 발명은 피부재생용 화장료 조성물에 관한 것이다.
피부는 외부에서부터 순서대로 표피, 진피, 피하 조직의 3개 층으로 크게 구분되며, 체내의 각 기관을 기온 및 습도변화, 자외선, 기타 물리화학적인 외부 환경의 자극으로부터 보호해 주는 기능이 있다.
피부는 외부환경으로부터 우리 몸을 보호하는 일차적인 기능과 미용적인 기능을 가지고 있다. 또한 화장품은 사람의 피부를 청결하게 하거나 아름답게 표현하는 미적 기능 및 유해 환경으로부터 피부를 보호하는 방어기능을 그 목적으로 한다.
피부의 역할은 자외선, 대기오염, 스트레스, 황사 등과 같은 다양한 유해환경으로 인해 쇠퇴하게 되는데 화장품들은 이러한 유해인자에 의해 발생하는 반응성이 높은 활성산소, 자유라디칼 및 과산화물 등에 의해 유발되는 다양한 피부의 트러블을 개선함은 물론 피부를 보호하고 피부가 정상적인 기능을 유지할 수 있도록 도와주고 있다.
그러나, 요즘과 같이 환경의 변화나 생활패턴의 변화에 따른 냉난방의 인위적인 온도조절, 사회생활에서 발생되는 각종 스트레스와 환경오염으로 인한 피부 스트레스, 화장 습관에 따른 잦은 세안 및 연령 증가에 따른 자연적인 피부 노화 등의 여러 가지 원인으로 인하여 피부가 건조해지고 표면이 거칠게 되며 피부가 푸석거리고 촉촉함을 잃어 생기가 없어지는 등의 현상이 발생하기 때문에 피부 노화방지 및 피부재생의 필요가 증가하고 있다.
피부재생 과정은 기저층에 위치한 각질형성세포가 분화과정을 거쳐 각화되어 새로운 세포로 교체되는 현상으로, 약 28일을 주기로 진행되는 것으로 알려져 있다. 노화된 피부에서는 각질형성세포의 기능이 저하되어 피부재생이 지연되는 것으로 알려져 있으며, 각질의 탈락이 제대로 이루어지지 않거나 새로운 세포로 교체되지 않아 거칠고 칙칙한 피부로 변하게 되는 것이다.
한편, 각질형성세포의 분화를 촉진하는 인자로써 칼슘(Calcium), 비타민D(Vitamin D), 비타민E(Vitamin E) 등이 관련되어 있는 것으로 알려져 있다. 칼슘은 각질형성세포의 증식, 분화조절, 항상성 유지에 기여하는데, 다른 영양소에 비해 흡수율이 낮으며, 카페인이 들어있는 커피 등의 섭취는 칼슘의 흡수를 더욱 방해하는 것으로 알려져 있다. 비타민D는 칼슘이온의 흡수에 관여하는데, 주로 햇빛을 쬐어 체내(표피)에서 합성된다. 현대인들은 실내활동 증가로 햇빛을 쬐는 일이 드물고, 일상적인 자외선차단제 사용으로 인해 야외 활동에서도 비타민D 합성이 적어져 비타민D 결핍 문제가 발생하고 있다. 비타민E는 칼슘 농도조절에 관여하는데, 세포 내 칼슘이온 농도를 증가시키는 것으로 알려져 있다.
종래 다양한 천연 추출물을 이용하여 보습, 피부진정 및 항노화 조성물을 제공하고 있었으나, 각질형성세포의 분화인자의 발현을 촉진하여 피부재생을 위한 효과적인 화장료 조성물에 대한 연구가 미흡하였다.
본 발명은 표피층에서 비타민D의 기능을 활성화시키고 각질형성세포의 분화인자의 발현을 촉진하여 피부재생을 촉진할 수 있는 조성물을 제공하는 것이다.
본 발명에 따른 피부재생용 화장료 조성물은 아몬드 (Almond) 추출물, 타히보 (Tabebuia) 추출물 및 흰목이버섯 (Tremella) 추출물으로부터 선택된 1종 이상의 추출물을 포함하는 것이 바람직하다.
또한, 본 발명에 따른 피부재생용 화장료 조성물은 타히보 추출물과; 아몬드 추출물 또는 흰목이버섯 추출물;을 포함하는 것이 보다 바람직하다.
또한, 본 발명에 따른 피부재생용 화장료 조성물은 아몬드 추출물, 타히보 추출물 및 흰목이버섯 추출물을 포함하는 것이 보다 더 바람직하다.
또한, 본 발명에 따른 피부재생용 화장료 조성물 중 상기 아몬드 추출물은 전체 조성물 중 0.1~5중량%로 포함되는 것이 바람직하다.
또한, 본 발명에 따른 피부재생용 화장료 조성물 중 상기 타히보 추출물은 전체 조성물 중 0.1~10중량%로 포함되는 것이 바람직하다.
또한, 본 발명에 따른 피부재생용 화장료 조성물 중 상기 흰목이버섯 추출물은 0.1~20 중량%로 포함되는 것이 바람직하다.
또한, 본 발명에 따른 피부재생용 화장료 조성물은 피부 재생과 관련된 피부세포 분화인자인 로리크린 (Loricrin), 인볼루크린 (Involucrin), 시토케라틴10 (Cytokeratin10) 및 데스모글레인2 (Desmoglein2) 중 2종 이상을 발현시키는 것이 바람직하다.
또한, 본 발명에 따른 피부재생용 화장료 조성물은 피부세포 분화와 관련된 인자인 비타민D(Vitamin D) 및 비타민D 수용체(Vitamin D Receptor) 중 1종 이상을 발현시키는 것이 바람직하다.
또한, 본 발명에 따른 피부재생용 화장료 조성물은 β-glucosidase 효소활성을 갖는 것이 바람직하다.
본 발명에 따른 피부재생용 화장료 조성물은 아몬드, 타히보, 흰목이버섯을 포함하여, 칼슘, 비타민D 및 비타민E를 충분히 공급하며, 노화된 피부의 재생을 촉진시켜 피부각질을 제거하고 피부결을 개선시키는 효과가 있다.

본 발명에 따른 피부재생용 화장료 조성물은 아몬드 (Almond) 추출물, 타히보 (Tabebuia) 추출물 및 흰목이버섯 (Tremella) 추출물으로부터 선택된 1종 이상의 추출물을 포함하는 것이 바람직하다.
이하, 본 발명이 속하는 기술분야에서 통상의 지식을 가진 자가 용이하게 실시할 수 있도록 본 발명의 실시형태를 들어 상세히 설명한다. 본 발명의 실시형태는 당업계에서 평균적인 지식을 가진 자에게 본 발명을 더욱 완전하게 설명하기 위해서 제공되는 것이다. 따라서, 본 발명의 실시형태는 여러 가지 다른 형태로 변형될 수 있으며, 본 발명의 범위가 이하 설명하는 실시형태로 한정되는 것은 아니다.

피부의 표피는 기저층 (Basal layer), 유극층 (Spinous layer), 과립층 (Granular layer), 각질층 (Cornified layer)으로 구성되어 있다. 표피의 가장 아래층인 기저층으로부터 생성되는 각질형성세포는 28일 동안 유극층, 과립층을 거쳐 각질층을 형성한다. 최종 분화가 진행되면 세포막의 안쪽에 인볼루크린 (Involucrin), 로리크린 (Loricrin) 등의 구조단백질을 결합시켜 물에 녹지 않는 단단한 구조물인 각질세포막 (Cornified cell envelope)을 형성하게 된다.
데스모글레인2 (Desmoglein2)는 데스모좀 (Desmosome)에 존재하는 당단백질로, 표피의 유극층 (Spinous layer)에서 발현된다. 각질형성세포의 분화 과정에서 세포간연접 (cell-cell junction)에 관여한다. 시토케라틴10 (Cyrokeratin10)은 분화 초기 세포 주기 조절에 직접적으로 관여하며 표피의 유극층과 과립층 사이에 주로 분포하는 것으로 알려져 있다. 로리크린 (Loricrin)과 인볼루크린 (Involucrin)은 표피의 각질층 (Cornified layer)에서 발현되는 표지인자이다. 로리크린 (Loricrin)은 세포의 분화, 증식 및 이동을 조절하는데 관여하며, 인볼루크린 (Involucrin)은 각질세포와 각질세포 사이의 지질을 이어주는 역할을 하는 단백질로 최종 분화가 진행됨에 따라 발현 수준이 증가하게 된다.
피부 재생을 촉진하기 위한 방안은 다양하게 있을 수 있지만, 화장료 조성물을 이용하는 것이 미용 및 편의성 측면에서 유리하다.
피부 재생을 위한 이러한 화장료 조성물에 사용될 수 있는 천연 추출물은 다양한 영양소 및 화합물을 포함하고 있으므로 그 작용이 복합적이다.
다양한 천연 추출물 중에서, 피부 재생을 위해서는 인체각질형성세포의 피부재생과 관련된 피부세포 분화인자인 로리크린 (Loricrin), 인볼루크린 (Involucrin), 시토케라틴10 (Cytokeratin10) 및 데스모글레인2 (Desmoglein2)의 발현을 촉진할 수 있는 것이 바람직하다.
본 발명은 다양한 천연물 중에서 아몬드 추출물, 타히보 추출물 및 흰목이버섯 추출물을 포함하여 표피에서 각질형성세포의 분화가 일어나는 전반적인 과정에 관여하여 단계별 분화인자의 발현을 촉진시키는 효과가 있다.
먼저, 본 발명에 따른 화장료 조성물의 효과를 확인하기 전에, 아래와 같이 젊은 피부와 노화 피부를 구분할 수 있는 세포노화인자를 확인하고, 젊은 피부와 노화 피부의 피부재생 주기를 확인하였다.
(실험예 1)
인체각질형성세포(Human Epidermal Keratinocyte neonatal, HEKn)에서 세포의 경로(passage)에 따라 젊은 피부와 노화피부의 모델로 구분할 수 있는지 확인하기 위해 웨스턴-블롯(western-blot)으로 경로변화(p4~p9)에 따른 p16INK4a(세포 노화의 표지인자) 발현 정도를 비교하였다.
구체적으로, P4~p9의 인체각질형성세포를 각각 60 mm plate에 100 x 104 cells 농도로 접종하고 24시간 배양한 후, 성장인자가 포함되지 않은 배지로 교체한 뒤 24시간 배양하였다. 배양 후 RIPA Buffer을 처리하여 단백질을 회수하고 BCA 정량을 통해 단백질량이 30㎍이 되도록 샘플을 제조하였다. 제조한 샘플을 SDS-PAGE(Sodiumdodecylsulfate-polyacrylamide gel electrophoresis)를 이용하여 단백질을 분리하고, Electrotransfer을 이용하여 PVDF 막(membrane)에 옮긴 후, 면역블로팅법(immune blotting)을 이용하여 세포 내 발현 수준을 확인하였다.
그 결과, 도 1에 도시된 바와 같이, 세포의 경로가 높아질수록 세포노화 표지인자 p16INK4a 발현이 증가하였다. 초기 경로(Early passage: p4~p5)와 후기 경로(Late passage: p8~p9)를 각각 젊은 피부와 노화 피부의 모델로 사용할 수 있음을 확인하였다.
(실험예 2)
젊은 피부보다 노화 피부에서 피부 재생 주기가 늦어지는지 확인하기 위해 초기 경로(p4)와 후기 경로(p8)에서 인체각질형성세포의 피부재생과 관련된 피부세포 분화인자인 로리크린 (Loricrin), 인볼루크린 (Involucrin), 시토케라틴10 (Cytokeratin10) 및 데스모글레인2 (Desmoglein2)의 발현 정도를 웨스턴-블롯으로 비교하였다.
구체적으로, 초기경로(p4)와 후기경로(p8)의 인체각질형성세포를 각각 60 mm plate에 100 x 104 cells 농도로 접종하고 24시간 배양한 후, 성장인자가 포함되지 않은 배지로 교체한 뒤 24시간 배양하였다. 배양 후 RIPA Buffer을 처리하여 단백질을 회수하고, BCA 정량을 통해 단백질량이 30㎍이 되도록 샘플을 제조하였다. 제조한 샘플을 SDS-PAGE(Sodiumdodecylsulfate-polyacrylamide gel electrophoresis)를 이용하여 단백질을 분리하고, Electrotransfer을 이용하여 PVDF 막(membrane)에 옮긴 후, 면역블로팅법(immune blotting)을 이용하여 세포 내 발현 수준을 확인하였다.
그 결과, 도 2에 도시된 바와 같이, 초기 경로에 비해 후기 경로에서 로리크린 (Loricrin), 인볼루크린 (Involucrin), 시토케라틴10 (Cytokeratin10) 및 데스모글레인2 (Desmoglein2)의 발현 시기가 더 늦은 것을 확인하였다. 즉, 노화 피부에서 피부 재생이 지연됨을 확인하였다.
본 발명은 칼슘, 비타민D 및 비타민E를 충분히 공급하여 피부재생을 촉진하는 화장료 조성물을 제공한다.
구체적으로, 본 발명에 따른 피부재생용 화장료 조성물은 아몬드 추출물, 타히보 추출물 및 흰목이버섯 추출물으로부터 선택된 1종 이상의 추출물을 포함하는 것이 바람직하다.
또한, 본 발명에 따른 피부재생용 화장료 조성물은 타히보 추출물과; 아몬드 추출물 또는 흰목이버섯 추출물;을 포함하는 것이 보다 바람직하다.
또한, 본 발명에 따른 피부재생용 화장료 조성물은 아몬드 추출물, 타히보 추출물 및 흰목이버섯 추출물을 포함하는 것이 보다 더 바람직하다.
상기 아몬드 추출물은 전체 조성물 중 0.1~5중량%로 포함되는 것이 바람직하다.
아몬드 추출물이 0.1중량% 미만으로 포함될 경우 피부재생과 관련된 피부세포 분화인자들의 발현 수준을 유의미하게 증가시킬 수 없다. 5중량% 초과하여 포함될 경우 함유량 증가에 따른 효과의 증가가 뚜렷하지 않으며, 고농도로 인해 세포독성 및 피부에 자극을 일으킬 수 있다. 또한 제형에 탁도를 유발하고 침전을 발생시켜 제형이 불안정해질 수 있다.
상기 타히보 추출물은 전체 조성물 중 0.1~10중량%로 포함되는 것이 바람직하다.
타히보 추출물이 0.1중량% 미만으로 포함될 경우 피부재생과 관련된 피부세포 분화인자들의 발현 수준을 유의미하게 증가시킬 수 없다. 10중량% 초과하여 포함될 경우 함유량 증가에 따른 효과의 증가가 뚜렷하지 않으며, 고농도로 인해 세포독성 및 피부에 자극을 일으킬 수 있다. 또한 제형에 탁도를 유발하고 침전을 발생시켜 제형이 불안정해질 수 있다.
상기 흰목이버섯 추출물은 전체 조성물 중 0.1~20중량%로 포함되는 것이 바람직하다.
흰목이버섯 추출물이 0.1중량% 미만으로 포함될 경우 피부재생과 관련된 피부세포 분화인자들의 발현 수준을 유의미하게 증가시킬 수 없다. 20중량% 초과하여 포함될 경우 함유량 증가에 따른 효과의 증가가 뚜렷하지 않으며, 고농도로 인해 세포독성 및 피부에 자극을 일으킬 수 있다. 또한 제형에 탁도를 유발하고 침전을 발생시켜 제형이 불안정해질 수 있다.
이하에서는 본 발명에 따른 피부재생용 화장료 조성물의 구체적인 실시예 및 실험예를 통해 본 발명을 보다 구체적으로 설명한다.
(실시예 1~3) 단일 추출물의 제조
건조된 아몬드 10~50g, 타히보 10~50g, 흰목이버섯 10~50g 각각을 분쇄하여 추출용매로 정제수와 부틸렌글라이콜 1~5 kg을 넣고 1M 염산 용액과 1M 수산화나트륨 용액을 이용해 pH를 5~6으로 조정하였다. 25~37℃에서 600 rpm으로 1시간 교반추출한 후, 1 ㎛ 필터페이퍼로 여과하였다. 상기 여액을 정제수 및 부틸렌글라이콜과 혼합한 뒤 0.2 ㎛ PES 멤브레인 필터로 최종 여과하여 제조하였다. 그 결과를 아래 표 1에 기재하였다.

(실험예 3) 단일 추출물 효소활성 평가
최적 추출 조건을 설정하기 위해 실시예 1~3로부터 얻어진 단일추출물에 대해 β-glucosidase 효소활성을 확인하였다. 단일추출물과 기질액(1 mM p-nitrophenyl-D-glucopyranoside, p-NPG)을 1:1 비율로 혼합하여 상온에서 2시간 안정화하였다. 50℃ 수욕상에서 30분간 반응한 후 반응정지용액(0.25 M sodium carbonate) 2 ml을 추가하여 반응을 종결하고 420 nm에서 흡광을 측정하였다. 표준용액(P-nitrophenol solution)과 비교하여 β-glucosidase 효소활성을 계산하였다. 그 결과를 아래 표 2에 기재하였다.

(실험예 4)
상기에서 제조된 본 발명의 실시예 1~3의 단일 추출물이 노화 피부에서 지연된 피부 재생 주기를 촉진할 수 있는지 확인하기 위해, 인체각질형성세포에 실시예 1~3을 처리한 뒤, 피부 재생과 관련된 피부세포 분화인자인 로리크린 (Loricrin), 인볼루크린 (Involucrin), 시토케라틴10 (Cytokeratin10) 및 데스모글레인2 (Desmoglein2)가 후기 경로에서 발현되는 정도를 웨스턴-블롯으로 확인하였다.
구체적으로, 후기경로(p9)의 인체각질형성세포를 각각 60 mm plate에 100 x 104 cells 농도로 접종하고 24시간 배양한 후, 성장인자가 포함되지 않은 배지에 실시예 1, 실시예 2, 실시예 3을 첨가하여 24시간 배양하였다. 배양 후 RIPA Buffer을 처리하여 단백질을 회수하고 BCA 정량을 통해 단백질량이 30㎍이 되도록 샘플을 제조하였다. 제조한 샘플을 SDS-PAGE(Sodiumdodecylsulfate-polyacrylamide gel electrophoresis)를 이용하여 단백질을 분리하고, Electrotransfer을 이용하여 PVDF 막(membrane)에 옮긴 후, 면역블로팅법(immune blotting)을 이용하여 세포 내 발현 수준을 확인하였다.
그 결과, 도 3에 도시된 바와 같이, 실시예 1은 인볼루크린 (Involucrin), 시토케라틴10 (Cytokeratin10) 및 데스모글레인2 (Desmoglein2)의 발현을 촉진하였고, 실시예 2는 로리크린 (Loricrin), 인볼루크린 (Involucrin), 시토케라틴10 (Cytokeratin10) 및 데스모글레인2 (Desmoglein2)의 발현을 촉진하였다. 실시예 3은 로리크린 (Loricrin), 인볼루크린 (Involucrin) 및 데스모글레인2 (Desmoglein2)의 발현을 촉진하는 효과가 있음을 확인하였다.
도 3에 도시된 바와 같이, 실시예 1~3은 각각 가장 큰 효과를 내는 인자들이 서로 다르다. 실시예 1은 시토케라틴10 (Cytokeratin10)의 발현을 촉진하는 효과가 가장 크며, 실시예 2는 로리크린 (Loricrin)과 인볼루크린 (Involucrin)의 발현을 촉진하는 효과가 크다. 실시예 3은 데스모글레인2 (Desmoglein2)의 발현을 촉진하는 효과가 크다.
실시예 1과 실시예 2만 조합할 경우 데스모글레인2 (Desmoglein2)의 발현을 촉진하는 효과가 낮을 수 있고, 실시예 2와 실시예 3만 조합할 경우 시토케라틴10 (Cytokeratin10)의 발현을 촉진하는 효과가 낮을 수 있다. 또한 실시예 1과 실시예 3만 조합할 경우 로리크린 (Loricrin)과 인볼루크린 (Involucrin)의 발현을 촉진하는 효과가 낮을 수 있다.
특히, 실시예 1은 분화의 가장 초기 단계에 관여하는 시토케라틴10 (Cytokeratin)의 발현을 촉진함으로써 하위 단계의 분화를 가속화 하는 효과가 있다.
따라서, 아래 실시예 4와 같이 실시예 1~3을 모두 포함하는 실시예를 실험하였다.
(실시예 4) 아몬드, 타히보, 흰목이버섯 혼합추출물 제조
건조된 아몬드 10g~50g을 분쇄하여 추출용매로 정제수와 부틸렌글라이콜 1~5 kg을 넣고 1M 염산 용액과 1M 수산화나트륨 용액을 이용해 pH를 5~6으로 조정하였다. 25~37℃에서 600 rpm으로 1시간 교반추출한 후, 1 ㎛ 필터페이퍼로 여과하여 아몬드 추출액을 제조하였다. 아몬드 추출액에 타히보 15g, 흰목이버섯 25g을 혼합 분쇄하여 넣고 상온에서 600 rpm으로 1시간 교반추출한 후, 1 ㎛ 필터페이퍼로 여과하였다. 상기 여액을 정제수 및 부틸렌글라이콜과 혼합한 뒤 0.2 ㎛ PES 멤브레인 필터로 최종 여과하여 제조하였다. 그 결과를 아래 표 3에 기재하였다.

상기에서 제조된 조성물의 외관 성상을 관찰하기 위해 사진 촬영하여 도 9에 도시하였다.
(실험예 5)
본 발명의 혼합 추출물이 노화 피부에서 지연된 피부 재생 주기를 촉진할 수 있는지 확인하기 위해. 인체각질형성세포에 실시예 4을 처리한 뒤, 피부 재생과 관련된 피부세포 분화인자인 로리크린 (Loricrin), 인볼루크린 (Involucrin), 시토케라틴10 (Cytokeratin10) 및 데스모글레인2 (Desmoglein2)가 후기 경로에서 발현되는 정도를 웨스턴-블롯으로 확인하였다.

즉, 실시예 1, 실시예 2 및 실시예 3이 적절한 비율로 혼합된 실시예 4는 실시예 1, 실시예 2 및 실시예 3의 각각이 단독으로 갖는 효과에 비해, 더 낮은 농도에서 높은 효과를 보인다. 또한, 실시예 4는 실시예 1, 실시예 2 및 실시예 3이 적절한 비율로 혼합됨으로써 피부의 기저층에서 형성된 각질형성세포가 유극층 및 과립층을 거쳐 최종적으로 각질층에 이르기까지 각 단계의 분화인자 발현을 촉진하여 결과적으로 피부 재생 주기를 촉진하는 효과를 나타낸다.
(실험예 6)
본 발명의 혼합 추출물이 노화 피부에서 비타민D의 기능에 영향을 미치는지 확인하기 위해. 인체각질형성세포에 실시예 4의 혼합추출물을 처리한 뒤, 정상적인 비타민D 기능에 관여하는 비타민D 수용체(Vitamin D receptor)가 후기 경로에서 발현되는 정도를 웨스턴-블롯으로 확인하였다.
구체적으로, 후기경로(p7)의 인체각질형성세포를 각각 60 mm plate에 100 x 104 cells 농도로 접종하고 24시간 배양한 후, 성장인자가 포함되지 않은 배지에 실시예 4의 혼합추출물을 첨가하여 24시간 배양하였다. 배양 후 RIPA Buffer을 처리하여 단백질을 회수하고, BCA 정량을 통해 단백질량이 30㎍이 되도록 샘플을 제조하였다. 제조한 샘플을 SDS-PAGE(Sodiumdodecylsulfate-polyacrylamide gel electrophoresis)를 이용하여 단백질을 분리하고, Electrotransfer을 이용하여 PVDF 막(membrane)에 옮긴 후, 면역블로팅법(immune blotting)을 이용하여 세포 내 발현 수준을 확인하였다.
그 결과, 도 5에 도시된 바와 같이, 실시예 4의 혼합추출물은 비타민D 수용체(Vitamin D receptor)의 발현을 촉진하는 효과가 있음을 확인하였다.
즉, 실시예 4의 혼합추출물은 피부세포에서 비타민D 수용체의 발현을 높임으로써 피부에서 합성되는 비타민D가 정상적인 기능을 하도록 하여 각질형성세포의 분화인자 발현을 촉진하여, 결과적으로 피부 재생 주기를 촉진하는 효과를 나타낸다.
(실험예 7) 피부 각질 개선 확인시험
본 발명의 실시예 4의 혼합 추출물이 피부 각질 개선에 도움을 주는지 확인하기 위해 40~60세 남성 5명을 대상으로 시험하였다.
실시예 4의 혼합 추출물을 오른쪽 얼굴에 아침 및 저녁으로 한번씩 총 4주간 도포하고, SOFT OLUS(Keratin probe) 장비를 이용해 2주마다 피부 각질 변화를 측정하였다.
그 결과, 도 6 및 도 7에 도시된 바와 같이, 아무것도 바르지 않은 왼쪽 얼굴(무처리)에 비해 실시예 4의 혼합 추출물이 도포된 오른쪽 얼굴에서 시간 경과에 따라 각질량이 감소함을 확인하였다.
(실험예 8) 피부결 개선 확인시험
본 발명의 실시예 4의 혼합 추출물이 피부결 개선에 도움을 주는지 확인하기 위해 40~60세 남성 5명을 대상으로 시험하였다.
실시예 4의 혼합 추출물을 오른쪽 얼굴에 아침 및 저녁으로 한번씩 총 4주간 도포하고, OBSERVE 장비를 이용해 2주마다 피부결 변화를 측정하였다.
그 결과, 도 8에 도시된 바와 같이, 아무것도 바르지 않은 왼쪽 얼굴에 비해 실시예 4의 혼합 추출물이 도포된 오른쪽 얼굴의 피부결이 개선됨을 확인하였다.
이상에서 본 발명의 실시예에 대하여 상세하게 설명하였지만 본 발명의 권리범위는 이에 한정되는 것은 아니고, 청구범위에 기재된 본 발명의 기술적 사상을 벗어나지 않는 범위 내에서 다양한 수정 및 변형이 가능하다는 것은 당 기술분야의 통상의 지식을 가진 자에게는 자명할 것이다.
Claims (9)
- 아몬드 추출물, 타히보 추출물 및 흰목이버섯 추출물으로부터 선택된 1종 이상의 추출물을 포함하는 것을 특징으로 하는 피부재생용 화장료 조성물.
- 제 1 항에 있어서, 타히보 추출물과; 아몬드 추출물 또는 흰목이버섯 추출물;을 포함하는 것을 특징으로 하는 피부재생용 화장료 조성물.
- 제 1 항에 있어서, 아몬드 추출물, 타히보 추출물 및 흰목이버섯 추출물을 포함하는 것을 특징으로 하는 피부재생용 화장료 조성물.
- 제 1 항 내지 제 3 항 중 어느 한 항에 있어서, 상기 아몬드 추출물은 전체 조성물 중 0.1~5중량%로 포함되는 것을 특징으로 하는 피부재생용 화장료 조성물.
- 제 1 항 내지 제 3 항 중 어느 한 항에 있어서, 상기 타히보 추출물은 전체 조성물 중 0.1~10중량%로 포함되는 것을 특징으로 하는 피부재생용 화장료 조성물.
- 제 1 항 내지 제 3 항 중 어느 한 항에 있어서, 상기 흰목이버섯 추출물은 0.1~20 중량%로 포함되는 것을 특징으로 하는 피부재생용 화장료 조성물.
- 제 1 항 내지 제 3 항 중 어느 한 항에 있어서, 상기 화장료 조성물이 피부 재생과 관련된 피부세포 분화인자인 로리크린 (Loricrin), 인볼루크린 (Involucrin), 시토케라틴10 (Cytokeratin10) 및 데스모글레인2 (Desmoglein2) 중 2종 이상을 발현시키는 것을 특징으로 하는 피부재생용 화장료 조성물.
- 제 1 항 내지 제 3 항 중 어느 한 항에 있어서, 상기 화장료 조성물이 피부세포 분화와 관련된 인자인 비타민D(Vitamin D) 및 비타민D 수용체(Vitamin D Receptor) 중 1종 이상을 발현시키는 것을 특징으로 하는 피부재생용 화장료 조성물.
- 제 1 항 내지 제 3 항 중 어느 한 항에 있어서, 상기 화장료 조성물이 β-glucosidase 효소활성을 갖는 것을 특징으로 하는 피부재생용 화장료 조성물.
