また、特許文献3には、ラクトバチルス・カルバタスあるいはラクトバチルス・アミロボラスにAtrogin-1の発現によって引き起こされる筋肉の分解を抑制する作用があることが開示されている。
また、非特許文献2(Nutirients)には、ビフィズス菌に筋肉を増加させる作用があることについて開示されている。
また、特許文献1は身体活動の促進に伴う副次的な効果として筋肉量を増加させるものであるが、直接筋合成を促進しまたは筋分解を抑制するものではない。特許文献2についても、損傷した筋の筋修復を促進させるものであって、筋合成を促進し又は筋分解を抑制するものではない。特許文献3は、筋肉の分解を抑制するものであるが限定的な2種類の乳酸菌が開示されるのみである。また非特許文献2は、特定のビフィズス菌に筋肉増加作用があることが開示されるのみで、乳酸菌に関する記載はない。
さらに上記いずれの文献にも、筋合成促進効果および筋分解抑制効果を併せ持つ乳酸菌については開示されていなかった。
一方、筋分解は主としてユビキチン-プロテアソーム系が担っており、筋特異的なユビキチンリガーゼとしてAtrogin-1およびMuRF1 (muscle ring finger 1)が知られている。すなわち、Atrogin-1およびMuRF1の遺伝子発現量が増加すると筋分解が促進され、Atrogin-1およびMuRF1の遺伝子発現量が低下すると筋分解が抑制される。
発明者らは鋭意研究の結果、ストレプトコッカス・サーモフィルス、ラクトコッカス・ラクティス・サブスピーシーズ・クレモリス、ラクトバチルス・ラムノーサスまたはラクトコッカス・ラクティス・サブスピーシーズ・ラクティスを筋細胞に添加することで筋細胞のリン酸化p70S6K量が増加すること、ラクトバチルス・ムコーサエ、ラクトバチルス・ファーメンタムおよびペディオコッカス・アシディラクティシを添加することで、Atrogin-1およびMuRF1の遺伝子発現量が低下することを見出すとともに、ラクトバチルス・デルブレッキー・サブスピーシーズ・ラクティス、ラクトバチルス・ガセリまたはラクトバチルス・ロイテリを添加すると、筋細胞のリン酸化p70S6K量が増加することに加えAtrogin-1及びMuRF1の遺伝子発現量が低下することを見出し、本発明を完成させた。

本発明の筋委縮予防剤に含まれる乳酸菌等は、筋合成促進効果を有する特定の乳酸菌等であり、ラクトバチルス・ガセリ、ストレプトコッカス・サーモフィルス、ラクトバチルス・ラムノーサス、ラクトコッカス・ラクティス、ラクトバチルス・デルブレッキー及びラクトバチルス・ロイテリからなる群から選ばれるいずれか1種以上から得られたものが好ましい。
このうちラクトコッカス・ラクティスは、ラクトコッカス・ラクティス・サブスピーシーズ・クレモリスまたはラクトコッカス・ラクティス・サブスピーシーズ・ラクティスであることがさらに好ましい。
また、上記菌種のうちでも、乳酸菌等がラクトバチルス・ガセリ SBT1848株、ストレプトコッカス・サーモフィルスSBT1021A株、ラクトコッカス・ラクティス・サブスピーシーズ・クレモリスSBT1393株、ラクトバチルス・ラムノーサス SBT2299株、ラクトコッカス・ラクティス・サブスピーシーズ・ラクティス SBT2397株、ラクトバチルス・ロイテリSBT2970株から得られたものであることがさらに好ましい。
また、上記菌種のうちでも、乳酸菌等がラクトバチルス・ロイテリ SBT2970株、ラクトバチルス・ムコーサエ SBT2958株、ラクトバチルス・ムコーサエ SBT10038株、ラクトバチルス・ムコーサエ SBT10043株、ラクトバチルス・ファーメンタム SBT1846株、ラクトバチルス・ファーメンタム SBT1859株、ペディオコッカス・アシディラクティシ SBT3331株またはラクトバチルス・ガセリ SBT1848株から得られたものであることがさらに好ましい。
筋合成促進効果及び筋分解抑制効果を有する乳酸菌等は、ラクトバチルス・デルブレッキー、ラクトバチルス・ロイテリまたはラクトバチルス・ガセリから得られたものが好ましい。ラクトバチルス・デルブレッキーは、さらに、ラクトバチルス・デルブレッキー・サブスピーシーズ・ラクティスから得られたものであることが好ましい。
また、上記菌種のうちでも、乳酸菌等がラクトバチルス・デルブレッキー・サブスピーシーズ・ラクティス SBT1371株、ラクトバチルス・デルブレッキー・サブスピーシーズ・ラクティス SBT2002株、ラクトバチルス・デルブレッキー・サブスピーシーズ・ラクティス SBT2080株、ラクトバチルス・ロイテリSBT2970株またはラクトバチルス・ガセリ SBT1848株から得られたものであることがよりいっそう好ましい。
筋合成促進作用の有無は、後述する実施例に示すように例えば、対象乳酸菌等について、無添加の場合に比べてリン酸化p70S6K量の増大がみられるかどうかにより判断できる。
また、筋分解抑制作用の有無は、実施例に示すように例えば、対象乳酸菌等について、無添加の場合に比べてAtrogin-1およびMuRF1の遺伝子発現量が低下するかどうかにより判断できる。
製剤化に際しては、賦形剤、結合剤、崩壊剤、矯味剤等、一般に用いられる添加物を適宜混合しても良い。
本発明の筋委縮予防用の飲食品としては、前記有効成分となる乳酸菌等が含まれる飲食品であればよく、機能性表示食品、特定保健用食品、栄養補助食品、サプリメントなどが挙げられる。本発明の飲食品は、乳飲料、ヨーグルト、チーズ、アイスクリーム、清涼飲料水、バター、練乳、クラッカー、調製粉乳、粉ミルク、液体ミルク、調味料等が挙げられる。また、既存の飲食品に本発明の有効成分を原料段階、製造途中、製造後に添加してもよい。また、本発明の有効成分は乳酸菌等であるから、例えば本発明の乳酸菌で発酵させたヨーグルト、チーズ、乳飲料などの発酵食品は、有効成分そのものが筋委縮予防用の飲食品の例である。
筋委縮予防剤のこれらの飲食品への配合量は、形態、剤型、投与対象の症状、体重、用途などによって異なるため、特に限定されないが、あえて挙げるなら例えば0.001~100(w/w)%で配合することができる。
筋委縮予防用の飼料としては、通常の飼料に前記有効成分を混合すればよい。
各菌株のグリセロールストックをMRS平板培地に画線し、37℃で3晩(64時間)培養した。次いで平板培地で得られた1コロニーを10 mLのMRS培地に植菌し、37℃または30℃で1晩培養(16時間)した。この菌体培養液 300 μLを新しいMRS培地 10 mLに植え継ぎ、再度37℃または30℃で1晩培養(16時間)した。次に、菌体培養液3 mLをMRS培地100 mLに植菌し、37℃または30℃で1晩培養(16時間)した。得られた菌体培養液を遠心分離し、沈殿した菌体をPBS、超純水で洗浄した。菌体は4 mLの超純水に再縣濁して、-80℃で凍結させた。これを凍結乾燥機(東京理科器械)を用いて凍結乾燥させることで、凍結乾燥菌体を得た。得られた凍結乾燥菌体をPBSまたはDMEM培地(gibco)に溶解し、マルチビーズショッカー(安井機械)を用いて破砕することで乳酸菌破砕物を調製した。
マウス筋芽細胞であるC2C12細胞を、10% FBS、1% penicillin-streptomycin含有DMEM培地を添加した48well 培養プレート(IWAKI)に1.8×104 cell/wellとなるよう播種した。これを37℃、5%CO2条件下で2日間、80%~90%コンフルエントまで培養した。
次に培地をFBSを含まない1% penicillin-streptomycin含有DMEM培地に交換し、4時間培養した。培養後、1% penicillin-streptomycin含有DMEM培地に懸濁した図1に示す各乳酸菌の破砕物を100 μg/mL となるよう添加した。コントロール群には乳酸菌破砕物を含まない1% penicillin-streptomycin含有DMEM培地を同量添加した。このC2C12細胞をさらに2時間培養後、PBSで洗浄し、p70S6K (pT389) SimpleStep ELISA (abcam)付属のCell Extraction Buffer PTRを用いて細胞を溶解した。細胞溶解液を回収し、遠心分離して上清を得ることで、次のELISAで用いるタンパク質溶液サンプルを得た。
ELISAは、リン酸化されたp70S6Kの濃度を検出できる、p70S6K (pT389) SimpleStep ELISAとPre-coated 384 well Microplate SimpleStep ELISA (abcam) を使用して行った。細胞溶解液の総タンパク濃度をBCA法によって定量し、各サンプルの濃度が一定となるようCell Extraction Buffer PTR で希釈した。これらを検量線作成用のコントロールサンプルとともに、25 μL /well ずつPre-coated 384 well Microplate SimpleStep ELISAに添加し、p70S6K (pT389) SimpleStep ELISAのマニュアル記載の方法により発色させた。VARIOSKAN (Thermo Scientific) を使用して、主波長として450 nm、副波長として 620 nmの吸光度をそれぞれ測定し、両波長における吸光度の差を求めた。この値をもとにコントロールサンプルから検量線を作成し、各サンプル中のリン酸化p70S6Kの相対濃度を算出した。
マウス筋芽細胞であるC2C12細胞を、10% FBS、1% penicillin-streptomycin含有DMEM培地を添加した48well 培養プレート(IWAKI)に1.8×104 cell/wellとなるよう播種した。これを37℃、5%CO2条件下で2日間、80%~90%コンフルエントまで培養した。
次に培地を、2%ウマ血清を含む1% penicillin-streptomycin含有DMEM培地に交換し、2から3日おきに培地を交換し、5日間培養して、C2C12細胞を分化させた。その後、同様の培地にAtrogin-1およびMuRF1の遺伝子発現を上昇させることが知られているデキサメタゾンを1μMの濃度で添加し、図2,図3に示す各乳酸菌破砕物を100μg/mL となるよう添加した。24時間培養後、培地を除去し、細胞にSepasol-RNA I Super G(ナカライテスク)を200μL/wellで加え、ピペッティングで細胞を破砕して回収した。回収した液からRNAを抽出し、約500 ngのRNAを鋳型にして、ReverTra Ace qPCR RT Master Mix with gDNA remover(TOYOBO)を用いてcDNAを合成した。その後、THUNDERBIRD SYBR qPCR Mix(TOYOBO)と、各種遺伝子用増幅プライマーを用いて、ViiA7 Real-time PCR system(Thermo Fisher Scientific)でAtrogin-1およびMuRF1の遺伝子発現量を定量した。内在性コントロールとしてはGapdhの遺伝子を使用した。使用したプライマーの配列を表1に示す。
