본 발명은 모낭세포 배양액을 포함하는 모낭 형성 촉진용 조성물 및 이를 이용한 다양한 용도에 관한 것이다.
Description
본 발명은 모낭조직배양액을 포함하는 탈모의 예방 또는 치료용 조성물 및 이의 다양한 용도에 관한 것이다.
모낭이란 모근을 싸고 있는 주머니 모양의 상피성 조직을 말하며, 모근이란 모발이 피부에 박힌 부분을 말한다. 모발은 모낭 근처에서 세포 분열이 활발히 일어나 생장하게 된다. 만약 이 세포 분열 능력이 저하되면 모낭은 위축하게 되고 피부의 얕은 부위로 이동한다. 정상적으로는 세포 분열 능력을 다시 획득하게 되어 생성된 신생 세포군이 피부의 깊은 곳으로 이동하여 새로운 모낭이 형성되고 모발이 자라나 묵은 모발을 밀어내는데, 이와 같은 '모발의 교환'이 일어나지 않으면 탈모증이 되는 것이다.
지금까지 개발된 탈모의 최적 치료방법은 모발이식이지만, 탈모 초기에는 FDA 승인을 받아 시중에 판매되는 외용제 미녹시딜 (Minoxidil) 이나 경구투여제인 피나스테라이드 (finasteride)를 주성분으로 하는 프로페시아 (Propecia) 등으로도 탈모 진행을 둔화시킬 수 있다.
초기 탈모 치료에 유용한 유효물질을 찾아내기 위해, 로(Roh)는 생체 실험으로, 쥐를 제모한 후 시료를 등에 도포한 후 발모현상을 관찰하였으며, 필폿(Philpott)은 쥐의 콧수염을 이용한 생체 외 기관배양을 통해 IGF (Insulin-like growth factor), EGF (Epidermal growth factor), HGF(Hepatocyte growth factor) 등의 성장인자의 영향을 확인하는 동물 실험을 진행해왔지만 일정한 주기로 털갈이를 하는 설치류와 달리 사람은 모낭마다 별개의 주기를 가지고 있기 때문에 성장인자에 대한 경향이 모두 일치하지는 않았다. 반면, 원숭이 모델의 경우 사람과 경향은 유사하지만 시간과 실험비용의 부담이 너무 크다. 또한, 최근 동물 보호 차원에서 동물을 이용한 생체 실험이 사회적으로 비판을 받고 있으며 법적으로도 규제가 되고 있어서, 동물 시험을 대체할 생체 외독성 검사 모델의 개발이 전 세계적인 문제로 대두되고 있다.
생체 실험과 달리 생체 외 실험의 경우에는, 세포 배양시 혈관을 통한 영양이나 산소 공급이 차단된 상황이므로 장기간의 실험에 어려움이 있다. 또한, 효능성분의 효과를 확인하기 전에 인공조직의 급속한 세포사로 인해 실험이 종결되는 상황이 생길 수 있으므로 생체 외 실험의 경우 인공조직의 기능을 생체와 유사한 환경으로 조성하여 장기간 유지시키는 것이 가장 중요하다.
모낭 기관 배양을 이용한 체외 모발 성장모델로는 액침 배양으로 DMEM (Dulbecco's Modified Eagle Medium)에 혈청을 10 중량% 첨가한 후 유리병 안에서 배양한 연구가 있으며, 산소압을 증가시키고 31℃에서 RPMI 1640 배지로 배양하는 경우, 일반적인 배양 온도인 37℃에서 배양하는 것보다 길이 성장속도와 길이 증가량은 감소되지만 모구의 형태변화는 적게 나타난 경우가 있다.
2000년 이후로 보편적으로 사용되는 모낭 기관 배양을 위한 배지는 윌리엄 배지 E(Williams' medium E)에 10 ㎍/㎖ 인슐린 (insulin), 10 ㎍/㎖ 트랜스페린 (transferrin), 10 ㎍/㎖ 소듐 셀레나이트(sodium selenite) 및 10 ㎍/㎖ 하이드로코티손 (hydrocortisone)이 첨가된 배지 조성을 가진다.
본 발명의 일 목적은 제1용기에 ROCK 저해제 및 기저막단백질을 첨가한 후, 모낭세포를 상기 제1용기에 부착하는 (a)단계; 상기 제1용기에 배지를 처리한 후, 세포 배양기에서 배양하는 (b)단계; 상기 모낭세포를 제2용기에 부착하여 계대하는 (c)단계; 및 상기 제2용기에 상기 배지를 첨가한 후 배양하는 (d)단계;에 의해 배양된 모낭세포 배양액을 포함하는 모낭 형성 촉진용 조성물에 관한 것이다.

본 발명에서, "ROCK 저해제"란 Rho 키나아제에 의해 매개되는 시그널을 저해할 수 있는 물질을 말한다. 본 발명에서 진피와 표피의 접합에 있어 기저막(basement membrane: BM)은 여러가지 기능을 가지며, 가장 명백한 기능은 표피와 진피를 근접 연결시키는 것이며, 이러한 방식으로 2층의 조직들의 기계적으로 안정적인 점착(cohesion)을 확보한다. 또한 기저막은 표피의 "극성"과 구성에도 영향을 주는 동시에 표피와 진피 사이에 명확한 경계가 된다. 기저막은 각질형성 세포의 표피 분화를 개시하고, 기저 세포층의 증식적인 상태를 유지시키는 것으로 추정된다. 정상 조건하에서는, 기저막은 또한 표피 세포와 진피의 직접 접촉을 방지한다. 그러나, 기저막에 손상이 가해지는 상처가 발생되면, 진피와의 직접 접촉이 이루어지게 되고, 그런 후 세포는 행태를 변화시켜 상처치유 과정을 개시하게 된다

본 발명에서 표피성장인자 (Epidermal growth factor; EGF)는 수용체 EGFR에 결합하여 세포 성장, 증식, 분화를 촉진하며, 인간 EGF는 53개의 아미노산으로 이루어진다 (Exp Cell Res., 284(1):2-13, 2003). 또한 이와 같은 EGF는 피부 및 헤어의 재생에 중요한 역할을 한다고 보고되고 있다(J Dermatol Sci., 72(2):81-86, 2013).
본 발명에서 상기 섬유아세포 성장인자(Fibroblast growth factor; FGF)는 FGF1 (aFGF), FGF2 (bFGF), FGF3 (int-2), FGF4 (HST), FGF5, FGF6 (HST2), FGF7 (KGF), FGF9, FGF10, FGF11, FGF12, FGF12B, FGF14, FGF16, FGF17, FGF19, FGF20, FGF21 및 FGF23으로 이루어진 군에서 선택된 1종 이상일 수 있으나 이에 제한되는 것은 아니다.
본 발명에서 상기 EGF와 상기 FGF4의 농도비는 1:10 내지 10:1일 수 있다. 상기 EGF의 농도는 10ng/ml 내지 500ng/ml이고, 상기 FGF4의 농도는 10ng/ml 내지 500ng/ml 일 수 있다.

본 발명에서, "세포외 매트릭스(ECM)"란 조성이 다양한 3차원 분자 복합체로, 성장인자 및 사이토킨 저장소 성질에 기인하여, 형태형성, 증식, 세포 분화 및 조직 회복에 관여한다. 이에 한정하지는 않지만, 예를 들어 상기 매트릭스는 매트리겔(Matrigel), NuFF 배양보조세포(feeder cell) 및 겔트렉스(Geltrax)에서 선택된 어느 하나 이상인 것이다.
본 발명에서 상기 (b) 및 (d)단계에서 배양하는 기간은 20 내지 40일일 수 있고, 바람직하게는 25 내지 35일일 수 있다.
본 발명의 다른 일 구현 예에 따르면, 제1용기에 ROCK 저해제 및 기저막단백질을 처리한 후, 모낭세포를 상기 제1용기에 부착하는 (a)단계; 상기 제1용기에 배지를 처리한 후 배양하는 (b)단계; 상기 모낭세포를 제2용기에 부착하여 계대하는 (c)단계; 및 상기 제2용기에 상기 배지를 처리한 후 배양하는 (d)단계;에 의해 배양된 모낭세포 배양액을 포함하는 탈모의 예방, 완화, 치료용 조성물; 또는 발모 또는 육모 촉진용 조성물에 관한 것이다.
본 발명의 상기 세포 배양 방법에 관한 기재는 앞서 기재된 바와 중복되어 명세서의 과도한 복잡을 피하기 위하여 이하 그 자세한 기재를 생략한다.
본 발명에서 용어 "탈모"는 정상적으로 모발이 존재해야 하는 부위에 모발이 결손된 상태를 의미하며, 일반적으로 두피의 성모가 빠지는 것을 의미한다. 성모는 색이 없고 굵기가 가는 연모와는 달리 결손 시 미용상의 문제를 일으킬 수 있다.
본 발명에서 상기 탈모는 임상적으로 흉터가 형성되는 것과 그렇지 않은 두 종류로 구분될 수 있으며, 흉터가 형성되는 탈모(반흔성)는 모낭이 파괴되므로 모발이 재생되지 않는 반면, 흉터가 형성되지 않는 탈모(비반흔성)는 모낭이 유지되므로 증상 부위가 사라진 후에 모발이 재생된다.
본 발명에서 상기 탈모는 원형 탈모(alopecia areata), 전두 탈모(alopecia totalis), 범발성 탈모(alopecia universalis), 남성형 탈모(androgenic alopecia), 휴지기 탈모(telogen effluvium), 성장기 탈모(anagen effluvium), 또는 화학치료-유발 탈모(chemotherapy-induced alopecia)일 수 있으나, 이에 한정되는 것은 아니다.
본 발명의 탈모의 예방, 완화, 치료 또는 발모 또는 육모 촉진용 조성물은 약학적 조성물, 화장료 조성물 또는 식품 조성물로도 제공될 수 있다.
본 발명의 또 다른 구현 예에 따르면, 본 발명에 따른 상기 모낭세포 배양액을 포함하는 조성물을 유효량으로 개체에 투여하는 단계를 포함하는, 탈모의 예방, 완화, 치료 방법; 또는 발모 또는 육모 촉진 방법에 관한 것이다.
본 발명에서 상기 "개체"란, 탈모, 상처의 예방 또는 치료가 필요한 개체 또는 피부 개선이 필요한 개체로서, 포유동물 및 비-포유동물을 모두 포함할 수 있다. 여기서, 상기 포유동물의 예로는 인간, 비-인간 영장류, 예컨대 침팬지, 다른 유인원 또는 원숭이 종; 축산 동물, 예컨대 소, 말, 양, 염소, 돼지; 사육 동물, 예컨대 토끼, 개 또는 고양이; 실험 동물, 예를 들어 설치류, 예컨대 래트, 마우스 또는 기니아 피그 등을 포함할 수 있으나, 이에 제한되는 것은 아니다.
본 발명에서 상기 "투여"란, 임의의 적절한 방법으로 개체에게 본 발명의 유효성분을 도입하는 과정을 의미하는 것으로서, 상기와 같이 투여되는 조성물의 제제는 특별히 제한하지 않으며, 고체 형태의 제제, 액체 형태의 제제 또는 흡인용 에어로졸 제제로 투여될 수 있으며, 사용하기 바로 전에 경구 또는 비경구 투여용 액체 형태 제제로 전환되도록 의도되는 고체 형태 제제로 투여될 수 있고, 예를 들면, 산제, 과립제, 캡슐, 정제, 수성 현탁액 등의 경구형 제형, 외용제, 좌제 및 멸균 주사용액의 형태로 제형화하여 투여될 수 있으나, 이에 제한되는 것은 아니다.
또한, 본 발명에서 상기 투여 시 본 발명의 조성물과 함께 약학적으로 허용 가능한 담체를 추가로 투여할 수 있다. 여기서, 상기 약학적으로 허용되는 담체는 경구 투여 시에는 결합제, 활탁제, 붕해제, 부형제, 가용화제, 분산제, 안정화제, 현탁화제, 색소, 향료 등을 사용할 수 있으며, 주사제의 경우에는 완충제, 보존제, 무통화제, 가용화제, 등장제, 안정화제 등을 혼합하여 사용할 수 있으며, 국소투여용의 경우에는 기제, 부형제, 윤활제, 보존제 등을 사용할 수 있다. 본 발명의 조성물의 제형은 상술한 바와 같은 약학적으로 허용되는 담체와 혼합하여 다양하게 제조될 수 있다. 예를 들어, 경구 투여시에는 정제, 트로키, 캡슐, 엘릭서(elixir), 서스펜션, 시럽, 웨이퍼 등의 형태로 제조할 수 있으며, 주사제의 경우에는 단위 투약 앰플 또는 다수회 투약 형태로 제조할 수 있다. 기타, 용액, 현탁액, 정제, 캡슐, 서방형 제제 등으로 제형할 수 있다.
한편, 제제화에 적합한 담체, 부형제 및 희석제의 예로는, 락토즈, 덱스트로즈, 수크로즈, 솔비톨, 만니톨, 자일리톨, 에리스리톨, 말디톨, 전분, 아카시아 고무, 알지네이트, 젤라틴, 칼슘 포스페이트, 칼슘 실리케이트, 셀룰로즈, 메틸 셀룰로즈, 미정질 셀룰로즈, 폴리비닐피롤리돈, 물, 메틸하이드록시벤조에이트, 프로필하이드록시벤조에이트, 탈크, 마그네슘 스테아레이트 또는 광물유 등이 사용될 수 있다. 또한, 충진제, 항응집제, 윤활제, 습윤제, 향료, 유화제, 방부제 등을 추가로 포함할 수 있다.
본 발명에 따른 조성물의 투여 경로는 이들로 한정되는 것은 아니지만 구강, 정맥 내, 근육 내, 동맥 내, 골수 내, 경막 내, 심장 내, 경피, 피하, 복강 내, 비강 내, 장관, 국소, 설하 또는 직장이 포함된다. 경구 또는 비경구 투하가 바람직하다.
본 발명에서, "비경구"는 피하, 피내, 정맥내, 근육내, 관절내, 활액낭내, 흉골내, 경막내, 병소내 및 두개골내 주사 또는 주입기술을 포함한다. 본 발명의 약학적 조성물은 또한 직장 투여를 위한 좌제의 형태로 투여될 수 있다.
본 발명에서, "약학적으로 유효한 양"은 바람직한 생물학적 결과를 제공하기 위한 작용제의 충분한 양을 지칭한다. 상기 결과는 질환의 징후, 증상 또는 원인의 감소 및/또는 완화, 또는 생물계의 임의의 다른 바람직한 변화일 수 있다. 예를 들어, 치료 용도를 위한 "유효량"은 질환에서 임상적으로 유의한 감소를 제공하는데 요구되는, 본 발명에 개시된 조성물의 양이다. 임의의 개별적인 경우에서 적절한 "효과적인" 양은 일상적인 실험을 사용하여 당업자에 의해 결정될 수 있다. 따라서, 표현 "유효량"은 일반적으로 활성 물질이 치료 효과를 갖는 양을 지칭한다. 본 발명의 경우에, 활성 물질은, 암의 예방, 개선 또는 치료제이다.
본 발명의 조성물은 사용된 특정 조성물의 활성, 연령, 체중, 일반적인 건강, 성별, 정식, 투여 시간, 투여 경로, 배출율, 약물 배합 및 예방 또는 치료될 특정 질환의 중증을 포함한 여러 요인에 따라 다양하게 변할 수 있고, 상기 조성물의 투여량은 환자의 상태, 체중, 질병의 정도, 약물 형태, 투여 경로 및 기간에 따라 다르지만 당업자에 의해 적절하게 선택될 수 있고, 1일 0.0001 내지 100mg/kg 또는 0.001 내지 100mg/kg으로 투여할 수 있다. 투여는 하루에 한번 투여할 수도 있고, 수회 나누어 투여할 수도 있다. 상기 투여량은 어떠한 면으로든 본 발명의 범위를 한정하는 것은 아니다. 본 발명에 따른 조성물은 환제, 당의정, 캡슐, 액제, 겔, 시럽, 슬러리, 현탁제로 제형될 수 있다.
본 발명의 또 다른 일 구현 예에 따르면, 제1용기에 ROCK 저해제 및 기저막단백질을 처리한 후, 모낭세포를 상기 제1용기에 부착하는 (a)단계; 상기 제1용기에 배지를 처리한 후 배양하는 (b)단계; 상기 모낭세포를 제2용기에 부착하여 계대하는 (c)단계; 및 상기 제2용기에 상기 배지를 처리한 후 배양하는 (d)단계;에 의해 배양된 모낭세포 배양액을 포함하는 상처의 예방, 치료 또는 완화용 조성물일 수 있다.

본 발명의 상처의 예방, 치료 또는 완화용 조성물은 약학적 조성물, 화장료 조성물 또는 식품 조성물로도 제공될 수 있다.
본 발명의 또 다른 구현 예에서 본 발명에 따른 상기 조성물을 유효량으로 개체에 투여하는 단계를 포함하는, 상처의 예방, 치료 또는 완화 방법에 관한 것이다.

본 발명에서 용어, "피부 개선"은 피부의 내재적 요인 또는 외인적인 요인에 의하여 유발되는 피부의 손상을 치료, 경감, 완화시키는 과정 또는 그의 효과 등을 포괄적으로 의미하는 것으로, 보다 상세하게는 피부 미백, 주름 개선, 탄력 증진, 피부 재생, 피부 보습, 노화 방지, 피부 자극 완화, 여드름 예방 또는 개선, 및/또는 아토피 예방 또는 개선 효과를 포함할 수 있으나, 이에 제한되는 것은 아니다.
본 발명의 조성물은 매우 우수한 줄기세포 증식 촉진능을 갖기 때문에 이를 인체에 적용하는 경우 피부에 존재하는 성체 줄기세포의 증식을 촉진하거나 활성화시킴으로써 피부 상태를 개선시킬 수 있다. 피부에 존재하는 성체 줄기세포는 예를 들어, 모낭과 모낭사이의 표피(interfollicular epidermis)의 기저층에 존재하는 줄기세포, 모낭(hair follicle)에 존재하는 줄기세포, 또는 피하 지방층에 존재하는 지방유래 줄기세포이다.
본 발명의 피부 개선용 조성물은 화장료 조성물, 식품 조성물 또는 약학적 조성물로도 제공될 수 있다.
본 발명의 또 다른 구현 예에서 본 발명에 따른 상기 조성물을 유효량으로 개체에 투여하는 단계를 포함하는, 피부 개선 방법에 관한 것이다.
본 발명의 상기 모낭세포 배양 방법, 상기 세포, 상기 화장료 조성물 및 상기 약학적 조성물에 관한 기재는 앞서 기재된 바와 중복되어 명세서의 과도한 복잡을 피하기 위하여 이하 그 자세한 기재를 생략한다.
본 발명에 따른 모낭세포 배양액은 상처 부위로 세포가 이동하는 것을 촉진하여 우수한 상처 치료 활성을 갖는다. 또한 섬유아세포의 활성도를 높여 피부 보습, 피부 미백, 주름 개선 및 노화 방지 등의 피부개선 효과가 우수하다. 또한, 본 발명에 따른 모낭세포 배양액은 발모 및 육모 효과가 우수하여 탈모의 방지 및 치료 효과가 뛰어나며, 이를 섭취하거나 피부에 도포하여도 부작용이 없이 안전하다.
본 발명에서, "예방"은 본 발명의 조성물을 이용하여 각종 질환의 증상을 차단하거나, 질환 증상의 억제 또는 지연시키는 모든 행위라면 제한없이 포함할 수 있다.
본 발명에서, "치료"는 본 발명의 조성물을 이용하여 각종 질환의 증상이 호전되거나 이롭게 되는 모든 행위라면 제한없이 포함할 수 있다.
본 발명의 조성물은 사용된 특정 조성물의 활성, 연령, 체중, 일반적인 건강, 성별, 정식, 투여시간, 투여경로, 배출율, 약물 배합 및 예방 또는 치료될 특정 질환의 중증을 포함한 여러 요인에 따라 다양하게 변할 수 있고, 상기 약학적 조성물의 투여량은 환자의 상태, 체중, 질병의 정도, 약무형태, 투여경로 및 기간에 따라 다르지만 당업자에 의해 적절하게 선택될 수 있고, 1일 0.0001 내지 50mg/kg 또는 0.001 내지 50mg/kg으로 투여할 수 있다. 투여는 하루에 한번 투여할 수도 있고, 수회 나누어 투여할 수도 있다. 상기 투여량은 어떠한 면으로든 본 발명의 범위를 한정하는 것은 아니다. 본 발명에 따른 약학적 조성물은 환제, 당의정, 캡슐, 액제, 겔, 시럽, 슬러리, 현탁제로 제형될 수 있다.
본 발명에서 화장료 조성물은 화장수, 영양로션, 영양에센스, 마사지 크림, 미용 목욕물 첨가제, 바디로션, 바디밀크, 배스오일, 베이비오일, 베이비파우더, 샤워겔, 샤워크림, 선스크린로션, 선스크린크림, 선탠크림, 스킨로션, 스킨크림, 자외선차단용 화장품, 크렌징밀크, 탈모제{화장용}, 페이스 및 바디로션, 페이스 및 바디크림, 피부미백크림, 핸드로션, 헤어로션, 화장용크림, 쟈스민오일, 목욕비누, 물비누, 미용비누, 샴푸, 손세정제(핸드클리너), 약용비누{비의료용}, 크림비누, 페이셜 워시, 전신 세정제, 두피 세정제, 헤어린스, 화장비누, 치아미백용 겔, 치약 등의 형태로 제조될 수 있다. 이를 위해 본 발명의 조성물은 화장료 조성물의 제조에 통상적으로 사용하는 용매나, 적절한 담체, 부형제 또는 희석제를 더 포함할 수 있다.
본 발명의 화장료 조성물 내에 더 추가될 수 있는 용매의 종류는 특별히 한정하지 않으나, 예를 들어, 물, 식염수, DMSO 또는 이들의 조합을 사용할 수 있고, 담체, 부형제 또는 희석제로는 정제수, 오일, 왁스, 지방산, 지방산 알콜, 지방산 에스테르, 계면활성제, 흡습제(humectant), 증점제, 항산화제, 점도 안정화제, 킬레이팅제, 완충제, 저급 알콜 등이 포함되지만, 이에 제한되는 것은 아니다. 또한, 필요에 따라 미백제, 보습제, 비타민, 자외선 차단제, 향수, 염료, 항생제, 항박테리아제, 항진균제를 포함할 수 있다.
상기 오일로서는 수소화 식물성유, 피마자유, 면실유, 올리브유, 야자인유, 호호바유, 아보카도유가 이용될 수 있으며, 왁스로는 밀랍, 경랍, 카르나우바, 칸델릴라, 몬탄, 세레신, 액체 파라핀, 라놀린이 이용될 수 있다.
지방산으로는 스테아르산, 리놀레산, 리놀렌산, 올레산이 이용될 수 있고, 지방산 알콜로는 세틸 알콜, 옥틸 도데칸올, 올레일 알콜, 판텐올, 라놀린 알콜, 스테아릴 알콜, 헥사데칸올이 이용될 수 있으며 지방산 에스테르로는 이소프로필 미리스테이트, 이소프로필 팔미테이트, 부틸 스테아레이트가 이용될 수 있다. 계면 활성제로는 당업계에 알려진 양이온 계면활성제, 음이온 계면활성제 및 비이온성 계면활성제가 사용가능하며 가능한 한 천연물 유래의 계면활성제가 바람직하다.
그 외에도 화장품 분야에서 널리 알려진 흡습제, 증점제, 항산화제 등을 포함할 수 있으며, 이들의 종류와 양은 당업계에 공지된 바에 따른다.
본 발명의 식품 조성물은 각종 식품류, 예를 들어, 음료, 껌, 차, 비타민 복합제, 분말, 과립, 정제, 캡슐, 과자, 떡, 빵 등의 형태로 제조될 수 있다.
본 발명의 조성물이 식품 조성물에 포함될 때 그 양은 전체 중량의 0.1 내지 50%의 비율로 첨가할 수 있다.
여기서, 상기 식품 조성물이 음료 형태로 제조되는 경우 지시된 비율로 상기 식품 조성물을 함유하는 것 외에 특별한 제한점은 없으며 통상의 음료와 같이 여러가지 향미제 또는 천연 탄수화물 등을 추가 성분으로서 함유할 수 있다. 즉, 천연 탄수화물로서 포도당 등의 모노사카라이드, 과당 등의 디사카라이드, 슈크로스 등의 및 폴리사카라이드, 덱스트린, 시클로덱스트린 등과 같은 통상적인 당 및 자일리톨, 소르비톨, 에리트리톨 등의 당알콜 등을 포함할 수 있다. 상기 향미제로서는 천연 향미제(타우마틴, 스테비아 추출물(예를 들어 레바우디오시드 A, 글리시르히진등) 및 합성 향미제(사카린, 아스파르탐 등) 등을 들 수 있다.
그 외 본 발명의 식품 조성물은 여러 가지 영양제, 비타민, 광물(전해질), 합성 풍미제 및 천연 풍미제 등의 풍미제, 착색제, 펙트산 및 그의 염, 알긴산 및 그의 염, 유기산, 보호성 콜로이드 증점제, pH 조절제, 안정화제, 방부제, 글리세린, 알콜, 탄산 음료에 사용되는 탄산화제 등을 함유할 수 있다.
이러한 성분은 독립적으로 또는 조합하여 사용할 수 있다. 이러한 첨가제의 비율은 그렇게 중요하진 않지만 본 발명의 조성물 100 중량부 당 0.1 내지 약 50 중량부의 범위에서 선택되는 것이 일반적이다.

본 발명의 일 목적은 제1용기에 ROCK 저해제 및 기저막단백질을 첨가한 후, 모낭세포를 상기 제1용기에 부착하는 (a)단계; 상기 제1용기에 배지를 처리한 후, 세포 배양기에서 배양하는 (b)단계; 상기 모낭세포를 제2용기에 부착하여 계대하는 (c)단계; 및 상기 제2용기에 상기 배지를 첨가한 후 배양하는 (d)단계;에 의해 배양된 모낭세포 배양액을 포함하는 모낭 형성 촉진용 조성물에 관한 것이다.
이하, 본 발명을 하기의 실시예에 의해 상세히 설명한다. 단, 하기 실시예는 본 발명을 예시하는 것일 뿐, 본 발명의 내용이 하기 실시예에 의해 한정되는 것은 아니다.
[준비예 1] 모낭세포의 배양

도 1 및 도 3에 따라 제조예 1의 모낭세포를 배양하였다. 도 2 및 도 4와 같이, 모낭조직을 6 웰 플레이트에 돌배코 변형 이글배지(Dulbecco Modified Eagle Medium; DMEM) + Y-27632 (10 μM) + I-매트릭스(재조합 라미닌 E8 프래그먼트) (5 μl/2ml) + 1% 페니실린에 첨가하여 부착시킨 후 세포 배양기에 넣어두었다. 약 30일간 배지 변경 없이 배지가 부족할 때 적당량 채워주었다.
30일 후 전체 모발(Whole-hair)을 포함한 모든 세포를 계대하는데, 이때는 도 5와 같이 겔트랙스(Geltrax)로 코팅을 한 뒤 Y-27632 (10 μM)을 넣고 세포를 부착시켰다. 이후에도 30일마다 겔트랙스로 코팅한후 Y-27632 (10 μM)을 첨가하고 세포를 부착시켜 배양했다. 배지는 돌배코 변형 이글배지(Dulbecco Modified Eagle Medium; DMEM) (5 μl/2ml) + 1% 페니실린 + 10% FBS를 사용하였고, 배지가 부족할 때 적당량 채워주었다.

모낭세포 배양액을 수득하기 위하여, 제조예 2 내지 4의 모낭세포를 배양하였다. 모낭세포를 6 웰 플레이트에 돌배코 변형 이글배지(Dulbecco Modified Eagle Medium; DMEM) + Y-27632 (10 μM) + I-매트릭스(재조합 라미닌 E8 프래그먼트) (5 μl/2ml) + 1% 페니실린에 첨가하여 부착시킨 후 세포 배양기에 넣어두었다. 이후 약 30일간 배지가 부족할 때 적당량 채워주었다. 이때 사용된 배지의 조성비는 하기 표 1과 같다.

30일 후 전체 모발(Whole-hair)을 포함한 모든 세포를 계대하는데, 이때는 도 5와 같이 겔트랙스(Geltrax)로 코팅을 한 뒤 Y-27632 (10 μM)을 넣고 세포를 부착시켰다. 이후에도 30일마다 겔트랙스로 코팅한후 Y-27632 (10 μM)을 첨가하고 세포를 부착시켜 배양했다. 마찬가지로 배지는 상기 표 1의 배지가 부족할 때 적당량 채워주었다.
[1-3] 모낭세포의 배양(3)

상기 표 2에서와 같이, 비교예 1의 배양액은 제조예 2의 배양액에서 Y-27632를 제외하고는 동일한 농도를 가지는 배양액이고, 비교예 2의 배양액은 제조예 3의 배양액에서 Y-27632를 제외하고는 동일한 농도를 가지는 배양액이며, 비교예 3의 배양액은 제조예 4의 배양액에서 Y-27632를 제외하고는 동일한 농도를 가지는 배양액에 해당한다.
30일 후 전체 모발(Whole-hair)을 포함한 모든 세포를 계대하는데, 이때는 도 5와 같이 겔트랙스(Geltrax)로 코팅을 한 뒤 세포를 부착시켰다. 이후에도 30일마다 겔트랙스로 코팅한후 세포를 부착시켜 배양했다. 마찬가지로 배지는 상기 표 2의 배지가 부족할 때 적당량 채워주었다.
[1-4] 모낭세포의 배양(4)
모낭조직에 코팅을 하지 않고 제조예 1과 같은 성분의 배지만 첨가하여 배양하였다. 그 결과, 도 6과 같이 모낭조직의 모낭세포가 부유하여 계대 과정에서 지속적으로 세포를 잃어 감소하는 것을 확인할 수 있었다.
[1-5] 모낭줄기세포의 확인
제조예 1의 모낭조직에서 상기 모간을 제거한 것에 도 7과 같이 미분화줄기세포마커인 OCT4 항체를 이용하여 세포의 핵(파란색)과 OCT4(빨강색)를 염색한 결과, 제조예 1의 모낭세포 배양배지에 모낭줄기세포가 존재함을 확인하였다
[준비예 2] 모낭세포 배양액의 분리
준비예 1의 과정으로 배양된 모낭세포를 계대배양하는 과정에서 배양액을 수득하였고, 이를 원심분리하여 상등액을 여과하여 모낭세포 배양액을 추출하였다. 구체적으로는 먼저 세포를 트립신 EDTA로 떼어낸후 3000RPM에서 5분동안 원심분리하였다. 다음으로 상층액만 걷어서 다시 3000RPM에서 5분동안 원심분리하였다. 다음으로 상층액만 걷어서 0.22 μM 크기의 필터시스템을 이용하여 세포를 포함한 이물질을 제거하였다.
[실험예 1] 모낭세포 배양액의 효과 확인(1)
[1-1] 섬유아세포에 모낭세포 배양액을 처리한 경우 이동능 비교
제조예 1의 모낭세포 배양액(Follicle로 표시)이 포함된 조성물의 효능을 확인하는 실험을 실시하였다. 도 8과 같이, 섬유아세포에 모낭세포 배양액이 포함된 조성물과 대조군을 처리하여, 섬유아세포 이동능을 확인하였다. 도 8의 점선은 섬유아세포가 이동하기 전을 표시한 것이고, 도 8의 상단 사진들은 섬유아세포에 배양액을 처리하여 이동하기 전을 촬영한 것이며, 도 8의 하단 사진들은 섬유아세포에 배양액을 처리한 후 48시간이 지난 후를 촬영한 것이다. 도 8과 같이 조성물을 처리하지 않은 대조군(CTRL)에 비하여 EGF+FGF4+모낭세포 배양액 조성물을 처리하였을 때 섬유아세포가 눈에 띄게 이동하는 것을 확인할 수 있었다. 상기 EGF의 농도는 200ng/ml, 상기 FGF4의 농도는 100ng/ml로 하였다.
[1-2] 섬유아세포에 모낭세포 배양액을 처리한 경우 유전자 발현 비교
제조예 1의 모낭세포 배양액(Follicle media로 표시)이 포함된 조성물의 효능을 확인하기 위해 섬유아세포에 모낭세포 배양액 조성물을 처리하는 실험을 실시하였다. 도 9는 섬유아세포에 모낭세포 배양액이 포함된 조성물을 처리한 경우 콜라겐 유전자 발현량을 그래프로 나타낸 것으로 EGF+FGF4+모낭세포 배양액 조성물을 처리한 경우, 대조군에 비해 콜라겐 유전자인 COLA1 및 COLA2의 발현량이 증가하였다.
도 10은 섬유아세포에 EGF+FGF4와 EGF+FGF4+모낭세포 배양액 조성물을 각각 처리한 경우, 엘라스틴 프로틴 유전자(ELN)의 발현량을 나타낸 것으로, EGF+FGF4에 비해 EGF+FGF4+모낭세포 배양액 조성물에서 ELN의 발현이 증가하였다.
도 11은 섬유아세포에 EGF, FGF4, EGF+FGF4, EGF+FGF4+모낭세포 배양액 조성물을 각각 처리한 경우, 각질세포 성장 인자(Keratinocyte Growth Factor; KGF)의 발현량을 나타낸 결과로, KGF 유전자의 발현량은 EGF+FGF4+모낭세포 배양액 성분의 배양액, EGF+FGF4, FGF4, EGF 순으로 발현량이 높았다. 이때, EGF의 농도는 200ng/ml, FGF4의 농도는 100ng/ml로 처리하였다.
도 12는 모낭세포 배양액이 포함된 조성물을 처리한 경우 섬유아세포의 증식량을 나타낸 것이다. 5만개의 섬유아세포를 파종(seeding)하여 7일 후의 세포양을 계산한 결과, 도 12와 같이 대조군은 변화가 없었으며, EGF+FGF4+모낭세포 배양액 조성물, EGF+FGF4, EGF, FGF4 순으로 세포가 증가하였다. 이때, EGF의 농도는 200ng/ml, FGF4의 농도는 100ng/ml로 처리하였다.
상기와 같이, 본 발명에 따라 EGF+FGF4+모낭세포 배양액 조성물을 처리하는 경우 EGF, FGF4를 단독으로 처리한 경우보다 모낭형성에 필요한 유전자 발현 및 세포의 이동 및 증식이 활성화되는 것을 확인하였다.
[실험예 2] 모낭세포 배양액의 효과 확인(2)
[2-1] 모낭세포 배양액에 따른 세포 활성 효과 비교(1)
모낭세포를 배양할 때 모낭세포 배양액에 Y-27632이 포함되었는지 여부에 따라 세포 활성 효과의 차이가 있는지 여부를 판단하기 위하여 FGF-10 발현 수준을 확인하였다. 구제척으로는, 제조예 2 내지 4에 의해 제조된 모낭세포 배양액과 비교예 1 내지 3에 의해 제조된 모낭세포 배양액을 진피유두세포에 처리한 경우 FGF-10 관련 mRNA 발현량을 비교하여 도 13에 나타내었다.
도 13에 나타난 것처럼, 제조예 2 내지 4에 의해 제조된 모낭세포 배양액의 경우 비교예 1 내지 3에 의해 제조된 모낭세포 배양액에 비하여 FGF-10 관련 mRNA의 발현량이 현저히 높은 것을 확인한 바, 모낭세포를 배양할 때 모낭세포 배양액에 Y-27632이 포함된 경우, 모낭세포 배양액의 세포 활성 효과가 현저한 것을 확인하였다.
[2-2] 모낭세포 배양액에 따른 세포 활성 효과 비교(2)
모낭세포를 배양할 때 모낭세포 배양액에 Y-27632이 포함되었는지 여부에 따라 세포 활성 효과의 차이가 있는지 여부를 판단하기 위하여 FGF-7 및 vEGF 발현 수준을 확인하였다. 구제척으로는, 제조예 4에 의해 제조된 모낭세포 배양액과 비교예 3에 의해 제조된 모낭세포 배양액을 진피유두세포에 처리한 경우 FGF-7 및 vEGF 관련 mRNA 발현량을 비교하여 도 14에 나타내었다.
도 14에 나타난 것처럼, 제조예 4에 의해 제조된 모낭세포 배양액의 경우 비교예 3에 의해 제조된 모낭세포 배양액에 비하여 FGF-7 및 vEGF 관련 mRNA의 발현량이 현저한 것을 확인한 바, 모낭세포를 배양할 때 모낭세포 배양액에 Y-27632이 포함된 경우, 모낭세포 배양액의 세포 활성 효과가 현저히 높은 것을 확인하였다.
이상에서 본 발명의 실시예에 대하여 상세하게 설명하였지만 본 발명의 권리범위는 이에 한정되는 것은 아니고, 청구범위에 기재된 본 발명의 기술적 사상을 벗어나지 않는 범위 내에서 다양한 수정 및 변형이 가능하다는 것은 당 기술분야의 통상의 지식을 가진 자에게는 자명할 것이다.
본 발명은 모낭조직배양액을 포함하는 탈모의 예방 또는 치료용 조성물 및 이의 다양한 용도에 관한 것이다.
Claims (25)
- 제1용기에 ROCK 저해제 및 기저막단백질을 첨가한 후, 모낭세포를 상기 제1용기에 부착하는 (a)단계;상기 제1용기에 배지를 처리한 후 배양하는 (b)단계;상기 모낭세포를 제2용기에 부착하여 계대하는 (c)단계; 및상기 제2용기에 상기 배지를 첨가한 후 배양하는 (d)단계;에 의해 배양된 모낭세포 배양액을 포함하는, 탈모의 예방 또는 치료용 약학적 조성물.
- 제1항에 있어서,상기 모낭세포는 모낭조직에서 유래한 세포인, 조성물.
- 제2항에 있어서,상기 모낭조직은 모간(hair shaft)이 부착된 것인, 조성물.
- 제1항에 있어서,상기 모낭세포를 제거하는 (e)단계;를 더 포함하는, 조성물.
- 제1항에 있어서,상기 ROCK 저해제는 Y-27632, HA-1077, Y-39983 및 Wf-536으로 이루어진 군에서 선택된 1종 이상인, 조성물.
- 제1항에 있어서,상기 기저막단백질은 라미닌인, 조성물.
- 제1항에 있어서,표피 성장인자(Epidermal growth factor; EGF) 및 섬유아세포 성장인자(Fibroblast growth factor; FGF)를 더 포함하는, 조성물.
- 제7항에 있어서,상기 FGF는 FGF1, FGF2, FGF3, FGF4, FGF5, FGF6, FGF7, FGF9, FGF10, FGF11, FGF12, FGF12B, FGF14, FGF16, FGF17, FGF19, FGF20, FGF21 및 FGF23으로 이루어진 군에서 선택된 1종 이상인, 조성물.
- 제8항에 있어서,상기 EGF와 상기 FGF4의 농도비는 1:10 내지 10:1인, 조성물.
- 제9항에 있어서,상기 EGF의 농도는 10ng/ml 내지 500ng/ml이고, 상기 FGF4의 농도는 10ng/ml 내지 500ng/ml인, 조성물.
- 제1항에 있어서,상기 배지는 돌배코 변형 이글배지(Dulbecco Modified Eagle Medium; DMEM), 항생제 및 태아소혈청(Fetal bovine serum; FBS)을 더 포함하는, 조성물.
- 제11항에 있어서,상기 항생제는 0.1% 내지 5%인 페니실린인, 조성물.
- 제1항에 있어서,상기 배지는 bFGF를 더 포함하는 것인, 조성물.
- 제1항에 있어서,상기 배지는 b27을 더 포함하는 것인, 조성물.
- 제1항에 있어서,상기 (c)단계에서 상기 모낭세포를 세포외 매트릭스로 코팅한 뒤 제2용기에 부착하는, 조성물.
- 제15항에 있어서,상기 세포외 매트릭스는 매트리겔(Matrigel), NuFF 배양보조세포(feeder cell) 및 겔트렉스(Geltrax)로 이루어진 군에서 선택된 1이상인 것인, 조성물.
- 제1항에 있어서,상기 (b) 및 (d) 단계에서 배양하는 기간은 20 내지 40일인, 조성물.
- 제1용기에 ROCK 저해제 및 기저막단백질을 첨가한 후, 모낭세포를 상기 제1용기에 부착하는 (a)단계;상기 제1용기에 배지를 처리한 후 배양하는 (b)단계;상기 모낭세포를 제2용기에 부착하여 계대하는 (c)단계; 및상기 제2용기에 상기 배지를 첨가한 후 배양하는 (d)단계;에 의해 배양된 모낭세포 배양액을 포함하는, 탈모의 예방 또는 완화용, 화장료 조성물.
- 제1용기에 ROCK 저해제 및 기저막단백질을 첨가한 후, 모낭세포를 상기 제1용기에 부착하는 (a)단계;상기 제1용기에 배지를 처리한 후 배양하는 (b)단계;상기 모낭세포를 제2용기에 부착하여 계대하는 (c)단계; 및상기 제2용기에 상기 배지를 첨가한 후 배양하는 (d)단계;에 의해 배양된 모낭세포 배양액을 포함하는, 상처의 예방 또는 치료용 약학적 조성물.
- 제1용기에 ROCK 저해제 및 기저막단백질을 첨가한 후, 모낭세포를 상기 제1용기에 부착하는 (a)단계;상기 제1용기에 배지를 처리한 후 배양하는 (b)단계;상기 모낭세포를 제2용기에 부착하여 계대하는 (c)단계; 및상기 제2용기에 상기 배지를 첨가한 후 배양하는 (d)단계;에 의해 배양된 모낭세포 배양액을 포함하는, 상처의 예방 또는 완화용 화장료 조성물.
- 제1용기에 ROCK 저해제 및 기저막단백질을 첨가한 후, 모낭세포를 상기 제1용기에 부착하는 (a)단계;상기 제1용기에 배지를 처리한 후 배양하는 (b)단계;상기 모낭세포를 제2용기에 부착하여 계대하는 (c)단계; 및상기 제2용기에 상기 배지를 첨가한 후 배양하는 (d)단계;에 의해 배양된 모낭세포 배양액을 포함하는, 피부 개선용 약학적 조성물.
- 제1용기에 ROCK 저해제 및 기저막단백질을 첨가한 후, 모낭세포를 상기 제1용기에 부착하는 (a)단계;상기 제1용기에 배지를 처리한 후 배양하는 (b)단계;상기 모낭세포를 제2용기에 부착하여 계대하는 (c)단계; 및상기 제2용기에 상기 배지를 첨가한 후 배양하는 (d)단계;에 의해 배양된 모낭세포 배양액을 포함하는, 피부 개선용 화장료 조성물.
- 제1용기에 ROCK 저해제 및 기저막단백질을 첨가한 후, 모낭세포를 상기 제1용기에 부착하는 (a)단계;상기 제1용기에 배지를 처리한 후 배양하는 (b)단계;상기 모낭세포를 제2용기에 부착하여 계대하는 (c)단계; 및상기 제2용기에 상기 배지를 첨가한 후 배양하는 (d)단계;에 의해 배양된 모낭세포 배양액을 포함하는 조성물을 유효량으로 개체에 투여하는 단계를 포함하는, 탈모의 예방 또는 치료 방법.
- 제1용기에 ROCK 저해제 및 기저막단백질을 첨가한 후, 모낭세포를 상기 제1용기에 부착하는 (a)단계;상기 제1용기에 배지를 처리한 후 배양하는 (b)단계;상기 모낭세포를 제2용기에 부착하여 계대하는 (c)단계; 및상기 제2용기에 상기 배지를 첨가한 후 배양하는 (d)단계;에 의해 배양된 모낭세포 배양액을 포함하는 조성물을 유효량으로 개체에 투여하는 단계를 포함하는, 상처의 예방 또는 치료 방법.
- 제1용기에 ROCK 저해제 및 기저막단백질을 첨가한 후, 모낭세포를 상기 제1용기에 부착하는 (a)단계;상기 제1용기에 배지를 처리한 후 배양하는 (b)단계;상기 모낭세포를 제2용기에 부착하여 계대하는 (c)단계; 및상기 제2용기에 상기 배지를 첨가한 후 배양하는 (d)단계;에 의해 배양된 모낭세포 배양액을 포함하는 조성물을 유효량으로 개체에 투여하는 단계를 포함하는, 피부 개선 방법.
