リジンのアミノ基がグリケーションされていることの確認は、NBT法(Nitroblue tetrazolium法)により比色定量することにより行うことができる。
リジンのアミノ基にグリケーションしている糖は、単糖、二糖、三糖以上の多糖のいずれであってもよいが、単糖又は二糖であることが好ましい。
単糖としては、グルコース、フルクトース、ガラクトース、マンノース、アラビノースを含むすべての糖が挙げられる。
二糖としては、マルトース、ラクトース等のマルトース型二糖が挙げられる。
三糖としては、マルトトリオース、セルトリオース、フコシルラクトース、シアリルラクトース等が挙げられる。
また、糖修飾タンパク質は穀類由来原料であるため、特許文献1に記載された風味改善剤のように香味に意図しない影響を与えることが防止される。また、穀類由来原料はノンアルコールビールテイスト飲料に普通に用いられる原料であるので、穀類由来原料をコク味を増強するための材料、又は、酸味を低減するための材料に使用することは、製造者及び飲料の需要者にとって抵抗が少ない。
また、酸味の有無や程度も、専門パネルによる官能評価によって評価することができる。
また、ノンアルコールビールテイスト飲料のコク味を増強する観点からは、グリケーションされたリジンの含有量が9.4nmol/mL以上であることが好ましい。
また、グリケーションされたリジンの含有量が50.0nmol/mL以上であることが好ましく、80.0nmol/mL以上であることも好ましく、100nmol/mL以上であることも好ましい。
また、グリケーションされたリジンの含有量が50.0~1000nmol/mLであることが好ましく、80.0~1000nmol/mLであることも好ましく、100~1000nmol/mLであることも好ましい。
グリケーションされたリジンの含有量の定量は、NBT法により比色定量することにより行うことができる。
本発明のノンアルコールビールテイスト飲料は、麦芽を原料として使用してもよい。
また、原料中の麦芽の比率が0重量%であってもよい。
ここで「麦芽の比率」とは、麦芽、米、トウモロコシ、コウリャン、バレイショ、デンプン、麦芽以外の麦、及び糖類など、水とホップ以外の原料中に占める麦芽の重量の比率をいう。ただし、酸味料、甘味料、苦味料、調味料、香料など、微量に添加され得る成分については上記比率の計算に含めない。

プリン体濃度が上記範囲にあるノンアルコールビールテイスト飲料について、糖修飾タンパク質を添加することによってコク味を特に増強させ、酸味を低減させることができる。
ビールテイスト飲料とは、ビール風味の炭酸飲料である。

穀類由来タンパク質を構成するリジンのアミノ基がグリケーションされた、分子量35~50kDaの糖修飾タンパク質を含む剤は、ノンアルコールビールテイスト飲料のコク味増強剤及び酸味低減剤として同時に機能する剤とすることができる。

コク味増強剤及び酸味低減剤が糖修飾タンパク質を含む組成物である場合、本発明の効果を阻害しない限りにおいて、飲料に添加することのできる公知の添加剤が任意に含まれていてもよい。
このような糖修飾タンパク質は、糖修飾タンパク質中においてグリケーションされたリジンの含有量が多いので、少量の使用でノンアルコールビールテイスト飲料のコク味を増強することができるコク味増強剤、又は、酸味を低減するための酸味低減剤として使用することができる。
また、穀類由来タンパク質を構成するリジンのアミノ基がグリケーションされた分子量35~50kDaの糖修飾タンパク質を含む麦芽又は大麦から精製することによって得ることもできる。
また、マッシングにより、リジンのアミノ基がグリケーションされた分子量35~50kDaの糖修飾タンパク質を多く含むようにタンパク質を抽出するようにしてもよい。
より具体的には、アルコール度数が1.0%未満であるノンアルコールビールテイスト飲料に、穀類由来タンパク質を構成するリジンのアミノ基がグリケーションされた分子量35~50kDaの糖修飾タンパク質を添加することにより、コク味が増強された及び/又は酸味が低減されたノンアルコールビールテイスト飲料を得る方法が挙げられる。
当該方法によるノンアルコールビールテイスト飲料の製造方法は、本発明のノンアルコールビールテイスト飲料のコク味増強方法及びノンアルコールビールテイスト飲料の酸味低減方法でもある。
また、糖修飾タンパク質が有する、グリケーションされたリジンの含有量が、50.0nmol/mL以上となるようにすることも好ましく、80.0nmol/mL以上となるようにすることも好ましく、100nmol/mL以上となるようにすることも好ましい。
この場合、コク味増強剤及び酸味低減剤の添加量は、コク味増強の観点からは糖修飾タンパク質の量として5μg/mL以上となるようにすることが好ましく、酸味低減の観点からは糖修飾タンパク質の量として10μg/mL以上となるようにすることが好ましい。
北米産麦芽を適当な粒度に粉砕し仕込み槽に入れた後、温水と混ぜ合わせた。麦芽酵素に適した温度で、糖化に十分な時間保持し、麦芽の酵素の働きででんぷん質を糖分に変換させ、糖化液を製造した。その後、糖化液をろ過してホップを加え、煮沸し、熱麦汁を製造し、発酵に備えた。熱麦汁を冷却し、これにビール酵母を加えた。数日の間に酵母の働きにより、麦汁中の糖分をアルコールと炭酸ガスに分解させ若ビールを製造した。若ビールを貯酒タンクに移し、低温で数十日間貯蔵した。この間にビールを熟成させ、ビールの味と香りを調和させた。熟成終了後、ビールをろ過し瓶詰を行った。
上記の方法で北米産麦芽から醸造したビールから、下記に従い糖修飾タンパク質の精製を行った。
(1)陽イオン交換樹脂による分画
陽イオン交換樹脂SP Sepharose50mLを空きカラムに入れた。北米産麦芽から醸造したビールを樹脂に吸着させた。その後、樹脂をカラムに移し替え、20mM酢酸ナトリウム緩衝液(pH4.5)で洗浄した。次いで、20mM酢酸ナトリウム(pH4.5)+0.5M-NaClで溶出し画分を集めた。得られた画分をSDS-PAGEで評価し、40kDaの糖修飾タンパク質が含まれる画分を集め陽イオン交換樹脂結合画分とした。
(2)限外濾過(バッファー交換)
水洗浄した限外ろ過ユニット(Merck製 Amicon Ultra-15 30K)に(1)で得た陽イオン交換樹脂結合画分を添加し、3500rpmで遠心し限外ろ過し濃縮液を得た。
(3)硫安分画
(2)で得た濃縮液を、20mMリン酸バッファー(pH7.0)+2M硫酸アンモニウムをビーカーに入れ濃縮液を滴下、撹拌した。次に懸濁液を遠心(2330g、10分間、室温)した。上清を別容器に集めた。集めた溶液は限外ろ過ユニットを用いて濃縮した。濃縮液を20mM酢酸ナトリウム(pH4.5)に加え遠心(2330g、10分間、室温)し、濃縮を行い、糖修飾タンパク質精製品(Bradford定量(ウシ血清アルブミン(BSA)換算)、20.4mg/mL、2.21mL)を得た。得られた糖修飾タンパク質精製品の純度はSDS-PAGEで確認した。
(1)ゲル片の調製
精製した糖修飾タンパク質溶液(濃度1mg/mL、20mM酢酸ナトリウムバッファー(pH4.5))10μLをSDS-PAGEで泳動しCBB(クマシーブリリアントブルー)で染色後、40kDa付近の糖修飾タンパク質のゲルを切り出した。
切り出したゲル片を細切し、ジチオスレイトールによる還元(56℃、1時間)、ヨードアセトアミドによるカルバミドメチル化(遮光下、室温、45分間)を行った。次いで0.01%ProteaseMax含有10ng/μLキモトリプシン溶液(5mM塩化カルシウム、50mM炭酸水素アンモニウム溶液)15μL、5mM塩化カルシウム、50mM炭酸水素アンモニウム溶液15μLを添加し一晩インキュベートした後、酵素消化液を回収した。回収した溶液を減圧乾固し、0.1%ギ酸溶液に再溶解した。
LC-MS/MSの測定は下記の条件で行った。
使用装置:ダイレクトフローnanoLCシステムEasy-nLC 1000TM (Thermo Scientific)
トラップカラム:Acclaim PepMap(登録商標)(Thermo Scientific)
分析カラム:NANO HPLC CAPILLARY COLUMN (日京テクノス(株))
液体クロマトグラフ質量分析計:Q Exactive Plus (Thermo Scientific)
移動相:A液:0.1%ギ酸/水、B液:0.1%ギ酸/アセトニトリル
流速:300nL/min
グラジエント:0-40%B/0-30min、40-60%B/30-35min、60-90%B/35-37min、90%B/37-45min
注入量:10μL
イオン化モード:ESI Positive
測定範囲:MS1(m/z 350-1750)
Data Dependent Scanモード
タンパク質同定および修飾検索は下記の条件で行った。
検索ソフト:Proteome Discoverer2.2.0.388(ThermoFisher製)
生物種:大麦(Hordeum vulgare)、ホップ(Humulus)、酵母(Saccharomyces cerevisiae)
検索条件:消化酵素:Chymotrypsin
修飾(Static):Carbamidomethyl (Cysteine)
修飾(Dynamic):Oxidation(Methionine), Hex(K, R, Protein N-term)、 Hex(2)(K, R, Protein N-term)、Hex(3)(K, R, Protein N-term)、Acetyl(Protein N-term)
プリカーサーイオン質量誤差範囲:Monoisotopic、±10ppm
プロダクトイオン質量誤差範囲:±0.02Da
最大ミスクリベージ数:5
コンフィデンスレベル(Percolator):High(確からしさ3段階のうち最も確率が高いレベル)
データベース:SwissProt
精製した糖修飾タンパク質の糖修飾度はFructosamine Assay kit(abcam社製品、ab228558)を用いてNBT法にて比色定量した。
ビール由来精製糖修飾タンパク質の糖修飾度を求めた。
各タンパク質につき試験数はn=2として、平均値を採用した。
糖修飾タンパク質1μgあたりのグリケーションされたリジンの量は、0.94nmol/μg(糖修飾タンパク質)であった。
市販ノンアルコールビールテイスト飲料A,Bの飲料中の糖修飾タンパク質の濃度はLabChipTM GXII装置(パーキンエルマー製)でHT Protein Expressチップを用いて、標準プロトコールに従い実施した(n=2)。その結果、市販ノンアルコールビールテイスト飲料Aおよび市販ノンアルコールビールテイスト飲料B中の糖修飾タンパク質の濃度は検出できなかった。
市販ノンアルコールビールテイスト飲料Aに、ビール由来精製糖修飾タンパク質を添加し、酸味及びコク味の官能評価を行った。
市販ノンアルコールビールテイスト飲料Aのアルコール度数は0.00%である。
市販ノンアルコールビールテイスト飲料Aのプリン体濃度は5.0mg未満/100mLである。総プリン体濃度は、日本食品分析センターで分解法にて実施した(以下同様)。
官能評価は、糖修飾タンパク質の添加をしていない市販ノンアルコールビールテイスト飲料Aの酸味を基準点の3.0点、コク味を基準点の0.5点とし、専門パネル5名により0.1点刻みでスコア化した。
糖修飾タンパク質の添加濃度を変化させて評価を行い、その結果を表1に示した。
酸味及びコク味は以下の基準とした。
0点:全く感じられない
1点:やや感じられる
2点:明確に感じる
3点:非常に強く感じる
表1には、ノンアルコールビールテイスト飲料における、糖修飾タンパク質が有するグリケーションされたリジンの含有量(nmol/mL)を示している。
市販ノンアルコールビールテイスト飲料Aに、エタノールを加え、アルコール度数が0.999%となるようにした。さらにビール由来精製糖修飾タンパク質を添加し、酸味及びコク味の官能評価を行った。
この場合もアルコール度数は1.0%未満であるので、飲料としてはノンアルコールビールテイスト飲料である。
官能評価は、市販ノンアルコールビールテイスト飲料Aにビール由来糖修飾タンパク質を添加した場合の評価と同様にした。
表2には、ノンアルコールビールテイスト飲料における、糖修飾タンパク質が有するグリケーションされたリジンの含有量(nmol/mL)を示している。
市販ノンアルコールビールテイスト飲料Bに、ビール由来精製糖修飾タンパク質を添加し、酸味及びコク味の官能評価を行った。
市販ノンアルコールビールテイスト飲料Bのアルコール度数は0.00%である。
市販ノンアルコールビールテイスト飲料Bのプリン体濃度は5.0mg未満/100mLである。
官能評価は、糖修飾タンパク質の添加をしていない市販ノンアルコールビールテイスト飲料Aの酸味を基準点の3.0点、コク味を基準点の0.5点とし、専門パネル5名により0.1点刻みでスコア化した。
糖修飾タンパク質の添加濃度を変化させて評価を行い、その結果を表3に示した。
酸味及びコク味は以下の基準とした。
0点:全く感じられない
1点:やや感じられる
2点:明確に感じる
3点:非常に強く感じる
4点:市販ノンアルコールビールテイスト飲料Aよりも強く感じる
表3には、ノンアルコールビールテイスト飲料における、糖修飾タンパク質が有するグリケーションされたリジンの含有量(nmol/mL)を示している。
Claims (10)
- アルコール度数が1.0%未満であるノンアルコールビールテイスト飲料であって、
穀類由来タンパク質を構成するリジンのアミノ基がグリケーションされた、分子量35~50kDaの糖修飾タンパク質を含む、ノンアルコールビールテイスト飲料。 - 前記糖修飾タンパク質が有する、グリケーションされたリジンの含有量が、4.7nmol/mL以上である請求項1に記載のノンアルコールビールテイスト飲料。
- 前記糖修飾タンパク質が有する、グリケーションされたリジンの含有量が、9.4nmol/mL以上である請求項1又は2に記載のノンアルコールビールテイスト飲料。
- 前記グリケーションされたリジンのアミノ基にグリケーションしている糖が、単糖又は二糖である請求項1~3のいずれか一項に記載のノンアルコールビールテイスト飲料。
- 前記穀類が、大麦、小麦、トウモロコシ、イネ及び大豆からなる群より選択される少なくとも1種である請求項1~4のいずれか一項に記載のノンアルコールビールテイスト飲料。
- 麦汁を含まない請求項1~5のいずれか一項に記載のノンアルコールビールテイスト飲料。
- 穀類由来タンパク質を構成するリジンのアミノ基がグリケーションされた、分子量35~50kDaの糖修飾タンパク質を含む、アルコール度数が1.0%未満であるノンアルコールビールテイスト飲料のコク味増強剤。
- アルコール度数が1.0%未満であるノンアルコールビールテイスト飲料に、穀類由来タンパク質を構成するリジンのアミノ基がグリケーションされた、分子量35~50kDaの糖修飾タンパク質を添加することにより、ノンアルコールビールテイスト飲料のコク味を増強する、ノンアルコールビールテイスト飲料のコク味増強方法。
- 穀類由来タンパク質を構成するリジンのアミノ基がグリケーションされた、分子量35~50kDaの糖修飾タンパク質を含む、アルコール度数が1.0%未満であるノンアルコールビールテイスト飲料の酸味低減剤。
- アルコール度数が1.0%未満であるノンアルコールビールテイスト飲料に、穀類由来タンパク質を構成するリジンのアミノ基がグリケーションされた、分子量35~50kDaの糖修飾タンパク質を添加することにより、ノンアルコールビールテイスト飲料の酸味を低減する、ノンアルコールビールテイスト飲料の酸味低減方法。
