Abstract
人為的な遺伝子導入を行うことなく、体細胞から神経様細胞を製造する方法、当該製造方法から得られる神経様細胞、又は当該製造方法のために使用することができる化学物質の組み合わせを含む組成物を提供することを主な課題とする。 本発明として、例えば、TGF-β阻害剤及びBMP阻害剤の2種、並びにcAMP誘導剤、GSK3阻害剤、Erk阻害剤、及びp53阻害剤からなる4種群から選択されるいずれか3種以上の存在下で体細胞を培養する工程を含み、当該TGF-β阻害剤が選択的ALK5阻害剤であることを特徴とする神経様細胞の製造方法や、かかる製造方法により得られる神経様細胞を挙げることができる。 本発明により得られた神経様細胞は、再生医療などにおいて有用である。
Description
本発明は、再生医療、ないし体細胞からのダイレクトリプログラミング(Direct Reprogramming)の技術分野に属する。本発明は、その技術分野において、主として低分子化合物により体細胞から神経様細胞を直接製造する方法、及びかかる製造方法によって製造される低分子化合物誘導性神経様細胞(CiNCs:Chemical compound-induced neuronal cells)に関するものである。本発明はさらに、当該神経様細胞、及び当該神経様細胞を製造する方法のために使用することができる組成物に関するものである。
近年の細胞関連研究の発展、特に多能性細胞に関する研究の発展により、治療用細胞を個体への移植に利用可能な品質及び量において入手することが可能になりつつある。幾つかの疾患については、治療に有効な細胞を患者に移植する試みが開始されている。
間葉系の細胞は、筋肉、骨、軟骨、骨髄、脂肪及び結合組織等の生体の各種器官を形成しており、再生医療の材料として有望である。間葉系幹細胞(mesenchymal stem cell:MSC)は、骨髄、脂肪組織、血液、胎盤及び臍帯等の組織に存在する未分化細胞である。間葉系に属す細胞への分化能を有しているため、間葉系幹細胞は、それらの細胞を製造する際の出発材料として注目されている。また、間葉系幹細胞自体を骨、軟骨、心筋等の再構築に利用する再生医療も検討されている。
一方、線維芽細胞のような体細胞を他の細胞に直接転換する方法も報告されている。例えば、非特許文献1では、CHIR99021(GSK3阻害剤)、PD0325901(Erk阻害剤)、SB431542(TGF-β阻害剤)、LDN193189(BMP阻害剤)、ピフィスリン-α(p53阻害剤)、及びフォルスコリン(cAMP誘導剤)といった6つの低分子化合物と共に線維芽細胞を培養することにより神経様細胞が得られることが報告されている。特許文献1には、上記6つの低分子化合物の中、ピフィスリン-α(p53阻害剤)をドルソモルフィン(AMPK阻害剤、BMP阻害剤)に代える以外は同様にして、ヒト線維芽細胞から神経様細胞へダイレクトリプログラミングする発明が開示されている。
また、非特許文献2では、バルプロン酸(HDAC阻害剤)、CHIR99021(GSK3阻害剤)、レプソックス(TGF-β阻害剤)、フォルスコリン(cAMP誘導剤)、Y-27632(ROCK阻害剤)、SP600125(JNK阻害剤)、及びDMH1(BMP阻害剤)といった低分子化合物の組み合わせにより、ヒト肺線維芽細胞から神経様細胞へのダイレクトリプログラミングが報告されている。

遺伝子導入を行うことなく体細胞から所望の細胞への転換を直接行う方法は、治療用細胞を取得する手段として有効な選択肢となる場合がある。神経様細胞についても、非特許文献1や特許文献1に記載の発明のように、ヒト線維芽細胞から直接誘導している報告例はあるが、当該誘導に2~3週間ほど要しており、より早期に誘導できる方法が望まれている。
本発明は、人為的な遺伝子導入を行うことなく、体細胞から神経様細胞をより速やかに直接誘導する方法、即ち、体細胞から神経様細胞をより速やかに直接製造することができる新たな製造方法を提供することを主な課題とする。

本発明者らは、鋭意検討した結果、TGF-β阻害剤として選択的ALK5阻害剤を用いることによって、体細胞から神経様細胞へより速やかに直接転換できることを見出し、本発明を完成するに到った。
本発明として、例えば、下記のものを挙げることができる。
[1]TGF-β阻害剤及びBMP阻害剤の存在下で体細胞を培養する工程を含み、当該TGF-β阻害剤が選択的ALK5阻害剤であることを特徴とする、神経様細胞の製造方法。

本発明によれば、体細胞(特に線維芽細胞)から神経様細胞を、例えば非特許文献1や特許文献1に記載の製造方法より短期間で製造することができる。本発明により得られた神経様細胞は、再生医療などにおいて有用である。
以下、本発明について詳細に説明する。
1 神経様細胞の製造方法
本発明に係る、神経様細胞の製造方法(以下、「本発明製法」という。)は、TGF-β阻害剤及びBMP阻害剤の存在下で体細胞を培養する工程を含み、当該TGF-β阻害剤が選択的ALK5阻害剤であることを特徴とする。あるいは、本発明製法は、TGF-β阻害剤及びBMP阻害剤の2種、並びにcAMP誘導剤、GSK3阻害剤、Erk阻害剤、及びp53阻害剤からなる4種群から選択されるいずれか3種以上の存在下で体細胞を培養する工程を含み、当該TGF-β阻害剤が選択的ALK5阻害剤であることを特徴とする。
本発明に係る、神経様細胞の製造方法(以下、「本発明製法」という。)は、TGF-β阻害剤及びBMP阻害剤の存在下で体細胞を培養する工程を含み、当該TGF-β阻害剤が選択的ALK5阻害剤であることを特徴とする。あるいは、本発明製法は、TGF-β阻害剤及びBMP阻害剤の2種、並びにcAMP誘導剤、GSK3阻害剤、Erk阻害剤、及びp53阻害剤からなる4種群から選択されるいずれか3種以上の存在下で体細胞を培養する工程を含み、当該TGF-β阻害剤が選択的ALK5阻害剤であることを特徴とする。
本発明製法において、BMP阻害剤はALK2・3阻害剤であることが好ましい。また、本発明製法において、TGF-β阻害剤はレプソックス(CAS No.:446859-33-2)、並びに/又はBMP阻害剤がLDN193189(CAS No.:1062368-24-4)及び/若しくはドルソモルフィン(CAS No.:866405-64-3)であることがより好ましい。また、cAMP誘導剤はフォルスコリン(CAS No.:66428-89-5)、GSK3阻害剤はCHIR99021(CAS No.:252917-06-9)、Erk阻害剤がPD0325901(CAS No.:391210-10-9)、又はp53阻害剤はピフィスリン-α(CAS No.:63208-82-2)であることがより好ましい。
本発明製法においては、少なくとも上記いずれかの阻害剤や誘導剤等の組み合わせの存在下で体細胞を培養すればよく、必要に応じて、任意にさらに他の阻害剤や誘導剤等を存在させて体細胞を培養し神経様細胞を製造することができる。
上記阻害剤や誘導剤は、それぞれにおいて、1種を用いても2種以上を併用してもよい。
具体的な上記阻害剤等においては、2種類以上の阻害作用等を有するものもあり得るが、その場合、一つで複数の阻害剤等が存在しているとみなすことができる。

1.1 体細胞について
生物の細胞は、体細胞と生殖細胞とに分類できる。本発明製法には、その出発材料として任意の体細胞を使用することができる。体細胞には特に限定はなく、生体から採取された初代細胞、又は株化された細胞の何れでもよい。本発明製法では、分化の種々の段階にある体細胞、例えば、最終分化した体細胞、最終分化への途上にある体細胞、又は初期化され多能性を獲得した体細胞を使用することができる。本発明製法に使用できる体細胞としては、任意の体細胞、例えば、造血系の細胞(各種のリンパ球、マクロファージ、樹状細胞、骨髄細胞等)、臓器由来の細胞(肝細胞、脾細胞、膵細胞、腎細胞、肺細胞等)、筋組織系の細胞(骨格筋細胞、平滑筋細胞、筋芽細胞、心筋細胞等)、線維芽細胞、神経細胞、骨芽細胞、軟骨細胞、内皮細胞、間質細胞、脂肪細胞(白色脂肪細胞等)、胚性幹細胞(ES細胞)等が挙げられる。また、これらの細胞の前駆細胞、癌細胞にも本発明製法を適用できる。好ましくは、線維芽細胞を使用することができる。
生物の細胞は、体細胞と生殖細胞とに分類できる。本発明製法には、その出発材料として任意の体細胞を使用することができる。体細胞には特に限定はなく、生体から採取された初代細胞、又は株化された細胞の何れでもよい。本発明製法では、分化の種々の段階にある体細胞、例えば、最終分化した体細胞、最終分化への途上にある体細胞、又は初期化され多能性を獲得した体細胞を使用することができる。本発明製法に使用できる体細胞としては、任意の体細胞、例えば、造血系の細胞(各種のリンパ球、マクロファージ、樹状細胞、骨髄細胞等)、臓器由来の細胞(肝細胞、脾細胞、膵細胞、腎細胞、肺細胞等)、筋組織系の細胞(骨格筋細胞、平滑筋細胞、筋芽細胞、心筋細胞等)、線維芽細胞、神経細胞、骨芽細胞、軟骨細胞、内皮細胞、間質細胞、脂肪細胞(白色脂肪細胞等)、胚性幹細胞(ES細胞)等が挙げられる。また、これらの細胞の前駆細胞、癌細胞にも本発明製法を適用できる。好ましくは、線維芽細胞を使用することができる。
上記の体細胞の供給源としては、ヒト、ヒト以外の哺乳動物、及び哺乳動物以外の動物(鳥類、爬虫類、両生類、魚類等)が例示されるが、これらに限定されるものではない。体細胞の供給源としては、ヒト、及びヒト以外の哺乳動物が好ましく、ヒトが特に好ましい。ヒトへの投与を目的として本発明製法により神経様細胞を製造する場合、好ましくは、レシピエントと組織適合性抗原のタイプが一致又は類似したドナーより採取された体細胞を使用することができる。レシピエント自身より採取された体細胞を本発明製法による神経様細胞の製造に供してもよい。
1.2 本発明に係る阻害剤等について
1.2.1 TGF-β阻害剤(選択的ALK5阻害剤)
TGF-β(transforming growth factor-β、形質転換増殖因子β)には、TGF-β1、TGF-β2、TGF-β3の3種類が存在し、ほぼ全ての細胞から生産されている。TGF-βは、上皮細胞をはじめ、多くの細胞の増殖を抑制するなど細胞増殖、形質転換、分化、発生、アポトーシスの制御等の多種多様な細胞機能に関与している。
1.2.1 TGF-β阻害剤(選択的ALK5阻害剤)
TGF-β(transforming growth factor-β、形質転換増殖因子β)には、TGF-β1、TGF-β2、TGF-β3の3種類が存在し、ほぼ全ての細胞から生産されている。TGF-βは、上皮細胞をはじめ、多くの細胞の増殖を抑制するなど細胞増殖、形質転換、分化、発生、アポトーシスの制御等の多種多様な細胞機能に関与している。
上記3種類のTGF-βにおいて、ALK5は、TGF-β1受容体とも称される。ALK5は、TGFβに結合した際にII型TGFβ受容体とヘテロ二量体複合体を形成するセリン/スレオニンキナーゼであり、TGFβシグナルを細胞表面から細胞質へと伝達する。
本発明では、TGF-β阻害剤の中、選択的ALK5阻害剤を用いることを特徴とするから、「TGF-β阻害剤の存在下」とは、「選択的ALK5阻害剤の存在下」と同義である。
「選択的ALK5阻害剤の存在下」とは、ALK5をほぼ限定的に阻害することができる培養条件下であることをいい、その手段は特に限定はなく、ALK5をほぼ限定的に阻害することができる任意の手段を利用することができる。本発明には、ALK5に限定的に直接作用してその機能を阻害する物質(例えば、抗ALK5抗体やその他の薬剤)、ALK5自体の産生を限定的に抑制する薬剤等を利用することができる。また、ALK5を限定的に阻害することができるのであれば、ALK5が関わるシグナル伝達をその上流で阻害することによってもよい。
「選択的ALK5阻害剤の存在下」とは、ALK5をほぼ限定的に阻害することができる培養条件下であることをいい、その手段は特に限定はなく、ALK5をほぼ限定的に阻害することができる任意の手段を利用することができる。本発明には、ALK5に限定的に直接作用してその機能を阻害する物質(例えば、抗ALK5抗体やその他の薬剤)、ALK5自体の産生を限定的に抑制する薬剤等を利用することができる。また、ALK5を限定的に阻害することができるのであれば、ALK5が関わるシグナル伝達をその上流で阻害することによってもよい。
従って、TGF-β阻害剤であっても、ALK5に加えALK4や7も広範囲に阻害する、例えば、SB431542(CAS No.:301836-41-9)、A83-01(CAS No.:909910-43-6)のような阻害剤については、本発明に係るTGF-β阻害剤には含まれない。
本発明で用いうる選択的ALK5阻害剤としては、その機能を発揮することができれば特に限定されないが、例えば、以下のレプソックスを挙げることができる。

上記ALKの中、ALK2は、ALKファミリーメンバーの受容体セリン/スレオニンキナーゼであり、SMADタンパク質、特にSMAD1/5/8が関与するシグナル伝達経路の上流に位置する。ALK2-Smad1経路の活性化を通じてエンドグリンにより前立腺癌細胞の運動性が低下する。ALK2遺伝子は、筋肉組織において進行性の異所性骨形成が特徴の珍しい常染色体性優性先天性疾患である進行性骨化性線維異形成症(FOP)に関与する主要遺伝子である。
ALK3は、膜貫通型セリン/スレオニンキナーゼファミリーメンバーである。ALK3遺伝子は、PTEN(phosphatase and tensin homologue deleted on chromosome 10)変異陰性カウデン病において微働感受性遺伝子として作用する。ALK3輸送は、FOPの病変形成において重要な役割を担い、またヒトT細胞分化にも関与する。
「BMP阻害剤の存在下」とは、BMPシグナル伝達経路を阻害することができる培養条件下であることをいい、その手段は特に限定はなく、BMPシグナル伝達経路を阻害することができる任意の手段を利用することができる。本発明には、BMPおよびBMP受容体に直接作用してその機能を阻害する物質(例えば、抗BMP抗体、その他の薬剤)、あるいはこれらの発現を抑制する薬剤等を利用することができる。また、BMPが関わるシグナル伝達の下流に位置するSMAD転写因子の発現およびその翻訳後修飾を阻害することによってもBMPシグナル伝達経路を阻害することができる。
本発明においては、BMP阻害剤の存在下で体細胞を培養するが、BMP阻害剤の中、ALK2・3阻害剤の存在下で体細胞を培養することが好ましい。

1.3 その他の好ましい条件
本発明においては特に限定されないが、製造工程において、成長因子及び/又はサイトカインの非存在下で体細胞を培養することが好ましい。成長因子及び/又はサイトカインの非存在下とは、成長因子及び/又はサイトカインが実質的に存在しないことを意味し、成長因子及び/又はサイトカインが全く存在しない場合だけでなく、成長因子及び/又はサイトカインが痕跡量で存在する場合を包含するものとする。成長因子及び/又はサイトカインの非存在下で体細胞を培養することの利点としては、安価に神経様細胞を製造できることが挙げられる。
成長因子とは、生体内において特定の細胞の増殖や分化を促進する内因性タンパク質の総称である。サイトカインとは、免疫系細胞から分泌されるタンパク質で、標的細胞は特定されない情報伝達を担う。サイトカインとしては、免疫や炎症に関係したものが多く、細胞の増殖、分化、細胞死、又は創傷治癒に関係するものもある。なお、成長因子にはサイトカインに含まれるものもあり、互いに排他的な概念ではない。

本発明においては特に限定されないが、製造工程において、ヒストンに関与する成分の非存在下で体細胞を培養することが好ましい。ヒストンに関与する成分の非存在下とは、ヒストンに関与する成分が実質的に存在しないことを意味し、ヒストンに関与する成分が全く存在しない場合だけでなく、ヒストンに関与する成分が痕跡量で存在する場合を包含するものとする。ヒストンに関与する成分としては、例えば、ヒストン脱アセチル化酵素阻害剤(例、バルプロン酸)等を挙げることができる。核初期化因子によるリプログラミングを促進すると言われるヒストン脱アセチル化酵素阻害剤を使用しない場合には、意図しない分化を起こしかねない多能性細胞が誘導されるリスクはより低くなる。
1.4 体細胞の培養
本発明製法における体細胞の培養は、使用する体細胞の種類に応じた培地、温度、その他の条件を選択し、上記の各種の阻害剤(及び、場合により誘導剤ないし活性化剤)の存在下において実施すればよい。培地は、公知の培地又は市販の培地から選択することができる。例えば、一般的な培地であるMEM(最少必須培地)、DMEM(ダルベッコ改変イーグル培地)、DMEM/F12、又はこれらを改変した培地に、適切な成分(血清、タンパク質、アミノ酸、糖類、ビタミン類、脂肪酸類、抗生物質等)を添加して使用することができる。
本発明製法における体細胞の培養は、使用する体細胞の種類に応じた培地、温度、その他の条件を選択し、上記の各種の阻害剤(及び、場合により誘導剤ないし活性化剤)の存在下において実施すればよい。培地は、公知の培地又は市販の培地から選択することができる。例えば、一般的な培地であるMEM(最少必須培地)、DMEM(ダルベッコ改変イーグル培地)、DMEM/F12、又はこれらを改変した培地に、適切な成分(血清、タンパク質、アミノ酸、糖類、ビタミン類、脂肪酸類、抗生物質等)を添加して使用することができる。
本発明においては、体細胞から神経様細胞を製造する。脳由来神経栄養因子(BDNF;Brain-Derived Neurotrophic Factor)、グリア細胞由来神経栄養因子(GDNF:Glial cell-Derived Neurotrophic Factor)、cAMP、アスコルビン酸、アスコルビン酸-2-リン酸等は、神経様細胞への分化の誘導に有効な物質として知られている。神経様細胞への分化の誘導に有効な物質としては、例えば、分化誘導剤として市販されているものを使用することもできる。本発明においては、上記した物質の存在下において体細胞を培養してもよい。
培養条件としては、一般的な細胞培養の条件を選択すればよい。37℃、5%CO2の条件などが例示される。培養中は適切な間隔(好ましくは1日から7日に1回、より好ましくは3日から4日に1回)で培地を交換することが好ましい。線維芽細胞を材料として本発明製法を実施する場合、37℃、5%CO2の条件では4日間から1週間で神経様細胞が出現する。使用する体細胞として培養が容易なものを選択することにより、あらかじめ細胞数を増加させた体細胞を神経様細胞に転換することも可能である。従って、スケールアップした神経様細胞の製造も容易である。
体細胞の培養には、プレート、ディッシュ、細胞培養用フラスコ、細胞培養用バッグ等の細胞培養容器を使用することができる。なお、細胞培養用バッグとしては、ガス透過性を有するものが好適である。大量の細胞を必要とする場合には、大型培養槽を使用してもよい。培養は開放系又は閉鎖系のどちらでも実施することができるが、得られた神経様細胞のヒトへの投与等を目的とする場合には、閉鎖系で培養を行うことが好ましい。
本発明製法においては、上記した各種の阻害剤等を含む培地において体細胞を培養することにより、一段階の培養によって体細胞から神経様細胞を製造することができる。

1.5 神経様細胞
上記した本発明製法により、神経様細胞を含有する細胞集団を得ることができる。本発明製法により製造される神経様細胞も本発明の範囲内である。本発明製法で製造される神経様細胞としては、特に限定されないが、神経細胞(ニューロン)、グリア細胞(アストロサイト、オリゴデンドロサイト、ミクログリア)、シュワン細胞などが例示される。上記した最終分化した細胞の他、神経様細胞に分化することが運命づけられた前駆細胞でもよい。
上記した本発明製法により、神経様細胞を含有する細胞集団を得ることができる。本発明製法により製造される神経様細胞も本発明の範囲内である。本発明製法で製造される神経様細胞としては、特に限定されないが、神経細胞(ニューロン)、グリア細胞(アストロサイト、オリゴデンドロサイト、ミクログリア)、シュワン細胞などが例示される。上記した最終分化した細胞の他、神経様細胞に分化することが運命づけられた前駆細胞でもよい。
本発明製法で製造される神経様細胞は、例えば、細胞の形態的変化により確認することができる。神経様細胞は、細胞の種類によって特徴的な形態をとることから、培養前後の細胞の形態を比較することによって神経様細胞の存在を知ることができる。また、神経様細胞に特徴的な分子、例えば、酵素、レセプター又は低分子化合物等を検出して、神経様細胞を確認することもできる。神経様細胞に特徴的な分子としては、β3-チューブリン、シナプシンI、ベシクル型グルタミン酸トランスポーター(vesicular glutamate transporter:vGULT)、微小管関連タンパク質(microtubule-associated protein:MAP)2、γ-アミノ酪酸(GABA)、チロシン水酸化酵素等が挙げられるが、これらに限定されるものではない。
上記分子の検出には、検疫的方法(抗体による検出)を利用できるが、タンパク質分子に関してはそのmRNA量の定量により検出を実施してもよい。神経様細胞に特徴的な分子を認識する抗体は、本発明の方法により得られた神経系細胞を単離及び精製する上でも有用である。
本発明製法で製造される神経様細胞は、例えば、神経系疾患の治療に有用である。神経系疾患としては、脊髄損傷、脳血管障害(脳梗塞等)、パーキンソン病、アルツハイマー病、筋萎縮性側索硬化症等が挙げられるが、これらに限定されるものではない。本発明製法で製造される神経様細胞を使用して、神経系疾患の治療のための医薬用組成物を製造することができる。
本発明製法で製造された神経様細胞を医薬用組成物とする場合には、常法により、神経様細胞を医薬的に許容される担体と混合するなどして、個体への投与に適した形態の製剤とすればよい。担体としては、例えば、生理食塩水、ブドウ糖やその他の補助薬(例えば、D-ソルビトール、D-マンニトール、塩化ナトリウム等)を加えて等張とした注射用蒸留水を挙げることができる。さらに、緩衝剤(例えば、リン酸塩緩衝液、酢酸ナトリウム緩衝液)、無痛化剤(例えば、塩化ベンザルコニウム、塩酸プロカインなど)、安定剤(例えば、ヒト血清アルブミン、ポリエチレングリコールなど)、保存剤、酸化防止剤等を配合してもよい。
本発明製法で製造される神経様細胞は、さらに神経様細胞の機能発揮や生着性向上に有効な他の細胞や成分と組み合わせた組成物とすることもできる。
本発明製法で製造される神経様細胞は、さらに神経様細胞の機能発揮や生着性向上に有効な他の細胞や成分と組み合わせた組成物とすることもできる。
さらに、本発明製法で製造される神経様細胞は、神経様細胞に作用する医薬候補化合物のスクリーニングや医薬候補化合物の安全性評価のために使用することもできる。本発明製法によれば、一度の操作で多くの神経様細胞を取得することができることから、細胞のロット差の影響を受けずに、再現性のある研究結果を得ることが可能になる。
2 組成物
本発明に係る組成物(以下、「本発明組成物」という。)は、体細胞から神経様細胞を製造するための組成物であって、TGF-β阻害剤及びBMP阻害剤を含み、当該TGF-β阻害剤が選択的ALK5阻害剤であることを特徴とする。あるいは、本発明組成物は、TGF-β阻害剤及びBMP阻害剤の2種、並びにcAMP誘導剤、GSK3阻害剤、Erk阻害剤、及びp53阻害剤からなる4種群から選択されるいずれか3種以上を含み、当該TGF-β阻害剤が選択的ALK5阻害剤であることを特徴とする。
本発明に係る組成物(以下、「本発明組成物」という。)は、体細胞から神経様細胞を製造するための組成物であって、TGF-β阻害剤及びBMP阻害剤を含み、当該TGF-β阻害剤が選択的ALK5阻害剤であることを特徴とする。あるいは、本発明組成物は、TGF-β阻害剤及びBMP阻害剤の2種、並びにcAMP誘導剤、GSK3阻害剤、Erk阻害剤、及びp53阻害剤からなる4種群から選択されるいずれか3種以上を含み、当該TGF-β阻害剤が選択的ALK5阻害剤であることを特徴とする。
本発明組成物において、BMP阻害剤はALK2・3阻害剤であることが好ましい。また、本発明組成物において、TGF-β阻害剤はレプソックス(CAS No.:446859-33-2)、並びに/又はBMP阻害剤がLDN193189(CAS No.:1062368-24-4)及び/若しくはドルソモルフィン(CAS No.:866405-64-3)であることが好ましい。また、cAMP誘導剤はフォルスコリン(CAS No.:66428-89-5)、GSK3阻害剤がCHIR99021(CAS No.:252917-06-9)、Erk阻害剤はPD0325901(CAS No.:391210-10-9)、又はp53阻害剤はピフィスリン-α(CAS No.:63208-82-2)であることがより好ましい。
本発明組成物においては、少なくとも上記いずれかの阻害剤や誘導剤等を含んでいればよく、必要に応じて、任意にさらに他の阻害剤や誘導剤等を含むことができる。
上記阻害剤や誘導剤は、それぞれにおいて、1種を用いても2種以上を併用してもよい。
具体的な上記阻害剤等においては、2種類以上の阻害作用等を有するものもあり得るが、その場合、一つで複数の阻害剤等を含んでいるとみなすことができる。
上記した阻害剤や誘導剤等の具体例や好ましい例などは、前記と同義である。
上記阻害剤や誘導剤は、それぞれにおいて、1種を用いても2種以上を併用してもよい。
具体的な上記阻害剤等においては、2種類以上の阻害作用等を有するものもあり得るが、その場合、一つで複数の阻害剤等を含んでいるとみなすことができる。
上記した阻害剤や誘導剤等の具体例や好ましい例などは、前記と同義である。
本発明組成物は、体細胞から神経様細胞を製造するための組成物として使用することができる。本発明組成物は、また、体細胞から神経様細胞を製造するための培地として使用することもできる。
体細胞から神経様細胞の製造に使用される培地としては、細胞の培養に必要な成分を混合して製造した基礎培地に、有効成分として、TGF-β阻害剤の一種である選択的ALK5阻害剤及びBMP阻害剤(好ましくはALK2・3阻害剤)、あるいはこれらに加えてcAMP誘導剤、GSK3阻害剤、Erk阻害剤、及びp53阻害剤からなる4種群から選択されるいずれか3種以上を含有させた培地を例示することができる。上記の有効成分は、神経様細胞の製造に有効な濃度で含まれていればよく、濃度は当業者が適宜決定することができる。基礎培地は、公知の培地又は市販の培地から選択することができる。例えば、一般的な培地であるMEM(最少必須培地)、DMEM(ダルベッコ改変イーグル培地)、DMEM/F12、又はこれらを改変した培地を、基礎培地として使用することができる。
培地にはさらに、本明細書中で上記した公知の培地成分、例えば、血清、タンパク質(アルブミン、トランスフェリン、成長因子等)、アミノ酸、糖類、ビタミン類、脂肪酸類、抗生物質等を添加してもよい。
培地にはさらに、本明細書中で上記した、脳由来神経栄養因子(BDNF;Brain-Derived Neurotrophic Factor)、グリア細胞由来神経栄養因子(GDNF:Glial cell-Derived Neurotrophic Factor)、cAMP、アスコルビン酸、アスコルビン酸-2-リン酸等の神経様細胞への分化の誘導に有効な物質を添加してもよい。
さらに本発明においては、例えば、TGF-β阻害剤の一種である選択的ALK5阻害剤及びBMP阻害剤(好ましくはALK2・3阻害剤)、あるいはこれらに加えてcAMP誘導剤、GSK3阻害剤、Erk阻害剤、及びp53阻害剤からなる4種群から選択されるいずれか3種以上を含有する組成物を生体に投与することによって、生体内において体細胞から神経様細胞を製造することもできる。即ち、本発明によれば、例えば、TGF-β阻害剤の一種である選択的ALK5阻害剤及びBMP阻害剤(好ましくはALK2・3阻害剤)、あるいはこれらに加えてcAMP誘導剤、GSK3阻害剤、Erk阻害剤、及びp53阻害剤からなる4種群から選択されるいずれか3種以上を含有する組成物を生体に投与することを含む、生体内において体細胞から神経様細胞を製造する方法が提供される。生体に投与する当該阻害剤等の好ましい組み合わせは、本明細書中に記載した通りである。
また、生体としては、ヒト、ヒト以外の哺乳動物、及び哺乳動物以外の動物(鳥類、爬虫類、両生類、魚類等)が例示されるが、ヒトが特に好ましい。例えば、TGF-β阻害剤の一種である選択的ALK5阻害剤及びBMP阻害剤(好ましくはALK2・3阻害剤)、あるいはこれらに加えてcAMP誘導剤、GSK3阻害剤、Erk阻害剤、及びp53阻害剤からなる4種群から選択されるいずれか3種以上を含有する組成物を生体内の特定部位に投与することによって、上記特定部位において、体細胞から神経様細胞を製造することができる。
以下、実施例により本発明を具体的に説明するが、本発明は実施例の範囲に限定されるものではない。
[実施例1]神経様細胞の製造
(1)ヒト線維芽細胞
材料としたヒト線維芽細胞はDSファーマバイオメディカル株式会社から購入した。38才のヒト皮膚に由来する線維芽細胞である。

材料としたヒト線維芽細胞はDSファーマバイオメディカル株式会社から購入した。38才のヒト皮膚に由来する線維芽細胞である。
(2)ヒト線維芽細胞からの神経様細胞への直接誘導
ヒト線維芽細胞を35mmディッシュに8×104個ずつ播種し、10%ウシ胎児血清(Fetal bovine serum;FBS)、100U/mlペニシリン、100μg/mlストレプトマイシンを添加したDMEM high glucose培地(和光純薬工業社製)で、37℃、5%CO2条件下で3日間培養した。なおDMEMは、ダルベッコ改変イーグル培地(Dulbecco’s Modified Eagle Medium)を示す。
ヒト線維芽細胞を35mmディッシュに8×104個ずつ播種し、10%ウシ胎児血清(Fetal bovine serum;FBS)、100U/mlペニシリン、100μg/mlストレプトマイシンを添加したDMEM high glucose培地(和光純薬工業社製)で、37℃、5%CO2条件下で3日間培養した。なおDMEMは、ダルベッコ改変イーグル培地(Dulbecco’s Modified Eagle Medium)を示す。
上記のヒト線維芽細胞のディッシュの培地を、5%ウシ胎児血清(FBS、Hyclone社製)、ITS-X(Cat#:51500056、Gibco社製)、非必須アミノ酸(NEAA:Non-essential amino acids;Cat#:11140050、Gibco社製)、ニコチンアミド(Cat#:72340-100G、Sigma-Aldrich社製;終濃度5mmol/L)、Dexamethasone(Cat#:047-18863、和光純薬工業社製;終濃度0.1μmol/L)、100U/mlペニシリン、100μg/mlストレプトマイシン、及び下記のサイトカインや低分子化合物を添加したIMDM(Cat#:12440053、Gibco社製)に培地を交換した。その後、3日毎に同組成の培地へ培地交換を行いながら、37℃、5% CO2条件下で培養した。
上記のヒト線維芽細胞のディッシュの培地を、0.5% N-2サプリメント(Thermo Fisher Scientific社製)、1% B-27サプリメント(Thermo Fisher Scientific社製)、50% DMEM/F12、50% Neurobasal Medium(Thermo Fisher Scientific社製)、および表1、2に従って添加した低分子化合物を含む培地に交換した。その後、3日毎に同組成の培地へ培地交換を行いながら、37℃、5%CO2条件下で培養した。表1及び2に記載の化合物の詳細は、本明細書中上記の通りである。
(3)結果
上記(2)に従って培養した結果を図1~5に示す。
図1に示す通り、TGF-β阻害剤として選択的ALK5阻害剤を用いて培養した実施例1及び2は、6日目(Day6)にして、TGF-β阻害剤として広範囲のALK4、5、7阻害剤を用いて培養した比較例1~3における11日目(Day11)に匹敵する同等な神経様細胞の誘導効果を示した。
上記(2)に従って培養した結果を図1~5に示す。
図1に示す通り、TGF-β阻害剤として選択的ALK5阻害剤を用いて培養した実施例1及び2は、6日目(Day6)にして、TGF-β阻害剤として広範囲のALK4、5、7阻害剤を用いて培養した比較例1~3における11日目(Day11)に匹敵する同等な神経様細胞の誘導効果を示した。
また、図2、3に示す通り、TGF-β阻害剤として選択的ALK5阻害剤を用いて培養した場合、実施例3の結果のように、ドルソモルフィンによるシグナル伝達の阻害が必要ないことが分かった。更に、図5に示す通り、p53阻害剤の非存在下で培養した比較例4に比べ、p53阻害剤の存在下で培養した比較例2の方が高い誘導効率を示したが、TGF-β阻害剤として選択的ALK5阻害剤を用いれば、実施例8のようにp53阻害剤が非存在下でも、高い誘導効率を示した。
次に神経細胞マーカーによる抗体染色を行った。比較例1については培養開始14日後に、実施例1については培養開始7日後に、細胞を2%パラホルムアルデヒドで固定した後、免疫染色を行った。染色には抗Neuronal Clas III β-Tubulin(TUJ1)抗体(Mouse anti-Tuj1:MMS-435P;旧コバンス社(現フナコシ);1,000倍希釈)を使用した。その結果を図4に示す。なお、図4はグレースケールであるため緑色や青色などは表示されないが、実際のオリジナル写真では、青がDAPIによる核染色、緑がβ3-チューブリンの染色を示す。
図4から明らかな通り、TGF-β阻害剤として選択的ALK5阻害剤を用いて培養した実施例1における培養開始7日後と、TGF-β阻害剤として広範囲のALK4、5、7阻害剤を用いて培養した比較例1における培養開始14日後とにおいて、ほぼ同等の神経様細胞が得られた。
図4から明らかな通り、TGF-β阻害剤として選択的ALK5阻害剤を用いて培養した実施例1における培養開始7日後と、TGF-β阻害剤として広範囲のALK4、5、7阻害剤を用いて培養した比較例1における培養開始14日後とにおいて、ほぼ同等の神経様細胞が得られた。
Claims (15)
- TGF-β阻害剤及びBMP阻害剤の存在下で体細胞を培養する工程を含み、当該TGF-β阻害剤が選択的ALK5阻害剤であることを特徴とする、神経様細胞の製造方法。
- TGF-β阻害剤及びBMP阻害剤の2種、並びにcAMP誘導剤、GSK3阻害剤、Erk阻害剤、及びp53阻害剤からなる4種群から選択されるいずれか3種以上の存在下で体細胞を培養する工程を含み、当該TGF-β阻害剤が選択的ALK5阻害剤であることを特徴とする、神経様細胞の製造方法。
- 前記BMP阻害剤がALK2・3阻害剤である、請求項1又は2に記載の神経様細胞の製造方法。
- 前記TGF-β阻害剤がレプソックス(CAS No.:446859-33-2)、並びに/又は前記BMP阻害剤がLDN193189(CAS No.:1062368-24-4)及び/若しくはドルソモルフィン(CAS No.:866405-64-3)である、請求項1~3のいずれか一項に記載の神経様細胞の製造方法。
- 前記cAMP誘導剤がフォルスコリン(CAS No.:66428-89-5)、前記GSK3阻害剤がCHIR99021(CAS No.:252917-06-9)、前記Erk阻害剤がPD0325901(CAS No.:391210-10-9)、又は前記p53阻害剤がピフィスリン-α(CAS No.:63208-82-2)である、請求項1~4のいずれか一項に記載の神経様細胞の製造方法。
- 前記工程が、成長因子及び/又はサイトカインの非存在下で体細胞を培養する工程である、請求項1~5のいずれか一項に記載の神経様細胞の製造方法。
- 前記工程が、ヒストンに関与する成分の非存在下で体細胞を培養する工程である、請求項1~6のいずれか一項に記載の神経様細胞の製造方法。
- 前記体細胞が線維芽細胞である、請求項1~7のいずれか一項に記載の神経様細胞の製造方法。
- 請求項1~8のいずれか一項に記載の神経様細胞の製造方法から製造される、神経様細胞。
- TGF-β阻害剤及びBMP阻害剤を含み、当該TGF-β阻害剤が選択的ALK5阻害剤であることを特徴とする、体細胞から神経様細胞を製造するための組成物。
- TGF-β阻害剤及びBMP阻害剤の2種、並びにcAMP誘導剤、GSK3阻害剤、Erk阻害剤、及びp53阻害剤からなる4種群から選択されるいずれか3種以上を含み、当該TGF-β阻害剤が選択的ALK5阻害剤であることを特徴とする、体細胞から神経様細胞を製造するための組成物。
- 前記BMP阻害剤がALK2・3阻害剤である、請求項10又は11に記載の組成物。
- 前記TGF-β阻害剤がレプソックス(CAS No.:446859-33-2)、並びに/又は前記BMP阻害剤がLDN193189(CAS No.:1062368-24-4)及び/若しくはドルソモルフィン(CAS No.:866405-64-3)である、請求項10~12のいずれか一項に記載の組成物。
