Abstract
本発明は、てんかん等の治療、予防及び/又は診断に有効な、式[I]: [式中、各記号は、明細書に記載のとおりである。]で表される新規ピリミジン化合物及びその塩並びにその医薬用途を提供する。
Description
本発明は、ピリミジン化合物及びその塩に関する。本発明はまた、ピリミジン化合物又はその塩を有効成分とする、てんかん等の疾患の診断、予防及び/又は治療に有用な医薬にも関する。
てんかん有病率は人口の約1%で、日本では約100万人の患者が存在する一般的な神経疾患であり生涯罹患率は3~4%とされ、年間数万人がてんかんを発症していると推測される。当該患者の約7割は、既存の抗てんかん薬の服用で発作を抑制することができ、問題なく日常生活を送ることができるが、残りの3割のてんかん患者では発作を十分にコントロールすることができず、いつ起こるかわからない発作に不安を抱えている。既存の抗てんかん薬は、神経細胞の興奮-抑制の不均衡を正すことを目的としているが、至適用量以上の用量では平衡が妨害され、運動障害や、てんかん発作が誘導されうる。
特許文献1は、てんかんの治療に有用な化合物としてジヒドロウラシル化合物を開示している。しかしながら、ジヒドロウラシルにピリミジンが結合した構造を有する化合物は、特許文献1には開示されていない。
本発明の目的の一つは、てんかん等の治療、予防及び/又は診断に有効な新規ピリミジン化合物及びその塩並びにその医薬用途を提供することである。
また、本発明の別の目的としては、てんかん発作を完全に抑制する用量においても、興奮-抑制の平衡を保ち、既存の抗てんかん薬に比べて幅広い治療スペクトラムを有する薬剤を提供することである。

本発明者らは、上記課題を解決すべく鋭意研究を重ねた結果、既存の抗てんかん薬に比べて幅広い治療スペクトラムを有する新規ピリミジン化合物の合成に成功した。本発明は、斯かる知見に基づき完成されたものである。
すなわち、本発明は、以下の態様を含む。

本発明の化合物及びその塩は、てんかん等の治療、予防及び/又は診断において優れた効果を有する。また、本発明の化合物及びその塩は、医薬品の有効成分として使用するのに優れた特徴を有しており、例えば、副作用が少なく、忍容性、安定性(保存安定性、代謝安定性など)などの点において優れた特徴も有している。更に、本発明の化合物及びその塩は、既存の抗てんかん薬に比べて幅広い治療スペクトラムを有する。
本明細書中で用いる語句及び用語について、以下に詳述する。
本明細書中、「C1-6アルキル」の例としては、炭素数1~6の直鎖又は分枝鎖状アルキル基を含み、その具体例としては、メチル、エチル、プロピル、イソプロピル、ブチル、イソブチル、sec-ブチル、tert-ブチル、ペンチル、イソペンチル、ネオペンチル、ヘキシル、イソヘキシル、3-メチルペンチル等を例示できる。

本明細書において、本発明の化合物、方法及び組成物の異なる特徴に関する好ましい態様及び選択肢の提示は、これらが組合せ可能であって矛盾のない限り、当該異なる特徴についての好ましい態様及び選択肢の組合せの提示も含む。
以下、本発明の化合物[I]の製造法について説明する。本発明の化合物[I]は、例えば下記に示す製造方法に基づき製造することができる。下記に示す製造方法は例示であって、化合物[I]の製造方法はこれらに限定されない。
以下の反応式において、アルキル化反応、加水分解反応、アミノ化反応、エステル化反応、アミド化反応、エーテル化反応、求核置換反応、付加反応、酸化反応、還元反応等を行う場合、これらの反応は、自体公知の方法に従って行われる。このような方法としては、例えば、実験化学講座(第5版、日本化学会編、丸善株式会社)、オーガニック・ファンクショナル・グループ・プレパレーションズ(ORGANIC FUNCTIONAL GROUP PREPARATIONS)第2版、アカデミックプレス社(ACADEMIC PRESS, INC.)1989年刊;コンプリヘンシブ・オーガニック・トランスフォーメーションズ(Comprehensive Organic Transformations)VCH Publishers Inc.,1989年刊、ウッツ(P.G.M.Wuts)及びグリーン(T.W.Greene)著、「Greene's Protective Groups in Organic Synthesis」(第4版、2006年)等に記載の方法等が挙げられる。
上記反応式-1で示される反応により、本発明の化合物[I]に含まれる化合物[Ia]を製造することができる。具体的には、化合物[II]のアミノ基に化合物[III](アクリル酸)を1,4-付加し、次いで、生成物のアミノ基を、尿素を用いてウレア誘導体に変換し、環化(分子内アミド化)することによって化合物[Ia]を製造することができる。
上記反応に使用する「溶媒」としては、反応に不活性な溶媒であればよく、例えば、水、エーテル(例えば、ジオキサン、テトラヒドロフラン、ジエチルエーテル、1,2-ジメトキシエタン、ジエチレングリコールジメチルエーテル、エチレングリコールジメチルエーテル)、ハロ炭化水素(例えば、塩化メチレン、クロロホルム、1,2-ジクロロエタン、四塩化炭素)、芳香族炭化水素(例えば、ベンゼン、トルエン、キシレン)、C1-6アルコール(例えば、メタノール、エタノール、イソプロパノール)、極性溶剤(例えば、N,N-ジメチルホルムアミド(DMF)、N-メチルピロリジン(NMP)、ジメチルスルホキシド(DMSO)、ヘキサメチルリン酸トリアミド、アセトニトリル)が挙げられる。これらの溶媒は、1種単独で又は2種以上混合して使用される。
上記反応に使用する「酸」としては、例えば、無機酸、有機酸等が挙げられる。「無機酸」の例としては、塩酸、硫酸、硝酸、臭化水素酸、リン酸等が挙げられる。「有機酸」の例としては、酢酸、トリフルオロ酢酸、シュウ酸、フタル酸、フマル酸、酒石酸、マレイン酸、クエン酸、コハク酸、メタンスルホン酸、p-トルエンスルホン酸、10-カンファースルホン酸等が挙げられる。
他の反応条件(反応温度、反応時間等)は、自体公知の1,4-付加反応、アミド化反応に基づいて適宜決定することができる。
上記反応式-2で示される反応により、本発明の化合物[I]に含まれる化合物[Ib]を製造することができる。具体的には、化合物[V]の脱離基Xを脱離させ、化合物[IV]と置換することによって化合物[Ib]を製造することができる。
上記反応に使用する「脱離基」としては、例えば、ハロ、C1-18アルカンスルホニル基、C1-6アルカンスルホニルオキシ、アリールスルホニルオキシ、アラルキルスルホニルオキシ、トリハロメタンスルホニルオキシ、スルホニオ、トルエンスルホキシ等を例示できる。本反応における好ましい脱離基としては、ハロが挙げられる。
上記「ハロ」の例としては、フッ素、塩素、臭素又はヨウ素が挙げられる。
「C1-18アルカンスルホニル」の例としては、炭素数1~18の直鎖又は分枝鎖状アルカンスルホニル基を含み、その具体例としては、メタンスルホニル、プロピルスルホニル、イソプロパンスルホニル、ブタンスルホニル、シクロヘキサンスルホニル、ドデカンスルホニル、オクタデカンスルホニル基等を例示できる。
「C1-6アルカンスルホニルオキシ」の例としては、炭素数1~6の直鎖又は分枝鎖状アルカンスルホニルオキシを含み、その具体例としては、メタンスルホニルオキシ、エタンスルホニルオキシ、n-プロパンスルホニルオキシ、イソプロパンスルホニルオキシ、n-ブタンスルホニルオキシ、tert-ブタンスルホニルオキシ、n-ペンタンスルホニルオキシ、n-ヘキサンスルホニルオキシ基等を例示できる。
「アリールスルホニルオキシ」の例としては、フェニル環上に置換基として炭素数1~6の直鎖又は分枝鎖状アルキル、炭素数1~6の直鎖又は分枝鎖状アルコキシ、ニトロ及びハロなる群より選ばれた基を1~3個有することのあるフェニルスルホニルオキシ、ナフチルスルホニルオキシ基等を含む。上記「置換基を有することのあるフェニルスルホニルオキシ」の具体例としては、フェニルスルホニルオキシ、4-メチルフェニルスルホニルオキシ、2-メチルフェニルスルホニルオキシ、4-ニトロフェニルスルホニルオキシ、4-メトキシフェニルスルホニルオキシ、2-ニトロフェニルスルホニルオキシ、3-クロロフェニルスルホニルオキシ基等を例示できる。上記「ナフチルスルホニルオキシ」の具体例としては、α-ナフチルスルホニルオキシ、β-ナフチルスルホニルオキシ基等を例示できる。
「アラルキルスルホニルオキシ」の例としては、例えば、フェニル環上に置換基として炭素数1~6の直鎖又は分枝鎖状アルキル基、炭素数1~6の直鎖又は分枝鎖状アルコキシ基、ニトロ及びハロなる群より選ばれた基を1~3個有することのあるフェニルで置換された炭素数1~6の直鎖又は分枝鎖状アルカンスルホニルオキシ、ナフチルで置換された炭素数1~6の直鎖又は分枝鎖状アルカンスルホニルオキシ等を含む。上記「フェニルで置換されたアルカンスルホニルオキシ」の具体例としては、ベンジルスルホニルオキシ、2-フェニルエチルスルホニルオキシ、4-フェニルブチルスルホニルオキシ、4-メチルベンジルスルホニルオキシ、2-メチルベンジルスルホニルオキシ、4-ニトロベンジルスルホニルオキシ、4-メトキシベンジルスルホニルオキシ、3-クロロベンジルスルホニルオキシ基等を例示できる。上記「ナフチルで置換されたアルカンスルホニルオキシ」の具体例としては、α-ナフチルメチルスルホニルオキシ、β-ナフチルメチルスルホニルオキシ基等を例示できる。
「ペルハロアルカンスルホニルオキシ」としては、具体的にはトリフルオロメタンスルホニルオキシ基を例示できる。
「スルホニオ」としては、具体的には、ジメチルスルホニオ、ジエチルスルホニオ、ジプロピルスルホニオ、ジ(2-シアノエチル)スルホニオ、ジ(2-ニトロエチル)スルホニオ、ジ-(アミノエチル)スルホニオ、ジ(2-メチルアミノエチル)スルホニオ、ジ-(2-ジメチルアミノエチル)スルホニオ、ジ-(2-ヒドロキシエチル)スルホニオ、ジ-(3-ヒドロキシプロピル)スルホニオ、ジ-(2-メトキシエチル)スルホニオ、ジ-(2-カルバモイルエチル)スルホニオ、ジ-(2-カルバモイルエチル)スルホニオ、ジ-(2-カルボキシエチル)スルホニオ、ジ-(2-メトキシカルボニルエチル)スルホニオ又はジフェニルスルホニオ基等を例示できる。
上記反応に使用する「溶媒」としては、反応に不活性な溶媒であればよく、例えば、水、エーテル(例えば、ジオキサン、テトラヒドロフラン、ジエチルエーテル、1,2-ジメトキシエタン、ジエチレングリコールジメチルエーテル、エチレングリコールジメチルエーテル)、ハロ炭化水素(例えば、塩化メチレン、クロロホルム、1,2-ジクロロエタン、四塩化炭素)、芳香族炭化水素(例えば、ベンゼン、トルエン、キシレン)、C1-6アルコール(例えば、メタノール、エタノール、イソプロパノール)、極性溶剤(例えば、N,N-ジメチルホルムアミド(DMF)、N-メチルピロリジン(NMP)、ジメチルスルホキシド(DMSO)、ヘキサメチルリン酸トリアミド、アセトニトリル)が挙げられる。これらの溶媒は、1種単独で又は2種以上混合して使用される。
上記反応に使用する「塩基」としては、例えば、無機塩基、有機塩基等が挙げられる。「無機塩基」の例としては、アルカリ金属水酸化物(例えば、水酸化ナトリウム、水酸化カリウム)、アルカリ土類金属水酸化物(例えば、水酸化マグネシウム、水酸化カルシウム)、アルカリ金属炭酸塩(例えば、炭酸ナトリウム、炭酸カリウム)、アルカリ土類金属炭酸塩(例えば、炭酸マグネシウム、炭酸カルシウム)、アルカリ金属炭酸水素塩(例えば、炭酸水素ナトリウム、炭酸水素カリウム)等が挙げられる。「有機塩基」の例としては、トリアルキルアミン(例えば、トリメチルアミン、トリエチルアミン)、ピコリン、1,5-ジアザビシクロ[4.3.0]ノン-5-エン、1,4-ジアザビシクロ[2.2.2]オクタン及び1,8-ジアザビシクロ[5.4.0]ウンデカ-7-エン等が挙げられる。
他の反応条件(反応温度、反応時間等)は、自体公知の求核反応に基づいて適宜決定することができる。
上記反応式における各反応において、生成物は反応液のまま、又は粗生成物として次反応に用いることもできるが、常法に従って反応混合物から単離することもでき、通常の分離手段により容易に精製することもできる。通常の分離手段としては、例えば、再結晶、蒸留、クロマトグラフィーが挙げられる。
上記各工程における出発原料化合物、中間体化合物及び目的化合物並びに本発明の化合物[I]には、幾何異性体、立体異性体、光学異性体及び互変異性体が含まれる。各種異性体は一般的な光学分割法により分離できる。また、適当な光学活性な原料化合物より製造することもできる。
本発明の化合物[I]は、上記の各反応式にて示された合成方法又はそれに準ずる方法により製造することができる。
本発明の化合物[I]の製造における原料化合物は具体的製法を述べない場合、市販のものを用いてもよく、自体公知の方法又はそれに準ずる方法に従って製造したものを用いてもよい。
上記各工程における出発原料化合物及び目的化合物を適切な塩形態で使用することができる。このような塩としては、下記に本発明の化合物[I]の塩として例示されるものと同様のものが挙げられる。
各工程で得られた化合物又は市販品が遊離化合物である場合には、自体公知の方法により、目的とする塩に変換することができる。各工程で得られた化合物又は市販品が塩である場合には、自体公知の方法により、遊離体又は目的とする他の種類の塩に変換することができる。
また、本発明の化合物[I]には、その医薬的に許容される塩の形態が含まれ、酸付加塩又は置換基の種類によっては塩基との塩を形成する場合もある。かかる「酸」の例としては、塩酸、臭化水素酸、硝酸、硫酸、リン酸等の無機酸;及びメタンスルホン酸、p-トルエンスルホン酸、酢酸、クエン酸、酒石酸、マレイン酸、フマル酸、リンゴ酸、乳酸等の有機酸等が挙げられる。かかる「塩基」の例としては、水酸化ナトリウム、水酸化カリウム、水酸化カルシウム、炭酸ナトリウム、炭酸カリウム、炭酸水素ナトリウム、炭酸水素カリウム等の無機塩基;及びメチルアミン、ジエチルアミン、トリメチルアミン、トリエチルアミン、エタノールアミン、ジエタノールアミン、トリエタノールアミン、エチレンジアミン、トリス(ヒドロキシメチル)メチルアミン、ジシクロヘキシルアミン、N,N’-ジベンジルエチレンジアミン、グアニジン、ピリジン、ピコリン、コリン等の有機塩基;及びアンモニウム塩等が挙げられる。また、例えばリジン、アルギニン、アスパラギン酸、グルタミン酸等のアミノ酸と塩を形成してもよい。
本発明は、化合物[I]及びその塩の各種の水和物や溶媒和物及び結晶多形の物質をも包含する。
本発明の化合物[I]には、1つ又は複数の原子を1つ又は複数の同位体原子で置換された化合物が含まれる。同位体原子の例としては重水素(2H)、三重水素(3H)、13C、15N、18O等が挙げられる。
本発明の化合物[I]には、薬学的に許容されるプロドラッグも含まれる。プロドラッグを形成するために修飾される置換基としては、-OH、-COOH、アミノ等の反応性官能基が挙げられる。これらの官能基の修飾基は、本明細書における「置換基」から適宜選択してもよい。
本発明の化合物[I]又はその塩としては、薬学的に許容され得る共結晶又は共結晶塩であってもよい。ここで、共結晶又は共結晶塩とは、各々が異なる物理的特性(例えば、構造、融点、融解熱など)を持つ、室温で2種若しくはそれ以上の独特な固体から構成される結晶性物質を意味する。共結晶及び共結晶塩は、公知の共結晶化法を適用して製造することができる。
本発明の化合物[I]及びその塩は、てんかん(例えば、単純部分発作、複雑部分発作若しくは二次性全般化発作等の部分発作;間代発作、強直発作、強直間代発作、熱性発作、欠神発作(小発作)、ミオクロニー発作、脱力発作、レノックス・ガストー(Lennox-Gastaut)症候群、ウエスト(West)症候群(点頭てんかん)、ドラベ(Dravet)症候群及び多剤耐性発作等からなる群から選ばれた全般発作)の治療、予防及び/又は診断において優れた効果を有する。更に、本発明の化合物[I]及びその塩は、医薬品の有効成分として使用するのに優れた特徴を有しており、例えば、副作用が少なく、忍容性、安定性(保存安定性、代謝安定性など)などの点において優れた特徴も有している。
次に、本発明の化合物[I]又はその塩を有効成分として含有する医療製剤(以下、「医薬組成物」ともいう)について説明する。
上記医療製剤は、本発明の化合物[I]又はその塩を通常の医療製剤の形態に製剤化したものであって、本発明の化合物[I]又はその塩と、薬学的に許容される担体とを用いて調製される。当該担体としては、通常使用される充填剤、増量剤、結合剤、付湿剤、崩壊剤、表面活性剤、滑沢剤等の希釈剤又は賦形剤が挙げられる。
このような医療製剤としては、治療目的に応じて種々の形態の中から選択でき、例えば錠剤、丸剤、散剤、液剤、懸濁剤、乳剤、顆粒剤、カプセル剤、坐剤、注射剤(液剤、懸濁剤等)等が挙げられる。
錠剤の形態に成形する際に用いられる担体としては、公知のものを広く使用でき、例えば、乳糖等の賦形剤;ポリビニルピロリドン等の結合剤;デンプン等の崩壊剤;ラウリル硫酸ナトリウム等の吸収促進剤;グリセリン、デンプン等の保湿剤;コロイド状ケイ酸等の吸着剤;ポリエチレングリコール等の滑沢剤等が挙げられる。
更に、錠剤は、必要に応じて通常の錠皮を施した錠剤、例えば、糖衣錠、ゼラチン被包錠、腸溶被錠、フィルムコーティング錠あるいは二重錠、多層錠とすることができる。
丸剤の形態に成形する際に用いられる担体としては、公知のものを広く使用でき、例えば、ブドウ糖等の賦形剤;アラビアゴム末等の結合剤;ラミナラン等の崩壊剤等が挙げられる。
坐剤の形態に成形する際に用いられる担体としては、公知のものを広く使用でき、例えば、カカオ脂等が挙げられる。
注射剤として調製される場合は、液剤、乳剤及び懸濁剤は殺菌され、かつ血液と等張であるのが好ましい。これらの液剤、乳剤及び懸濁剤の形態に成形する際に用いられる希釈剤としては、公知のものを広く用いられているものを使用することができ、例えば、水等が挙げられる。なお、この場合、等張性の溶液を調製するのに十分な量の食塩等を医療製剤中に含有させてもよく、また通常の溶解補助剤、緩衝剤、無痛化剤等を、更に必要に応じて着色剤、保存剤、香料、風味剤、甘味剤等や他の医薬品を含有させてもよい。
医療製剤中に含有される本発明の化合物[I]又はその塩の量は、特に限定されず広い範囲内から適宜選択することができるが、通常、医療製剤中に本発明の化合物[I]又はその塩を1~70重量%含有させるのが好ましい。
本発明に係る医療製剤の投与方法としては特に制限はなく、各種製剤形態、患者の年齢、性別、疾患の状態、その他の条件に応じた方法で投与してもよい。例えば、錠剤、丸剤、液剤、懸濁剤、乳剤、顆粒剤及びカプセル剤の場合には経口投与してもよい。また、注射剤の場合には、単独であるいはブドウ糖、アミノ酸等の通常の補液と混合して静脈内に投与したり、更には必要に応じて単独で筋肉内、皮内、皮下もしくは腹腔内に投与することができる。坐剤の場合には、直腸内に投与してもよい。
上記医療製剤の投与量は、用法、患者の年齢、性別、疾患の程度、その他の条件に応じて適宜選択すればよく、通常、1日あたり体重1 kgに対して0.01~100 mg、好ましくは0.1~50 mgを1回~数回に分けて投与してもよい。
上記投与量は、種々の条件で変動するので、上記範囲より少ない投与量で充分な場合もあるし、また上記範囲を超えた投与量が必要な場合もある。
本発明の化合物[I]又はその塩は、前述の化合物[I]が有効と考えられる疾患の種々の治療又は予防剤と併用することができる。当該併用は、同時投与、或いは別個に連続してもしくは所望の時間間隔をおいて投与してもよい。同時投与製剤は、配合剤であっても別個に製剤化されていてもよい。
本発明のある態様において、本発明の化合物[I]又はその塩及び薬学的に許容可能な担体を含む、医薬組成物が提供される。
別の態様において、本発明の化合物[I]又はその塩及び薬学的に許容可能な担体を含む、てんかん等を診断、予防及び/又は治療するための剤が提供される。
更に別の態様において、てんかん等を診断、予防及び/又は治療するための本発明の化合物[I]又はその塩が提供される。
更に別の態様において、てんかん等の診断薬、予防薬及び/又は治療薬の製造における本発明の化合物[I]又はその塩の使用が提供される。
更に別の態様において、有効量の本発明の化合物[I]又はその塩をヒトに投与することを特徴とする、てんかん等を診断、予防及び/又は治療する方法が提供される。

1-[2-(メチルチオ)ピリミジン-5-イル]ピリミジン-2,4(1H,3H)-ジオン(440 mg)をDCM(10 mL)に懸濁させ、含水77%MCPBA(1002 mg)を加え窒素雰囲気下、室温で終夜撹拌した。反応溶液にジメチルスルフィドを加えて撹拌後、減圧濃縮、残渣をDMF(4 mL)に溶解しフェノール(0.33 mL)、K2CO3(772 mg)を加え、室温で終夜撹拌した。その後、70℃で3時間撹拌した。反応溶液に水を加えAcOEtにて抽出した。有機層を分取、水、飽和食塩水で洗浄、硫酸ナトリウムにて乾燥し、濃縮して粗生成物を得た。粗生成物を中圧分取液体クロマトグラフィー(Hexane:AcOEt=1:1→0:1)にて精製後、得られた固体をIPEにて洗浄、含水EtOHから再結晶を行い、目的化合物(270 mg)を得た。

1-[2-(メチルスルホニル)ピリミジン-5-イル]-5,6-ジヒドロピリミジン-2,4(1H,3H)-ジオン(224 mg)、K2CO3(172 mg)、チオフェノール(0.10 mL)をDMF(5 mL)に懸濁させ窒素雰囲気下室温で撹拌した後、70℃で10時間撹拌した。残渣に水を加えAcOEtにて抽出した。有機層を分取、水、飽和食塩水で洗浄、硫酸ナトリウムにて乾燥し、濃縮して粗生成物を得た。粗生成物を中圧分取液体クロマトグラフィー(Hexane:AcOEt=1:1→0:1)にて精製後、EtOHにて洗浄して目的化合物(47 mg)を得た。

[試験例1]聴原性てんかん発作モデル
この試験で用いる動物モデルは臨床予測性が高く、部分発作(二次性全般化を含む)や全般性強直間代発作の表現系である。本試験はDe Sarroらの報告(Br J Pharmacol. 1988 Feb; 93(2): 247-56. Anticonvulsant effects of some calcium entry blockers in DBA/2 mice. De Sarro GB, Meldrum BS, Nistico G.)に準じて行った。
本試験例においては、試験化合物として、以下の表3に示す実施例化合物を使用した。また、比較例化合物としては、置換位置を勘案した場合に最も近似な次の化合物(国際公開第2004/009559号公報の実施例5の化合物)を使用した。
試験化合物を5%アラビアゴム/蒸留水(w/v)に懸濁し、30 mg/kgの用量で雌雄DBA/2マウス(日本エスエルシー株式会社、3週齢、1群8例)に強制経口投与した。その1時間後にマウスを直径23 cm 高さ30 cmの透明アクリルシリンダー内に静置し、30秒の馴化後に12.6 kHz、100-110 dBの音刺激を 1分間又は強直発作が発現するまで与えた。
発作の程度を、0:発作なし、1:Wild running、2:間代性発作、3:強直性発作、4:呼吸停止とし、これらのポイントのうち最大のポイントを発作重篤度スコアとして記録した。
以下の数式により各化合物投与群の発作抑制率を算出した。
結果を表3に示す。

試験化合物を5%アラビアゴム/蒸留水(w/v)に懸濁し、30 mg/kgの用量で雄性ICRマウス(日本エスエルシー株式会社、5-6週齢、1群8例)に強制経口投与する。その1時間後に両耳介に装着した電極より、電気刺激(100 Hz、30 mA、0.2秒間)を、小動物用電撃けいれん刺激装置(UgoBasile社製)を用いて与え、誘発される後肢の強直性伸展痙攣の発現の有無を観察する。
上記試験において、溶媒投与群では全例で後肢の強直性伸展痙攣が誘発されるのに対して、試験化合物投与群では痙攣の発現が抑制されることを確認できる。

上記試験において、溶媒投与群では全例で後肢の強直性伸展痙攣が誘発されるのに対して、試験化合物投与群では痙攣の発現が抑制されることを確認できる。
[試験例3]皮下注射ペンチレンテトラゾールモデル(scPTZ)
本試験は、試験例2)と同様に化合物の抗痙攣作用を評価する試験である。この試験で用いる動物モデルは、試験例2)の表現系とは異なり、全般性の欠神発作やミオクロニー発作の表現系である。
試験化合物を5%アラビアゴム/蒸留水(w/v)に懸濁し、30 mg/kgの用量で雄性ICRマウス(日本エスエルシー株式会社、5-6週齢、1群5例)に強制経口投与する。その1時間後に、生理食塩水に溶解したペンチレンテトラゾール90 mg/kgを皮下投与し、30分間における間代性痙攣の発現の有無を観察する。
上記試験において、溶媒投与群では全例で間代性痙攣が誘発されるのに対して、試験化合物投与群では痙攣の発現が抑制されることを確認できる。
本試験は、試験例2)と同様に化合物の抗痙攣作用を評価する試験である。この試験で用いる動物モデルは、試験例2)の表現系とは異なり、全般性の欠神発作やミオクロニー発作の表現系である。

[試験例4]ロータロッド試験
本試験は、化合物の協調運動障害作用を評価する試験である。
雄性ICRマウス(日本エスエルシー株式会社、5-6週齢、1群8例)をロータロッド装置(室町機械株式会社)に乗せ、15回転/分の速度で回転する棒から2分間落下しないで歩行できるように訓練する。試験化合物を5%アラビアゴム/蒸留水(w/v)に懸濁し、30 mg/kgの用量で訓練済みのマウスに強制経口投与する。1時間後に上記と同様のロータロッド装置に乗せ、5分間で4回転/分から40回転/分に加速する棒の回転に合わせて歩行できるかを、200秒間観察し、落下潜時を計測する。溶媒投与群の落下潜時の平均値に対する、化合物投与群の落下潜時の相対値を算出する。
上記試験において、試験化合物投与群では協調運動障害が認められないことを確認できる。
本試験は、化合物の協調運動障害作用を評価する試験である。

上記試験において、試験化合物投与群では協調運動障害が認められないことを確認できる。
以上のように、本発明の化合物は抗てんかん薬の評価に用いられる複数のモデル動物において、いずれにも抗痙攣作用を示すので、広範な治療スペクトルを示す抗てんかん薬(例えば、単純部分発作、複雑部分発作若しくは二次性全般化発作等の部分発作;間代発作、強直発作、強直間代発作、熱性発作、欠神発作(小発作)、ミオクロニー発作、脱力発作、レノックス・ガストー症候群、ウエスト症候群(点頭てんかん)、ドラベ症候群及び多剤耐性発作等からなる群から選ばれた全般発作に対する予防薬及び/又は治療薬)として有用である。更に、本発明の化合物は、てんかんの診断薬としても有用である。

R3は、ハロ;C2-6アルキニル;ハロで置換されていてもよいC1-6アルキル;重水素又はハロで置換されていてもよいC1-6アルキル-O-;ハロで置換されていてもよいC1-6アルキル-S-;-CN;又はベンジル-O-であり;
Lは、直接結合、C1-6アルキレン、-O-又は-S-であり;
m及びnは、それぞれ0又は1であり;
qは、0、1又は2であり、qが2の場合、R3は各々独立して、同一又は異なった置換基を示す;
は、一重結合又は二重結合を示す]
で表される化合物又はその塩。 - 環Aが、フェニルであり;
Lが、-O-であり;
m及びnが、それぞれ0である、請求項1に記載の化合物又はその塩。 - R3がハロ又はC1-6アルキルである、請求項1~2のいずれかに記載の化合物又はその塩。
- 有効成分としての上記請求項1~5のいずれかに記載の化合物又はその塩と、薬学的に許容される担体を含む、医薬組成物。
