Abstract

A preparation method of extracting Aronia melanocarpa fruit comprises the following steps: selecting of Aronia melanocarpa fruit, washing and disinfecting of the said fruit, fermenting of the said fruit, fermentation neutralization by small cluster water with reducing powder, separating and extracting of active components of the said fruit according to concentrations of alcohol, filtering, controlling an alcoholic strength and tightly packing. The extract of Aronia melanocarpa fruit has an effect of clearing blood and anti-aging.

Extract of Aronia melanocarpa fruit and the preparation method thereof
Field of invention
The present invention relates to extract of Aronia melanocarpa fruit that is one of natural health food and grown in DPR Korea, which has an effect of clearing blood and anti-aging, and the preparation method thereof. In particular, the invention relates to high purity extract of Aronia melanocarpa fruit, which maintains its proper taste, scent and color as it is without destroying active components in the fruit; and the preparation method thereof.
Background and purpose of invention
As fruit of Aronia melanocarpa has rich vitamin P (hesperidin or rutin) which is a kind of phenolic compound and other physiological active materials, it attracts the most attention among fruits.
As Aronia melanocarpa fruit contains a lot of useful components, it is widely used in various fields such as food, medicine, etc.
The fresh fruit of Aronia melanocarpa contains 8-13% of sugar and 1.8% of organic acid, and has rich vitamin and microelements, thereby becoming a very good health food.
The fruit contains plant active materials such as anthocyan of pigment series, catechol, tannin, carotene, etc, and In particular, the content of anthocyan and catechol takes the first place among the natural fruits known up to now.
Polyphenol in Aronia melanocarpa fruit improves the structure and function of blood capillary and blood vessal. Anthocyan and flavonol elevate the function of heart, urinary system and immunity system, are effective in the treatment of internal disease, in particular gastritis and makes eyesight good. The drawback of the prior arts is that as the active components contained in Aronia melanocarpa fruit are not extracted fully, the content of active component in extract of Aronia melanocarpa fruit is low.
If dried Aronia melanocarpa fruit is used to obtain essence of Aronia melanocarpa fruit, many active components including components of vitamin P series of the fruit are destroyed in the course of drying.
The drawback is, further that as extracting of Aronica melanocarpa fruit is carried out in high concentration alcohol of over 60%, protein component of fruit is coagulated, thereby not fully extracting active components of fruit.
The drawback of the method for obtaining the extract by crushing Aronia melanocarpa fruit quenched is that its process is complicated and some vitamins of fruit are destroyed in the course of cooling.
The present invention uses fresh fruit instead of dried fruit and increases gradually concentration of alcohol using in extracting, thereby providing the extract of Aronia melanocarpa fruit, which maintains its proper taste, scent and color as it is without destruction or loss of pharmacologic components of the fruit, and has an effect of clearing blood and anti-aging.

Pharmacologic function of Aronia melanocarpa fruit is to have a special effect on treatment of arteriosclerosis, cardiovascular disease, hypertension and cerebrovascular disease.

The extract of Arohia melanocarpa fruit according to the present invention has the following advantages.
First, proper taste, scent and colour of Aronia melanocarpa fruit are maintained as it is, extracting efficiency of active components of the fruit is high without destruction or loss, and absorption ratio into the human body is high.
Second, the extract of Aronia melanocarpa contains solubilized mineral materials and various useful proteins such as vitamin B1; vitamin B2, vitamin C, vitamin P, anthocyan, β-carotene, total flavonoid, quercetin, total organic acid, calcium, magnesium, kalium, phosphorous, sodium, iron, zinc, copper, etc.
Third, as the active components of Aronia melanocarpa fruit are extracted with a high purity, the extract is effective in prevention and treatment of various diseases including cardiovascular system, cerebrovascular system, immune system, urinary system, etc, and reduces the period of treatment.
Forth, the extract is convenient for human to use daily for improvement of health. The preparation method of the extract of Aronia melanocarpa fruit comprises (1) selecting of Aronia melanocarpa fruit, (2) washing and disinfecting of the said fruit, (3) fermenting of the said fruit, (4) neutralizing fermentation, (5) separating and extracting active components of the said fruit according to concentrations of alcohol, (6) filtering, (7) controlling alcoholic strength and (8) tightly packaging.

Detailed description of the invention
The preparation method of the extract of Aronia melanocarpa fruit is described in detail through the figure 1.
As shown in figure 1, the preparation method of the extract of Aronia melanocarpa according to the invention comprises the following steps: (1) selecting of Aronia melanocarpa fruit, (2) washing and disinfecting of the said fruit, (3) fermenting of the said fruit, (4) neutralizing fermentation, (5) separating and extracting active components of the said fruit according to concentrations of alcohol, (6) filtering, (7) controlling alcoholic strength and (8) tightly packaging.
The said preparation method according to the steps is described below.
In selecting step of Aronia melanocarpa fruit (1), branches, leaves and other impurities are removed from the fresh fruits of Aronia melanocarpa yielded, unripe, overripe or broken fruits are removed, and full and half-ripe fresh fruits are optionally used.
Aronia melanocarpa fruit is tinged with black purple and it is preferable to be a little juicy.
In washing and disinfecting step (2), Aronica melanocarpa fruit is washed by water and is disinfected by using antiseptic solution. When disinfecting, the fruit is disinfected by soaking it in oxidizing water (pH=2~2.7) which has not direct influence on the fruit and has strong germicidal power, for about 15 minutes.
In fermenting step (3), the washed and disinfected fruit is stored in an air-tight container, and is fermented for 5 days in a dark place of 25^-30°C by adding 25% of sugar liquid thereto. In the course of fermentation, the proper taste and color of the fruit are maintained at it is, absorption ratio of active components into the human body is high. Also, in the course of fermentation, change of anthocyan into tannin is restrained.
After 3-4 days from starting to be fermented, the fruit begins to be inflated to the full, and after 5 days, the fruits are broken and the juice tinged with bright red is come from the fruit.
After 7 days from starting to be fermented, many small bubbles are formed in the upper layer of container, and after 8 days, getting musty, tingeing with purple and beginning to be decomposed. Therefore, it is desirable to go over to the next process step in 5 days after starting to be fermented.
In neutralizing fermentation (4), the fermented Aronia melanocarpa fruit is neutralized for 1 day by small cluster water with reducing power, of which pH is not less than 10.
The small cluster water neutralizes active components oxidized in the course of fermentation to suspend a decomposition process, thereby maintaining a proper taste and quality of Aronia melanocarpa fruit.
Also, as the small cluster water permeates quickly cells of Aronia melanocarpa fruit, its extracting effect is over 2 times as compared with that of normal water.
If Aronia melanocarpa fruit is directly extracted from alcohol not less than 60%, protein component of the fruit is coagulated, thereby not extracting fully the active components contained in the flesh of fruit. Therefore, it is desirable to gradually increase the concentration of alcohol from 25% to extract fully the active components of the fruit.
In separating and extracting step of active components according to concentrations of alcohol (5), medicinal alcohol of 25% is added to the fruit soaked by small cluster water at the rate of 1 : 1, leaving it for 5 days at over 30°C, separating and extracting the active components. Then, supernatant fluid is drained off, medicinal alcohol of 40% is added thereto at the rate of 1 : 1, leaving it for 5 days at over 30°C, separating and extracting the active components. Then, supernatant liquid is drained off, medicinal alcohol of 60% is added thereto at the rate of 1 : 1, leaving it for 5 days at over 30°C, separating and extracting the active components. In filtering step (6), three extracts of Aronia melanocarpa fruit extracted in medicinal alcohol of 25%, 40% and 60% respectively are mixed and filtered three times, thereby obtaining clear filtrate.
In controlling step of alcoholic strength (7), alcohol is recovered and the extract is concentrated into a quarter, thereby the extract with 5-7% of alcoholic strength.
In tightly packaging step (8), the extract of Aronia melanocarpa fruit obtained in the above step is tightly packaged by 100 m£.
The contents of the active components contained in the extract of Aronia melanocarpa fruit show in table 3.

The main component of the extract of Aronia melanocarpa fruit is anthocyan of which anti-oxidizing function is quite strong.
The extract of Aronia melanocarpa fruit having 5-7% of alcohol strength is adjusted to be 7.5-8.5 of pH, then when storing it more than 1 year in sterilized and tightly packaged state, the extract is never decomposed.
After patients suffered from cerebral thrombosis, angina cordis, arteriosclerosis and hyperlipemia drank the extract of Aronia melanocarpa fruit in the form of undiluted solution or solution diluted with water, by 10-15mg a time before meals three times a day for 60 days, finding of experimental check was recorded in checklist, and the condition after treatment was compared with that before treatment. The comparison data shows in table 4.

As seen in table 4, the remarkable effect in the treatment was proved.
After the extract of Aronia melanocarpa fruit was administered into the stomach of the experimental animals (white rats) in the amount of lOml/kg, when the said animals were observed for 15 days, there was no any death or abnormal reaction. After the extract was administered into the white rats by 0. Iml/kg a day for 20 days, when the indexes indicated in the table 5 were tested, significant difference between the test group and control group is not proved.

Claims
1. The extract of Aronia melanocarpa fruit which is grown in DPR Korea and has an effect of clearing blood and anti-aging, which comprises vitamin C of 44-80 mg/£, vitamin
Bi of 85-160 DIg/£, vitamin B2 of below 2.0 mg/£, vitamin P of 560-1000 mg £, tannin of 1540-2000 mg/e, anthocyan of 163-200 mg/£, total flavonoid of 892-1600 mg/£, quercetin below 20 mg/£, triterpenoid of 10-20 mg/£, β-carotene below 20 mg/£, total organic acid of
1000-2000 mg/£; mineral components such as calcium of 202-400 fl]g/£, magnesium of
85-160 mg/£, kalium of 215-400 mg/£, sodium of 38- 60 mg/£, iron of 0.25-0.5 mg/£, manganese of 0.7-1.5 mg/£, zinc of 0.06-0.12 mg/£ copper of below 0.1 mg/£, nickel of below 0.1 mg/£, phosphorus of below 0.2 mg/£, cadmium of below 0.06 mg/£ and cobalt of below 0.1 mg/£.

The preparation method of the extract of Aronia melanocarpa fruit according to claim 1, is characterized in that it comprises the following steps: selecting of Aronia melanocarpa fruit; washing and disinfecting of the said fruit; fermenting of the said fruit; fermentation neutralization by small cluster water with reducing power; separating and extracting of active components of the said fruit according to concentrations of alcohol; filtering; controlling an alcoholic strength; and tightly packaging.

The preparation method of the extract of Aronia melanocarpa fruit according to claim 2, is characterized in that, in selecting step of Aronia melanocarpa fruit, branches, leaves and other impurities are removed from the fresh Aronia melanocarpa fruit; unripe, overripe or broken fruit is removed; and full and half-ripe fresh fruits are optionally used. The fruit is tinged with black purple and is a little juicy.
4. The preparation method of the extract of Aronia melanocarpa fruit according to claim 2, is characterized in that, in washing and disinfecting step, the fruit is soaked in oxidation water with pH of 2-2.7 for 15 minutes to disinfect it.

The preparation method of the extract of Aronia melanocarpa fruit according to claim 2, is characterized in that, in fermenting step, the washed and disinfected fruit is stored in an air-tight container; 20% of sugar solution is added thereto; fermenting the fruit in a dark place of 25~30°C for 5 days.

The preparation method of the extract of Aronia melanocarpa fruit according to claim 2, is characterized in that, in neutralizing fermentation by small cluster water, after 5 days from starting to be fermented, the fermented fruit is neutralized for 1 day by the small cluster water of which pH is not less than 10.

The preparation method of the extract of Aronia melanocarpa fruit according to claim 2, is characterized in that separating and extracting of active components of the said fruit according to concentrations of alcohol comprises the following steps: medicinal alcohol of 25% is added to the neutralized fruit at the rate of 1: 1, leaving it fo 5 days at the temperature over 30°C, separating and extracting the active components; then supernatant liquid is drained off, adding medicinal alcohol of 40% thereto at the rate of 1 : 1, leaving it for 5 days at the temperature over 30°C, separating and extracting the active components; then supernatant liquid is drained off, adding medicinal alcohol of 60% to thereto at the rate of 1 : 1, leaving it for 5 days at the temperature over 30 °C, separating and extracting the active components.

The preparation method of the extract of Aronia melanocarpa fruit according to claim 2, is characterized in that, in filtering step, three extracts of Aronia melanocarpa fruit extracted in medicinal alcohol solutions of 25%, 40%; and 60% respectively are mixed and filtered three times, thereby obtaining clear filtrate.
9. The preparation method of the extract of Aronia melanocarpa fruit according to claim 2, is characterized in that, in controlling step of an alcoholic strength, the alcohol is recovered and the extract is concentrated into a quarter, thereby obtaining the extract with 5-7% of alcoholic strength.
10. The preparation method of the extract of Aronia melanocarpa fruit according to claim 2, is characterized in that, the filtered extract of Aronia melanocarpa fruit is tightly packaged by 100 mi.
