Abstract
본 발명의 판두라틴 유도체 또는 보에센베르기아 판두라타 추출물을 유효성분으로 포함하는 조성물에 관한 것으로, 상기 조성물은 파골세포 분화를 억제하여 골 손실 질환의 치료, 예방 또는 개선 효과를 가지고, 천연물로서 부작용 없이 안전하게 사용될 수 있어 의약품, 의약외품 또는 식품에 효과적으로 사용될 수 있다.
Description
본 발명은 판두라틴(panduratin) 유도체 또는 보에센베르기아 판두라타 추출물을 포함하는 골 손실 질환의 예방, 개선 또는 치료용 조성물에 관한 것이다.
정상적인 골의 기능을 위해서는 파골 세포(osteoclast)에 의한 골 흡수(bone resoprtion)와 조골 세포(osteoblast)에 의한 골 형성(bone formation)의 항상성 작용에 따른 뼈 리모델링(bone remodeling) 과정이 필요하다. 그러나 파골 세포의 지나친 활성이나 조골 세포의 활성 저하는 리모델링 과정의 불균형을 초래하여, 골다공증(osteoprosis)과 같은 골 손실 질환(골격계 질환)을 유도한다.
골다공증은 다발성 골수증, 골관절염 등의 골 관련 질병들 중 가장 흔한 골 손실 질환으로서, 일반적으로 골절의 증가 및 골 강도의 감소가 특징이다(Nat. Rev. Endocrinol. 8: 212-227, 2011; J. Dent. Res. 91: 736-744, 2012).
한편, 구강 내에서는 치조골의 파골 세포 활성이 급격히 증가하여 골흡수 반응이 초래되어 치주 조직에서 치조골 손실이 일어나 치아를 손실하는 치조골 손실 질환이 나타나게 된다(J. Immunol. Res. 2015: 1-10, 2015). 이러한 불균형을 해소하기 위해서 일반적으로 파골 세포의 지나친 활성을 억제하거나, 조골 세포의 활성을 촉진시키거나, 또는 파골 세포의 활성을 억제하고 조골 세포의 활성을 촉진하는 방법이 사용되고 있다(Nat. Rev. Rheumatol. 7: 631-638, 2011).
파골 세포는 조혈모세포에서 형성되는데 분화된 파골 세포는 무기질화된 뼈를 분해하기 때문에 신체 골격의 성장 및 항상성 유지에 매우 중요한 기능을 한다. 파골 세포의 분화는 TNF (tumor necrosis factor) 리간드인 RANKL(receptor activator of nuclear factor kappa-B ligand), RANK(receptor activator of nuclear factor kappa-B) 및 OPG(osteoprotegerin)와 같은 수용체에 의해 조절된다. RANKL은 조골세포나 인간 치주섬유아세포(human gingival fibroblast)와 같은 세포에서 분비되며, 파골세포나 파골세포 전구체 세포(osteoclast precursor cell)에 발현되어 있는 RANK에 부착하게 된다. 이 부착신호에 의해 주요전사인자인 NFATc1(nuclear factor of activated T-cells, cytoplasmic 1)의 활성이 증가되고 이에 따라 골 분해 및 흡수에 주요한 역할을 하는 TRAP(tartrate-resistant acid phosphatase), 카뎁신 케이(cathepsin K)와 같은 효소와 파골세포 분화 특이적 생체지표인 칼시토닌 수용체(calcitonin receptor)의 합성이 촉진된다. OPG는 디코이(decoy) 수용체로써 RANKL과 부착되어 RANKL이 RANK에 직접 부착되지 못하게 막게 된다. 따라서 RANKL과 OPG의 상대적 농도가 골격 항상성 유지에 매우 중요한 역할을 하게 된다(J. Immunol. Res. 2015: 1-10, 2015).
현재까지 개발된 대표적인 골다공증 치료제는 파골세포의 기능을 약화시켜 뼈 손실을 막아주는 비스포스포네이트(bisphosphonates) 제제가 주류를 이루고 있다. 하지만, 이러한 화학 합성품은 위장장애, 신장독성, 근골격통증, 턱뼈 괴사와 같은 부작용을 유발시키는 단점이 있다. 따라서 적은 부작용과 높은 안전성의 특징을 지닌 천연물을 기반으로 골다공증 또는 치조골 손실을 예방하고 치료하는 것은 매우 중요한 연구과제이다(Datamonitor Research Reports, 2007; Korean J. Fam. Pract. 3: 16-24, 2013).
보에센베르기아 판두라타(Boesenbergia
pandurata
, 동명-Boesenbergia
rotunda)는 핑거루트(fingerroot)라고 불리는 생강과(zingiberaceae family)식물로서, 근경(rhizome) 부위는 감기, 장염, 피부질환 및 요됴통증에 널리 사용되고 있다. 보에센베르기아 판두라타에는 피노셈브린 칼콘(pinocembrin chalcone), 카르다모닌(cardamonin), 피노셈브린(pinocembrin), 피노스트로빈(pinostrobin), 4-히드록시판두라틴 에이(4-hydroxypaduratin A), 판두라틴 에이(panduratin A)등이 함유되어 있는데, 이러한 성분들은 항암효과를 나타내는 것으로 보고된 바 있고(Trakoontivakorn, G.,등, J.Arig. Food chem.., 49, 3046-3050, 2001), 플라보노이드 계통의 디하이드로칼콘(dihydrochalcone) 화합물들은 살충효과를 나타내는 것으로 보고되고 있다(Phytochemistry, 34, 415-419, 1993).
그러나 본 발명의 이전에는 판두라틴 유도체 또는 보에센베르기아 판두라타 추출물의 골다공증 또는 치조골 손실을 포함하는 골 손실 질환에서 예방, 개선 혹은 치료 효과에 대해 상세히 보고된 바 없다.
본 발명은 파골세포의 분화를 억제하여, 뼈 리모델링 과정에서 뼈의 생성과 흡수 작용의 균형을 회복 및 개선 시키는 효과를 가지면서도 안전하게 적용될 수 있는 천연물인 판두라틴 유도체 및/또는 보에센베르기아 판두라타 추출물을 포함하는 조성물을 제공하는 것을 목적으로 한다.
상기 목적을 달성하기 위하여, 본 발명은 보에센베르기아 판두라타(Boesenbergia
pandurata) 추출물 또는 이의 분획물을 유효성분으로 포함하는 조성물을 제공한다.
구체적으로, 본 발명은 보에센베르기아 판두라타(Boesenbergia
pandurata) 추출물 또는 이의 분획물을 유효성분으로 포함하는 골 손실 질환 예방 또는 치료용 약학 조성물을 제공한다.

본 발명의 판두라틴 유도체 또는 보에센베르기아 판두라타 추출물은 파골세포 분화를 억제하여 골 손질 질환의 치료, 예방 또는 개선 효과를 가지고, 천연물로서 부작용 없이 안전하게 사용될 수 있어 의약품, 의약외품 또는 식품 등의 제조에 효과적으로 사용될 수 있다.

본 발명자들은 파골세포의 분화를 억제하여, 뼈 리모델링 과정에서 균형을 회복 및 유지시키는 효과를 가지며 안전하게 적용될 수 있는 천연물질을 탐색 및 연구한 결과, 판두라틴 유도체 및/또는 보에센베르기아 판두라타 추출물 또는 이의 분획물이 골 손실 질환 예방, 개선 또는 치료 효과를 가짐을 확인하고 본 발명을 완성하였다.
이하, 본 발명을 구체적으로 설명한다.
본 발명은, 보에센베르기아 판두라타(Boesenbergia
pandurata) 추출물 또는 이의 분획물을 유효성분으로 포함하는 골 손실 질환 예방, 치료 또는 개선용 조성물에 관한 것이다.
또한, 본 발명은 판두라틴 유도체 또는 이의 염을 유효성분으로 포함하는 골 손실 질환 예방, 치료 또는 개선용 조성물에 관한 것이다.
상기 '보에센베르기아 판두라타(Boesenbergia
pandurata, 동명-Boesenbergia
rotunda)'는 핑거루트(fingerroot)라고도 하며, 동남아시아에서 자생하는 생강과(zingiberaceae family) 식물이다. 근경 부위는 감기, 장염, 피부질환 등에 널리 사용되고 있다.
본 발명의 추출물의 제조는 상기 핑거루트의 모든 부위를 사용할 수 있고, 추출 부위의 제한을 받는 것은 아니나, 보에센베르기아 판두라타 식물의 근경을 바람직하게 사용할 수 있다. 추출물을 제조하기 위해서는 상기 식물체 형태에 의하여 제한되지 않고, 상기 식물체는 건조, 분쇄 등의 가공 과정을 거친 것을 모두 포함하는 의미이다.
본 발명의 일 실시예에서, 상기 보에센베르기아 판두라타로부터 추출한 추출물의 파골세포 억제효과를 확인하여, 이를 이용하여 뼈가 손실 및 생성되는 리모델링 과정에서 과도한 손실을 억제하여 균형을 맞춤으로서 골 손실 질환을 치료 또는 개선할 수 있음을 확인하였다.
본 발명의 보에센베르기아 판두라타 추출물은 바람직하게는 판두라틴(padurtin) 유도체를 포함하는 것 일 수 있다. 본 발명의 일 실시예에서 보에센베르기아 판두라타 추출물로부터 판두라틴 화합물을 분리 및 동정하고, 상기 추출물뿐 아니라, 분리된 판두라틴 화합물도 파골세포의 분화를 억제하는 효과가 있음을 실험적으로 확인하였다.
상기 '판두라틴(padurtin)'은 핑거루트(보에센베르기아 판두라타)라는 식물의 뿌리에서 추출된 성분으로, AMPK 활성 효과를 가진다고 알려져있다.
상기 판두라틴 유도체는 판두라틴 A(panduratin A), 4-히드록시판두라틴 A(4-hydroxypaduratin A), 및 이소판두라틴 A(isopanduratin A)를 포함한다. 상기 판두라틴 A는 (2,6-디히드록시-4-메톡시페닐)[3-메틸-2-(3-메틸부트-2-에닐)-5-페닐시클로헥스-3-에닐]메타논으로, 분자식은 C26H30O4이며, 바람직하게는 하기 화학식 1의 구조를 갖는 화합물 일 수 있다. 상기 4-히드록시판두라틴 A는 (2,4,6-트리히드록시페니[3-메틸-2-(3-메틸부트-2-에닐)-6-페닐시클로헥스-3-에닐]메타논으로, 분자식은 C25H28O4이며, 바람직하게는 하기 화학식 2의 구조를 갖는 화합물 일 수 있다. 또한, 상기 이소판두라틴 A(isopanduratin A)는 (2-메톡시-4,6-디하이드록시페닐)-[3-메틸-2-(3-메틸부트-2-에닐)-6-페닐시클로헥스-3-에닐]메타논으로, 분자식은 C26H30O4이며, 바람직하게는 하기 화학식 3의 구조를 갖는 화합물 일 수 있다.

본 발명의 조성물은 상기 판두라틴 유도체의 염을 유효성분으로 포함할 수 있다. 상기 염은 약학적으로 허용 가능한 염 일 수 있다.
본 발명의 일 실시예에서, 보에센베르기아 판두라타 추출물로부터 활성 성분을 분리 및 동정한 결과 판두라틴 A임을 확인하였고, 상기 판두라틴 A가 파골세포 분화를 억제하는 효과가 있음을 확인 하였다. 따라서 본 발명의 판두라틴 유도체는 보에센베르기아 판두라타(Boesenbergia
pandurata)로부터 추출된 것 일 수 있다.
본 명세서에서, '추출물'은 어떤 물질을 용매에 녹여 그 활성성분 또는 특성 성분을 분리한 것을 의미한다. 구체적으로 식물에 추출용매를 가하여 추출한 추출물 및 추출용매로 추출하여 제조한 추출물에 분획용매를 가하여 분획한 분획물을 포함한다. 따라서, 본 발명의 보에센베르기아 판두라타(Boesenbergia
pandurata) 추출물은 보에센베르기아 판두라타에 용매를 가하여 추출한 추출물과 이에 다시 분획용매를 가하여 분획한 분획물을 모두 포함하는 의미이다.
구체적으로, 상기 추출물은 보에센베르기아 판두라타의 근경을 이용한 에탄올 추출물, 열수 추출물, 헥산 추출물, 에틸아세테이트 추출물 또는 초고압 추출물 일 수 있다.
상기 추출용매는 물, 유기용매, 아임계수 및 초임계 유체로 이루어진 군에서 선택된 하나 이상 일 수 있다. 상기 유기용매는 극성용매, 비극성 용매, 극성 및 비극성 혼합용매 또는 물 일수 있다. 구체적으로, 탄소수 1 내지 6의 알코올(alcohol), 아세톤(acetone), 에테르(ether), 벤젠(benzene), 클로로포름(chloroform), 에틸아세테이트(ethyl acetate), 메틸렌 클로라이드(methylene chloride), 헥산(hexane), 시클로헥산(cyclohexane), 석유에테르(petroleum ether) 및 물로 이루어진 군에서 선택된 어느 하나 일 수 있다.
본 발명의 일 실시예에서, 보에센베르기아 판두라타에 에탄올을 가하여 추출물을 제조하고, 상기 보에센베르기아 판두라타 에탄올 추출물의 파골세포 활성을 억제하는 효과를 확인하였는바, 에탄올을 추출용매로 바람직하게 사용할 수 있다.
상기 식물 추출물은 통상의 식물 추출물의 제조방법에 따라 제조된 것일 수 있다. 보다 구체적으로, 불순물을 제거한 상기 식물에 추출용매를 가하고 추출과정을 수행하는 방법으로 수행할 수 있다. 상기 추출과정은 냉침추출법, 온침추출법, 가압추출법 또는 초음파 분쇄 추출법일 수 있다. 예를 들어, 건조시킨 식물체를 식품가공에 적합한 정제수, 에탄올 및 아임계수 또는 초임계 이산화탄소를 이용하여 추출, 정제하거나, 보에센베르기아 판두라타 식물을 100MPa 이상의 초고압 조건하에서 초고압 추출 장치를 이용하여 추출, 정제하여 얻을 수 있으며, 또는 식물체를 직접 압착하여 얻은 오일로부터 분리 정제하여 얻을 수 있다.
상기 분획용매는 물, 부탄올, 에틸아세테이트, 클로로포름, 헥산 또는 이들의 혼합물일 수 있다. 상기 분획물은 상기 추출법으로 제조한 추출물, 구체적으로 조추출액에 분획과정을 더욱 실시한 분획물 일 수 있다. 상기 분획용매는 에틸아세테이트, 에테르, 클로로포름, 벤젠, 헥산, 메틸렌클로라이드 및 이들의 혼합용매로 이루어진 군에서 선택된 용매일 수 있다. 상기 분획과정은 구체적으로 상기 조추출액에 혼합용매를 순서로 가한 후에, 층 분리한 분획물을 순차적으로 수득하는 방법으로 수행할 수 있다.
본 발명의 일 실시예에서, 상기 추출물은 보에센베르기아 판두라타 에탄올 추출물에 헥산, 클로로포름 및 에틸아세테이트 혼합 용매를 가하여 분획한 헥산-클로로포름-에틸아세테이트 제 1 분획물 일 수 있다. 또한, 상기 제 1 분획물에 다시 헥산, 에틸아세테이트 및 메탄올을 전개 용매로 사용하여 분획한 헥산-에틸아세테이트-메탄올 제 2 분획물 일 수 있다. 상기 헥산, 클로로포름 및 에틸아세테이트 혼합 용매는 1 내지 5 : 1 : 0.1 내지 0.5의 부피비로 혼합된 것 일 수 있고, 상기 헥산, 에틸아세테이트 및 메탄올은 15 내지 20 : 0.5 내지 4 : 1의 부피비로 혼합된 것 일 수 있다. 상기 본 발명의 분획물에 판두라틴 성분이 가장 많이 함유되어 있어, 골 손실 질환의 예방, 치료 또는 개선 효과에 우수한 장점이 있다.
상기 추출물 또는 분획물은 추출 또는 분획과정을 수행한 이후, 감압 여과 과정을 수행하거나 추가로 농축 및/또는 동결건조를 수행하여 농축하거나 용매를 제거할 수 있다. 상기 수득한 추출물은 사용시까지 급속 냉동 냉장고(deep freezer)에 보관할 수 있다.
상기 분획은 용매 분획, 실리카 겔 크로마토그라피(silica gel chromatography), prep-HPLC 등의 기술을 이용하여 활성물질이 농축된 특정 분획물을 제조할 수 있다.
본 명세서에서, '골 손실'은 파골세포와 조골세포의 불균형에 의하여 초래되는 뼈가 손실되는 증상을 의미하는 것으로, '골 손실 질환'은 상기 증상과 관련된 질병을 모두 포함하는 의미이다. 따라서, 파골세포의 활성이 지나치게 높아져 뼈가 손실되어 뼈의 밀도가 낮아지거나, 조골세포의 활성이 저하되어 뼈의 생성이 원활하게 일어나지 않아 초래되는 질환을 모두 포함한다. 구체적인 예로, 골다공증, 파제트병(Paget's disease), 치조골 손실, 골연화증 및 골이영양증을 포함한다.
상기 골다공증(osteoporosis)은 뼈의 양이 감소하고 질적인 변화로 인해 뼈의 강도가 약해져서 골절이 일어날 가능성이 높은 상태를 의미하는 것으로, 파골세포의 기능을 떨어트리거나, 골모세포를 활성화시키는 방법으로 증상을 완화, 개선, 치료하거나 해당 질환을 예방할 수 있다.
상기 파제트병(Paget's disease)은 골 재형성(bone remodeling)이 과도하게 항진되어 광범위한 부위의 골격계가 침범되는 골 질환을 의미하는 것으로, 골흡수 및/또는 골형성을 억제하는 방법으로 증상을 완화, 개선, 치료하거나 해당 질환을 예방할 수 있다.
상기 치조골 손실 치조(치주) 부위의 골이 손실되는 증상을 모두 포함하는 의미로, 치주염 또는 치은염에 의해 발생할 수 있으나, 이에 한정되는 것은 아니다.
상기 골연화증(Osteomalacia)은 새로 생성되는 골기질의 석회화 이상으로 골밀도의 감소를 보이는 질환을 의미하는 것으로, 구루병(rickets)라고도 한다. 파골세포의 활성을 억제함으로써 골의 연화를 지연하거나, 골연화에 의한 골절 등을 막을 수 있다.
상기 골이영양증(osteodystrophy)은 영양불균형에 의한 뼈의 성장저해증으로, 골다공증이 수반될 수 있다. 뼈형성장애, 골형성장애 또는 골형성이상증으로도 불릴 수 있다. 상기 골이영양증은 신성골이영야증, 비대성골이영양증(Hypertrophic Osteodystrophy) 및 태자성연골이영양증(chondrodystrophia foetalis, fetal chonchodystrophy)을 포함한다.
본 발명의 일 실시예에서, 파골세포에 판두라틴 또는 본 발명의 추출물을 처리한 결과, 파골세포의 활성이 억제되는 것을 실험적으로 확인하였다. 따라서, 본 발명의 조성물은 파골세포의 활성을 억제함으로써 골손실을 유발하는 과다한 뼈의 흡수를 억제 및 지연함으로써 골 손실 질환을 예방, 치료 또는 개선하는데 효과적이다.
본 명세서에서, '예방'은 질병 또는 병증의 발병을 억제하거나 지연시키는 모든 행위를 의미한다. 본 발명에 있어서는 파골세포의 활성을 억제하여, 골 손실 질환의 발병 시기를 지연시키거나, 발병을 억제하는 것을 의미한다.
본 명세서에서, '개선'은 질병 또는 병증 상태를 호전 또는 이롭게 변경하는 모든 행위를 의미하는 것으로, 본 발명에 있어서는 파골세포의 활성을 억제하는 작용을 통해서 골다공증의 증상 또는 치조골 손실과 같은 증상을 호전시키는 것을 의미한다.
본 명세서에서, '치료'는 질병 또는 병증의 진행을 지연, 중단 또는 역전시키는 모든 행위를 의미하는 것으로, 본 발명에 있어서는 파골세포의 활성을 억제하는 작용을 통해서 치조골 또는 뼈 등의 손실을 중단, 경감, 완화 또는 없애거나, 역전시키는 것을 의미한다.
본 발명의 일 실시예에서 보에센베르기아 판두라타(Boesenbergia
pandurata) 추출물 또는 판두라틴을 처리하여 골다공증의 발병원인이 되는 파골세포 분화를 억제할 수 있음을 확인하였다.
따라서, 이러한 측면에서 본 발명의 조성물은 골 손실 질환 예방 또는 치료용 약학 조성물 일 수 있다.
본 발명의 조성물은 골 손실 질환의 예방 또는 치료를 위하여 상기 유효성분 이외에, 추가로 동일 또는 유사한 기능을 가지는 유효성분을 1 종 이상 포함할 수 있다. 예컨대, 골 손실 질환의 예방 또는 치료에 효과가 있는 공지의 골다공증 치료제, 치조골 질환 치료제 등을 더 포함할 수 있다. 추가적인 성분을 포함하게 되면 본 발명의 조성물의 골 손실 질환에 대한 예방 또는 치료효과는 더욱 증진될 수 있다. 상기 성분 추가 시에는 복합 사용에 따른 안정성, 제형화의 용이성, 유효성분들의 안정성을 고려할 수 있다. 상기 추가의 성분은 전체 조성물 중량에 대하여, 0.0001 중량% 이상 내지 10 중량% 이하로 포함될 수 있다. 예를 들어, 0.0001 중량% 이상 내지 1 중량% 이하, 0.0001중량% 이상 내지 0.1중량% 이하, 0.0001 중량% 이상 내지 0.001 중량% 이하, 0.001중량% 이상 내지 10중량% 이하, 0.001중량% 이상 내지 1중량% 이하, 0.001 중량% 이상 내지 0.1중량% 이하, 0.01 중량% 이상 내지 10중량% 이하, 0.01중량% 이상 내지 1 중량% 이하일 수 있다. 상기 함량 범위는 안전성, 본 발명의 보에센베르기아 판두라타 추출물 또는 판두라틴 화합물의 제형화 시의 용이성 등의 요건에 따라 조절될 수 있을 것이다.
본 발명의 약학 조성물은 판두라틴의 약제학적으로 허용 가능한 염을 더 포함할 수 있다. 본 명세서에서 용어 '약학적으로 허용 가능한'이란 생리학적으로 허용되고 인간에게 투여될 때, 통상적으로 알레르기 반응 또는 이와 유사한 반응을 일으키지 않는 것을 말하며, 상기 염으로는 약제학적으로 허용 가능한 유리산(free acid)에 의하여 형성된 산 부가염이 바람직하다.
상기 약제학적으로 허용 가능한 염은 유기산 또는 무기산을 이용하여 형성된 산 부가염일 수 있으며, 상기 유기산은 예를 들면 포름산, 아세트산, 프로피온산, 락트산, 부티르산, 이소부티르산, 트리플루오로아세트산, 말산, 말레산, 말론산, 푸마르산, 숙신산, 숙신산 모노아미드, 글루탐산, 타르타르산, 옥살산, 시트르산, 글리콜산, 글루쿠론산, 아스코르브산, 벤조산, 프탈산, 살리실산, 안트라닐산, 디클로로아세트산, 아미노옥시 아세트산, 벤젠술폰산, p-톨루엔술폰산 또는 메탄술폰산을 포함한다. 무기산은 예를 들면 염산, 브롬산, 황산, 인산, 질산, 탄산 또는 붕산을 포함한다. 산 부가염은 바람직하게는 염산염 또는 아세트산염 형태일 수 있으며, 보다 바람직하게는 염산염 형태일 수 있다.
상기 언급된 산 부가염은 a) 판두라틴 및 산을 직접 혼합하거나, b) 이들 중 한 가지를 용매 또는 함수 용매 중에 용해시키고 혼합시키거나, 또는 c) 판두라틴을 용매 또는 수하 용매 중의 산에 위치시키고 이들을 혼합하는 일반적인 염 제조방법으로 제조된다.
위와는 별도로 추가적으로 염이 가능한 형태는 가바염, 가바펜틴염, 프레가발린염, 니코틴산염, 아디페이트염, 헤미말론산염, 시스테인염, 아세틸시스테인염, 메티오닌염, 아르기닌염, 라이신염, 오르니틴염 또는 아스파르트산염 등이 있다.
또한, 본 발명의 약학 조성물은 약학적으로 허용 가능한 담체를 더 포함할 수 있다.
약학적으로 허용되는 담체로는 예컨대, 경구 투여용 담체 또는 비경구 투여용 담체를 추가로 포함할 수 있다. 경구 투여용 담체는 락토스, 전분, 셀룰로스 유도체, 마그네슘 스테아레이트, 스테아르산 등을 포함할 수 있다. 또한 비경구 투여용 담체는 물, 적합한 오일, 식염수, 수성 글루코스 및 글리콜 등을 포함할 수 있다. 또한, 안정화제 및 보존제를 추가로 포함할 수 있다. 적합한 안정화제로는 아황산수소나트륨, 아황산나트륨 또는 아스코르브산과 같은 항산화제가 있다. 적합한 보존제로는 벤즈알코늄 클로라이드, 메틸- 또는 프로필-파라벤 및 클로로부탄올이 있다. 그 밖의 약학적으로 허용되는 담체로는 다음의 문헌에 기재되어 있는 것을 참고로 할 수 있다(Remington's Pharmaceutical Sciences, 19th ed., Mack Publishing Company, Easton, PA, 1995).
본 발명의 약학 조성물은 인간을 비롯한 포유동물에 어떠한 방법으로도 투여할 수 있다. 예를 들어, 경구 또는 비경구로 투여할 수 있으며, 비경구적인 투여방법으로는 이에 제한되는 것은 아니나, 정맥내, 근육내, 동맥내, 골수내, 경막내, 심장내, 경피, 피하, 복강내, 비강내, 장관, 국소, 설하 또는 직장 내 투여일 수 있다.
본 발명의 약학 조성물은 상술한 바와 같은 투여 경로에 따라 경구 투여용 또는 비경구 투여용 제제로 제형화 할 수 있다. 제형화할 경우에는 하나 이상의 완충제(예를 들어, 식염수 또는 PBS), 카보하이드레이트(예를 들어, 글루코스, 만노스, 수크로스, 또는 덱스트란 등), 항산화제, 정균제, 킬레이트화제(예를 들어, EDTA 또는 글루타치온), 충진제, 증량제, 결합제, 아쥬반트(예를 들어, 알루미늄 하이드록사이드), 현탁제, 농후제 습윤제, 붕해제 또는 계면활성제, 희석제 또는 부형제를 사용하여 조제될 수 있다.
경구투여를 위한 고형제제에는 정제, 환제, 산제, 과립제, 액제, 겔제, 시럽제, 슬러리제, 현탁액 또는 캡슐제 등이 포함되며, 이러한 고형제제는 본 발명의 약학 조성물에 적어도 하나 이상의 부형제 예를 들면, 전분(옥수수 전분, 밀 전분, 쌀 전분, 감자 전분 등 포함), 칼슘카보네이트(Calcium carbonate), 수크로스(Sucrose), 락토오스(Lactose), 덱스트로오스, 솔비톨, 만니톨, 자일리톨, 에리스리톨 말티톨, 셀룰로즈, 메틸 셀룰로즈, 나트륨 카르복시메틸셀룰로오즈 및 하이드록시프로필메틸-셀룰로즈 또는 젤라틴 등을 섞어 조제될 수 있다. 예컨대, 활성성분을 고체 부형제와 배합한 다음 이를 분쇄하고 적합한 보조제를 첨가한 후 과립 혼합물로 가공함으로써 정제 또는 당의정제를 수득할 수 있다.
단순한 부형제 이외에 마그네슘 스티레이트 탈크 같은 윤활제들도 사용된다. 경구를 위한 액상 제제로는 현탁제, 내용액제, 유제 또는 시럽제 등이 해당되는데 흔히 사용되는 단순 희석제인 물 또는 리퀴드 파라핀 이외에 여러 가지 부형제, 예를 들면 습윤제, 감미제, 방향제 또는 보존제 등이 포함될 수 있다.
또한, 경우에 따라 가교결합 폴리비닐피롤리돈, 한천, 알긴산 또는 나트륨 알기네이트 등을 붕해제로 첨가할 수 있으며, 항응집제, 윤활제, 습윤제, 향료, 유화제 및 방부제 등을 추가로 포함할 수 있다.
비경구적으로 투여하는 경우 본 발명의 약학 조성물은 적합한 비경구용 담체와 함께 주사제, 경피 투여제 및 비강 흡입제의 형태로 당 업계에 공지된 방법에 따라 제형화 될 수 있다. 상기 주사제의 경우에는 반드시 멸균되어야 하며 박테리아 및 진균과 같은 미생물의 오염으로부터 보호되어야 한다. 주사제의 경우 적합한 담체의 예로는 이에 한정되지는 않으나, 물, 에탄올, 폴리올(예를 들어, 글리세롤, 프로필렌 글리콜 및 액체 폴리에틸렌 글리콜 등), 이들의 혼합물 및/또는 식물유를 포함하는 용매 또는 분산매질일 수 있다. 보다 바람직하게는, 적합한 담체로는 행크스 용액, 링거 용액, 트리에탄올 아민이 함유된 PBS(phosphate buffered saline) 또는 주사용 멸균수, 10% 에탄올, 40% 프로필렌 글리콜 및 5% 덱스트로즈와 같은 등장 용액 등을 사용할 수 있다. 상기 주사제를 미생물 오염으로부터 보호하기 위해서는 파라벤, 클로로부탄올, 페놀, 소르빈산, 티메로살 등과 같은 다양한 항균제 및 항진균제를 추가로 포함할 수 있다. 또한, 상기 주사제는 대부분의 경우 당 또는 나트륨 클로라이드와 같은 등장화제를 추가로 포함할 수 있다.
경피 투여제의 경우 연고제, 크림제, 로션제, 겔제, 외용액제, 파스타제, 리니멘트제, 에어롤제 등의 형태가 포함된다. 상기에서 '경피 투여'는 약학적 조성물을 국소적으로 피부에 투여하여 약학적 조성물에 함유된 유효한 양의 활성성분이 피부 내로 전달되는 것을 의미한다.
흡입 투여제의 경우, 본 발명에 따라 사용되는 화합물은 적합한 추진제, 예를 들면, 디클로로플루오로메탄, 트리클로로플루오로메탄, 디클로로테트라플루오로에탄, 이산화탄소 또는 다른 적합한 기체를 사용하여, 가압 팩 또는 연무기로부터 에어로졸 스프레이 형태로 편리하게 전달 할 수 있다. 가압 에어로졸의 경우, 투약 단위는 계량된 양을 전달하는 밸브를 제공하여 결정할 수 있다. 예를 들면, 흡입기 또는 취입기에 사용되는 젤라틴 캡슐 및 카트리지는 화합물, 및 락토오스 또는 전분과 같은 적합한 분말 기제의 분말 혼합물을 함유하도록 제형화할 수 있다. 비경구 투여용 제형은 모든 제약 화학에 일반적으로 공지된 처방서인 문헌(Remington's Pharmaceutical Science, 15th Edition, 1975. Mack Publishing Company, Easton, Pennsylvania 18042, Chapter 87: Blaug, Seymour)에 기재되어 있다.
본 발명의 약학 조성물은 판두라틴 또는, 보에센베르기아 판두라타 추출물 또는 이의 분획물을 유효량으로 포함할 때 바람직한 골 손실 질환 예방, 개선 또는 치료 효과를 제공할 수 있다. 본 명세서에서, '유효량'이라 함은 음성 대조군에 비해 그 이상의 반응을 나타내는 양을 말하며 바람직하게는 골 손실 질환을 예방, 개선 또는 치료하기에 충분한 양을 말한다. 본 발명의 약학적 조성물에 판두라틴 또는 보에센베르기아 판두라타 추출물이 0.01 내지 99.99% 포함될 수 있으며, 잔량은 약학적으로 허용 가능한 담체가 차지할 수 있다. 본 발명의 약학 조성물에 포함되는 판두라틴 또는 보에센베르기아 판두라타 추출물 또는 이의 분획물의 유효량은 조성물이 제품화되는 형태 등에 따라 달라질 것이다.
본 발명의 약학적 조성물의 총 유효량은 단일 투여량(single dose)으로 환자에게 투여될 수 있으며, 다중 투여량(multiple dose)으로 장기간 투여되는 분할 치료 방법(fractionated treatment protocol)에 의해 투여될 수 있다. 본 발명의 약학 조성물은 질환의 정도에 따라 유효성분의 함량을 달리할 수 있다. 예를 들어, 판두라틴 또는 보에센베르기아 판두라타 추출물을 기준으로 하루에 체중 1 kg당 바람직하게 0.001 내지 100 mg, 더 바람직하게는 0.01 내지 10 mg의 양으로 투여되도록 1 내지 수회에 나누어 투여할 수 있다. 그러나 상기 판두라틴 또는 보에센베르기아 판두라타 추출물의 용량은 약학적 조성물의 투여 경로 및 치료 횟수뿐만 아니라 환자의 연령, 체중, 건강 상태, 성별, 질환의 중증도, 식이 및 배설율 등 다양한 요인들을 고려하여 환자에 대한 유효 투여량이 결정되는 것이므로, 이러한 점을 고려할 때 당 분야의 통상적인 지식을 가진 자라면 상기 판두라틴 또는 보에센베르기아 판두라타 추출물을 골 손실 예방, 치료 또는 개선을 위한 특정한 용도에 따른 적절한 유효 투여량을 결정할 수 있을 것이다. 본 발명에 따른 약학 조성물은 본 발명의 효과를 보이는 한 그 제형, 투여 경로 및 투여 방법에 특별히 제한되지 아니한다.
본 발명의 골 손실 질환 예방 또는 치료용 약학 조성물은 단독으로, 또는 수술, 방사선 치료, 호르몬 치료, 화학 치료 또는 생물학적 반응조절제를 사용하는 방법들과 병용하여 사용할 수 있다.
본 발명의 골 손실 질환 예방 또는 치료용 약학 조성물은 또한 판두라틴 또는, 보에센베르기아 판두라타 추출물을 유효성분으로 포함하는 외용제의 제형으로 제공할 수 있다. 이러한 측면에서, 본 발명의 조성물은 골 손실 예방 또는 개선용 의약외품 조성물 및 상기 조성물을 포함하는 의약외품 일 수 있다.
상기 외용제는 피부 또는 구강 내에 직접 적용될 수 있다. 본 발명의 골 손실 질환 예방 또는 치료용 약학 조성물을 외용제로 사용하는 경우, 추가로 지방 물질, 유기 용매, 용해제, 농축제 및 겔화제, 연화제, 항산화제, 현탁화제, 안정화제, 발포제(foaming agent), 방향제, 계면활성제, 물, 이온형 유화제, 비이온형 유화제, 충전제, 금속이온봉쇄제, 킬레이트화제, 보존제, 비타민, 차단제, 습윤화제, 필수 오일, 염료, 안료, 친수성 활성제, 친유성 활성제 또는 지질 소낭 등 피부 외용제에 통상적으로 사용되는 임의의 다른 성분과 같은 피부 과학 분야에서 통상적으로 사용되는 보조제를 함유할 수 있다. 또한, 상기 성분들은 피부 과학 분야에서 일반적으로 사용되는 양으로 도입될 수 있다.
본 발명의 조성물이 외용제로 제공될 경우, 이에 제한되는 것은 아니나, 액제, 연고, 패취, 겔, 크림 또는 분무제 등의 제형일 수 있다. 본 발명의 일 구현예에 따르면, 본 발명의 외약외품은 치약, 양치액 및 마우스 스프레이를 포함하는 구강관리 제품, 연고제, 마스크, 습포제, 첩부제 및 경피흡수제 등을 포함할 수 있다.
본 발명의 일 실시예에서, 판두라틴 또는 보에센베르기아 판두라타 추출물을 처리하는 경우 파골세포의 분화가 억제되었는바, 상기 유효성분을 구강관리 제품에 적용하는 경우, 파골세포의 골 흡수가 과다하게 일어나 문제가 되는 치조골 질환의 예방 또는 개선에 효과가 있다. 따라서, 상기 의약외품 조성물은 골 손실 예방 또는 개선을 위한 구강 관리용 조성물 일 수 있다.
본 발명의 조성물을 의약외품 조성물로 사용하는 경우, 보에센베르기아 판두라타 추출물 또는 판두라틴을 그대로 첨가하거나 다른 의약외품 성분과 함께 통상적인 방법에 따라 적절하게 사용될 수 있다. 유효성분의 혼합양은 사용 목적(예방, 건강 또는 치료적 처치)에 따라 적합하게 결정될 수 있다.
본 발명의 의약외품 조성물 및 의약외품에 대하여 상기 약학 조성물 및 하기 식품 조성물의 내용이 준용될 수 있다.
본 발명의 조성물은 다른 측면에서 골 손실 예방 또는 개선용 식품 조성물 일 수 있다.
상기 식품 조성물은 건강 기능성 식품 조성물 일 수 있다.
본 발명의 일 실시예에서, 보에센베르기아 판두라타 추출물 또는 판두라틴 유도체를 처리한 경우, 파골세포의 분화 및 활성을 억제함으로써, 골 손실을 직접적으로 예방 또는 지연시킬 수 있음을 확인하였다. 따라서, 상기 골 손실은 골 손실을 유발하는 질환에 의한 것, 바람직하게는 파골세포의 과다한 활성 등에 의해 나타나는 골 손실 질환에 의한 것 일 수 있다.
본 발명의 식품 조성물은 기능성 식품(functional food), 영양 보조제(nutritional supplement), 건강식품(health food), 식품 첨가제(food additives) 및 사료 등의 모든 형태를 포함하며, 인간 또는 가축을 비롯한 동물을 취식대상으로 한다.

또한, 영양보조제로는 이에 한정되지 않지만 캡슐, 타블렛, 환 등에 상기 판두라틴 또는 보에센베르기아 판두라타 추출물을 첨가하여 제조할 수 있다.
또한, 건강기능식품으로는 이에 한정되지 않지만 예를 들면, 상기 판두라틴 또는 보에센베르기아 판두라타 추출물 자체를 차, 쥬스 및 드링크의 형태로 제조하여 음용(건강음료)할 수 있도록 액상화, 과립화, 캡슐화 및 분말화하여 섭취할 수 있다. 또한, 상기 판두라틴 또는 보에센베르기아 판두라타 추출물을 식품 첨가제의 형태로 사용하기 위해서는 분말 또는 농축액 형태로 제조하여 사용할 수 있다. 또한, 상기 판두라틴 또는 보에센베르기아 판두라타 추출물과 골 손실 예방 또는 개선에 효과가 있다고 알려진 공지의 활성 성분과 함께 혼합하여 조성물의 형태로 제조할 수 있다.
본 발명의 식품 조성물이 건강음료 조성물로 이용되는 경우, 상기 건강음료 조성물은 통상의 음료와 같이 여러 가지 향미제 또는 천연 탄수화물 등을 추가 성분으로 함유할 수 있다. 상술한 천연 탄수화물은 포도당, 과당과 같은 모노사카라이드; 말토스, 수크로스와 같은 디사카라이드; 덱스트린, 사이클로덱스트린과 같은 폴리사카라이드; 자일리톨, 소르비톨, 에리트리톨 등의 당알콜일 수 있다. 감미제는 타우마틴, 스테비아 추출물과 같은 천연 감미제; 사카린, 아스파르탐과 같은 합성 감미제 등을 사용할 수 있다. 상기 천연 탄수화물의 비율은 본 발명의 조성물 100 mL 당 일반적으로 약 0.01 ~ 0.04 g, 바람직하게는 약 0.02 ~ 0.03 g 이다.
판두라틴 또는 보에센베르기아 판두라타 추출물은 골 손실 예방 또는 개선용 식품 조성물의 유효성분으로 함유될 수 있는데, 그 양은 골 손실 질환 예방, 개선 효과를 얻기에 유효한 양으로 특별히 한정되는 것은 아니나, 전체 조성물 총 중량에 대하여 0.01 내지 100 중량%인 것이 바람직하다. 본 발명의 식품 조성물은 판두라틴 또는 보에센베르기아 판두라타 추출물과 함께 골 손실 예방 또는 억제에 효과가 있는 것으로 알려진 다른 활성 성분과 함께 혼합하여 제조될 수 있다.
상기 외에 본 발명의 건강식품은 여러 가지 영양제, 비타민, 전해질, 풍미제, 착색제, 펙트산, 펙트산의 염, 알긴산, 알긴산의 염, 유기산, 보호성 콜로이드 증점제, pH 조절제, 안정화제, 방부제, 글리세린, 알코올 또는 탄산화제 등을 함유할 수 있다. 그 밖에 본 발명의 건강식품은 천연 과일주스, 과일주스 음료, 또는 야채 음료의 제조를 위한 과육을 함유할 수 있다. 이러한 성분은 독립적으로 또는 혼합하여 사용할 수 있다. 이러한 첨가제의 비율은 크게 중요하진 않지만 본 발명의 조성물 100 중량부당 0.01 ~ 0.1 중량부의 범위에서 선택되는 것이 일반적이다.

추출물의 제조
건조시킨 보에센베르기아 판두라타를 믹서로 분쇄한 다음, 분쇄한 보에센베르기아 판두라타 시료 100g을 에탄올 500mL에 넣고, 50℃에서 30분간 교반하면서 추출물을 제조하였다. 추출된 시료는 와트만(Whatman) 2번 여과지로 여과하고, 여과된 추출액을 진공 회전 농축기로 농축하여 용매성분을 제거한 후, 동결 건조하여 수분을 제거하여 보에센베르기아 판두라타 추출물을 얻었다.

A의 분리
상기 실시예 1에서 얻은 농축된 보에센베르기아 판두라타 추출물과 에틸아세테이트를 혼합하여 에틸아세테이트 용해성 성분을 추출하고, 감압 하에서 에틸아세테이트를 제거하여 에틸아세테이트 용해성 성분만을 농축하였다. 그 다음, 실리카겔을 6 X 15cm로 충진한 컬럼에 상기 농축된 성분을 적재하고 헥산, 클로로포름, 에틸아세테이트를 15:5:1.5(v/v/v)의 비율로 혼합한 용매시스템을 이용하여 분취하였다. 상기 분취 순서에 따라서 총 6개의 분획으로 나누어 각각의 분획을 농축 건조하였다. 6개의 분획 중 3번 분획(분획3)을 헥산, 에틸아세테이트, 메탄올을 각각 18:2:1(v/v/v)의 비율로 혼합한 용매를 전개 용매로 이용하여 박층 크로마토그래피(TLC, silica gel 60F254, Merck)를 수행 하고, 분취한 순서에 따라서 총 3개의 분획으로 나누어 농축 건조하였다. 최종적으로 상기 3개의 분획 중 2번 분획(분획3-2)을 이용하여 리사이클링 고성능 액체 크로마토그래피(recycling HPLC, column: W252, 20.0 mm ID X 500 mm L)를 수행하고, 분취 순서에 따라서 총 2개의 분획으로 나누어 각각의 분획을 농축 건조하였다. 최종적으로 상기 2개의 분획 중 2번 분획(분획3-2-2)을 농축 건조시켜 순수한 단일 활성 물질을 분리하였다.

판두라틴
A의 구조결정
상기 실시예 2-1에서 분리된 단일 활성물질의 구조결정을 위하여 1H-NMR 스펙트럼과 13C-NMR 스펙트럼을 각각 500 MHz 와 125 MHz(용매: CDCl3)에서 측정하였다. 수득된 13C-NMR 스펙트럼과 1H-NMR 스펙트럼의 결과를 토대로 1H-1H의 상관관계와 1H-13C의 상관관계를 측정하기 위해 1H-1H COSY 스펙트럼과 13C-HSQC 스펙트럼을 측정하고, 탄소공명을 통해 나오는 파장으로 각각의 탄소 신호를 구별하여 그 결과를 측정하였다.
또한 상기 분리된 단일물질의 질량분석을 위해 EI/MS를 측정하였다. 본 화합물은 EI/MS에서 [M+H+]가 m/z 407에서 관측되어 분자량이 406으로 판명되었고, 분자식은 C26H30O4이었다.
이상의 1H-NMR, 13C-NMR, 1H-1H COSY, 1H-13C HSQC 및 EI/MS에 대한 결과와 기존에 발표된 연구보고(Phytochemistry, 26: 1542-1543, 1987)를 비교 분석하여 동정한 결과, 상기 실시예 2-1에서 분리된 단일물질은 (2,6-디히드록시-4-메톡시페닐)[3-메틸-2-(3-메틸부트-2-에닐)-5-페닐시클로헥스-3-에닐]메타논으로서, 하기 화학식 1로 표시되는 판두라틴 A(panduratin A) 화합물로 확인되었다.

RAW264.7 파골세포 전구세포(American Type Culture Collection (ATCC), Manassas, VA, USA)를 6-웰 평판배양기(6-well microtiter plate)에 10%의 우태아 혈청이 함유된 DMEM(Dulbecco's Modified Eagle's Medium, Hyclone, Logan, UT, USA) 배지를 이용하여 8 X 104 세포/웰(cells/well)이 되도록 넣었다. 24시간 후, 10%의 우태아 혈청이 함유된 α-MEM(α-modified Eagle's Medium, Gibco, Grand Island, NY, USA) 배지에 25 ng/ml의 RANKL(Peprotech Inc., Rocky Hill, NJ, USA)과 보에센베르기아 판두라타 추출물 1 ㎍/ml, 10 ㎍/ml 또는 상기 실시예 2에서 얻은 판두라틴 A 0.1 μM, 1 μM씩을 각각 처리하여 4일 동안 분화를 유도하였다. 배지는 2일에 한번씩 교체하였다.
파골세포 분화 시 발현되는 주요 전사인자인 NFATc1(nuclear factor of activated T-cells, cytoplasmic 1)과 효소 TRAP, 카뎁신 케이(cathepsin K)의 mRNA 발현량을 알아보기 위해 RT-PCR을 수행하였다. 분화한 세포로부터 TRIzol시약(Takara, Tokyo, Japan)을 사용하여 총 RNA를 수확하여, cDNA 합성을 위해 상기 RNA를 역전사효소로 역전사 시킨 후, 다음의 프라이머를 이용하여 RT-PCR을 수행하였다. 그 결과는 도 1 또는 도 2에 나타내었다.

RAW264.7 파골세포 전구세포(ATCC)를 6-웰 평판배양기에 10%의 우태아 혈청이 함유된 DMEM(Hyclone) 배지를 이용하여 1 X 105 세포/웰(cells/well)이 되도록 넣었다. 24시간 후, 10%의 우태아 혈청이 함유된 α-MEM(Gibco) 배지에 25 ng/ml의 RANKL(Peprotech)과 보에센베르기아 판두라타 추출물 1 ㎍/ml, 10 ㎍/ml 또는 판두라틴 A 0.1 μM과 1 μM씩 각각 처리하여 24시간 동안 배양하였다. 그 후 파골세포 분화 시 발현되는 주요 전사인자 NFATc1과 효소 TRAP, 카뎁신 케이의 단백질 발현량을 알아보기 위해 웨스턴 블랏(western blot)을 수행하였다.
구체적으로, 프로테이나제 인히비터 칵테일(proteinase inhibitor cocktail)이 포함된 NP-40 완충용액(ELPIS-Biotech, Daejeon, Korea)으로 세포들을 용해시켰다. 완충용액에 용해된 세포를 1.5㎖ 튜브(tube)로 옮겨 13,000rpm으로 10분간 원심분리하고, 상등액 만을 취하였다. 상등액을 브래드포드(Bradford, Bio-Rad Laboratories Inc., Hercules, CA, USA)법을 이용하여 정량 하였다. 정량된 단백질을 5분간 끓인 후, 10% SDS-PAGE로 전기영동하여 분리하였으며, 분리된 단백질들을 니트로셀룰로스 막으로 전달하였다. NFATc1과 TRAP, 카뎁신 케이의 1차 항체를 2.5% bovine serum albumin(BSA)에 1:1000의 비율로 희석하여 니트로셀룰로스 막에 전달된 단백질과 20시간 동안 상온에서 반응시켰다. 1차 항체를 반응시킨 다음 Tris-buffer Saline Tween20(TBST)을 이용하여 니트로셀룰로스 막을 10분간 3회 세척하였다. 세척 후, 1차 항체를 인지하는 2차 항체를 2.5% BSA에 1:5000이 되도록 희석하여 니트로셀룰로스 막과 2시간 동안 상온에서 반응시켰으며, TBST를 이용하여 10분씩 3회에 걸쳐 세척하였다. 단백질 밴드(Protein band)는 ECL western blotting detection reagents(Amersham, Tokyo, Japan)를 사용하여 발색하였으며, G;BOX EF imaging system(Syngene, Cambridge, UK)을 이용하여 발색된 단백질 밴드(Protein band)를 확인하였다. 그 결과를 도 3에 나타내었다.
도 3 또는 도 4에 나타낸 바와 같이, 보에센베르기아 판두라타 추출물 또는판두라틴 A 처리에 의해 분화된 파골세포에서 NFATc1과 TRAP, 카뎁신 케이의 단백질 발현량이 감소한 것을 확인할 수 있었다. 특히, 보에센베르기아 판두라타 추출물 10 ㎍/ml 또는 판두라틴 A 1 μM 처리군에서 NFATc1과 카뎁신 케이의 단백질 발현이 거의 나타나지 않았다. 이는 본 발명의 보에센베르기아 판두라타 추출물 또는 판두라틴 A가 파골세포의 뼈 흡수 작용을 억제하는 효능이 우수하다는 것을 의미한다.

상기 시험예 1-1과 동일한 방법으로 RAW264.7 파골세포 전구세포를 6-웰 평판기에 부작시킨 후, 10%의 우태아 혈청이 함유된 α-MEM(Gibco)에 25 ng/ml의 RANKL(Peprotech)과 보에센베르기아 판두라타 추출물 1 ㎍/ml, 10 ㎍/ml 또는 상기 실시예 2에서 얻은 판두라틴 A 0.1 μM과 1 μM을 각각 처리하여 4일 동안 분화를 유도하였다. 배지는 2일에 한번씩 교환하였다. 그 후 상등액을 모두 제거한 뒤, 0.1% Triton X-100(Oriental chemical industries, Seoul, Korea)으로 세포들을 용해시켰다. 완충용액에 용해된 세포를 1.5 ml 튜브로 옮겨 13,000rpm으로 15분간 원심분리하고, 상등액 만을 취하였다. TRAP을 포함하는 이 상등액은 백혈구 산성 인산화효소 키트(Leukocyte acid phosphatase kit)(Sigma-Aldrich, St. Louis, MO, USA)를 이용하여 제조사의 가이드라인에 따라 분석하였다. 그 결과를 도 5 또는 도 6에 나타내었다.
도 5 또는 도 6에 나타낸 바와 같이, 보에센베르기아 판두라타 추출물 또는 판두라틴 A 처리에 의해 분화된 파골세포에서 TRAP 효소활성이 유의적으로 감소한 것을 확인할 수 있었다(##p < 0.01, **p < 0.01). 특히, 보에센베르기아 판두라타 추출물 10 ㎍/ml 또는 판두라틴 A 1 μM 처리군에서 현저하게 TRAP 효소 활성이 낮아짐을 확인하였다. 이는 본 발명의 보에센베르기아 판두라타 추출물 또는 판두라틴 A가 골세포의 뼈 흡수 작용을 억제하는 효능이 우수하다는 것을 의미한다.

상기 시험예 1-1에서와 동일한 방법으로 RAW264.7 파골세포 전구세포를 6-웰 평판기에 부착시킨 후, 10%의 우태아 혈청이 함유된 α-MEM(Gibco)에 25 ng/ml의 RANKL(Peprotech)과 상기 실시예 2에서 얻은 판두라틴 A 0.1 μM과 1 μM을 각각 처리하여 4일 동안 분화를 유도하였다. 배지는 2일에 한번씩 교환하였다. 4일 뒤 상등액을 모두 제거하고 2% 포름알데하이드(Junsei chemical, Tokyo, Japan)로 세포들을 고정하였다. 그 후 백혈구 산성 인산화효소 키트(Leukocyte acid phosphatase kit, Sigma-Aldrich)의 제조사의 가이드라인에 따라 TRAP 염색을 진행하여 보에센베르기아 판두라타 추출물 또는 판두라틴 A의 파골세포 분화억제 효능을 현미경 분석을 통해 분석하였다. 그 결과를 도 7 또는 8에 나타내었다.
그 결과, 도 7 또는 도 8에 나타낸 바와 같이 보에센베르기아 판두라타 추출물 또는 판두라틴 A 처리에 의해 파골세포 분화가 억제된 것을 확인할 수 있었다. 특히, 보에센베르기아 판두라타 추출물 10 ㎍/ml 또는 판두라틴 A 1 μM 처리군에서 대조군(control)과 거의 유사한 수준까지 분화를 억제하였음을 확인하였다. 이는 본 발명의 보에센베르기아 판두라타 추출물 또는 판두라틴 A가 파골세포의 뼈 흡수 작용을 억제하는 효능이 우수하다는 것을 의미한다.
이하, 본 발명에 따른 상기 실시예 1 내지 2의 보에센베르기아 판두라타 추출물 및/또는 판두라틴 A를 유효성분으로 포함하는 의약품 및 식품의 제조예를 설명하나, 본 발명은 이를 한정하고자 함이 아니라, 단지 구체적으로 설명하고자 함이다. 상기 골다공증 또는 치조골 손실 예방, 개선 또는 치료 효과가 우수한 보에센베르기아 판두라타 추출물 및/또는 판두라틴 A를 가지고 하기와 같은 조성성분 및 조성비에 따라 제조예 1 내지 2의 의약품 및 식품 조성물을 통상적인 방법에 따라서 제조하였다.

상기 실시예 1 내지 2의 보에센베르기아 판두라타 추출물 또는 판두라틴 A 1000 ㎎, 구연산 1000 ㎎, 올리고당 100 g, 매실농축액 2 g, 타우린 1 g에 정제수를 가하여 전체 900 ml 통상의 건강음료 제조방법에 따라 상기의 성분을 혼합한 다음, 약 1시간동안 85℃에서 교반 가열한 후, 만들어진 용액을 여과하여 멸균된 2 L용기에 취득하여 밀봉 멸균한 뒤 냉장 보관한 다음 건강음료 조성물 제조에 사용할 수 있다.

Claims (23)
- 보에센베르기아 판두라타(Boesenbergia pandurata) 추출물 또는 이의 분획물을 유효성분으로 포함하는 골 손실 질환 예방 또는 치료용 약학 조성물.
- 제1항에 있어서,상기 추출물은 물, 탄소수 1 내지 6의 유기용매, 아임계수 및 초임계 유체로 이루어진 군에서 선택되는 하나 이상의 용매의 추출물인, 약학 조성물.
- 제2항에 있어서,상기 탄소수 1 내지 6의 유기용매는 탄소수 1 내지 6의 알코올(alcohol), 아세톤(acetone), 에테르(ether), 벤젠(benzene), 클로로포름(chloroform), 에틸아세테이트(ethyl acetate), 메틸렌 클로라이드(methylene chloride), 헥산(hexane), 시클로헥산(cyclohexane) 및 석유에테르(petroleum ether)로 이루어진 군으로부터 선택되는 하나 이상인, 약학 조성물.
- 제1항에 있어서,상기 분획물은 보에센베르기아 판두라타 추출물에 대한 헥산, 클로로포름 및 에틸아세테이트로 이루어진 군에서 선택된 하나 이상을 포함하는 용매에 의한 분획물인, 약학 조성물
- 제1항 내지 제5항 중 어느 한 항에 있어서,상기 골 손실 질환은 골다공증, 파제트병(Paget's disease), 치조골 손실, 골연화증 및 골이영양증(osteodystrophy)으로 이루어진 군에서 선택된 것 인, 약학 조성물.
- 보에센베르기아 판두라타(Boesenbergia pandurata) 추출물 또는 이의 분획물을 유효성분으로 포함하는 골 손실 예방 또는 개선용 의약외품 조성물.
- 제7항에 있어서,상기 골 손실은 골다공증, 파제트병(Paget's disease), 치조골 손실, 골연화증 및 골이영양증(osteodystrophy)으로 이루어진 군에서 선택된 질환에 의한 것인, 의약외품 조성물.
- 보에센베르기아 판두라타(Boesenbergia pandurata) 추출물 또는 이의 분획물을 유효성분으로 포함하는 골 손실 예방 또는 개선용 의약외품.
- 보에센베르기아 판두라타(Boesenbergia pandurata) 추출물 또는 이의 분획물을 유효성분으로 포함하는 골 손실 예방 또는 개선용 식품 조성물.
- 제10항에 있어서,상기 추출물은 물, 탄소수 1 내지 6의 유기용매, 아임계수 및 초임계 유체로 이루어진 군에서 선택되는 하나 이상의 용매의 추출물인, 식품 조성물.
- 제11항에 있어서,상기 탄소수 1 내지 6의 유기용매는 탄소수 1 내지 6의 알코올(alcohol), 아세톤(acetone), 에테르(ether), 벤젠(benzene), 클로로포름(chloroform), 에틸아세테이트(ethyl acetate), 메틸렌 클로라이드(methylene chloride), 헥산(hexane), 시클로헥산(cyclohexane) 및 석유에테르(petroleum ether)로 이루어진 군으로부터 선택되는 하나 이상인, 식품 조성물.
- 제10항에 있어서,상기 분획물은 보에센베르기아 판두라타 추출물에 대한 헥산, 클로로포름 및 에틸아세테이트로 이루어진 군에서 선택된 하나 이상을 포함하는 용매에 의한 분획물인, 식품 조성물.
- 제10항 내지 제14항 중 어느 한 항에 있어서,상기 골 손실은 골다공증, 파제트병(Paget's disease), 치조골 손실, 골연화증 및 골이영양증(osteodystrophy)으로 이루어진 군에서 선택된 질환에 의한 것 인, 식품 조성물.
- 제16항에 있어서,상기 판두라틴 유도체는 보에센베르기아 판두라타(Boesenbergia pandurata)로부터 추출된 것인, 약학 조성물.
- 제16항 내지 제17항 중 어느 한 항에 있어서,상기 골 손실 질환은 골다공증, 파제트병(Paget's disease), 치조골 손실, 골연화증 및 골이영양증(osteodystrophy)으로 이루어진 군에서 선택된 것 인, 약학 조성물.
- 제21항에 있어서,상기 판두라틴 유도체는 보에센베르기아 판두라타(Boesenbergia pandurata)로부터 추출된 것인, 식품 조성물.
- 제21항에 있어서,상기 골 손실은 골다공증, 파제트병(Paget's disease), 치조골 손실, 골연화증 및 골이영양증(osteodystrophy)으로 이루어진 군에서 선택된 질환에 의한 것 인, 식품 조성물.
