本発明のPICは、ポリカチオン性ポリマーとしての式(I)のブロック共重合体(PEG-b-P(L-Arg)またはP(L-Arg)-b-PEG-b-P(L-Arg))とポリアニオン性ポリマーを含んでなるが、好ましくは、該共重合体と該ポリアニオン性ポリマー、から本質的になる(consisting essentially of)か、或いは、からなる(consisting of)ものである。
<PICミセル>
さらに、式(I)のブロック共重合体(PEG-b-P(L-Arg)またはP(L-Arg)-b-PEG-b-P(L-Arg))-とポリアニオン性ポリマーのPICは、水性媒体中で形成され、該共重合体中のポリ(L-アルギニン)(P(L-Arg)とも記載する。)セグメントとポリアニオン性ポリマー間の静電相互作用を介してPIC部を形成することにより、該PIC部をコアとし、一方、該共重合体のポリ(エチレングリコール)部をシェルとするナノサイズのポリマーミセルを形成するものと理解されている(例えば、実施例9の「ポリイオンコンプレックスミセルの調製」における、PICミセル水溶液についてDLSの測定を行った結果を参照。)。ここで、水性媒体は、純水もしくはイオン交換水、またはそれらの緩衝化溶液、水溶性有機溶媒(例えば、N,N-ジメチルホルムアミド、ジメチルスルホキシド等)含有水溶液等であることができる。
PICを形成するための式(I)で示されるブロック共重合体は、本発明の目的に沿うものであれば如何なる方法によって製造される相当するPEGセグメント及びP(L-Arg)セグメントを含むものであってもよい。しかし、該ブロック共重合体は、分子量分布が狭く、かつ、水性媒体中で該ブロック共重合体とポリアニオン性ポリマーから形成されるPICミセルのナノ粒子の平均径が腫瘍組織への取り込みに適するサイズとし得る該ブロック共重合体であることが好ましい。かような共重合体は、上記本発明の態様(8)に従って製造できる。
予め、式(II)
の前駆体である共重合体を用意し、次いで、該式中のL-オルニチンに由来するセグメント中のδ-アミノ基をグアニジノ化するためのグアニジノ化試薬、N,N’-ビス(tert-ブトキシカルボニル)-1H-ピラゾール-1-カルボキサミジンを用いて前記グアノジニノ化を行うことで製造することにより提供できる。このような製造は、典型的には、下記の合成スキームに従って、式(II)で示される共重合体とトリフルオロ酢酸(TFA)の塩を形成しておき、適当な塩基の存在下の不活性溶媒または塩基と溶媒との性質を備えた溶媒、例えば、N-メチルピロリドン、ジメチルスルホキシド等の溶媒中でグアニジノ化を行い、
次いで、グアニジノ化試薬に由来する部分に存在する保護基tert-ブトキシカルボニル(Boc)を脱離することによって目的の式(I)のブロック共重合体が製造される。脱離反応は、該共重合体のペプチド結合等に悪影響を及ぼさない条件下であれば如何なる条件下で行うこともできるが、TFAを用いる条件下で行うことが好ましい。
これに対し、式(II)のAが(II)’で定義される場合には、上記反応スキーム中の出発原料である化合物1としては、NH2CH2CH2-(OCH2CH2)nNH2を用い、以下,化合物2~4のA部分が、それぞれ、m個の反復単位に相当する反復単位であることができる。
で表され、本発明者の知る限り、従来技術文献未載の化合物である。
本発明に従うPICミセルは、前述した式(I)で示されるブロック共重合体と前述したポリアニオン性ポリマーを、前者のアミノ基もしくはグアニジノ基の数(Cという)/後者のカルボキシル基及び/もしくはスルホ基の数(A)の比が、0.5~2.50になるように調整し、緩衝化した水性溶液(pH=7.2~7.5)に溶解混合し、一定時間(通常、室温で30分間以上)静置することで調製するこができる。この調製に際し、前述の式(I)のAが(i)で定義されるブロック共重合体を使用して得られるミセル水溶液について動的光散乱(DLS)の測定を行うと、通常、平均粒径が約20nm~約50nmのミセル粒子を得ることができ、一方、式(I)のAが(ii)で定義されるトリブロック共重合体または式(III)で表されるトリブロック共重合体とポリアニオン性ポリマーのPICミセル水溶液についてDLS測定すると、通常、平均粒径が約 40 ~70のミセル粒子を得ることができる。
一方、式(III)のトリブロック共重合体単独もしくは当該共重合体及び実施例11に記載されるようにWO2015/118993Aに記載の、典型的には、PMNT-PEG-PMNTまたはそれを包含する下記一般式で表されるトリブロク共重合体との混合物(前者のL-Arg単位と後者のTEMPO単位の比は5~1:1~5であることができる。)と特定のアニオン性ポリマーのPICミセル水性溶液は、生体条件下のpH及び温度37℃以上でゲル化を起こす。したがって、当該PICを滞留させることが望まれる生体部位(例えば、心筋、関節など)に直接投与し、炎症が関与する疾患または障害を処置にすることができる。投与の態様については、本発明者らによる特許出願に基づく、WO2013/111801A、WO2013/111801A、WO2014/199982Aを参照できる。これらの特許文献は、引用することにより、その内容全体が本願明細書に組み込まれる。

実施例2に従って得られるL-Orn(Z)-NCAの量を3.9gから7.8gにした以外、実施例2から5までのスキームと同様の方法でPEG-b-P(L-Arg)の合成を行った。この時、得られたブロック共重合体のP(L-Arg)セグメントの重合度m=62であり、グアニジノ基の置換度は99%であった。1H-NMRを図11に示す。
L-Orn(Z)の代わりに、D-Orn(Z)を使用した以外実施例1から6までのスキームと同様の方法でPEG-b-P(D-Arg)の合成を行った。こうして得られたブロック共重合体のP(D-Arg)セグメントの重合度m=58であり、グアニジノ基の置換度は99%であった。1H-NMRを図12に示す。
L-Orn(Z)の代わりに、Nδ-ベンジルオキシカルボニル-L-リシン〔Nδ-Benzyloxycarbonyl-L-lysine(L-Lys(Z))〕を使用した以外実施例1から6までのスキームと同様の方法でPEG-b-P(L-Lys-Gua)の合成を行った。このブロック共重合体はPEG-b-poly(L-Lysine)ブロック共重合体のポリリジンセグメント中のε-アミノ基をグアニジノ基に置換したグアニジル置換ポリリジンまたはポリ(L-ホモアルギニン)である。こうして得られたブロック共重合体のP(L-Lys-Gua)セグメントの重合度m=56であり、グアニジノ基の置換度は99%であった。1H-NMRを図13に示す。
PEG-b-P(L-Arg)ブロック共重合体およびコンドロイチン硫酸Cナトリウム(CS:分子量50,000)をそれぞれTris-HClバッファ(10mM,pH7.4)に溶解させ、濃度を2mg/mLにしたPEG-b-P(L-Arg)ポリカチオン水溶液とCSポリアニオン水溶液を調製した。PEG-b-P(L-Arg)ポリカチオン水溶液とCSポリアニオン水溶液を任意のカチオン/アニオン比(C/A 比)で混合し、ボルテックスした後、30分静置することでPICミセル水溶液を調製した。ここで、C/Aとは[ポリカチオン中のアミノ基またはグアニジノ基の数]/[[ポリアニオン中のカルボキシル基またはスルホ基]である。得られたPICミセル水溶液に動的光散乱(DLS)測定を行ったところ、C/A=1の時に、最も散乱強度が大きく、多分散度(PdI)の小さなナノ粒子が得られた。この時の平均粒径は約35nm程度である(図14)。
iNOS酵素とポリアルギニンの反応性
実施例6から9で合成したブロック共重合体の粉末をHEPES-NaOHバッファ(10mM,pH7.4)にそれぞれ溶解させ、2mg/mLのポリマー水溶液を調製した。調製したポリマー水溶液を1mg/mLのトリプシン酵素水溶液と混合し37℃で24時間インキュベートした。またコントロールとして、トリプシン酵素を含んでいない単なるバッファとポリマー水溶液を同様に混合し、37℃で24時間インキュベートした。インキュベート後の混合溶液を98℃で5分間インキュベートし、トリプシン酵素を失活させた。この混合溶液とiNOS酵素(Sigma-Aldrich)を含んだバッファを混合し、37℃で24時間インキュベートした後、亜硝酸イオン(NO2 -)をグリース法(Dojindo,製品コードNK05)で定量した。その結果、実施例6で合成したPEG-b-P(L-Arg)ブロック共重合体のみトリプシン処理後に有意にNO産生が確認された(図15)。このNO産生量はPEGホモポリマー(分子量12,000)とL-アルギニンを同濃度含んだ系と同じ程度であることがわかった。
マクロファージ細胞からのNO産生評価
RAW264.7マクロファージ細胞(American Type Culture Collection,ATCC(U.S.A.))を使用した。培地は10%ウシ胎児血清(FBS)と抗生物質(アンピシリン、ストレプトマイシン、ネオマイシン)を加えているダルベッコ改変イーグル培地(DMEM)を使用した。24ウェルプレートに5,000cells/wellの細胞密度でRAW264.7マクロファージを播種し、37℃、5%CO2濃度で一晩インキュベートした。その後、実施例9の方法で調製したPICミセル水溶液をミセル中のアルギニン濃度が1mMになる濃度でウェルに投与し、72時間インキュベートした。その後、10ng/mLの濃度のリポ多糖(LPS)を加えて6時間インキュベートし、マクロファージを活性化させた。インキュベート後、ウェル中の上澄み培地を回収し、試験1と同様の手順で亜硝酸イオンを定量した。その結果、実施例6で合成したPEG-b-P(L-Arg)ブロック共重合体とコンドロイチン硫酸からなるPICミセル(L-PArg-CS/m,C/A=1)のみ、LPSで活性化したマクロファージから有意にNO産生が確認された(図16)。実施例7、8で合成し他PEG-b-P(D-Arg)ブロック共重合体およびPEG-b-P(L-Lys-Gua)ブロック共重合体とコンドロイチン硫酸からなるPICミセル(D-PArg-CS/mおよびL-PLys-Gua/m)を投与してもLPSで活性化したマクロファージからはNOの有意な産生は認められなかった。以上より、PEG-b-P(L-Arg)ブロック共重合体のみ有意にマクロファージ中のiNOS酵素と反応してNOを産生することを見出した。
担がんマウスへの抗腫瘍活性評価
実施例6で合成したPEG-b-P(L-Arg)ブロック共重合体とコンドロイチン硫酸Cから成るPICミセル(L-PArg-CS/m,C/A=1)の静脈投与による担がんマウスへの抗腫瘍活性を評価した。L-PArg-CS/m水溶液は実施例9の通り調製した。5週齢のBalb/cマウス(♂)を1群4匹に分けて、入荷から本実験終了までの期間中、室温25℃(±1℃))、湿度50%、12時間明暗サイクルの条件下で飼育を行い、餌及び水は自由に摂取させた。このマウスの右後脚大腿部に1×105cells/mLの細胞密度のColon-26がん細胞を100μL播種した。腫瘍組織の大きさはノギスで測定し、腫瘍体積(mm3)=[短軸(mm)]2×[長軸(mm)]×0.52で表す。腫瘍体積の平均が100mm3を超えた時に、下記の通りL-PArg-CS/m水溶液を投与した。

1.ポリ(L-Arg)-ポリ(エチレングリコール)-ポリ(L-Arg)(PArg-PEG-PArg)の合成
PArg-PEG-PArgは、ポリ(L-オルニチン)-ポリ(エチレングリコール)-ポリ(L-オルニチン)(POrn-PEG-POrn)(のグアニジニル化により製造した。
当該POrn-PEG-POrnはPEG(NH2-PEG-NH2;Mn=10,000)のα,ω-NH2基へのN-δ-カルボベンゾキシ-L-オルニチンのN-カルボン酸無水物(L-Orn(Z)-NCA)の開環重合、次いで、保護基の脱離を介して合成した。
PArg-b-PEG-b-PArgは、ポリ(L-オルニチン)-ポリ(エチレングリコール)-ポリ(L-オルニチン)(POrn-b-PEG-b-POrn)(のグアニジニル化により製造した。なお、ブロック共重合体を表示する「-b-」は、以下、単に「-」と略記する。
当該POrn-PEG-POrnはPEG(NH2-PEG-NH2;Mn=10,000)のα,ω-NH2基へのN-δ-カルボベンゾキシ-L-オルニチンのN-カルボン酸無水物(L-Orn(Z)-NCA)の開環重合、次いで、保護基の脱離を介して合成した。
(1)POrn-PEG-POrnの合成
窒素雰囲気下の三方停止栓を備えた300mL丸底フラスコ中で撹拌させることでNH2-PEG-NH2(2g,0.2mmol)をジメチルホルムアミド(DMF,20mL)に溶解した。別の100mLフラスコ中で20mLのDMFに溶解したNδ-ベンジルオキシカルボニル-L-オルニチン酸無水物(L-Orn(Z)-NCA;2g,6.85mmol)をNH2-PEG-NH2溶液にN2パージドシリンジを用いて加えた。乾燥窒素雰囲気下の反応混合物を30℃油浴で48時間撹拌した。反応後、氷浴中で15倍過剰のジエチルエーテル(600mL)を用いて反応混合物を沈殿させ、次いで濾過した。回収したポリマーをジエチルエーテルで2度再沈殿させ、不純物を除去した。白色固体としてPOrn(Z)-PEG-POrn(Z)(2.6g)が得られた。該固体の1H-NMRの分析結果を図19に示す。
得られたPOrn(Z)-PEG-POrn(Z)(2.6g)をトリフルオロ酢酸(TFA,15mL)に溶解させ、臭化水素酸(HBr,4.5mL)を加え、氷浴中で4時間撹拌した。次に、トリエチルアミン(TEA.15mL)を滴下し、過剰の酸を除去した。引き続き、懸濁物蒸留水(100mL)で希釈し、次いで、予め膨潤させた半透膜チューブ(MWCO=3500)を用い水に対して3日間透析した。さらに、連続して0.01HCl、蒸留水に対して透析を続け、TFA型POrn-PEG-POrnをHCl型に変換した後、凍結乾燥した。POrn-PEG-POrnを白色粉末として得た(2.2g)。該粉末の1H-NMRの分析結果を図19に示す。
(2)PArg-PEG-PArgの合成
PArg-PEG-PArgは、POrn-PEG-POrnのPOrnセグメントの側鎖δ-アミノ基のN,N’-ビス(tert-ブトキシカルボニル)-1H-ピラアゾール-1-カルボキサミジン(PCX(Boc2))を用いるグアニジニル化、次いでBoc基の脱保護によって取得した。
簡潔には、POrn-PEG-POrn(1g,0.7mmol)とPCX(Boc2)(1g、3.2mmol)をN,N-ジイソプロピルエチルアミン(0.5mL,3mmol)含有N-メチルピロリドン(20mL)に溶解し、室温で24時間撹拌した。反応後、混合物を氷浴中で15倍過剰のジエチルエーテル(300mL)で沈殿させ、次いで濾過した。回収したポリマー(PArg(Boc2)-PEG-PArg(Boc2))をジエチルエーテルにより2度再沈殿させ、不純物を除去した。ポリマー(1.1 g)が得られ、その1H-NMRの分析結果を図20に示す。こうして得られたPArg(Boc2)-PEG-PArg(Boc2)をTFA(10mL)に溶解し、室温で8時間撹拌した。次に、TEA(15mL)を滴下し、過剰の酸を除去した。引き続き、懸濁物を蒸留水(100mL)で希釈し、0.01HCl、次いで蒸留水に対して透析した後、凍結乾燥した。PArg-PEG-PArgを白色粉末として0.9g得た。この粉末の1H-NMRの分析結果を図20に示す。
ポリ(クロロメチルスチレン)-ポリ(エチレングリコール)-ポリ(クロロメチルスチレン)(PCMS-PEG-PCMS)を、スルファニル(sulphanyl?)-ポリ(エチレングリコール)-スルファニル(HS-PEG-SH;Mn=10,000)をテローゲンとして用いるクロロメチルスチレンのラジカルテロメリゼーションによって合成した。次いで、4-アミノ-2,2,6,6-テトラメチルピペリジン1オキシル(4-amino-TEMPO)のアミノ基とPCMS-PEG-PCMSブロック共重合体中のベンジルクロダイド基間の相互作用を介してクロロメチル基をTEMPOに転化した。具体的には、WO2015/118993A を参照されたい。
ポリカチオン性ポリマーのPArg-PEG-PArg及びPMNT-PEG-PMNTをリン酸緩衝液(pH6.2,100mM)に溶解した。ポリアニオン性ポリマーのポリ(アクリル酸)(PAAc)又はコンドロイチン硫酸(CS)もリン酸緩衝液(pH6.2,100mM)に溶解した。ポリカチオン性ポリマー溶液にポリアニオン性ポリマーを加えてポリイオンコンプレックス(アニオン:カチオン比 1:1)を調製した。混合物を室温にて30分間撹拌し、こうして形成しPICミセル粒子の動的光散乱(DLS)測定を行った。結果を下記表1に示す。
<試験4>
インビトロでの血管新生の評価
Arg-PEG-PArg/CSコンプレックスの血管新生促進性を、グロース-ファクター-リデュースド マトリゲル(growth-factor-reduced-Matrigel)(BD Biosciences)を用いるヒト臍帯静脈内皮細胞(HUVEC)チューブ検定により評価した。
簡潔には、製造業者のプロトコールに従い冷室温(4℃)でマトリゲルを夜どうし解かした。マトリゲル液(10μL)をμ-スライド(μ-slide)(同上)に加え、30分37℃のインキュベーター中に維持し、重合させた。次に、ダルベッコ改変イーグル培地(DMEM)中HUVEC懸濁液(5×103細胞/ウエル)を、Arg-PEG-PArg/CSコンプレックスの存在下又は不存在下のマトリゲル層上にゆっくり加えた。血管内皮細胞増殖因子(VEGF)を陽性血管新生促進剤として用いた。5%CO2下37℃にて4時間インキュベーションした後、DMEMを慎重に取り除いた後、カルセイン(calcein)AM含有(10μg/mL)新鮮DMEMをマトリゲル層上に加えた。15分インキュベーション後、チューブ形成を蛍光顕微鏡下で観察し、イメージジェイ(ImageJ)ソフトウエア―の血管新生解析装置プラグインにより解析した。得られた結果はPEG-PArg/CS及びPArg-PEG-PArg/CSがHUVEC中でチューブを形成することを示した。結果を図22に示す。なお、図中、PIC(di)は、上記実施例9にしたがって調製されるPEG-PArgジブロックコポリマーに由来するPICを意味し、PIC(tri)はPArg-PEG-PArgトリブロックコポリマーに由来するPICを意味する。
図22に示されるようにPEG-PArgもPArg-PEG-PArgもHUVECのチューブ形成を誘発することが分かる。
<試験5>
心筋梗塞マウスにおけるPMNT-PEG-PMNT/PArg-PEG-PArgをベースとするインジェクタブルハイドロゲルの評価
心筋梗塞(MI)マウスを心臓の左前下行枝(LAD)の連結を誘発させた。雄7~8週齢ICRマウス(体重32~35g)をチャールスリバージャパン(Charles River Japan)から購入した。これらのマウスを温度(23±1℃)、湿度(50±5%)及び明暗(12時間明暗周期)の制御下の筑波大学実験動物施設で飼った。これらの動物に飼料と水を自由に採ることができるようにした。すべての実験は筑波大学の動物実験用規則に従って実施した。LADを8-0シルク縫合糸で連結した後、濃縮PIC溶液(40μL,PMNT-PEG-PMNT/PArg-PEG-PArg+PAAc)を縫合糸部位に心臓内注入した。MI後1週目及び4週目に心エコー検査を行った。心機能は左室駆出率の測定により評価し、梗塞のサイズは組織学的評価により決定した。図23及び24に示されるとおり、PMNT-PEG-PMNT/PArg-PEG-PArgをベースとするインジェクタブルゲル処置マウスは未処置マウスに比べ有意に心機能を改善した。なお、図中、Gel1はPMNT-PEG-PMNT/PArg-PEG-PArg+PAAC(30mg/mL)のデータであり、Gel2はPMNT-PEG-PMNT/PArg-PEG-PArg+PAAC(60mg/mL)のデータである。
図24に関しては、1日間4%(v/v)緩衝化ホルマリンおよび2日間70%(v/v)アルコールで心臓組織を固定化した後パラフィン埋め込みを行い、次いで、これらの組織の5μm厚薄片を調製し、マッソン(Masson)トリクローム染色を行った。こした生物試料の組織学的特徴を光学顕微鏡下で試験した結果である。

- ポリカチオン性ポリマーとポリアニオン性ポリマーを含んでなるポリイオンコンプレックス(PIC)であって、
ポリカチオン性ポリマーが、式(I)

(i)水素、非置換または置換C1-C12アルキル基を表し、置換されている場合の置換基は、ホルミル基、式R1R2CH-(ここで、R1及びR2は独立して、C1-C4アルコキシまたはR1とR2は一緒になって-OCH2CH2O-、-O(CH2)3O-もしくは-O(CH2)4O-を表す。)の基を表すか、或は
(ii)式
を表し、
L及びL’は独立して、連結基を表し
Y及びY’は独立して、H、C1-21アルキルカルボニル、置換C1-4アルキルカルボニル、非置換もしくは置換C3-7シクロアルキルカルボニル、非置換もしくは置換アリールカルボニル、または非置換もしくは置換5もしくは6員ヘテロアリールカルボニルを表し、ここで、置換C1-4アルキルカルボニルの置換基は、ハロゲン原子、ヒドロキシル、カルボキシル、非置換もしくは置換C3-7シクロアルキル、非置換もしくは置換アリール及び非置換もしくは置換5もしくは6員ヘテロアリール、非置換もしくは置換アダマンチル、非置換もしくは置換コレステロール残基からなる群より選ばれ、これらの置換基が置換されている場合の置換基はC1-4アルキル、C1-4アルキルオキシ、ヒドロキシル、カルボキシル、シアノ、ニトロ、ハロゲン原子、またはモノ-もしくはジC1-4アルキルアミノであることができ、
m及びm’は独立して、5~300の整数であり、
nは5~1,000の整数であり、
式中のmまたはm’個のアミジノ基(C(=NH)NH2)の80%まではHであってもよい、
で示される共重合体であり、
ポリアニオン性ポリマーはポリアニオン性多糖、ポリアニオン性ポリペプチド、ポリ(アクリル酸)、及びポリ(メタアクリル酸)からなる群より選ばれ、かつ、
前記PICは水に溶解または分散させたとき、平均粒径がナノメートル(nm)オーダーのPICミセルの形態にある、
前記ポリイオンコンプレックス。 - 請求項1に記載のポリイオンコンプレックスであって、Aが(i)で定義される、ポリイオンコンプレックス。
- 請求項1に記載のポリイオンコンプレックスであって、Aが(ii)で定義される、ポリイオンコンプレックス。
- 請求項1~3のいずれか1に記載のポリイオンコンプレックスを有効成分として含む、哺乳動物組織における活性化された細胞に由来する誘導型NOシンターゼの基質L-Argを提供するための組成物。
- 哺乳動物組織における活性化された細胞が腫瘍組織内または近傍のマクロファージである、請求項4に記載の組成物。
- 哺乳動物組織における活性化された細胞が心臓筋肉内の炎症に伴い活性化されたマクロファージである、請求項4に記載の組成物。
- 請求項1~3のいずれか1に記載のポリイオンコンプレックスを有効成分として含む、哺乳動物組織における腫瘍を予防または治療するための組成物。
