Abstract
一种如式(I)所示的异丙氧苯胍的系列盐及其在制备动物饲用生长促进剂中的应用,其中R为DL-乳酸、甲磺酸、2-羟乙基磺酸、柠檬酸、酒石酸、苯甲酸、琥珀酸、富马酸、马来酸、醋酸、硫酸、磷酸或草酸。
Description
本发明属于畜禽动物饲料领域,具体涉及异丙氧苯胍的系列盐及其在制备动物饲用生长促进剂中的应用。
饲用生长促进剂包括多种作用机制的多种化学结构的生物活性物质及其制剂,对于提高饲料转化率、提高畜牧业生产效率有重要贡献。但是由于耐药性传递问题,许多与人类临床医学应用的抗生素具有结构相关性或交叉耐药性的饲用抗生素类生长促进剂已经或即将被禁用。
所以如何筛选到没有人类医学用途、与人用抗生素无交叉耐药性的安全、高效的动物专用的饲用生长促进剂,尤其是非抗生素类的、非激素类型的饲用生长促进剂是个问题,本研究解决了这个问题。
发明内容:
本发明的第一个目的是提供能够高效促进动物生长的异丙氧苯胍的系列盐。
本发明的异丙氧苯胍的系列盐,其结构式如式(1)所示:
其中:R为DL-乳酸、甲磺酸、2-羟乙基磺酸、柠檬酸、酒石酸、苯甲酸、琥珀酸、富马酸、马来酸、醋酸、硫酸、磷酸或草酸。
本发明的第二个目的是提供如式(1)所示的异丙氧苯胍的系列盐在制备动物饲用生长促进剂中的应用。
所述的动物为各个生长阶段的鸭、猪、鸡等养殖动物。
所述的异丙氧苯胍的系列盐,其在动物饲料中的添加剂量为0.1~500ppm,所述的饲料为全价配合饲料。
本发明的如式(1)所示的异丙氧苯胍的系列盐对动物低毒或者无毒,因此适合作为饲用生长促进剂,在养殖行业具有非常好的应用前景。
本研究通过饲养试验首次发现鸭在应用异丙氧苯胍的系列盐后,生产性能改善明显。本研究同样通过动物饲养试验发现应用此类异丙氧苯胍的系列盐后猪、鸡生产性能得到提高,因此本发明的异丙氧苯胍的系列盐能够用于制备饲用生长促进剂,具备广阔的应用前景。
以下实施例是对本发明的进一步说明,而不是对本发明的限制。
实施例1:游离的2,2’-双[[4-(异丙氧基)苯基]亚甲基]-二氨基胍的制备:
取2,2’-双[[4-(异丙氧基)苯基]亚甲基]-二氨基胍盐酸盐100g(含水约15%),加750ml 95%乙醇,加12.5gNaOH,2,2’-双[[4-(异丙氧基)苯基]亚甲基]-二氨基胍逐渐溶解,析出NaCl。搅拌0.5h,抽滤。滤液滴加到3L水中,析出固体。抽滤,得到的固体在45度烘干,得70g产品(2,2’-双[[4-(异丙氧基)苯基]亚甲基]-二氨基胍)。

其中R为DL-乳酸、甲磺酸、2-羟乙基磺酸、柠檬酸、酒石酸、苯甲酸、琥珀酸、富马酸、马来酸、醋酸、硫酸、磷酸或草酸。
实施例2:2,2’-bis[[4-(1-methylethoxy)phenyl]methylene]-carbonimidic dihydrazide.2-hydroxypropanoic acid/2,2’-双[[4-(异丙氧基)苯基]亚甲基]-二氨基胍.DL-乳酸盐的制备
取游离的2,2’-双[[4-(异丙氧基)苯基]亚甲基]-二氨基胍1.6g,加8ml 95%乙醇,室温搅拌,加0.4g DL-乳酸盐,搅拌2h,过滤,滤饼用95%乙醇洗,水洗,然后45℃干燥过夜,得1.1g产品(2,2’-双[[4-(异丙氧基)苯基]亚甲基]-二氨基胍.DL-乳酸盐,或名异丙氧苯胍乳酸盐),产率55%。1H NMR(500MHz,d-DMSO)δ7.99(s,2H),δ7.67(d,4H),δ6.92(d,4H),δ6.56(br,2H),δ4.63-4.67(m,2H),δ4.01(q,1H),δ1.28(d,12H),δ1.22(d,3H).
实施例3:2,2’-bis[[4-(1-methylethoxy)phenyl]methylene]-carbonimidic dihydrazide.methanesulfonic acid/2,2’-双[[4-(异丙氧基)苯基]亚甲基]-二氨基胍.甲磺酸盐的制备
取游离的2,2’-双[[4-(异丙氧基)苯基]亚甲基]-二氨基胍1.6g,加8ml 95%乙醇,室温搅拌,加0.4g甲磺酸,搅拌2h,过滤,滤饼用95%乙醇洗,水洗,然后45℃干燥过夜,得1.4g产品(2,2’-双[[4-(异丙氧基)苯基]亚甲基]-二氨基胍.甲磺酸盐,或名异丙氧苯胍甲磺酸盐),产率70%。1H NMR(500MHz,d-DMSO)δ11.8(br,1H),δ8.39(s,2H),δ8.26(br,2H),δ7.85(d,4H),δ7.01(d,4H),δ4.70-4.75(m,2H),δ2.41(s,3H),δ1.28(d,12H).
实施例4:2,2’-bis[[4-(1-methylethoxy)phenyl]methylene]-carbonimidic dihydrazide.2-hydroxyethanesulfonic acid/2,2’-双[[4-(异丙氧基)苯基]亚甲基]-二氨基胍.2-羟乙基磺酸盐的制备
取游离的2,2’-双[[4-(异丙氧基)苯基]亚甲基]-二氨基胍1.6g,加8ml 95%乙醇,室温搅拌,加0.5g羟乙基磺酸,搅拌2h,过滤,滤饼用95%乙醇洗,水洗,然后45℃干燥过夜,得1.4g产品(2,2’-双[[4-(异丙氧基)苯基]亚甲基]-二氨基胍.2-羟乙基磺酸盐,或名异丙氧苯胍2-羟乙基磺酸盐),产率66%。1HNMR(500MHz,d-DMSO)δ11.58(br,1H),δ8.26(s,2H),δ8.11(br,2H),δ7.82(d,4H),δ7.01(d,4H),δ4.69-4.74(m,2H),δ3.64(t,2H),δ2.62(t,2H),δ1.29(d,12H).

取游离的2,2’-双[[4-(异丙氧基)苯基]亚甲基]-二氨基胍1.6g,加8ml 95%乙醇,室温搅拌,加0.6g酒石酸,搅拌2h,过滤,滤饼用95%乙醇洗,水洗,然后45℃干燥过夜,得1.5g产品(2,2’-双[[4-(异丙氧基)苯基]亚甲基]-二氨基胍.酒石酸盐,或名异丙氧苯胍酒石酸盐),产率68%。1H NMR(500MHz,d-DMSO)δ10.15(br,1H),δ8.08(s,2H),δ7.65(d,4H),δ7.47(br,2H),δ6.94(d,4H),δ4.65-4.70(m,2H),δ4.18(s,1H),δ1.28(d,12H).

取游离的2,2’-双[[4-(异丙氧基)苯基]亚甲基]-二氨基胍1.6g,加8ml 95%乙醇,室温搅拌,加0.28g琥珀酸,搅拌2h,过滤,滤饼用95%乙醇洗,水洗,然后45℃干燥过夜,得1.6g产品(2,2’-双[[4-(异丙氧基)苯基]亚甲基]-二氨基胍.琥珀酸盐,或名异丙氧苯胍琥珀酸盐),产率76%。1H NMR(500MHz,d-DMSO):δ11.80(br,1H),δ8.00(s,2H),δ7.66(d,4H),δ6.92(d,4H),δ6.56(br,2H),δ4.63-4.68(m,2H),δ2.40(s,2H),δ1.28(d,12H).
实施例9:2,2’-bis[[4-(1-methylethoxy)phenyl]methylene]-carbonimidic dihydrazide.fumaric acid/2,2’-双[[4-(异丙氧基)苯基]亚甲基]-二氨基胍.富马酸盐的制备
取游离的2,2’-双[[4-(异丙氧基)苯基]亚甲基]-二氨基胍1.6g,加8ml95%乙醇,室温搅拌,加0.24g富马酸,搅拌过夜。过滤,滤饼用95%乙醇洗,然后45℃干燥过夜,得1.78g产品(2,2’-双[[4-(异丙氧基)苯基]亚甲基]-二氨基胍.富马酸盐,或名异丙氧苯胍富马酸盐),产率96.7%。1H NMR(500MHz,d-DMSO)δ8.06(s,2H),δ7.72(d,4H),δ7.19(br,2H),δ6.94(d,4H),δ6.60(s,1H),δ4.63-4.71(m,2H),δ1.28(d,12H).
实施例10:2,2’-bis[[4-(1-methylethoxy)phenyl]methylene]-carbonimidic dihydrazide.maleic acid/2,2’-双[[4-(异丙氧基)苯基]亚甲基]-二氨基胍.马来酸盐的制备
取游离的2,2’-双[[4-(异丙氧基)苯基]亚甲基]-二氨基胍1.6g,加8ml 95%乙醇,室温搅拌,加0.24g马来酸,搅拌过夜。过滤,滤饼用95%乙醇洗,然后45℃干燥过夜,得1.64g产品(2,2’-双[[4-(异丙氧基)苯基]亚甲基]-二氨基胍.马来酸盐,或名异丙氧苯胍马来酸盐),产率89.1%。1H NMR(500MHz,d-DMSO)δ8.17(s,2H),δ7.77(d,4H),δ7.54(br,2H),δ6.97(d,4H),δ6.02(s,1H),δ4.66-4.73(m,2H),δ1.28(d,12H).
实施例11:2,2’-bis[[4-(1-methylethoxy)phenyl]methylene]-carbonimidic dihydrazide.acetic acid/2,2’-双[[4-(异丙氧基)苯基]亚甲基]-二氨基胍.醋酸盐的制备
取0.25g醋酸溶于20ml水,加游离的2,2’-双[[4-(异丙氧基)苯基]亚甲基]-二氨基胍1.6g,室温搅拌,加2ml95%乙醇。搅拌过夜,过滤,滤饼水洗,然后45℃干燥过夜,得1.31g产品(2,2’-双[[4-(异丙氧基)苯基]亚甲基]-二氨基胍.醋酸盐,或名异丙氧苯胍醋酸盐),产率70.8%。1H NMR(500MHz,d-DMSO)δ7.98(s,2H),δ7.65(d,4H),δ6.91(d,4H),δ6.44(br,2H),δ4.63-4.67(m,2H),δ1.90(s,3H),δ1.28(d,12H).

1500只1日龄快大黄羽肉鸡按100只/组,分15组,在同一基础日粮中,加入不同种类的异丙氧苯胍的盐,试验期间自由采食及饮水、连续饲喂30天,统计生产性能及成活率。试验设计及生产性能试验结果见表1。从试验结果看不同异丙氧苯胍的盐对肉鸡生产性能的影响差异较大,其中乳酸盐、甲磺酸盐和2-羟乙基磺酸盐对肉鸡生产性能的提高最明显,效果优于盐酸盐,而其它盐的效果差于盐酸盐。

实施例16:不同剂量异丙氧苯胍盐对肉鸡生产性能的影响
800只1日龄快大黄羽肉鸡按100只/组,分8组,在同一基础日粮中,加入不同剂量的异丙氧苯胍甲磺酸盐,试验期间自由采食及饮水、连续饲喂30天,统计生产性能及成活率。试验设计及生产性能试验结果见表2。从试验结果看不同剂量异丙氧苯胍甲磺酸盐对肉鸡生产性能均有明显的提高,且对生产性能的改进呈剂量效应,剂量越高,效果越明显。而且高剂量对生产性能没有任何负面影响。

实施例17:异丙氧苯胍系列盐在猪料中的应用效果
120头体重相近的65日龄杜大长三元杂瘦肉型小猪随机分为6组,每组20头。按表3每组添加不同种类的异丙氧苯胍系列盐。试验期自由采食和饮水,试验期共28天。试验结果显示不同种类的异丙氧苯胍盐较对照组均能提高试验猪的增重,并对料比有不同程度的改善,但异丙氧苯胍的乳酸盐、甲磺酸盐和2-羟乙基磺酸盐效果优于盐酸盐的效果,其次为柠檬酸盐。试验分组及结果见表3。

实施例18异丙氧苯胍系列盐在鸭料中的应用效果
600只1日龄樱桃谷肉鸭随机分为6组,每组100只。按表4每组添加不同种类的异丙氧苯胍系列盐。试验期自由采食和饮水,试验期共28天。试验结果显示不同种类的异丙氧苯胍盐较对照组均能提高试验鸭的增重,并对料比有不同程度的改善,其中异丙氧苯胍的乳酸盐、甲磺酸盐和2-羟乙基磺酸盐效果最优,且优于盐酸盐的效果,其次为磷酸盐。试验分组及结果见表4。
