Abstract
Description
前記複数の捕捉領域を走査して、前記被検出物質に標識された蛍光物質を励起して得られる蛍光シグナルの検出により該被検出物質を検出する光学検出手段と、を少なくとも有するイムノアッセイシステムであって、
検出処理順序に関する情報が記憶された情報記憶媒体を備えるとともに、
前記情報記憶媒体に記憶された前記検出処理順序に関する情報を処理して前記検出処理順序を決める検出処理順序決定手段を有する。
以下、図1~図12を参照しながら本発明に係る第1実施形態のイムノアッセイ法、およびイムノアッセイシステムについて詳細に説明する。
第1の実施形態のイムノアッセイ装置100は、表面プラズモン共鳴(Surface Plasmon Resonance:SPR)現象を応用して検体溶液中に含まれる検体の検出を行うSPFS装置である。
まず、本実施形態のイムノアッセイ装置100に用いられるセンサーチップ1について説明する。
図1に示したように、イムノアッセイ装置100の筐体3には、センサーチップ1の情報を読み取るためのチップ情報読取手段(不図示)と、ユーザが所定の操作をして情報を入力するため入力手段(操作ボタン等)を有する操作部22と、ユーザに対して情報を表示するための表示部23、分析結果を出力するためのプリント出力部24等が設けられている。ユーザは、表示部23を確認しながら操作部22のボタンを操作することで、イムノアッセイ反応の開始処理、イムノアッセイ反応による分析の結果の出力処理(表示処理や印刷処理)、センサーチップ1から読み取ったセンサーチップ1の情報の確認または編集(値の変更等)等の各種操作が行えるようになっている。
チップ情報読取手段(一部不図示)は、センサーチップ1の情報を読み取って制御手段9に伝送する手段であり、チップ情報読取機構25と制御手段9の一部とから構成される。
投光光学系7は、従来のSPFS装置と同様であり、例えば図3に示したように、光源26と、光源26の照射角度を調整する角度走査機構27と、光源26を制御して光源26からの発する励起光6の強度等を調節するための光源制御機構28等とを備えている。
受光光学系8は、従来のSPFS装置と同構成の光学系であり、例えば、図3に示したように、光学レンズ群29と、励起光6の成分をカットするための励起光カットフィルタ30と、蛍光を受光して検出する光学検出手段21と、該光学検出手段21の動作を制御するためのセンサー制御機構31と、励起光カットフィルタ30を光学レンズ群29の光軸に配置または光軸から退避させる位置切り替え機構32等を有している。
センサーチップ搬送機構4(図3参照)は、制御手段9の制御により、スロット2Aに挿入されたセンサーチップ1を所定の位置に移動させる公知の手段である。
送液機構5は、図2及び図3に示したように、送液手段としての送液ポンプ19と、送液ポンプ19を駆動する送液ポンプ駆動機構33等と、を有している。
制御手段9は、一般的なパーソナルコンピュータ等であり、前述したセンサーチップ搬送機構4、送液機構5、投光光学系7、受光光学系8、および筐体3の各部(操作部22、表示部23、プリント出力部24等)等に接続されており、それらの動作を制御する機能を有する。
以下、第1実施形態のイムノアッセイ装置100を用いてイムノアッセイ法を行った例を、図9~図12を参照しながら具体的に説明する。
検出準備工程S1(図9参照)のステップS1-1では、図10(A)に示したように、ユーザにより、イムノアッセイ装置100の筐体3にある操作部22が操作されて、検査開始ボタンがONとされたか否かを判断する。YesであればステップS1-2に進み、Noであれば、ステップS1-1に戻る。
検出処理順序決定工程S2(図9参照)のステップS2-1(図10(B)参照)では、制御手段9が、検出処理順序(Dtc_Aln)の小さい順にデータ構造体の配列をソートする(図12(A)と(B)を対比して参照)。このソート処理は、図7に示した解離定数の相対値の情報(Rtv_val)または解離定数の値(Diss_cst)そのものの情報を用いて、前記相対値または解離定数の値が大きい順にソートする処理であってもよい。
送液反応工程S3(図9参照)のステップS3-1(図10(C)参照)の洗浄工程では、センサーチップ1を送液可能な位置に移動させて(図3参照)、送液機構5により緩衝液を送液して保湿剤を除去した後、洗浄液を送液する処理を行う。
検出処理工程S4(図9参照)のステップS4-1(図10(D)参照)では、整数値nを0に設定する。
出力工程S5(図9参照)では、上記検出した結果をプリント出力部24または表示部23に表示する出力処理を行う。
以下、図13~図27を参照しながら本発明に係る第2実施形態のイムノアッセイ法、およびイムノアッセイシステムについて詳細に説明する。
第2の実施形態のイムノアッセイ装置100Aは、表面プラズモン共鳴(Surface Plasmon Resonance:SPR)現象を応用して検体溶液中に含まれる検体の検出を行う表面プラズモン励起増強蛍光分析装置(SPFS装置)である。
まず、本実施形態のセンサーチップは、以下の点を除き第1実施形態のセンサーチップ1と同様であるので、その部分についての説明は省略する。
図13に示したように、イムノアッセイ装置100Aの筐体3Aには、チップ情報読取手段(不図示)と、操作部22と、表示部23、プリント出力部24等が設けられている。また、イムノアッセイ装置100Aの筐体3Aには、上述したように、センサーチップSC1~SC4を挿入するためのスロット2B~2Eが形成されている。この筐体3Aには、例えば、センサーチップSC1~SC4がスロット2B~2Eに挿入された状態か否かを検知する前述と同様の公知の検知手段がそれぞれ設けられている。
図13Aに示したように、制御手段9Aは、一般的なパーソナルコンピュータ等であり、センサーチップ搬送機構4A、送液機構5A、投光光学系7、受光光学系8、および筐体3Aの各部(操作部22、表示部23、プリント出力部24等)等に接続されており、それらの動作を制御する機能を有する。
データ構造体は、一般的なプログラムでいう構造体の配列として構成され、イムノアッセイ装置に挿入されたセンサーチップのチップ情報等を格納するためのものである。このデータ構造体は、以下の点を除いて図8に示す第1実施形態のデータ構造体と同じであるので共通部分の説明は省略する。
前記記憶手段10には、上述したように、各捕捉領域の検出処理の順序を決定するために用いられるデータベースaz1~az3が記憶されている。
モード表(図16参照)は、上記データベースaz1~az3とは異なるデータベースとして、上記記憶手段10に記憶されており、モード別の参照先のデータベース名が、少なくとも記憶されている。また、図16の例では、モード別に設定されている処理内容が備考として保存されている。
以下、本発明に係る第2実施形態のイムノアッセイ装置100Aを用いてイムノアッセイ法を行った例を、図13~図27を主に参照しながら具体的に説明する。
検出準備工程S1A(図20参照)のステップS1A-1では、図21(A)に示したように、ユーザにより、イムノアッセイ装置100Aの筐体3Aにある操作部22が操作されて、検査開始ボタンがONとされたか否かを判断する。YesであればステップS1A-2に進み、Noであれば、ステップS1A-1に戻る。
ステップS1A-2では、制御手段9Aが検知手段からの情報に基づいて、各センサーチップSC1~SC4が各スロット2B~2Eに挿入されているか否かを判断する。センサーチップが挿入されておりYesと判断された場合には、ステップS1A-3に進み、センサーチップが各スロット2B~2Eに挿入されておらずNoと判断された場合は、ステップS1A-1に戻る。
検出処理順序決定工程S2A(図20参照)のステップS2A-1では、図21(B)に示したように、センサーチップSC1~SC4に保存されている検出モード(「1」~「3」)、または保存されていない情報「-」(便宜的にデータがないことを「-」で示す)という情報に応じて、データベースaz1~az3が参照される、または参照されずに、各データ構造体が更新される。
送液反応工程S3A(図20参照)のステップS3A-1の洗浄工程(図21(c)参照)では、各センサーチップを送液可能な位置に移動させて(図13A参照)、送液機構5Aにより緩衝液を送液して保湿剤を除去した後、洗浄液を送液する処理を行う。その後、各センサーチップSC1~SC4を検出可能位置DTに移動させる(図13A参照)。
検出処理工程S4A(図20参照)のステップS4A-1(図22参照)では、整数値mを0に設定する。
出力工程S5A(図20参照)では、上記検出した結果をプリント出力部24に印刷処理させるか、または表示部23に表示する出力処理を行う。

Claims (14)
- 被検出物質を第1の捕捉体により捕捉する捕捉領域を互いに分離して複数配置したセンサーチップを用いて、該第1の捕捉体に捕捉された該被検出物質を個別に検出するイムノアッセイ法であって、
複数の捕捉領域は、捕捉する被検出物質の種類に応じて、異なる種類の第1の捕捉体を用いて形成されており、
複数の前記捕捉領域間の検出処理順序に関する情報に基づいて、該捕捉領域間の検出処理順序を決定する検出処理順序決定工程と、
該決定された検出処理順序に従って前記捕捉領域の各々を検出処理する検出処理工程と、
を有する、イムノアッセイ法。 - 前記検出処理順序に関する情報は、前記センサーチップに保存された情報、あるいは前記センサーチップに保存された情報に対応して参照される検出処理する装置に保存された情報である、請求項1に記載のイムノアッセイ法。
- 前記検出処理順序に関する情報は、前記被検出物質の解離定数に関する情報である、請求項1または2に記載のイムノアッセイ法。
- 前記被検出物質の解離定数に関する情報は、前記被検出物質と前記第1の捕捉体との間の第1解離定数か、前記被検出物質と、蛍光物質で標識された第2の捕捉体との間の第2解離定数である、請求項3に記載のイムノアッセイ法。
- 前記検出処理順序に関する情報は、前記被検出物質の種類に関する情報である、請求項1から4のいずれか1項に記載のイムノアッセイ法。
- 前記検出処理順序に関する情報は、前記検出処理順序を予め定めた情報である、請求項1から5のいずれか1項に記載のイムノアッセイ法。
- 前記検出処理順序に関する情報は、検出精度の必要性の高さを示す情報である、請求項1から6のいずれか1項に記載のイムノアッセイ法。
- 前記検出処理順序に関する情報は、前記蛍光シグナルの測定値が小さいと予測される順である、請求項1から7のいずれか1項に記載のイムノアッセイ法。
- 前記検出処理順序に関する情報が、前記センサーチップに保存された情報がない場合には、検出処理順序決定工程にて、検出処理工程での装置の動作が最小となる順に検出処理順序を決定する、請求項1に記載のイムノアッセイ法。
- 前記第1の捕捉体による前記被検出物質の捕捉、および前記第2の捕捉体による前記被検出物質の蛍光標識を、前記複数の捕捉領域において同時に行う、請求項1から9のいずれか1項に記載のイムノアッセイ法。
- 前記蛍光シグナルの検出処理は、表面プラズモン励起増強蛍光分光法により行う、請求項1から10のいずれか1項に記載のイムノアッセイ法。
- 被検出物質を捕捉する第1の捕捉体を固定した複数の捕捉領域を互いに分離して有するセンサーチップと、
前記複数の捕捉領域を走査して、前記被検出物質に標識された蛍光物質を励起して得られる蛍光シグナルの検出により該被検出物質を検出する光学検出手段と、
を少なくとも有するイムノアッセイシステムであって、
検出処理順序に関する情報が記憶された情報記憶媒体を備えるとともに、
前記情報記憶媒体に記憶された前記検出処理順序に関する情報を処理して前記検出処理順序を決める検出処理順序決定手段を有する、イムノアッセイシステム。 - 前記検出処理順序に関する情報は、前記センサーチップに保存された情報、あるいは前記センサーチップに保存された情報に対応して参照される検出処理する装置に保存された情報である、請求項12に記載のイムノアッセイシステム。
- 被検出物質を第1の捕捉体により捕捉する捕捉領域が互いに分離して複数配置され、被検出物質が第1の捕捉体に捕捉された状態で、被検出物質を個別に検出する処理が行われるセンサーチップであって、
複数の捕捉領域は、捕捉する被検出物質の種類に応じて、異なる種類の第1の捕捉体を用いて形成されており、
複数の前記捕捉領域間の処理順序に関する情報を記憶した情報記憶媒体を有する、センサーチップ。
