すなわち、 本発明は、 以下の通りである :
( 1 ) イソロイシン、 ロイシン、 及びバリンから選ばれる少なくとも 1種の分岐 鎖アミノ酸を有効成分とする、 A k t活性化抑制剤。
(2) 少なくともロイシンを含む、 (1) に記載の A k t活性化抑制剤。
(3) イソロイシン、 ロイシン、 及びパリンの 3種からなる、 (1) に記載の A k t活性化抑制剤。
(4) イソロイシン、 ロイシン、 及びパリンの重量比が、 1 : 1. 5〜2. 5 : 0. 8〜1. 7であることを特徴とする、 (3) に記截の A k t活性化抑制剤。
(5) 高インスリン血症を示すか又は高ィンスリン血症の危険性のある患者にお ける癌の発生及び 又は進展を抑制するために用いられる、 (1) 〜 (4) のいず れかに記載の A k t活性化抑制剤。 .

ところで、 癌は細胞の生と死のバランスが崩れた病態である。 すなわち、 正常 な細胞では DN Aに損傷が蓄積すると自らアポトーシスを誘導し'細胞死に至る力 癌細胞においてはアポトーシスに対して耐性を獲得し細胞死を免れていることが 明らかになつてきており、 これが癌発生進展の原因の 1つと考えられている。 ァ ポトーシスを制御する経路は、 TNF-ひなどのデスレセプター、 JUNK、 p 3 8などの MAPキナーゼ、 B e 1 -2フアミリータンパク質、 p 53など各種知ら れているが、 P I 3 K-Ak t経 も重要である。 B a d (Bel- 2 antagonist of cell death) は、 B c 1 -2や B e 1 - x Lと結合してそれらを不活性化することで アポトーシスを誘導するが、 Ak tは B a dの S e r l 36をリン酸化して不活 性化する。 また Ak tは、 J NKの上流である AS K 1 (apoptosis
signal-regulating kinase 1) の S r 8 3をリン酸化することでその下流で誘 導されるアポトーシスを抑制する。 また、 Ak tは膜上で活性化すると一部が核 に移行し、 FOXO 1, 3, 4をリン酸化して不活性化するが、 FOXOは、 F a sリガンドゃ B i m (Bcl-2 interacting mediator of cell death) といったァ ポトーシスを誘導する因子の転写を活性化するため、 Ak tによる FOXOの不 活性化は、 アポトーシスの抑制につながる。 また p 53や p 73は、 B a x、 P
uma s N o x aなどのアポトーシス促進因子の転写を活性化するが、 Ak tは p 5 3や p 7 3に対して抑制的に働くことが知られている。 すなわち、 p 5 3を 負に制御している MDM2 (mouse double minute 2) を Ak tがリン酸化して活 性を上昇させることや、 ρ 7 3のコアクチベータ一である YAP (yes-associated protein)を A k tはリン酸化して核外移行を促すことによりアポトーシス促進因 子の転写活性化が抑制される。 さらには、転写因子 N F- K Bは阻害因子である I κ Bが I KKによりリン酸化されることにより活性化されるが、 Ak tは I KK をリン酸化して活性化する。 N F- K Bは I A Pや A 1および B c 1 _xLを転写活 性化することでアポトーシスを抑制する. Proc Natl Acad Sci USA 94: 10057 10062)。 また、 Ak tは CREB (c AMP-responsive element binding protein) の S e r l 3 3をリン酸化して活性化することが知られているが、 CR EBはアポトー ス抑制因子である B c 1 - 2や M'c 1 -1の転写を活性化する

このように、 A k tは多くの段階、 多様なターグットでアポトーシスを抑制し ている。 したがって、 Ak tの活性化を抑制することは、 アポトーシスの抑制を 解除することになり、 癌発生抑制、 進展抑制につながることが期待できる。 よつ て、 本発明の Ak t活性化抑制剤は、 アポトーシスの促進、 癌発生または進展の 抑制に用いることもでき、 これらの態様も本発明の権利に包含される。
本発明の有効成分 (分岐鎖アミノ酸) 'である、 イソロイシン、 ロイシン及びバ リンは、 それぞれ、 L一体、 D—体、 D L—体のいずれも使用可能であるが、 好 ましくは、 L一体、 DL—体であり、 さらに好ましくは、 L一体である。
また、イソロイシン、 ロイシン、及びバリンは、それぞれ、遊離体のみならず、 塩の形態でも使用することができる。 塩の形態としては、 酸付加塩や塩基との塩 等を挙げることもでき、 イソロイシン、 ロイシン、 及びパリンの医薬品として許 容される塩を選択することが好ましい。
イソロイシン、 ロイシン、 及びパリンにそれぞれ付加して医薬として許容され る塩を形成する酸としては、 例えば、 塩化水素、 臭化水素、 硫酸、 リン酸等の無 機酸;酢酸、 乳酸、 クニン酸、 酒石酸、 マレイン酸、 フマル酸、 モノメチル硫酸 等の有機酸が挙げられる。 .
イソロイシン、 ロイシン、 及びパリンにそれぞれ付加して医薬として許容され る塩基を形成する塩基の例としては、 例えば、 ナトリウム、 カリウム、 カルシゥ ム等の金属の水酸化物または炭酸化物、 あるいはァ.ンモニケ等の無機塩基;ェチ レンジァミン、 プロピレンジァミン、 エタノールァミン、 モノアルキルエタノー ノレアミン、 ジァノレキノレエタノ一ノレアミン、 ジ タノーノレアミン、 'トリエタノーノレ ァミン等の有機塩基が挙げられる。
上記塩は水和物 (含水塩) であってもよい。 また、 水和物としては、 例えば 1 〜 6水和物などが挙げられる。 これらの塩、 水和物も本発明の権利に包含される ものである。
本発明の薬剤は、 イソロイシン、 ロイシン、 及ぴパリンのいずれか 1種以上が 分岐鎖アミノ酸として含有されていればよく、 イソロイシン、 ロイシン、 及びバ リンの 3種全てが含有されていてもよい。
好適な実施形態では、 本発明の薬剤は、 少なくともロイシンを含有する。
別の好適な実施形態では、 本発明の製剤は、 イソロイシン、 ロイシン、 及ぴバ リンの 3種全てを含有する。
イソロイシン、 ロイシン、 及ぴバリンの 3種全てを用いる場合、 イソロイシン と、 ロイシンと、 バリンの重量比は、 1 : 1 . 5〜2 . 5 : 0 . 8〜1 . 7であ り、 特に 1 : 1 . 9〜2 . 2 : 1 . 1〜1 . 3の範囲が好ましい。
本発明の薬剤の投与形態 ·剤型は、 経口投与、 非経口投与のいずれでもよく、
経口投与剤としては、 散剤、 顆粒剤、 カプセル剤、 錠剤、 チユアブル剤などの固 形剤、溶液剤、 シロップ剤などの液剤が、また、非経口投与剤としては、注射剤、 スプレー剤などが挙げられる。
本発明の薬剤は、 動物 (好ましくは、 ヒ ト等の哺乳動物) に投与することがで さる。 .
本発明の薬剤は、 経口、 注射、 あるいは局所投与等の任意の投与方法によって 投与することができる。
投与量は、 対象患者の年齢、 体重も.しくは病態、 剤形、 または投与方法などに よっても異なるが、 通常 1日量として、 イソロイシン 0. 5〜30. O g、 ロイ シン 1. 0〜60. O g、 ノ リン 0. 5〜30. 0 gを目安とする。 一般の成人 の場合、 好ましくは、 ー 量として、 ィソロイシン 2. 0〜1 0. 0 g、 口イシ ン 3. 0〜20. 0 g、 ノ リ ン 2. 0〜 : L O. 0 g、 より好ましくは、 イソロイ シン 2. 5~3. 5 g、 ロイシン 5. 0〜7., O g、 ノ リン 3. 0〜4. O gで あり、 アミノ酸 全体量としては 1日当たり 2. 0 g〜50. O g程度が好まし く、 これを 1〜6回、 好ましくは 1~3回に分割して投与する。
本発明の有効成分である分岐鎖アミノ酸の投与量 (摂取量) について算出する 際、 その値は、 本発明が目的とする疾患の治療または予防等の目的で使用される 薬剤の有効成分の量として前記の算定方法が決められているので、 これとは別目 的で、例えば、通常の食生活の必要から、あるいは別の疾患の治療目的のために、 摂取又は投与される分岐鎖ァミノ酸については、 前記算定に含める必要はない。 例えば、 通常の食生活から摂取される一日あたりの分岐鎖アミノ酸の量を、 前 記本発明における有効成分の 1日あたりの投与量から控除して算定する必要はな い。 . .
本発明の有効成分であるイソロイシン、 ロイシン、 及ぴバリンは、 それぞれが 単独で、 若レくは任意の組合わせで製斉 ljに含有されていてもよいし、 全てが 1種 の製剤中に含有されていてもよい。 別途製剤化して投与する場合、 それらの投与 経路、 投与剤形は同一であっても、 異なっていてもよい。 また各々を投与するタ
ィ ングも、 同時であっても別々であってもよい。 これらは、 併用する薬剤の種 類や効果なども考慮して、 適宜決定され得る。
即ち、 本発明の薬剤は、 2種以上の分岐鎖アミノ酸を含有する場合、 複数の分 岐鎖アミノ酸を同時に含有する製剤であってもよいし、 それぞれを別途製剤化し て併用するような併用剤であってもよく、 これらの形態全てを包含するものであ る。 特に、 同一製剤中に全ての分岐鎖アミノ酸を含有する態様が、 簡便に投与で きるため好ましい。
本発明において、 「重量比」 とは、 製剤中のそれぞれの成分の重量の比を示す。 例えば、 イソロイシン、 ロイシンおよび.バリンの各有効成分を 1つの製剤中に含 めた場合には、 個々の含有量の比であり、 各有効成分のそれぞれを単独でまたは 任意の組み合わせで複数製剤中に含めた場合には、 各製剤【こ含められる各有効成 分の重量の比である。 なお、 本明細書中において各アミノ酸の重量は、 遊離アミ ノ酸としてのものである。 .
また本発明において、 実際の投与量の比は、 投^対象 (即ち患者) あたりの各 有効成分 1回投与量あるいは 1日投与量の比である。 例えば、 イソロイシン、 口 イシンおよびパリンの各有効成分を 1つの製剤中に含め、 それを投与対象に投与 する場合には、 重量比が投与量比に相当する。 各有効成分を単独でまたは任意の 組み合わせで複数の製剤中に含めて投与する場合には、 1回あるいは 1日投与,し た各製剤中の各有効成分の合計量の比が重量比に相当する。
イソロイシン、 ロイシン、 およぴバリンは既に、 医薬 '食品分野において広く 用いられていて、 安全性は確立している。 例えば、 これらのアミノ酸を 1 : 2 : 1 . 2の比で含有する製剤の急性毒 '14 ( L D 50) は、 マウスの経口投与において 1 0 g / K g以上である。
本発明の薬剤は、 通常の方法によって、 散剤、 顆粒剤、 カプセル剤、 錠剤、 チ ユアブル剤などの固形剤、 溶液剤、 シロップ剤などの液剤、 または、 注射剤、 ス プレー剤などに製剤化することができる。
このような製剤は、必要に応じて、薬学的に許容される担体、例えば、賦形剤、
結合剤、 滑沢剤、 溶剤、 崩壊剤、 溶解補助剤、 懸濁化剤、 乳化剤、 等張化剤、 安 定化剤、 無痛化剤、 防腐剤、 抗酸化剤、 矯味矯臭剤、 着色剤などを配合される。 賦形剤としては、乳糖、ブドウ糖、 D—マンニトールなどの糖類、でんぷん類、 結晶セルロースなどのセルロース類などの有機系賦形剤、 炭酸カルシウム、 カオ リンなどの無機系賦形剤などが挙げられる。 .
結合剤としては、 α化デンプン、ゼラチン、アラビアゴム、 メチルセルロース、 カノレボキシメチノレセノレロース、 カノレポキシメチノレセノレロースナトリ ウム、 結晶セ ノレ口一ス、 D—マンニ ト—ノレ、 ト レノ、.ロース、 ヒ ドロキシプロピノレセノレロース、 ヒ ドロキシプロピルメチルセルロース、 ポリ ビ、ニルピロリ ドン、 ポリ ビニルアル コールなどが挙げられる。
滑沢剤としては、 ステアリン酸、 ステアリン酸塩などの脂肪酸塩、 タルク、 珪 酸塩類などが挙げられる。
溶剤としては、 精製水、 生理的食塩水などが挙げられる。
崩壌剤としては、 低置換度ヒ ドロキシプロピルセルロース、 化学修飾されたセ ルロースやデンプン類などが挙げられる。
溶解補助剤としては、 ポリエチレングリコール、' プロピレングリコール.、 トレ ハロース、 安息香酸ベンジル、 エタノール、 炭酸ナトリウム、 クェン酸ナトリウ ム、 サリチル酸ナトリウム、 酢酸ナトリウムなどが挙げられる。
懸濁化剤あるいは乳化剤としては、 ラウリル硫酸ナトリウム、 アラビアゴム、 ゼラチン、 レシチン、 モノステアリン酸グリセリン、 ポリ ビニルアルコール、 ポ リ ビエノレピロリ ドン、 カノレポキシメチノレセノレロースナトリ ウムなどのセノレロース 類、 ポリソルベート類、 ポリオキシエチレン硬化ヒマシ油などが挙げられる。 等張化剤としては、 塩化チトリウム、 塩化カリウム、 糖類、 グリセリン、 尿素 などが挙げられる。
安定化剤としては、 ポリエチレングリコール、 デキス トラン硫酸ナトリウム、 その他のァミノ酸類などが挙げられる。
無痛化剤としては、 ブドウ糖、 ダルコン酸カルシウム、 塩酸プロ力インなどが
挙げられる。
防腐剤としては、 パラォキシ安息香酸エステル類、 クロロブタノール、 ベンジ ノレアルコール、 フエネチルアルコール、 デヒドロ酢酸、 ソルビン酸などが挙げら れる。 '
抗酸化剤としては、 亜硫酸塩、 ァスコルビン酸などが挙げられる。
矯味矯臭剤としては、 医薬および食品分野において通常に使用される甘味料、 香料などが挙げられる。 '
着色剤としては、 医薬および食品分野において通常に使用される着色料が挙げ られる。
本発明の A k t抑制剤には、他の薬剤、例えば、糖尿病用剤、高脂血症治療剤、 高血圧用治療剤、肝臓病用剤、抗がん剤(例えば、 力.ルポプラチン、テガフール、 パクリタキセル、 酢酸リュープロレリンなど) などを配合してもよい。
以下の実施例により本発明をより具体的に説明するが、 実施例は本発明の単な る例示を示すも にすぎず、 本発明の範囲を何ら限定するものではない。 実施例
(実施例 1 )
S u b c o n f l u e n tに培養した HE K 29 3細胞をトリプシン処理レ、 DMEM+ 10 % F C S培地中に 1 χ 105 c e l l s /m 1で懸濁し、得られ た懸濁液を l w e l lあたり Ι Ο Ο μ Ιずつ 96w e l l l a t eにまいた, DMEM+ 1◦ %F C S培地で 1晚培養後、 コントロール群 (n = 4) は、 DM EM+ 1 0 %F C S培地に交換し、 5mM BCAA群 (n = 4) は、 イソロイ シン、 ロイシン、 及びバリンを重量比 1 : 2 : 1. 2で含有する 5'mM BCA A混合物を DMEM+ 10% FC Sに添加した培地に交換し、 さらに 3日間培 養した。 培養終了後、 C e l l C o u n t i n g K i t— 8 (和光純薬) を 用いてプロトコール通りに細胞数を測定した。
結果を、 コントロール群の OD値からバックグラウンドの OD値を引いた値を
1 0 0 %として、 % o f c o n t r o 1 として表す(図 1)。 図 1に示される ように、イソロイシン、ロイシン、及びパリンから構成される B CAA混合物は、 有意に HE K 2 9 3細胞の増殖を抑制した。
(実施例 2 )
S u b c o n f l u e n tに培養した H 4 I I E細胞をトリプシン処理し、 D MEM+ 1 0 %F C S培地中に 1 X 1 05 c e l l s /m 1で懸濁し、得られた 懸濁液を 1 w e 1 1あたり 1 m 1ずつ 2 4 w e 1 1 l a t eにまいた。 DM EM+ 1 0 %F C S培地で 4時間培養後、 以下の 5群に分けて、 1晚培養した: ( 1 ) DMEM F C S (—) 培地 (n = 2) ;
(2) DMEM F C S (-) +インスリン nM (n = 2) ;
( 3 ) DMEM F C S (-) +インスリン n'M+ロイシン 3 mM (n 2) ;
(4) DMEM F C S (-) +インスリ ン 0 nM (n = 2) ; .
( 5) DMEM , F C S (-) +インスリン O nM+ロイシン 3 mM (n = 2)。
培養終了後、 P a t h S c a n P h o s p h d -A k t l ( S e r 4 7 3 ) S a n d w i c h E L I SA k i t (C e l l S i g n a l i n g T e c h n o 1 o g y)を用いて、プロ トコ一ノレ通りにリン酸化 A k tを定量した.。 結果を図 2に示す。 A k tは、 インスリ ン 1 nM、 1 0 nM (図 2の (2) 及び (4)) で用量依存的に活性化されたが、 ロイシンは、 これらの活性化を抑制 した (図 2の (3) 及び (5.))。
(実施例 3 )
H4 I I E細胞を D-MEM+ 1 0 % F B S培地で 4 X 1 05個/ w e l 1で 1 2ゥエルプレートにまき、 ー晚培養した。 翌日 D- MEM F C S (―) 培地に交 換し、 さらにー晚培養した。
翌日 D-MEMから B C AAのみ除去した培地( iB C AA培地) に交換して 1 時間培養した後に、 B CAA添加条件では 2 mMの B CAAを添加してさらに 0.
5日き間培養した。その後 10 nMのインスリンあるいは 14 nMの I GF- 1を添 加して、 8時間後に細胞中の mRNAを抽出し、 c DNAを合成した後に、 以下 のプライマーを用いて R e a 1 -T i m e P CR法によりアポトーシス抑制遺伝 子 M c l - 1および I AP_1、 h o u s e k e e p i n g g e n eである R p 1 36の発現量を定量した。 号: 1)

結果は h o u e k e e i n g g e n e (R p 1 36) の発現量で割り返 して、 インスリンなし、 BCAAなし、 又は I GF— 1なし、 B CAAなしの時 の値を 1 00 %として0/ 0 o f c o n t r o lで表示した (図 3 ) 。 ィソロィシ ン、 ロイシン、 及びパリンから構成される BCAA混合物は、 アポトーシス抑制 遺伝子 Mc 1 - 1および I AP- 1の発現を抑制した。 産業上の利用可能性
本発明により提供される、 イソロイシン、 ロイシン、 及ぴバリンから選ばれる 少なくとも 1種の分岐鎖アミノ酸を有効成分とする Ak t活性化抑制剤は、 高ィ ンスリン血症を示すか又は高インスリン血症の危険性のある患者における癌の発 生及び 又は進展を抑制する。
本発明の薬剤は、 アミノ酸を有効成分とすることから、 安全性が高く、 副作用 がほとんどないため長期間にわたって投与可能であり、 高インスリン血症を示す か又は高ィンスリン血症の危険性のある患者が癌発生に至るまでの長期経過にお
け 予防または治療に有利に用いることができる。
本出願は、 日本で出願された特願 200 5- 227780 (出願曰 : 2005 年 8月 5日) を基礎としており、 その内容は本明細書に全て包含されるものであ る。
Claims
1. イソロイシン、 ロイシン、 及びパリンから選ばれる少なくとも 1種の分岐鎖 アミノ酸を有効成分とする、 Ak t活性化抑制剤。
2. 少なくともロイシンを含む、 請求の範囲 1に記載の A k t活性化抑制剤。
3. イソロイシン、 ロイシン、 及ぴバリンの 3種からなる、 請求の範囲 1に記載 の Ak t活性化抑制剤。
4. ィソロイシン、 ロイシン、 及びバリンの重畺比が、 1 : 1. 5〜2. 5 : 0. 8~1. 7であることを特徴とする、請求の範囲 3に記載の A k t活性化抑制剤。
5. 高インスリン血症を示すか又は高ィンスリン血症の危険性のある患者におけ る癌の発生及び/又は進展を抑制するために用いられる、 請求の範囲 1〜4のい ずれか 1項に記載の A k t活性化抑制剤。
6.前記高インスリン血症を示すか又は高インスリン血症の危険性のある患者が、 インスリン抵抗性、 糖代謝異常、 及び Z又は脂質代謝異常の症状を有する、 請求 の範囲 5に記載の A k t活性化抑制剤。
7.前記高インスリン血症を示すか又は高インスリン血症の危険性のある患者が、 肥満、 糖尿病、 非アルコ^"ル性脂肪性肝炎に罹患している、 請求の範囲 5に記载 の Ak t活性化抑制剤。
8. 前記癌が、 肝癌である、 請求の範囲 5〜7のいずれか 1項に記載の Ak t活 性化抑制剤。
9. アポトーシスを促進するために用いられる、 請求の範困 1〜4のいずれか 1 項に記載の A k t活性化抑制剤。
10. アポトーシスの促進がアポトーシス抑制遺伝子発現の抑制によるものであ る、 請求の範囲 9に記載の A k t活性化抑制剤。
1 1. アポト シス抑制遺伝子が Mc 1— 1または I AP— 1である、 請求の範 囲 1 0に記載の A k t活性化抑制剤。 '
1 2. 高インスリン血症を示すか又は高ィンスリン血症の危険性のある患者にお ける癌の発生及び Z又は進展を抑制するために用いられる、 請求の範囲 9に記載
の Ak t活性化抑制剤。
1 3. 前記高インスリン血症を示すか又は高インスリン血症の危険性のある患者 、 インスリン抵抗性、 糖代謝異常、 及びノ又は脂質代謝異常の症状を有する、 請求の範囲 1 2に記載の A k t活性化抑制剤。
14. 前記高インスリン血症を示すか又は高インスリン血症の危険性のある患者 力 肥満、 糖尿病、 非アルコール性脂肪性肝炎に罹患している、 請求の範囲 1 2 に記載の A k t活性化抑制剤。
1 5. 前記癌が、 肝癌である、 請求の範囲 1 2〜14のいずれか 1項に記載の A k t活性化抑制剤。
1 6. 有効量のイソロイシン、 ロイシン、 及びパリンから選ばれる少なくとも 1 種の分岐鎖アミノ酸を被験体に投与することを含む、 Ak t活性化の抑制方法。
1 7. Ak t活性化抑制剤の製造における、 イソロイシン、 ロイシン、 及びバリ ンから選ばれる少なくとも 1種の分岐鎖アミノ酸の使用。
1 8. イソロイ ン、 ロイシン、 及びパリンから選ばれる少なくとも 1種の分岐 鎖アミノ酸および医薬として許容される担体を含有してなる医薬钽成物、 ならび に当該組成物を A k t活性化の抑制に使用し得るまたは使用すベきであることを 記載した記載物を含む、 商業的パッケージ。 ·
