Abstract

The immunoenzyme assay kit for detecting IgG against herpes viruses types I and II in human blood serum (DIA-HSV 1/2-IgG) consists of immune sorbent and conjugate. The immune sorbent comprises the mixture of the purified envelope protein antigens of herpes viruses types I and II. The conjugate comprises peroxidase labeled anti-human IgG monoclonal antibodies. The sera being assayed are used at a 1:50 dilution.

Винахід належить до біотехнології, імунохімії та медицини і може бути використаний для виявлення імуноглобулінів класу їх проти вірусу герпесу 1-го і 2-го типів у сироватці крові людини; винахід може бути застосований у клініці для постановки та підтвердження діагнозу первинної або рецидивної герпетичної інфекції, а також для проведення сероепідемічних досліджень.The invention belongs to biotechnology, immunochemistry and medicine and can be used to detect immunoglobulins of class I against herpes virus 1 and 2 in human blood serum; the invention can be applied in the clinic to establish and confirm the diagnosis of primary or recurrent herpes infection, as well as to conduct seroepidemic studies.

В основу винаходу покладено завдання створити імуноферментну тест-систему на основі імуносорбенту з сумішшю очищених мембранних білків-антигенів вірусу герпесу 1-го і 2-го типів. Антигени сорбують на поверхні полістиролових планшетів. Досліджувані сироватки використовують в розведенні 1:50. Визначення проводять за відомою методикою |2| твердофазного імуноферментного аналізу. Використовуючи названі тест-системи, досліджували сироватки крові (по 20 достеменно позитивних та 100 достеменно негативних сироваток). Усі сироватки досліджували у двох повторностях кожною тест-системою за описаною методикою. Усі 20 позитивних сироваток показали наявність антитіл проти вірусу герпесу, що складає 10095 чутливості. З 100 негативних сироваток у пропонованій тест-системі виявилися справді негативними всі 100 (специфічність - 10090). В порівнюваній тест-системі з Росії з 20 достеменно позитивних сироваток 20 були позитивними (чутливість 10095), але з 100 негативних сироваток 2 сироватки були визначені як хибно-позитивні.Blood sera were examined using the named test systems (20 definitely positive and 100 definitely negative sera).
