Description of the invention
The method refers to biotechnology, namely to the methods of obtaining plants of grape ip migo.
с. СРСР Мо810160,There is a known method of growing sunflower plants, according to which embryos are isolated from the seeds and cultivated in a nutrient medium with the addition of growth regulators (A. s. USSR Mo810160,
Common features of the analogue and the claimed technical solution: the cultivation of the embryos of ip mygo on a nutrient medium with the addition of growth regulators. 70 The disadvantage of the analogue method is the low yield of plants due to the fact that the embryos receive mechanical damage when they are separated from the seeds and stress as a result of the change in the environment, which leads to suppression and slowing down of the development of the embryo.
Найбільш близьким по технічній сутності до корисної моделі, що заявляється, є, прийнятий як прототип, спосіб вирощування рослин із важкопророщуваного насіння, що включає культивування зародків з частиною 72 оболонки насіння іп уйго та добір стійких генотипів зародків під впливом селективного фактору живильного середовища, для чого недорозвинені зародки з частиною оболонки насіння послідовно пересаджують на живильні середовища з додаванням регуляторів росту, змінюючи їх зміст в залежності від стадії розвитку зародку (Патент України Мо17919 А, АОТНА/00; АОТНІ/04, 1997).The closest in technical essence to the useful model that is claimed is, accepted as a prototype, a method of growing plants from difficult-to-germinate seeds, which includes the cultivation of embryos with part 72 of the seed coat of ip uygo and the selection of resistant genotypes of embryos under the influence of the selective factor of the nutrient medium, for which underdeveloped embryos with part of the seed coat are successively transplanted into nutrient media with the addition of growth regulators, changing their content depending on the stage of embryo development (Patent of Ukraine Mo17919 A, AOTNA/00; AOTNI/04, 1997).
Common features of the prototype method and the claimed technical solution: cultivation of embryos together with a part of the seed coat of ip myo and planting them on nutrient media with the addition of growth regulators.
The disadvantages of the prototype method are the low yield of plants due to frequent transplants, which can lead to mechanical damage or infection of the material at one of the stages of cultivation, as well as the high labor intensity associated with the identification of seeds by the degree of embryo development and the sequential change in the content of various growth regulators in the nutrient medium.
The basis of a useful model is the task of improving the method of obtaining grape plants from low-fertility initial forms and, by selecting initial forms, determining the stages of cultivation on a nutrient medium of ip mygo and hormonal stimulation of the formation of shoots, to increase the yield of plants from seeds. The method is simple and reliable in execution.
The task is solved by the fact that in the method of obtaining grape plants from low-fertility initial forms, which includes the cultivation of embryos with a part of the seed coat and planting them on a nutrient medium with the addition of growth regulators, according to a useful model, the initial forms are first differentiated according to the class of seedlessness and the category of expression in seed culture, cultivation of ip mygo is carried out in two or three stages: germination of seeds, formation of plants and regeneration of plants, and the expediency of conducting the third stage is determined by the formation of callus, and as a growth regulator is used
Зо б-бензиламінопурин (БАП) в кількості, мг/дм: на першій стадії - 0,5-1,0; на другій стадії - не більше 0,5; - на третій стадії - 0,2-0,25.With b-benzylaminopurine (BAP) in quantity, mg/dm: at the first stage - 0.5-1.0; at the second stage - no more than 0.5; - at the third stage - 0.2-0.25.
Differentiation of initial forms of plants according to the class of seedlessness and the category of expression in seed culture allows to identify polyembryonic seeds, record the formation of multiple seedlings and guarantee their survival. When polyembryonic seeds germinate, plant development can occur in several ways (direct and indirect embryogenesis, hemogenesis, formation of a single plant). By changing the concentration of BAP in the nutrient medium, it is possible to regulate the path of development of multiple seedlings. Thus, one seed can give from one to hundreds of juvenile plants, which may have genetic heterogeneity.
An example of a specific implementation of the method.
Stenospermocarpic grape varieties and hybrids have been singled out, which have valuable properties that they cannot - 395 pass on to the offspring during normal generative reproduction due to low fertility and embryo degeneration. They drew up a hybridization plan, taking into account the reproductive features of the original forms. Like mother forms
Для проростання насіння використовували живильне с середовище (середовище Ніча, Ніч, 1969), яке мало наступний склад (концентрація, мг/дм З): мінеральні елементи МНАМО»з - 720, КМОз - 950, Ма5О,.7НЬО - 185, КНоРО, - 68, СасСі» - 166, Реб5О,.7НЬО - 28,ChK" varieties and hybrids belonging to the 1st class of seedlessness were used. Also, the data of the original forms by expression in the culture were taken into account. Seedless varieties and hybrids were used as pollinators. When selecting parental forms, the ability to transmit specific positive qualities to the offspring (yield, aroma, stability and (about) 50 others) was taken into account, depending on the task. Each crossing combination was represented by 5 inflorescences. sp. Grapes were collected during the period of physiological ripeness. The seeds were separated from the berries, washed in distilled water and placed in boxes for sterilization. Externally thin, deformed seeds were discarded. In the conditions of the laminated box, the seeds were sterilized for 40 seconds with 9095% ethyl alcohol, then for 8 minutes with 0.195% diocide and washed with sterile distilled water 3-4 times for 10 minutes. Then 99 seeds were cut. Seeds that did not have endosperm were discarded. For seed germination, a nutrient medium was used (Nicha medium, Nich, 1969), which had the following composition (concentration, mg/dm Z): mineral elements MNAMO»z - 720, KMOz - 950, Ma5O,.7НЭО - 185, KNoRO, - 68, SasSi" - 166, Reb5O,.7NYO - 28,

У 1095 аномальних проростків, які мають відхилення у розвитку сім'ядольних листків та підсім'ядольного коліна, на даному середовищі сформувався калюс, який пересадили на середовище, що містить БАП у концентрації 0,25мг/дму, внаслідок чого розвилося 495 рослин (третя стадія).In 1095 abnormal seedlings, which have deviations in the development of cotyledon leaves and subcotyledon knee, a callus was formed on this medium, which was transplanted to a medium containing BAP at a concentration of 0.25 mg/dmu, as a result of which 495 plants developed (third stage ).
As a result, 5295 seedlings developed into plants, which were propagated by micrografting and received 10-20 70 plants of each form. Adaptation was carried out according to known technology using a two-stage preparatory period followed by planting in a greenhouse. The first stage is replacing the foil with a cellophane film pressed with a ring, and cultivating plants in this state for 3-4 weeks; the second stage - cultivation of plants in open vessels with the addition of a weak solution of potassium permanganate for 3 weeks. At the end of May, the plants were transplanted into a greenhouse, shaded for 3 weeks.
