Claims (23)

(a) culturing the host cell according to claim 12 under conditions that enhance the expression of thermostable DNA polymerase; and
(b) isolation of thermostable DNA polymerase from the host cell or from the culture medium.

(г) разделение продуктов реакции со стадии (в); и
(д) определение последовательности нуклеиновой кислоты-мишени.19. A method for sequencing a target nucleic acid, comprising the following steps
(a) preparing an unusual nucleotide and a corresponding conventional nucleotide for a DNA sequencing reaction, wherein the unusual and corresponding conventional nucleotides are present in a ratio of less than about 1: 1;
(b) treating the reaction mixture from step (a) in the presence of a thermostable DNA polymerase according to any one of claims. 1-9 under conditions suitable for primer extension, to obtain primer extension products containing this unusual nucleotide;
(c) processing the primer extension products from step (b) under conditions suitable for hydrolysis of these primer extension products;
(d) separation of reaction products from step (c); and
(e) determining the sequence of the target nucleic acid.
