Abstract
Description
The invention relates to veterinary medicine, specifically to methods for the treatment and prevention of infectious diseases, in particular staphylococcosis of chickens, and can find application in industrial poultry farms, veterinary laboratories and veterinary research institutions.
Bacterial infections of the skeletal system cause serious economic losses in poultry production worldwide. Among infectious factors, staphylococci are one of the main bacterial pathogens that contribute to the occurrence of skeletal infections in birds. Although Staphylococcus aureus is the leading causative agent of avian staphylococcosis, other staphylococcal species such as S. agnetis, S. cohnii, S. epidermidis, S. hyicus, S. simulans also have a high frequency of isolation from affected bones and joints. Staphylococcosis is characterized by a low percentage of morbidity and mortality, but the culling of birds is always increasing. Treatment of staphylococcal infections with antimicrobial drugs is effective, however, due to the high rate of acquisition of resistance of staphylococci to drugs of various pharmacological groups, it is necessary to conduct antibiotic sensitivity tests of the pathogen.
- 2012. - №7. - С. 35-37)There are various methods used to treat patients with staphylococcal infection of birds, where 100 mg/ml (10%) enrofloxacin and 1,000,000 IU/ml colistin (Enronit OR) are used as active ingredients (Toropov A.A. et al. Prevention of infectious diseases diseases with the drug Enronit OR. - Poultry farming. - 2012. - No. 7. - P. 35-37)
- 2015. - №9. - С. 18-19.).Sulfonamides, lincomycin, gentamicin, chlortetracycline, chloramphenicol, biovit-80 and combinations are used, for example, the drug lenovit - the synergistic effect of chloramphenicol, norsulfazole and chlortetracycline at a dose of 2.5 kg/t of feed for 10-12 days. (N.V. Kozhemyaka Prevention and treatment of major bacterial diseases of poultry. Effective animal husbandry. - 2015. - No. 9. - P. 18-19.).
- 2019. - №6. - С. 12-15.).The drug Florox OR based on florfenicol is given to birds individually or in a group with water at a daily dose of 20 mg of florfenicol per 1 kg of body weight, daily for 3 days (Balyshev A.V. et al. Florox OR in the treatment of birds with bacterial infections. - Veterinary Science. - 2019. - No. 6. - pp. 12-15.).
(Патент RU 2570389).There is also a known method for treating farm animals and birds with an antibacterial pharmaceutical composition which includes a compound from the group of fluoroquinolones and apramycin sulfate in the following ratio (in parts by weight): compound from the group of fluoroquinolones: apramycin sulfate 1:2-1:10. (Patent RU 2570389).
(Патент RU 2554797).As a treatment for infectious diseases of birds, a drug is used, which contains compounds from the fluoroquinolone group and from the pleuromutilin group in a ratio (in parts by weight) of 1:3.0-1:225. The claimed preparation contains tiamulin hydrogen fumarate or tiamulin as pleuromutilins. Sugar is used as excipients - powdered sugar, or glucose, or fructose, or dextrose, or lactose, or lactulose. (Patent RU 2554797).
- 2016. - №8. - С. 15-18).Lexoflon OR is administered orally individually or in a group with water for drinking birds at a dose of 1 ml per 20 kg of animal weight (7.5 mg of levofloxacin per 1 kg of animal weight), for 3-5 days. It has a wide spectrum of antibacterial activity and is capable of destroying strains of pathogens resistant to enrofloxacin (Lysko S.B. et al. Antibacterial properties of Lexoflon OR - a drug based on a new generation fluoroquinolone. - Veterinary Medicine. - 2016. - No. 8. - P. 15- 18).
(Mosleh N. etall. Comparative evaluation of therapeutic efficacy of sulfadiazine-trimethoprim, oxytetracycline, enrofloxacin and florfenicol on Staphylococcus aureus-induced arthritis in broilers. - Br Poult Sci. - 2016. - №57. - Р. 179-84).A number of authors have conducted a comparative assessment of the therapeutic effectiveness of drugs for infectious arthritis of birds: sulfadiazine-trimethoprim 250 ml/1000 l of drinking water, oxytetracycline 20% 1 mg/l of drinking water, florfenicol 10%, 1/1000 IV in drinking water, enrofloxacin 10% 1/1000 IV in drinking water. As a result, it was found that sulfadiazine-trimethoprim showed higher therapeutic efficacy according to most of the analyzed criteria (Mosleh N. etall. (Nematollahi F et all. Effect of prophylactic administration of vitamin C in chickens with staphylococcal septic arthritis. - Vet Med Sci. - 2022. - №8. - Р. 245-253).Prophylactic administration of vitamin C (15 g/100 L of water), especially over a long period, can attenuate oxidative stress and histopathological changes caused by staphylococcal arthritis in chickens (Nematollahi F et all. Effect of prophylactic administration of vitamin C in chickens with staphylococcal septic arthritis. - Vet Med Sci. - 2022. - No. 8. - R. 245-253).
(Патент RU 2227741).To prevent infectious diseases of animals and birds caused by gram-positive microflora, a drug containing clindamycin, spectinomycin or an aminoglycoside antibiotic, as well as solvents or fillers, stabilizers and preservatives is used in the following ratio of components, wt. %: clindamycin 0.25-90.0, spectinomycin or aminoglycoside antibiotic 2.5-90.0, solvents or fillers, stabilizers, preservatives the rest. The drug has high preventive and therapeutic effectiveness and a wide spectrum of antimicrobial action. (Patent RU 2227741).
(Патент 2019188).For the treatment of avian staphylococcosis, the proteolytic enzyme lysoamidase is used as an antimicrobial drug, which is administered first in hatchers on the 19th - 20th day of incubation after transfer of embryos for hatching, and then 1 hour before sampling chickens and after sorting them indoors, at a dose of 1800 -450 LE/m 3 , with a total exposure in all cases of 30 - 60 minutes. (Patent 2019188).
The objective of the invention was to expand the arsenal of methods for treating experimental staphylococcosis in chickens.
The problem is solved by the proposed method of treating experimental staphylococcosis in chickens, including oral administration of a drug for five days, and a combination of a sorbent enriched with montmorillonite and water-soluble vievitin is used as a drug. The specified drug is administered in mixtures with food in doses: vievitin at concentrations of 100-300 mg/kg of feed together with a sorbent at a concentration of 4000 mg/kg of feed.
The technical result is that the complex use of vievitin in combination with an enriched montmorillonite-containing sorbent increases the effectiveness of treatment of experimental staphylococcosis in chickens, the risks of overdose are reduced, the rapid recovery of sick animals, and the reduction of vievitin consumption and its side effects on the body.

Сорбент является продуктом обогащения глинистого сырья месторождений Белгородской области (Патент на изобретение №2471549).The sorbent is a product of enrichment of clayey raw materials from deposits in the Belgorod region (Patent for invention No. 2471549).
Giving selectivity to the sorbent by immobilizing specific vievitin ligands on its surface is a new direction in sorption therapy. The resulting loosely immobilized vievitin ligands will be quite easily desorbed from the surface of the sorbent during enterosorption. In this case, the sorbent functions as a carrier for delivery, dosed release and prolongation of the action of bioactive ligands, and also additionally performs the function of protecting immobilized ligands from inactivating factors of the external (during long-term storage) and internal (for example, gastric juice) environment. In addition, in this embodiment, the sorbent-immobilized vievitin system has a certain buffer capacity, that is, it works as a depot from which the body, as needed, selects an antibiotic for its needs in the required quantities, thereby reducing the risk of overdose.
The adsorption capacity of certain types of montmorillonite-containing clays is due to their layered structure. This kind of sorbents usually have a large specific surface area - up to several hundred m2/g. The distance between the microscopic layers that make up the structural basis of clay is a fraction of a nanometer. If you increase this distance, you can significantly increase the specific surface area of the clay and thereby its adsorption properties.

The experiment was carried out on 15-day-old Hisex Brown cross chickens. The research involved 300 chickens, which were divided into 6 groups of 50 birds each. Experimental staphylococcosis was reproduced by intraperitoneal injection of Staphylococcus aureus at a concentration of 3.0x10 8 CFU/0.5 ml (2 McFarland/0.5 ml). Chickens were prescribed vievitin and sorbent orally for 5 days in a mixture with feed. The concentration of sorbent in the feed was the same for all groups and equaled 4000 mg/kg of feed. The concentration of vievitin in each group was different. The chickens of the first group were given food containing 300 mg/kg of feed, the chickens of the second group - 200 mg/kg of feed, the third - 100 mg/kg of feed, the fourth - 50 mg/kg of feed and the fifth - 25 mg/kg of feed. Chickens of the sixth (control) group were not prescribed the drug.

The comparative effectiveness of various treatment methods was studied experimentally. The therapeutic effectiveness of various concentrations of vievitin, administered both separately and in a mixture with a sorbent at a concentration of 4000 mg/kg of feed, was compared. Chickens of the first group were prescribed vievitin at a concentration of 300 mg/kg of feed and a sorbent at a concentration of 4000 mg/kg of feed; chickens of the second group were prescribed only vievitin at a concentration of 300 mg/kg of feed. Chickens of the third group were prescribed vievitin at a concentration of 200 mg/kg of feed and a sorbent at a concentration of 4000 mg/kg of feed; chickens of the fourth group were prescribed only vievitin at a concentration of 200 mg/kg of feed. Chickens of the fifth group were prescribed vievitin at a concentration of 100 mg/kg of feed and a sorbent at a concentration of 4000 mg/kg of feed; chickens of the sixth group were prescribed only vievitin at a concentration of 100 mg/kg of feed. Chickens of the seventh group were prescribed vievitin at a concentration of 50 mg/kg of feed and a sorbent at a concentration of 4000 mg/kg of feed; chickens of the eighth group were prescribed only vievitin at a concentration of 50 mg/kg of feed. Chickens of the ninth group were prescribed vievitin at a concentration of 25 mg/kg of feed and a sorbent at a concentration of 4000 mg/kg of feed; chickens of the tenth group were prescribed only vievitin at a concentration of 25 mg/kg of feed.
4-6%.The table data shows that high activity of the drug was observed in groups where chickens were prescribed vievitin at concentrations of 300, 200, and 100 mg/kg of feed in combination with a sorbent at a concentration of 4000 mg/kg of feed. The incidence in the experimental groups was only 4-6%.
12-14%.When comparing the effectiveness of the composite drug (vievitin in combination with a sorbent) and only vievitin in the above groups, we see that in the second, fourth and fifth groups, which were prescribed the drug without a sorbent, the number of sick chickens increased to 12-14%.
В восьмой группе цыплят, где назначали только виэвитин в концентрации 50 мг/кг корма, заболело 10 цыплят (20%), причем 4 из них пали (40% по отношению к заболевшим).Similar results were obtained in other experimental groups. So, in the seventh group, 7 chickens (14%) fell ill, of which 2 (29% in relation to the sick) died. In the eighth group of chickens, where only Vievitin was prescribed at a concentration of 50 mg/kg of feed, 10 chickens (20%) fell ill, and 4 of them died (40% of the sick ones).
In the ninth group of chickens that were prescribed vievitin at a concentration of 25 mg/kg of feed and sorbent at a concentration of 4000 mg/kg of feed, the incidence rate was 28% (14 chickens), of which 4 died, which is 29% in relation to sick chickens. In the group where only Vievitin was prescribed at a concentration of 25 mg/kg of feed, 17 chickens (34%) fell ill, and the mortality rate was 35% in relation to the sick.
Comparing the results of studies in groups where vievitin was prescribed together with a sorbent and in groups of chickens in which vievitin was used separately from the sorbent, it should be noted that better treatment results were obtained from the use of the drug composition.

The experiment was carried out on 30-day-old Hisex Brown cross chickens.
These studies showed that the lowest incidence was in the groups of chickens that were prescribed vievitin at concentrations of 300, 200, 100 mg/kg of feed and sorbent at a concentration of 4000 mg/kg of feed. Among the chickens who were prescribed the drug at a dose of 300 mg/kg, one chicken fell ill. When Vievitin was used at a dose of 200 mg/kg of feed, there were no sick chickens. In the group of chickens that were prescribed Vievitin at a dose of 100 mg/kg of feed, three chickens fell ill.
В этой группе пал один цыплёнок (14.3% по отношению к заболевшим).When Vievitin was prescribed at a concentration of 50 mg/kg of feed, 7 (14%) chickens became ill. In this group, one chicken died (14.3% of the sick ones).
В этой группе заболело 14 (28%) цыплят, из которых 3 - пало (21,4% по отношению к заболевшим).The highest incidence was observed in the group of chickens that were prescribed Vievitin at a concentration of 25 mg/kg of feed and sorbent at a concentration of 4000 mg/kg of feed. In this group, 14 (28%) chickens fell ill, of which 3 died (21.4% of the sick ones).
Падеж в данной группе составил 31,6% по отношению к заболевшим (6 цыплят).In the control group, 19 (38%) chickens became ill. The mortality rate in this group was 31.6% in relation to the sick (6 chickens).
