Технология получения Scm A, Scm В основана на культивировании штамма-продуцента КММ 9044 и извлечении субстанции из культуральной жидкости (КЖ) указанного штамма методом твердофазной экстракции и жидкостной хроматографии. Культивирование осуществляли при 28°С в течение 5 сут на термостатируемой качалке Innova 40 (New Brunswick Scientific) при 150 об/мин.2. Growing seed material of the 1st generation (first inoculum). 4. Культивирование в ферментере. Культивирование проводили в ферментере BIOSTAT A plus (Sartorius BBI Systems) объемом 2 л. В ферментер, содержащий 1350 мл стерильной среды SY, вносили посевной материал в объеме 10% (150 мл культуры на 1.5 л среды) и 150 мл стерильного раствора глюкозы с концентрацией 20 г/л (конечная концентрация 2 г/л). Культивирование проводили при температуре 28°С. Концентрацию растворенного кислорода (pO2) поддерживали на уровне 90%, скорость перемешивания составила 300 об/мин, количество стерильного воздуха, подаваемого в аппарат, - 1.5 л/мин, рН среды поддерживали в интервале 6.8 титрованием 25% раствором аммиака. Пеногаситель силикон (0.01%) вносили в конце первого дня культивирования. Продолжительность ферментации составила 43 ч.4. Cultivation in a fermenter. The concentration of dissolved oxygen (pO2) was maintained at 90%, the stirring speed was 300 rpm, the amount of sterile air supplied to the apparatus was 1.5 L/min, and the pH of the medium was maintained in the range of 6.8 by titration with 25% ammonia solution. Silicone defoamer (0.01%) was added at the end of the first day of cultivation. The duration of fermentation was 43 hours.
5. Отделение КЖ от биомассы. Выращенную культуру центрифугировали на скорости 10000 rpm (центрифуга Beckman J2-21), после чего супернатант фильтровали. Картридж промывали 150 мл ацетонитрила (АЦН) и активировали 150 мл дистиллированной воды с помощью перистальтического насоса Masterflex Easy-Load 11 при скорости потока 15 мл/мин. При той же скорости наносили КЖ; по окончании картридж промывали 150 мл воды. Картридж можно использовать повторно, но не более 5 раз.7. Application of CL on a polymeric sorbent. The cartridge was washed with 150 ml of acetonitrile (ACN) and activated with 150 ml of distilled water using a Masterflex Easy-Load 11 peristaltic pump at a flow rate of 15 ml/min. QOL was applied at the same speed; when finished, the cartridge was washed with 150 ml of water. The cartridge can be reused, but not more than 5 times.

Содержание АЦН в элюенте было задано по следующей программе: начальный состав - 5%; 1 мин - 45%, 10 мин - 45%, 11 мин - 100%, 22 мин - 100%. Состав элюата контролировали с помощью УФ детектора при длине волны 290 нм. УФ профили имели два пика: мажорный пик в области 5-6 мин, отвечающий сумме нецелевых соединений, и минорный пик в области 13-15 мин, содержащий целевые соединения Scm А и Scm В, УФ спектр которых имеет максимум при 290 нм. Данную фракцию объемом около 30 мл собирали с помощью коллектора фракций.8. Elution from the cartridge using a flash chromatograph. 9. Очистка субстанции. Очистку Scm А и Scm В проводили методом ВЭЖХ (хроматограф Шимадзу Нексера). Весь объем фракции после картриджа помещали в колбу роторного испарителя (Buchi Rotavapor), концентрировали примерно в 5 раз. К раствору добавляли раствор 0.1% уксусной кислоты - половину от полученного объема (в конкретном случае 30 мл концентрировали до 6 мл и добавляли 3 мл раствора уксусной кислоты). При наличии осадка раствор центрифугируют на скорости 10000 rpm, супернатант переносят в пробирки. Разделение проводят колонке полупрепаративного размера на сорбенте типа С18, фракция 5 или 10 мкм, при том же составе элюента, температуре термостата колонки и длине волны УФ детектора, как в п. 5. Расходы элюента и объем пробы, элюируемой за один цикл разделения, выбирают в зависимости от размера колонки. На колонке 9.2 х 150 мм проводили разделения порциями по 1.5 мл при скорости потока 3 мл/мин. Продолжительность каждого разделения составляет 20 мин. Для очистки всего объема фракции требуется повторить операцию несколько раз, в данном случае 6 раз.9. Purification of substance. Scm A and Scm B were purified by HPLC (Shimazu Necker chromatograph). Елякова, Arthrobacter sp. 21022 (АТСС) и Mycobacterium smegmatis АТСС 700084 из коллекции Lytech Co. Ltd.For testing, strains of Micrococcus sp. Минимальные ингибирующие концентрации (МИК) бактерий определяли серийными двукратными разведениями (0.1-10000 нг/мл) в бульоне 2YT (BD, США). МИК определяли, как наименьшую концентрацию, препятствующую росту тестируемого организма в 96-луночном планшете после 16 ч культивирования. Для Mycobacterium smegmatis МИК определяли через 60 ч культивирования. Минимальную бактерицидную концентрацию (МБК) определяли, как наименьшую концентрацию, препятствующую повторному культивированию тест-организма через 24 ч инкубации. Длительное культивирование приводит к разложению Scm и к завышенным значениям МИК. Временной ход роста бактерий отслеживали при 800 нм с использованием многорежимного считывающего устройства Varioskan Flash (Thermo Fisher Scientific, Уолтем, Массачусетс, США).Minimal inhibitory concentrations (MICs) of bacteria were determined by serial twofold dilutions (0.1–10000 ng/mL) in 2YT broth (BD, USA). MIC was defined as the lowest concentration that interferes with growth of a test organism in a 96-well plate after 16 hours of culture. For Mycobacterium smegmatis, MICs were determined after 60 hours of cultivation. The minimum bactericidal concentration (MBC) was defined as the lowest concentration preventing re-cultivation of the test organism after 24 hours of incubation. Prolonged cultivation results in Scm degradation and inflated MIC values. The temporal course of bacterial growth was monitored at 800 nm using a Varioskan Flash multimode reader (Thermo Fisher Scientific, Waltham, MA, USA).
Оба варианта структур Scm А (I) и Scm В (II) имеют противомикробную активность, причем для репортерного штамма Micrococcus sp. KMM 1467 величины минимальных ингибирующих концентраций (МИК) были на уровне нг/мл: 6 нг/мл для Scm А и 2 нг/мл для Scm В, т.е. значения для эпокси-производного в несколько раз ниже, чем значения для хлор-производного.Both variants of structures Scm A (I) and Scm B (II) have antimicrobial activity, and for the reporter strain Micrococcus sp.
