Abstract
Description
The invention relates to the field of veterinary medicine, and in particular to methods for the prevention of respiratory infections of broiler chickens.
Respiratory infections account for at least 40% of the total number of infectious diseases of birds, in the etiology of which pathogenic and conditionally pathogenic microorganisms play a leading role. In most cases, respiratory infections occur in associations, which complicates their diagnosis, reduces the effectiveness of anti-epizootic measures and causes significant economic damage to poultry enterprises.
М., 1977. с. 184).Known methods for the prevention of respiratory infections of birds of bacterial etiology using broad-spectrum antimicrobials - antibiotics, sulfonamides and nitrofurans (Mozgov I.E. Pharmacology. M .: Agropromizdat, 1985. P. 338-380; Aerosol therapy for respiratory diseases. 7 N . S. Pilipchuk [et al.] Kiev, 1988. p. 113), complex antibiotics (see patent RU No. 2287989, published on November 27, 2006 bull. No. 33 A61K 31/22, A61K 31/65, A61K 38 / 12, A61P 31/00, A61P 31/04, 02/25/2004; No. 2227741, published April 27, 2004 bulletin No. 12 A61K 31/00, A61K 31/7036, A61P 31/04) and chemical disinfectants (Vashkov B .And. A antimicrobial agents and disinfection methods for infectious diseases. M., 1977. p. 184).
However, their use leads to the development of drug resistance in pathogens, the development of dysbacteriosis in birds, a decrease in natural resistance and post-vaccination immunity, and a deterioration in the quality and environmental safety of poultry products.

However, the known methods of using propolis are intended for the treatment and prevention of a specific disease - bronchopneumonia and only in calves, piglets, and in birds - only to increase their productivity. The prophylactic effect of propolis tincture in respiratory infections of broiler chickens has not been studied.
The technical result of the invention is the creation of a new highly effective, environmentally friendly way to prevent respiratory infections of boiler chickens, as well as expanding the range of products for the prevention of infectious diseases of birds of bacterial etiology.
The claimed technical result of a method for the prevention of respiratory infections of broiler chickens includes feeding a 10% propolis tincture to a bird at a dose of 1 ml / 1 l of water in age periods of 1-5, 14-18, 25-30 days, an additional aerosol treatment of the house air 1 once a day with 10% propolis tincture diluted with water 1:20 500 ml / 100 m 3 of the room for 60 minutes for 1, 7, 14, 21-22, 28-29, 35-36 days of cultivation.
№31, A23L 1/076).Propolis is a biologically active substance, it is a solid mass of brown-brown color with a greenish tint, it is soluble in alcohol. The composition of propolis includes: a mixture of resins and balms 55%, wax 30%, essential oil 10%, pollen 5%, vitamins A, C, group B and other trace elements. Propolis contains cinnamon, cinnamic and benzoic acids, tannins, etc. Glycocol, aspartic and glutamic acids, alanine, tryptophan, phenylalanine, leucine were found in its composition. More than 20 compounds are isolated from propolis, which are represented by three groups of biologically active substances: acids, polyphenols and compounds of an isoprenoid structure (Method for preparing an aqueous extract of propolis: see patent RU No. 2402923 publ. 10.11.2010; Bull. No. 31, A23L 1/076 )
In the experiments we used a 10% alcohol tincture of propolis produced by Hippocrates LLC, Russia, Samara.
A dilution of propolis tincture 1:20 and exposure for treatments are established on the basis of studies. The choice of age periods for aerosol treatments is due to the fact that, with an increase in the age of the bird, pathogenic, conditionally pathogenic microorganisms accumulate in the air of the house and the risk of respiratory infections increases many times. The use of aerosol treatments is associated with a violation of the humidity regime. Increased air humidity in the house can provoke the development of a number of pathological conditions, and especially the respiratory system. The optimal amount of sprayed drug, which has a preventive effect on birds and the environment, while not increasing the humidity of the house is 500-1000 ml / 100 m 3 of the room. The high bactericidal activity of propolis tincture made it possible to use it for aerosol treatments at the rate of 500 ml / 100 m 3 of the room. When an aerosol is inhaled, it affects a large surface of the mucous membranes, which creates conditions for increased activity of the drug and, accordingly, a more rapid development of the pharmacological effect. The drug goes directly to those tissues where it should have the maximum effect - the mucous membranes of the respiratory tract. In addition, it has a bactericidal effect on the microflora of the air.
Together with aerosol treatment, propolis tincture was drunk at the dose established on the basis of the studies. The choice of periods for watering is due to the fact that in chickens of the given age, against the background of an unformed immune system, numerous technological stresses and vaccinations, natural resistance decreases and their body is most susceptible to respiratory diseases. It is during these periods that poultry enterprises use antibiotics to prevent respiratory infections.
The proposed method for the prevention of respiratory infections of broiler chickens reduces the number of pathogenic, conditionally pathogenic microorganisms in the air of the premises for their maintenance and on the mucous membrane of the larynx of birds, activates the immune system, metabolism, which increases safety by 2-3%, live weight by 236 , 6-342.7 g, profitability - by 18.3% and allows you to get safe poultry products without the use of chemicals.
The proposed method is confirmed by in vitro experience, experimental and production experiments on broiler chickens of the cross "Ross-308", performed in the poultry farm and the department of veterinary medicine of the poultry SibNIIP-branch of the Federal State Budget Scientific Institution "Omsk ANC".
Example 1. The study of the bactericidal properties of tincture of propolis in vitro on pathogens of respiratory infections in birds
На основании микробиологического мониторинг установлено, что доминирующими микроорганизмами при респираторных инфекциях птиц являются Escherichia coli, Staphylococcus spp. Streptococcus spp., Enterobacter agglomerans (Гофман A.A., Лыско СБ., Сунцова О.А. Н.Э. - 2016. - Т. 225. №1.. - С. 25-28).Based on microbiological monitoring, it was found that the dominant microorganisms in respiratory infections of birds are Escherichia coli, Staphylococcus spp. Streptococcus spp., Enterobacter agglomerans (Hoffmann AA, Lysko SB., Suntsova O.A. et al., Species composition of pathogens of infectious diseases of birds with pathology of the respiratory tract of birds // Scientific notes of the Kazan State Academy of Veterinary Medicine named after N.E.Bauman. - 2016. - T. 225. No. 1 .. - S. 25-28).
When studying in a comparative aspect the bactericidal properties of the antibiotic tilmipul and 10% propolis tincture by serial dilutions on the pathogens of respiratory diseases of birds, it was found that propolis tincture had the highest bactericidal activity (table 1). So, the minimum inhibitory concentration (MIC) of propolis tincture is higher than the antibiotic tilmipul for cultures of Escherichia coli, Enterobacter agglomerans and Streptococcus spp. 2 times, in cultures of Staphylococcus aureus - 64 times. Thus, a 10% tincture of propolis has a high antibacterial activity on pathogens of respiratory diseases of birds, which exceeds the activity of the antibiotic by 2 or more times, which makes it one of the most promising, effective and at the same time environmentally friendly means for the prevention of respiratory infections of birds.
The most effective dilution of propolis tincture and exposure for aerosol treatment of house air were selected. In vitro, a 10% tincture of propolis was diluted with water in a ratio of 1:15, 1:20, 1:25. Test cultures of Escherichia coli, Staphylococcus aureus, Streptococcus spp, Enterobacter agglomerans were introduced. After exposure of 30, 60, 90 minutes, the viability of microorganisms was assessed by plating on differential media.
Table 2 shows that propolis tincture at a 1:25 dilution showed antibacterial activity only to Staphylococcus aureus after 60 minutes exposure and to Streptococcus spp. After 90 minutes of contact. Propolis tincture in dilutions 1:20 and 1:15 after 60 minutes of interaction caused the death of all test cultures (Escherichia coli, Staphylococcus aureus, Streptococcus spp, Enterobacter agglomerans). Thus, an effective dilution of a 10% propolis tincture with water is 1:20, with an exposure of 60 minutes it has an antimicrobial effect on all studied cultures.

Example 2. Determination of the optimal dose of 10% propolis tincture by watering broiler chickens for the prevention of respiratory infections in an experimental experiment
3.Of the daily broiler chickens, the control and three experimental groups of 20 animals each were completed. Chickens in the control group received the antibiotic tilmipul (d.v. tilmicosin) 0.3 ml / l of water in the age periods 1-3, 14-16, 25-27 days. The birds of the first, second and third groups were fed a 10% propolis tincture, respectively, in doses of 0.5; 1 and 1.5 ml / 1 liter of water in the age periods of 1-5, 14-18, 25-30 days. Preservation, live weight were taken into account, hematological (the number of leukocytes, erythrocytes, hemoglobin), biochemical (total protein, albumin, globulin), immunological (bactericidal activity of blood serum - BASK) blood tests were performed. The duration of the experiment is 42 days. The research results are presented in table 3.
Based on the data obtained, it was found that the use of propolis tincture in a dose of 1 ml / l of water (second group) increased the natural resistance and metabolism of broilers, as evidenced by the increase in the number of red blood cells, white blood cells, hemoglobin, total protein and globulins compared with the BASK control in the blood of birds. This had a positive effect on the zootechnical indicators of broiler rearing — an increase in safety by 10% and live weight by 252 g. When using the drug at a dose of 500 ml / l of water (first group), there was no significant difference with the control for the studied parameters. The use of a higher dose of 1.5 ml / l of water (the third troupe) also contributed to a significant increase in all studied parameters, but they did not exceed the indicators of the second group, therefore, it is not economically feasible.

Thus, the most effective use of a 10% propolis tincture for the prevention of respiratory infections by the method of drinking water at a dose of 1 ml / l of water in the age periods of 1-5, 14-18, 25-30 days, which increases the natural resistance and metabolism of broilers, provides high levels of safety, productivity and helps to prevent respiratory infections.
Example 2. The study of the application of tincture of propolis for the prevention of respiratory infections of boiler chickens in an experimental experiment
Of the daily broiler chickens, the control and two experimental groups were completed, 100 animals each. Chickens in the control group received the antibiotic tilmipul 0.3 ml / l of water in the age periods 1-3, 14-16, 25-27 days, aerosol treatment was carried out by ecocide-C (0.5%, 1 l / 100 m 3 of the house, exposure 60 minutes) on 1, 7, 14, 21-22, 28-29, 35-36 days of cultivation. In the first group, an antibiotic was used according to the same scheme as in the control, aerosol treatment was carried out with propolis tincture (1:20 dilution, 500 ml / 100 m 3 , 60 minutes exposure) in the same age periods as in the control group. The bird of the second group was fed tincture of propolis at a dose of 1 ml / 1 l of water in the age periods of 1-5, 14-18, 25-30 days, an additional aerosol treatment was carried out with tincture of propolis (dilution 1:20, 500 ml / 100 m 3 , exposure 60 minutes) on 1, 7, 14, 21-22, 28-29, 35-36 days of cultivation.
The prophylactic efficacy of the regimens was evaluated by the manifestation of clinical signs, safety and live weight of chickens. Bacteriological studies of scrapings from the mucous membrane of the larynx and air of the houses were carried out (total microbial number - TBC, bacteria of the Escherichia coli group - BGKP, staphylococci, enterococci, microscopic fungi), hematological (number of leukocytes, erythrocytes, hemoglobin), biochemical (total protein, albumin , globulins) and immunological (BASK) blood tests.
Аэрозольное применение настойки прополиса позволило снизить количество патогенной и условно-патогенной микрофлоры в соскобах со слизистой оболочки гортани цыплят опытных групп по сравнению с контролем, при наименьшем спектре выделенных микроорганизмов у цыплят второй группы (таблица 4).Aerosol application of propolis tincture allowed to reduce the number of pathogenic and conditionally pathogenic microflora in scrapings from the mucous membrane of the larynx of the chickens of the experimental groups compared to the control, with the smallest spectrum of microorganisms isolated in chickens of the second group (table 4).
Note: "+" - isolated microorganisms; “-” - microorganisms not detected
Based on the results of microbial contamination of the air, it was found that the number of microorganisms under study in the air of halls with experimental birds decreased over the entire period of broiler rearing compared to the control (table 5). So by the age of 42 days, the difference between the first and second groups in comparison with the control was: OMC - 1109 and 1169, BGKP - 108 and 122, staphylococci - 141 and 132, enterococci - 120 and 122, microscopic fungi - 229 and 236 CFU / m 3 , which confirmed the bactericidal activity of propolis on the air microflora.

Thus, the most effective for the prevention of respiratory infections of birds is the use of a 10% propolis tincture according to the scheme of the second group, where it was used aerosol in combination with water without chemotherapeutic agents. This reduces the microbial contamination of the air in the house during the entire period of broiler growing, sanitizes the upper respiratory tract of birds, activates the natural resistance and metabolism in the birds, which increases the safety, productivity of broilers and allows you to get safe poultry meat products.
Example 3. Testing a new method for the prevention of respiratory infections in a production experiment
Of the daily broiler chickens, the control and experimental groups of 400 animals each were completed. The poultry of the experimental group received a 10% tincture of propolis at a dose of 1 ml / 1 l of water during the age periods of 1-5, 14-18, 25-30 days, an additional aerosol treatment was carried out with 10% tincture of propolis (dilution 1:20, 500 ml / 100 m 3 , exposure 60 minutes) for 1, 7, 14, 21-22, 28-29, 35-36 days. Chickens in the control group received the antibiotic tilmipul 0.3 ml / l of water in the age periods of 1-3, 14-16, 25-27 days, aerosol treatment of the air of the house was carried out by Ecocide-C (0.5%, 1 liter / 100 m 3 , exposure 60 minutes) in the same age periods as in the experimental group. The duration of the experiment is 1-42 days.
