Abstract
Description
The invention relates to the field of veterinary medicine, namely to poultry farming, can be used to disinfect hatching chicken eggs.
In incubators, with a maximum concentration of eggs and hatched young animals, a large amount of pathogenic and conditionally pathogenic microflora accumulates, which contributes to the emergence and spread of infectious diseases. The still not strong bird organism becomes a "target" for pathogens.
As a result of the increase in microbial potential during the incubation period, embryonic mortality increases, hatchability of eggs and hatching of young animals decreases, and chickens are infected at hatching, which is always accompanied by their death on the first day of life.
The most common methods for processing hatching eggs are chemical, which are based on the use of various disinfectants (formaldehyde, ozone, hydrogen peroxide, desmol, virocid, ecocide C, bactericide, etc.) in the form of aqueous solutions, aerosols, gas.
A known method of processing eggs, including the use of formaldehyde vapor, per 1 m 3 : 30 ml of formalin (40%), 15 ml of water and 20 g of potassium permanganate, exposure time 30 minutes (see Veterinary law. T. 4, M. , 1988, pp. 411-415; Veterinary and sanitary rules for poultry farms (farms) and requirements for their design. Approved GUV Ministry of Agriculture of the USSR 07.23.1973).
However, the use of formaldehyde leads to pathological changes in internal organs and an increase in mortality of embryos in the second half of incubation, in addition, it causes allergic reactions in staff, increases the number of respiratory diseases. When using formaldehyde, its vapors are released into the environment, violating the environment.
- 24 с.).In order to prevent the appearance of wastes of the “cuff” category, a 1% aqueous propolis extract was used to process duck eggs. The effectiveness of propolis in these experiments was compared with the action of antibiotics (penicillin and streptomycin) and a solution of bleach. Antibiotics were used in the following concentrations: penicillin 1000 units and streptomycin 500 units per 1 ml of water, bleach in the form of a 5% solution (containing 1.4% active chlorine). For processing, eggs were immersed in propolis extract and antibiotic solutions for 3 minutes, and in a solution of bleach for 5 minutes. Treatment of eggs with antibiotics, a solution of bleach and propolis extract before laying in the incubator made it possible to prevent, at the very first mirage, the appearance of incubation waste of the “cuff” category. At the second mirage (on the 27th day), in batches of eggs treated with antibiotics and a 1% aqueous extract of propolis, incubation waste of the “tumak” category was also not detected. In batches treated with a solution of bleach, the amount of the “cuff” category was 20%, and in the control — 26%. In bacteriological examination of shell scrapings and samples from dead embryos, mold fungi were isolated in 90% of cases, and Sal bacteria in 6%. typhimurium and in 4% vulgar microflora (Pokrovsky S.G. Results of laboratory and industrial studies of the prophylactic and therapeutic effect of propolis in duck paratyphoid: abstract, dissertation of the candidate of economic sciences: - St. Petersburg: Ministry of Agriculture of the USSR - 1966. - 24 pp.) .
However, the processing of eggs in a known manner is laborious, requires a significant investment of time and money.
The objective of the invention is to increase the efficiency of processing of hatching eggs, which provides high viability of embryos, increase hatchability of eggs, hatching of young animals, preservation in embryonic and postembryonic periods of growing. The problem is solved by using propolis alcohol tincture, from which a working solution is prepared in the following ratio of components: 1 part propolis tincture 10 parts water, use aerosol at the rate of 250 ml per 1 m 3 of the incubator, the first treatment is carried out before incubation, then at 11.5 , 18.5, 21.5 days of incubation.
Features of the formation of the organism at the beginning of its development are important not only during the incubation period, but also for the entire subsequent viability and productivity of the bird. During biological control (11.5, 18.5, 21.5 days of incubation), eggs are cooled, microorganisms can penetrate through the pores of the shell. In order to reduce the risk of their ingestion, we proposed periods of biological control for the treatment of hatching eggs.
Processing of hatching eggs is carried out until their surface is completely moistened, so as not to increase the percentage of moisture in the setter. The optimum relative humidity in it should be maintained within 40-60% in the incubation and 50-80% in the hatcher. Constant high air humidity delays the evaporation of moisture, low - stimulates its strong evaporation from the egg. It is known that in the initial period of incubation, for normal development of the embryo, elevated temperature and humidity are required to prevent more intense evaporation and drying of the egg. In the second incubation period, on the contrary, more intensive evaporation should be promoted. At high humidity during the incubation period, the shrinkage of the egg is too small because the evaporation of moisture from the egg is limited. This is evidenced by the size of the air chamber, which does not increase during all incubation periods, which leads to a difficult conclusion, and the hatched chickens remain edematous and are poorly preserved during rearing.
3.These circumstances were taken into account when calculating the drug used aerosol to process 1000 pieces of hatching eggs and it was found that 250 ml per 1 m 3 is optimal.

Example 1. Determination of the bactericidal activity of propolis tincture and effective in vitro concentration.
2009. - №8. - С. 43-44).The bactericidal effect of propolis tincture and the alcohol used for its preparation were compared. Serial dilutions were prepared from propolis tincture and alcohol: 1: 5, 1:10, 1:15, 1:20, the test strain was introduced, the viability of microorganisms was determined after 24 hours of interaction by plating on solid nutrient media (timing agar, staphylococcagar ) Microbiological monitoring in hatcheries made it possible to establish that the dominant microorganisms are Escherichia coli and Staphylococcus spp. (Lysko SB, Makarova OA Microbiological monitoring in hatcheries // Poultry farming. 2009. - No. 8. - P. 43-44) .
Field cultures of Escherichia coli, Staphylococcus aureus isolated in the hatchery were used as test strains.
1:5.From table 1 it is seen that propolis tincture exhibits high antibacterial activity against the studied pathogens, causing their death at dilutions of 1:10 - Escherichia coli and 1:15 - Staphylococcus aureus, compared to alcohol, which does not show antibacterial activity even in breeding 1 :5.
Note: "+" - the presence of culture growth; “-” - lack of culture growth
Based on our experience, we offer the optimal dilution of propolis tincture, which has a bactericidal effect on E. coli and staphylococcus in the following ratio of components: 1 part propolis tincture 10 parts water.
Example 2. The study of various methods of processing hatching eggs. The control and three experimental groups of 1000 pieces each, which were placed in separate, identical incubators, were composed of hatching eggs of the Ross-308 cross. To control microbial contamination, microbiological studies were carried out: swabs from the shell of the incubation eggs (for the presence of staphylococci, bacteria of the E. coli group) - before laying and on the 11.5, 18.5 days of incubation; air in incubators (total microbial number, bacteria of the Escherichia coli group, staphylococci, enterococci, microscopic fungi) - on the 11.5, 18.5 and 21.5 days of incubation; scrapings from the mucous membrane of the larynx of the removed young. When evaluating the results of incubation, the hatchability of eggs and the causes of death of the embryos were taken into account. In order to study the viability of young animals, the first 10 days of cultivation carried out daily safety records with registration of the causes of death.
In the control group, before the start of incubation, formaldehyde vapor was treated (30 ml of formalin, 20 g of potassium permanganate and 15 ml of water per 1 m 3 of the chamber), exposure time 30 min; during transfer and until withdrawal (18.5-21.5 days of incubation) - formalin diluted half with water. In the experimental groups, the processing was carried out by aerosol tincture of propolis diluted with water 1 part propolis and 10 parts water at the rate of 250 ml per 1 m 3 . In the first experimental group, the treatment was carried out three times: the first treatment before the start of incubation, the subsequent on the 18.5 and 21.5 days of incubation. In the second group, it is similar to the first, only four times: before the start of incubation, the subsequent ones on the 11.5, 18.5 and 21.5 days of incubation. In the third group: the first treatment - before the start of the incubation, it was carried out with formaldehyde vapors, and then a double treatment with propolis tincture (1:10) on the 18.5 and 21.5 days of incubation.
From table 2 it is seen that in the study of swabs from the surface of the shell of the incubation eggs by 18.5 days of incubation, a decrease in the frequency of isolation of staphylococci by 80-90%, bacteria of the group of Escherichia coli (BHEC) by 20% in the 1st, 2nd, 3rd experimental groups compared with control.
The data in table 3 confirm that propolis tincture reduces microbial airborne contamination by 21.5 days of incubation in the 1st, 2nd and 3rd groups compared with the control: the total microbial number - by 83, 94 (P <0.01 ) and 30 CFU / m 3 ; coliform bacteria - at 78, 85 (P <0.01) and 57 CFU / m 3 ; staphylococci - by 81 (P <0.01), 81 (P <0.01) and 79 (P <0.01) CFU / m 3 ; enterococci - at 20, 26 (P <0.05) and 12 CFU / m 3 ; microscopic fungi - by 17 (P <0.05), 21 (P <0.01) and 11 (P <0.05) CFU / m 3 . The smallest contamination was observed in the second experimental group.

Прижизненным исследованием соскобов со слизистой оболочки гортани у выведенного молодняка установлено, что наибольший спектр микроорганизмов выделен у цыплят контрольной группы, наименьший у цыплят второй группы (таблица 4).By a lifetime study of scrapings from the mucous membrane of the larynx in hatched young animals, it was found that the largest spectrum of microorganisms was isolated in chickens of the control group, the smallest in chickens of the second group (table 4).
Enterococcus faecalis+Escherichia coli.At the same time, 80% of microorganisms are isolated in associations. The largest number of associations of microorganisms was isolated in the control group: Enterococcus faecalis + Pseudomonas aeruginosa + Citrobacter freundii, Staphylococcus spp + Enterococcus faecalis + Citrobacter freundii and Staphylococcus spp + Enterobacter cloacea + Citrobacter freundii. The following two associations were distinguished in the first group: Staphylococcus spp + Enterococcus faecium and Staphylococcus spp + Enterococcus faecalis. In the second group, one association was identified - Staphylococcus spp + Enterococcus faecalis + Escherichia coli, and in the third two - Staphylococcus spp + Citrobacter freundii and Enterococcus faecalis + Escherichia coli.

From table 5 we observe that the largest number of eggs of the category “embryo death up to 48 hours of incubation” in the control and third groups, which were treated with formaldehyde vapor before laying, is associated with the negative effect of formalin on embryo development on the first day of incubation. The largest waste of the suffocated egg category was also noted in the control group, in which formaldehyde treatment was carried out on the 18.5 day of incubation. A decrease in this category in the experimental groups can be associated with a decrease in the microbial background and the absence of a negative effect of propolis tincture on the embryo during repeated treatments. The highest hatchability of eggs with the lowest values of the category of eggs “embryo death up to 48 hours” and “suffocates” was noted in the second group.
2,6%.In the first group, hatchability was 0.6% higher than in the control, due to a decrease in the category of eggs “embryo death up to 48 hours” - 0.4% and “suffocates” - 1.4%. In the second group, the hatchability of eggs was 3.3% higher than the control due to a decrease in the category of eggs “embryo death up to 48 hours” by 1.4%, and “suffocates” - by 3.4%. In the third group, the hatchability of eggs is 0.7% higher compared to the control due to a decrease in the category of eggs of "suffocated" - 2.6%. The highest hatchability of eggs was in the second group, the difference from the first was 2.7%, from the third 2.6%.
In the chickens of the experimental groups, the safety in the first 10 days of growing was 1-3% higher compared to the control, while the best result was noted in the second group. The main causes of death of chickens in the control group were pneumonia, ampholytes and gout, experienced - trauma. In chickens of the experimental groups with pathoanatomical autopsy, pathologies of the respiratory tract were not found, in contrast to the control.
Example 3. Testing methods for processing hatching eggs
The control and experimental groups of 1000 eggs each, which are placed in separate, identical incubators, are composed of hatching eggs of Ross-308 cross-country birds. To process hatching eggs in an incubator with a control group, they were disinfected with formaldehyde vapors (30 ml formalin, 20 g potassium permanganate and 15 ml water) per 1 m 3 of chamber; during transfer and until withdrawal - formalin diluted half with water. In an incubator with an experimental group, the treatment of hatching eggs was carried out by aerosol tincture of propolis diluted with water 1:10 at the rate of 250 ml of solution in 1 m 3 of the chamber before the start of incubation and on the 11.5, 18.5, 21.5 days of incubation.
When evaluating the effectiveness of the treatments, the development of embryos during the incubation process, the hatchability of eggs, the withdrawal of young animals were taken into account, pathological analysis of the dead embryos and the causes of their death in each group were carried out. Bacteriological studies were carried out of the washings from the shell of incubation eggs (bacteria of the Escherichia coli group, Staphylococcus aureus), air (total microbial count, bacteria of the Escherichia coli group, Staphylococcus aureus, Enterococci, microscopic fungi), the presence of pathogenic and conditionally pathogenic microflora in the incubation waste.
From table 6 we see that when studying the swabs from the shell surface, it was found that treatment of the hatching eggs with propolis tincture reduced the rate of staphylococcus excretion by 20-70%), BGKP - by 50-70% from the shell surface and prevented their reproduction throughout the incubation.
The results of microbiological studies of air in incubators indicate the bactericidal effect of the propolis preparation on pathogenic, conditionally pathogenic microflora and microscopic fungi during the incubation period (table 7). Aerosol treatment of air with tincture of propolis by 21.5 days contributed to a decrease in TBC by 341 CFU / m 3 , BGKP by 48 CFU / m 3 , staphylococcus by 129 CFU / m 3 , enterococcus by 79 CFU / m 3 , and microscopic fungi 51 CFU / m 3 compared with the control group.

При бактериологическом исследовании соскобов со слизистой оболочки гортани наиболее широкий спектр микроорганизмов был выделен у цыплят контрольной группы (таблица 8).During bacteriological examination of scrapings from the mucous membrane of the larynx, the widest spectrum of microorganisms was isolated in chickens of the control group (table 8).
The largest number of types of associations was identified in the control group: Enterococcus faecalis + Citrobacter freundii + Staphylococcus aureus + Enterobacter agglomerans, Enterococcus faecalis + Enterobacter agglomerans + Escherichia coli, Staphylococcus aureus + Enterococcus faecalisococerocerocerocerocerocericobecerociferocobecerociferocobius faecalis . In the experimental one association was identified - Enterococcus faecalis + Staphylococcus aureus.

From table 9 we see the comparative incubation results for the groups: the hatchability of eggs in the experimental group was 4.8% higher compared to the control due to the decrease in the number of eggs in the category “embryo death up to 48 hours” - 0.9%, “blood ring” - 2.6%, “frozen bird embryos” - 0.9%, “suffocates” - 1.3%, the chickens in the experimental group were 4.2% higher. The safety of chickens in the first 10 days of growing in the experimental group was 2% higher than in the control group.
Thus, the proposed method for processing hatching eggs allows to reduce the growth of pathogenic and conditionally pathogenic microflora during the incubation period, does not adversely affect the development of embryos, increases hatchability of eggs by 4-5%, hatch of chickens by 4-5%, and the safety of chickens in the first 10 days of cultivation by 2-3%, disinfects the mucous membranes of the respiratory tract of hatched young animals, and thereby contributes to the prevention of respiratory infections in the hatchery.
