Abstract
Description
The invention relates to the field of experimental biology and medicine and relates to a method for stimulating the production of granulocyte colony stimulating factor in vitro.
[1, 2].There are various ways to stimulate the production of cytokines, including granulocyte colony stimulating factor, by various bone marrow cells [1, 2].
[2].The closest in technical essence, mechanism of action and technical result (prototype) is a method of stimulating the production of granulocyte colony stimulating factor in vitro by adherent bone marrow cells in vitro using a p38 protein kinase inhibitor, which is added at a concentration of 300 μM in tissue culture and incubated at 37 ° C , 5% CO 2 and 100% air humidity within 24 hours [2].
The disadvantage of this method is the lack of stimulation of the production of colony stimulating factor under the action of this inhibitor.
The problem solved by this invention is to expand the arsenal of methods for stimulating the production of granulocyte colony stimulating factor in bone marrow cells in vitro and to increase the efficiency of the method.
Поставленная задача достигается тем, что in vitro культивируют фракцию непри-липающих миелокариоцитов в СO2-инкубаторе при 37°С, 5% СО2 и 100% влажности воздуха в течение 24 часов в полной культуральной среде следующего состава: 90% среды RPMI-1640, 10% ЭТС, 280 мг/л L-глутамина, 50 мг/л гентамицина, при добавлении ингибитора фосфатидилинозитол-3-киназы (PI3K) в концентрации 50 мкМ (микромоль).This object is achieved in that a non-adherent myelocaryocyte fraction is cultured in vitro in a CO 2 incubator at 37 ° C, 5% CO 2 and 100% air humidity for 24 hours in a complete culture medium of the following composition: 90% RPMI-1640 medium , 10% ETS, 280 mg / l L-glutamine, 50 mg / l gentamicin, with the addition of a phosphatidylinositol-3-kinase inhibitor (PI3K) at a concentration of 50 μM (micromol).
New in the present invention is the use of bone marrow cells producing granulocyte colony stimulating factor, non-adherent fraction of bone marrow and the use of PI3K inhibitor as a stimulator of production of granulocyte colony stimulating factor.
Today, among the various pharmacological agents used as hemostatic stimulants, the most widely used are preparations based on endogenous hemopoietic stimulants (colony-stimulating factors and erythropoietin), which, when bound to specific receptors, trigger numerous signaling mechanisms involved in the implementation of key cell functions [1, 3, 4]. Particularly noteworthy is the preparation of granulocyte colony stimulating factor (G-CSF) [3]. An in vitro study of the synthesis of this hematopoietin by bone marrow cells — elements of the hematopoietic-inducing microenvironment — is an important element of the methodology for creating hemostatic agents [4] with targeted action by activation / inhibition of intracellular signaling [5-8]. These studies are carried out as part of a new direction of targeted therapy in regenerative medicine, proposed at the NIIIFiRM them. E.D. Goldberg (Tomsk Scientific and Research Center) - “Strategies for the pharmacological regulation of intracellular signal transduction in regeneratively competent cells" [5]. In this regard, the development of effective methods for the production of granulocyte colony stimulating factor using modifiers of the activity of intracellular signaling seems relevant.
A number of signaling molecules are known to be involved in determining the functional, including secretory, activity of some bone marrow cells [1, 8]. However, the participation and role of phosphatidylinositol-3-kinase (PI3K) in the production of granulocyte colony stimulating factor by non-adherent bone marrow cells has not yet been studied.
The fact of using a non-adherent bone marrow fraction when a PI3K inhibitor is added to the culture medium to achieve a new technical result, which is to effectively stimulate the production of granulocyte colony stimulating factor in vitro, is not obvious for a specialist. The experiment showed unpredictable results.
New properties do not follow explicitly from the prior art in this field. An identical set of features was not found in the study of the prior art in patent and medical literature.
The present invention can be used in experimental biology and medicine.
Based on the foregoing, the claimed technical solution should be considered relevant criteria: "Novelty", "Inventive step", "Industrial applicability".
The method is as follows.
A non-adherent fraction of cells is obtained from bone marrow cells. Non-adherent bone marrow cells are incubated in a complete culture medium of the following composition: 90% RPMI-1640 medium, 10% ETS, 280 mg / l L-glutamine, 50 mg / l gentamicin, 50 μM PI3K inhibitor, in a CO 2 incubator at 37 ° C, 5% CO 2 and 100% air humidity for 24 hours. At the end of the incubation, the conditioned media after centrifugation at 1500 rpm for 10 minutes are collected using a pipette.
Animals were obtained from the Department of Experimental Biological Models of the NIIIFiRM im. E.D. Goldberg Tomsk Scientific and Technical Center. The studies were carried out in accordance with the rules of laboratory practice (GLP), Order of the Ministry of Health and Social Development of the Russian Federation No. 708n dated 08/23/2010 “On approval of the rules of laboratory practice”.
1.Example 1
A suspension of bone marrow cells obtained under sterile conditions (a box equipped with a laminar) was incubated for 45 min in RPMI-1640 medium (Sigma, USA) containing 10% ETS (Sigma, USA) in 90 mm plastic cups Petri (Corning, USA) with a diameter of 35 mm for 40-50 minutes at 37 ° C, 5% CO 2 and 100% humidity to isolate a non-adherent fraction of myelokaryocytes. Then non-adherent myelokaryocytes were collected with a pipette, which (at a concentration of 2 × 10 6 cells / ml) were incubated for 24 hours in a complete culture medium of the following composition: 90% RPMI-1640 medium (Sigma, USA), 10% ETS (Sigma, USA), 280 mg / L L-glutamine (Sigma, USA), 50 mg / L gentamicin (Sigma, USA), 50 μM PI3K inhibitor (Calbiochem, USA), at 37 ° C, 5% CO 2 and 100% humidity [4]. The used concentration of the PI3K inhibitor was selected in preliminary experiments as the most effective.
In this case, control No. 1 and No. 2 were conditioned media from non-adherent and non-adherent bone marrow cells (respectively) without an inhibitor, respectively, and the comparison group — conditioned media from non-adherent bone marrow cells with the addition of a p38 protein kinase inhibitor, obtained by the prototype method.
To obtain adherent bone marrow cells for control No. 2 and the prototype method [2], a suspension of bone marrow cells obtained under sterile conditions (a box equipped with a laminar) was incubated for 45 min in RPMI-1640 medium (Sigma, USA) containing 10% ETS (Sigma, USA), in 90 mm plastic Petri dishes (Corning, USA) with a diameter of 35 mm for 40-50 minutes at 37 ° C, 5% CO 2 and 100% humidity to isolate the adhesive fraction myelocaryocytes. Then non-adherent myelokaryocytes were collected with a pipette, and adherent cells were incubated for 24 hours in a complete culture medium of the following composition: 90% RPMI-1640 medium (Sigma, USA), 10% ETS (Sigma, USA), 280 mg / L L-glutamine (Sigma, USA), 50 mg / L gentamicin (Sigma, USA), 300 μM p38 protein kinase inhibitor (Calbiochem, USA), at 37 ° C, 5% CO 2 and 100% humidity.
At the end of the incubation, conditioned media were collected and the level of G-CSF was determined in them by ELISA using a kit of R&D systems (USA) according to the manufacturer's guidelines.
1).During the experiment, an increase in the production of granulocyte colony-stimulating factor is shown only with the addition of a stimulator of production to adherent bone marrow cells (prototype method) and with the implementation of the proposed method. Moreover, when a PI3K inhibitor was added to the culture of non-adherent bone marrow cells, the level of production of granulocyte colony stimulating factor was statistically significantly higher than that of the prototype method (culturing adherent bone marrow cells with the addition of the p38 inhibitor) (Table 1).
Thus, incubation of the non-adherent fraction of bone marrow cells with a PI3K inhibitor leads to the most significant stimulation of G-CSF production in vitro.
The proposed method can effectively stimulate the production of granulocyte colony stimulating factor non-adherent myelocaryocytes in vitro.
