Abstract
Description
The invention relates to a new strain of Lactococcus lactis 1b-MI and can be used in the production of starter preparations and fermented milk products of mass and therapeutic purposes.
The bacterial strains of the species L. lactis in modern production are used as part of multicomponent starter cultures. In order to increase the efficiency and reliability of the production process of milk fermentation, strains to be combined in yeast are selected in such a way that their technologically significant properties are complementary and have high activity. The strains of lactic acid bacteria L. lactis are producers of nisin, which inhibits the development of gram-positive microorganisms: streptococci, staphylococci, sarcinomas, spore-forming bacteria, including thermophilic microflora, which is an advantage when starter cultures are included in the consortium.
Known strain of Streptococcus lactis VKPM B-6793 - a component of lactic acid starter culture and producer of bacteriocin nisin [1]. The disadvantages of this known strain is a rather narrow spectrum of fermentation of sugars and alcohols. Closest to the claimed invention according to the technical solution and production indicators is a strain of Streptococcus lactis VKPM B-6795 [2]. The disadvantages of its use in production are low productivity, low ability to ferment carbohydrates other than lactose.
Unlike the prototype, the proposed strain of Lactococcus lactis 1b-MI is able to ferment a wider range of various carbohydrates and alcohols and has optimal properties for inclusion in the consortium of lactic acid starter culture.
[3].The strain was isolated from collection samples by direct seeding on liquid nutrient media, followed by seeding on agar media. Identification of the strain was carried out according to the determinant of Bergey [3].
RCAM 02909.Lactococcus lactis 1b-MI is deposited in the Departmental collection of beneficial microorganisms for agricultural purposes (VKSM) GNU VNIISCHM under the number RCAM 02909.

Example 2. Determination of the viability of the claimed strain in the process of long-term cultivation.
Bacteria of the inventive strain of Lactococcus lactis 1b-MI and a control strain from collection samples of MI PTU were grown for 24 hours at 37 ° C after plating in Petri dishes with enriched agarized MA20 medium (Table 4). A bacterial suspension was prepared from the grown biomass of each strain and diluted in such a way that when plating on the surface of a minimal agarized medium MA4, a comparable number of individual colonies of the compared strains on one plate could be observed.
After prolonged (for 96 hours) incubation at 37 ° C, the average colony area of the claimed strain exceeded that of the control strain (3 and 1.5 mm 2 , respectively). This confirms the ability of the claimed strain 1b-MI to accumulate a greater amount of biomass in conditions of a minimum composition of the nutrient medium and long incubation.
The properties of the claimed strain indicate the feasibility of its use as part of a multicomponent sourdough, since under unfavorable conditions (lowering the pH of the medium, increasing temperature, as well as the gradual depletion of the composition of the nutrient medium as the process proceeds), it is capable of fermenting milk. Increased bacterial viability allows to increase the content of living cells in the resulting fermented milk product.
