Abstract
Description
The invention relates to medicine, namely to toxicology, relates to a method for assessing the toxic hazard of anticholinesterase compounds (ANCES), and can be used in unforeseen emergencies (industrial accidents, terrorist acts, etc.) to identify substances that are extremely dangerous for humans.
At present, ANHES are widely used in medicine, agriculture and other areas of the national economy. Some of them belong to the first hazard class [1] and are potential toxic substances of the nerve-paralytic type of action. In this regard, the assessment of the toxic hazard of such extremely dangerous chemical compounds and their identification in unforeseen emergencies is of great practical importance.
The currently known and widely used method for determining the degree of danger of any substances, including ANXES, is based on the assessment of their toxicity indicators (for example, the average lethal dose (LD 50 )) and involves the use of biological models, which are rats, mice, cats, rabbits and other animals. The specified method is adopted as a prototype method.
The prototype method has a number of significant disadvantages that do not allow its use in unforeseen emergencies. So, a very important condition of the prototype method is the need to use the test compound during the experiments in a pure form in order to then prepare its various dilutions and to ensure that the experimental animal is given the exact amount of the toxicant. In the same cases when, in emergency situations, there are solutions of unknown concentrations of ANCHES composition unknown, it is practically impossible to conduct experiments using experimental animals and determine the toxicity indices of the studied compounds. The determination of toxicity indicators using the prototype method becomes possible only after analysis of the qualitative and quantitative composition of the studied solutions, that is, after conducting their chemical-analytical examination.
In addition, when implementing the prototype method only to determine the order of the level of toxicity index, a number of experiments using a sufficiently large number of experimental animals are required [2, 3, 4]. At the same time, experimental animals should be kept in a certain way, and studies should be carried out in a stationary laboratory with the involvement of specially trained personnel, which leads to their high complexity, implementation time (up to 24 hours) and high material costs.
As you can see, the use of the prototype method for assessing the toxic hazard of ANES in unforeseen emergencies, when the studied compounds are in the form of solutions of unknown concentration and their compositions are not known, is practically not possible.
Thus, the aim of the invention is to develop a method for assessing the toxic hazard of compounds with an anticholinesterase type of action under various conditions, including in unforeseen emergencies, when their composition is not known and they are in the form of solutions of unknown concentration, which is easy to carry out and allows identify extremely dangerous compounds in a short time.
Поставленная цель достигается за счет использования способа, основу которого составляет определение антихолинэстеразного эффекта исследуемого соединения на препарат холинэстеразы, в качестве которого используют фермент пропионилхолин-эстераза мозговой ткани кальмара (ПХЭ).This goal is achieved through the use of a method based on the determination of the anticholinesterase effect of the test compound on a cholinesterase preparation, using the enzyme propionylcholine esterase of squid brain tissue (PCE).
Фермент ПХЭ используют в двух фиксированных концентрациях - малой (0,04 Е/мл) и высокой (4 Е/мл).The essence of the proposed method is to determine the degree of inhibition of PCE activity under the influence of a solution of the test compound. The PCE enzyme is used in two fixed concentrations - small (0.04 U / ml) and high (4 U / ml).
During the experiment, first a dilution of the test compound solution is found that provides 30-60% inhibition of PCE activity in a low concentration solution. Then, the same dilution of the solution of the test compound is used to determine the effect of its effect on the activity of PCE in a high concentration solution.
By the difference in the effect of the studied ANCES on the activity of PCEs in solutions of low and high concentrations, it is determined whether the test compound belongs to highly toxic or low toxic substances. In the event that the percentage of inhibition of PCE activity in a solution of high concentration drops to zero or does not exceed 5-10%, it is concluded that the analyzed solution contains a highly toxic anticholinesterase compound of hazard class 1, i.e., a compound extremely dangerous for humans is detected. If the magnitude of the inhibitory effect remains constant or drops by no more than 10%, a conclusion is made on the presence of low-toxic ANES.
The ability to achieve the objectives of the invention is confirmed by the results of the studies presented in the following examples.
Example 1. The implementation of the proposed method for assessing the toxic hazard of anticholinesterase compounds.
When implementing the proposed method, the anticholinesterase effect of the test compound on the PCE enzyme is determined, namely, the degree of inhibition of PCE activity under the influence of the test compound solution is evaluated. In this case, the PCE enzyme is used in solutions of two concentrations differing 100 times in the enzyme content. Two solutions of PCE are prepared in 0.2 M phosphate buffer pH 7.5 with an enzyme activity of 0.04 U / ml and 4.0 U / ml.
An aqueous solution of acetylthiocholinium iodide (ATX) in a conception of 5.78 mg / ml and a solution of Ellman's reagent in a concentration of 0.792 mg / ml are also used.
Затем это же разведение исследуемого соединения используют для определения степени угнетения активности ПХЭ в его растворе высокой концентрации (4 Е/мл).During the experiment, first a dilution of the test compound solution is found which, when mixed with an equal volume of a low concentration enzyme solution (0.04 U / ml), inhibits PCE activity by 30-60%. Then, the same dilution of the test compound is used to determine the degree of inhibition of PCE activity in its high concentration solution (4 U / ml).
For research, to 1 ml of an enzyme solution with an activity of 0.04 U / ml add 1 ml of a solution of the test compound and incubated for 3 minutes at room temperature. The resulting mixture was mixed with a pre-prepared solution containing 1 ml of distilled water, 1 ml of phosphate buffer, 0.5 ml of ATX solution and 0.5 ml of Ellman's reagent solution. The solution is stirred and after 3 minutes, its optical density is measured using a spectrophotometer at a wavelength of 412 nm. In parallel, a control sample is prepared in which distilled water is used instead of diluting the solution of the test compound.
The calculation of the percentage of inhibition of enzyme activity (i%) is carried out according to the formula:
где Dоп - оптическая плотность опытной пробы;where D op - the optical density of the experimental sample;
D to - the optical density of the control sample.
Then, a dilution of the solution of the test compound is selected, which causes inhibition of PCE activity by 30-60% at an enzyme solution concentration of 0.04 U / ml. The same dilution of the solution of the test compound is subsequently used to inhibit the activity of PCE in a concentration of 4 U / ml. For this purpose, to 1 ml of an enzyme solution with activity of 4 U / ml add 1 ml of a solution of the test compound and incubated for 3 minutes at room temperature. Then, 0.02 ml was taken and poured into a pre-prepared mixture containing 2.98 ml of distilled water, 1 ml of phosphate buffer, 0.5 ml of ATX solution and 0.5 ml of Ellman's reagent solution. The resulting mixture was stirred and after 3 minutes, its optical density was measured at a wavelength of 412 nm. To calculate the percentage of inhibition of PCE activity, an optical density index is used in parallel with a control sample, in which distilled water is used instead of diluting the solution of the test compound.
In the event that the percentage of inhibition of PCE activity in a solution of high concentration drops to a bullet or does not exceed 5-10%, it is concluded that the analyzed solution contains a highly toxic anticholinesterase compound of hazard class 1, that is, a compound extremely dangerous for humans is detected. If the value of the inhibitory effect remains constant or drops by no more than 10%, a conclusion is drawn on the presence of low-toxic ANES.
As you can see, the inventive method is quite simple to implement, and in a variety of conditions. For research takes no more than 30-60 minutes. In contrast to the prototype method, a long, laborious and expensive determination of toxicity indicators using a large number of experimental animals is excluded.
Example 2. Assessment of the toxic hazard of anticholinesterase compounds using the proposed method.
When using the proposed method, an assessment of the toxic hazard of various anticholinesterase compounds was carried out, including:
- O-изопропил-метил-фтор-фосфонат (вещество 1);- O-isopropyl methyl fluorophosphonate (substance 1);
- O-пинаколил-метил-фтор-фосфонат (вещество 2);O-pinacolyl methyl fluorophosphonate (substance 2);
- O-этил-S-диизопропил-аминоэтил-метил-тио-фосфоиат (вещество 3);- O-ethyl-S-diisopropyl-aminoethyl-methyl-thio-phosphoate (substance 3);
- O-изобутил-S-диэтил-аминоэтил-метил-тио-фосфонат (вещество 4);- O-isobutyl-S-diethyl-aminoethyl-methyl-thio-phosphonate (substance 4);
- O,O-диметил-O,2,2-дихлорвинил-фосфат (ДДВФ);- O, O-dimethyl-O, 2,2-dichlorvinyl phosphate (DDVF);
- O-паранитрофенил-O,O-диэтилфосфат (фосфакол);- O-paranitrophenyl-O, O-diethyl phosphate (phosphacol);
- О-паранитрофенил-O-этил-этилфосфонат (армии);- O-paranitrophenyl-O-ethyl-ethylphosphonate (army);
- Дихлоргидрат-N,N-диметил-(2-диметиламиноэтил-пиридил-3) карбамат (аминостигмин);- Dichlorohydrate-N, N-dimethyl- (2-dimethylaminoethyl-pyridyl-3) carbamate (aminostigmine);
- N-метил-N-децил-(2-N,N-диметил-амино-метил-пиридил-3) карбамат (оксалат) (ИХ-118);- N-methyl-N-decyl- (2-N, N-dimethyl-amino-methyl-pyridyl-3) carbamate (oxalate) (IH-118);
- 1,3A,8-триметил-2-3-a,8a-тетрагидропирроло[3'2b]-индол-5ил-N-метил-карбамат салициловокислый (эзерин).- 1,3A, 8-trimethyl-2-3-a, 8a-tetrahydropyrrolo [3'2b] -indole-5yl-N-methyl-carbamate salicylic acid (eserin).
The inclusion of these widely known compounds in the experiment is primarily due to the fact that there is information in the literature on the assessment of their toxicity using the prototype method [5, 6, 7, 8]. In addition, it is known that according to the requirements of GOST [9] for toxicity of 0.1 mg / kg and below, the compound belongs to the first hazard class, and for toxicity of 1.1-10.0 mg / kg to the third class. Information on the hazard assessment of all considered compounds based on the assessment of their toxicity indicators is also available in available information sources. So, it is known that substances 1-4 are highly toxic and belong to hazard class 1, and the remaining compounds used in the experiment are low toxic substances of hazard class 3.
The results of the assessment of the toxic hazard of anticholinesterase compounds using the proposed method indicate that solutions of substances 1, 2, 3 4 should be classified as extremely dangerous ANCES hazard class 1, since they cause inhibition of PCE activity in low concentration solutions (0.04 E / ml) by 30-60%. The degree of their inhibition of PCE activity in high concentration solutions (4 U / ml) is 5-8%. Such an assessment fully confirms the available information on the toxicity and hazard of the highly toxic substances under consideration.
Это свидетельствует о том, что указанные соединения не относятся к 1 классу опасности, что также совпадает с имеющимися сведениями об указанных малотоксичных веществах, относящихся к 3 классу опасности (таблица).At the same time, other anticholinesterase compounds used in the experiment more significantly inhibit the activity of PCEs and high concentration solutions (4 U / ml) by 28-42%, while the decrease in inhibitory activity for such compounds (compared to low concentration solutions - 0 , 04 U / ml) did not exceed 10%. This indicates that these compounds do not belong to hazard class 1, which also coincides with the available information about these low-toxic substances belonging to hazard class 3 (table).
п.).Thus, the inventive method allows you to quickly assess the toxic hazard of anticholinesterase compounds of unknown composition in the form of solutions of unknown concentrations, which is almost impossible to carry out in animal experiments using the prototype method. Moreover, the claimed method is quite simple to carry out and can be used in a variety of situations, including unforeseen emergencies (industrial accidents, terrorist acts, etc.).
The claimed invention meets the criterion of "novelty", since for the first time to assess the toxic hazard and identify extremely dangerous substances of anticholinesterase type of action, it is proposed to use a method involving determining the degree of inhibition of the activity of the enzyme propionylcholinesterase of squid brain tissue taken in two different concentrations with solutions of anticholinesterase compounds.
The claimed invention meets the criterion of "inventive step", since there are no data in the well-known and accessible sources of information containing descriptions of methods for determining the toxic hazard of anticholinesterase compounds that indicate the feasibility and advisability of determining anticholinesterase effects on the squid brain tissue propionylcholinesterase enzyme for this purpose, and the use of in this case, enzyme solutions of two concentrations.
Compliance with the criterion of "suitability for use" is confirmed by the results of the study, which showed that the proposed method is quite simple to carry out, allows you to quickly and reliably distinguish between highly toxic and low toxic anticholinesterase compounds and identify substances that are extremely dangerous for humans.
