Abstract
Description
The invention relates to biotechnology, veterinary virology and microbiology, expands the range of adjuvants and can be used in the biological industry and research work in the development and production of immunizing drugs for the diagnosis and prevention of infectious diseases in various types of domestic and farm animals.
The immunogenicity and effectiveness of vaccines is significantly increased with the use of non-specific immunity stimulants, adjuvants, in the technology for their manufacture.
Known use as adjuvants in the manufacture of various antigens of aluminum hydroxide Al (OH) 3 , which is a weak adjuvant in combination with antigens.
Oil adjuvants are known (such as Freund's adjuvant, etc.), which have pronounced immunostimulating properties, but have increased reactogenicity for animals.
[1].Separate substances from the group of saponins are known, which are currently widely used as adjuvants in the production of various types of vaccines [1].
In the Russian Federation, imported saponin manufactured by Merck Germany is most often used in the manufacture of vaccines.
[2].Saponins are surface active agents and cause hemolysis of red blood cells in vitro. In 1995, Kensil et al. Isolated a pure fraction of Quil A saponin with low toxicity (QS 21) and determined the structural fractions responsible for adjuvant activity [2].
[2].QS 21 has been proven to be an adjuvant for the production of cytotoxic T lymphocytes (CTL), inducing cytokines (IL-2 and IFN-y) and Ig G antibodies of the 2a isotype [2].
However, all known saponins with adjuvant action (described in the literature and imported from Germany and England) are made from plants that do not grow on the territory of our country. The high cost of imported drugs and the annual increase in saponin consumed in the bio-industry and research work increase the cost of purchasing it abroad.
The task of creating the present invention was to search for a new adjuvant that has high immunostimulating properties and is able to replace the imported drug saponin in the manufacture of various antigens used to obtain vaccines against infectious diseases of animals and diagnostic sera. In this case, the adjuvant should be free of ballast substances, not have antigens similar to host antigens, not have oncogenic, allergenic and toxic properties, easily metabolized in animals.
The technical result from the use of the well-known drug etonium as an adjuvant is to expand the range of adjuvants for the production of effective, safe and cost-effective sorbed FMD vaccines.
The specified technical result was achieved by the use of the drug etonium as an adjuvant in the manufacture of drugs for active immunization and for the diagnosis of viral infections of animals.

Д. [4].Onischuk F.D. et al. used etonium as part of an immunomodulating drug, which was used as a therapeutic agent to enhance immunity [4].
The essence of the invention is illustrated by examples of its use, which do not limit the scope of the invention.
7,4÷7.6.As a test object, foot and mouth disease virus was used. An inactivated sorbed foot and mouth disease vaccine is prepared from a virus grown in a suspension culture of BHK-21 cells. As a supporting medium, an Earle solution without serum is used with the addition of 0.15% hemohydrolyzate and antibiotics at a pH of 7.4–7.6.
[5].To inactivate the virus, aminoethylethyleneimine (AEEI) is used, which is added to the virus-containing suspension to a concentration of 0.025–0.05%. At the end of inactivation, AEEI is neutralized by adding sodium thiosulfate to the suspension [5].
0,005÷0,007% [6].The resulting antigen is purified from ballast impurities using polyhexamethylene guanidine (PHMG), which is added to the suspension to a concentration of 0.005–0.007% [6].
[7, 8].Avirulent and purified antigenic material is a suspension containing predominantly 146S and 75S immunogenic components of foot and mouth disease virus. At the same time, the production strain is presented with the requirements for the stability of the virus during purification from tissue components and concentration, as well as the preservation of the virus during inactivation and long-term storage [7, 8].
For the preparation of the vaccine, viral material is used that contains at least 0.5 μg of 146S and 75S immunogenic components of foot and mouth disease virus in 1 cm 3 .
The required concentration of 146S and 75S immunogenic components of foot and mouth disease virus in the proposed vaccine, comprising at least 3 μg in 1 cm 3 of the finished product, is obtained by adding the calculated amount of GOA sorbent adjuvant to the avirulent and purified antigenic material. The optimal GOA content in 1 cm 3 of the finished product in the range from 1132.0 μg to 2108.0 μg.
An additional 10% aqueous solution of ethonium or saponin is added to the resulting concentrate to a final concentration of 0.075%, which corresponds to 750.0 μg of its content in 1 cm 3 of the finished preparation.
Prepare a 10% solution of ethonia or saponin in distilled water. The solution is autoclaved at a temperature of 105 ° C for no more than 90 minutes. The sterile solution is stored at a temperature of 4-6 ° C.
1.The optimal component composition of the obtained sorbed vaccine against foot and mouth disease is presented in table 1.
It is permissible to change the composition of the vaccine and the percentage of its individual components, including etonia. It is permissible to change the prescription of the preparations themselves.
The resulting vaccine is a light yellow liquid with a loose precipitate of sorbent, which is formed at the bottom of the vial during storage and is easily broken into a homogeneous suspension with shaking.
After manufacture, the vaccine is subjected to control for sterility, safety and immunogenicity, after which it is released for practical use. Virulence, harmlessness and immunogenicity of the vaccine are tested on cattle.
The adjuvant properties of ethonia were determined on cattle weighing 250–300 kg. For this purpose, 2 preparations were prepared.
The drug No. 1 (content per vaccine dose):
- антиген культурального вируса ящура (КВЯ) типа Азия-1 Шамир 3/89 в количестве 6 мкг иммуногенных компонентов;- antigen of the culture of foot and mouth disease virus (CVD) type Asia-1 Shamir 3/89 in the amount of 6 μg of immunogenic components;
Этоний использовали ФС 42-1599-94, ОТК 6, партия 2 - Шосткинский завод химреактивов.- etonium in an amount of 3 mg. Etonius was used by FS 42-1599-94, OTK 6, batch 2 - Shostkinsky Chemical Reagents Plant.
Drug No. 2 (content per vaccine dose):
- антиген культурального вируса ящура типа Азия-1 Шамир 3/89 в количестве 6 мкг иммуногенных компонентов без этония (контроль).- FMD virus antigen of type Asia-1 Shamir 3/89 in the amount of 6 μg of immunogenic components without etony (control).
Preparations at a dose of 2 cm 3 were injected subcutaneously in the neck. The intensity of immunity was judged by titers of neutralizing antibodies in the blood serum of animals with homologous foot and mouth disease virus of type Asia-1 Shamir 3/89. It was believed that the higher the titers of neutralizing antibodies, the stronger the adjuvant effect due to etonium.
2.The results of the experiment are presented in table 2.
В эти же сроки титр антител у животных, привитых препаратом №2, не содержащим этония (контроль), был равен: через 5 суток - 1,92±0,08 log2, 10 суток - 4,00±0,14 log2, 17 суток - 4,17±0,22 log2, 30 суток - 3,00±0,14 log2.From table 2 it is seen that the introduction of etonium into the viral preparation contributes to the earlier formation of virus-neutralizing antibodies. So, the preparation No. 1 with etonium in the inoculated dose of 3 μg causes the formation of virus-neutralizing antibodies after 5 days in the titer of 3.33 ± 0.68 log 2 , 10 days - 4.25 ± 0.38 log 2 , 17 days 5, 08 ± 0.73 log 2 , 30 days - 4.00 ± 0.43 log 2 . At the same time, the antibody titer in animals inoculated with Etonium-free preparation No. 2 (control) was equal: after 5 days - 1.92 ± 0.08 log 2 , 10 days - 4.00 ± 0.14 log 2 , 17 days - 4.17 ± 0.22 log 2 , 30 days - 3.00 ± 0.14 log 2 .
Thus, the results of Table 2 demonstrate the good adjuvant properties of etonium when evaluated in cattle.
1.In the following experiments on cattle, the adjuvant properties of etonium were determined in comparison with imported saponin, manufactured by Merck Germany, as part of a sorbed FMD vaccine. Was prepared 4 series of vaccines, as described previously in example 1.
Series No. 1 (content per vaccination dose):
- антиген культурального вируса ящура типа Азия-1 Шамир 3/89 в количестве 5,5 мкг иммуногенных компонентов (146S+75S);- antigen of FMD virus of type Asia-1 Shamir 3/89 in the amount of 5.5 μg of immunogenic components (146S + 75S);

- этоний в количестве 1,5 мг.- etonium in an amount of 1.5 mg.
Series No. 2 (content per vaccination dose):
- антиген культурального вируса ящура Азия-1 Шамир 3/89 в количестве 5,5 мкг иммуногенных компонентов (146S+75S);- antigen of culture virus of foot and mouth disease Asia-1 Shamir 3/89 in the amount of 5.5 μg of immunogenic components (146S + 75S);
- ГОА в количестве 20 мг;- GOA in an amount of 20 mg;
- сапонин в количестве1,5 мг.- saponin in an amount of 1.5 mg.
Series No. 3 (content per vaccination dose):
- антиген культурального вируса ящура типа C564 в количестве 6 мкг иммуногенных компонентов (146S+75S);- antigen of the culture virus of foot and mouth disease type C 564 in the amount of 6 μg of immunogenic components (146S + 75S);
- ГОА в количестве 20 мг;- GOA in an amount of 20 mg;
- этоний в количестве 1,5 мг.- etonium in an amount of 1.5 mg.
Series No. 4 (content per vaccination dose):
- антиген культурального вируса ящура типа C564 в количестве 6 мкг иммуногенных компонентов (146S+75S);- antigen of the culture virus of foot and mouth disease type C 564 in the amount of 6 μg of immunogenic components (146S + 75S);
- ГОА в количестве 20 мг;- GOA in an amount of 20 mg;
- сапонин в количестве 1,5 мг.- saponin in an amount of 1.5 mg.
3, 4.The preparations were administered at a dose of 2 cm 3 to cattle subcutaneously in the neck. On tension immunity judged by neutralizing antibody titers in serum of animals with homologous FMD virus type Asia-1 and 3/89 Shamir type C 564. The results are presented in tables 3, 4.
As shown by the data presented in table 3, the adsorbed FMD vaccine of type Asia-1 Shamir 3/89 series No. 1 with etonium 1.5 mg in a vaccine dose causes the formation of virus-neutralizing antibodies: after 10 days in a titer of 5.80 ± 0.40 log 2 and after 21 days 5.75 ± 0.24 log 2 . The titer of virus-neutralizing antibodies in animals vaccinated with the Asia-1 Shamir 3/89 sorbed vaccine of series No. 2 with a 1.5 mg saponin in the vaccination dose was equal: after 10 days 5.70 ± 0.56 log 2 and 21 days 5, 60 ± 0.32 log 2 . By analyzing the titers of neutralizing antibodies, there is practically no significant difference between the adjuvant activity of etonium and saponin.
Table 4 shows the results of immunogenic activity of type C 564 adsorbed vaccine. Thus, the titer of virus-neutralizing antibodies in animals vaccinated with a vaccine of series 3 with etonium 1.5 mg in a vaccination dose is 5.40 ± 0.39 log 2 after 10 days and 5.90 ± 0.30 log 2 after 21 days. The titer of virus-neutralizing antibodies in animals vaccinated with the vaccine of series 4 with saponin 1.5 mg at the vaccination dose was equal: after 10 days 5.75 ± 0.64 log 2 and after 21 days 5.81 ± 0.41 log 2 . As can be seen from table 4, there is no significant difference between the adjuvant activity of etonium and saponin in this experiment either.
Thus, the drug etonium has good adjuvant activity in the composition of the sorbed FMD vaccine and meets all the requirements for adjuvants.
