Изобретение относится к области медицины, в частности к онкологии, дерматологии, косметологии, ветеринарной медицине и может быть использовано при разработке препаратов для лечения поверхностных опухолей кожи.The invention relates to medicine, in particular to oncology, dermatology, cosmetology, veterinary medicine and can be used in the development of drugs for the treatment of superficial skin tumors.
К настоящему моменту опубликовано значительное число работ по использованию наночастиц золота в различных областях медицины [8, 9, 10, 11]. Среди лечебных средств существуют препараты, в состав которых включены частицы золота или его соединения. Наночастицы золота представляют гаптены и антигены, усиливающие иммунный ответ и являющиеся носителями вакцин и лекарственных агентов для доставки в организм [12]. Известно, что золото обладает противовоспалительным, иммунодепресантным, противоопухолевым, антиферментным и некоторыми другими лечебными эффектами. Препараты золота используются в терапии больных ревматоидным и псориатическими артритами, синдромом Фелти, красной волчанкой [13, 14, 15, 16, 17, 18, 19, 20, 21, 22, 23, 24, 25, 26, 27, 28, 29, 30, 31, 32]. Исследованиями показана высокая эффективность использования наноразмерных частиц золота в терапии злокачественных новообразований кожи и ее придатков при помощи управляемой локальной гипертермии в эксперименте [4, 5].To date, a significant number of works on the use of gold nanoparticles in various fields of medicine have been published [8, 9, 10, 11]. Among the therapeutic agents, there are preparations, the composition of which includes particles of gold or its compounds. Gold nanoparticles represent haptens and antigens that enhance the immune response and are carriers of vaccines and drug agents for delivery to the body [12]. It is known that gold has anti-inflammatory, immunosuppressive, antitumor, antienzyme and some other therapeutic effects. Gold preparations are used in the treatment of patients with rheumatoid and psoriatic arthritis, Felty’s syndrome, lupus erythematosus [13, 14, 15, 16, 17, 18, 19, 20, 21, 22, 23, 24, 25, 26, 27, 28, 29 , 30, 31, 32]. Studies have shown the high efficiency of using nanoscale particles of gold in the treatment of malignant neoplasms of the skin and its appendages using controlled local hyperthermia in the experiment [4, 5].
Исследован парентеральный способ введения наночастиц золота в организм животных и человека, а также кинетика распределения наночастиц золота во внутренних органах [1, 2, 3, 33]. Однако этот способ введения и сами растворимые лекарственные формы наночастиц имеют ряд выраженных побочных эффектов. Наряду с положительными, терапевтическими эффектами накопления их в опухолевых тканях, включая кожные эпидермальные, наблюдаются и негативные - их накопление во внутренних органах (печень, селезенка, легкие, почки), приводят к нарушению органной и тканевой микроциркуляции, развитию в них дистрофических изменений [1, 2].The parenteral method of introducing gold nanoparticles into animals and humans has been studied, as well as the kinetics of the distribution of gold nanoparticles in internal organs [1, 2, 3, 33]. Along with the positive, therapeutic effects of their accumulation in tumor tissues, including epidermal skin, negative ones are also observed - their accumulation in internal organs (liver, spleen, lungs, kidneys) leads to disruption of organ and tissue microcirculation, development of dystrophic changes in them [1 , 2].

Для улучшения трансдермальной проницаемости дополнительно применяют ультрафонофорез [6]. Участки кожи после нанесения наночастиц золота в комплексе с сероорганическими соединениями подвергают ультразвуковому воздействию с частотой 1 МГц, мощностью 2 Вт и продолжительностью 2 минуты (ультразвуковой излучатель "Dinatron 125", фирмы Dinatronics.Co, USA).Ultrafonophoresis is additionally used to improve transdermal permeability [6]. В результате анализа описаний изобретений к патентам, отобранных при проведении патентного поиска, и на основании проведенных патентных исследований установлено, что лекарственные формы и составы для местного нанесения на кожу, содержащие наночастицы золота в комплексе с сераорганическими соединениями в патентной литературе не встречаются. На основании чего, можно считать, что различные решения в данной области могут быть патентоспособными и оформлены заявками на изобретения. При этом удалось избежать побочных эффектов накопления наночастиц во внутренних органах (печень, селезенка, легкие, почки), нарушения органной и тканевой микроциркуляции, развитие в них дистрофических изменений, которые обычно наблюдаются при внутривенном введении [6].The authors first studied the transdermal permeability of intact and damaged skin for gold nanoscale materials in combination with organosulfur compounds. В проведенном исследовании использовались мужские особи беспородных белых крыс, масса которых в среднем составляла 150-200 грамм. Животные содержались в соответствии с «Правилами Европейской конвенции по защите позвоночных животных, используемых для экспериментальных и иных научных целей». После депиляции на спине каждой из крыс проводилась разметка, в результате которой на коже животных из каждой группы был выделен участок, на который наносили гель с наночастицами и раствором диметилсульфоксида, либо тиофансульфоксида, после чего подвергали ультразвуковому воздействию.The study used male mongrel white rats, whose mass averaged 150-200 grams. По данным световой микроскопии разной степени увеличения, на препаратах кожи животных, подвергшихся ультразвуковому воздействию, как с нанесением только одного геля, так и в комплексе с сероорганическими соединениями, можно увидеть, что сероорганические соединения и ультразвуковое воздействие способствуют усилению проницаемости наноразмерных материалов через трансдермальный барьер (фиг.1, 2, 5, 6).
