Abstract
Description
The claimed technical solution relates to veterinary medicine, agriculture, medicine, biotechnology and the microbiological industry and can be used to protect against infections caused by DNA - and RNA-containing viruses.
The closest to the claimed technical solution according to the set of coinciding features and the achieved technical result is the drug "Betadin" - one of the few antiseptic and disinfectants for local and external use, characterized by the absence of type and species specificity for microorganisms. For example, Betadine, containing 10% of the active substance (povidone iodine), has a rapid bactericidal effect on gram-positive and gram-negative bacteria, and is also effective against fungi, protozoa and viruses [1-3], including human immunodeficiency viruses [3] . The tool is non-toxic and does not cause a mutagenic effect in relation to higher animals due to the peculiarities of their structure at the cellular level.
[3].The disadvantage of "Betadine" is that it can cause irritation of organs and tissues in contact with it, which limits the scope of its application. For example, Betadine is undesirable for use in pulmonary asthma, bronchitis, diseases of the mucous membrane of the eyes and hypersensitivity to iodine in general [1, 2]. Meanwhile, it is known that both the microbicidal effect and the irritating effect of Betadine depend on the same indicator: the content of active iodine [3]. In this regard, the dilution of "Betadine", which reduces its irritating effect, simultaneously leads to a decrease in its microbicidal effect, which is combined with the manifestation of type and species specificity. For example, a 10-1000-fold decrease in the content of the active substance did not adversely affect the level of bactericidal activity, while lowering its virucidal activity, especially with respect to the human immunodeficiency virus [3]. A further decrease in the content of the active substance also lowered its bactericidal effect [3].
It is probably possible to compensate for the decrease in effectiveness by supplementing the effect of one drug with another. For example, often improving the effect of biologically active substances is achieved by combining one substance with another.
However, “Betadine”, according to its description [1], is incompatible with other agents, especially those containing alkalis, enzymes and mercury, due to, apparently, worsening conditions for its activity to appear under the influence of alkali or a decrease in the content of active iodine under the action of mercury or biological catalysts - enzymes. Despite this, during the study, the result of which was the subject of the claimed technical solution, the applicant was able to successfully combine Betadine with an enzyme - bacterial endonuclease - and to obtain a combined preparation with a reduced irritation effect, the virucidal activity of which was higher than the virucidal activity of the repeatedly diluted Betadine ". The latter became possible, obviously, because endonuclease, itself possessing antiviral activity [4, 5], was not a biological catalyst with respect to active iodine.
Bacterial endonuclease, a product of the structural gene of genesis originally found in the bacterial genome of S. marcescens and later cloned and expressed in S. marcescens and E. coli cells [6], is a deoxyribonucleate (ribonucleate) - 5'-nucleotidohydrolase, which is why may exhibit catalytic activity with respect to active iodine. Bacterial endonuclease hydrolyzes nucleic acids by the endonucleolytic mechanism to mono-, di-, tri-, tetra- and oligonucleotides, with which its antiviral activity is probably associated. Bacterial endonuclease is known to inhibit the development of RNA genomic bacteriophages [4]. As part of the Endoglukin preparation, endonuclease is used for the treatment and prevention of bee viral paralysis [5]. In addition, as one of the active components, bacterial endonuclease is a part of an antimicrobial preparation, which has a wide spectrum of action, including an antiviral effect without pronounced species and type specificity [7]. Bacterial endonuclease is non-toxic and harmless to humans, animals and plants, as it is an enzyme, that is, a natural biological catalyst capable of spontaneous inactivation.
The objective of the claimed technical solution was the creation of a new drug based on Betadine, namely a drug with both reduced irritating effect and high virucidal activity.
[1-3].The claimed problem is solved by the applicant through the use of widely known from the prior art, as such, drugs "Betadine" and bacterial endonuclease. The claimed technical solution provides the possibility of solving a problem (goal), which was not solvable in principle for specialists in the claimed field of technology, since the results are not obvious to specialists, due to the contradictions described in the prior art [1-3].
The applicant has established that the half-inactivation time of the endonuclease upon contact with Betadine is 6 minutes, and therefore, to obtain the claimed technical solution, the components are mixed immediately before using an antiviral drug with a contact action in order to prevent a decrease in the enzymatic activity of the endonuclease due to preliminary contact with Betadine . An increase in the contact time with Betadine leads to a further decrease in the enzymatic activity of the endonuclease, up to its complete loss.
[1]."Betadine" is a solution for local and external use containing 10% povidone-iodine, as well as excipients: glycerin, nanoxynol 9, citric acid, disodium hydrogen phosphate, caustic soda and purified water [1].
[3]."Betadine" with a low content of active substance (povidone iodine) and, accordingly, auxiliary substances, was obtained by dilution with distilled water. The dilution effect on virucidal activity against phage λ bII was previously determined, the results of which are presented in Table 1. Published data were used to evaluate the content of active iodine [3].
2.An endonuclease preparation is a solution with an activity of at least 5000 units / ml, containing 0.01-0.10 M Tris-HCl buffer, pH 7.0-9.0, or 0.5-0.8% NaCl, and not less than 0.01 mol / l of magnesium cations. The applicant has established that the use of an endonuclease preparation with an activity of less than 5000 units / ml will also lead to the achievement of the result of the claimed technical solution. But this result will be worse than the results presented in table 2.
2.Applicant knows that endonuclease exhibits enzymatic activity at pH 7-10, with the best effect at pH 8.5. The applicant found that when mixing diluted Betadine with an equal volume of 0.5-0.8% NaCl or 0.10 M Tris-HCl buffer, pH 8.5, it is possible to obtain a composition with a pH of 7.3 or 7.9, which is sufficient to achieve the total effect of the claimed technical solution, which can be seen from the results presented in table 2. A decrease in the concentration of NaCl to 0.2% worsens the result of the claimed technical solution by lowering the pH of the medium. Increasing the concentration of NaCl to 17.5% does not reduce the result of the claimed technical solution, however, it takes the composition of the claimed technical solution beyond the physiological values for the content of NaCl. A further increase in the content of NaCl is impractical, firstly, for the reason noted above, and secondly, because it leads to a decrease in the enzymatic activity of endonuclease. 0.01-0.10 M concentration of Tris-HCl buffer is optimal to achieve the result of the claimed technical solution. Increasing or decreasing the concentration of the buffer will also lead to the achievement of the result of the claimed technical solution. But this result will be worse than the results presented in table 2.
The applicant is aware that the increase in the content of magnesium cations in the claimed technical solution will not have a negative effect on the result. On the contrary, a decrease in the magnesium content will worsen the result up to approaching the result of the action of diluted Betadine. In the absence of magnesium cations, the result achieved will be equal to the result of the action of diluted Betadine and is due to this action. Thus, the applicant was able to combine two drugs belonging to different classes of compounds, which together gave a mutual improvement in the biological effect - virucidal activity - and allowed to reduce the irritant effect of Betadine.
The claimed technical solution is illustrated by examples and information given in tables 1 and 2. Table 1 presents the virucidal activity of Betadine diluted with distilled water 10-1000000 times, in relation to phage λ bII. Table 2 shows the results of the claimed technical solution in comparison with the action of endonuclease and diluted "Betadine" in various ratios.
The claimed technical solution is implemented as follows.
Антивирусный препарат контактного действия, который содержал 0,005-0,000005% повидон-йод, 5,0 ммоль/л катионов магния, 25 мМ Трис-HCl буфер, рН 8,0, или 0,20% NaCl и бактериальную эндонуклеазу, готовили непосредственно перед использованием, смешивая в соотношении 1:1 «Бетадин» с пониженным содержанием повидон-йода и препарат эндонуклеазы (пример 1).A contact-action antiviral preparation that contained 0.005-0.000005% povidone iodine, 5.0 mmol / L magnesium cations, 25 mM Tris-HCl buffer, pH 8.0, or 0.20% NaCl and bacterial endonuclease, were prepared directly before use, mixing in a ratio of 1: 1 "Betadine" with a low content of povidone-iodine and the preparation of endonuclease (example 1).
The antiviral effect of the contact preparation was tested in direct contact with a suspension of phage λ bII, mixing them in a 1: 1 ratio (Example 1) and incubating the mixture at room temperature for 6 minutes. Similarly and simultaneously with an antiviral drug with a contact action, the virucidal effect of a bacterial endonuclease solution containing 5.0 mmol / L magnesium cations, 25 mM Tris-HCl buffer, pH 8.0, or 0.20% NaCl, and Betadine with a reduced the content of povidone iodine taken separately. Virucidal activity was evaluated by monitoring the change in the number of sterile spots determined by the Grazia method using E. coli BL21 (DE3) pLysS as a test culture. An increase in the incubation time improved the results presented in Table 2. In particular, incubation for 6 hours improved the result of the diluted Betadine by 4.5 times, the bacterial endonuclease solution containing 5.0 mmol / L magnesium cations, 25 mM Tris- HCl buffer, pH 8.0, or 0.20% NaCl - 3.5 times, and the contact-action antiviral drug, which contained 0.005-0.000005% povidone-iodine, 5.0 mmol / L magnesium cations, 25 mM Tris-HCl buffer, pH 8.0, or 0.20% NaCl - 8.5 times. Incubation for 24 hours allowed to achieve 100% effect. Reducing the incubation time worsened the result. So, incubation for 4 minutes led to a deterioration of the result by 1.5 times, and within 2 minutes - by 1.87 times.
2.Virucidal activity of the claimed technical solution (antiviral drug contact action) and its active components are presented in table 2.
7-9.From the above examples in table 2 it is seen that the changes in the ratios “Betadine: with a low content of povidone iodine: the preparation of Endonuclease” by 5 and 500 times, as well as the “preparation of Endonuclease to Betadine with a low content of povidone-iodine” by 5 and 500 times does not affect the result - a decrease in viability, provided that the content of povidone iodine in the diluted Betadine is 0.005-0.000005%, and the pH of the solution is 7-9.
Thus, the results of the experiments shown in the materials of table 2, confirms the fact that the claimed technical solution can be achieved with different quantitative ratios of the components of the claimed drug.
The ratio "Betadine: low in povidone iodine: Endonuclease preparation" can vary widely.
Since endonuclease is an inhibitor of RNA genomic bacteriophages [4], and phage λ bII refers to DNA genomic viruses, a contact-action antiviral drug (the claimed technical solution) is effective against DNA and RNA viruses.
The claimed technical solution (antiviral drug of contact action) is a fundamentally new drug and is a non-toxic environmentally friendly substance that is active under mild physiological conditions. Thus, the data obtained show the possibility of using the drug as a universal remedy for infections of viral etiology in various areas of the national economy.

The claimed technical solution meets the criterion of "inventive step" for inventions, since the claimed technical solution does not follow explicitly from the prior art for a person skilled in the art, moreover, the applicant was able to combine two drugs in one preparation, belonging to different classes of compounds, basically incompatible, which is known from the studied prior art, while the applicants obtained results that are not obvious to a specialist from a known level of technology ki, which is additional proof of compliance of the claimed solution to the criterion "Inventive Step" since the results obtained were given in combination the relative improvement of the biological effect - virucidal activities - and allowed to reduce the irritant effect of the agent "Betadine".
The claimed technical solution meets the criterion of "industrial applicability", as it can be implemented in the national economy using standard equipment and tools.
