Description
The invention relates to the field of ceramic materials for medicine, namely to traumatology and reconstructive surgery, dentistry and to a drug delivery system.
В результате получают пористые сферические гранулы с регулируемым размером и открытой пористостью 20-80 об.%.A known method of manufacturing granules of hydroxyapatite (HA) (RF patent No. 2299869, СВВ 35/447, 35/626, A61L 27/12, publ. 2007.05.27.), Which consists in the preliminary synthesis of calcium phosphate powder with a ratio of Ca / P from 1 , 5 to 1.67, preparing a suspension with a 10% gelatin solution per 1 g of powder at a solution temperature of 10-39 ° С, a suspension of HA in an aqueous gelatin solution was dispersed in a neutral liquid medium of vegetable oil, the mixture was stirred with a paddle mixer with a rotation speed 100-1500 rpm Under the influence of surface tension, spherical granules were formed, which were washed, dried and subjected to heat treatment at a temperature of 900-1250 ° C. The result is porous spherical granules with an adjustable size and open porosity of 20-80 vol.%.
A disadvantage of the known invention is the production of granules, the sizes of which depend on the surface tension forces in the medium of vegetable oil and are limited by their size. In addition, artificial HA is used for the manufacture of hydroxyapatite granules, for the preparation of which it is necessary to synthesize it.
В результате получают керамику с бимодальным распределением пор в суммарном количестве от 41 до 70 об.%.Closest to the proposed invention is a method for the manufacture of hydroxyapatite ceramics with a bimodal pore distribution (RF patent No. 2303580, С04В 35/447, A61L 27/12, publ. 2007.07.27.), Which consists in the manufacture of spherical granules with a diameter of 400-600 microns, containing hydroxyapatite powder and gelatin by the method using the effect of immiscible liquids, pressing spherical granules under a pressure of 10-100 MPa and heat treatment at temperatures of 900-1250 ° C with a holding time of 30 to 300 minutes. The result is ceramics with a bimodal pore distribution in a total amount of from 41 to 70 vol.%.
A disadvantage of the known invention is that the granules obtained by dispersing a liquid into a free volume (into oil) have certain sizes. In addition, it is necessary to filter and then wash the granules from the oil. The present invention uses artificial (synthetic) hydroxyapatite.
The objective of the invention is to obtain granules of hydroxyapatite of natural origin, the sizes of which can be set in advance in a wide range. The use of hydroxyapatite of natural origin increases its biocompatibility with a living organism.
Технический результат предлагаемого изобретения - гранулированный порошок гидроксиапатита естественного происхождения с заданным размером гранул в широком диапазоне от 0,1 мм до 2 мм, которые содержат сообщающиеся между собой внутригранульные поры размером 100-350 мкм в количестве от 50 до 70 об.%.The technical result of the invention is a granular powder of natural hydroxyapatite with a given granule size in a wide range from 0.1 mm to 2 mm, which contain intergranular pores of 100-350 microns in size in an amount of 50 to 70 vol.% Communicating with each other.
The specified technical result is achieved by the fact that in the method of manufacturing porous hydroxyapatite granules containing intergranular pores interconnected, consisting in the formation of granules with subsequent heat treatment, the granules are formed by extrusion of a plastic mass containing 1 parts by weight paraffin and 3 parts by weight hydroxyapatite powder obtained from natural bone biomaterial by annealing at a temperature of 950-1000 ° C and its subsequent grinding to obtain powder particles with a size of not more than 35 microns, then the obtained granules are subjected to heat treatment at a temperature of 750-800 ° C.
To prepare the plastic mass, paraffin is used, heated to a melting point of 50-55 ° C
Grinding of bone biomaterial is carried out in a ball mill for 45-50 hours.
Extruding the plastic mass is carried out through a matrix with a mesh size of 800-2000 microns.
Heat treatment of granules is carried out within 1-2 hours
Granules contain interconnected pores with a size of 100-350 microns in an amount of from 50 to 70 vol.%.
- 311 с.].The main requirements for medical supplies are their biocompatibility and the presence of interconnected pores with an average size of mainly 100-350 microns, which do not impede the mandatory penetration of blood cells to the contact surfaces of bone tissue with its subsequent germination [I.F . Obraztsov, IS Adamovich and others. Problems of strength in biomechanics. - M .: Higher school, 1988. - 311 p.].
Based on numerous studies on the use of ceramic materials in medicine, it has been established that hydroxyapatite belongs to the class of bioactive materials with surface-active properties and high biocompatibility. The most convenient form for use are granules. Granulation is carried out in order to improve the technological properties of the powder, flowability, fluidity, prevent sticking, delamination of molded products, increase the bulk density of the powders, facilitates loading and transportation.
The production of granules from synthesized hydroxyapatite is currently quite well mastered on a large scale. Obtaining granules from natural hydroxyapatite suggests increasing biocompatibility.
For this, Ca 5 (PO 4 ) 3 OH powder was prepared from a biomaterial representing cattle bones, which was annealed in order to remove the organic component at a temperature of 950-1000 ° С.
This temperature is sufficient to completely remove the organic component and complete crystallization of amorphous forms of calcium phosphate compounds.
Then, the annealed biomaterial was ground for 45-50 hours in a ball mill to obtain particles no larger than 35 microns in size. This particle size of the powder is optimal for subsequent activation of the granulation process.
ч. To obtain a plastic mass of 3 parts by weight annealed obtained natural hydroxyapatite powder of natural origin is introduced heated to a melting point (55 ° C) of 1 hour paraffin wax.
Such a ratio of components is necessary for optimal consolidation of powder particles for the formation of porous hydroxyapatite granules during heat treatment, as well as the production of hydroxyapatite granules containing interconnected intragranular pores in the required quantity and certain sizes.
Next, the extrusion of the plastic mass is carried out through a matrix, the cell size of which is selected depending on the required size of the finished granules, taking into account shrinkage during the annealing.
In order to remove paraffin, consolidate powder particles and ensure the mechanical properties of the granules, heat treatment is carried out in an oven at a temperature of 750-800 ° C with an isostatic exposure for 1-2 hours.
The selected temperature is necessary and sufficient, in addition, for the formation of pore space in the granules.
The formed pore space in the granules provides unobstructed mandatory penetration of blood cells to the contact surfaces of bone tissue with its subsequent germination.
Оценка размеров пор и их количества проводилась методом хорд, получаемых пересечением поверхности шлифа случайными секущими согласно ГОСТ-5639-82.The obtained hydroxyapatite granules contain intergranular pores interconnected with a size of 100-350 microns in an amount of from 50 to 70 vol.%. Evaluation of pore sizes and their number was carried out by the method of chords obtained by crossing the surface of a thin section with random secants according to GOST-5639-82.
