Суть метода состоит в том, что обнаружение антигенов вирусов оспы овец, находящихся в цитоплазме клеток органов и тканей овец, пораженных оспой, проводят непосредственно в отобранных образцах. Для этого пораженные оспой органы и ткани овец фиксируют формалином и готовят парафиновые блоки. После приготовления срезов из тканей и органов и их депарафинизации проводят детекцию антигенов. С этой целью вначале на поверхность среза наносятся специфические к антигенам вируса оспы овец иммуноглобулины, выделенные из гипериммунной сыворотки кролика. Затем для усиления положительного сигнала на поверхность среза наносятся биотинилированные анти-кроличьи антитела. К ним относятся применение поликлональных иммуноглобулинов, выделенных из гипериммунных сывороток, которые вступают в перекрестные реакции с антигенами вируса контагиозного пустулезного дерматита овец и коз и могут быть причинами фонового неспецифического окрашивания, вследствие применения низкоочищенных антигенов при получении гипериммунных сывороток. Данные причины приводят к снижению диагностической ценности метода, однако устранить их возможно, используя моноклональные антитела, обладающие специфичностью к антигенным детерминантам вирусов оспы овец и коз. Способ получения пригодных для этих целей моноклональных антител и их характеристика описаны ранее (Машнин А.В., , Стрижаков А.А., Стрижакова О.М., Новикова М.Б., Середа А.Д. Разработка средств диагностики оспы овец на основе моноклональных антител // Диагностика, профилактика и меры борьбы с особо опасными, экзотическими и зооантропонозными болезнями животных. При постановке гистохимического иммуноферментного анализа готовили тест-препараты. Для этого к монослойным первично-трипсинизированной культуре клеток тестикул ягненка (ТЯ) и перевиваемой культуры клеток почки овцы (ПО), выращенным на покровных стеклах в пробирках, после удаления культуральной среды и трехкратного омовения монослоя средой для культивирования без сыворотки добавляли осветленные суспензии двукратных разведении контрольных культуральных (нормального и специфического) и исследуемых органотканевых антигенов. Контакт клеток с исследуемыми материалами осуществляли в течение 1 ч при 37(±1)°С. После чего супернатант удаляли, клетки двукратно омывали средой для культивирования без сыворотки. Дальнейшее культивирование клеток осуществляли с добавлением 2% сыворотки овцы и антибиотиков (100 ЕД/см3 пенициллина, 100 мг/см3 стрептомицина) при 37(±1)°С на протяжении 5-6 сут. Затем клеточный монослой, выращенный на предметных стеклах, фиксировали в течение 10 мин при комнатной температуре 100%-ным ацетоном, охлажденным до минус 20°С. Конъюгат инкубировали в течение 1 ч при 37(±1)°С в условиях влажной камеры. Непровзаимодействовавший конъюгат двукратно отмывали по 3-5 мин в отмывочном буфере и однократно ЗФР. В работе по определению чувствительности и специфичности данного метода использовали тест-препараты, представляющие фиксированные интактные и инфицированные культуры клеток ТЯ и ПО гомологичными вирусами, выделенными из подкожной клетчетки с мест оспенного поражения, хоан и пораженных участков легких инфицированных животных, а также гетерологичными вирусами мелких жвачных (вирусом контагиозного пустулезного дерматита овец и коз, катаральной лихорадки овец, чумы мелких жвачных).
