Изобретение относится к медицинской биотехнологии и биологической промышленности, ветеринарии и касается усовершенствованного способа получения ферментативного гидролизата, а также питательной среды на его основе, предназначенной для выращивания клеток эукариотов продуцентов биологически активных веществ, диагностических и профилактических биологических и медицинских препаратов. Известен способ получения ферментативного гидролизата путем гидролиза в водопроводной воде измельченных органов и тканей ластоногих в присутствии ферментсодержащих органов животных того же вида при нагревании до 50oC [1] Полученный продукт предназначен для культивирования только микроорганизмов и, соответственно, содержит высокомолекулярные полипептиды. Недостатком является сложная технология ее изготовления из дорогостоящих, главным образом импортных реагентов. В условиях биотехнологического производства, где требуется большое количество питательной среды для получения клеточной биомассы, ПС 199 нерентабельна.
Задачей изобретения является упрощение технологии получения ферментативного гидролизата и снижение себестоимости конечного продукта - основы питательной среды, а также создание новой питательной среды Целат для культивирования клеток эукариотов. Измельченную мышечную ткань ластоногих (например, нерпы), разведенную дистиллированной водой в массовом соотношении 1:2, подвергают гидролизу ферментсодержащими органами животных того же вида, взятыми в количестве 15 об. Процесс ведут при температуре 40oC в щелочной зоне рН 7,6 в течение 5 суток в присутствии 1,0 1,5 об. хлороформа. Гидролиз прекращают прогреванием при температуре 80oC в течение 10 минут. Осадок удаляют центрифугированием. Оставшиеся в надосадочной жидкости высокомолекулярные соединения осаждают сначала в присутствии 20%-ного раствора хлористого кальция, а затем в изоэлектрической точке белка. Ферментативный гидролиз мышечной ткани ластоногих проводят аналогично примеру 1 с тем отличием, что процесс ведут при температуре 42oC в щелочной зоне 7,8 в течение 4 суток. Гидролиз прекращают прогреванием биомассы при температуре 90oC в течение 5 минут.
