Abstract
Description
The invention relates to the production of biologically active substances obtained by microbiological synthesis, and can be used for the production of purine-containing products (inosine, guanosine, hypoxanthine). These products can be used to obtain riboxin, adenosine and derivatives based on them, as well as growth-promoting feed additives for farm animals and poultry.
Known methods for producing purine-containing products and nucleosides by microbiological synthesis followed by isolation and chemical purification.
In the method for producing purine-containing products (ed. St. N 675651), taken as a prototype, purine-containing products are obtained by treating the culture fluid with aluminum sulfate to bring the pH to 5.2-5.5, separating the supernatant (native solution), extracting purines using carbon-containing sorbents by concentration by evaporation of the alcohol-ammonia eluate and crystallization of the product upon cooling to obtain a crude product, which is then dissolved and purified to produce riboxin using sulfotcationites.
невысокий выход целевых продуктов из-за многоступенчатой очистки;
The disadvantages of this and other known methods are:
low yield of target products due to multi-stage purification;
the presence of non-utilizable wastes in the form of depleted purine-containing solutions and wastes in the form of acid-base wastes generated from solutions used in the regeneration of ion-exchange resins.
The purpose of the invention is to increase the yield of target products.
For this purpose, a method for producing purine products by fermentation of strains - producing, separating the biomass and concentration, the concentration of the native solution is carried out until the kinematic viscosity of the concentrate with further 2-10 cst its crystallization at a temperature of
It was possible to establish that the concentration of the native solution and subsequent crystallization under the selected conditions make it possible to obtain the first purine-containing product with a purine content of 60-90%. Such a simple and effective process is caused by the fact that crystallization of purines and related impurities substantially depends, on the one hand, on the viscosity of the concentrate obtained, on the other hand, on the temperature and pH during crystallization. Depending on the relative content of purines and impurities in the initial native solution, the optimal degree of concentration was selected, which is limited by the limiting values of the kinematic viscosity coefficient, as well as the temperature and pH of crystallization. Justification of the optimal conditions for concentration and crystallization are given in table. 1-4. At a high content of purine and low impurity concentration degree can be high and the crystallization temperature of 0-20 C, otherwise (the inverse ratio of the target product and impurities) the degree of concentration below the crystallization temperature - 20-0 ° C
Thus, due to the selective crystallization of the target product, it is possible to obtain the first purine-containing product, which can be purified by known methods to obtain inosine (riboxin), guanosine, hypoxanthine or transformed with obtaining a simple purification and purification procedure with at least 60% yield, to obtain adenosine, ribose and other products based on it.
The remaining product from the first crystallization mother liquor containing the purine compound, dehydrated at 60-180 ° C to produce a second product purine, wherein the purine content of 5-15%. The specified product can be used as a feed additive. Thus, the yield of the target products increases to 90% (in the prototype up to 60%), production waste and saline effluent are excluded.

With an increase in the kinematic viscosity coefficient of one stripped off native solution of more than 10 cSt, the crystallization time of the first purine-containing product sharply increases and the yield decreases.
