Abstract

The expression vector replicable in a eukaryotic host cell comprises: a) a transcription promoter; b) a first DNA segment encoding a secretory leader; c) a second DNA segment encoding a thrombopoietin polypeptide backbone consisting of C-X-B, wherein C is a human thrombopoietin cytokine domain polypeptide; X is a peptide bond or a linker consisting of one or two amino acid residues, subject to the limitation that X, alone or in combination with C or B, does not provide a dibasic amino acid pair; and B is a polypeptide consisting of residues 1 to y of SEQ ID NO:3, wherein y is an integer from 5 to 18, and wherein up to 35 % of the amino acid residues of B are individually replaced by other amino acid residues and d) a transcription terminator. The polypeptides are characterized by an amino acid backbone consisting of C-X-B wherein: C is a human thrombopoietin cytokine domain polypeptide; X is a peptide bond or a linker consisting of one or two amino acid residues, subject to the limitation that X, alone or in combination with C or B, does not provide a dibasic amino acid pair; and B is a polypeptide consisting of residues 1 to y of SEQ ID NO:3, wherein y is an integer from 5 to 18, and wherein up to 35 % of the amino acid residues of B are individually replaced by other amino acid residues. The polypeptides are useful with methods of stimulating proliferation or development of hematopoietic cells in vitro and in vivo.
