Description
1. OPPFINNELSENS BAKGRUNN
Den foreliggende oppfinnelsen er relatert til (-)-l-(3,4-diklorfenyl)-3-azabicyklo[3.1.0]heksan eller farmasøytisk akseptable salter derav, fremgangsmåter for deres fremstilling, sammensetninger samt nevnte forbindelse for behandling eller forebyggelse av en lidelse lindret ved inhibering av dopamin-reopptak.
2. OPPFINNELSENS BAKGRUNN
Dopamin er en monoamin-neurotransmitter som spiller en kritisk rolle i funksjon av hypotalamus-hypofysen-adrenalaksen og ved integreringen av informasjon i sensoriske, limbiske og motorsystemer. Den primære mekanismen for terminering av dopamin-neurotransmisjon er gjennom gjenopptak av frigitt dopamin ved Na<+>/Cl"avhengig plasmamembrantransportere (Hoffman et al, Front. Neuroendocrinol. 19(3):187-231). Avhengig av de omkringliggende ioniske betingelsene kan dopamintransporteren fungere som en mediator av både innoverrettet direkte dompamintransport (dvs "gjenopptak") og utoverrettet direkte dopamintransport (dvs "frigivelse"). Den funksjonelle betydningen av dopamintransporteren er dens regulering av dopamin-neurotransmisjon ved terminering av virkningen av dopamin i en synapse via gjenopptak (Hitri et al, 1994, Clin. Pharmacol. 17:1-22).
"Attention deficit disorder" er en læringsvanskesykdom som involverer utviklingsmessig uegnet uoppmerksomhet med eller uten hyperaktivitet. De primære tegnene på "Attention deficit disorder" (AAD) er en pasients uoppmerksomhet og impulsivitet. Dcke-passende uoppmerksomhet forårsaker økt rate av aktivitet eller ulyst til å delta eller respondere. En pasient lidende av ADD viser et konsistent mønster av uoppmerksomhet og/eller hyperaktivitet-impulsivitet som er mer frekvent og uttalt enn typisk observert hos individer på et sammenlignbart utviklingsnivå. Pasienter med Parkinson's sykdom har skjelvende bevegelser av lemmene, hode og kjeve. Parkinson's sykdom er forbundet med bradykinsi, stivhet og fall (Stacy et al, 1996, Am. Fam. Phys. 53:1281-1287). Bevegelsesforstyrrelsene observert hos Parkinson's sykdom-pasienter er et resultat av degenerering av dopaminneuroner, tap av nerveterminaler og dopaminmangel. Det er antatt at årsaken til degenerering av dopaminneuronene er et resultat av apoptose på grunn av forøkte nivåer av cytokiner (Nagatsu et al, 2000, J. Neurak Transm. Suppl 60:277-290). Abnormaliteter i dopamintransporteren har blitt implikert i Parkinson's sykdom (Hitri et al, 1994, Clin. Neuropharmacol 17:1-22). Symptomer på Parkinson's sykdom kan svekkes ved forbindelser som pergolid som etterligner virkningene av dopamin eller av forbindelser som inhiberer dopaminmetabolisme (f.eks. karbidopa) eller av dopaminforløpere (f.eks. L-DOPA ± karbidopa).
Appetittsuppresjon er en reduksjon, eller en minskning eller, i tilfeller av overdreven matkonsumpsjon, en lindring av appetitt. Denne suppresjon reduserer lysten eller trangen til mat. Appetittsuppresjon kan resultere i vekttap eller vektkontroll som ønsket. Appetittsuppresjon kan regulere matinntak gjennom legemiddeladministrasjon direkte til en eller flere systemer kjent å spille en rolle ved matfordøyelse. Andedoni er en av de prinsipielle (kjerne) symptomene på depresjon. Dopamin-reaksjonsveiene har blitt forbundet med lystsøkende oppførsel og strategier for å øke synaptiske konsentrasjoner av dopamin har blitt foreslått som antidepressive terapier. (Se f.eks. Mange legemidler kan forårsake fysisk og/eller psykologisk avhengighet. De mest kjente legemidlene inkluderer opiater, slik som heroin, opium og morfin; sympatomimetiks, inkluderende kokain og amfetaminer; sedative-hypnotika, inkludert alkohol, enzodiazepiner og barbiturater; og nikotin, som har en virkning lik opioider og sympatomimetiks. Legemiddelavhengig karakteriseres av en trang eller tvang til å ta legemiddelet og en manglende evne til å begrense dets opptak. I tillegg er legemiddelavhengighet forbundet med legemiddeltoleranse, tap av virkningen av legemiddelet etter gjentatt administrering, og abstinens, tilsynekomsten av fysiske og adferdssymptomer når legemiddelet ikke konsumeres. Toleranse, sensitivisering og abstinens er fenomener som påviser en endring i sentralnervesystemet som et resulat av kontinuerlig anvendelse av legemiddelet. Denne endring motiverer det avhengige individet til å fortsette konsumering av legemiddelet tross alvorlige sosiale, legale, fysiske og/eller yrkesmessige konsekvenser. (Se, f.eks. U.S. patent nr. 6.109.269 til Rise et al). Kokainavhengighet er en av de mest alvorlige helseproblemene i de Forente Stater. Fundamentale studier fra mange laboratorier har vist at kokain blokkerer opptaket av dopamin fra den synaptiske kløften til dopamintransporteren (Kreek, 1996, J. Addict. Dis. 15:73-96). For eksempel, inhiberingsvirkningen av kokain på reopptak av frigitt dopamin forklarer imidlertid ikke fullt ut utviklingen og opprettholdelsen av addiktiv oppførsel. Visse farmasøytiske midler har vært administrert for behandling av avhengighet. U. S.
Patent nr. 5.556.838 til Mayer et al. Beskriver anvendelsen av ikke-toksiske NMDA-blokkerende midler ko-administrert med en avhengighetsskapende substans for å forebygge utviklingen av toleranse eller abstinenssymptomer. U.S. patent nr. 5.574.052 til Rose et al beskriver ko-administrering av en avhengighetsskapende substans med en antagonist for å partielt blokkere de farmakologiske virkningene av substansen. Beskriver anvendelse av en serotoninantagonist forbehandling av kjemisk avhengighet. U.S. patent nr 5.556.837 til Nestler et al Viser infusjon av BDNF eller NT-4-vekstfaktorer for å inhibere eller reversere neurologiske adaptive endringer som korrelerer med adferdsendringer hos et avhengig individ. U.S. patent nr. 5.762.925 til Sagan fremlegger implantasjon av innkapslede adrenalmedullare celler inn i en pasients sentralnervesystem for å inhibere utviklingen av opoidintoleranse. Bupropion har dopamin-reopptaksinhiberingsegenskaper og anvendes for å behandle nikotinavhengighet.
Dopaminerge "reward" reaksjonsveier har blitt implikert i lidelser forårsaket av addiktiv adferd. Varianter av D2-reseptorgenet har blitt forbundet med alkoholisme, fedme, patologisk spilling, ADD, Tourette-syndrom, kokainavhengighet, nikotinavhengighet, polysubstansmisbruk og andre stoffavhengigheter (Noble, 1994, Alcohol Supp. 2:35-43 og Blum et al, 1995, Pharmacogenetics 5:121-141). Ettersom reduserte dopaminerge funksjoner har blitt funnet hos individer med et lite Al allel av dopamin D2-reseptoren har blitt foreslått at dopamin D2-reseptoren kan være et forsterkings- eller belønnings-("reward") gen. (Noble, 1994, Alcohol Supp. 2:35-43). Videre antyder flere studier at en samling dopamin D2-reseptorgenpolymorfismer assosieres med impulsiv-avhengighets-tvangsadferd, dvs "Reward Deficiency Syndrome" (gjennomgått av Blum et al, 1995, Pharmacogenetics 5:121-141).
U.S. patent nr. 4.435.419 til Epstein et al. fremlegger racemisk (+)-l-(3,4-diklorfenyl)-3-azabicyklo[3.1.0]heksan for anvendelse som et anti-depressivt middel.
U.S. patent nr. 6.204.284 til Bee et al. fremlegger racemisk (±)-l-(3,4-diklorfenyl)-3-azabicyklo[3.1.0]heksan for anvendelse ved forebyggelse eller lindring av abstinens-syndrom forårsaket av avhengighet av legemidler og for behandlingen av kjemiske avhengigheter.
Administrering av racemisk, dvs 50:50, blanding av (+)- og (-)-enantiomerer av et hvilket som helst legemiddel, f.eks. (±)-l-(3,4-diklorfenyl)-3-azabicyklo[3.1.0]heksan til en pasient kan være ufordelaktig. For det første, den racemiske blandingen kan være mindre farmagologisk aktiv enn en av dens enantiomerer, noe som gjør racemiske legemidler iboende ineffektive. For det andre, den racemiske blandingen kan være mer toksisk for en pasient enn en av dens enantiomerer, slik at administrering av en racemisk blanding kan føre til uønskede bivirkninger hos en pasient.
I henhold til dette er det et klart behov innenfor faget for en enantiomer, hvor enantiomeren fortrinnsvis er i det vesentlig fri for tilsvarende motsatte enantiomerer, som ville overstige en eller begge av de nevnte ulempene.
3. OPPSUMMERING AV OPPFINNELSEN
Et første aspekt ved foreliggende oppfinnelse vedrører (-)-l-(3,4-diklorfenyl)-3-azabicyklo[3.1.0] heksan eller et farmasøytisk akseptabelt salt derav, hvor hver ikke har mer enn 5% w/w av dets korresponderende (+)-enantiomer.
Et andre aspekt ved foreliggende oppfinnelse vedrører en sammensetning omfattende en effektiv mengde av forbindelsen ifølge det første aspekt ved oppfinnelsen.
Et tredje aspekt ved oppfinnelsen vedrører forbindelsen ifølge det første aspekt ved oppfinnelsen for behandling eller forebyggelse av en lidelse lindret ved inhibering av dopamin-reopptak.
Et fjerde aspekt ved foreliggende oppfinnelse vedrører en fremgangsmåte for oppnåelse av forbindelsen ifølge det første aspekt ved foreliggende oppfinnelse, hvor fremgangsmåten omfattende trinnene: (a) føre en løsning av en organisk eluent og (±)-l-(3,4-diklorfenyl)-3-azabicyklo[3.1.0]heksan over en kiral polysakkaridstasjonær fase for å frembringe en første fraksjon inneholdende (-)-l-(3,4-diklorfenyl)-3-azabicyklo[3.1.0]heksan; og (b) føre den første fraksjonen over den kiral polysakkaridstasjonære fasen for å
frembringe en andre fraksjon inneholdende (-)-l-(3,4-diklorfenyl)-3-azabicyklo[3.1.0]heksan som ikke har mer enn 5% w/w av det korresponderende (+)-enantiomer.
Et femte aspekt ved oppfinnelsen vedrører en fremgangsmåte for oppnåelse av forbindelsen ifølge det første aspekt ved foreliggende oppfinnelse, hvor fremgangsmåten omfattende trinnene: (a) føre en løsning av den organiske eluent og (±)-l-(3,4-diklorfenyl)-3-azabicyklo[3.1.0]heksan over en kiral polysakkaridstasjonær fase for å frembringe en første fraksjon inneholdende (-)-l-(3,4-diklorfenyl)-3-azabicyklo[3.1 .Ojheksan;

Det har uventet blitt oppdaget at bivirkninger slik som kardiovaskulære virkninger, søvnforstyrrelser, hypertensjon eller seksuell dysfunksjon forbundet med norepinefrin-eller serotoninopptaksinhibitorer kan unngås ved bruk av (-)-l-(3,4-diklorfenyl)-3-azabicyklo[3.1.0]heksan eller et farmasøytisk akseptabelt salt derav i det vesentlig fri for dets korresponderende (+)-enantiomer.

4. DETALJERT BESKRIVELSE AV OPPFINNELSEN
4.1. DEFINISJONER
Begrepet "i det vesentlig fri for dets tilsvarende (+)-enantiomer" betyr inneholder ikke mer enn omkring 5% v/v av den tilsvarende (+)-enantiomer, fortrinnsvis ikke mer enn omkring 2% ved v/v av den tilsvarende (+)-enantiomer, mer foretrukket ikke mer enn omkring 1% v/v av den tilsvarende (+)-enantiomer.
Begrepet "tilsvarende (+)-enantiomer" når anvendt i forbindelse med (-)-l-(3,4-diklorfenyl)-3-azabicyklo[3.1.0]heksan eller et farmasøytisk akseptabelt salt derav betyr "(+)-'l-(3,4-diklorfenyl)-3-azabicyklo[3.1.0]heksan" eller et farmasøytisk akseptabelt salt derav.
En "pasient" er et dyr, inkludert, men ikke begrenset til, et dyr slik som ku, ape, hest, sau, gris, kylling, kalkun, vaktel, katt, hund, mus, rotte, hare og marsvin, og er mer foretrukket et pattedyr, mest foretrukket et menneske.
Frasen "farmasøytisk akseptabelt salt" som anvendt heri er et salt dannet fra en syre og den basiske nitrogengruppen til (-)-l-(3,4-diklorfenyl)-3-azabicyklo[3.1.0]heksan. 4.2. (-)-l-(3,4-DIKLORFENYL)-3-AZABICYKLO[3.1.0]HEKSAN (-)-l-(3,4-diklorfenyl)-3-azabicyklo[3.1.0]heksan fortrinnsvis det i det vesentlig fri for dets tilsvarende (+)-enantomer, kan oppnås fra (±)-l-(3,4-diklorfenyl)-3-azabicyklo[3.1.0]heksan ved å bruke kiralkromatografimetoder, slik som høyytelsevæskekromatografi ("HPLC") med en egnet, fortrinnsvis kiral, kolonne. Føringen av løsningen av den organiske eluenten (+)-l-(3,4-diklorfenyl)-3-azabicyklo[3.1.0]heksan over den kiralt polysakkaridstasjonære fasen frembringer en første fraksjon (dvs en eller flere fraksjoner) inneholdende (-)-l-(3,4-diklorfenyl)-3-azabicyklo[3.1.0]heksan. Utrykkene "behandling" eller "behandle" viser til en forbedring av en lidelse lindret ved inhibering av dopamin-reopptak, eller minst ett skjelnbart symptom derav. Videre kan utrykkene "behandling" eller "behandle" vise til en bedring av minst ett målbart fysisk parameter, ikke nødvendigvis skjelnbart av pasienten. 4.3.1 LIDELSER LINDRET VED ANVENDELSE AV (-)-l-(3,4-DIKLORFENYL)-3-AZ ABIC YKLO [3.1.0] HEKS AN (-)-l-(3,4-diklorfenyl)-3-azabicyklo[3.1.0]heksan og farmasøytisk akseptable salter derav er nyttige ved behandling eller forebyggelse av endogene lidelser lindret ved inhibering av dopamin-reopptak. Slike sykdommer inkluderer, men er ikke begrenset til, ADD, depresjon, fedme, Parkinson's sykdom, tic-sykdommer og addiktive lidelser.
Lidelser lindret ved inhibering av dopamin-reopptak er ikke begrenset til spesifikke lidelser beskrevet heri, ettersom mange typer lidelser kan manifesteres fra den primære lidelsen. 4.4. TERAPEUTISK/PROFYLAKTISK ADMINISTRERING OG
SAMMENSETNING AV OPPFINNELSEN
På grunn av deres aktivitet er (-)-l-(3,4-diklorfenyl)-3-azabicyklo[3.1.0]heksan og farmasøytisk akseptable salter derav fordelaktig nyttige innen veterinær- og humanmedisin. Når administrert til en pasient er (-)-l-(3,4-diklorfenyl)-3-azabicyklo[3.1.0]heksan eller et farmasøytisk akseptabelt salt derav fortrinnsvis administrert som en del av en sammensetning som eventuelt omfatter en farmasøytisk akseptabel bærer. Foreliggende sammensetninger, som-omfatter en effektiv mengde av (-)-l-(3,4-diklorfenyl)-3-azabicyklo[3.1.0]heksan eller et farmasøytisk akseptabelt salt derav, administreres fortrinnsvis oralt. I en foretrukket utførelsesform formuleres (-)-l-(3,4-diklorfenyl)-3-azabicyklo[3.1.0]heksan eller et farmasøytisk akseptabelt salt derav i henhold til rutineprosedyrer som en farmasøytisk sammensetning tilpasset oral administrering til mennesker. Sammensetninger for oral levering kan være i form av tabletter, sugetabletter, vandige eller oljesuspensjoner, granulater, pulvere, emulsjoner, kapsler, sirup, eller eliksirer, for eksempel. Oralt administrerte sammensetninger kan inneholde en eller flere midler, for eksempel, søtningsmidler, slik som fruktose, aspartam eller sakkarin; smakstilsetningsmidler slik som peppermynte, olje av wintergreen, eller kirsebær; fargemidler; og preserveirngsmidler for å frembringe et farmasøytisk spiselig preparat. 4.5 KOMBINASJONSTERAPI
I visse utførelsesformer av den foreliggende oppfinnelsen kan (-)-l-(3,4-diklorfenyl)-3-azabicyklo[3.1.0]heksan eller et farmasøytisk akseptabelt salt derav anvendes i kombinasjon med minst ett annet terapeutisk middel. (-)-l-(3,4-diklorfenyl)-3-azabicyklo[3.1.0]heksan eller et farmasøytisk akseptabelt salt derav og det andre terapeutiske middelet kan virke additivt eller, mer foretrukket, synergistisk. (-)-l-(3,4-diklorfenyl)-3-azabicyklo[3.1.0]heksan eller et farmasøytisk akseptabelt salt derav kan administreres samtidig med administrering av et annet terapeutisk middel som kan være del av den samme sammensetningen eller i en forskjellig sammensetning fra den som omfatter (-)-l-(3,4-diklorfenyl)-3-azabicyklo[3.1.0]heksan eller et farmasøytisk akseptabelt salt derav. Det andre terapeutiske middelet kan være et anti-opiatmiddel. Nyttige anti-opiatmidler inklduerer, men er ikke begrenset til, metadon, klonidin, lofeksidin, levometadylacetat HC1, naltrekson og buprenorfin.
Det andre terapeutiske middelet kan være et anti-kokainmiddel. Nyttige anti-kokainmidler inkluderer, men er ikke begrenset til, desipramin, amantadin, fluoksidin og buprenorfin.
Det andre terapeutiske middelet kan være en appetittsenker. Nyttige appetittsenkere inkluderer, men er ikke begrenset til, fenfluramin, fenylpropanolarnin og mazindol.
Det andre terapeutiske middelet kan være et anti-lyserginsyredietylamid ("anti-LSD") middel. Nyttige anti-LSD-midler inkluderer, men er ikke begrenset til diazepam.
Det andre terapeutiske middelet kan være et anti-fencyklidin ("anti-PCP") middel. Nyttige anti-PCP-midler inkluderer, men er ikke begrenset til, haloperidol.
Det andre terapeutiske middelet kan være et anti-Parkinson's sykdom-middel. Nyttige
anti-Parkinson's midler inkluderer, men er ikke begrenset til, dopaminprekursorer, slik som levodopa, L-fenylalanin, og L-tyrosin; neurobeskyttende midler; dopaminagonister; dopaminreopptakinhibitorer; antikolinære midler slik som amantadin og memantin; og
1,3,5-trisubstituerte adamantaner, slik som l-amino-3,5-dimetyl-adamantan (U.S. patent nr 4.122.193 til Sherm et al).
Det andre terapeutiske middelet kan være et anti-depresjonsmiddel. Nyttige anti-depressiva inkluderer, men er ikke begrenset til, amitriptylin, klomipramin, doksepin, imipramin, trimipramin, amoksapin, desipramin, maprotilin, nortriptylin, protripylin, fluoksetin, fluvoksamin, paroketin, setralin, venlafaksin, bupropion, nefazodon, trazodon, fenelzin, tranylcypromin og selegilin.
Det andre terapeutiske middelet kan være et anksiolyttisk middel. Nyttige anksiolyttiske midler inkluderer, men er ikke begrenset til, benzodiazepiner, slik som alprazolam, klordiazepoksid, klonazepam. Klorazepat, diazepam, halazepam, lorazepam, oksazepam, og prazepam; ikke-benzodiazepinmidler, slik som buspiron; og tranquilizers slik som barbituater.
Det andre terapeutiske middelet kan være et anti-psykotisk legemiddel. Nyttige anti-psykotiske legemidler inkluderer, men er ikke begrenset til, fenotiaziner, slik som klorpromazin, mesoridazinbesylat, tioridazin, acetofenazinmaleat, flufenazin, perfenazin, og trifluoperazin; tioksantener, slik som klorprotiksen, og tiotiksen; og andre heterocykliske forbindelser, slik som klozapin, halperidol, lokapin, molindon, pimozid og risperidon. Foretrukne anti-psykotiske legemidler inkluderer klorpromazin Hel, tioridazin HC1, flufenazin HC1, tiotiksen HC1 og molindon HC1.
Det andre terapeutiske middelet kan være et anti-fedmelegemiddel. 5. EKSEMPEL: (-)-l-(3,4-DIKLORFENYL)-3-AZ ABIC YKLO [3.1.0] HEKS ANH YDROKLORID
Til 279 mg av (±)-l-(3,4-diklorfenyl)-3-azabicyklo[3.1.0]heksanhydroklorid oppnådd ved fremgangsmåtene beskrevet i Esptein et al, J. Med. Chem., 25:481.490 (1981) ble det lagt 7 ml 9:1 heksan:ispropylalkohol, fulgt av 8 dråper av dietylamin. Til den resulterende blandingen ble det tilsatt isopropylalkohol, dråpevis, til løsningen var oppnådd. Løsningen ble konsentrert til et volum på 6 ml ved å bruke en strøm av heliumgass, og seks 1 -ml-porsjoner av konsentratet ble underlagt high-performance-væskekromatografi ved å bruke et HPLC-instrument utstyrt med en 1 cm x 25 cm Diacel CHIRALPAK AD-kolonne (Chiral Technologies, Inc., Exton, Pennsylvania). Eluering ble utført ved en ambient temperatur ved å bruke 95:5 (v/v) heksan:isopropylalkoholløsning inneholdende 0,05% dietylamin som en mobil fase ved en flowrate på 6 ml/min. Fraksjonen eluerte ved omkring 26,08 til 34 minutter ble samlet og konsetnrert for å frembringe et første residium, som ble løst i en minimal mengde med etylacetat. Ved å bruke en strøm av nitrogen ble etylacetatløsningen fordampet for å frembringe et andre residium, som ble løst i 1 ml dietyleter. Til dietyleterløsningen ble det tilsatt 1 ml dietyleter mettet med gassformsaltsyre. Et presipitat ble dannet som ble filtrert, vasket med 2 ml dietyleter og tørket for å frembringe 33 mg (-)-l-(3,4-diklorfenyl)-3-azabicyklo[3.1.0]heksanhydroklorid med 88% enantiomeroverskudd. Dette materialet ble gjenrenset vd å bruke kromatografibetingelsene beskrevet ovenfor. Fraksjonen som eluerte ved omkring 28 til omkring 34 minutter ble konsentrert, asidifisert og tørket, som beskrevet ovenfor, til å frembringe 16,0 mg av (-)-l-(3,4-diklorfenyl)-3-azabicyklo[3.1.0]heksanhydroklorid: optisk rotering [a]<25>o= -56° i metanol ved 2 mg/ml; 99,1% enantomerisk overskudd.
6. EKSEMPEL: AKTIVITETSSAMMENLIGNING AV (-)-l-(3,4-DIKLORFENYL)-3-AZABICYKLO[3.1.0]HEKSAN HCL OG (±)-l-(3,4-DIKLORFENYL)-3-AZABIC YKLO [3.1.0] HEKS AN HCL I ET DOPAMIN, NOREPINEFRIN, OG SEROTONINTRANSPORTER BINDINGSASSAY
Dopamin, norepinefrin, og serotonin optaksinhibeirngaktivitet til (-)-l-(3,4-diklorfenyl)-3-azabicyklo[3.1.0]heksanhydroklorid ble sammenlignet med (±)-l-(3,4-diklorfenyl)-3-azabicyklo[3.1.0]heksanhydroklorid ved å bruke et standard dopamin-transporterbindingsassay.
6.1 MATERIAL OG METODER
6.1.1 DOPAMIN-TRANSPORTERASSAY
Dopamin-transporterbindingsassay ble utført i henhold til fremgnagnsmåtene beskrevet i Madras et al, 1989, Mol. Pharmacol. 36(4):518-524 og Javitch et al, 1984, Mol. Pharmacol 26(l):35-44. Reseptorkilden var marsvin-striatalmembraner; radioligandet var [<3>H]WIN 35,428 (DuPont-NEN, Boston, Massachussetts) (60-87 Ci/mmol) ved en endelig ligandkonsentrasjon på 2,0 nM; den ikke-spesifikke determinanten 1 uM l-[2-[bis(4-fluorfenyl)metoksy]etyl]-4-[3-fenylpropyl]piperazindihydroklorid("GBR

(-)-l-(3,4-diklorfenyl)-3-azabicyklo[3.1.0]heksan HC1 ble oppnådd i henhold til fremgangsmåten i eksempel 5 ovenfor. Reaksjonene ble utført i 50 mM TRIS-HC1 (pH 7,4), inneholdende 120 mM natriumklorid og 5 mM Kcl og ved 25°C i en time. Reaksjonen ble terminert ved rask vacuumfiltrering på glassfiberfiltere. 6.2 RESULTATER
Data i tabell 1 viser at både (-)-l-(3,4-diklorfenyl)-3-azabicyklo[3.1.0]heksan HC1 og (±)-l-(3,4-diklorfenyl)-3-azabicyklo[3.1.0]heksan HC1 har affinitet for dopamin-opptakssetet. På motsatt måte viser data i tabellene 2 og 3 at (+)-l-(3,4-diklorfenyl)-3-azabicyklo[3.1.0]heksan HC1 har affinitet for norepinefrin- og serotonin-opptakssetene, hvor (-)-l-(3,4-diklorfenyl)-3-azabicyklo[3.1.0]heksan HC1 har ingen målbar affinitet for norepinefrin- og serotonin-opptakssetene. Selv om (±)-l-(3,4-diklorfenyl)-3-azabicyklo[3.1.0] heksan HC1 har en høyere bindingsaffinitet for dopamin-reopptakssetet enn (±)-l-(3,4-diklorfenyl)-3-azabicyklo[3.1.0] heksan Hel så kan anvendelsen av (-)-l-(3,4-diklorfenyl)-3-azabicyklo[3.1.0] heksan HC1 være mer fordelaktig enn (±)-l-(3,4-dikIorfenyl)-3-azabicyklo[3.1.0] heksan HCI for inhibering av dopamin-opptak på grunn dets spesifitet for inhibering av dopamin-opptak. Med andre ord, anvendelsen av (-)-l-(3,4-diklorfenyl)-3-azabicyklo[3.1.0] heksan HCI kan forebygge uønskede bivirkninger forbundet med inhibering av norepenefirn-opptak og serotinin-opptak, slik som hypertensjon og seksuell disfunksjon, henholdsvis.
Vellykket inhibering av dopamin-reopptak har blitt assosiert med behandingen av ADD, depresjon, fedme, Parkinson's sykdom, tic-lidelser og addiktive lidelser (Hitri et al, 1994, Clin. Pharmacol 17:1-22; Noble, 1994, Alcohol Supp. 2:35-43; og Blum et al, 1995, Pharmacogenetics 5:121-141). På grunn av dets spesifitet for inhibering av dopamin-opptak er (-)-l-(3,4-diklorfenyl)-3-azabicyklo[3.1.0]heksan eller et farmasøytisk akseptabelt salt derav mer fordelaktig enn (±)-l-(3,4-diklorfenyl)-3-azabicyklo[3.1.0]heksan eller dets farmasøytisk akseptabelt salt derav for behandling eller forebyggelse av lidelser forbundet med inhibering av dopamin-reopptak hos en pasient.
De spesifikke utførelsesformer fremlagt i eksemplene er kun ment som illustrasjoner av noen få aspekter ved oppfinnelsen.

8. (-)-l-(3,4-diklorfenyl)-3-azabicyklo[3.1.0] heksan eller et farmasøytisk akseptabelt salt derav, hvor hver ikke har mer enn 5% w/w av dets korresponderende (+)-enantiomer, for behandling eller forebyggelse av en lidelse lindret ved inhibering av dopamin-reopptak.
9. (-)-l-(3,4-diklorfenyl)-3-azabicyklo[3.1.0]heksan eller det farmasøytisk akseptable salt derav ifølge krav 8, som ikke har mer enn omkring 2% w/w av den korresponderende (+)-enantiomer.
10. (-)-l-(3,4-diklorfenyl)-3-azabicyklo[3.1.0]heksan eller det farmasøytisk akseptable salt derav ifølge krav 8, som ikke har mer enn omkring 1% w/w av den korresponderende (+)-enantiomer.

(-)-l-(3,4-diklorfenyl)-3-azabicyklo[3.1.0]heksan eller det farmasøytisk akseptable salt derav ifølge hvilket som helst av kravene 8-10, for behandling eller forebyggelse av en lidelse lindret ved inhibering av dopamin-reopptak, hvor lidelsen velges fra gruppen bestående av ADD, depresjon, fedme, Parkinson's sykdom og tic-lidelse.
12. (-)-l-(3,4-diklorfenyl)-3-azabicyklo[3.1.0]heksan eller det farmasøytisk akseptable salt derav ifølge hvilket som helst av kravene 8-10, for behandling eller forebyggelse av en lidelse lindret ved inhibering av dopamin-reopptak, hvor lidelsen er en vanedannende lidelse.

(-)-l-(3,4-dikIorfenyl)-3-azabicyklo[3.1.0]heksan eller det farmasøytisk akseptable salt derav ifølge krav 11, hvor ADD velges fra gruppen bestående av ADHD, hovedsakelig uoppmerksom type; ADHD, hovedsakelig hyperaktiv-impulsiv type; ADHD, kombinert type; adferdslidelse; og "oppositional defiant" lidelse.
14. (-)-l-(3,4-diklorfenyl)-3-azabicyklo[3.1.0]heksan eller det farmasøytisk akseptable salt derav ifølge krav 11, hvor depresjonen velges fra gruppen bestående av større depressive lidelser, gjentagende; dystymisk lidelse; og større depressive lidelser, enkeltepisode.
15. (-)-l-(3,4-diklorfenyl)-3-azabicyklo[3.1.0]heksan eller det farmasøytisk akseptable salt derav ifølge krav 11, hvor Parkinsons's sykdom er neuroleptisk-indusert parkinsonisme.

(-)-l-(3,4-diklorfenyl)-3-azabicyklo[3.1.0]heksan eller det farmasøytisk akseptable salt derav ifølge krav 11, hvor tic-lidelsen velges fra gruppen bestående av Tourette's syndrom, kronisk motorlidelse, stemme-tic-lidelse, transient-tic-lidelse, stamming, autistisk lidelse og somatiseringslidelse.

(-)-l-(3,4-diklorfenyl)-3-azabicyklo[3.1.0]heksan eller det farmasøytisk akseptable salt derav ifølge krav 12, hvor den vanedannende lidelsen velges fra gruppen bestående av spiseforstyrrelser, impulskontroll-lidelser, alkohol-relaterte lidelser, nikotin-relaterte lidelser, amfetamin-relaterte lidelser, kannabis-relaterte lidelser, kokain-relaterte lidelser, hallusinogen-anvendelse lidelser, inhalant-relaterte lidelser, og opioid-relaterte lidelser.

Fremgangsmåte for oppnåelse av (-)-l-(3,4-diklorfenyl)-3-azabicyklo[3.1.0]heksan, omfattende trinnene: (a) føre en løsning av en organisk eluent og (±)-l-(3,4-diklorfenyl)-3-azabicyklo[3.1.0]heksan over en kiral polysakkaridstasjonær fase for å frembringe en første fraksjon inneholdende (-)-l-(3,4-diklorfenyl)-3-azabicyklo[3.1.0]heksan; og (b) føre den første fraksjonen over den kiral polysakkaridstasjonære fasen for å frembringe en andre fraksjon inneholdende (-)-l-(3,4-diklorfenyl)-3-azabicyklo[3.1.0]heksan som ikke har mer enn 5% w/w av det korresponderende (+)-enantiomer.
