Description
Foreliggende oppfinnelse vedrører væskefylte rriykgelatinkapsler som kan oppnås ved en fremgangsmåte for innkapsling av en flytende blanding i en gelatinsammensetning. The present invention relates to liquid-filled soft gelatin capsules which can be obtained by a method for encapsulating a liquid mixture in a gelatin composition.

Enkelte farmakologisk aktive substanser kan ha biofarmasøytiske og/eller fysisk-kjemiske egenskaper som gjør at det er vanskelig å forme dem til kommersielt aksepter-bare formuleringer. Slike substanser kan imidlertid passende administreres i flytende form, for eksempel i et komplekst bærermedium dannet av en rekke komponenter. Opp-løsningsmidler som 1,2-propylenglykol og dimetylisosorbid har et stort potensiale som slike bærermedier. Certain pharmacologically active substances may have biopharmaceutical and/or physicochemical properties that make it difficult to shape them into commercially acceptable formulations. However, such substances can conveniently be administered in liquid form, for example in a complex carrier medium formed from a number of components. The carrier can be designed to form an emulsion in the stomach, which thereby facilitates absorption of the pharmacologically active substance. The carrier must be prepared precisely and even small variations in the composition cannot be tolerated without the system being irreversibly disturbed and its beneficial properties destroyed. Thus, the dissolving properties of the capsule filling material can be changed and the active substance can precipitate. This precipitation process can be irreversible and the patient is thereby underdosed. The emulsifying properties of the capsule filling material can be changed and upon administration no emulsion will be formed in the stomach and the pharmacologically active substance will not be absorbed correctly or in a reproducible manner.

It has been found that microemulsion preconcentrates comprising 1,2-propylene glycol as the hydrophilic phase in soft gelatin capsules tend to cause migration of the 1,2-propylene glycol into the capsule shell from the capsule filling material. There is not only a softening of the capsule shell, but also a destruction of the microemulsion pre-concentrates because the hydrophilic component was removed therefrom.
