Description
Foreliggende oppfinnelse angår nye beta-karbolin derivater, en fremgangsmåte for deres fremstilling og medisinske produkter som inneholder derivatene.
Melatonin, N-acetyl-5-metoxytryptamin er et hormon fra den pineale kjertel (furukonglekjertel), som ble isolert av Lemer & al. (J. Am. Chem. Sos., 80, 1958, 2587), og som har vært utgangspunkt for en rekke undersøkelser med hensyn til dets cirkadianske aktivitet, på søvnrytmen, på dets effekt på produksjonen av testosteron, for dets aktivitet på hypotalamus og i forskjellige psykiatriske lidelser.
Det har således vært foreslått å anvende melatonin og dens analoger, spesielt for behandling av depresjon og psykiatriske lidelser, da spesielt stress, angst, depresjon, søvnløshet, schizofreni, psykoser og epilepsi, og dessuten for behandlingen av søvnforstyrrelser som er forbundet med såkalt »jet-lag», neurodegenererende sykdommer i det sentrale nervesystemet, for eksempel Parkinsons sykdom eller Alzheimers sykdom, for behandlingen av kreft eller alternativt som et befruktningshemmende middel eller som et smertestillende middel.
Den direkte bruken av melatonin in vivo har imidlertid ikke vist seg å være særlig tilfredsstillende, noe som dels skyldes at den første passasjen gjennom leveren ekstraherer mer enn 90% av det aktive prinsippet, og at melatoninets hypnotiske aktivitet ikke fullt ut er påvist.
Forskjellige melatonin analoger har blitt beskrevet, og viser to synteseveier som enten angår melatonin substituenter (WO-A-89/01472, US-A5 283 343, US-A-5 093 352 eller WO-A-93/11761) eller til den aromatiske ringen ved å erstatte indolyl gruppen med en naftyl gruppe (FR-A-2 658 818, FR-A-2 689 124).
Vi har nå påvist at melatonin ikke viser noen hypnotisk aktivitet, men er bioforløperen til acetylerte metabolitter som induserer søvn.
Det er derfor en hensikt å tilveiebringe nye fremgangsmåter til fremstilling av beta-karbolin derivater. Denne hensikt er oppnådd med foreliggende oppfinnelse kjennetegnet ved det som fremgår av de vedlagte krav.
Den foreliggende patentsøknad foreslår en ny vei for utvikling av nye karbol i nderi vater som er analoge til de endogene acetylerte metabolittene til melatonin.

Foreliggende oppfinnelse angår racemiske blandinger av slike forbindelser, deres ,rene enantiomerer eller blandinger i alle mengdeforhold, foruten deres terapeutiske akseptable salter.
Mer spesielt kan karbonatomet som bærer R8-gruppen være et asymmetrisk sentrum, og foreliggende oppfinnelse innbefatter alle slike enantiomerer og blandinger.
Med begrepene lavere alkyl, lavere alkoksy eller perhalogen(lavere)alkyl forstås generelt radikaler hvor alkylresten inneholder fra et til seks karbonatomer.
Disse er fortrinnsvis lineære eller grenete C1-C4 alkylgrupper valgt mer spesielt fra metyl, etyl, n-propyl, i-propyl, n-butyl, i-butyl og t-butyl grupper. Med uttrykket umettet alifatisk kjede forestås fortrinnsvis en umettet C2-C6 hydrokarbonkjede.
Begrepet aryl betegner generelt aromatiske og heteroaromatiske grupper, og da spesielt arylgrupper valgt fra fenyl, tienyl, furanyl, pyridyl og naftylgrupper.. Arylradikalene kan også være substituert med en eller flere grupper valgt spesielt fra et hydrogenatom, et lavere alkyl, et lavere sykloalkyl, halogen(lavere)alkyI, perhalogen(lavere)alkyl, aryl, aralkyl, et hydroksyradikal, lavere alkoksy, lavere sykloalkoksy, mono eller polyhalogenalkoksy, aryloksy, aralkylaoksy, et tiolradikal, lavere alkyltio, lavere sykloalkyltio, mono eller polyhalogenalkyltio, aryltio, aralkyltio, format, lavere alkylkarbonyloksy, et lavere sykloalkylkarbonyloksy, halogen(lavere)alkylkarbonyloksy, arylkarbonyloksy, aralkylkarbonyloksy, dialkylaminalkyl, amino, alkylamino, dialkylamino, arylamino, diarylamino, aralkylamino, diaralkylamino, eventuelt substituert karboksamid, formamid, et lavere alkylkarbonylamino, et lavere sykloalkylkarbonylamino, halogen(lavere)alkylkarbonylamino, perhalogen(lavere)alkylkarbonylamino, lavere arylkarbonylamino, lavere aralkylkarbonyl amino, formyl, et lavere alkylkarbonyl, et lavere sykloalkylkarbonyl, halogen(lavere)alkylkarbonyl, perhalogen(lavere)alkylkarbonyl, arylkarbonyl, aralkylkarbonyl, karboksyl, lavere alkoksykarboriyl, lavere sykloalkoksykarbonyl, halogenkoksykarbonyl, aryloksykarbonyl, aralkyloksykarbonyl, et lavere akylsulfonyl, et lavere sykloalkylsulfonyl, halogen(lavere)alkylsulfonyl, perhalogen(lavere)alkylsulfonyl,. arylsulfonyl, aralkylsulfonyl, halogen, cyano eller nitroradikaler.
Uttrykket lavere aralkyl refererer seg til kombinasjonen av lavere alkyl og en arylgruppe som definert ovenfor. En slik gruppe vil fortrinnsvis være benzylradikalet, som eventuelt kan være substituert.
Halogenradikalene blir fortrinnsvis valgt fra fluor, klor, brom og jod atomer.
Perhalogenradikalene er fortrinnsvis perfluor radikaler.
Når R1-R2, R2-R3 og R3-R4 danner en del av en annen aromatisk ring, med eller uten et heteroatom, så er dette fortrinnsvis en annen benzenring, som eventuelt er substituert, eller en pyridylring som eventuelt er substituert.
Når Ri:R2, R2-R3 og R3-R4 danner en del av en annen ikke-aromatisk ring, så danner de fortrinnsvis et di val ent radikal med formelen -0-(CH2)m-, hvor m er 2 eller 3, og som eventuelt er substituert, eller et divalent radikal med formelen
(CH2)p-0-, hvor p er 1 eller 2 og som eventuelt er substituert. Når derivatene består av minst et asymmetrisk karbonatom, så angår oppfinnelsen de tilsvarende kjemiske blandinger så vel som de rene enantiomerene eller blandinger i alle typer mengdeforhold.
De terapeutisk akseptable saltene av derivatene ifølge foreliggende oppfinnelse er de vanlige kjente organiske og uorganiske salter innen den farmasøytiske industri, da spesielt hydrokloridene, tosylatene, mesylatene og sitratene, så vel som solvater sånn som hydratene eller hemihydratene av forbindelsene med den generelle formel
I.
Ved sammenligning av de derivater som er beskrevet i WO 96/08490, så er derivatene ifølge foreliggende oppfinnelse karakterisert ved at de inneholder en R8 gruppe som er forskjellig fra et hydrogenatom, og dette øker dramatisk forbindelsenes stabilitet i mavesekkens sure miljø og tillater en oral administrering. Uten å være bundet til en eller flere teorier, så har en nå funnet at den gruppe som nå er ansvarlig for den hypnotiske aktiviteten i de foreliggende forbindelser, er en-amidet eller dihydroen-amid gruppen. Dette en-amidet eller dihydroen-amidgruppen er gruppen bestående av Ca-N-Cb i formel I.

Hvor R2, R3, Rg, Rh og * er som definert ovenfor. ;Derivatet med formel lb kan fremstilles direkte ved å reagere oppfinnelsen med den generelle formel Ila. ;Hvor R2, R3, Rg, R]4 og x er som definert ovenfor, med en karboksylsyre (for eksempel akrylsyre) i nærvær eller fravær av difenylfosforazid eller med akryl oni tri 1. ;For å fremstille derivatene med den generelle formel Ila, så utføres det en Bischler-Napieralski reaksjon med å reagere forbindelser med den generelle formel Illa. Derivatene ifølge foreliggende oppfinnelse har myoavslappende egenskaper slik at de også kan brukes for behandlingen av depresjon og psykiatriske lidelser, da spesielt stress, angst, depresjon, søvnløshet, schizofreni, psykose og epilepsi, men også for behandlingen av søvnlidelser forbundet med reiser (»jet-lag»), neurodegenererende sykdommer i det sentrale nervesystemet, sånn som Parkinsons sykdom eller Altzheimers sykdom, eller for behandling av kreft eller alternativ av som et befruktningshemmende middel eller som et smertestillende middel. De har alle hypnotiske og sedative effekter. ;STABILITET I SURT MEDIUM OG FARMAKOLOGISK AKTIVITET ;Stabilitet i surt medium og oral biotilgjengelighet i Beagle hunder ;For å undersøke virkningen av R8-gruppen på stabiliteten av syklus A, ble det utført en sammenligning mellom forbindelsene EKSEMPEL A og EKSEMPEL B ;(Rs = et hydrogenatom) og forbindelsene fra eksemplene 1, 4, 8 og 11, og hvor 30 mM av hver av disse forbindelsene ble løst i 5 ml ved pH=l eller pH=2 eller pH=3 eller pH=7 (referanse) buffer. Løsningen ble rørt i 15 minutter ved 37°C. Deretter ble det utført målinger av de mengdene av forbindelsene som ikke var hydrolysert, og dette ble utført ved hjelp av høytrykksvaeske kromatografi. ;;Resultatet i tabell 1 er angitt som prosentmengde av forbindelsene som ble hydrolysert. Som påvist i tabellen så ble forbindelsene fra EKSEMPEL A og B totalt hydrolyset mellom pH=l og pH=3. ;Forbindelsene I, 4, 7, 8 og 11 er ganske stabile fra pH=2 ettersom mindre enn 27% av disse forbindelsene ble hydrolysert ved en pH som er lik eller lavere enn ved pH=2. ;Som konklusjon kan en angi at forbindelser hvor Rg-gruppen (i den generelle formel 1) er forskjellig fra et hydrogenatom, ikke underkastes total hydrolyse av syklus A. Stabiliteten i surt mavesekk miljø for forbindelsene fra eksempel 1, 4 og 8 gjør at disse kan administreres oralt. Farmakokinetiske undersøkelser av disse forbindelsene (tabell 1) er blitt utført i Beagle hunder, og forbindelsene 1, 4 og 8 viste absolutt biotilgjengelighet ved oral administrering i forhold til intravenøs administrering og denne tilgjengeligheten var større enn 35% (forbindelse 1) 20% ;(forbindelse 4) og lik 100% (forbindelse 8). Ettersom forbindelsen fra eksempel A og eksempel B har en absolutt biotilgjenglighet som er lavere enn 1% (Eksempel A) og 5% (Eksempel B). ;Forbindelse nr. 1 (se tabell II) så vel som forbindelsene 4 og 7 (se tabell III) viser signifikant hypnotisk effekt i Beagle hunder når forbindelsene ble gitt oralt. ;HYPNOTISK AKTIVITET I BEAGLE HUNDER: EFFEKT PÅ VÅKENHET/SØVNTILSTANDER I BEAGLE HUNDER ;Beagle hunder ble holdt i metallbur forbundet med en 2 Schwartzer ED 24 polygraf som var utstyrt med Brainlab programvare gjennom en bøylekabel. ;Prøve og kontrollproduktene ble administrert via den orale eller intravenøse vei ved hjelp av mavesekkrør. Det ble utført målinger i tidsrom på 30 sekunder for aktiv våkenhet, sløvhet, langsom bølgesøvn (lett søvn + dyp søvn) og REM søvn. Disse kriteria ble utført som beskrevet av Shelton et al. (i Shelton J., Nishino S. Vaugt J:, Dement W.C. og Mignot E. Comparative effects of modafinil and amphetamine on daytime sleepiness and cataplexy of narcoleptic dogs. Sleep 19(1); 29-38, 1996.), basert på frekvens og empletydemønstrene for EEG hjernebarksporing (fronto-frontal), EMG (øvre nakkemuskel) og EOG (bilateralt). Adferden ble også kontinuerlig observert for hver periode på 120 minutter. En akklimatisering før forsøkene var 5 døgn, og for å etterligne det stress som kunne forårsakes av mavesekkrøret, ble en blanding vann/PEG (50/50; V/V) administrert oralt sammen med intravenøse boluser. ;Våkenhet: dette innbefatter alle episoder med lav spenning blandet med frekvenssporing hvor EMG ikke var inhibert. I våkenhetstilstanden står hundene, de sitter eller ligger ned og øynene er åpne. ;Søvnighet: tilstanden ble angitt når hundene ligger rolig med øynene lukket (50% eller mer pr. et tidsrom) og hjernebarkens EEG viser spor av relativt lange bølger (4-7 Hz) uten utvikling av søvnspindler. Sykrone bølger ved 4-7 Hz og 50-100 myV opptrer på en bakgrunn av lagspennings-hurtigbølgeaktivitet. ;EMG er moderat nedsatt sammenlignet med våken tilstand. ;Lett dyp søvn - langsombølgesøvn (SWS) (lett søvn + dyp søvn): hjørnetennene er avslappet med nedadrettet stilling. EEG mønstrene er av høyere amplityde enn i det foregående trinn og søvnspindelen (10-14 Hz) og/eller kompleksene må være tilstede i lett søvn, mens EOG viser langsom øyebevegelse eller ingen bevegelse. Dyp søvn noteres når langsomme deltabølger (<4 Hz) utgjør mer enn 20% av et tidsrom eller mer under dyp søvn. ;REM: rask øyebevegelsessøvn blir angitt når hundene legger seg ned, øynene er lukket med periodevis raske muskelkramper. En lavspennings med blandet frekvens EEG sporing kan observeres sammen med raske øyebevegelser og et fall i EMG aktivitet. ;Gjenoppvåkning og varighet av søvnighet/søvntilstander hos Beagle hundene ble målt etter oral (og intravenøs administrering av forbindelsen fra Eksempel 1) administreringer av placebo (væske = 10 ml blanding av etanol/PEG 400/vann; 10/40/50 i V/V/V) og for forbindelsene fra eksempel 1, 4 og 7. ;Resultatene er angitt i tabellene II og III som Varigheter og Latenser for hvert trinn. ;Forbindelsene fov eksempel 1} 4 og 7 viser sterk hypnotisk aktivitet og induserer en søvn som er karakterisert ved en høy proporsjon av langsom bølgesøvn. Når det gjelder varighet og latens med hensyn til våkenhet, søvnighet og SWS tilstander, så induserer disse tre forbindelsene en signifikant hypnotisk effekt. ;TABELL II: Søvntilstander og søvnlatenser i en 20 minutters observasjonsperiode fulgt av eksempel 1 (prøve), og væske alene (Placebo: 10 ml etanol/PEG/vann; 10/40/50; V/V/V) administreringer via den orale eller den intravenøse vei ;Tabell III: Søvntilstandér og søvnlatenser i en 120 minutters observasjonsperiode, etter administrering av forbindelsen for eksempel 4 (prøve) og forbindelse eksempel 7 (prøve) og væske alene (Placebo : 10 ml etanol/PEG/vann; 10/40/50; V/V/V) via den orale vei. ;HYPNOTISKE OG SEDATIVE EFFEKTER I KYLLINGER ;De hypnotiske og sedative effektene av derivatene ifølge foreliggende oppfinnelse, fremstilt som beskrevet ovenfor (prøveresultatene er gitt i tabell IV nedenfor) ble sammenlignet med tilsvarende effekter for tre referanseprodukter, dvs. diazepam, pentobarbitalnatrium og melatonin, så vel som med 2 psykostimulerende forbindelser med hallusinogene egenskaper: 10-metoksyharmalan og harmalin, som er 3,4-dihydro-beta-karboliner, i 10 til 14 dager gamle kyllinger av rase nr. JA657. Dyrene ble underkastet to alternerende programmer med hensyn til belysning, bestående av 12 timers mørke (fra 20.00 til 08.00) og 12 timers lys (fra 08.00 til 20.00). Romtemperaturen var 25 grader C under den første uken av kyllingenes liv, og deretter 22 grader C og utover. Under dagen ble lyset tilveiebragt ved hjelp av en halogenlampe (30 W) plassert 30 cm over gulvet i kyllingburet. Under prøvene var vekten av kyllingene mellom 85 og 120 g. Prøvene ble utført mellom 14.00 og 15.00. Kyllingene ble delt i grupper på 3 innenfor identisk bur på 30 X 50 X 30 cm. Prøveproduktene ble administrert intramuskulært (IM) i den store lårmuskelen, som en løsning i 25/50/25 (V/V/V) etanol/PEG 400/vannblanding, i en mengde på 0,2 ml løsning pr. 100 g kroppsvekt. De doser som ble administrert for prøveproduktene (de nye forbindelsene ifølge foreliggende oppfinnelse og referanseforbindelsene) varierte fra 0,25 mmol til 2 mmol pr. 100 g kroppsvekt. Placeboproduktene tilsvarte 0,2 ml av nevnte 25/50/25 (V/V/V) etanol/PEG 400/vannblandingen. ;Løsningene av prøveproduktene i nevnte 25/50/25 (V/V/V) etanol/PEG 400/vannblanding ble fremstilt når de skulle brukes ved en suksessiv fortynning fra en lagerløsning, fremstilt fra 2,5 til 20 mmol av et nøyaktig oppveid produkt, som deretter suksessivt ble tilsatt 0,25 ml ren etanol og deretter 1 ml PEG 400, ble så rørt ved hjelp av ultralyd og så justert opp til 2 ml med 0,5 destillert vann for fremstilling av et injiserbart preparat. Tabell IV gir resultatene etter en IM administrering av doser mellom 0,25 og 2 mmol av prøveproduktene løst i 0,2 ml av etanol/PEG 400/destillert vannblåndingen (25/50/25:V/V/V), pr. 100 g kroppsvekt. For hver kylling ble det injiserte volumet justert som en funksjon av den virkelige kroppsvekten til 0,2 ml pr 100 g kroppsvekt. ;De observerte parametrene var den lokomotoriske aktiviteten og bevisthetstilstanden hos kyllingene i 2 timer, og dette tilsvarer 6 teoretiske oppvåkninger/søvnsykluser for en kylling av denne alder. Disse ble registrert ved hjelp av et videokamera i 90 minutter, og hvor de første 30 minuttene ble tatt som en tilpasningstid for forsøket. ;De hypnotiske og sedative effektene av prøveproduktene på døgnrytmen for 10 til 14 dager gamle kyllinger som var underkastet et program med permanent belysning fra klekking og i 48 timer, og deretter et alternerende program med 12 timers dagslys (08.00 til 20.00) og 12 timers mørke (20.00 til 08.00), opp til den dato da prøven ble foretatt, er gitt i tabell 1 nedenfor. Prøvene ble utført mellom kl. 14.00 og 15.00. ;For hvert prøveprodukt ble det utført flere serie målinger på grupper på 3 dyr, og hver verdi som er angitt er et middel for hver gruppe bestående av 3 kyllinger. Når antallet grupper er større enn eller lik 2, så er de angitte tall den observerte midlere grenseverdi. ;Nøkkel: ;NA: ikke anvendbar da dyrene forble beviste under hele observasjonsperioden; ;FAT: innsovningstid som er lik det tidsrom som er nødvendig for å gå fra en tilstand av aktiv bevissthet til en ikke-bevi sst'tilstand; ;ST: søvntid tilsvarer varigheten av søvnperioden fra begynnende søvn til oppvåkning; ;Sedasjonstid: etter oppvåkning innen aktivitetstidsrommet som tilsvarer trinn 2 som definert ovenfor. ;Under de betingelser ved hvilke prøven ble utført (tidspunkt for administrering under den fase hvor dyrene fikk lys, dvs. mellom 14.00 og 15.00, så hadde melatonin ingen hypnotisk aktivitet. ;Ved suksessivt å underkaste kyllingene programmer med hensyn til alternerende og permanent belysning, kunne et eksperiment påvise at melatonin ikke hadde noen direkte hypnotisk aktivitet som skyldes dens struktur. Dens hypnotiske aktivitet er avhengig av aktiviteten av enzymet N-acetyltransferase (NAT) i furukonglekjertelen hos kyllingene og administreringstidspunktet for melatonin. NAT enzymet er et acetyleringsenzym. I nærværet av NAT enzymet i furukonglekjertelen hos kyllingnene, vil en IM administrering av melatonin indusere en hypnotisk effekt med sterk intensitet (søvntid på mellom 250 og 350 minutter for en dose som tilsvarer et mmål melatonin pr. 100 g kroppsvekt). Melatonin er således forløperen for acetylerte metabolitter med direkte hypnotisk aktivitet. Forbindelser ifølge oppfinnelsen er analoger for de hypnotiske acetylmetabolittene for melatonin. ;I kontrast til melatonin så har alle derivatene ifølge foreliggende oppfinnelse en direkte hypnotisk og sedativ aktivitet, og disse er uavhengig av administreringstidspunktet, dvs. nivået av N-acetyltransferaseenzymet i det sentrale nervesystemet. ;De oppnådde resultater for derivater ifølge oppfinnelsen viser en hypnotisk effekt som er høyere enn for referanseproduktene (pentobarbital, melatonin) og er lik eller endog bedre enn tilsvarende resultater for diazepam. ;Derivatene ifølge oppfinnelsen er således spesielt fordelaktige ved en behandling av søvnforstyrrelser og sykdommer som er forbundet med en ubalanse med hensyn til melatoninaktiviteten. ;EKSEMPLER ;Eksempel 1: ETCARB07 ;Formel: CigH2oN202 M=296,36 g.mol-1 ;Struktur: ;9-metoksy-1 -etyl-2,3,4,6,7,12heksahydroindolo(2,3-a)kinolizin-4-one Fremstilling: Parametoksyfenylhydrazinsulfonat (5 g - 20,7 mmol) og N(4,4-dietoksybutyl)butanamid (4,8 g - 20,7 mmol) ble blandet i 85 ml kommersiell THF i en 500 ml kolbe. Blandingen ble så oppvarmet med tilbakeløp av THF, hvoretter 35 ml 25% eddiksyre ble dråpevis tilsatt. En svak gul blanding ble så rørt i 6 timer ved en temperatur mellom 80 og 85 grader C. Etter avkjøling ble reaksjonsblandingen overfør til en 2 liters Erlenmeyerkolbe og gjort basisk ved å tilsette ca. 100 ml av en mettet natriumkaibonatløsning ved pH>7. Reaksjonsblandingen ble så oppvarmet under tilbakeløp i 2 timer. Etter avkjøling ble det tilsatt 25 ml vann, og det ble utfelt et fast stoff. Dette ble fortynnet med etylacetat og vasket med vann. Den resulterende organiske fasen ble tørket over magnesiumsulfat, hvor løsemiddelet ble fordampet under redusert trykk.

En Bischler-Napieralski reaksjon på 2,1 g Nl-(butyryl)-5-metoksy-(L)-tryptofan-etyl ester fører til 1,5 g (3S)-l-propyl-3-karbetoksy-6-metoksy-3,4-dihydro-2-karbolin. En løsning av sistnevnte produkt i 20 ml DMF ble suksessivt dråpevis tilsatt 0,35 ml akrylsyre, I ml difenylfosforyl azid og 2 ml trietylamin. Etter separasjon på silicagel (elueringsmiddehetylacetat/petroleumeter - 30/70), ble det fremstil 400 mg (6S)-6-karbetoksy-9-metoksy-l-etyl-2,3,4,6,7,12-heksahydroindolo[2,3-a]kinolizin-4-en (utbytte 17%).
NMR: <!>H (CDCI3) : 1,1 (t, 3H); 1,25 (t, 3H); 2,3 (m, IH); 2,62 (m, 5H); 3,08 (dd, 6 og 16 Hz, 1H); 3,53 (d, 16 Hz, 1H); 3,85 (s, 3H), 4,03 (m, 2H); 6,00 (d, 6 Hz, 1H); 6,85 (dd, 2,3 og 8,7 Hz, 1H); 6,94 (d, 2,3 Hz, 1H); 7,24 (d, 8,7 Hz, 1H); 8,13 (bred s, 1H)
Smeltepunkt: 192°C
Claims (15)
1. Forbindelse, karakterisert ved at den generelle formel Ia (Ia' og Ia"):
hvor
R2 er (Ci-Cg) alkoksyradikal og fortrinnsvis et metoksy radikal,
R3 er et hydrogenatom eller et (Ci-C6) alkylradikal, fortrinnsvis et metylradikal, R8 er et (Ci-Ce) alkylradikal, (Ci-Ce) alkyloksykarbonyl, en fenylgruppe, eventuelt subsituert med (Ci-Ce) alkyl, halogen, nitro, di (Ct-C6) alkylamino, en (C]-C6) alkyloksygruppe, eller en aminogruppe, Ci-Ce alkoksykarbonyl eller en pyridylgruppe,
X er fortrinnsvis et divalent radikal med formelen
Ri i er et oksygenatom eller et svovelatom,
Rh og R15 er uavhengige av hverandre, et hydrogenatom, (C]-C6) alkyl, eller (Ci-Qe) alkoksykarbonyl.
2. Derivat ifølge krav 1, karakterisert ved at Rg er et etyl, heksyl eller isopropylradikal, et fenyl, fluorfenyl, metoksyfenyl, aminofenyl, dimetylaminifenyl, nitrofenyl, p. metylfenylradikal, et etoksykarbonylradikal eller et pyridinradikal.
