Nabih og Elsheikh (J. Pharm. Sei. 54, 1672-1673 (1965)) beskriver l-[[2-dimetylamino)etyl]amino]-4-[(dietylamino )-metyl]tioksanten-9-on. Ingen nyttevirkning er vist for forbindelsen.
Collins og Rosi, US patent 3.745.172 beskriver som et mellomprodukt i syntesen av antifungus- og antibakterielle midler, forbindelsen: og som et antelmintisk og antibakterielt middel, forbindelsen :
Rosi og Peruzotti, US patent 3.312.598 beskriver l-[[2-(dietylamino)etyl]amino]-9-okso-9H-tioksanten-4-karboksylsyre som et fermenteringsbiprodukt som ikke har noen angitt nyttevirkning.

Chem. 31, 707-712 (1988)) beskriver N-[2-(dimetylamino )etyl] -9-okso-9H-tioksanten-4-karboksamid-monohydroklorid som ble testet in vitro mot murin leukemi (L1210) og in vivo mot P388 leukemi celler og ble funnet "sannsynligvis ikke verdt å forfølge" som et potensielt antitumor middel.
Ifølge foreliggende oppfinnelse er det tilveiebragt nye forbindelser som er kjennetegnet ved formelen:

R<3>er hydrogen eller lavere alkyl;
R<4>er hydrogen eller lavere alkyl;

R<7>er lavere alkyl eller Ar;
R<8>er hydrogen, lavere alkyl, lavere alkoksy, hydroksy eller halogen; og
Ar er fenyl eller fenyl substituert med metyl, metoksy, halogen eller nitro;
eller et farmasøytisk akseptabelt syreaddisjonssalt eller solvat derav.
Forbindelsene er nyttige for behandling av tumorer i pattedyr.
Den heri benyttede betegnelse lavere alkyl omfatter lineære, forgrenete eller cykliske hydrokarboner inneholdene 4 karbonatomer eller mindre. Halogen omfatter brom, klor eller fluor. Det er foretrukket at Ar er fenyl.
Videre er det ifølge oppfinnelsen tilveiebragt et farma-søytisk preparat som er kjennetegnet ved at det omfatter en forbindelse som definert i medfølgende krav 1, og en farmasøytisk akseptabel bærer eller fortynningsmiddel.
Oppfinnelsen tilveiebringer også anvendelse av en forbindelse som definert i medfølgende krav 1 for fremstilling av et legemiddel for behandling av en tumor i et pattedyr.
Syntesen av forbindelser ifølge foreliggende oppfinnelse kan illustreres som vist i reaksjonsskjemaer A og B:
Forbindelsene med formel III (formel I hvor Q er -CH=N-Ar) kan syntetiseres ved oppvarming av et aldehyd med formel II sammen med omkring en ekvivalent av den passende anilin-forbindelsen i et inert, aseotropgivende oppløsningsmiddel, fortrinnsvis xylen eller toluen, ved tilbakeløp.
Forbindelsene med formel IV kan fremstilles fra et aldehyd med formel II ved oppvarming ved 150-185<*>C i nærvær av omtrent 10 ekvivalenter maursyre i formamid eller N-alkyl-formamid som oppløsningsmiddel. Betingelsene er velkjente innen teknikken for Leuckart-reaksjonen. Forbindelsene med formel V oppnås deretter ved syrehydrolyse av formamidet. Forbindelsene med formel VI (formel I hvor Q er -CH2H(R<4>)-S02R<7>) kan syntetiseres ved sulfonylering av aminet V med et lite overskudd av et lavere alkylsulfonyl- eller arylsulfo-nylklorid i et egnet oppløsningsmiddel, fortrinnsvis pyridin, ved 0-50°C.
Forbindelsene med formel VI, hvor R<4>er lavere alkyl, kan alternativt syntetiseres ved sulfonylering av aminet V, hvor R<4>er hydrogen, som beskrevet ovenfor, fulgt av behandling av det resulterende sulfonamid VI, hvor R<4>er hydrogen, med et overskudd av en base, fortrinnsvis natriumhydroksyd, i et egent oppløsningsmiddel, slik som tetrahydrofuran, fulgt av behandling med et overskudd av et passende lavere alkyl-halogenid ved en temperatur i området fra 0°C opp til kokepunktet for det benyttede oppløsningsmiddel.
Karboksamidene med formel IX kan syntetiseres ved omsetning av aldehydet II med et 5-6 gangers overskudd av hydroksylaminhydroklorid i pyridin, eventuelt inneholdene et ko-oppløsningsmiddel, fulgt av dehydratisering av oksimet (VII) ved behandling med et overskudd av eddiksyreanhydrid og oppvarming i et inert, høytkokende oppløsningsmiddel slik som xylen, og til slutt delvis hydrolyse av nitrilet (VIII) i konsentrert svovelsyre.
Forbindelser med formel X (formel I hvor Q er CH2N(R4)C(0 )R7) kan syntetiseres ved acylering av aminet V med et overskudd av et lavere alkylsyreklorid, eller arylsyreklorid i et egnet oppløsningsmiddel, fortrinnsvis pyridin, eventuelt i nærvær av et ko-oppløsningsmiddel, fortrinnsvis diklormetan, ved en temperatur i området fra 0 til 50"C.
Forbindelsene med formel XI (formel- hvor Q er CH2N(H)P(0)-(0-lavere alkyl)2) kan syntetiseres ved behandling av aminet V med et overskudd av et passende di-lavere alkyl fosforo-kloridat i et egnet oppløsningsmiddel, slik som diklormetan, i nærvær av et overskudd av en base, fortrinnsvis trietylamin, ved en temperatur i området fra 0 opp til 50<*>C.
Aldehydet II er tilgjengelig ut fra fremgangsmåten beskrevet i US patent 3.294.803 og ved Mn02-oksydasjon av alkoholen oppnådd ved fremgangsmåten i US patent 3.711.512.
Forbindelsene med formel I er nyttige både i den frie baseformen og i form av syreaddisjonssalter, og begge former omfattes av foreliggende oppfinnelse. Syreaddisjonssaltene er i noen tilfeller en mer hensiktsmessig bruksform, og i praksis er bruken av saltformen ifølge sakens natur ens-betydende med bruken av baseformen. Syrene som kan benyttes for å fremstilles syreaddisjonssaltene inkluderer fortrinnsvis de som ved kombinasjon med den frie basen gir medisinsk akseptable salter, det vil si salter hvis anioner er relativt uskadelige for dyreorganismen i medisinske doser av saltene slik at nytteegenskapene som naturlig er forbundet med den frie basen ikke forringes av bivirkninger som kan tilskrives anionene. Ved utførelse av foreliggende oppfinnelse er det hensiktsmessig å danne hydroklorid-, fumarat-, toluen-sulfonat-, metansulfonat- eller maleatsaltene. Andre passende medisinsk akseptable salter som omfattes av oppfinnelsen er imidlertid de som er avledet fra andre mineralsyrer og organiske syrer. Syreaddisjonssaltene av de basiske forbindelsene fremstilles enten ved oppløsning av den frie basen i vandig alkoholoppløsning inneholdene den passende syren og isolering av saltet ved fordampning av oppløsningsmidlet, eller ved omsetning av den frie basen og en syre i et organisk oppløsningsmiddel, hvorved saltet utsepareres direkte, utfelles med et annet organisk oppløsningsmiddel, eller kan oppnås ved konsentrering av oppløsningen. Selv om medisinsk akseptable salter av de basiske forbindelsene er foretrukket så omfattes alle syreaddisjonssaltene av foreliggende oppfinnelse. Alle syreaddisjonssalter er nyttige som kilder for den frie baseformen selv om det spesielle saltet per se er ønsket kun som et mellomprodukt, som for eksempel når saltet dannes kun for rensingsformål eller identifikasjon, eller når det benyttes som et mellomprodukt ved fremstilling av et medisinsk akseptabelt salt ved ioneutvekslingsmetoder.
Strukturene til foreliggende forbindelser ble fastslått ved syntese, elementanalyse og infrarød, ultrafiolett og kjernemagnetisk ressonans-spektroskopi. Reaksjonenes forløp og identiteten og homogeniteten til produktene ble bestemt ved tynnsjiktskromatografi (TLC) eller gass-væskekromatografi (GLC). Smeltepunktene er angitt i grader C og er ukorrigerte. Utgangsmaterialene er enten kommersielt tilgjengelige eller kan fremstilles ved hjelp av velkjente metoder.
Oppfinnelsen vil nå bli beskrevet mer spesielt under henvisning til følgende eksempler.
EKSEMPEL 1
l-[[2-(dietylamino)etyl]amino] - 4-(N-fenyl f ormimidoyl )-tioksanten-9-on
(I:R1=R<2>=Et; Q=CH=N-C6H5;R8=H; n=2) (III; Ar=Ph)
En blanding av 17,7 g (50 mmol) av l-[[2-(dietylamino)-etyl]amino]tioksanten-4-karboksyaldehyd og 15,1 g (150 mmol) anilin i 100 ml toluen, ble tilbakeløpskokt i 8 timer med en Dean-Stark-felle. TLC på aluminiumoksyd med kloroform/heksan-/isopropylamin 10:10:2 viste ufullstendig reaksjon. Toluenet ble avdestillert, 25 ml anilin ble tilsatt og blandingen tilbakeløpskokt i 4 timer. 50 ml xylen ble tilsatt og reaksjonsblandingen tilbakeløpskokt på nytt i 3 timer. Oppløsningsmidlet og overskudd anilin ble fjernet i vakuum og resten omkrystallisert fra benzen hvilket ga 19,9 g rå-produkt. Dette ble omkrystallisert fra ca 1,5 1 heksan og dette ga 15,8 g (86 %) produkt, smp. 125-126°.
EKSEMPEL 2
N-[[2[[2-(dietylamino)etyl]amino]-9-oksotioksonaten-4-yl]metyl]formamid
(I: R<1=>R<2>=Et; Q=CH2NHCH0; R<8>=H; n=2) (IV; R<4=>H)
En oppløsning på 35,4 g (0,1 mol) av l-[ [2-(dietylamino )-etyl]amino]tioksanten-4-karboksyaldehyd, 420 ml formamid og 50 ml (1 mol) maursyre ble oppvarmet ved 160° i 1 time. Reaksjonsblandingen ble avkjølt, helt 12 1 vann og gjort basisk med ca 50 ml 35 # natriumhydroksydoppløsning. Det gummiaktige bunnfallet ble frafiltrert og tørket i vakuum. Det tørkede bunnfallet ble oppløst i ca 1,5 1 varm etylacetat, behandlet med trekull og krystallisert ved avkjøling. Produktet ble frafiltrert, vasket med etylacetat og tørket og dette ga 29,0 g (75 %) produkt, smp. 154-155°.
EKSEMPEL 3
N-[[l-[[2-(dietylamino)etyl]amino] -9-oksot ioksanten-4-yl]metyl]-N-metylformamid
(IV:R1=R<2=>Et;R<4=>Me; R<8>=H; n=2)
Ved en fremgangsmåte analog med den i eksempel 2 ble 24,6 g av N-metylformamidet fremstilt fra 35,4 g (0,1 mol) av l-[[2-(dietylamino)etyl]amino]-tioksanten-4-karboksyaldehyd, 394 g N-metylformamid og 50 ml maursyre. Produktet ble omkrystallisert fra 150 ml aceton til et smp. på 127-130°.
EKSEMPEL 4
4-(aminometyl)-l-[[2-(dietylamino )etyl]amino]-tioksanten-9-on (I: R<1>=R2=Et; Q=CH2NH2; R<8>=H; n=2) (V;R<4=>H)
En oppløsning av 24,4 g (64 mmol) av formamidet i eksempel 2 i 240 ml 2N saltsyre ble oppvarmet på et dampbad i 1 time. Reaksjonsblandingen ble avkjølt til romtemperatur, gjort basisk med 35 £ vandig natriumhydroksyd og det resulterende gule bunnfall ble oppsamlet ved filtrering. Produktet ble oppløst i benzen, behandlet med trekull, tørket med magnes-iumsulfat, filtrert og aceotropbehandlet for å fjerne spor av vann. Den tørkede resten ble krystallisert fra metanol og isopropanol ved tilsetning av eterisk hydrogenklorid. Det resulterende faste stoff ble omkrystallisert i flere omganger fra metanol og dette ga 10,6 g produkt, smp. 270-272°, som dihydrokloridsaltet.
EKSEMPEL 5
l-[[2-(dietylamino)etyl]amino]-4-[(metylamino )-metyl] - tioksanten-9-on
(I: R<1>=R2=Et; Q=CE2NHCH3; R<8>=H; n=2) (V; R<4=>Me)
Ved en fremgangsmåte helt analog med den i eksempel 4 ble 10,5 g av metylaminet oppnådd som dihydrokloridhemihydratet fra 14,6 g (37 mmol) av N-metylformamidet i eksempel 3 og 150 ml 2N saltsyre. Produktet smeltet ved 241-243°.
EKSEMPEL 6
N-[[l-[[2-(dietylamino)etyl]amino]-9-oksotioksanten-4-yl]metyl]metansulfonamid
(I:R1=R2=Et; Q=CH2NHS02CH3; R<8=>H; n=2) (VI; R<4=>H;R<7=>Me)
En oppløsning av 10,65 g (30 mmol) av den frie basen av aminet i eksempel 4 i 100 ml pyridin ble avkjølt i et isbad og 4 g (35 mmol) metansulf onylklorid ble tilsatt i en porsjon. Blandingen ble omrørt i 2 timer ved romtemperatur og helt i 750 ml vann inneholdene 2 g natriumhydroksyd. Det mørke gule bunnfallet ble oppsamlet, vasket med vann og tørket i vakuum natten over. Et annet utbytte ble oppnådd ved tilsetning av overskudd natriumhydroksyd til filtratet og filtrering av det resulterende faste stoff. De kombinerte utfellingene etter tørking ble omkrystallisert fra benzen og dette ga 6,4 g av metansulfonamidet, smp. 169-170°.
EKSEMPEL 7
l-[[2'-(dietyl amino ) etyl] amino] -9-oksotioksanten-4-karboks-amid
(I:R1=R<2=>Et; Q=CONH2; R<8>=H; n=2) (IX)
En suspensjon av 74 g (0,23 mol) l-[[2-(dietylamino )-etyl]amino]tioksanten-4-karboksaldehyd og 74 g (1,06 mol) hydroksylaminhydroklorid i 400 ml pyridin og 400 ml etanol ble tilbakeløpskokt i 0,5 timer og 70 ml vann ble tilsatt for å gi en homogen oppløsning. Oppløsningen ble oppvarmet i ytterligere 2 timer og ble hensatt ved romtemperatur i 14 timer. Det resulterende krystallinske oksim (VII) ble frafiltrert for oppnåelse av et kvantitativt utbytte, smp 215-128°.
120 g av oksimet ble kort oppvarmet på et dampbad i 180 ml eddiksyreanhydrid for oppnåelse av en oppløsning. Oppløs-ningen ble avkjølt, 100 ml 1,8 M EC1 i eter ble tilsatt og den resulterende suspensjon ble fortynnet med 500 ml eter. Suspensjonen ble hensatt i 14 timer ved 0° og filtrert. Resten (123 g, smp. 109-112°) ble oppslemmet i 250 ml xylen og tilbakeløpskokt i 20 min. Blandingen ble avkjølt og 71,3 g av nitrilet (VIII) ble frafiltrert, smp. 265°. 10 g av nitrilet ble omrørt i 200 ml kons. H2SO4ved romtemperatur i 3 dager. Reaksjonsblandingen ble nøytralisert med kons. NH4OH og resten frafiltrert. Resten ble behandlet i varm EtOAc/EtOH, filtrert og produktet krystallisert fra den avkjølte oppløsningen, smp. 241-243°. Det ble oppløst i etanol og en ekvivalent HC1 i etanol ble tilsatt. 6 g av amidhydrokloridet ble oppnådd, smp. 271-272°.
EKSEMPEL 8
N-[[l-[[2-(dietylamino)etyl)amino]-9-oksotioksanten-4-yl]metyl]N-metylmetansulfonamid
(I:R1=R<2>=Et; Q=CH2N(CH3)S02CH3;R8=H; n=2) (VI; R<4=>Me; R<7>=Me)
En oppløsning av 1,5 g (3,5 mol) av metansulf onamidet i eksempel 6 i THF (60 ml) ble avkjølt til 0°C i et isbad og NaH 0,16 g (4,0 mmol) ble tilsatt. Reaksjonsblandingen ble oppvarmet til romtemperatur, omrørt i 10 minutter og deretter ble 0,25 ml (4,0 mmol) metyliodid tilsatt. Reaksjonsblandingen ble omrørt ved romtemperatur i 24 timer og oppløsnings-midlet ble fjernet i vakuum. Resten ble renset ved kolonnekromatografi på silisiumdioksyd ved eluering med kloroform (100 %) deretter med 1 % isopropylamin/klorof orm til oppnåelse av 1,15 g (74 <$ >) av N-metylmetansulfonamidet som et gult pulver, smp 175-177°C.
EKSEMPEL 9
N-[[l-[[2-dietylamino )etyl]am ino]-9-oksotioksanten-4-yl]metyl]fenylsulfonamid

Ved å følge en fremgangsmåte vesentlig lik den beskrevet i eksempel 6 ble 2,4 g (57 $ >) av fenyl sul f onamidet oppnådd som metansulf onsyresaltet fra 2,54 g (7,15 mmol) av den frie basen av aminet i eksempel 4, pyridin (50 ml) og benzen-sulfonylklorid (1,1 ml, 8,62 mmol), fulgt av behandling av den således oppnådde rest med metansulfonsyre i metanol. Produktet ble omkrystallisert fra etanol.
EKSEMPEL 10
N-[[l-[[2-(dietylamino)etyl]amino]-9-oksotioksanten-4-yl]metyl]acetamid

Ved å følge en fremgangsmåte vesentlig lik den beskrevet i eksempel 6 ble 2,3 g (52 Sé) av acetamidet oppnådd som et orangefarget fast stoff fra 4,15 g (11,7 mmol) av den frie basen av aminet i eksempel 4, pyridin (60 ml) og acetylklorid (0,82 ml, 11,53 mmol). Produktet ble omkrystallisert fra aceton og smeltet ved 182-183°C.
EKSEMPEL 11
N-[[l-[[2-(dietylamino)etyl]amino]-9-oksotioksanten-4-yl]metyl]benzamid
(I: R<1>=R<2>=Et; Q=CH2NHC(0)Ph;R8=H; n=2) (X; R<4=>H;R<7=>Ph)
Ved å følge en fremgangsmåte vesentlig lik den beskrevet i eksempel 6 ble 1,02 g (68 $ >) av benzamidet oppnådd som et gult pulver fra 1,17 g (3,29 mmol) av den frie basen av aminet i eksempel 4, pyridin (25 ml) og benzoylklorid (0,42 ml, 3,62 mmol). Produktet ble renset ved kolonnekromatografi på silisiumdioksyd ved eluering med kloroform (100 K>) til 1 % isopropyl/kloroform, fulgt av omkrystallisering fra etylacetat. Produktet smeltet ved 161-163°C.
EKSEMPEL 12
N-[[l-[[2-(dietylamino)etyl]amino]-9-oksotioksanten-4-yl]dietylfosformaid
(I: R<1>=R2=Et; Q=CH2NHP(0)(OEt)2; R<8>=H; n=2) (XI;R<4=>H)
En oppløsning av 2,28 g (6,41 mmol) av den frie basen av aminet i eksempel 4, CH2C12(50 ml), og trietylamin (2 ml) ved 0°C ble behandlet med dietylfosforokloridat (1,0 ml, 6,9 mmol). Reaksjonsblandingen ble omrørt ved 0°C i 2 timer, deretter ved romtemperatur i 1 time. Oppløsningsmidlet ble fjernet i vakuum og resten ble renset ved kolonnekromatografi på silisiumdioksyd ved eluering med etylacetat (deretter 5 % metanol/etylacetat og til slutt metanol/isopropylamin/etylacetat (5/5/90) hvilket ga 2,28 g (72 %) av dietylfosfor-amindet som et gult fast stoff, smp 108-110°C omkrystallisert fra etylacetat.
EKSEMPEL 13
N-[[l-[[2-(dietylamino)etyl]amino] -9-oksot ioksanten-4-yl]metyl]-N-etylformamid
(I: R<1>=R2=Et; Q=CH2NEtCH0; R<8>=H; n=2) (IV; R<4=>Et)
En oppløsning av 2,0 g (5,6 mmol) l-[[2-(dietylamino)etyl]-amino]tioksanten-4-karboksaldehyd, N-etylformamid (24,0 ml) og maursyre (3,0 ml, 79,5 mmol) ble oppvarmet ved 170° C i 4 timer. Reaksjonsblandingen ble avkjølt, helt i vann og gjort basisk med 10 % natriumhydroksyd. Det ble oppnådd et fast stoff som ble oppsamlet ved filtrering og vasket med vann. Den faste resten ble opptatt i kloroform/vann og det organiske laget ble separert og tørket over Na2S04. Oppløs-ningsmidlet ble fjernet i vakuum og resten renset ved radlalkromatografi ved eluering med isopropylamin/metanol/- etylacetat (0,5/1/98,5) hvilket ga 1,32 g (57 %) av N-etylformamidet som et orangefarget fast stoff, smp. 75-77°C.
EKSEMPEL 14
l-[[2-(dietylamino)etyl]amino]-4-[( etyl amino )-metyl] tioks-anten-9-on
(I: R<1>=R<2=>Et; Q=CH2NHC2E5; R<8>=H; n=2) (V;R<4=>Et)
Ved en fremgangsmåte vesentlig lik den beskrevet i eksempel 4 ble 1,29 g (92 av etylaminet oppnådd som dihydrokloridet fra 1,3 g (3,2 mmol) av N-etylformamidet i eksempel 13 og 10,8 ml 2N saltsyre. Produktet ble omkrystallisert fra etanol/tetrahydrofuran og smeltet ved 160°C (dekomp.).
Andre representative forbindelser av typen I kan fremstilles analogt med det som beskrives i eksemplene 1-14, ved å benytte det passende l-[[2-(dialkylamino)etyl]amino]- eller l-[ [3-(dialkylamino )propyl]amino]-tioksanten-4-karboksy-aldehyd i steden for l-[[2-(dietyl amino)etyl]amino]tiok-santen-4-karboksaldehyd. Aldehydene og deres forløpere er beskrevet i TJS patent 3.294.803.
Representative eksempler på forbindelser ifølge oppfinnelsen ble testet med henblikk på antitumoraktivitet i mus ifølge nedenstående metode: Dyrene ble samlet, implantert subkutant med 30-60 mg tumorfragmenter ved hjelp av en nr. 12 hulnål og igjen samlet før ikke-selektiv fordeling til de forskjellige behandlings-og kontrollgruppene. For behandling i et tidlig trinn ble kjemoterapi startet i løpet av 1-5 dager etter tumorimplan-tering mens antallet celler var relativt lite (10<7->10<8>celler). For behandling i et fremskredet trinn ble kjemoterapi utsatt inntil tumoren ble relativt stor (2-300 mg i størrelse) en 300 mg tumor inneholder omtrent 3 x 10<8>totale celler. Tumorer innenfor et gitt forsøk i et fremskredet trinn befinner seg innenfor et 2,5-gangers størrelsesområde for 90 % av dyrene. Tumorer ble målt med en passer en gang i uken (eller to ganger i uken for de mer hurtigvoksende tumorer). Mus ble avlivet da deres tumorer nådde 1500 mg (det vil si før de kunne forårsake ubehag for dyret). Tumorvekter ble anslått utfra todimensjonale målinger.
Behandlings- og kontrollgruppene ble målt da tumorene hos kontrollgruppen nådde en størrelse på ca 700-1200 mg (gruppemedian). Tumormedianvekten for hver gruppe ble bestemt (inkludert null-verdier). T/C-verdien (vekt av behandlede tumorer over vekten av kontrolltumorer) i prosent er en indikasjon på antitumoreffektivitet: en T/C-verdi lik eller mindre enn 42 % anses som signifikant antitumoraktivitet av Drug Evaluation Branch of the Division of Cancer Treatment (NCI). En T/C-verdi på <10 % er anses å indikere meget signifikant antitumoraktivitet. Et legemsvekt-tapsnadir (gruppemiddel) på over 20 £ eller 20 % legemiddeldød anses å indikere en usedvanlig toksisk dosering.
Resultatene er vist i tabell I for pankreaktisk ductal adenocarsinom nr 03 og i tabell 2 for colon-adenocarcinom nr 38.
Forbindelsen i eksempel 5 ble testet ved intravenøs infusjon mot et antall andre tumorer som vist i tabell 3 og var aktiv ved 300 mg/kg p.o. mot colon-adenocarcinom nr 38.
Forbindelsen i eksempel 6 ble testet ved intravenøs bolus-injeksjon mot et antall andre tumorer som vist i tabell 4. Foreliggende farmasøytiske preparater innbefatter en eller flere av forbindelsene ifølge oppfinnelsen formulert til preparater sammen med en eller flere ikke-toksiske, fysiologisk akseptable bærere eller hjelpemidler som kollektivt referes til som bærere i foreliggende sammenheng, for parenteral injeksjon, for oral administrasjon i fast eller flytende form, for rektal eller topisk administrasjon, og lignende.
Preparatene kan administreres til mennesker og dyr, enten oralt, rektalt, parenteralt (intravenøst, intramuskulært eller subkutant), intrasisternalt, intravaginalt, intrapere-tonealt, lokalt (pulvere, salver eller dråper), eller som en bukal eller nasal spray.
Preparater egnet for parenteral injeksjon kan omfatte fysiologisk akseptable sterile vandige eller ikke-vandige oppløsninger, dispersjoner, suspensjoner eller emulsjoner og sterile pulvere for rekonstituering til sterile injiserbare oppløsninger eller dispersjoner. Eksempler på egnede vandige og ikke-vandige bærere, fortynningsmidler eller oppløsnings-midler innbefatter vann, etanol, polyoler (propylenglykol, polyetylenglykol, glyserol og lignende) egnede blandinger derav, vegetabilske oljer (slik som olivenolje) og injiserbare organiske estere slik som etyloleat. Hensiktsmessig fluiditet kan opprettholdes for eksempel ved bruk av et belegg slik som lecitin, ved opprettholdelse av den nødven-dige partikkelstørrelse i tilfelle for dispersjoner og ved bruk av overflateaktive midler.
Disse preparatene kan også inneholde hjelpemidler slik som preserverings-, fukte-, emulgerings- og utleveringsmidler. Hindring av virkningen av mikroorganismer kan sikres ved hjelp av forskjellige antibakterielle og antifungale midler, for eksempel parabener, klorbutanol, fenol, sorbinsyre og lignende. Det kan også være ønskelig å innbefatte isotoniske midler, for eksempel sukkere, natriumklorid og lignende. Forlenget absorbsjon av den injiserbare farmasøytiske formen kan bevirkes ved bruk av midler som forsinker absorbsjon, for eksempel aluminiummonostearat og gelatin.
Forbindelsene kan om ønsket og for mer effektiv fordeling inkorporeres i langsomt frigivende eller målrettede avlev-er ingssystemer slik som polymermatriser, liposomer og mikrosfærer. De kan steriliseres for eksempel ved filtrering gjennom et bakterie-tilbakeholdene filter eller ved inkor-porering av steriliseringsmidler i form av sterile faste preparater som kan oppløses i sterilt vann, eller et eller annet sterilt injiserbart medium umiddelbart før bruk.
Faste doseringsformer for oral administrasjon innbefatter kapsler, tabletter, piller, pulvere og granuler. Den aktive forbindelsen blir i slike faste doseringsformer sammenblandet med minst en inert konvensjonell eksipiens (eller bærer) slik som natriumsitrat eller dikalsiumfosfat eller
(a) fyllstoffer eller drøyemidler slik som for eksempel stivelser, laktose, sukrose, glukose, mannitol og
kiselsyre,
(b) bindemidler som for eksempel karboksymetylcellulose,
alginater, gelatin, polyvinylpyrolidon, sukrose og
akasie,
(c) fuktemidler som for eksempel glyserol,
(d) desintegreringsmidler som for eksempel agar-agar,
kalsiumkarbonat, potet- eller tapiokastivelse, alginsyre, visse komplekse silikater og natrium-karbonat,
(e) oppløsningsretarderende midler som for eksempel
paraffin,
(f) absorbsjonsakselleratorer som for eksempel kvaternære
ammoniumforbindelser,
(g) fuktemidler som for eksempel cetylalkohol og
glyserolmonostearat,
(h) adsorbsjonsmidler som for eksempel kaolin og (i) smøremidler som for eksempel talk, kalsiumstearat, magnesiumstearat, faste polyetylenglykoler, natrium-laurylsulfat eller blandinger derav.
I tilfelle for kapsler, tabletter og piller kan doserings-formene også omfatter buffermidler.
Prosentandelen av aktiv komponent i preparatet for behandling av tumorer eller kreft kan varieres slik at det oppnås en egnet dose. Dosen som administrers til en spesiell pasient kan variere avhengig av legens bedømmelse ved anvendelse av følgende kriterier: administrasjonsvei, behandlingens varighet, pasientens størrelse og tilstand, virkningsgraden til den aktive komponenten, og pasientens respons til denne. En effektiv doseringsmengde av aktiv komponent kan således lett bestemmes av legen ved å betrakte alle kriterier å anvende sin beste bedømmelse på pasientens vegne.

R<3>er hydrogen eller lavere alkyl;
R<4>er hydrogen eller lavere alkyl;
R55 er hydrogen;
r<6>er hydrogen;
R<7>er lavere alkyl eller Ar;
R<8>er hydrogen, lavere alkyl, lavere alkoksy, hydroksy eller halogen; og
Ar er fenyl eller fenyl substituert med metyl, metoksy, halogen eller nitro;
eller et farmasøytisk akseptabelt syreaddisjonssalt eller solvat derav.
