Description
Fremgangsmåte ved fremstilling av anthelmintisk
aktive 5- eller 6-substituerte benzimidazoler.
Foreliggende oppfinnelse angår fremstilling av nye anthelmintisk virksomme forbindelser som er derivater av benzimidazoler som i 5- eller 6-stilling har en amino- eller substituert aminogruppe, eller halogen i 5- og/eller 6-stilling.
Infeksjonen kjent som helminthiasis innbefatter befengelsen med forskjellige
arter av parasittormer i det menneskelige legeme, særlig i fordøyelseskanalen. Det er en alminnelig, vidt utbredt og alvorlig sykdom og metodene for behandling og forhindring av dem har ikke vært helt tilfredsstillende. Det er et mål ved foreliggende oppfinnelse å skaffe fremgangsmåter for syntetisering av nye forbindelser for fremstilling av anthelmintica, og særlig slike som er nyttige mot ascarider.
Ifølge foreliggende oppfinnelse har det nu vist seg at forbindelser av den
generelle formel:
hvor R er furyl eller thiazolyl, Rj er hydrogen eller benzoyli og den ene av substi-tuentene R^ og R^ er amino, lavere-alkylamino eller di-lavere-alkylamino, og den annen er hydrogen, eller R^ og/eller R4 er halogen, og syreaddisjonssalter derav, har anthelmintisk aktivitet, og at noen av dem har en forbausende grad av aktivitet mot svineascarider.
Blant de helmintiske parasitter er nematodene av stammene Haemonchus, Trichostrongylus, Ostertagia, Hematodirus, Cooperia Bunostromum, Oesophago-stromum, Chabertia, Trichuris, Ascaris, Capillaria, Heterakis og Ancylostoma de mest alminnelige parasitter i husdyr. Sykdommer som tilskrives slike infeksjoner, såsom ascariasis, trichostrongylosis og gross parasitism, er meget utbredt og al-vorlige, idet den angrepne vert vanligvis lider av slike tilstander som underernæring, anemi og blødning. Dessuten kan mere fremskredne og ubekjempede tilfeller av helminthiasis føre til henligging og død.
Generelt har forbindelsene ifølge foreliggende oppfinnelse en forbausende
høy aktivitet mot den parasittiske rundorm Ascaris lumbricoides var. Suum. Den nevnte ascarid utgjør et alvorlig økonomisk problem ved svineoppdrett, og død, stanset vekt og andre permanente skader til verten er vanlige resultater av infek-sjon.
Den voksne orm holder til i tynntarmen hos svin. Et stort antall egg legges daglig og føres ut i avføringen av det infiserte dyr og fortsetter og utvikles inntil det tas opp. Når larvene er utklekket, finner de veien til blodstrømmen, vandrer til leveren, lunger og andre organer og finner igjen veien til tarmene hvor de vokser opp. Leveren og nyrene kan vise sterke forandringer på grunn av passasjen av larvene gjennom dem.
Når begge nitrogenatomer på benzimidazolkjernen er usubstituerte, vil hydro-genatomet festet til et av de to nitrogenatomer i ringen vandre mellom dem, og på grunn av tautomeri vil 5- og 6-stillingene i ringen være ekvivalente. Det er vanlig å betegne disse spesielle forbindelser som f. eks. 5(6)-fluor-benzimidazol eller 5 (6) - (dimethylamino) -benzimidazol.
Som eksempler på de nye substituerte benzimidazoler som fremstilles ifølge oppfinnelsen, kan nevnes 2-(4'-thiazolyl)-5(6)-fluor-benzimidazol, 2-(2'-thiazolyl) - 5 (6) -klor-benzimidazol, 2- (2 '-furyl) -5 (6) -dimethylamino-benzimidazol, 2- (4T-thiazolyl)-5(6)-brom-benzimidazol og l-benzoyl-2-(2'-thiazolyl)-5-dimethylamino-benzimidazol.
Ifølge oppfinnelsen fremstilles forbindelsene av formel I ved at enten
a) en forbindelse av den generelle formel:
hvor R 1 , R„ o og R 4 er som ovenfor angitt, omsettes under oppvarmning med en forbindelse av den generelle formel: R - D^, hvor R er som ovenfor angitt, og er carboxy, carbalkoxy, carbamoyl, cyano eller halogen, i nærvær av en mineralsyre, eller
b) en forbindelse av den generelle formel:
hvor Rg og R^ er som ovenfor angitt, omsettes med en forbindelse av den generelle
formel: R - D^, hvor R er som ovenfor angitt, og Dg er carboxy, carbalkoxy eller halogen, i et inert oppløsningsmiddel, hvorpå nitrogruppen i det dannede anilid reduseres, og anilidet ringsluttes ved reduksjon, eller
c) en forbindelse av den generelle formel:
hvor Rg og R^ er som ovenfor angitt, omsettes med en forbindelse av den generelle
formel: R - Dg, hvor R er som ovenfor angitt, og D^ er carboxy, carbalkoxy, carbamoyl, cyano eller halogen, i et inert organisk oppløsningsmiddel, den erholdte forbindelse nitreres, og den innførte nitrogruppe reduseres, og det dannede anilid ringsluttes, eller
d) en forbindelse av den generelle formel:
hvor R, Rg og R^ er som ovenfor angitt, omsettes med et positivt halogenerings-middel som er i stand til å halogenere nitrogenatomet i amidingruppen, eventuelt under tilsetning av en base, hvorpå det dannede N-halogen-N'-fenylamidin omsettes med en base, eller
e) når Rj i trinn a) - d) fremstillede forbindelser er hydrogen, omsettes eventuelt den dannede forbindelse med et alkalimetallhydrid og derpå med et benzoylhalogenid
i et inert oppløsningsmiddel, og at de dannede forbindelser eventuelt overføres til syreaddisjonssalter på i og for seg kjent vis.
Ved alternativ a) ifølge oppfinnelsen kan et passende substituert o-fenylendiamin, som 2-amino-4-fluoranilin, omsettes med en heterocyclisk carboxylsyre, eller et derivat derav, som syrehalogenidet, nitrilet, esteren eller amidet,
i en mineralsyre, såsom svovelsyre, fosforsyre eller saltsyre. Fremgangsmåten utføres ved temperaturer fra ca. 120 - 180°C i ca. -3 - 10 timer. Den optimale tid og temperatur vil selvsagt avhenge i noen grad av de spesielle reaksjoner som anvendes. Benzen, toluen, xylen og lignende kan anvendes som oppløsningsmidler for en eller flere av reaktantene.
Ekvimolare mengder av den heterocycliske forbindelse og diaminet foretrekkes, og fra ca. 5-20 vektdeler syre pr. del heterocyclisk forbindelse er ønskelig. Det vil forståes at de relative mengder av syre ikke er kritisk. De ønskede benzimidazoler utvinnes ved å kjøle reaksjonsblandingen og fortynne den med vann. Når benzimidazolene ikke krystalliserer lett efter ovenstående behandling, felles
de ved å behandle den avkjølede blanding med en base.
Ved alternativ b) ifølge oppfinnelsen fremstilles 5- eller 6-substituerte benzimidazoler ved å omsette et passende substituert nitroanilin med en heterocyclisk carboxylsyre eller den tilsvarende ester eller syrehalogenid derav, i et passende inert oppløsningsmiddel, såsom pyridin, benzen eller lignende. Nitrogruppen på det dannede anilin reduseres så og benzimidazoldannelsen bevirkes ved å behandle anilidet med et reduksjonssystem, som zink-saltsyre, zink-eddiksyre, jern-saltsyre og lignende eller ved katalytisk reduksjon.
Dessuten kan et passende substituert anilin, som 2,3-difluoranilin, omsettes med en carboxylsyre eller et derivat derav, og derpå nitreres og reduseres for å få en annen aminosubstituent. Det dannede o-fenylendiamin kan så overføres til det ønskede benzimidazol, som beskrevet ovenfor.
Ved alternativ d) kan utgangsmaterialet fremstilles ved at et passende substituert anilin omsettes med et heterocyclisk nitril i nærvær av en passende katalysa-tor, som aluminiumklorid, under dannelse av et N'-fenylamidin-derivat av den heterocycliske forbindelse. Dette N'-fenylamidinet blir så ved foreliggende fremgangsmåte klorert eller bromert for å få et N-klor- eller N-brom-N'-fenylamidin. Denne halogenering utføres ved å omsette nevnte N'-fenylamidin med et positivt halogener-ingsmiddel som er i stand til å halogenere nitrogenatomet i amidingruppen. Passende reagenser for dette formål er N-haloamider eller N-haloimider, f. eks. N-klorsuccin-imid, N-bromacetamid og lignende. Når et N-haloamid eller N-haloimid anvendes, anvendes en base i en tilstrekkelig mengde til å nøytralisere syreaddisjonssaltet av N'-fenylamidinet. Egnet for dette formål er et alkalimetallcarbonat eller -hydroxyd.
De foretrukne halogeneringsmidler er imidlertid underklor og underbromsyr-ling. Disse dannes bekvemt in situ ved tilsetning av et alkali- eller jordalkalimetall-hypohalit til en oppløsning av N'-fenylamidin-syreaddisjonssaltet, hvorved nøytrali-sasjon av syreaddisjonssaltet og dannelsen av halogeneringsmidlet inntrer samtidig. Typiske hypohaliter som er nyttige for dette formål, er natrium- eller kaliumhypo-klorit, natriumhypobromit og calciumhypobromit.
N-halogen-N'-fenylamidinet som fåes ved ovenstående halogenering, over føres til benzimidazolen ved behandling med en base, som alkali-eller jordalkali-metallhydroxyd, såsom natriumhydroxyd, kaliumhydroxyd eller calciumhydroxyd.
Ved alternativ e) ifølge oppfinnelsen fremstilles forbindelsene av formel I ovenfor ved å overføre den 1-usubstituerte forbindelse til et alkalimetallsalt, fortrinnsvis natriumsaltet, ved å bringe nevnte forbindelse i intim kontakt med natriumhydrid i et passende oppløsningsmiddel. Et lite molart overskudd av natriumhydrid gir tilfredsstillende resultater og ekvimolare mengder av benzimidazol og natriumhydrid kan også anvendes om ønskes. Reaksjonen utføres bekvemt ved å oppvarme reaktantene til en noe forhøyet temperatur, men værelsetemperatur gir tilfredsstillende resultater .
En 1-substituert benzimidazol kan så erholdes ved å bringe benzimidazol-al-kalimetallsaltet i kontakt med et acylhalogenid, som benzoylklorid, acetylklorid, ace-tylbromid, propionylklorid og lignende, et aralkylhalogenid, som benzylklorid, eller et alkylhalogenid, som methylklorid, ethylklorid, methylbromid eller lignende. Vanligvis tilsettes acylhalogenidet, aralkylhalogenidet eller alkylhalogenidet direkte til en oppløsning eller suspensjon av benzimidazolsaltet i inert oppløsningsmiddel, og reaksjonen får lov til å foregå ved en temperatur fra ca. værelsetemperatur opptil 100°C. Reaksjonstemperaturer i området 50 - 75°C foretrekkes. Oppløsningsmidlet som anvendes som reaksjonsmedium, er fortrinnsvis et hydrocarbonoppløsningsmid-del, som benzen, toluen, xylen, petroleter og lignende, enten alene eller i blanding med andre oppløsningsmidler som er blandbare dermed, som dimethylformamid.
De substituerte benzimidazoler som fremstilles ifølge oppfinnelsen er nyttige ved behandling og/eller forhindring av helminthiasis i husdyr. For dette formål kan de gies oralt som en bestanddel av dyreforet, eller i drikkevannet, i saltblokker eller i enhetsdoseformer, såsom boluser og tvangsinnføring. Mengdene av aktiv bestanddel som kreves for optimal motvirkning av helminthiasis varierer selvsagt med slike faktorer som den spesielle forbindelse som anvendes, arten av dyr som behandles, arten av den infiserende parasitt, graden av infeksjonen, og enten for-bindelsen anvendes terapeutisk eller profilaktisk. Generelt for forbindelsene beskrevet her, når de gies oralt til husdyr i dagsdoser på fra 0,1 mg til ca. 500 mg pr. kg dyrelegemsvekt, meget virksomme for å motvirke helminthiasis uten uak-septabel giftvirkning. Når forbindelsene skal anvendes som terapeutiske midler,
fåes gode resultater når dyrene gies en dagsdose på fra ca. 5 mg til ca. 500 mg, og fortrinnsvis 15 mg til 250 mg pr. kg legemsvekt. Tilførselen kan være i en enkelt dose eller fordelt over flere mindre doser over et tidsrom av 24 timer. Når profilaktisk behandling ønskes, og forbindelsene gies kontinuerlig, fåes tilfredsstillende resultater når de av dyret daglig opptatte doser er fra ca. 0,1 mg til 100 mg pr. kg legemsvekt.
Benzimidazolene som fremstilles ifølge oppfinnelsen kan gies dispergert i
eller blandet med de vanlige bestanddeler av dyreernæringen, dvs. f6ret, drikkevannet eller andre væsker som normalt inntas av dyrene. Disse metoder foretrekkes når det ønskes å gi de aktive forbindelser kontinuerlig enten som et terapeutisk eller profilaktisk middel, over en periode av flere dager eller mere.
Som tidligere anført har de 5- og 6-substituerte benzazoler som fremstilles ifølge oppfinnelsen, i alminnelighet en usedvanlig høy aktivitet mot svineascarider, som Ascaris iumbricoides. Når anthelmintica som de som fremstilles ifølge foreliggende oppfinnelse, anvendes ved behandling av dyr som svin, og særlig når behand-lingen er for å forhindre eller befri verten for ascaridinfeksjon, blir de aktive midler fortrinnsvis gitt som en bestanddel av føden. Når de beskrevne forbindelser ifølge formel I anvendes som en bestanddel av føden, kan den nødvendige dose gies med f6r-blandinger inneholdende fra ca. 0,001 til 3 vekt% aktiv forbindelse. Slike medisintilsatte f6rblandinger kan gjøres i stand for direkte anvendelse ved å blande ovenstående mengde aktiv bestanddel direkte med f6ret. Det medisintilsatte f6r kan også fremstilles ved å anvende for-tilsetningsmidler inneholdende høyere konsentrasjoner av aktiv bestanddel jevnt dispergert i en fast spiselig bærer som nevnt ovenfor. Fértilset-ningene kan så fortynnes med foret for å få den ønskede konsentrasjon av aktiv bestanddel for matning. Da det er bekvemt for férfremstillere å anvende ca. 450 g (1 pund) fértilsetning for hvert tonn ferdig f6r, er den foretrukne konsentrasjon av aktiv bestanddel i for-tilsetnirigen delvis en funksjon av mengden av aktiv bestanddel som ønskes i ferdige for. I alminnelighet kan fårtilsetninger inneholdende fra ca. 5% til ca. 50 vekt% aktiv bestanddel anvendes tilfredsstillende for å gi den nødvendige dose i det ferdige f6r.
Eksempel 1
2- ( 4'- thiazolyl) - 5 ( 6) - dimethylamino- benzimidazol
En oppløsning av 4,1 g (0.03 mol) N,N-dimethyl-p-fenylendiamin 50 ml toluen ble tilsatt i løpet av 30 minutter ved værelsetemperatur til en blanding av thiazol-4-carboxylsyre-klorid fremstilt ved å koke 3,9 g (0,03 mol) thiazol-4-carboxylsyre og 3,3 ml thionylklorid i 50 ml toluen under tilbakeløp. Det faste stoff som utskiltes ble frafiltrert, vasket med toluen og tørret, og hadde smeltepunkt 181 - 183°C.
En liten prøve ble overført til den frie base ved å oppløse den i vann, tilsette et overskudd av kalium bi carbonat og ekstrahere med ether.
Til en kold oppløsning av 1,1 g (0,005 mol) N-(p-dimethylamino)-4'-thiazol-carboxanilid-hydroklorid i 5 ml svovelsyre ble der i løpet av 4 - 5 minutter tilsatt 0,2ml (0,0044 mol) rykende salpetersyre i 2,5 ml svovelsyre. Efter 10 minutter ble blandingen helt på knust is. Isen ble nøytralisert ved tilsetning av kaliumbicarbonat og det kastanjebrune faste stoff som dannedes, [N-(p-dimethylamino-o-nitro)-4'-thia-zol-carboxanilid], ble frafiltrert, vasket og tørret og hadde smeltepunkt 209 - 210°C. Efter omkrystallisasjon fra alkohol smeltet materialet ved 210,5 - 211°C.
En oppløsning av 460 mg (0,00157 mol) N-(p-dimethylamino-o-nitro)-4'-thia-zolcarboxanilid og 0,2 ml (0,002 mol) konsentrert saltsyre i 100 ml methanol ble redusert ved værelsetemperatur ved 2,8 kg/cm 2 med 0,2 g 5% palladium på aktivert trekull. Trekullet ble fjernet ved filtrering og filtratet konsentrert til tørrhet ved ned-satt trykk. Residuet ble oppløst i en blanding av 12 ml ethanol, 11 ml vann og 1,2 ml konsentrert saltsyre og oppløsningen kokte under tilbakeløp i 4 timer. Overskuddet av syre ble nøytralisert ved tilsetning av ammoniakkvann, og fast materiale begynte å krystallisere. Blandingen ble avkjølt og det gulbrune faste stoff, [2-(4'-thiazolyl) - 5(6)-dimethylamino-benzimidazol], ble frafiltrert, vasket og tørret og hadde smeltepunkt 231 - 234°C.
Eksempel 2
2- ( 2 '- furyl) - 5 ( 6) - fluorbenzimidazol
Til 50 ml tørr toluen ble tilsatt 60 g 2-nitro-4-fluoranilin og 10 ml furoyl-klorid. Den røde oppløsning ble oppvarmet under tilbakeløp i 1 time. Oppløsnings-midlet ble fjernet under vakuum, benzen ble tilsatt til residuet, derpå inndampet og tilsatt og fordampet påny. Det gjenværende faste røde stoff ble filtrert og vasket med n-butanol. Filtratet ble fortynnet med ether hvorved man fikk ca. 1 g av et gult hygro-skopisk fast stoff som ble spaltet i løpet av få minutter når det ble utsatt for luft,
smeltepunkt 113 - 117°C.
Det ovenfor dannede anilid ble så redusert med en palladium på kullkatalysa-tor og tilsatt til en blanding av 75 ml vann, 75 ml ether-alkohol og 10 ml saltsyre,'
og oppløsningen ble kokt under tilbakeløp i 4 timer. Den røde oppløsning ble behand-let med trekull, filtrert og filtratet inndampet til halvt volum og nøytralisert med konsentrert ammoniumhydroxyd. Det erholdte gulbrune faste stoff hadde efter filtrering smeltepunkt pa 210 - 214°C. Omkrystallisasjon fra ethanol-vann ga det ønskede produkt med smeltepunkt 220 - 221°C.
Eksempel 3
2- ( 4 r- thiazoIyl) - 5 ( 6) - fluor- benzimidazol
En blanding av 1,0 g (0,07 mol) p-fluoranilin, 1,1 g (0,07 mol) 4-cyanothiazol, 1,33 g (0,07 mol) vannfritt aluminiumklorid og 11 ml tetraklorethan ble omrørt og kokt under tilbakeløp i 20 minutter. Den ovenstående væske ble dekantert, residuet oppløst i 25 rn! methanol og derpå tilsatt til 50 ml av en 5 N oppløsning av natriumhydroxyd. En stor mengde vann ble tilsatt og blandingen ekstrahert med ether. Ved fjernelse av etheren ga dette ekstrakt N-4'-fluorfenyl-(thiazol-4-amidin) med smeltepunkt 100 - 102°C. Omkrystallisasjon fra ethanol-vann (1:2) ga et produkt som smeltet ved 103,5 - 104,5°C.
En suspensjon av 4,4 g N-4Mluorfenyl-(thiazol-4-amidin) i 25 ml methanol
og 25 ml vann ble innstilt på pH 4,5 ved tilsetning av konsentrert saltsyre. Til opp-løsningen ble tilsatt 7,3 ml (1 ekvivalent) 2,8 M natriumhypoklorit. Efter 3 minutter ved værelsetemperatur ble en oppløsning av 1 g natriumhydroxyd i 4 ml vann tilsatt. Blandingen ble kokt under tilbakeløp i 10 minutter. Et lyst farvet fast stoff, [2-(4'-thiazolyl)-5(6)-fluor-benzimidazol],. viste seg og ble efter avkjøling, filtrert, vask-
et og tørret og hadde et smeltepunkt på 251 - 253°C.
Eksempel 4
2- ( 4 '- thiazolyl) - 5 ( 6) - dif luor- benzimidazol
En blanding av 5.2 g (0,04 mol) 3,4-difluoranilin, 4,4 g (0,04 mol) 4-cyanothiazol og 5,4 g (0,04 mol) vannfritt aluminiumklorid i 50 ml tetraklorethan ble om-rørt og kokt under tilbakeløp i 25 minutter og tillatt å avkjøle. Oppløsningsmidlet ble dekantert fra og det mørke residuum ble oppløst i 60 ml methanol og tilsatt til en om-rørt oppløsning av 40 g natriumhydroxyd i 600 ml vann. N-3,4-difluorfenyl-(thiazol-4-amidin), med smeltepunkt 116 - 118°C, feltes som reaksjonsprodukt.
En blanding av 1,2 g N-3,4-difluorfenyl-(thiazol-4-amidin) og 15 ml methanol ble innstilt på pH 4,5 med konsentrert saltsyre. Til denne oppløsning ble tilsatt 3,2 ml (1, ek vi va lent) 1,6 M natriumhypoklorit og oppløsningen fikk stå ved værelsetemperatur i 3 minutter. En oppløsning av 0,5 g natriumcarbonat i 12 - 15 ml vann ble tilsatt, og blandingen ble kokt under tilbakeløp i 10 minutter. Blandingen ble avkjølt, mere vann tilsatt og produktet ble frafiltrert, vasket og tørret hvilket ga 2-(4'-thiazolyl)-5,6-difluor-benzimidazol med smeltepunkt 250°C.
Eksempel 5
Syreaddisjonssalter
Når de heri beskrevne benzimidazoler ble isolert som de frie baser, kan de lett overføres til syreaddisjonssalter ved behandling med syre. Typiske salter som kan dannes på denne måte, er mineralsaltene såsom hydrohalogenidene, f. eks. hydro-kloridet, hydrobromidet, hydrojodidet, sulfatene, nitratene, fosfatene og lignende, alifatiske syresalter, såsom acetåtet, trimethylacetatet og propionatet, salter poly-carboxylsyrer såsom citratet, oxalatet, succinatet og lignende, og salter av andre uoppløselige organiske syrer såsom pamoatet, embonatet og hydroxynaftoatsaltene. Noen av disse salter er mere vannoppløselige og noen mindre vannoppløselige enn den frie base. Det vil derfor sees at oppløselighetsegenskapene av et spesielt benzimidazol i alminnelighet kan avpasses ved kritisk valg av et salt. Når benzimidazolene ifølge oppfinnelsen anvendes i saltform som anthelmintica, er det ønskelig at den spesielle syre som anvendes er spiselig og ikke-giftig.
Når 2-(4'-thiazolyl)-5 (6)-fluor-benzimidazol oppløses i ethanol og et overskudd av ethanolisk hydrogenklorid og ethylether tilsettes, og blandingen avkjøles og filtreres fåes krystaller av hydrokloridsyreaddisjonssaltet av benzimidazol en.
Eksempel 6
l- benzoyl- 2- ( 4'- thiazolyl) - 5 ( 6) - fluor- benzimidazol
Til 400 ml benzen og 175 ml dimethylformamid ble tilsatt 20 g 2-(4'-thiazolyl)-5 (6)-fluor-benzimidazol og 3 g natriumhydrid. Natriumhydridet ble tilsatt som en 50%-ig emulsjon i olje blandet med 10 ml benzen. Tilsetningen ble utført sakte ved 60°C og omrøringen ble fortsatt i 45 minutter efter tilsetningen. 14 g benzoylklorid ble så sakte tilsatt, og den dannede blanding ble oppvarmet i ytterligere 40 minutter ved 60°C. Reaksjonsblandingen ble så avkjølt til ca. 10°C, 15 ml vann ble tilsatt, og blandingen ble så vasket med 5%-ig natriumbicarbonatoppløsning. Oppløsningen ble så filtrert, og filtratet ble konsentrert til tørrhet i vakuum. Residuet ble omkrystal-lisert fra ethylether hvorved man fikk l-benzoyl-2-(4'-thiazolyl)-5 (6)-fluor-benzimidazol. t
Forbindelsene ble undersøkt på effektivitet mot ascarider ved å fore grupper på 10 mus hver på dietter inneholdende 0,3 vekt% av den forbindelse som skulle prøves. Efter én dag på dietten inneholdende den aktive forbindelse ble musene indku-lert oralt med 40.000 larvale egg av Ascaris lumbricoides var. Suum. Tilførselen av aktiv forbindelse ble så fortsatt i ytterligere ni dager hvorpå prøvedyrene ble av-
livet og deres lunger undersøkt på tegn på blødnings skader frembragt av migrerende ascarid-larver. Resultatene av prøvene var som følger:
Som det sees av ovenstående tall har fremgangsmåteforbindelsene bedre ak-
tivitet mot ascarider enn den som utvises av det vellykkede kommersielle produkt.
Ingen av disse forbindelser var giftige overfor forsøksdyrene.
Claims (1)
- Fremgangsmåte ved fremstilling av anthelmintisk virksomme forbindelser av den generelle formel: ,hvor R er furyl eller thiazolyl, R^ er hydrogen eller benzoyl, og den ene av substi-tuentene R^ og R^ er amino, lavere-alkylamino eller di-lavere-alkylamino, og den annen er hydrogen, eller R^ og/eller R^ er halogen, og syreaddisjonssalter derav,karakterisert ved at enten a) en forbindelse av den generelle formel: hvor R^, R3 og R^ er som ovenfor angitt, omsettes under oppvarmning med en forbindelse av den generelle formel: R - D^. hvor R er som ovenfor angitt, og Dj er carboxy, carbalkoxy, carbamoyl, cyano eller halogen, i nærvær av en mineralsyre, eller b) en forbindelse av den generelle formel: hvor R„ og R er som ovenfor angitt, omsettes med en forbindelse av den generelle formel: R - D„, hvor R er som ovenfor angitt, og D er carboxy, carbalkoxy eller halogen, i et inert oppløsningsmiddel, hvorpå nitrogruppen i det dannede anilid reduseres, og anilidet ringsluttes ved reduksjon, eller c) en forbindelse av den generelle formel: hvor R_ «3 og R4. er som ovenfor angitt, omsettes med en forbindelse av den generelle formel: R - D , hvor R er som ovenfor angitt, og D„ er carboxy, carbalkoxy, carbamoyl, cyano eller halogen, i et inert organisk oppløsningsmiddel, den erholdte forbindelse nitreres og den innførte nitrogruppe reduseres og det dannede anilid ringsluttes, eller d) en forbindelse av den generelle formel: hvor R, R og R. er som ovenfor angitt, omsettes med et positivt halogenerings-middel som er istand til å halogenere nitrogenatomet i amidingruppen, eventuelt under tilsetning av en base, hvorpå det dannede N-halogen-N'-fenylamidin omsettes med en base, eller e) når i trinn a) - d) fremstillede forbindelser er hydrogen, omsettes eventuelt den dannede forbindelse med et alkalimetallhydrid og derpå med et benzoylhalogenid i et inert oppløsningsmiddel, og at de dannede forbindelser eventuelt overføres til syreaddisjonssalter på i og for seg kjent vis.
