Abstract
Analogifremgangsmåte til fremstilling av terapeutisk virksomme cefalosporansyrederivater.
Description
Foreliggende oppfinnelse angår fremstilling av nye cefalosporansyrederivater som er virksomme mot både Gram-positive og Gram-negative patogene bakterier.
De nye forbindelser som fremstilles ifølge foreliggende oppfinnelse har den generelle formel:
hvor M og M' kan være like eller forskjellige og betyr hydrogen, alkalimetaller, jordalkalimetaller eller organiske aminer, og X er -OCOCH3 eller
M og M<1> kan være kalium, kalsium, magnesium eller andre ugiftige alkali- eller jordalkalimetaller. Videre kan M og M' være slike ugiftige organiske aminer som trietylamin, procain, dibenzylamin, N-benzyl-g-fenetylamin, 1-efenamin, N,N'-bisdehydroabietyletylendiamin og lignende. Andre aminer kan brukes i den grad de har vært brukt til saltdannelse med benzylpenicillin.
De fremstilte forbindelser har både sulfo- og karboksylgrupper, og kan være enten sure eller nøytrale salter avhengig av ovenstående syregruppers surhetsgrad. Forbindelsene som fremstilles i henhold til oppfinnelsen,
kan eksistere i to optisk aktive former (D- og L-diastereo-isomerel da a-karbonatornet er asymmetrisk.
Forbindelsene er aktive mot Psudomonas aeruginosa, Escherichia coli, Proteus vulgaris, Proteus morganii,
Proteus mirabilis, Klebsiella pneumoniae, Staphylococcus aureus, Bacillus subtilis, Sarcina lutea og lignende. Forbindelsene kan gis ved oral administrasjon, subkutant eller intravenøst. Dosene ligger i området 5-500 mg pr. kg pr. dag, fortrinnsvis ca. 10-200 mg pr. kg pr. dag, ved 2-4 gangers administrasjon pr. dag, mot f.eks. pseudomonas-infeksjon.
De fremstilte forbindelser har meget høyere aktivitet mot Streptomyces aureus, som representerer en penicillin G-resistent stamme, enn konvensjonelt a-sulfo-penicillin. Videre har de nye forbindelsene en sterk virkning overfor Pseudomonas aeruginosa mens den beslektede kjente a-karboksy-cefalosporinforbindelsen, som det frem-går av nedenstående tabell, ikke er virksom overfor denne mikroorganisme. Dette er uventet på bakgrunn av den kjente forbindelses struktur. Det ble oppnådd følgende data over verdier for minimal inhiberende konsentrasjon (MIC):
Ifølge foreliggende oppfinnelse fremstilles de virksomme nye cefalosporansyrederivatene ved at en 7-aminocefalosporansyre med formelen: hvor X har den ovenfor angitte betydning, salter derav eller lett dekomponerbare estere derav, omsettes med a-sulfofenyl-eddiksyre med formelen:
eller med funksjonelle derivater derav,
hvoretter, om ønsket, omsettes en erholdt forbindelse hvor X er -OCOCH^, med 2-merkaptopyridinoksyd, en eventuell estergruppe avspaltes, og, om ønsket, omdannes erholdte forbindelser til et ønsket salt.
Utgangsforbindelsen a-sulfofenyleddiksyre kan fås
i handelen og fremstilles ved å sulfonere tilsvarende
karboksylsyrer med svovelsyreanhydrid. Sulfogruppen kan,
om ønsket, beskyttes i form av sulfonamid eller sulfonat under kondensasjonstrinnet med 7-aminocefalosporansyre.
Funksjonelle derivater av a-sulfofenyleddiksyre
er karboksylsyrehalogenid som -klorid eller -bromid,
blandede anhydrider som lavere alkyloksy- eller aralkyloksy-karbonsyreanhydrid, ot-toluensulfonsyreanhydrid, azid eller aktiverte estere slik som med p-nitrofenol, 2,4-dinitro-fenol eller pentaklorfenol eller karboazid. Disse derivater kan fremstilles fra a-sulfofenyleddiksyre på kjent måte. F.eks. fremstilles syrehalogenidet ved å behandle a-sulfofenyleddiksyre med et halogeneringsmiddel som f.eks. tionylklorid, fosforoksyklorid eller fosforoksybromid.
Salter av 7-aminocefalosporansyre er salter
dannet ved aminogruppen som hydrokloridet, sulfater, ace-tater og lignende, eller salter dannet ved karboksylsyre-gruppen som natrium-, kalium-, trietylamin-salter eller lignende.
De lett dekomponerbare estere, f.eks. trimetyl-silylester, kan fremstilles på en hvilken som helst kjent måte fra 7-aminocefalosporansyre.
Foreliggende fremgangsmåte omfatter som nevnt kondensasjon av aminogruppen i 7-aminocefalosporansyren med a-sulfofenyleddiksyrens karboksylgruppe. Metoder for opp-nåelse av dette er kjent for fagfolk, og foretrukne metoder er: 1. 7-aminocefalosporansyre eller dennes salter kondenseres med a-sulfofenyleddiksyre i nærvær av et dehydrati-seringsmiddel. Dehydratiseringsmidlet er f.eks. N,N'-dicykloheksylkarbodiimid, N,N'-diisopropylkarbodiimid eller N,N'-karbonyltriazol, 2. funksjonelle derivater av a-sulfofenyleddiksyre kondenseres med 7-aminocefalosporansyre eller dennes salter og 3. lett dekomponerbare estere av 7-aminocefalosporansyre kondenseres med funksjonelle derivater av a-sulfofenyl-eddiksyre .
Disse kondensasjoner kan gjennomføres i nærvær av oppløsningsmidler. Oppløsningsmidlene kan være vann, dioksan, aceton, dimetylformamid, tetrahydrofuran, diklormetan, kloroform, etylendiklorid, toluen, benzen eller en blanding av disse. Ved metodene 1 og 3 ovenfor er vann ikke gunstig. Ved metode 2 og 3 ovenfor foretrekkes nærvær av baser. Basene er organiske baser som f.eks. trietylamin, tributyl-amin, dimetylanilin, pyridin, pikolin, chinolin. Eventuelt er basene uorganiske baser som natriumkarbonat, kaliumkar-bonat, natriumbikarbonat, kaliumbikarbonat, natriumhydroksyd, kaliumhydroksyd, kalsiumhydroksyd eller lignende. Kondensasjonen utføres ved en temperatur på mellom -5 og 40°C, fortrinnsvis mellom -2 og 10°C. Den øvre grense er 50°C på grunn av at cefalosporansyreproduktet lett dekom-poneres .
Ved anvendelse av silylesteren i metode 3 ovenfor be-handles det fremstilte produkt med vann eller en lavere alifatisk alkohol for å dekomponere og fjerne estergruppen.
Etter at kondensasjonen er ferdig kan produktet renses på kjente måter. Slike fremgangsmåter er oppløsnings-middelekstraksjon, konsentrasjon, kromatografering, krystal-lisasjon, omkrystallisasjon og lignende. Således kan man få produktet i form av frie baser eller salter. Saltene kan om ønskes omdannes til andre ugiftige salter på forøvrig kjent måte.
Oppfinnelsen skal nå beskrives mer detaljert ved hjelp av de nedenstående eksempler.
Eksempel 1
Fremstilling av 7-( a- sulfofenylacetamido)- cefalosporansyre
I en kolbe forsynt med rører fylte man 2,5 ml IN NaOH-oppløsning og 5 ml vann. Blandingen ble avkjølt til 0-5°C i et bad av isvann og under mekanisk omrøring innførte man 680 mg 7-aminocefalosporansyre. Deretter tilsatte man dråpevis en separat fremstilt oppløsning av 585 mg a-sulfo-fenylacetylklorid i 7 ml dietyleter i løpet av 15 minutter. Blandingen ble omrørt i 15 minutter, og vannsjiktet ble separert fra reaksjonsblandingen og dette sjiktets pH regu-lert til 1,5 med IN HC1. Deretter ble sjiktet ekstrahert to ganger med 15 ml n-butanol, ekstraktet vasket to ganger med hver gang 5 ml vann og deretter ekstrahert ytterligere med en mettet vandig NaHCO-^-oppløsning.. Ekstraktet ble innstilt på pH 6,5. Etter rensing på kromatografikolonne pakket med polystyrenpulver ble eluatfraksjoner som inne-holdt den aktuelle forbindelse frysetørket til et hvitt pulver i form av et natriumsalt. Produktet ble tørket og analysert, og man fikk følgende resultater:
Eksempel 2
Fremstilling av 7-( a- sulfofenylacetamido)- cefalosporansyre
3,78 g 7-aminocefalosporansyre ble suspendert i 30 ml kloroform og 2,3 g heksametyldisilazan ble tilsatt. Blandingen ble oppvarmet ved 65°C i 1,5 timer i varmt vann-bad. Den gjennomsiktige oppløsning ble destillert under nedsatt trykk ved 40°C. Restforbindelsen ble oppløst på nytt i 50 ml kloroform og man tilsatte 1,94 g trietylamin. Deretter ble denne oppløsning avkjølt og 30 ml kloroform-oppløsning inneholdende 4,3 g a-sulfofenyleddiksyreklorid ble tilsatt dråpevis. Blandingen ble omrørt ved 15°C i 15 minutter. Etter avsluttet reaksjon ble reaksjonsblandingen konsentrert ved romtemperatur under nedsatt trykk. Deretter oppløste man produktet i 50 ml vann. Den vandige oppløsning ble innstilt på pH 1,5 og ekstrahert med to ganger 35 ml n-butanol. Butanolsjiktet ble vasket med en liten mengde vann og deretter ekstrahert igjen med en vandig
natriumhydrogenkarbonatoppløsning. Etter innstilling av pH på 6, ble den aktuelle forbindelse utkrystallisert fra oppløsningen og tørket. Man fikk 4,3 g natriumsalt. Resul-tatene av absorbsjonsspektra var som i eksempel 1.
Eksempel 3
Fremstilling av 7-( a- sulfofenylacetamido)- 3-[ 2-( 1- oksydo-pyridyl)- tiometyl]- A 3- cefem- 4- karboksylsyre:
0,217 g 7-(a-sulfofenylacetamido)-cefalosporansyre og 0,164 g 2-merkaptopyridinoksyd ble oppløst i 15 ml vann, og væskens pH innstilt på 6,4. Blandingen ble oppvarmet til 4 0°C i 18 timer. Etter avsluttet reaksjon ble produktet frysetørket til 0,350 g pulver. Det ønskede produktet ble opparbeidet ved hjelp av kolonnekromatografi på cellulose-pulver og eluert med et blandet oppløsningsmiddel av n-propa-nol, vann og trikloreddiksyre (75 : 24 : 1). Eluatet destillert under nedsatt trykk og vasket med heksan. Uopp-løselig stoff ble kombinert og oppløst i vann. Etter innstilling av den vandige oppløsningens pH på 6,4, ble den søkte forbindelsen frysetørket. Produktutbyttet var 0,080 g. Resultatet av IR-spektrometri var som følger: IR v <KBr> (cm-1): 3450(OH), 1765(laktam), 1680

- Analogifremgangsmåte til fremstilling av terapeutisk virksomme cefalosporansyrederivater med den generelle formel:HVOR M og M<1> kan være like eller forskjellige og betyr hydrogen, alkalimetaller, jordalkalimetaller eller organiske aminer, og X er -OCOCH^ ellerkarakterisert ved at en 7-aminocefalosporansyre med formelen:hvor X har den ovenfor angitte betydning, salter derav eller lett dekomponerbare estere derav, omsettes med a-sulfofenyleddiksyre med formelen:eller med funksjonelle derivater derav,hvoretter, om ønsket, omsettes en erholdt forbindelse hvor X er -OCOCH3, med 2-merkaptopyridinoksyd, en eventuell estergruppe avspaltes, og, om ønsket, omdannes erholdte forbindelser til et ønsket salt.
