Abstract
Drøvtygger-for.
Description
Foreliggende oppfinnelse angår et nytt for for
ernæring av drøvtyggere som har en utviklet vomfunksjon, inne-holdende karbohydrater, proteiner, kilder for grovt for og uorganiske salter. Foret karakteriseres ved at det også inne-
holder en vom-virksom konsentrasjon av et vekstfremmende antibiotikum valgt fra A28695A, A28695B og de fysiologisk akseptable salter derav.
De to nevnte antibiotiske stoffer og fremstilling av
disse er beskrevet i vårt patent 136.981.
Foreliggende oppfinnelse anvendes i forbindelse med drøvtyggere som har en utviklet vom-funksjon.. Unge drøvtyggere, hovedsakelig de som ikke er avvendt, fungerer som éri-mavede dyr.
De utnytter sitt enkle, flytende for slik som én-mavede dyr gjør. gjør. Når de unge drøvtyggere begynner å spise fast føde som inneholder cellulose, stivelse og andre karbohydrater, begynner vom-funksjonen å utvikles, og den mikrobiologiske populasjon i vommen begynner å øke. Efter at dyret har spist fast føde en tid, når vom-funksjonen sin fulle utvikling og fortsetter å funksjonere gjennom dyrets hele levetid.
Foreliggende oppfinnelse er anvendbar for alle drøv-tyggere, dvs. dyr som har flere maver hvorav én er vommen.
Økonomisk viktige drøvtyggere er kveg, sauer og geiter.
Det vil forstås at anvendbarheten av metoden ikke er begrenset til dyr som skal fetes eller til unge dyr som vokser. Når metoden anvendes på voksne dyr, slik som melkekyr eller.avlsdyr, får man fordelene i form av redusert for-forbruk.
Dyrene må gis en mengde antibiotikum som er virksom
for vommen for å få fordelene som metoden gir. Minste mengde som er virksom, varierer avhengig av dyrenes diett, tilstand og alder.
Et tilfredsstillende område for tilsetning er fra 0,02 mg/kg/dag til 2,0 mg/kg/dag. Det foretrukne området er fra 0,05 mg/kg/dag til 1 mg/kg/dag.
Eksempel 1
Vomvæske som ble oppnådd fra en stut som hadde en kirurgisk innsatt røråpning i vommen, ble anvendt. Stuten ble holdt på et for med høyt korn-innhold, og sammensetningen var som følger:

En prøve av vomvæske filtreres gjennom 4 skikt av osteklede og eluatet samles opp i en vakuumflaske. Det partikkelformede materiale som holdes tilbake på ostekledet, suspenderes igjen i nok fysikalsk buffer for å få det opprinnelige volum av vomvæske, og eluatet filtreres enda en gang. Den anvendte buffer er beskrevet nedenfor.

De to eluatene kombineres i en skilietrakt og får stå tii det partikkelformede materiale skiller seg ut på toppen.
Det klare skiktet fortynnes derefter 1:1 med den samme buffer og justeres til pH 7,0. 10 ml av fortynnet vomvæske plasseres i en 25 ml kolbe med 40 mg av det samme for som vist ovenfor. 5 mg av soyabønne-protein tilsettes også pr. kolbe. Forbindelsen som skal under-søkes, innveies til hver forsøkskolbe. Fire paralleller anvendes for hver behandling.
To sett som består av fire ubehandlede blindprøve-flasker, gjøres også i stand. Ett sett på fire flasker inkuberes i 16 timer ved 38°C med prøveflaskene. Det andre settet på fire ubehandlede blindprøveflasker er null-tidskontroll, og til disse tilsettes 2 ml 25%ig metafosforsyre så snart flaskene er klare for å stoppe fermenteringen.
Fermenteringen i den inkuberte prøven og flaskene ved blindprøven stoppes efter 16 timer ved tilsetning av 2 ml 25%ig metafosforsyre til hver kolbe.
Alle prøvene får lov til å bunnfelle og overliggende væske analyseres ved hjelp av gasskromatografiske metoder på acetat, propionat og butyrat.
Analysetallene for hver flyktig fettsyre (VFA) som man finner i null-tidsblindprøvene, trekkes fra analyseverdiene i de ubehandlede blindprøvene og i prøvekolbene. De resulterende verdier angir mengden av hver VFA fremstilt i løpet av den 16 timers lange fermenteringsperioden. Man finner gjennomsnitts-verdien fra verdiene fra de fire parallellkolbene.
Tallene i det følgende angir forholdet mellom VFA fremstilt i behandlede flasker, og VFA fremstilt i ubehandlede blind-prøvekolber. Denne metoden å angi dataene på viser klart resultatene av de kjemiske forandringer i vommen, som skyldes forbedring av forutnyttelsen.
Tallene ovenfor, hvor en rekke forsøk er utført med en gitt tilsetning, kan kombineres for å gi de følgende gjennom-snittsverdier .
Tallene ovenfor viser at ved hvert forsøk ble propionat-nivået øket over blindprøveverdien. I 19 av de 29 angitte forsøk var acetatnivået redusert sammenlignet med den ubehandlede blind-prøve. økningen i propionat i forhold til acetat er spesielt merkbart når man ser på tabellen for de kombinerte analysetall, hvor den naturlige variasjon i biologiske data er utjevnet. Disse tabellene viser at behandling med A28695 ved alle tilsetninger merkbart øker propionat i forhold til acetat sammenlignet med blindprøveverdiene.
Butyratnivåene er jevnt redusert i forhold til blind-prøveverdiene. Som det tidligere er blitt forklart, siden butyrat fremstilles av rumen fra lite effektivt fremstilt acetat, er reduksjonen i butyratnivåene ikke skadelig for forutnyttelsen.
Vi har funnet at antibiotika-forbindelsene øker fremstillingen av propionat i forhold til fremstillingen av acetat i drøvtyggere når disse tilsettes oralt til dyrene. Den enkleste måte å tilsette antibiotika er ved å blande disse i dyreforet.
I det følgende er gjengitt en tabell med resultater av sammenligningsforsøk, som viser at antibiotikum A28695A er mer effektivt enn det i østerriksk patent 292.194 beskrevne antibiotikum A204 med hensyn til å øke produksjonen av propionsyre i vommen.

Disse data er for 5 eller 6 undersøkelsesdager i løpet av en
14 dagers behandlingsperiode.

Forblandingene er nye på grunn av tilstedeværelsen av A28695; dyreforene som sammensetningen er basert på, kan være et hvilket som helst for som er egnet som næring for drøvtyggere som har en utviklet vom-funksjon.
Den passende formulering av dyrefor som gir den nødvendige ernæring til drøvtyggere, er velkjent. Fagmannen er velkjent med formuleringen av for som gir dyrene næring og som utnytter den økonomiske situasjon i de forskjellige områder, og de er vel i stand til å formulere for uten at man skal nærmere omtale dette her. Den forbedrede forblanding inneholder minst en vom-virksom konsentrasjon av A 28695. En vom-virksom konsentrasjon er den som gir en vom-virkende dose til dyret når det konsumerer sin daglige rasjon av for. Den vom-virksomme konsentrasjon varierer selvfølgelig med dyrets vekt og den mengde f5r som det konsumerer daglig. Vanligvis ligger den vom-virksomme konsentrasjon i området fra 2 g pr. tonn for til 200 g A28695 pr. tonn for. Det foretrukne området for konsentrasjoner av A28695 er fra
5 g pr. tonn til 100 g pr. tonn f5r. Konsentrasjoner av A28695 innenfor disse områder gir generelt effektive mengder av A28695 til de behandlede dyr. Fagmannen på området vil imidlertid innse
at i spesielle tilfeller, f.eks. når de dyrene som skal behandles, konsumerer spesielt små eller spesielt store mengder for i forhold til sin vekt, kan det være passende med konsentrasjoner som ligger utenfor de angitte områder,,
Metodene for formulering av antibiotika i dyrefor er velkjent» Det er vanligvis alminnelig å fremstille en forblanding som et råmateriale for forene som er behandlet med lege-midlet. Egenskapene av forblandingen overføres til egenskapene i foret som fremstilles fra forblandingen. Ved formuleringen av forblandingen tar man hensyn til at det skal være lett å fremstille f6r fra forblandingen og til økonomien. Naturen av de aktive ingredienser i forblandingen og den biologiske virkning av den aktive ingrediens er irrelevant ved formuleringen av forblandingen. Forblandingen kan inneholde 2,2-880 g/kg A28695, alt efter hva som er praktisk når det gjelder å blande for som inneholder den ønskede konsentrasjon av A28695. Forblandingene kan enten være flytende eller faste.
Den forbedrede for-forblanding, som er ny på grunn av tilstedeværelsen av antibiotikumet A28695, formuleres i hvilke som helst av de alminnelig anvendte, fysiologisk aksepterbare bærere„ Flytende bærere som er anvendbare for forblandingen, omfatter glykoler slik som polyetylenglykoler av forskjellige molekylvekter og propylenglykol, inerte oljer som omfatter vegetabilske oljer og høyt raffinerte mineraloljer og fysiologisk aksepterbare alkoholer slik som etanol. Vann og vandig alkohol er en virskom, flytende bærer for forblanding for mange av formene av A28695. Faste forblandingsbærere omfatter vermikulit, diatomé-jord, fysiologisk aksepterbare leirer slik som attapulgit og montmorillonit og granulerte eller pulveriserte f6r-komponenter slik som knust mais, soyamel, alfalfamel, risskolmer, maiskolber, knust hvete og havre og avfallsstoffer ved kornfremstillingen.
Alle metodene for formulering, blanding og tablett-fremstilling av for som vanligvis anvendes for drøvtyggere, er egnet for fremstillingen av f6r som inneholder de antibiotiske forbindelser ifølge foreliggende oppfinnelse.
Det er vanligvis økonomisk å behandle dyr, også drøv-tyggere, med en rekke vekstøkende midler, sykdomshindrende midler og sykdomsbehandlende midler gjennom hele livet. Slike medika-menter anvendes ofte i kombinasjon. Foreliggende for kan anvendes i kombinasjon med andre forblandinger eller tilsetninger.
Som vist forandrer oral tilsetning av A28695 fordelaktig fremstillingen av propionater i forhold til fremstillingen av acetater i vommen. De én-mavede dyr som det refereres til er de som konsumerer fiberformet, vegetabilsk føde og som fordøyer minst deler av det ved hjelp av mikrobiologisk fermentering i cekum. Den cekale fermentering følger et kjemisk forløp tilsvarende vom-fermenteringen.
Hester, griser og kaniner er eksempler på dyr som for-døyer en del av føden ved hjelp av cekal fermentering. Den totale forutnyttelse for slike dyr forbedres ved oral tilsetning av A28695 ved hjelp av en gunstig forandring i propionat/acetat-forholdetu Hester og kaniner er eksempler på dyr hvor cekal-fermenteringen utgjør hoveddelen av den totale fordøyelsesprosess,
og hvor A28695 er spesielt gunstig.
Claims (4)
1. Drøvtyggerfor som inneholder karbohydrater, proteiner og kilder for grovt f5r og uorganiske salter, for drøvtyggere med ferdig utviklet vom-funksjon, karakterisert ved at det også inneholder en vekstfremmende mengde av antibiotikum A28695A, A28695B eller fysiologisk aksepterbare salter derav.
2. For som angitt i krav 1,
karakterisert ved at antibiotikumet er til stede i en mengde fra 2 til 200 g pr. tonn f6r.
3. For som angitt i krav 1 eller 2, karakterisert ved at antibiotikumet er det blandede natrium-kaliumsalt av A28695A.
4. For som angitt i krav 1 eller 2, karakterisert ved at antibiotikumet er det blandede natrium-kaliumsalt av A28695B„
