Description
Foreliggende oppfinnelse angår en analogifremgangsmåte ved fremstilling av 1'-(2-hydroxyethyl)-1'-demethylclindamycin [ 7-halogen-7-desoxy-1 * -(2-hydroxyethyl) -1.' -demethyllincomycin] og analoger derav, såvel som estere og ethere derav.
Disse
forbindelser har clindamycin-lignende antibakteriell aktivitet og kan anvendes på lignende-måte. De har dessuten forbedret gram-
negativ aktivitet og har lavere giftighet overfor pattedyr.
De nye forbindelser som fremstilles ifølge oppfinnelsen, kan
betegnes med følgende strukturformel:
hvor én eller alle av 2-, 3- og 4-hydroxygruppene kan være for-
esteret eller foretheret , Halo er klor, brom eller jod, R er alkyl med inntil 4 carbonatomer eller 2-hydroxyethyl, og Ac er acyl-
gruppen av en L-2-pyrrolidin-carboxylsyre av formel:
hvor R^ er alkyl med inntil 8 carbonatomer.
De nye forbindelser fremstilt ifølge oppfinnelsen med formel I fremstilles ved at en forbindelse av formel I hvor hvilke som helst eller alle av 2-, 3- °9 4-hydroxygruppene kan være foresteret eller foretheret, og hvor Halo og R er som ovenfor angitt og Ac er acylgruppen av en L-2^pyrrolidin-carboxylsyre av formelen:
hvor R^ er som ovenfor angitt, N-alkyleres med et 2-hydroxyethyl-alkyleringsmiddel, og en erholdt forbindelse med formel I hvor 2-, 3- og/eller 4-hydroxygruppene er frie, kan derpå eventuelt foresteres eller foretheres på i og for seg kjent vis.
Fremstilling av forbindelsene av formel IAD er beskrevet i U.S. patenter 3 496 163 og 3 502 648-
N-alkyleringen av forbindelser åv formel IAD kan utføres ved å omsette utgangsforbindelsen med ethylenoxyd. Utgangsmaterialet, fortrinnsvis som hydrokloridsaltet, oppvarmes med ethylenoxydet, fortrinnsvis i et trykk-kar, i hvilket reaksjonen forløper som følger:
Reaksjonsblandingen opparbeides efter avkjøling på vanlig måte under anvendelse av fremgangsmåter som destillasjon, oppløs-ningsmiddelekstraksjon og krystallisasjon.
Mengdene er.ikke kritiske, men et overskudd av ethylenoxyd
er ønskelig. Et overskudd på fra 2.- 100 ganger den støkiometriske mengde er passende. Temperaturen er heller ikke kritisk. Ved temperaturer under ca. 35°C er reaksjonen imidlertid uønsket lang-som, og vanligvis vil det ikke være nødvendig eller ønskelig å overstige ca. 200°C.. Reaksjonen forløper godt ved 100°C.
Reaksjonen utføres med fordel i et inert felles oppløsnings-middel for reaktantene. Passende slike oppløsningsmidler innbe-fatter methylalkohol, ethylalkohol, propylalkohol, benzen, toluen, cyclohexan og tetrahydrofuran.
Alternativt kan alkyleringen utføres ved en reduserende alkylering som beskrevet i U.S. patent 3 49° 163, del Gl-4. ved å erstatte formaldehydet (formalin) med hydroxyacetaldehyd.
En hvilket som helst av eller alle 2-, 3- °9 4-hydroxygruppene kan forestres før reaksjonen, f.eks. med hydrocarbon-carboxylsyrer , fortrinnsvis med inntil. 18 carbonatomer , eller halogen-, nitro-, hydroxy-, amino-, cyano-, thiocyano- eller alkpxy-substituerte hydrocarbon-carboxylsyrer, fortrinnsvis med inntil 18 carbonatomer. Disse acylater har også antibakteriell aktivitet .
De kan også foresteres efter reaksjonen, men i det tilfelle blir også hydroxygruppen av 2-hydroxyethylgruppen foresteret. I virkeligheten kan man ved å anvende en ekvimolar mengde, eller mindre, av foresteringsmidlet få et 1 *-(2-acyloxyethyl)-1 *-demethyl-clindamycin eller analog derav uten at 2-, 3- eller 4-hydroxygruppene acyleres. De således erholdte monoacylater har også clindamycinlignende antibakteriell aktivitet og kan anvendes på lignende måte.
En eller alle av 2-, 3- og 4-hydroxygruppene kan også foretheres, f.eks. med alkyl, fortrinnsvis med.ikke over 20 carbonatomer; cycloalkyl, fortrinnsvis med 3 - 12 carbonatomer; eller yliden (f.eks. 3,4-O-yliden)-gruppen av f.eks. alkyliden, fortrinnsvis med ikke over 20 carbonatomer og aralkyliden og vinyl-oger derav, fortrinnsvis med ikke over 12 carbonatomer» Disse ethere har også antibakteriell aktivitet.
Forbindelsene av formel I foreligger enten i den protonerte eller ikke-protonerte form avhengig av omgivelsenes pH. Når den protonerte form menes, betegnes forbindelsen som et syreaddisjons - salt, og når den ikke-protonerte form menes, betegnes den som den frie base. Den frie base kan overføres til stabile syreaddisjons - salter ved å nøytralisere den frie base med den passende syre til under ca. pH 7,0, og fortrinnsvis til ca. pH 2 - 6.
Syreaddisjonssaltene kan anvendes til de samme formål som
den frie base, og de kan anvendes til å rense disse. Eksempelvis kan den frie base overføres til et uoppløselig salt, som picratet, som kan underkastes rensebehandlinger, f.eks. oppløsningsmiddel-ekstraksjoner og vaskinger, kromatografi, fraksjonert væske-væskeekstraksjoner og krystallisasjon, og så anvendes til å regenerere den frie baseform ved behandling med alkali eller for å fremstille et annet salt ved metathese. Eller den frie base kan overføres til et vannoppløselig salt, som hydrokloridet eller sulfatet, og den vandige oppløsning av saltet kan ekstraheres med forskjellige med vann ublandbare oppløsningsmidler før regenerering av den frie baseform ved behandling av den således ekstraherte syreoppløsning eller overføres til et annet salt ved metathese.
De frie baser av formel I er gode medier for giftige syrer. Eksempelvis er fluorkiselsyre-addisjonssaltene nyttige som møll-impregneringsmidler ifølge U.S. patenter 1 915 334 og 2 075 359, og hexafluorarsensyre- og hexafluorfosforsyre-addisjonssaltene er nyttige som parasitticider ifølge U.S. patenter 3 122 536 og 3 122 552. Thiocyansyreaddisjonssaltet danner, når det kondenseres med formaldehyd, harpiksaktige materialer som er nyttige som beis-ningsinhibitorer i henhold til U.S. patenter 2 425 320 og 2 6o6 155.
De nære analoger av clindamycin, dvs. hvor -R^ er cis- eller trans-alkyl med inntil 8 carbonatomer, og R er methyl, ethyl eller 2-hydroxyethyl, har antibakterielle egenskaper, og noen er sammen-lignbare med eller bedre enn lincomycin og kan anvendes til de samme formål som lincomycin. De andre analoger og isomerer har lignende antibakterielle egenskaper og kan anvendes til de samme
formål som lincomycin hvor større mengder ikke er uheldige.
De fem forbindelser:
forbindelse 1 = forbindelsen ifølge eksempel 1,
forbindelse 2 = forbindelsen ifølge eksempel 2,
forbindelse 3 = forbindelsen ifølge eksempel 3, cis-isomeren, forbindelse 4 = forbindelse 3, trans-isomeren, og forbindelse 5 = 7-bromanalogen av forbindelsen fra eksempel 1,
ble prøvet side om side med clindamycin på forskjellige tidspunkter. Det er vanlig at MIC (minimum inhiberingskonsentrasjon, p.g/ml) vil. variere litt fra ett forsøk til et annet på grunn av forsøksfeil. Det viktige er at forbindelsene ifølge oppfinnelsen og standarden, clindamycin, lble sammenlignet side om side under identiske beting-elser når de ble sammenlignet. Data for disse sammenligninger som viser den forbausende aktivitet sammenlignet med clindamycin, er angitt i tabell 1-5- De numeriske data er MIC.
Disse data viser den forbausende og bedre aktivitet for:
a) forbindelse 1 sammenlignet med clindamycin for fem av de seks forsøksorganismer, b) forbindelse 2 sammenlignet med clindamycin for fire av de åtte forsøksorganismer, c) forbindelse 3 sammenlignet med clindamycin for fem av de seks forsøksorganismer, d) forbindelse 4 sammenlignet med clindamycin for fem av de åtte forsøksorganismer, og e) forbindelse 5 sammenlignet med clindamycin for fire av de syv forsøksorganismer.
Tabell 5 viser videre den forbausende og bedre aktivitet av forbindelse 1. Tabellen viser at forbindelse 1 konsentreres i urinen i større mengder enn clindamycin. Dette indikerer at denne forbindelse sannsynligvis ville være meget nyttig ved behandling av bakterielle urinveisinfeksjoner.
Figur 1 viser også den forbausende og bedre aktivitet av forbindelse 1. Figuren viser de høyere blodnivåer av forbindelse 1 sammenlignet med clindamycin når de gies oralt til rotter. Denne forbausende og bedre egenskap er meget fordelaktig fra et farma-søytisk og terapeutisk synspunkt.
De følgende eksempler er illustrerende for fremgangsmåten ifølge oppfinnelsen. Delene og prosentene er angitt i vekt, og oppløsningsmiddelforholdene er angitt i volum hvor annet ikke er anført.
Eksempel 1
1'-( 2- hydroxyethyl)- 1' - demethyl- clindamycin
En blanding av 10 g 1 *-demethyl-clindamycin.HCl, lOO ml ethylalkohol og 50 ml ethylenoxyd ble forseglet i en trykkolbe og oppvarmet ved 100°C i 1 time. Kolben ble så avkjølt til 0°C og åpnet og reaksjonsblandingen inndampet under vakuum til en viskøs ravfarvet olje. Oljen ble oppløst i 2O0 ml vann, og pH innstilt på 10 med vandig KOH-oppløsning. Ekst raksjon med kloro-form fulgt av inndampning av ekstraktene ga ca. IO g råprodukt. Dette materiale ble renset ved kromatografi over silicagel under anvendelse av et oppløsningsmiddelsystem bestående av ethyi-acetat :aceton:vann (8:5:1) for eluering. Prodiiktf raksjonene ble identifisert ved tynnskiktskroraatografi, forenet og inndampet, hvorved man fikk 2,0 g 1<*->(2-hydroxyethyl)-1<1->demethyl-clindamycin.

Eksempel 2
1'-( 2- hydroxyethyl)- 11- demethyl- cis- clindamycin
Reaksjonen ble utført på samme måte som i eksempel 1 idet der som utgangsmateriale ble anvendt den passende cis-isomer. Et utbytte på 1,1 g 1'-(2-hydroxyethyl)-1'-demethyl-cis-clindamycin ble erholdt.

Eksempel 3
1 *-(2-hydroxyethyl)-1 *-demethyl-4'-depropyl-4'-cis- og trans - pentyl - clindamycin
Reaksjonen ble utført på samme måte som i eksempel 1 idet der som utgangsmateriale ble anvendt 1'-demethyl-4'-depropyl--4'-cis/trans-pentyl-clindamycin (eksempel IO av U.S. patent 3 496 163). Man fikk et utbytte på 5,0 g 1'-(2-hydroxyethyl)-1<*->demethyl-4'-depropyl-4'-cis/trans-pentyl-clindamycin.

Claims (1)
- Analogifremgangsmåte ved fremstilling av terapeutisk aktive lincomycinderivater med formelen:hvor én eller alle av 2-, 3- og 4-hydroxygruppene kan være foresteret eller foretheret, Halo er klor, brom eller jod, R er alkylmed inntil 4 carbonatomer eller 2-hydroxyethyl, og Ac er acylgruppen av en L-2-pyrrolidin-carboxylsyre av formelen:hvor R^ er alkyl med inntil 8 carbonatomer,karakterisert ved at en forbindelse av formel Ihvor hvilke som helst eller alle av 2-, 3_ og 4-hydroxygruppenekan være foresteret eller foretheret , og hvor Halo og R er somovenfor angitt og Ac er acylgruppen av en L-2-pyrrolidin-carboxyl-syre av formelen:hvor R1 er, som ovenfor angitt, N-alkyleres med et 2-hydroxyethyl-alkyleringsmiddel, og en erholdt forbindelse med formel I hvor 2-, 3_ og/eller 4-hydroxygruppene er frie, kan derpå eventuelt foresteres eller foretheres på i og for seg kjent vis.
